51. Comparative pangenome analysis of capsulated Haemophilus influenzae serotype f highlights their high genomic stability

Abstract: Abstract Haemophilus influenzae is an opportunistic pathogen adapted to the human respiratory tract. Non-typeable H. influenzae are highly heterogeneous, but few studies have analysed the genomic variability of capsulated strains. This study aims to examine the genetic diversity of 37 serotype f isolates from the Netherlands, Portugal, and Spain, and to compare all capsulated genomes available on public databases. Serotype f isolates belonged to CC124 and shared few single nucleotide polymorphisms (SNPs) (n = 10,999), but a high core genome (> 80%). Three main clades were identified by the presence of 75, 60 and 41 exclusive genes for each clade, respectively. Multi-locus sequence type analysis of all capsulated genomes revealed a reduced number of clonal complexes associated with each serotype. Pangenome analysis showed a large pool of genes (n = 6360), many of which were accessory genome (n = 5323). Phylogenetic analysis revealed that serotypes a, b, and f had greater diversity. 52. Genetic Adaptation and Acquisition of Macrolide Resistance in Haemophilus spp. during Persistent Respiratory Tract Colonization in Chronic Obstructive Pulmonary Disease (COPD) Patients Receiving Long-Term Azithromycin Treatment

Abstract: ABSTRACT Patients with chronic obstructive pulmonary disease (COPD) benefit from the immunomodulatory effect of azithromycin, but long-term administration may alter colonizing bacteria. Our goal was to identify changes in Haemophilus influenzae and Haemophilus parainfluenzae during azithromycin treatment. Fifteen patients were followed while receiving prolonged azithromycin treatment (Hospital Universitari de Bellvitge, Spain). Four patients (P02, P08, P11, and P13) were persistently colonized by H. influenzae for at least 3 months and two (P04 and P11) by H. parainfluenzae. Isolates from these patients (53 H. influenzae and 18 H. parainfluenzae) were included to identify, by whole-genome sequencing, antimicrobial resistance changes and genetic variation accumulated during persistent colonization. All persistent lineages isolated before treatment were azithromycin-susceptible but developed resistance within the first months, apart from those belonging to P02, who discontinued the treatment. H. influenzae isolates from P08-ST107 acquired mutations in 23S rRNA, and those from P11-ST2480 and P13-ST165 had changes in L4 and L22. In H. parainfluenzae, P04 persistent isolates acquired changes in rlmC, and P11 carried genes encoding MefE/MsrD efflux pumps in an integrative conjugative element, which was also identified in H. influenzae P11-ST147. Other genetic variation occurred in genes associated with cell wall and inorganic ion metabolism. Persistent H. influenzae strains all showed changes in licA and hgpB genes. Other genes (lex1, lic3A, hgpC, and fadL) had variation in multiple lineages. Furthermore, persistent strains showed loss, acquisition, or genetic changes in prophage-associated regions. Long-term azithromycin therapy results in macrolide resistance, as well as genetic changes that likely favor bacterial adaptation during persistent respiratory colonization. IMPORTANCE The immunomodulatory properties of azithromycin reduce the frequency of exacerbations and improve the quality of life of COPD patients. However, long-term administration may alter the respiratory microbiota, such as Haemophilus influenzae, an opportunistic respiratory colonizing bacteria that play an important role in exacerbations. This study contributes to a better understanding of COPD progression by characterizing the clinical evolution of H. influenzae in a cohort of patients with prolonged azithromycin treatment. The emergence of macrolide resistance during the first months, combined with the role of Haemophilus parainfluenzae as a reservoir and source of resistance dissemination, is a cause for concern that may lead to therapeutic failure. Furthermore, genetic variations in cell wall and inorganic ion metabolism coding genes likely favor bacterial adaptation to host selective pressures. Therefore, the bacterial pathoadaptive evolution in these severe COPD patients raise our awareness of the possible spread of macrolide resistance and selection of host-adapted clones.

