Abstract: The present study analyses the network that regulates and guards against oxidative imbalances in common dentex, poorly adapted to culture conditions, in comparison to the successfully cultured and closely related gilthead sea bream. Both sparid fish were grown from fingerlings to market size, and several molecular and metabolic biomarkers were analysed in fish sampled at the end of the trial. For such purpose, partial cDNA sequences coding for tumour necrosis factor-α (TNFα), interleukin-1β (IL-1β), growth hormone receptors (GHR-I and II), insulin-like growth factors (IGF-I and II), cytochrome P450 1A1 (CYP1A1), glucose regulated protein-75 (GRP75), metallothionein (MT), glutathione reductase (GR) and glutathione peroxidase (GPX-1) were first PCR amplified and sequenced in common dentex. The hepatic gene expression profile was then analysed by real-time PCR assays, using β-actin as housekeeping gene. Metabolic biomarkers included total plasma antioxidant capacity, ROS production, and circulating levels of GH, IGF-I, cortisol, glucose and blood lactate. Overall growth rates to attain market size were similar in both species. However, common dentex exhibited accelerated growth over the course of larval and initial juvenile stages that was not retained thereafter. They also exhibited higher levels of plasma peroxidases and secondary stress metabolites (glucose, lactate) in combination with an increased expression of pro-inflammatory cytokines (TNFα, IL-1β), which is in accordance with an inflammatory over activity in chronically stressed fish. The activity of oxidative CYP1A1 and GH/IGF pathways was down-regulated in common dentex, possibly to equilibrate this oxidative pressure. In the same way, hepatic transcripts of enzymatic (GPX-1) and non-enzymatic (GRP75, MT) antioxidant genes were up-regulated, remaining the total plasma antioxidant capacity within the reference values of gilthead sea bream. Thus, common dentex seems to orchestrate in the last juvenile stages several biological strategies, which may serve to preserve redox homeostasis at the expense of reduced growth and low detoxifying capacity for pollutants and endogenous substrates. Polyethylene microparticles are common contaminants in the marine environment, which are easily ingested by fish and can provoke physical damage or toxic reactions, resulting in inflammation, stress-related alterations and behavioural disorders. This work focuses on the extend of the alterations by microplastic ingestion on the intestinal health of Sparus aurata , in an integrative effort to relate the effect on gut microbiota and histopathological damage. Virgin, low density, polyethylene microplastic (LDPE-MP) particles were included into a control diet (D1, without MP), at a 0.12% in two different size ranges in diet D2 (200–500 μm) and D3 (501–1000 μm). Gilthead sea bream were fed D1, D2 or D3 diets for 30 days, and then sacrificed, biometrical data recorded, anterior intestine samples taken for histological analyses and anterior intestine mucus collected for Illumina sequencing of the V3-V4 region of the 16S rRNA, to evaluate the adherent bacterial communities. Intestines of fish fed D2 and D3 contained plastic aggregates and showed macroscopic signs of distension, liquid accumulation and inflammation. Their histopathological analysis revealed severe mechanical lesions (haemorrhages and epithelial desquamation and necrosis), inflammatory reaction (eosinophilia, lymphocytosis, and melanomacrophage presence) and severe epithelial vacuolization, caused by erosion and accumulation of MPs with sharp and rough-edges. Illumina sequencing resulted in over 7 million high-quality reads that were assigned to 589 OTUs. No significant differences were found in richness, diversity or composition at the phylum level among all three experimental groups. However, small but significant differences were found in differential expression analyses, with the D2 diet changing 6.2% and D3 changing 2.9% of the total bacterial composition in anterior intestine. The altered microbial composition seems to be directed as an attempt to counteract the mechanical damage and inflammation inflicted by MPs ingestion, in agreement with metagenome prediction and pathway analysis, which revealed that the slight effect of diets containing MPs (D2 and D3) on gut microbiota has the potential to change the metabolic capacity of these populations, including a decrease in pathways related to bacterial infection and inflammation. Thus, virgin LDPE-MPs ingestion in gilthead sea bream did not significantly affect growth and gut microbiota composition over a 30-day feeding period, but the slight alterations observed could derive in growth problems and dysbiosis upon prolonged microplastic ingestion, which are mainly driven by the severe mechanical damage inflicted to the digestive tract. • Microplastic ingestion produces intestinal damage in cultured gilthead sea bream. • Intestinal damage is caused by mechanical more than chemical effects. • Microplastic ingestion slightly changes intestinal microbiota in gilthead sea bream. • Prolonged microplastic ingestion could cause growth problems and dysbiosis. B Abstract: b Introduction: The Paisa project aims to use genetics to develop novel approaches for classifying severe mental illness (SMI) that have a stronger scientific foundation than the systems currently used in both research and clinical practice (see other abstracts in this symposium). For this project, electronic health records (EHR) are used to ascertain cases who have received inpatient treatment at a single psychiatric hospital that serves the entire 1 million inhabitants of the state of Caldas, Colombia. Using natural language processing of EHR text combined with neural network-based classification algorithms, we are able to classify substance use phenotypes with great accuracy (AUCROC Tabacco 0.93, Alcohol 0.90, Cannabis 0.95, Cocaine 0.96, others 0.91), in all cases improving upon linear naïve Bayes classifiers. Elongation of very long-chain fatty acid 4 (Elovl4) proteins participate in the biosynthesis of long-chain and very long-chain polyunsaturated fatty acids (LC-PUFA and VLC-PUFA). In the present study, an elovl4 cDNA was cloned from the swimming crab Portunus trituberculatus by PCR techniques and functionally characterized using recombinant expression in yeast Saccharomyces cerevisiae. The elovl4 cDNA sequence contained an open reading frame of 1038 base pairs, encoding a protein of 346 amino acids. The elovl4 has typical Elovl structures, with transmembrane domains (6) and a histidine box. The elovl4 was expressed in various tissues analyzed, with the highest expression found in intestine and hepatopancreas, followed by stomach and eyestalk. The functional characterization of Elovl4 yeast showed that the P. trituberculatus Elovl4 can elongate C 18 – 22 polyunsaturated fatty acids (PUFA), reaching in some cases products of C 24 and C 26. Along its ability to elongate PUFA, the P. trituberculatus Elovl4 was also efficient in the elongation of saturated fatty acids, with 28:0 and 30:0 being prominent elongation products. These results provide insight into the LC-PUFA biosynthetic capability of commercially important species of crustaceans. Changes in the ecosystems of vector-borne diseases can make the environment more or less favorable for mosquito vectors, animal reservoirs, and disease transmission. We report for the 1st time Anopheles marajoara in Argentina. In addition, An. deaneorum is reported for the 1st time in northwestern area of the country. Aedes fulvus, Culex eduardoi, Limatus durhamii, Mansonia pseudotitillans, Psorophora albigenu, Ps. cingulata, and Uranotaenia nataliae are recorded for the 1st time in Tucumán Province. Patients with bipolar disorder type I (BP-I) often present with impairments in cognitive function. Offspring unaffected by the disorder can also present with cognitive dysfunction. The objective of this study was to compare the cognitive function of BP-I patients, their unaffected offspring (UO) and healthy control subjects (HC)., Methods: Verbal memory, working memory index, processing speed, attention, verbal and phonological fluency and executive function were evaluated through the application of a neuropsychological battery to three groups made up of BP-I patients that attended the Bipolar Disorder Outpatient Clinic of Clínica San Juan de Dios de Manizales [San Juan de Dios de Manizales Clinic] (n=30), UO (n=32) and control group (n=31). The UO group and the control group were matched by gender, age and level of education., Results: Major differences between the three groups were found in the measures of cognitive functions (except in semantic fluency). The HC group showed better cognitive performance in all the functions. Post-hoc analysis showed similar results in the cognitive performance between BP-I and UO except in verbal learning and executive function tasks where the results were better in UO. A better performance in the control group was found, compared to the UO group, in executive function, attention, working memory, and semantic fluency and phonological areas., Conclusions: These results indicate that the offspring of patients with BP-I present with cognitive impairments without suffering from the disorder. The Annelida phylum is composed of a myriad of species exhibiting key phenotypic adaptations. They occupy key ecological niches in a variety of marine, freshwater and terrestrial ecosystems. Importantly, the increment of omic resources is rapidly modifying the taxonomic landscape and knowledge of species belonging to this phylum. Here, we comprehensively characterised and annotated a transcriptome of the common ragworm, Hediste diversicolor (OF Müller). This species belongs to the family Nereididae and inhabits estuarine and lagoon areas on the Atlantic coasts of Europe and North America. Ecologically, H. diversicolor plays an important role in benthic food webs. Given its commercial value, H. diversicolor is a promising candidate for aquaculture development and production in farming facilities, under a circular economy framework. We used Illumina next-generation sequencing technology, to produce a total of 105 million (M) paired-end (PE) raw reads and generate the first whole-body transcriptome assembly of H. diversicolor species. This high-quality transcriptome contains 69,335 transcripts with an N50 transcript length of 2313 bp and achieved a BUSCO gene completeness of 97.7% and 96% in Eukaryota and Metazoa lineage-specific profile libraries. Introduction: The safety of electroconvulsive therapy has improved greatly over the last decades, making the potentially adverse effects on memory and other neurocognitive functions the main clinical aspect of concern in the present. In Colombia, the general population and healthcare professionals (even some psychiatrists) seem to have mostly negative opinions towards electroconvulsive therapy treatment, but maybe this could be reconsidered if more information is provided; therefore, the aim of the present study was to evaluate the changes in memory and the severity of the symptoms in a group of patients with severe depression before and after electroconvulsive therapy., Methods: Twenty-three patients ranging in age from 23 to 70 years from the electroconvulsive therapy service at the San Juan de Dios Clinic (Manizales, Colombia) were recruited in order to assess the effect of electroconvulsive therapy on memory in patients with severe depression. Depressive symptoms and memory were assessed with the Hamilton Depression Scale (HAMD) and Rey Auditory Verbal Learning Test (RAVLT), respectively. The assessment was administered to participants before the initial treatment of electroconvulsive therapy series (0-1 day) and 2 days after their last treatment., Results: Electroconvulsive therapy resulted in significant improvement in the rating of depression. There were no significant differences in the five learning trials, delayed recall, learning and forgetting scores from pre-treatment to post-treatment. Significant pre-treatment/post-treatment differences were found in the delayed recognition trial., Conclusions: Pre- and post- electroconvulsive therapy cognitive assessment is a feasible and useful procedure. In general, memory performance does not worsen after electroconvulsive therapy in patients with depression. The plasma membrane and autoinhibited Ca 2+ -ATPases contribute to the Ca 2+ homeostasis in a wide variety of organisms. The enzymatic activity of these pumps is stimulated by calmodulin, which interacts with the target protein through the calmodulin-binding domain (CaMBD). Most information about this region is related to all calmodulin modulated proteins, which indicates general chemical properties and there is no established relation between Ca 2+ pump sequences and taxonomic classification. Thus, the aim of this study was to perform an in silico analysis of the CaMBD from several Ca 2+ -ATPases, in order to determine their diversity and to detect specific patterns and amino acid selection in different species. Patterns related to potential and confirmed CaMBD were detected using sequences retrieved from the literature. The occurrence of these patterns was determined across 120 sequences from 17 taxonomical classes, which were analyzed by a phylogenetic tree to establish phylogenetic groups. Predicted physicochemical characteristics including hydropathy and net charge were calculated for each group of sequences. 22 Ca 2+ -ATPases sequences from animals, unicellular eukaryotes, and plants were retrieved from bioinformatic databases. These sequences allow us to establish the Patterns 1(GQILWVRGLTRLQTQ), 3(KNPSLEALQRW), and 4(SRWRRLQAEHVKK), which are present at the beginning of putative CaMBD of metazoan, parasites, and land plants. A pattern 2 (IRVVNAFR) was consistently found at the end of most analyzed sequences. The amino acid preference in the CaMBDs changed depending on the phylogenetic groups, with predominance of several aliphatic and charged residues, to confer amphiphilic properties. The results here displayed show a conserved mechanism to contribute to the Ca 2+ homeostasis across evolution and may help to detect putative CaMBDs., (© 2022. The Vesicular Stomatitis Virus (VSV) is an arbovirus causing vesicular stomatitis (VS) in livestock. There are two serotypes recognized: New Jersey (VSNJV) and Indiana (VSIV). The virus can be transmitted directly by contact or by vectors. In 2018, Ecuador experienced an outbreak of Vesicular Stomatitis (VS) in cattle, caused by VSNJV and VSVIV, with 399 cases reported distributed over 18 provinces. We determined the phylogenetic relationships among 67 strains. For the construction of phylogenetic trees, the viral phosphoprotein gene was sequenced, and trees were constructed based on the Maximum Likelihood method using 2004 outbreak strains from Ecuador (GenBank) and the 2018 sequences (this article). We built a haplotype network for VSNJV to trace the origin of the 2004 and 2018 epizootics through topology and mutation connections. These analyses suggest two different origins, one related to the 2004 outbreak and the other from a transmission source in 2018. Our analysis also suggests different transmission patterns; several small and independent outbreaks, most probably transmitted by vectors in the Amazon, and another outbreak caused by the movement of livestock in the Andean and Coastal regions. We recommend further research into vectors and vertebrate reservoirs in Ecuador to clarify the mechanisms of the reemergence of the virus.

