Arbuscular mycorrhizal (AM) fungi and plant growth-promoting rhizobacteria (PGPR) have potential for the biocontrol of soil-borne diseases. The objectives of this study were to quantify the interactions between AM fungi [Glomus versiforme (Karsten) Berch and Glomus mosseae (Nicol. & Gerd.) Gerdemann & Trappe] and PGPR [Bacillus polymyxa (Prazmowski) Mace and Bacillus sp.] during colonization of roots and rhizosphere of tomato (Lycopersicon esculentum Mill) plants (cultivar Jinguan), and to determine their combined effects on the root-knot nematode, Meloidogyne incognita, and on tomato growth. Three greenhouse experiments were conducted. PGPR increased colonization of roots by AM fungi, and AM fungi increased numbers of PGPR in the rhizosphere. Dual inoculations of AM fungi plus PGPR provided greater control of M. incognita and greater promotion of plant growth than single inoculations, and the best combination was G. mosseae plus Bacillus sp. The results indicate that specific AM fungi and PGPR can stimulate each other and that specific combinations of AM fungi and PGPR can interact to suppress M. incognita and disease development., (© Springer-Verlag 2012)

Homology-based cloning was used to obtain a new gene (named FIP-tvc) from the genomic DNA of the Chinese traditional medicinal mushroom Trametes versicolor. FIP-tvc is a member of the fungal immunomodulatory protein family, is composed of 336bp encoding 111 amino acids, and is highly homologous with other fungal immunomodulatory proteins. In addition, an expression system for FIP-tvc was constructed. The results indicated that recombinant protein FIP-tvc could be expressed in Escherichia coli and that about 20% of the expressed protein was in soluble form. Recombinant FIP-tvc was capable of agglutinating mouse and rat red blood cells. Furthermore, recombinant protein FIP-tvc could selectively enhance the expression of interleukin (IL)-1α, IL-2, IL-5, IL-6, tumor necrosis factor-alpha (TNF-α), and lympotoxin (LT) in mouse spleen cells., (Crown Copyright © 2012. Background: Species in the ascomycete fungal genus Cordyceps have been proposed to be the teleomorphs of Metarhizium species. The latter have been widely used as insect biocontrol agents. Cordyceps species are highly prized for use in traditional Chinese medicines, but the genes responsible for biosynthesis of bioactive components, insect pathogenicity and the control of sexuality and fruiting have not been determined., Results: Here, we report the genome sequence of the type species Cordyceps militaris. Phylogenomic analysis suggests that different species in the Cordyceps/Metarhizium genera have evolved into insect pathogens independently of each other, and that their similar large secretomes and gene family expansions are due to convergent evolution. However, relative to other fungi, including Metarhizium spp., many protein families are reduced in C. militaris, which suggests a more restricted ecology. Consistent with its long track record of safe usage as a medicine, the Cordyceps genome does not contain genes for known human mycotoxins. We establish that C. militaris is sexually heterothallic but, very unusually, fruiting can occur without an opposite mating-type partner. Transcriptional profiling indicates that fruiting involves induction of the Zn2Cys6-type transcription factors and MAPK pathway; unlike other fungi, however, the PKA pathway is not activated., Conclusions: The data offer a better understanding of Cordyceps biology and will facilitate the exploitation of medicinal compounds produced by the fungus.

Tricladins A and B (1 and 2, resp.), new imidazolone-containing alkaloids, together with five known metabolites, bacillamides A, B (3 and 4, resp.), 20-hydroxyaflavinine (5), N-(2-phenylethyl)acetamide (6), and N(b)-acetyltryptamine (7), have been isolated from the crude extract of the ascomycete fungus Tricladium sp. The structures of 1 and 2 were elucidated primarily by NMR and MS methods. Six new secondary metabolites including two aphidicolin analogues, inflatins A (1) and B (2), and four chlamydosporol derivatives, inflatins C-F (3-6), have been isolated from the crude extract of Tolypocladium inflatum. The structures of 1-6 were determined mainly by NMR experiments, and 4 and 5 were further confirmed by X-ray crystallography. The absolute configurations of C-16 in 1 and C-5 in 3 were deduced via the circular dichroism data of the in situ formed [Rh₂(OCOCF₃)₄] complexes, whereas that of 4 was assigned by X-ray crystallography using Cu Kα radiation. Compounds 1 and 2 showed modest cytotoxicity against a panel of eight human tumor cell lines.