Abstract: This paper examines the influence of weaving variables such as yarn count, number of layers, warp and weft ratio, materials of the top layer, weft density and interlocking cell shape, and size on the thermal performance of multilayer interlocked woven fabrics. A split-plot design was used to construct a total of 64 fabric structures, which were assessed for thermal performance in terms of resistance to convective, conductive, and radiative heat. It was found that, for equal weft density and yarn number, protective performance improved with the number of fabric layers and with the presence of air cells between these layers, especially if air was not trapped within and could rather pass freely between the cells. An optimal combination of factors for the thermal response to the three types of heat was established via a Derringer–a much needed desirability function. The results of this paper are useful for identifying the interaction between configuration parameters and thermal performance, and hence for the design of improved heat protective clothing.

Abstract: BiD: Texts of University of Library and Information Science is a magazine founded by the Faculty of Library and Information Science in 1998. He is currently indexed by various databases, among which LISA, LIST, ISOC and Scopus. Since its launch, the magazine has positioned itself as an open forum for disseminating scientific papers on the area and acting speaker of the experiences of professional librarians (Pérez-Montoro and Ollé, 2013) channel. 15 years later, from June 2013 issue, the magazine has become co-published by the University of Barcelona and the University of Catalonia. In the UOC existed, meanwhile editing experience in a multidisciplinary journal and was UOC Papers (now defunct). In the poster the findings of this period of collaboration will be presented, highlighting the strengths and less strong in the same, emphasizing that each organization has gained in collaboration. Because collaboration between institutions is not only the division of the budget, but also a non-competitive collaborative vision and academia. As Rodríguez-point Yunta and Giménez-Toledo (2013), the merger is more than ever an option and a necessity. BiD Des advice for process and some will point to consider issues we have encountered along the way.

Abstract: There are different approaches to the study of the evolution of Library and Information Science (LIS) over the last 20 years. This paper does so by telling the story of an academic journal. We wanted to explore whether the discipline and its evolution could be understood in terms of the journal BiD: Textos universitaris en Biblioteconomia i Documentació. We chose to track different parameters for the articles published in BiD in the period from 1998 to 2014: the number of keywords used, the frequency variation in word use and the evolution of the signatory institutions in the papers regarding the ratio of professional to academic profiles of authors. This research is intended to complement the programme of the LIS-ER Seminar. Results will be presented for the first time at this seminar.

70. Palmitoylation in neurodegeneration : analysis of cysteine-string protein mutants linked with neuronal ceroid lipofuscinosis

Abstract: Neuronal Ceroid Lipofuscinoses (NCLs) are neurodegenerative lysosomal-storage disorders characterized by intracellular accumulation of autofluorescent material. Mutations in the DNAJC5 gene encoding Cysteine-String Protein alpha (CSPα) cause autosomal-dominant adult-onset NCL (ANCL). The disease-causing mutations occur within the cysteine-stringdomain (CSD), a region of the protein that is extensively palmitoylated. It has been shown that the ANCL CSPα mutants form aggregates and that this is dependent on palmitoylation. As aggregation is a common feature of neurodegenerative disorders, aggregates formed in ANCL are likely to contribute, together with loss-of-function effects, to disease pathology. The aims of this project were to: (i) investigate features of CSPα that are important for aggregate formation; (ii) identify molecular changes that occur in ANCL; and (iii) identify mechanisms regulating CSPα turnover to provide insight into pathways that might be perturbed in ANCL. A cluster of palmitoylated cysteines in the CSD were identified as essential for aggregation of the ANCL CSPα mutants, further supporting an association between palmitoylation and aggregation. Analysis of the expression levels of different proteins in post-mortem brain revealed a massive increase in expression of the palmitoyl thioesterase enzyme PPT1 in ANCL samples, which is intriguing as PPT1 functions to depalmitoylate proteins during their degradation and mutations in the PPT1 gene cause infantile NCL. Further analysis identified several other proteins that had altered expression levels including α-synuclein. Degradation of both wild-type and mutant CSPα proteins was mediated by the proteasome but was independent of lysine ubiquitination. This pathway mediated rapid degradation of non-palmitoylated protein, and blocking proteasome activity enhanced the formation of mutant aggregates. It will be important in future work to determine how mutations in CSPα affect protein degradation pathways to cause the characteristic NCL lysosomal morphology, and whether there is a role for the lysosome in degradation of palmitoylated or aggregated CSPα.