Trypanosomes are a group of pathogens distributed in the continents of Africa, America, Asia and Europe, and they affect all vertebrates including the neotropical primate group. Information about the trypanosome's diversity, phylogeny, ecology and pathology in non-human primates (NHPs) from the neotropical region is scarce. The objective of the study was to identify Trypanosoma and Babesia molecularly in NHPs under the phylogenetic species concept. We extracted DNA from a total of 76 faecal samples collected between 2019 and 2021, from a total of 11 non-human primate species of which 46 are from captive NHPs and 30 are free-living NHPs in the Western Amazon region of Ecuador. We did not detect DNA of Babesia sp. by polymerase chain reaction test in any of the faecal samples. However, the nested-PCR-based method revealed Trypanosoma parasites by ITS gene amplification in two faecal samples; one for the species Leontocebus lagonotus (from the captive population) and a second one for Cebus albifrons (from the free-ranging population). Maximum parsimony and likelihood methods with the Kimura2+G+I model inferred the evolutionary history of the two records, which showed an evolutionary relationship with the genus Trypanosoma . Two sequences are monophyletic with Trypanosoma. However, the number of sequences available in GenBank for their species identification is limited. The two samples present different molecular identifications and evolutionary origins in the tree topology. We are most likely referring to two different species, and two different localities of infection. We suggest that health management protocols should be implemented to prevent the transmission of blood-borne pathogens such as Trypanosoma sp. among captive populations. In addition, these protocols also protect the personnel of wildlife rehabilitation centers working in close proximity to NHPs and vice versa.

