Background: Ischemia reperfusion injury (IRI) plays a major role in solid organ transplantation. The length of warm ischemia time is critical for the extent of tissue damage in renal IRI. In this experimental study we hypothesized that local release of labile heme in renal tissue is triggered by the duration of warm ischemia (15 vs. 45 min IRI) and mediates complement activation, cytokine release, and inflammation.Methods: To induce IRI, renal pedicle clamping was performed in male C57BL/6 mice for short (15 min) or prolonged (45 min) time periods. Two and 24 h after experimental ischemia tissue injury labile heme levels in the kidney were determined with an apo-horseradish peroxidase assay. Moreover, renal injury, cytokines, and C5a and C3a receptor (C5aR, C3aR) expression were determined by histology, immunohistochemistry and qPCR, respectively. In addition, in vitro studies stimulating bone marrow-derived macrophages with LPS and the combination of LPS and heme were performed and cytokine expression was measured.Results: Inflammation and local tissue injury correlated with the duration of warm ischemia time. Labile heme concentrations in renal tissue were significantly higher after prolonged (45 min) as compared to short (15 min) IRI. Notably, expression of the inducible heme-degrading enzyme heme oxygenase-1 (HO-1) was up-regulated in kidneys after prolonged, but not after short IRI. C5aR, the pro-inflammatory cytokines IL-6 and TNF-α as well as pERK were up-regulated after prolonged, but not after short ischemia times. Consecutively, neutrophil infiltration and up-regulation of pro-fibrotic cytokines such as CTGF and PAI were more pronounced in prolonged IRI in comparison to short IRI. In vitro stimulation of macrophages with LPS revealed that IL-6 expression was enhanced in the presence of heme. Finally, administration of the heme scavenger human serum albumin (HSA) reduced the expression of pro-inflammatory cytokines, C3a receptor and improved tubular function indicated by enhanced alpha 1 microglobulin (A1M) absorption after IRI.Conclusions: Our data show that prolonged duration of warm ischemia time increased labile heme levels in the kidney, which correlates with IRI-dependent inflammation and up-regulation of anaphylatoxin receptor expression. more...

The bioeconomy can be integral to transforming the current economic system into one with reduced environmental and social impacts of material consumption. This work describes a bio-based multi-layer panel that is based on residual coniferous bark. To ensure that the presented bio-based panel positively contributes to environmental protection while remaining competitive with conventional products and meeting high social standards, the development of the panel is accompanied by a life cycle sustainability assessment. This study performs a comparative LCA and LCC of the developed panel to conventional benchmark panels, as well as a qualitative social life cycle assessment. While the panel performs only economically marginally weaker than the benchmarks, the results are more heterogeneous for the environmental dimension with benefits of the bio-based panel in categories such as climate change, acidification, and ozone formation and detriments in categories including eutrophication. The S-LCA analysis shows that all of the involved companies apply social principles in direct proximity; however, social responsibility along the supply chain could be further promoted. All results need to be viewed with the caveat that the manufacturing processes for the new panel have been implemented, to date, on a pilot scale and further improvements need to be achieved in terms of upscaling and optimisation cycles. more...

Supply risk assessments are an integral part of raw material criticality assessments frequently used at the country or company level to identify raw materials of concern. However, the indicators used in supply risk assessments to estimate the likelihood of supply disruptions vary substantially. Here, we summarize and evaluate the use of supply risk indicators and their normalization to supply risk scores in 88 methods published until 2020. In total, we find 618 individual applications of supply risk criteria with 98 unique criteria belonging to one of ten indicator categories. The most often used categories of supply risk indicators are concentration, scarcity, and political instability. The most frequently used criteria are the country concentration of production, depletion time of reserves, and geopolitical risk. Indicator measurements and normalizations vary substantially between different methods for the same criterion. Our results can be used for future raw material criticality assessments to screen for suitable supply risk indicators and generally accepted indicator normalizations. Nickel-based superalloys contain various elements which are added in order to make the alloys more resistant to thermal and mechanical stress and to the adverse operating environments in jet engines. In particular, higher combustion temperatures in the gas turbine are important, since they result in higher fuel efficiency and thus in lower CO2 emissions. In this paper, a semi-quantitative assessment scheme is used to evaluate the relative supply risks associated with elements contained in various Ni-based superalloys: aluminium, titanium, chromium, iron, cobalt, niobium, molybdenum, ruthenium, tantalum, tungsten, and rhenium. Twelve indicators on the elemental level and four aggregation methods are applied in order to obtain the supply risk at the alloy level. The supply risks for the elements rhenium, molybdenum and cobalt are found to be the highest. For three of the aggregation schemes, the spread in supply risk values for the different alloy types (as characterized by chemical composition and the endurance temperature) is generally narrow. The fourth, namely the cost-share’ aggregation scheme, gives rise to a broader distribution of supply risk values. This is mainly due to the introduction of rhenium as a component starting with second-generation single crystal alloys. The resulting higher supply risk appears, however, to be acceptable for jet engine applications due to the higher temperatures these alloys can endure. more...

In this study we present the results of an emission study of different phytoplankton samples in aqueous media treated with elevated ozone levels. Halocarbon measurements show that the samples tested released bromoform and different iodocarbons, including iodomethane, iodochloromethane and diiodomethane. Iodide and iodate levels in the liquid phase were representative of concentrations of surface water in a natural environment. Measurement of volatile iodine (I2) emissions from two diatom samples (Mediopyxis helysia and Porosira glacialis) and the background sample (F/2 medium from filtered natural seawater) showed that the quantity of evolved I2 depends on the ozone concentration in the air. This behaviour was assumed to be caused by the oxidation reaction mechanism of iodide with ozone. The I2 emission flux agrees with model calculations at different iodide concentrations. The I2 emission of a natural plankton concentrate sample was, however, very low compared to other samples and showed no dependence on ozone. The reason for this was shown to be the low iodide concentration in the algal suspension, which seems to be the limiting factor in the oxidative formation of I2. more...

Based on the results of a pilot study in 2007, which found high mixing ratios of molecular iodine (I2) above the intertidal macroalgae (seaweed) beds at Mweenish Bay (Ireland), we extended the study to nine different locations in the vicinity of Mace Head Atmospheric Research Station on the west coast of Ireland during a field campaign in 2009. The mean values of chem{I_2} mixing ratio found above the macroalgae beds at nine different locations ranged from 104 to 393 ppt, implying a high source strength of I2. Such mixing ratios are sufficient to result in photochemically driven coastal new-particle formation events. Mixing ratios above the Ascophyllum nodosum and Fucus vesiculosus beds increased with exposure time: after 6 h exposure to ambient air the mixing ratios were one order of magnitude higher than those initially present. This contrasts with the emission characteristics of Laminaria digitata, where most I2 was emitted within the first half hour of exposure. Discrete in situ measurements (offline) of I2 emission from ambient air-exposed chamber experiments of L. digitata, A. nodosum and F. vesiculosus substantially supported the field observations. Further online and time-resolved measurements of the I2 emission from O3-exposed macroalgal experiments in a chamber confirmed the distinct I2 emission characteristics of A. nodosum and F. vesiculosus compared to those of L. digitata. The emission rates of A. nodosum and F. vesiculosus were comparable to or even higher than L. digitata after the initial exposure period of ~20–30 min. We suggest that A. nodosum and F. vesiculosus may provide an important source of photolabile iodine in the coastal boundary layer and that their impact on photochemistry and coastal new-particle formation should be reevaluated in light of their longer exposure at low tide and their widespread distribution. more...
