Workplace gossip, a ubiquitous organizational behavior broadly defined as talking about an absent target in an evaluative way, has received increasing scientific attention over the past few years. The complexity and dynamism of the workplace gossip phenomenon create challenging research conditions such that studies focusing on the allegedly same type of workplace gossip can produce differential findings. To address this problem and better align theory and methodology, our manuscript first proposes a framework of workplace gossip that accounts for the interdependencies of the context-embeddedness and dynamism of workplace gossip. Guided by this framework, we systematically evaluate extant workplace gossip research, spanning a total of N = 104 empirical research articles. Highlighting current methodological challenges that indicate a neglect of the dynamic nuances and contexts of workplace gossip, we argue that previous organizational research painted an overly simplistic picture of workplace gossip. By looking beyond traditional organizational research designs, we derive recommendations to advance research on workplace gossip and, ultimately, to establish it as a complex and dynamic social interaction behavior.

77. The association between antibodies to neurotropic pathogens and bipolar disorder : A study in the Dutch Bipolar (DB) Cohort and meta-analysis.

Exposure to neurotropic pathogens has been hypothesized to be a risk factor for the development of bipolar disorder (BD). However, evidence so far is inconsistent. We, therefore, analyzed the seroprevalence and titer levels of IgG antibodies against several herpesviruses and Toxoplasma gondii (T. gondii) in plasma of 760 patients with a bipolar disorder, 144 first-degree matched relatives and 132 controls of the Dutch Bipolar (DB) Cohort using ELISA. In addition, we performed a literature-based meta-analysis on the seroprevalence of IgG antibodies against these pathogens (n = 14). Our results in the DB Cohort and subsequent meta-analysis (n = 2364 BD patients, n = 5101 controls) show no association between exposure to herpesviruses and bipolar disorder (HSV-1 [adjusted OR 0.842, 95% CI 0.567-1.230], HSV-2 [adjusted OR 0.877, 95% CI 0.437-1.761], CMV [adjusted OR 0.884 95% CI 0.603-1.295], EBV [adjusted OR 0.968 95% CI 0.658-1.423]). In the DB Cohort, we did not find an association between bipolar disorder and T. gondii titer or seroprevalence either [adjusted OR 1.018, 95% CI 0.672-1.542]. The overall OR was not significant for T. gondii [OR: 1.4, 95% CI 0.95-1.90, p = 0.09), but subgroup analyses in age groups below 40 years showed a significantly increased seroprevalence of T. gondii IgGs in BD [OR: 1.8 (95% CI 1.10-2.89, p = 0.021]. Our meta-analysis indicates that T. gondii exposure may be a risk factor for BD in certain subpopulations.

Genome-wide association studies (GWASs) are highly effective at identifying common risk variants for schizophrenia. Rare risk variants are also important contributors to schizophrenia etiology but, with the exception of large copy number variants, are difficult to detect with GWAS. Exome and genome sequencing, which have accelerated the study of rare variants, are expensive so alternative methods are needed to aid detection of rare variants. Here we re-analyze an Irish schizophrenia GWAS dataset (n = 3,473) by performing identity-by-descent (IBD) mapping followed by exome sequencing of individuals identified as sharing risk haplotypes to search for rare risk variants in coding regions. We identified 45 rare haplotypes (>1 cM) that were significantly more common in cases than controls. By exome sequencing 105 haplotype carriers, we investigated these haplotypes for functional coding variants that could be tested for association in independent GWAS samples. We identified one rare missense variant in PCNT but did not find statistical support for an association with schizophrenia in a replication analysis. However, IBD mapping can prioritize both individual samples and genomic regions for follow-up analysis but genome rather than exome sequencing may be more effective at detecting risk variants on rare haplotypes.

Clinical outcomes after a first-episode of psychosis (FEP) are heterogeneous. Many patient-related factors such as gender and comorbidity have been studied to predict symptomatic outcomes. However, psychiatrist-related factors such as prescription behaviour and gender have received little attention. We assessed the relationship between patients' psychiatrists, psychosis severity and daily functioning in 201 patients remitted from an FEP for a duration of one year, treated by 18 different psychiatrists. We controlled for baseline severity, dose and type of antipsychotic medication, frequency of visits, and patients' education. Symptom severity, daily functioning, and antipsychotic drug use were assessed at baseline and at 3, 6, and, 12 months follow-up. We found that psychiatrists accounted for 9.1% of the explained variance in patients' symptom severity and 10.1% of the explained variance in daily functioning.These effects persisted even when controlling for factors such as baseline severity and the prescribed dose. The effect of prescribed dose on symptom severity and daily functioning differed between psychiatrists. Treatment centre, session frequency, and medication nonadherence were not related to symptom severity. Our results emphasize the importance of individual psychiatrist factors in symptomatic outcomes after an FEP. Further identification of psychiatrist-related factors such as the quality of therapeutic alliances and shared decision-making, may optimize psychiatrists' training with the goal of improving patient outcomes. Ocular complications of antineoplastic agents can have a profound effect on the quality of life of cancer patients. New oncologic treatments like monoclonal antibodies, immunotherapies, antibody-drug conjugates, checkpoint inhibitors and growth factor receptors have resulted in increased ocular complications. These ocular complications differs in respect to distinct mechanisms of actions and lead to significant challenges in the management of cancer patients. In this review, we reviewed literature, clinical studies and cases detailing ocular complications due to administration of antineoplastic agents and emphasized the need for communication between oncologists and ophthalmologists toward early detection and management of ocular complications.