Non-tuberculous mycobacteria that cannot be identified at the species level represent a challenge for clinical laboratories, as proper species assignment is key to implementing successful treatments or epidemiological studies. We re-identified forty-eight isolates of Ziehl-Neelsen (ZN)-staining-positive "acid-fast bacilli" (AFB), which were isolated in a clinical laboratory and previously identified as Mycobacterium species but were unidentifiable at the species level with the hsp65 PCR restriction fragment length polymorphism analysis (PRA). As most isolates also could not be identified confidently via 16S , hsp65 , or rpoB DNA sequencing and a nBLAST search analysis, we employed a phylogenetic method for their identification using the sequences of the 16S rDNA, which resulted in the identification of most AFB and a Mycobacterium species diversity not found before in our laboratory. Most were rare species with only a few clinical reports. Moreover, although selected with the ZN staining as AFB, not all isolates belonged to the genus Mycobacterium , and we report for the first time in Latin America the isolation of Nocardia puris , Tsukamurella pulmosis , and Gordonia sputi from sputum samples of symptomatic patients. We conclude that ZN staining does not differentiate between the genus Mycobacterium and other genera of AFB. Moreover, there is a need for a simple and more accurate tree-based identification method for mycobacterial species. For this purpose, and in development in our lab, is a web-based identification system using a phylogenetic analysis (including all AFB genera) based on 16S rDNA sequences (and in the future multigene datasets) and the closest relatives.