To determine a suitable DNA barcode for the genus Neonectria, the internal transcribed spacer rDNA, β-tubulin, EF-1α, and RPB2 genes were selected as candidate markers. A total of 205 sequences from 19 species of the genus were analyzed. Intra- and inter-specific divergences and the ease of nucleotide sequence acquisition were treated as criteria to evaluate the feasibility of a DNA barcode. Our results indicated that any single gene among the candidate markers failed to serve as a successful barcode, while the combination of the partial EF-1α, and RPB2 genes recognized all species tested. We tentatively propose the combined partial EF-1α and RPB2 genes as a DNA barcode for the genus. During this study, two cryptic species were discovered, based on the combined data of morphology and DNA barcode information. We described and named these two new species N. ditissimopsis and N. microconidia.

The Amsterdam Declaration on Fungal Nomenclature was agreed at an international symposium convened in Amsterdam on 19-20 April 2011 under the auspices of the International Commission on the Taxonomy of Fungi (ICTF). The purpose of the symposium was to address the issue of whether or how the current system of naming pleomorphic fungi should be maintained or changed now that molecular data are routinely available. The issue is urgent as mycologists currently follow different practices, and no consensus was achieved by a Special Committee appointed in 2005 by the International Botanical Congress to advise on the problem. The Declaration recognizes the need for an orderly transitition to a single-name nomenclatural system for all fungi, and to provide mechanisms to protect names that otherwise then become endangered. That is, meaning that priority should be given to the first described name, except where that is a younger name in general use when the first author to select a name of a pleomorphic monophyletic genus is to be followed, and suggests controversial cases are referred to a body, such as the ICTF, which will report to the Committee for Fungi. If appropriate, the ICTF could be mandated to promote the implementation of the Declaration. In addition, but not forming part of the Declaration, are reports of discussions held during the symposium on the governance of the nomenclature of fungi, and the naming of fungi known only from an environmental nucleic acid sequence in particular. Possible amendments to the Draft BioCode (2011) to allow for the needs of mycologists are suggested for further consideration, and a possible example of how a fungus only known from the environment might be described is presented.

The epipolythiodioxopiperazines (ETPs) are fungal secondary metabolites proven to trigger both apoptotic and necrotic cell death of tumor cells. However, the underlying mechanism of their regulatory role in macroautophagy and the interplay between autophagy and apoptosis initiated by the ETPs, remain unexplored. In the current work, we found that 11'-deoxyverticillin A (C42), a member of the ETPs, induces autophagosome formation, accumulation of microtubule-associated protein 1 light chain 3-II (LC3-II ) and degradation of sequestosome 1 (SQSTM1/p62). In addition, the LC3-II accrual and p62 degradation occur prior to caspase activation and coincide with PARP activation. Inhibition of autophagy by either chemical inhibitors or by RNA interference single knockdown of essential autophagic genes partially reduces the cell death and the cleavage of both caspase 3 and PARP. Necrostatin-1, a specific inhibitor of necroptosis, inhibits both the augmentation of LC3-II and the cleavage of caspase 3, which was confirmed by depletion of receptor-interacting protein 1 (RIP-1), a crucial necrostatin-1-targeted adaptor kinase mediating cell death and survival. Moreover, inhibition of PARP by either chemical inhibitors or RNA interference provides obvious protection for cell viability and suppresses the LC3-II accretion caused by C42 treatment. Interestingly, double silencing of LC3 and p62 completely suppressed PARP cleavage and concurrently and maximally augmented the PAR formation induced by C42. Collectively, we have demonstrated that C42 enhances the cellular autophagic process, which requires both PARP and RIP-1 participation, preceding and possibly augmenting, the caspase-dependent apoptotic cell death.

Four new cyclic tetradecapeptides, verrucamides A-D (1-4), have been isolated from the solid-substrate fermentation culture of the ascomycete fungus Myrothecium verrucaria. The structures of these compounds, each featuring six N-methylated amino acid residues, were elucidated primarily by NMR and MS methods. The absolute configurations of 1-4 were assigned by application of Marfey's method on their acid hydrolysates. Compounds 1-4 showed antimicrobial activity against the Gram-positive bacterium Staphylococcus aureus.