Abstract: The digital transition that drives the new industrial revolution is largely driven by the application of intelligence and data. This boost leads to an increase in energy consumption, much of it associated with computing in data centers. This fact clashes with the growing need to save and improve energy efficiency and requires a more optimized use of resources. The deployment of new services in edge and cloud computing, virtualization, and software-defined networks requires a better understanding of consumption patterns aimed at more efficient and sustainable models and a reduction in carbon footprints. These patterns are suitable to be exploited by machine, deep, and reinforced learning techniques in pursuit of energy consumption optimization, which can ideally improve the energy efficiency of data centers and big computing servers providing these kinds of services. For the application of these techniques, it is essential to investigate data collection processes to create initial information points. Datasets also need to be created to analyze how to diagnose systems and sort out new ways of optimization. This work describes a data collection methodology used to create datasets that collect consumption data from a real-world work environment dedicated to data centers, server farms, or similar architectures. Specifically, it covers the entire process of energy stimuli generation, data extraction, and data preprocessing. The evaluation and reproduction of this method is offered to the scientific community through an online repository created for this work, which hosts all the code available for its download.

Abstract: Objectives This study aimed to explore the prevalence of macrolide resistance and the underlying resistance mechanisms in Haemophilus influenzae (n = 2556) and Haemophilus parainfluenzae (n = 510) collected between 2018 and 2021 from Bellvitge University Hospital, Spain. Methods Antimicrobial susceptibility was tested by microdilution. Whole-genome sequencing was performed using Illumina MiSeq and Oxford Nanopore technologies, and sequences were examined for macrolide resistance determinants and mobile genetic structures. Results Macrolide resistance was detected in 67 H. influenzae (2.6%) and 52 (10.2%) H. parainfluenzae strains and associated with resistance to other antimicrobials (co-trimoxazole, chloramphenicol, tetracycline). Differences in macrolide resistance existed between the two species. Acquired resistance genes were more prevalent in H. parainfluenzae (35/52; 67.3%) than in H. influenzae (12/67; 17.9%). Gene mutations and amino acid substitutions were more common in H. influenzae (57/67; 85%) than in H. parainfluenzae (16/52; 30.8%). Substitutions in L22 and in 23S rRNA were only detected in H. influenzae (34.3% and 29.0%, respectively), while substitutions in L4 and AcrAB/AcrR were observed in both species. The MEGA element was identified in 35 (67.3%) H. parainfluenzae strains, five located in an integrative and conjugative element (ICE); by contrast, 11 (16.4%) H. influenzae strains contained the MEGA element (all in an ICE). A new ICE HpaHUB8 was described in H. parainfluenzae. Conclusions Macrolide resistance was higher in H. parainfluenzae than in H. influenzae , with differences in the underlying mechanisms. H. parainfluenzae exhibits co-resistance to other antimicrobials, often leading to an extensively drug-resistant phenotype. 78. A Review of the Electrical Conductivity Test Methods for Conductive Fabrics.

Abstract: With the substantial growth of the smart textiles market, electrical properties are becoming a basic requirement for most of the advanced textiles used in the development of wearable solutions and other textile-based smart applications. Depending on the textile substrate, the test method to determine the electrical properties can be different. Unlike smart fibers and yarns, the characterization of the electrical properties of fabrics cannot be tested between two connection points because the result would not represent the behavior of the entire fabric, so the electrical properties must be related to an area. The parameters used to characterize the electrical properties of the fabrics include resistance, resistivity, and conductivity. Although all of them can be used to indicate electrical performance, there are significant differences between them and different methods available for their determination, whose suitability will depend on the function and the textile substrate. This paper revises the main parameters used to characterize the electrical properties of conductive fabrics and summarizes the most common methods used to test them. It also discusses the suitability of each method according to several intervening factors, such as the type of conductive fabric (intrinsically or extrinsically conductive), its conductivity range, other fabric parameters, or the final intended application. For intrinsically conductive woven fabrics, all the methods are suitable, but depending on the requirements of conductivity accuracy, the contact resistance from the measuring system should be determined. For intrinsically conductive knitted fabrics, two-point probe, Van der Pauw, and eddy current methods are the most suitable. And for intrinsically conductive nonwoven fabrics, two-point probe and four-point probe methods are the most appropriate. 79. Insights into the recognition of hypermucoviscous Klebsiella pneumoniae clinical isolates by innate immune lectins of the Siglec and galectin families.