The COVID-19 pandemic has led to a global disruption of several services, including routine immunizations. This effect has been described in several countries, but there are few detailed studies in Latin America and no reports in Ecuador. Therefore, this work aims to quantify the reduction in routine immunizations for infants during the 2020 COVID-19 pandemic in Ecuador. 2018, 2019, and 2020 data were obtained from the Ministry of Health, Ecuador. The number of doses and the extent of immunization coverage was descriptively compared for four vaccines: rotavirus (ROTA), poliovirus (PV), pneumococcal (PCV), and pentavalent (PENTA) vaccines. There was no significant difference in doses applied during the 2018 and 2019 years. However, a significant (p < 0.05) drop of 137,000 delivered doses was observed in 2020 compared to the pre-pandemic years. Reductions in the percentage of coverage were more pronounced for the PENTA vaccine (17.7%), followed by PV (16.4%), ROTA (12%), and PCV vaccines (10.7%). Spatial analysis shows a severe impact on vaccination coverage on provinces from the Coast and Highland regions of the country. The pandemic has significantly impacted the immunization programs for infants across Ecuador. This retrospective analysis shows an urgent need to protect vulnerable zones and populations during public health emergencies.

Photo-thermal catalysis has recently emerged as a viable strategy to produce solar fuels or chemicals using sunlight. In particular, nanostructures featuring localized surface plasmon resonance (LSPR) hold great promise as photo-thermal catalysts given their ability to convert light into heat. In this regard, traditional plasmonic materials include gold (Au) or silver (Ag), but in the last years, transition metal nitrides have been proposed as a cost-efficient alternative. Herein, we demonstrate that titanium nitride (TiN) tubes derived from the nitridation of TiO 2 precursor display excellent light absorption properties thanks to their intense LSPR band in the visible-IR regions. Upon deposition of Ru nanoparticles (NPs), Ru-TiN tubes exhibit high activity towards the photo-thermal CO 2 reduction reaction, achieving remarkable methane (CH 4 ) production rates up to 1200 mmol g -1 h -1 . Mechanistic studies suggest that the reaction pathway is dominated by thermal effects thanks to the effective light-to-heat conversion of Ru-TiN tubes. This work will serve as a basis for future research on new plasmonic structures for photo-thermal applications in catalysis.