Bioactive proteins represent an important group of functional agents in medicinal mushrooms. Trametes versicolor (L.) Lloyd is a mushroom frequently used in traditional Chinese medicine for its anti-tumor and immunomodulatory activities. A new immunomodulatory protein from T. versicolor, named TVC, was purified by ammonium sulfate precipitation, ion-exchange chromatography and gel filtration chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of the purified protein revealed a single band with a molecular weight of 15.0 kD. Native polyacrylamide gel analysis revealed a band at 30 kD, indicating that TVC exists in solution as a homodimer. Isoelectric focusing showed that TVC was an acidic protein with an isoelectric point of 4.0. TVC was found to lack carbohydrate modifications (based on periodic acid/Schiff staining) and it does not agglutinate mouse red blood cells, suggesting that TVC is not a lectin-like protein. Biological activity assays demonstrated that TVC can enhance the proliferation of splenocytes, while it has no stimulatory effects on CD4(+) and CD8(+) T cells. TVC markedly increases the proliferation of human peripheral blood lymphocytes in a dose-dependent manner and enhances the production of both nitric oxide and tumor necrosis factor-alpha by lipopolysaccharide-induced murine macrophages. The results indicate that TVC is an immunostimulant that can boost immune response. Comparison of the N-terminal amino acid residues and mass spectrometry results with the protein database revealed no homologous proteins.

New N-hydroxypyridones, militarinones E (1) and F (2), phenylhydrazones, farylhydrazones A (3) and B (4), a quinazolinone, 2-(4-hydroxybenzyl)quinazolin-4(3H)-one (5), and the known militarinones A (6) and B (7) were isolated from cultures of the Cordyceps-colonizing fungus Isaria farinosa. The structures of 1-5 were elucidated by spectroscopic methods, and 3 was confirmed by X-ray crystallography. The absolute configuration of the C-4' secondary alcohol in 1 was deduced via the circular dichroism data of the in situ formed [Rh(2)(OCOCF(3))(4)] complex. Compounds 1 and 6 showed significant cytotoxicity against A549 cells, whereas 7 was active against Staphylococcus aureus, Streptococcus pneumoniae, and Candida albicans.

Background: Ophiocordyceps sinensis (syn. Cordyceps sinensis), which is a parasite of caterpillars and is endemic to alpine regions on the Tibetan Plateau, is one of the most valuable medicinal fungi in the world. "Natural O. sinensis specimens" harbor various other fungi. Several of these other fungi that have been isolated from natural O. sinensis specimens have similar chemical components and/or pharmaceutical effects as O. sinensis. Nevertheless, the mycobiota of natural O. sinensis specimens has not been investigated in detail., Methodology/principal Findings: Based on the technique of PCR-single-strand conformation polymorphism (PCR-SSCP), the mycobiota of three different sections (stromata, sclerotia, and mycelial cortices) from natural O. sinensis specimens were investigated using both culture-dependent and -independent methods. For the culture-dependent method, 572 fungal strains were isolated, and 92 putative operational taxonomic units (OTUs) were identified from 226 sequenced strains with the threshold of 97%. For the culture-independent method, 490 fungal clones were identified from about 3000 clones of ITS fragments from the whole-community DNA; based on PCR-SSCP analyses, 266 of these clones were selected to be sequenced, and 118 putative OTUs were detected. The overwhelming majority of isolates/clones and OTUs were detected from mycelial cortices; only a few were detected from stromata and sclerotia. The most common OTUs detected with both methods belonged to Ascomycota; however, only 13 OTUs were detected simultaneously by both methods. Potential novel lineages were detected by each of the two methods., Conclusions/significance: A great number of fungal species present in the mycobiota of naturally-occurring O. sinensis specimens were detected, and many of them may represent undescribed lineages. That only a few of the same OTUs were detected by both methods indicated that different methods should be used. This study increased our understanding about the fungal community structure of this valuable medicinal herb.