Abstract: Klebsiella pneumoniae is an opportunistic bacterium that frequently colonizes the nasopharynx and gastrointestinal tract and can also cause severe infections when invading other tissues, particularly in immunocompromised individuals. Moreover, K. pneumoniae variants exhibiting a hypermucoviscous (HMV) phenotype are usually associated with hypervirulent strains that can produce invasive infections even in immunocompetent individuals. Major carbohydrate structures displayed on the K. pneumoniae surface are the polysaccharide capsule and the lipopolysaccharide, which presents an O-polysaccharide chain in its outermost part. Various capsular and O-chain structures have been described. Of note, production of a thick capsule is frequently observed in HMV variants. Here we examined the surface sugar epitopes of a collection of HMV and non-HMV K. pneumoniae clinical isolates and their recognition by several Siglecs and galectins, two lectin families of the innate immune system, using bacteria microarrays as main tool. No significant differences among isolates in sialic acid content or recognition by Siglecs were observed. In contrast, analysis of the binding of model lectins with diverse carbohydrate-binding specificities revealed striking differences in the recognition by galactose- and mannose-specific lectins, which correlated with the binding or lack of binding of galectins and pointed to the O-chain as the plausible ligand. Fluorescence microscopy and microarray analyses of galectin-9 binding to entire cells and outer membranes of two representative HMV isolates supported the bacteria microarray results. In addition, Western blot analysis of the binding of galectin-9 to outer membranes unveiled protein bands recognized by this galectin, and fingerprint analysis of these bands identified several proteins containing potential O-glycosylation sites, thus broadening the spectrum of possible galectin ligands on the K. pneumoniae surface. Moreover, Siglecs and galectins apparently target different structures on K. pneumoniae surfaces, thereby behaving as nonredundant complementary tools of the innate immune system. Abstract: Tetracycline resistance in streptococci is mainly due to ribosomal protection mediated by the tet(M) gene that is usually located in the integrative and conjugative elements (ICEs) of the Tn916-family. In this study, we analyzed the genes involved in tetracycline resistance and the associated mobile genetic elements (MGEs) in Streptococcus dysgalactiae subsp. equisimilis (SDSE) causing invasive disease. SDSE resistant to tetracycline collected from 2012 to 2019 in a single hospital and from 2018 in three other hospitals were analyzed by whole genome sequencing. Out of a total of 84 SDSE isolates, 24 (28.5%) were resistant to tetracycline due to the presence of tet(M) (n = 22), tet(W) (n = 1), or tet(L) plus tet(W) (n = 1). The tet(M) genes were found in the ICEs of the Tn916-family (n = 10) and in a new integrative and mobilizable element (IME; n = 12). Phylogenetic analysis showed a higher genetic diversity among the strains carrying Tn916 than those having the new IME, which were closely related, and all belonged to CC15. In conclusion, tetracycline resistance in SDSE is mostly due to the tet(M) gene associated with ICEs belonging to the Tn916-family and a new IME. This new IME is a major cause of tetracycline resistance in invasive Streptococcus dysgalactiae subsp. equisimilis in our settings.