The characterization of very long-chain (>C24) polyunsaturated fatty acids (VLC-PUFAs), which are essential in the vision, neural function, and reproduction of vertebrates, is challenging because of the lack of reference standards and their very low concentrations in certain lipid classes. In this research, we have developed a new methodology for VLC-PUFA identification based on gas chromatography coupled to quadrupole/time-of-flight mass spectrometry with an atmospheric pressure chemical ionization source (GC-APCI-QTOF MS). The mass accuracy attainable with the innovative QTOF instrument, together with the soft ionization of the APCI source, provides valuable information on the intact molecule, traditionally lost with electron ionization sources due to the extensive fragmentation suffered. We have identified, for the first time, VLC-PUFAs with chains up to 44 carbons in eyes, brain, and gonads of gilthead sea bream, a commercially important fish in the Mediterranean. The added value of ion mobility-mass spectrometry (IMS), recently developed in combination with GC-QTOF MS, and the contribution of the collisional cross section (CCS) parameter in the characterization of novel VLC-PUFAs (for which reference standards are not available) have been also evaluated. The methodology developed has allowed assessing qualitative differences between farmed and wild fish, and opens new perspectives in a still scarcely known field of research.

Salinity has been demonstrated to influence the biosynthesis of long-chain (C 20-24 ) polyunsaturated fatty acids (LC-PUFAs) in teleost fish. Since LC-PUFAs are essential nutrients for vertebrates, it is central to understand how fish cope with an acute change in salinity associated with natural events. We herein report on the cloning and functional characterization of two elongation of very-long-chain fatty acid (Elovl)4 proteins, namely, Elovl4a and Elovl4b, and study the roles that these enzymes play in the biosynthesis of LC-PUFAs and very-long-chain (>C 24 ) polyunsaturated fatty acids (VLC-PUFAs) in marine teleost Pampus argenteus . The P. argenteus Elovl4 displayed all of the typical features of Elovl-like enzymes and have eyes and brain as major sites through which they exert their functions. Moreover, functional studies showed that the P. argenteus Elovl4 can effectively elongate C 18-22 substrates to C 36 VLC-PUFA. Because both P. argenteus Elovl4 are able to produce 24:5 n - 3 from shorter precursors, we tested whether the previously reported Δ6 Fads2 from P. argenteus was able to desaturate 24:5 n - 3 to 24:6 n - 3, a key step for docosahexaenoic acid (DHA) synthesis. Our results showed that P. argenteus can indeed bioconvert 24:5 n - 3 into 24:6 n - 3, suggesting that P. argenteus has the enzymatic capacity required for DHA biosynthesis through the coordinated action of both Elovl4 and Fads2. Furthermore, an acute salinity test indicated that low-salinity stress (12 ppt) upregulated genes involved in LC-PUFA biosynthesis, with 12 ppt salinity treatment showing the highest hepatic LC-PUFA content. Overall, our results unveiled that the newly characterized Elovl4 enzymes have indispensable functions in LC- and VLC-PUFA biosynthesis. Moreover, acute salinity change influenced the biosynthesis of LC-PUFA in P. argenteus . This study provided new insight into the biosynthesis of LC- and VLC-PUFAs in vertebrates and the physiological responses that teleosts have under acute salinity stress.

Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has rapidly spread throughout Latin America, a region swept by multiple previous and ongoing epidemics. There are significant concerns that the arrival of COVID-19 is currently overlapping with other viruses, particularly dengue, in various endo-epidemic regions across South America. In this report, we analyzed trends for both viral infections in Colombia during the first 20 epidemiological weeks (EWs) of 2020. From 1st January to 16th May 2020 (EWs, 1-20), a total of 52 679 cases of dengue and 14 943 cases of COVID-19 have been confirmed in Colombia. As both conditions may potentially lead to fatal outcomes, especially in patients with chronic co-morbidities, overlapping infections, and co-occurrence may increase the number of patients requiring intensive care and mechanical ventilation. In regions, such as Valle del Cauca, intensified preparation for such scenarios should be pondered, and further studies should be performed to address this critical issue in a timely matter., (© 2020 Wiley Periodicals LLC.)

Microplastics are one of the main environmental pollutants in marine ecosystems, and their presence in seawater is a consequence of the widespread use of plastic materials in modern commodities. This wide usage of plastics includes the employment of microspheres in common personal care products, which end up being ultimately released into the aquatic compartment. Known ecotoxicological effects of microplastics favoured the search for technologically viable and environmentally safer alternatives, such as paraffin wax microparticles, whose ecotoxicological risks have not been entirely characterized. To address this gap, the present study exposed mussels (Mytilus sp.) for 96 h to three densities (5 mg/L, 20 mg/L and 80 mg/L) of four size ranges (100-300 μm, 300-500 μm, 400-850 μm, and 800-1200 μm) of paraffin wax particles. Toxicological endpoints were the activities of four enzymes involved in key cellular processes, including antioxidant defence (catalase (CAT), glutathione reductase (GRed) and glutathione peroxidase (GPx)) and phase II metabolism (glutathione S-transferases (GSTs)), as well as lipid content and fatty acid profiles of the digestive gland. Significant interactions between the presence/absence of food and paraffin particle density were found, as food sometimes worked as a confounding factor in the analysed biomarkers. Despite this uncertainty, some overall patterns emerged. In general, smaller paraffin wax particles (100-300 μm) caused little effects on the activity of the four enzymes tested, whereas larger particles (800-1200 μm) caused significant effects on almost all biomarkers. CAT activity was enhanced in animals exposed to larger paraffin particles, whilst GPx activity was depressed; GRed activity was not affected by the exposure to paraffin particles. The activity of GSTs was enhanced, but only in one tested condition. No effects were observed in terms of the total lipid content and fatty acids of exposed animals. Overall, data obtained in this work suggest that, at densities of paraffin wax particles comparable to the levels found in the environment for microplastic beads, no toxicity is expected to occur in the tested mussel species, by measuring the here-assessed toxicological endpoints.