82. Antibiotic policies in the UK dairy industry : a voluntary industry-led approach in action

Today a major topic of concern is the use of antibiotics in food animals and its link with the development of antimicrobial resistance (AMR) in humans. Public health agencies across the globe frame the ‘misuse and overuse’ of antibiotics in agriculture as major human driver to the development of AMR in animals, humans and the environment. In 2016, the United Kingdom (UK) has made its livestock sectors and food supply chains responsible to reduce and achieve responsible antibiotic use. Since, antibiotic surveillance systems are being implemented across livestock sectors to measure and monitor antibiotic practices. This is complemented with educational interventions and guidelines to ‘rationalise’ antibiotic use by farmers and veterinarians. According to the latest UK veterinary antibiotic surveillance and sales report, the sales of veterinary antibiotics for use in food-producing animals have dropped with 40% between 2013 in 2017. It is believed by governmental and agricultural policymakers that the antibiotic policies undertaken by the livestock sectors is taking force. This PhD thesis has reconsidered the progress claimed by the UK veterinary antibiotic surveillance and sales reports. Moving beyond statistical realities, it examined how the UK’s industry-led approach is taking shape in practice. A multi-sited ethnographic methodological framework has been used to examine at first how the UK has consolidated around an industry-led approach, in contrast to some other European countries who used legislation to tackle persistent antibiotic practices. Second, taking the UK dairy industry as case study, interview and observational methods were used to understand how dairy policies are formulated by the dairy sector and dairy supply chains and how the policies are practised by farmers and veterinarians. Findings reveal that the policies in the dairy industry only partially address the complex network of people, animals and the environment in which dairy antibiotics circulate. Although milk processors and retailers in the UK have taken up the lead to produce dairy antibiotic policies, the policies seem to benefit market purposes rather than addressing structural issues in UK dairy production systems. Some of the antibiotic policies produce new travel routes of antibiotics between systems resulting in new public health risks. Other antibiotic policies fail to assess how veterinarians and farmers are constrained in their antibiotic choices by their agricultural actor-networks. As a result, the UK’s industry-led approach maintains and reproduces irresponsible antibiotic practices in the UK dairy industry. This study reveals how antibiotic ‘misuse and overuse’ in agriculture is far from a behavioural matter, with solely farmers and veterinarians to blame. Instead, antibiotic use in food animals is embedded in complex economic networks that constrain radical changes in dairy husbandry management and antibiotic use on farms. To achieve responsible farming and improve antibiotic practices, the UK government should take responsibility and work more closely with the UK livestock sectors to understand what regulatory and financial support is needed.

One largely neglected focus in the analysis of mission-oriented innovation policies is mission cocreation between stakeholder groups advocating different solution directions. In this paper, we introduce the innovation management concept of ambidexterity to study how mission cocreation in different mission arenas is influenced by actors aiming to continue existing innovation pathways (exploitative innovation) and actors advocating alternative pathways (explorative innovation). Our case study on the Dutch circular agriculture mission highlights how an initial top-down strategy development was dominated by exploitative forces, despite ambitions to secure broad stakeholder participation. Government-led efforts to still engage exploration-minded actors, and restore ambidexterity, were hampered by potential contributors feeling excluded from the policy process. These findings underline the risk that openness of missions, required for broad stakeholder involvement, might also reinforce pre-existing contestation and innovation pathways. Overall, our findings demonstrate a trade-off between maximizing efficiency and speed versus obtaining novel perspectives and wide societal legitimacy. [ABSTRACT FROM AUTHOR]