The development of fungal biopesticides requires the efficient production of large numbers spores or other propagules. The current study used published information concerning carbon concentrations and C:N ratios to evaluate the effects of carbon and nitrogen sources on sporulation of Paecilomyces lilacinus (IPC-P and M-14) and Metarhizium anisopliae (SQZ-1-21 and RS-4-1) in a two-stage cultivation system. For P. lilacinus IPCP, the optimal sporulation medium contained urea as the nitrogen source, dextrin as the carbon source at 1 g/L, a C:N ratio of 5:1, with ZnSO(4)·7H(2)O at 10 mg/L and CaCl(2) at 3 g/L. The optimal sporulation medium for P. lilacinus M-14 contained soy peptone as the nitrogen source and maltose as the carbon source at 2 g/L, a C:N ratio of 10:1, with ZnSO(4)·7H(2)O at 250 mg/L, CuSO(4)·5H(2)O at 10 mg/L, H(3)BO(4) at 5 mg/L, and Na(2)MoO(4)·2H(2)O at 5 mg/L. The optimum sporulation medium for M. anisopliae SQZ-1-21 contained urea as the nitrogen source, sucrose as the carbon source at 16 g/ L, a C:N ratio of 80:1, with ZnSO(4)·7H(2)O at 50 mg/L, CuSO(4)·5H(2)O at 50 mg/L, H(3)BO(4) at 5 mg/L, and MnSO(4)·H(2)O at 10 mg/L. The optimum sporulation medium for M. anisopliae RS-4-1 contained soy peptone as the nitrogen source, sucrose as the carbon source at 4 g/L, a C:N ratio of 5:1, with ZnSO(4)·7H(2)O at 50 mg/L and H(3)BO(4) at 50 mg/L. All sporulation media contained 17 g/L agar. While these results were empirically derived, they provide a first step toward low-cost mass production of these biocontrol agents.

LZ-8 protein, isolated from a well known Chinese traditional medicinal fungus Ganoderma lucidum, is the first member of fungal immunomodulatory protein, members of which have been isolated from a variety of medicinal and edible mushrooms in the last two decades. The protein plays a multifunctional and important role in modulating immune system. In this report, in order to get LZ-8 protein crystals, the LZ-8 gene was expressed and purified by affinity chromatography, gel filtration chromatography and anion exchange chromatography subsequently. The protein was then crystallized using the hanging-drop vapour-diffusion method. The LZ-8 crystals were obtained and the phase information was calculated by X-ray diffraction. The resolution of LZ-8 crystals is 3.2A. This study will provide an insight into the structure of fungal immunomodulatory proteins.

To develop fungal biopesticides, the nutritional requirements on available culture media that maximize mycelia and spore yields at low cost are essential for their mass-production. The objective of present study was to find the optimal combinations for the most mycelia yield in submerged and the highest spore yield on solid. Have a better understanding of their basic physiology, and have a better yield of mycelia and conidia with short time and low cost will help to provide valuable information for mass production and commercialization.

Cytosporolides A-C (1-3), three caryophyllene-derived meroterpenoids with a unique peroxylactone skeleton, were isolated from cultures of the fungus Cytospora sp. Their structures were elucidated by NMR spectroscopy, and the absolute configuration of the 5,6-diol moiety in 1 was assigned using Snatzke's method. Compounds 1-3 showed significant antimicrobial activity against the Gram-positive bacteria Staphylococcus aureus and Streptococcus pneumoniae.

This study measured trap induction and trapping on agar disks as affected by juvenile stages (J1, J2, J3, and J4) of the nematode Caenorhabditis elegans and by species of nematode-trapping fungi. Eight species of nematode-trapping fungi belonging to the family Orbiliaceae and producing four kinds of traps were studied: adhesive network-forming Arthrobotrys oligospora, A. vermicola, and A. eudermata, constricting ring-forming Drechslerella brochopaga, and Dr. stenobrocha, adhesive column-forming Dactylellina cionopaga, and adhesive knob-forming Da. ellipsospora, and Da. drechsleri. The number of traps induced generally increased with increasing juvenile stages of C. elegans. The ability to capture the juveniles tended to be similar among isolates that produced the same kind of trap but differed among species that produced different kinds of traps. Trapping by Dr. stenobrocha and Da. cionopaga was correlated with trap number and with juvenile stage. A. oligospora and A. vermicola respectively captured more than 92 and 88% of the J1, J3, and J4 but captured a lower percentage of J2. The knob-producing isolates captured more younger than elder juveniles. Partial correlation analyses demonstrated that the trap induction of the most fungal species positively correlated with the juvenile size and motility, which was juvenile stage dependent. Overall, trap induction and trapping correlated with C. elegans juvenile stage (size and motility) in six species of trapping fungi.