Very long-chain fatty acids (VLC-FA) play critical roles in neural tissues during the early development of vertebrates. However, studies on VLC-FA in fish are scarce. The biosynthesis of VLC-FA is mediated by elongation of very long-chain fatty acid 4 (Elovl4) proteins and, consequently, the complement and activity of these enzymes determines the capacity that a given species has for satisfying its physiological demands, in particular for the correct development of neurophysiological functions. The present study aimed to characterize and localize the expression of elovl4 genes from Sparus aurata and Solea senegalensis , as well as to determine the function of their encoded proteins. The results confirmed that both fish possess two distinct elovl4 genes, named elovl4a and elovl4b . Functional assays demonstrated that both Elovl4 isoforms had the capability to elongate long-chain (C 20-24 ), both saturated (SFA) and polyunsaturated (PUFA), fatty acid precursors to VLC-FA. In spite of their overlapping activity, Elovl4a was more active in VLC-SFA elongation, while Elovl4b had a preponderant elongation activity towards n-3 PUFA substrates, particularly in S. aurata , being additionally the only isoform that is capable of elongating docosahexaenoic acid (DHA). A preferential expression of elovl4 genes was measured in neural tissues, being elovl4a and elovl4b mRNAs mostly found in brain and eyes, respectively.

The regulation of lipogenesis and lipolysis mechanisms related to consumption of lipid has not been studied in swimming crab. The aims of the present study were to evaluate the effects of dietary lipid levels on growth, enzymes activities and expression of genes of lipid metabolism in hepatopancreas of juvenile swimming crab. Three isonitrogenous diets were formulated to contain crude lipid levels at 5·8, 9·9 and 15·1 %. Crabs fed the diet containing 15·1 % lipid had significantly lower growth performance and feed utilisation than those fed the 5·8 and 9·9 % lipid diets. Crabs fed 5·8 % lipid had lower malondialdehyde concentrations in the haemolymph and hepatopancreas than those fed the other diets. Highest glutathione peroxidase in haemolymph and superoxide dismutase in hepatopancreas were observed in crabs fed 5·8 % lipid. The lowest fatty acid synthase and glucose 6-phosphate dehydrogenase activities in hepatopancreas were observed in crabs fed 15·1 % lipid, whereas crabs fed 5·8 % lipid had lower carnitine palmitoyltransferase-1 activity than those fed the other diets. Crabs fed 15·1 % lipid showed lower hepatopancreas expression of genes involved in long-chain-PUFA biosynthesis, lipoprotein clearance, fatty acid uptake, fatty acid oxidation, lipid anabolism and lipid catabolism than those fed the other diets, whereas expression of some genes of lipoprotein assembly and fatty acid oxidation was up-regulated compared with crabs fed 5·8 % lipid. Overall, high dietary lipid level can inhibit growth, reduce antioxidant enzyme activities and influence lipid metabolic pathways to regulate lipid deposition in crab.

Introduction: Previous studies suggest that the severity of alterations in cognitive functions in people with a diagnosis of bipolar I disorder (BP-I) are directly related to the deterioration in overall functioning and life quality. Altered cognitive function is associated with a worse prognosis of BP-I, however little is known about the relationship between cognitive functions and the clinical features of BP-I., Objective: To establish possible associations between cognitive function and the clinical and socio-demographic characteristics of patients with BP-I., Methods: Cognitive function was evaluated by applying a neuropsychological battery to a group of patients with a BP-I diagnosis, who did not have affective episodes for at least 6 months, and who were outpatients treated at the San Juan de Dios Clinic in Manizales. The statistical analysis involved using clustering methocology in order to divide those patients with the best and worst cognitive function and it was later correlated with the clinical and socio-demographic variables., Results: A statistically significant corrrelation was found between the number of years of education and the age the disorder started with alterations in the level of cognitive function (P=.002 and P=.017 respectively). No significant correlations were found with other variables., Conclusions: An early onset of the pathology and fewer years of education seems to be risk factors associated with poorer cognitive function in patients with BP-I., (Copyright © 2017 Asociación Colombiana de Psiquiatría. Aedes aegypti is the vector of the arboviruses causing dengue, chikungunya and zika infections in Mexico. However, its presence in public places has not been fully evaluated. In a cemetery from Merida, Yucatan, Mexico, the productivity of Ae. aegypti, the gonotrophic cycle, and the presence of Ae. aegypti females infected with arboviruses were evaluated. Immature and adult mosquitoes were inspected every two months between April 2016 to June 2017. For the gonotrophic cycle length, the daily pattern of total and parous female ratio was registered and was analyzed using time series analysis. Ae. aegypti females were sorted into pools and assayed for flavivirus RNA by RT-PCR and Sanger sequencing. Aedes aegypti immatures represented 82.86% (8,627/10,411) of the collection. In total, 1,648 Ae. aegypti females were sorted into 166 pools. Two pools were positive; one for dengue virus (DENV-1) and the other for zika virus (ZIKV). The phylogenetic analysis revealed that the DENV-1 is more closely related to isolates from Brazil. While ZIKV is more closely related to the Asian lineage, which were isolates from Guatemala and Mexico. We report some evidence of vertical transmission of DENV-1 in nulliparous females of Ae. aegypti. The gonotrophic cycle was four and three days in the rainy and dry season, respectively. The cemetery of Merida is an important focus of Ae. aegypti proliferation, and these environments may play a role in arboviruses transmission; probably limiting the efficacy of attempts to suppress the presence of mosquitoes in domestic environments.