Abstract Although epidemiological studies report that hallucinations occur in 6–15% of the general population, little is known about their phenomenology. To overcome this paucity, this study investigates the phenomenological characteristics of hallucinations in the general population, by using a nationally promoted online survey to assess hallucination phenomenology in four sensory modalities, through a self-report version of the Questionnaire for Psychotic Experiences (QPE), in 10,448 participants (aged 14–88 years). The phenomenology of hallucinations was assessed if hallucinations reportedly occurred in the past month. In the past month, auditory hallucinations were reported most frequently (29.5%), followed by visual (21.5%), tactile (19.9%), and olfactory hallucinations (17.3%); hallucinations in two or more modalities were reported by 47.6%. Substantial numbers of participants rated their hallucinations as severe, due to negative content (16.0–31.6%), previous bothersome experiences (14.8–20.2%), ensuing distress (10.5–16.8%), and/or ensuing disfunctioning (12.7–17.3%). Decreased insight was found in 10.2–11.4%. Hypnagogia was reported by 9.0–10.6%, and bereavement hallucinations by 2.8%. Spondyloepimetaphyseal dysplasia (SEMD) is characterized by vertebral, epiphyseal, and metaphyseal alterations. Patients become predominantly apparent with disproportionate short stature. The genetic background of SEMD is heterogeneous, with different modes of inheritance (autosomal dominant, autosomal recessive, and X-linked disorders). Amongst the genes in which variants are known to cause SEMD, UFM1-specific protease 2 (UFSP2) encodes a cysteine protease involved in the maturation of Ubiquitin-fold modifier 1 (UFM1). Heterozygous pathogenic variants affecting the C-terminal catalytic domain of UFSP2 are related to two entities of skeletal dysplasia, Beukes hip dysplasia (BHD) and SEMD type Di Rocco (SEMDDR). This is the first report of a de novo heterozygous variant affecting the catalytic Cys302 residue of UFSP2 (NM_018359.3:c.905G>C, p.(Cys302Ser)) causing SEMDDR. According to previously described patients with SEMDDR, our patient presented with disproportionate short stature, genu varum, gait instability, and radiologically detected epiphyseal and metaphyseal alterations. Additionally, a bell-shaped thorax, lumbar hyperlordosis, muscular hypotonia, and coxa vara were observed in the patient described in this study. Our findings underline the fundamental importance of an intact catalytic triad of the human UFSP2 for normal skeletal development and extend the phenotypical features of patients with UFSP2-related skeletal dysplasia.

The article argues that the imperial expansion of the Late Roman Republic is reflected in Cicero’s philosophical texts by example of De divinatione. The expansion of Rome made it necessary to consider what it meant to be Roman and what Roman practices are, especially confronted with other, alien practices that might seem similar. Cicero offers his texts as an admonition to consider religious practices – here: divination – the Romans might encounter in the provinces and how to deal with them, to consider their usefulness while also bearing in mind the latent danger in not doing them right or exceeding the religious need which upholds the pax deorum. Being put in the context of the expanding empire, the article makes sense of the multiple non-Roman examples cited especially in Book 1 of the treatise De divinatione.

Abstract Background As the technology of next generation sequencing rapidly develops and costs are constantly reduced, the clinical availability of whole genome sequencing (WGS) increases. Thereby, it remains unclear what exact advantage WGS offers in comparison to whole exome sequencing (WES) for the diagnosis of genetic diseases using current technologies. Methods Trio‐WGS was conducted for 20 patients with developmental or epileptic encephalopathies who remained undiagnosed after WES and chromosomal microarray analysis. Results A diagnosis was reached for four patients (20%). However, retrospectively all pathogenic variants could have been detected in a WES analysis conducted with today's methods and knowledge. Conclusion The additional diagnostic yield of WGS versus WES is currently largely explained by new scientific insights and the general technological progress. Nevertheless, it is noteworthy that whole genome sequencing has greater potential for the analysis of small copy number and copy number neutral variants not seen with WES as well as variants in noncoding regions, especially as potentially more knowledge of the function of noncoding regions arises. We, therefore, conclude that even though today the added value of WGS versus WES seems to be limited, it may increase substantially in the future.

Telomere biology disorders (TBD) result from premature telomere shortening due to pathogenic germline variants in telomere maintenance-associated genes. In adults, TBD are characterized by mono/oligosymptomatic clinical manifestations (cryptic TBD) contributing to severe underdiagnosis. We present a prospective multi-institutional cohort study where telomere length (TL) screening was performed in either newly diagnosed patients with aplastic anemia (AA) or if TBD was clinically suspected by the treating physician. TL of 262 samples was measured via flow-fluorescence in situ hybridization (FISH). TL was considered suspicious once below the 10th percentile of normal individuals (standard screening) or if below 6.5 kb in patients >40 years (extended screening). In cases with shortened TL, next generation sequencing (NGS) for TBD-associated genes was performed. The patients referred fell into 6 different screening categories: (1) AA/paroxysmal nocturnal hemoglobinuria, (2) unexplained cytopenia, (3) dyskeratosis congenita, (4) myelodysplastic syndrome/acute myeloid leukemia, (5) interstitial lung disease, and (6) others. Overall, TL was found to be shortened in 120 patients (n = 86 standard and n = 34 extended screening). In 17 of the 76 (22.4%) standard patients with sufficient material for NGS, a pathogenic/likely pathogenic TBD-associated gene variant was identified. Variants of uncertain significance were detected in 17 of 76 (22.4%) standard and 6 of 29 (20.7%) extended screened patients. Expectedly, mutations were mainly found in TERT and TERC. In conclusion, TL measured by flow-FISH represents a powerful functional in vivo screening for an underlying TBD and should be performed in every newly diagnosed patient with AA as well as other patients with clinical suspicion for an underlying TBD in both children and adults.