The nematophagous fungus Pochonia chlamydosporia (Clavicipitaceae) and entomopathogenic fungus Beauveria bassiana (Cordycipitaceae) have great potential for biological control. However, a significant barrier to their commercial development as mycopesticides is the high costs associated with production. Carbon (C) concentration and C to nitrogen ratio (C:N ratio) greatly affect fungal growth and sporulation. Effects of C concentration and C:N ratio differed when the fungi were cultivated using two different methods: the conventional (continuous cultivation) method and a novel "two-stage" method. Sporulation of P. chlamydosporia (HSY-12-14) was the highest when the media contained 6 g l(-1) C and a C:N ratio of 40:1 or 8 g l(-1) C and C:N ratios of 20:1 or 40:1 for the conventional method but 8 g l(-1) C and a C:N ratio of only 10:1 with the novel "two-stage" method. Sporulation of B. bassiana (IBC1201) was the highest when the media contained 12 g l(-1) C and a C:N ratio of 40:1 with the conventional method but only 4 g l(-1) C and a C:N ratio of 5:1 with the novel "two-stage" method. In addition, the nutritional requirements as determined by the conventional method differed for mycelial growth and sporulation. Understanding the effects of nutrition on sporulation can help programs seeking to use these organisms as biological control agents and is essential for their mass production and commercialization.

The new polyketides leptosphaerins A-G (1-7) and the known compounds monodictysin B (8), 2-(2,6-dihydroxybenzoyl)-3-hydroxy-5-methylbenzoic acid (9), and 2-(2,6-dihydroxy-4-methylbenzoyl)-6-hydroxybenzoic acid (10) have been isolated from solid cultures of the ascomycete fungus Leptosphaeria sp. The absolute configurations of compounds 1, 2, and 6 were assigned using the modified Mosher method, whereas that of C-2 in 3 was determined via the circular dichroism data of the [Rh(2)(OCOCF(3))(4)] complex. Compound 7 showed antifungal activity against Aspergillus flavus.

Environmental factors greatly affect the activity of soil microorganisms. However, not enough is known about how the environment affects the abundance and activity of many beneficial microorganisms after they are released into soil. Hirsutella minnesotensis is an important fungal parasite of nematodes and has shown great potential in nematode control. The objective of this study was to measure the effects of environmental factors on the abundance and activity of H. minnesotensis in soil. Fungal mycelium was mixed with dry soil (1% wet weight per dryweight) and placed into 50-mL plastic tubes. Soil temperature ranged from 5 to 30 degrees C; soil water content ranged from 6% to 22%; and the soil was supplemented with 0-70% of fine soil particles (silt and clay) or sand. After 24 days, fungal abundance was quantified by real-time PCR and its activity was inferred from the numbers of second-stage juveniles (J2) parasitized. The amount of H. minnesotensis DNA g(-1) soil and the percentage of J2 parasitized by H. minnesotensis were higher with lower soil temperature and water content, and higher fine particle content, demonstrating that H. minnesotensis has a greater potential to multiply and control pest nematodes in cooler, drier and heavier soils.