The aim of this study is to report 11 cases of erector spinae plane (ESP) block used for unilateral or bilateral abdominal surgery, adding to the overall limited experience with abdominal ESP block. The procedures were carried out at a teaching hospital in 11 patients (eight males, three females, ages 36-80 years) requiring abdominal surgery, including laparoscopic surgery. Each patient required surgery under different physical circumstances and likely different conceptions of what constituted pain. Two of the eleven patients were administered the ESP block and did not require general anaesthesia. Most of the patients with the ESP block maintained a numerical rating scale (NRS) for pain of 0-2/10 postoperatively. An occasional patient required paracetamol analgesia. There were no cases of opiate rescue. Obesity in a 46-year-old woman was believed to cause unclear ultrasonographic visualisation, interfering with entry of the ESP catheter. She, however, had no post-operative pain. She was given the usual intravenous metamizole 2 g for 10 h and required only analgesics at 16 h. ESP block, which produces analgesia by blocking trunk nerves, is an appropriate approach to patients requiring abdominal surgery, whether laparoscopic or open., Competing Interests: There are no conflicts of interest.

Introduction: The province of Pichincha in Ecuador is an endemic area of cutaneous leishmaniasis, where anthropophilic sand flies with natural infection by Leishmania, have been reported as vectors. However, the role in transmission of zoophilic species has not been evaluated., Objective: To evaluate natural infection by Leishmania in two zoophilic phlebotomine sand fly species, Lutzomyia reburra and Lu. barrettoi majuscula, and one anthropophilic species, Lu. trapidoi, as well as the endophagy and synanthropism of these species in the northwest of Pichincha., Materials and Methods: Phlebotomines were collected using CDC light traps in different habitats and altitudes with presence of cutaneous leishmaniasis. Leishmania infection was detected using genomic DNA from females of the collected sand flies. We amplified the internal transcribed spacer gene of ribosomal RNA I (ITS1), the mitochondrial topoisomerase II gene (mtTOPOII), and the nuclear topoisomerase II gene (TopoII). Percentages of positivity for Leishmania, at spatio-temporal scale, proportion of endophagy and synanthropism index were calculated., Results: Natural infection was determined for Le. amazonensis in Lu. reburra (9.5%) and Lu. b. majuscula (23.8%), while in Lu. trapidoi we detected Le. amazonensis, Le. brazilienis and Le. naiffi-lainsoni. Phlebotomines were asynanthropic and with low endophagy., Conclusion: Natural infection with Le. amazonensis was recorded for the first time in Lu. reburra and Lu. b. majuscula, demonstrating the importance of zoophilic phlebotomines in the maintenance of the Leishmania transmission cycle in endemic foci.