Schizophrenia and bipolar disorder are two distinct diagnoses that share symptomology. Understanding the genetic factors contributing to the shared and disorder-specific symptoms will be crucial for improving diagnosis and treatment. In genetic data consisting of 53,555 cases (20,129 bipolar disorder [BD], 33,426 schizophrenia [SCZ]) and 54,065 controls, we identified 114 genome-wide significant loci implicating synaptic and neuronal pathways shared between disorders. Comparing SCZ to BD (23,585 SCZ, 15,270 BD) identified four genomic regions including one with disorder-independent causal variants and potassium ion response genes as contributing to differences in biology between the disorders. Polygenic risk score (PRS) analyses identified several significant correlations within case-only phenotypes including SCZ PRS with psychotic features and age of onset in BD. For the first time, we discover specific loci that distinguish between BD and SCZ and identify polygenic components underlying multiple symptom dimensions. These results point to the utility of genetics to inform symptomology and potential treatment.

Anthropogenic litter is omnipresent in terrestrial and freshwater systems, and can have major economic and ecological impacts. Monitoring and modeling of anthropogenic litter comes with large uncertainties due to the wide variety of litter characteristics, including size, mass, and item type. It is unclear as to what the effect of sample set size is on the reliability and representativeness of litter item statistics. Reliable item statistics are needed to (1) improve monitoring strategies, (2) parameterize litter in transport models, and (3) convert litter counts to mass for stock and flux calculations. In this paper, we quantify sample set size requirement for riverbank litter characterization, using a database of more than 14,000 macrolitter items (>0.5 cm), sampled for 1 year at eight riverbank locations along the Dutch Rhine, IJssel, and Meuse rivers. We use this database to perform a Monte Carlo based bootstrap analysis on the item statistics, to determine the relation between sample size and variability in the mean and median values. Based on this, we present sample set size requirements, corresponding to selected uncertainty and confidence levels. Optima between sampling effort and information gain is suggested (depending on the acceptable uncertainty level), which is a function of litter type heterogeneity. We found that the heterogeneity of the characteristics of litter items varies between different litter categories, and demonstrate that the minimum required sample set size depends on the heterogeneity of the litter category. This implies that more items of heterogeneous litter categories need to be sampled than of heterogeneous item categories to reach the same uncertainty level in item statistics. For example, to describe the mean mass the heterogeneous category soft fragments (>2.5 cm) with 90% confidence, 990 items were needed, while only 39 items were needed for the uniform category metal bottle caps. Finally, we use the heterogeneity within litter categories to assess the sample size requirements for each river system. All data collected for this study are freely available, and may form the basis of an open access global database which can be used by scientists, practitioners, and policymakers to improve future monitoring strategies and modeling efforts.

A detailed understanding of the immune response to SARS-CoV-2 is critical to unravel the causative factors underlying differences in disease severity and to develop optimal vaccines against new SARS-CoV-2 variants.MethodsWe combined single-cell RNA and T cell receptor sequencing with CITE-seq antibodies to characterize the CD8+ T cell response to SARS-CoV-2 infection at high resolution and compared responses between mild and severe COVID-19.ResultsWe observed increased CD8+ T cell exhaustion in severe SARS-CoV-2 infection and identified a population of NK-like, terminally differentiated CD8+ effector T cells characterized by expression of FCGR3A (encoding CD16). Further characterization of NK-like CD8+ T cells revealed heterogeneity among CD16+ NK-like CD8+ T cells and profound differences in cytotoxicity, exhaustion, and NK-like differentiation between mild and severe disease conditions.DiscussionWe propose a model in which differences in the surrounding inflammatory milieu lead to crucial differences in NK-like differentiation of CD8+ effector T cells, ultimately resulting in the appearance of NK-like CD8+ T cell populations of different functionality and pathogenicity. Our in-depth characterization of the CD8+ T cell-mediated response to SARS-CoV-2 infection provides a basis for further investigation of the importance of NK-like CD8+ T cells in COVID-19 severity.