Abstract: The extremely flammable cellulose macromolecules in cotton fabrics seriously affect their application in the field of flame retardancy. In this paper, the surface of cotton fabric was modified with citric acid, and then metal-organic framework (UiO-66), cationic starch (St) and polyphosphonitrile (PZS) microspheres containing the transition metal element Zr were sequentially deposited to prepare an efficient and economical cotton fabric. St-PZS double-impregnated cotton fabrics have better thermal stability, flame retardancy and mechanical properties. And the coating does not affect the whiteness and stiffness of the cotton fabric. The results of thermogravimetric analysis showed that cotton/UiO-66/2St-PZS had a higher residual char rate (25.34%) under N2 atmosphere, which was 200.1% higher than that of control cotton (8.28%). In addition, cotton/UiO-66/2St-PZS had a 46.6% reduction in peak heat release, a 31.7% reduction in total heat release, and a 35.3% reduction in the peak of CO2 production compared to control cotton. It is worth noting that the average gross calorific value is 63.3% lower than that of the control cotton, indicating that flame retardant cotton fabrics can effectively inhibit the production of flammable gases. In addition, the tensile strength and elongation at break of cotton/UiO-66/2St-PZS are 458.79 N and 11.04%, respectively. Abstract: The use of lime-based composites reinforced with fabrics made of natural fibers, is a promising solution for the strengthening of historical masonry structures owing to its moderate mechanical strength, similar color and chemical compatibility, avoiding damage on the historical structure and contributing to crack-width control. In this study, a new lime-based composite reinforced with flax nonwoven fabrics is mechanically characterized to study its reinforcement potential in terms of strength, and stress distribution. To this end, laminate plates with lime/metakaolin matrix and four to six layers of fabric reinforcement were produced and totally carbonated in a CO2 chamber. Both the mechanical (flexural and tensile) and durability (against wet-dry cycles) properties of the composite were subsequently assessed. Furthermore, Digital Image Correlation (DIC) was carried out on tensile testing to study the stress distribution. The 6-layer composites displayed the best performance (with flexural and tensile strengths of approximately 5.3 MPa and 2.5 MPa, respectively), followed by the 5-layer and the 4-layer composites. The DIC analysis revealed a higher stress distribution in the 6-layer composites, with an increased number of cracks, although having a lower severity. As for durability, a decrease of 30–45% is observed in flexural strength, and of 6–18% in tensile strength, depending on the number of reinforcing layers. SEM analysis refers to fiber/matrix debonding as the cause of this decrease. No damage has been observed on the fiber surface, which retains its reinforcing capacity. All of the composites displayed strain-hardening behavior in both the unaged and aged conditions. Abstract: This project consists on the development of an automatic audiovisual advertisement generator, based on state-of-the-art deep learning models. The project was developed as a prototype during an internship period, with the intention of turning it into an API in the future. It combines a depth map and segmentation to achieve a parallax effect in generated images, and includes a text-to-speech model that achieves dynamic and realistic results. The project works as an effective tool for video generation, while text-to-video models improve until they reach commercial quality within a few years., Este proyecto consiste en el desarrollo de un generador automático audiovisual de anuncios, basado en modelos de aprendizaje profundo de última generación. El proyecto fue desarrollado como prototipo durante un período de prácticas, con la intención de convertirlo en una API en un futuro. 98. HACKTEX MOOC on smart textiles tested on practical experience: a workshop about raw materials for the development of smart textiles

Abstract: Smart textiles have gathered an increasing interest in the industry in recent years. Nevertheless, due to its uneven development and its novelty, the presence of this topic in the educational curricula is scarce and mainly devoted to master's degrees. HACKTEX project, an Erasmus+ project funded by the European Union, aims to develop open-access resources to complement the knowledge of European engineering students’ curricula in the field of smart textiles through different pedagogical tools. In that sense, in the framework of the project, a virtual training based on a MOOC (Massive Online Open Course) and a Virtual Guide on Entrepreneurship has been developed. In order to test the produced virtual materials, a pilot bootcamp training was carried out, hosted at the facilities of the University of Borås (Sweden). The bootcamp involved students from all the universities participating in the project: the hosting university (UB), TUIASI (Romania), UNIWA (Greece) and UPC (Spain). Previously to its start, the students accessed the online virtual training and studied the materials available. Then, during the bootcamp, the project partners designed as trainers tested the knowledge from the virtual training acquired by the students with different learning methodologies. The TECTEX Research group from the Universitat Politècnica de Catalunya (UPC), was responsible for the first day of the bootcamp training, where a workshop about raw materials and products for functional and smart textiles (corresponding to the learning unit 2 of the MOOC: Raw materials and products for functional and smart textiles) was performed. This workshop started assessing the knowledge of the students about the raw materials presented on the MOOC with an interactive quiz. Then, the students were mixed and grouped to interact with four workstations, each one focussed on one of the raw materials discussed on the MOOC.
