Objective To construct lipid nanoparticles DLin-LNP for mRNA delivery. Methods DLin-LNP was prepared by thin film hydration method, and DLin-LNP/mRNA was further constructed by using EGFP-mRNA as model drug. The particle size, zeta potential, and appearance morphology were measured. Furthermore, the intracellular distribution and transfection of DLin-LNP/mRNA in RM-1 cells was investigated by laser scanning confocal microscope. Results DLin-LNP was successfully prepared. The average particle size was about (151.1±2.1) nm, the no-load potential was (23.7±0.5) mV. The cytotoxicity of DLinLNP was far lower than that of the commercially available liposomal Lipo8000. The results of transfection experiment indicated that DLin-LNP has high transfection efficiency for mRNA delivery with low cytotoxicity and good stability. Objective To delineate RNA binding profile of NAT10 (N-acetyltransferase 10) in human chronic myelogenous leukemia cell line K562. Methods Enhanced UV crosslinking and immunoprecipitation (eCLIP) were used to capture the transcript collection bound by NAT10 in K562 cells. Data were obtained through high throughput sequencing and then analyzed with bioinformatics. The type and regions of NAT10 binding genes were identified via peak-calling and annotation. The function of the binding RNA was further explored by gene function enrichment analysis. Results NAT10 binding transcripts which were enriched in 3′UTR region, were mainly protein coding genes. Further analysis showed that they were functionally associated with DNA damage and repair. Conclusions NAT10 may regulate gene expression by binding to the 3′UTR region of mRNA associated with DNA damage repair. [ABSTRACT FROM AUTHOR]

Calcium phosphate cements (CPCs) with macropores and micropores for high bone regeneration ability are desired. CPCs with macropores and micropores which were prepared by combining porous granules have been reported. In this study, the cement system composed of β-tricalcium phosphate (β-TCP) granules and phosphoric acid solution was selected, and the effects of the characteristics of granules on the microstructures and properties of the cements were examined. The spherical porous β-TCP granules with high porosity and irregular porous β-TCP granules with low porosity were prepared, and they were mixed with a phosphoric acid solution. When the spherical porous granules with high porosity were used, the granules were packed uniformly in the cement, and the size distribution of the macropores formed among the granules was narrower than that of the cement prepared using the irregular porous β-TCP granules with low porosity. The cement with high porosity can be obtained from the spherical porous granules with high porosity. We investigated Kansei hierarchical structures of protective films for smartphones to clarify user's evaluation criteria. The hierarchical structure was hypothesized to be divided into three layers: physical element, impression, and value. For physical elements, optical characteristics were measured. For impression, evaluation words for protective films were collected by the evaluation grid experiment, and selected words were used in the affective evaluation experiment. Then, factor analyses were performed to extract factors. For value, three words were adopted: "Good", "Like", and "Premium". Hierarchical structures were modeled using the structural equation modeling to clearly relationships among three layers. The results showed that Tt and Haze influenced the factor of natural, and the factor influenced values. Finally, we also investigated individual difference in hierarchical structures, the results showed that participant types were divided into "Clear" and "Clear and Matt", and the type of "Clear and Matt" had additional evaluation criteria compared type of "Clear". To achieve rapid on-site detection of aflatoxin B1 (AFB1) in agricultural and sideline products, a portable detection system based on differential pulse voltammetry (DPV) and STM32F103ZET6 as the core processor was designed. The system consists of two main parts: hardware detection devices and a mobile App, which are connected through Wi-Fi communication. The hardware detection equipment includes a DPV waveform generation circuit, constant potential circuit, and micro current detection module. The upper computer App was developed in an Android environment and completed tasks such as signal acquisition and data storage. After completing the design, experiments were conducted to verify the accuracy of the constant potential circuit and micro current detection module. The constant potential circuit accurately applied the voltage set by the program to the electrode, with a maximum error of 4 mV. The micro current detection module converts the current into a voltage signal according to the theoretical formula and amplifies it according to the theoretical amplification factor. The laboratory-made AFB1 sensor was used to effectively detect AFB1 in the range of 0.1 fg/ml to 100 pg/ml. The maximum relative error between the test results in the standard solution and the electrochemical workstation CHI760e was 7.37%. Furthermore, peanut oil samples with different concentrations of AFB1 were tested, and the results were compared to the CHI760e detection results as the standard, with a recovery rate of 96.8%~106.0%. Peanut samples with different degrees of mold were also tested and compared with CHI760e, with a maximum relative error of 7.10%. The system's portability allows it to be easily transported to different locations for on-site testing, making it an ideal solution for testing in remote or rural areas where laboratory facilities may be limited. Furthermore, the use of a mobile App for data acquisition and storage makes it easy to track and manage testing results. The fracture of WC-Co cemented carbides with different WC particle sizes was investigated experimentally, and then a discrete element method (DEM) simulation was performed considering such experiments. The bending strength of ultra-fine grained cemented carbide with a WC particle size of 0.4 μm was extremely high. The ultrafine grained cemented carbide sample after bending test had numerous fracture pieces and the short length of the specimens without fracture. Initial structures consisting of matrix and defect (d) particles were used in the DEM simulations. The DEM simulation results for a sample of 2% d particle, which was considered ultra-fine grained cemented carbide by comparison with experimental results, were as follows: The sample with the highest load at fracture, the most d particle bond-breaking, and the shortest length of remaining specimen without particle bond breaking was the 2% d sample.