Background: Ophiocordyceps sinensis (syn. Cordyceps sinensis), endemic to alpine regions on the Tibetan plateau, is one of the most valuable medicinal fungi in the world. Huge commercial demand has led to excessive harvest and a dramatic decline in its numbers. The diversity of terrains and climates on the Tibetan Plateau and the broad insect host range (more than 50 species in the family Hepialidae) may have resulted in substantial intraspecific genetic diversity for this fungus. The objective of this study was to evaluate the population distribution of O. sinensis from geographically diverse regions of the Tibetan Plateau based on nrDNA ITS and MAT1-2-1 gene sequences. Understanding of the genetic diversity and genesis of O. sinensis will provide important information for the evolution and conservation of this fungus., Results: Significant sequence variations in the ITS and MAT1-2-1 genes (27 and 23 informative sites, eight and seven haplotypes, respectively) were observed. Phylogenetic analysis based on ITS sequences, MAT1-2-1 sequences, or their combined data set, clustered isolates from northern regions in one clade (clade I), whereas isolates from southern regions were dispersed in all four clades (clade I-IV). Single-strand conformation polymorphism (SSCP) analyses of 2639 ITS clones from seven samples revealed 91 different SSCP patterns that were subsequently sequenced. ITS heterogeneity was found in XZ-LZ07-H1 (Nyingchi population), and 17 informative sites and five haplotypes were detected from 15 clones. The five haplotypes clustered into three clades (clade I, II, and IV)., Conclusions: Significant genetic divergence in O. sinensis was observed and the genetic diversification was greater among southern isolates than that among northern isolates. The polymorphism of nrDNA ITS sequences suggested that O. sinensis spread from a center of origin (the Nyingchi District) to southern regions and subsequently to northern areas. These results suggest that southern populations are important reservoirs of genetic diversity and should be taken into account in conservation programs.

New diketopiperazines, epicoccins E-H (1-4) and diphenylalazines A (5) and B (6), have been isolated from the solid-substrate fermentation culture of the Cordyceps-colonizing fungus Epicoccum nigrum. The structures of 1-6 were determined primarily by NMR experiments, and the structure of 1 was confirmed by X-ray crystallography. The absolute configurations of 3 and 5 were assigned using the modified Mosher (3) and Marfey's (5) methods, respectively. Compounds 3-5 showed inhibitory effects on HIV-1 replication in C8166 cells.

Cycloaspeptides F (1) and G (2), two new cyclic pentapeptides, and the known cycloaspeptides A (3), C (4), and bisdethiodi(methylthio)hyalodendrin (5) have been isolated from the crude extract of the fungus Isaria farinosa that colonizes Cordyceps sinensis. The structures of 1 and 2 were elucidated primarily by NMR and MS methods. The absolute configuration of 1 was assigned using Marfey's method on its acid hydrolysate. Compounds 1 and 2 showed cytotoxic effects against HeLa and MCF7 cell lines, displaying the same magnitude of activity toward the MCF7 cells as the positive control 5-fluorouracil.

Hirsutella rhossiliensis OWVT-1 has substantial potential as a biocontrol agent against plant-parasitic nematodes. Serine proteases have emerged as a potentially useful factor in the nematode-fungus interactions. When grown in liquid culture with the nematode Panagrellus redivivus as the sole nitrogen source, an extracellular alkaline protease (Hasp) was produced by the OWVT-1. The purified Hasp killed the juveniles of the soybean-cyst nematode (Heterodera glycines) and degraded proteins of the nematode cuticle. The molecular mass of Hasp was estimated to be 33 kDa. The optimum pH and temperature for enzyme activity were pH 9 and 75 degrees C. The amino acid sequence obtained by the N-terminal sequence analysis was applied for the primer design to isolate the Hasp cDNA gene, which consists of 1170 bp open reading frame. Analysis of the cDNA and corresponding genomic sequence revealed that Hasp included four exons (279, 186, 513, and 192 bp) divided by three introns (65, 99, and 93 bp). Southern blotting showed that Hasp was a single-copy gene in the genome. The deduced amino acid sequence was very similar to other serine proteases of endoparasitic and egg-parasitic fungi of nematodes and of entomopathogenic fungi but was less similar to the serine proteases of nematode-trapping fungi. In a phylogenetic analysis of the amino acid sequences of serine proteases, the serine protease of H. rhossiliensis OWVT-1 clustered with the serine proteases of parasites of nematode eggs rather than with those of the trapping fungi., (2009 Elsevier GmbH.)

Five new asterric acid derivatives, ethyl asterrate (3), n-butyl asterrate (4), and geomycins A-C (6-8), have been isolated from cultures of an isolate of the Antarctic ascomycete fungus Geomyces sp. The structures of these metabolites were elucidated by NMR spectroscopy. The absolute configuration of 8 was determined by application of the CD excitation chirality method. Compound 7 displayed antifungal activity against Aspergillus fumigatus, whereas 8 showed antimicrobial activities against gram-positive and gram-negative bacteria.