Tapeworms Taenia solium and Taenia saginata are the causative agents of taeniasis/cysticercosis. These are diseases with high medical and veterinary importance due to their impact on public health and rural economy in tropical countries. The re-emergence of T. solium as a result of human migration, the economic burden affecting livestock industry, and the large variability of symptoms in several human cysticercosis, encourage studies on genetic diversity, and the identification of these parasites with molecular phylogenetic tools. Samples collected from the Ecuadorian provinces: Loja, Guayas, Manabí, Tungurahua (South), and Imbabura, Pichincha (North) from 2000 to 2012 were performed under Maximum Parsimony analyses and haplotype networks using partial sequences of mitochondrial DNA, cytochrome oxidase subunit I (COI) and NADH subunit I (NDI), from Genbank and own sequences of Taenia solium and Taenia saginata from Ecuador. Both species have shown reciprocal monophyly, which confirms its molecular taxonomic identity. The COI and NDI genes results suggest phylogenetic structure for both parasite species from south and north of Ecuador. In T. solium, both genes gene revealed greater geographic structure, whereas in T. saginata, the variability for both genes was low. In conclusion, COI haplotype networks of T. solium suggest two geographical events in the introduction of this species in Ecuador (African and Asian lineages) and occurring sympatric, probably through the most common routes of maritime trade between the XV-XIX centuries. Moreover, the evidence of two NDI geographical lineages in T. solium from the north (province of Imbabura) and the south (province of Loja) of Ecuador derivate from a common Indian ancestor open new approaches for studies on genetic populations and eco-epidemiology., (Copyright © 2016 Elsevier Inc. Oropouche virus (OROV), genus Orthobunyavirus, family Bunyaviridae, is an important cause of human illness in tropical South America. Herein, we report the isolation, complete genome sequence, genetic characterization, and phylogenetic analysis of an OROV species reassortant, Madre de Dios virus (MDDV), obtained from a sick monkey (Cebus olivaceus Schomburgk) collected in a forest near Atapirire, a small rural village located in Anzoategui State, Venezuela. Mayaro virus produces nonspecific, sublethal disease symptoms, often confused with dengue, but with symptoms of arthalgias that can cause incapacitating disability. Outbreaks have been localized and sporadic in the Pan-Amazonia forest since its first isolation in 1954 (Trinidad and Tobago). The literature available is scarce, diverse and dispersed. Mayaro virus is an alphavirus, phylogenetically related to the Semliki forest antigenic complex. UNA and Mayaro viruses are the only viruses of this complex that have been isolated in the New World. Mayaro consists of single-stranded RNA of positive charge, length of 12 kb, subdivided into genomic and subgenomic regions, which encode nonstructural and structural proteins respectively. Mayaro shows a great plasticity in vertebrate host infection, whereas high specificity in the family Culicidae (mosquitoes). Risk factors of infection are associated with forest areas of northern South America and the rainy season. Two genotypes of MAYV have been identified, L (Belterra, Brazil) and D (widely distributed in the Pan-Amazonia). The enzootic cycle is similar to the jungle cycle of yellow fever, which involves Haemagogus mosquitoes and monkeys as reservoirs. However, the involvement of other secondary vectors and other hosts may be important in spread of the virus. Humans may have high levels of viremia, and efficient experimental transmission has been demonstrated in Aedes aegypti, Aedes albopictus and Aedes scapularis, suggesting a significant risk to public health in urban, rural and peridomestic locations close to enzootic foci of Mayaro virus.

Mosquitoes (Diptera: Culicidae) are holometabolous insects with aquatic immature stages, which use a broad variety of larval habitats, from ground water bodies to Phytothelmata (water deposits in plants) and artificial deposits. The availability of breeding sites often determines the upper limits of mosquito ranges. We built a database with 9,607 records with 432 localities, 19 genera and 254 species. The Andean mountains have 77% of the highest mosquito records including Aedes euris with record at 3,133 m, followed by three species of Anopheles--subgenera Kerteszia--with the upper limit of 2,680 m. Wyeomyia bicornis and Culex daumastocampa at 2,550 m were the highest records in the Central-Coastal cordillera, while the highest record in Pantepui was Wyeomyia zinzala at 2,252 m. The species associated with phytothelmata (Bromeliaceae and Sarraceniaceae) represent 60% of the records. The upper limits of Culex quinquefasciatus and Anopheles (Kerteszia) species could represent the theoretical limit for transmission of filariasis or arboviruses, by Culex, and malaria by Anopheles (Kerteszia) in Venezuela. Similarly, a vector of Dengue, Aedes aegypti, has not been not recorded above 2,000 m.

Age-related changes in the balance between endogenous pro-oxidative and antioxidative processes in the freshwater cladoceran Daphnia magna (Crustacea) were assessed. The activities of key antioxidant enzymes including catalase, superoxide dismutase and glutathione peroxidase and levels of lipid peroxidation measured as thiobarbituric acid-reactive substances (TBARS) were determined in eight age classes, covering juvenile, young and senescent adults. Age-related changes in fatty acid composition were also measured to examine the contribution of polyunsaturated fatty acids (PUFA) in the peroxidation status of animals. Biochemical responses depicted in this study demonstrated that age-related decline in survival was accompanied by increasing oxidative stress and oxidative damage. Enhanced oxidative stress in aging D. magna was suggested by the significant increase in the formation of lipid peroxides, and a concomitant reduction of unsaturated fatty acids of 20 or more carbon atoms. Because aging was accompanied by selective loss of key antioxidant enzymes and small changes in the amount of PUFA, the breakdown of antioxidant defences might have directly contributed to oxidative stress, membrane lipid peroxide and a decline of survival. Indeed, the results reported here, indicate that age-related increases of lipid peroxides were at least partially due to the functional imbalance of enzymatic antioxidant defences.