The entomopathogenic fungus Cordyceps sinensis has been important in traditional Chinese medicine, but is yet to be commercially cultivated. Difficulty in cultivation results in part from the low percentage of fungal infection on artificially inoculated host insects. To better understand the infection mechanism, we cloned two cuticle-degrading serine protease genes (csp1 and csp2) from C. sinensis. These enzymes are novel members of the S8A subfamily of proteases. Identities of cDNA or amino acid sequences between Csp1 and Csp2 were 72.9% and 68.9%, respectively. After successful expression in the yeast Pichia pastoris, recombinant enzymes were purified and characterized using the synthetic substrate N-suc-AAPF-p-NA. Both were chymotrypsin-like serine proteases with an optimum pH of 7.0 and an optimal temperature of 40 degrees C (Csp1) or 50 degrees C (Csp2). Bioassay revealed that Csp1 and Csp2 degraded the cuticle proteins of larval Hepialus sp. in vitro. This is the first report of serine proteases from C. sinensis.

Alachalasins A-G (1-7), seven new cytochalasin-type of metabolites, have been isolated from cultures of an isolate of Stachybotrys Podospora vesticola [corrected] The structures of these compounds were determined mainly by analysis of their NMR spectroscopic data, and the absolute configuration of 1 was assigned by application of the modified Mosher method. Alachalasin A (1) displayed inhibitory effect on HIV-1LAI replication in C8166 cells with an EC50 of 8.01 microM; alachalasins D (4) and G (7) showed modest antimicrobial activity.

Epicoccins A-D (1-4), four unique epipolythiodioxopiperazines possessing unusual sulfur bridges, have been isolated from cultures of a Cordyceps-colonizing isolate of Epicoccum nigrum. The structures of these compounds were determined mainly by analysis of their NMR spectroscopic data. The relative and absolute configuration of epicoccin A (1) was established by single-crystal X-ray diffraction analysis. Epicoccin A (1) showed modest antimicrobial activity.

Gliocladinins A (1) and B (2), two new p-terphenyl derivatives, have been isolated from solid cultures of an isolate of Gliocladium sp. that colonizes Cordyceps sinensis. The structures of these compounds were determined mainly by analysis of their NMR spectroscopic data. In standard disk assays, compounds 1 and 2 showed modest antimicrobial activity against Staphylococcus aureus (ATCC 6538). In addition, these compounds displayed inhibitory effects on the growth of two human tumor cell lines, HeLa and HCT116.

Among fungi, the basic life strategies are saprophytism, parasitism, and predation. Fungi in Orbiliaceae (Ascomycota) prey on animals by means of specialized trapping structures. Five types of trapping devices are recognized, but their evolutionary origins and divergence are not well understood. Based on comprehensive phylogenetic analysis of nucleotide sequences of three protein-coding genes (RNA polymerase II subunit gene, rpb2; elongation factor 1-alpha gene, ef1-alpha; and ss tubulin gene, bt) and ribosomal DNA in the internal transcribed spacer region, we have demonstrated that the initial trapping structure evolved along two lineages yielding two distinct trapping mechanisms: one developed into constricting rings and the other developed into adhesive traps. Among adhesive trapping devices, the adhesive network separated from the others early and evolved at a steady and gentle speed. The adhesive knob evolved through stalk elongation, with a final development of nonconstricting rings. Our data suggest that the derived adhesive traps are at a highly differentiated stage. The development of trapping devices is felicitous proof of adaptive evolution.

Serine protease plays an important role in fungal infection to invertebrate hosts. An extracellular protease (Hnsp) was detected in liquid culture of Hirsutella rhossiliensis OWVT-1 with nematodes (Panagrellus redivivus) as the unique nitrogen source and purified to homogeneity by ammonium sulphate precipitation, anion exchange chromatography and gel filtration. Its molecular mass was about 32 kDa, and the optimal reaction pH value and temperature were pH 7 and 40 degrees C, respectively. The Hnsp activity was stable at pH 6-8 and decreased radically at 50 degrees C for 10 min. Hnsp was highly sensitive to inhibitor of PMSF and well decomposed the substrate N-succinyl-Ala-Ala-Pro-Phe-p-nitroanilide, suggesting that it belonged to the chymotrypsin/subtilisin of serine proteases. The N-terminal amino acid sequence of Hnsp was SVTDQQGADCGLARISHRE, which showed high homology with other serine proteases from nematophagous fungi. Ability to kill nematode and degrade its cuticle in vitro indicated that Hnsp could be involved in the infection of nematode.

Decaspirones F-H (1-3), three new spirobisnaphthalene derivatives, and one new non-spirobisnaphthalene, decaspirone I (4), have been isolated from cultures of an isolate of the saprophytic fungus Helicoma viridis. The structures of these compounds were determined mainly by analysis of their NMR spectroscopic data. The absolute configuration of decaspirone F (1) was established by X-ray crystallographic analysis of its mono-bromobenzoate derivative. In standard disk assays, compounds 1-4 showed modest activity against Pseudomonas aeruginosa (multiple antibiotic-resistant strain, ATCC 27853), and decaspirone G (2) also displayed activity against Lactococcus lacti (PCM 2379).

Thirty-three carbon sources were evaluated for their effects on spore germination, hyphal growth and sporulation of 11 fungal biocontrol agents, i.e. the nematophagous fungi Paecilomyces lilacinus, Pochonia chlamydosporia, Hirsutella rhossiliensis, H. minnesotensis and Arkansas Fungus 18, the entomopathogenic fungi Lecanicillium lecanii, Beauveria bassiana and Metarhizium anisopliae, and the mycoparasitic fungus Trichoderma viride. Variations in carbon requirements were found among the fungal species or strains tested. All strains studied except for T. viride grew on most carbon sources, although B. bassiana had more fastidious requirements for spore germination. Monosaccharides and disaccharides were suitable for fungal growth. For most isolates, D-glucose, D-mannose, sucrose and trehalose were superior to pectin and soluble starch among the polysaccharides and lactic acid among the organic acids. Both ethanol and methanol could accelerate growth of most isolates but not biomass. D-mannose, D-fructose and D-xylose were excellent carbon sources for sporulation, while D-glucose, sucrose, cellobiose, trehalose, chitin, dextrin, gelatin and lactic acid were better for some isolates. Neither sorbic acid nor linoleic acid could be utilized as a single carbon source. These findings provided a better understanding of the nutritional requirements of different fungal biocontrol agents that can benefit the mass production process.

Twenty carbohydrates (C), 18 nitrogen compounds (N), and 9 vitamins were examined for their effects on the growth and conidiation of the nematode-egg-parasitic fungi Arkansas Fungus 18 (ARF18, isolate 908) and Pochonia chlamydospora var. chlamydospora in solid and liquid cultures. Glycogen was the best, and inulin, D-(+)-galactose, and soluble starch were good C sources for the growth of ARF18 in both solid and liquid cultures. ARF18 could not utilize alpha-cellulose in liquid; alpha-lactose and D-mannitol in solid; and D-(+)-xylose, L-(-)-sorbose, and D-(-)-ribose in both solid and liquid cultures. Casein was the best N source for ARF18 growth in both solid and liquid cultures and L-aspartic acid, DL-glutamic acid, peptone, and L-histidine were good in solid culture. ARF18 could not utilize L-cystine and L-tyrosine in solid culture, and L-cystine, DL-methionine, peptone, L-proline, and ammonium nitrate in liquid culture. Supplement of vitamins appeared to be unnecessary for ARF18 to grow. For P. chlamydospora var. chlamydospora growth, all test C sources, except L-(-)-sorbose, alpha-cellulose, citric acid, and D-(+)-glucose, were good in both solid and liquid cultures. Most N compounds were good for P. chlamydospora var. chlamydospora growth with casein and peptone the best. Vitamins had limited effect on P. chlamydospora var. chlamydospora growth. P. chlamydospora var. chlamydospora conidiation was well supported by D-(-)-ribose, D-(-)-fructose, melibiose, and D-(+)-galactose as C sources and by L-aspartic acid, DL-glutamic acid, and L-arginine as N sources. Excluding myo-inositol from the medium containing all other test vitamins increased P. chlamydospora var. chlamydospora conidiation, while excluding pyridoxine appeared to reduce its conidiation.
