Global urbanization leads to the loss of periurban farming land and increases dependency on distant agriculture systems. This provokes greenhouse gas emissions associated with transportation and storage while disconnecting nutrient cycles, as urban organic waste is not recycled into the agricultural system. Urban food production based on composted local biomass could reduce these problems, but currently used hybrid crops rely strongly on inorganic fertilizers. On the contrary, open-pollinated varieties were bred for productivity under organic fertilization, such as compost. Hypothesising that open-pollinated varieties retain high nutritional value under low nutrient conditions, a commercial hybrid and a local open-pollinated variety of maize were cultivated in non-fertilized soil and under two compost applications: Municipal compost as high nutrient input or locally produced green waste compost and municipal compost mix, as medium nutrient input. Unfertilized plots exhibited low grain production (1.9 t/ha), but yields under green waste compost/municipal compost (6.1 t/ha) and municipal compost (7.8 t/ha) treatments were comparable to observations from maize under inorganic fertilization. Contrary to the commercial variety, the open-pollinated variety exhibited higher grain micronutrient concentrations, e.g. 220% higher zinc concentrations and lower accumulation of heavy metals, e.g. 74% lower nickel concentrations. This variety-related effect was found in all treatments and was independent of soil micronutrient concentrations. In conclusion, both compost mixes were effective in increasing grain yield in both maize varieties. However, the open-pollinated variety produced grain with higher nutritional values in soil and all treatments, indicating it is potentially better suited for compost-based sustainable urban agriculture.

SummaryAlthough poly(ADP-ribose) polymerase (PARP) inhibitors are active in homologous recombination (HR)-deficient cancers, their utility is limited by acquired resistance after restoration of HR. Here, we report that dinaciclib, an inhibitor of cyclin-dependent kinases (CDKs) 1, 2, 5, and 9, additionally has potent activity against CDK12, a transcriptional regulator of HR. In BRCA-mutated triple-negative breast cancer (TNBC) cells and patient-derived xenografts (PDXs), dinaciclib ablates restored HR and reverses PARP inhibitor resistance. Additionally, we show that de novo resistance to PARP inhibition in BRCA1-mutated cell lines and a PDX derived from a PARP-inhibitor-naive BRCA1 carrier is mediated by residual HR and is reversed by CDK12 inhibition. Finally, dinaciclib augments the degree of response in a PARP-inhibitor-sensitive model, converting tumor growth inhibition to durable regression. These results highlight the significance of HR disruption as a therapeutic strategy and support the broad use of combined CDK12 and PARP inhibition in TNBC.

Ammonia (NH3) empirical critical levels for Europe were re-evaluated in 2009, based mainly on the ecological responses of lichen communities without acknowledging the physiological differences between oligotrophic and nitrophytic species. Here, we compare a nitrogen sensitive lichen (Evernia prunastri) with a nitrogen tolerant one (Xanthoria parietina), focussing on their physiological response (Fv/Fm) to short-term NH3 exposure and their frequency of occurrence along an NH3 field gradient. Both frequency and Fv/Fm of E. prunastri decreased abruptly above 3 μg m(-3) NH3 suggesting direct adverse effects of NH3 on its photosynthetic performance. By contrast, X. parietina increased its frequency with NH3, despite showing decreased capacity of photosystem II above 50 μg m(-3) NH3, suggesting that the ecological success of X. parietina at ammonia-rich sites might be related to indirect effects of increased nitrogen (NH3) availability. These results highlight the need to establish NH3 critical levels based on oligotrophic lichen species.

Adiponectin (adipoQ) gene variants have been associated with type 2 diabetes mellitus and insulin resistance. Our aim was to examine whether the presence of several polymorphisms at the adipoQ gene locus (-11391 G > A, -11377 C > G, 45 T > G, and 276 G > T) influences the insulin sensitivity to dietary fat. Healthy volunteers (30 men and 29 women) consumed 3 diets for 4 wk each: an initial period during which all subjects consumed a SFA-rich diet (38% total fat, 20% SFA), followed by a carbohydrate-rich diet (CHO) (30% total fat, 55% carbohydrate) or a monounsaturated fatty acid (MUFA)-rich diet (38% total fat, 22% MUFA) following a randomized, crossover design. After participants consumed each diet, we tested peripheral insulin sensitivity with the insulin suppression test and measured plasma adiponectin concentrations. C/C homozygous men for the -11377 C > G single nucleotide polymorphism (SNP) had a significantly greater decrease in the steady-state plasma glucose concentrations when changing from the SFA-rich (8.95 +/- 0.6 mmol/L) to the MUFA-rich (6.04 +/- 0.31 mmol/L) and CHO-rich (6.35 +/- 0.38 mmol/L) diets than did those carrying the minor G allele (SFA, 6.65 +/- 0.4 mmol/L; MUFA, 6.45 +/- 0.4 mmol/L; CHO, 5.83 +/- 0.3 mmol/L) (P sex x gene x diet interaction = 0.016). These differences did not occur in female participants. Furthermore, C/C men had lower plasma adiponectin concentrations than did C/C women (P sex x gene interaction = 0.015), independently of the dietary fat consumed. None of the variables examined were significantly associated with -11426 A > G, 45T > G, or 276 G > T SNP. In conclusion, C/C homozygous men for the -11377 C > G SNP at adipoQ gene were significantly less insulin resistant after consumption of the MUFA- and CHO-rich diets compared with the SFA-rich diet. This information should help in the identification of vulnerable populations or persons who will benefit from more personalized and mechanism-based dietary recommendations.

Hydrogen peroxide (H2O2) functions as an important signaling molecule in plants during biotic interactions. However, the extent to which H2O2 accumulates during these interactions and its implications in the development of disease symptoms is unclear. In this work, we provide a step-by-step optimized protocol for in situ quantification of relative H2O2 concentrations in wheat leaves infected with the pathogenic bacterium Pseudomonas syringae pv. atrofaciens (Psa), either alone or in the presence of the beneficial bacterium Herbaspirillum seropedicae (RAM10). This protocol involved the use of 3-3'diaminobenzidine (DAB) staining method combined with image processing to conduct deconvolution and downstream analysis of the digitalized leaf image. The application of a linear regression model allowed to relate the intensity of the pixels resulting from DAB staining with a given concentration of H2O2. Decreasing H2O2 accumulation patterns were detected at increasing distances from the site of pathogen infection, and H2O2 concentrations were different depending on the bacterial combinations tested. Notably, Psa-challenged plants in presence of RAM10 accumulated less H2O2 in the leaf and showed reduced necrotic symptoms, pointing to a potential role of RAM10 in reducing pathogen-triggered H2O2 levels in young wheat plants.

13 páginas.-- 4 figuras.-- 85 referencias, Many publications highlight the importance of nitric oxide (NO) in plant–bacteria interactions, either in the promotion of health and plant growth or in pathogenesis. However, the role of NO in the signaling between bacteria and plants and in the fate of their interaction, as well as the reconstruction of their interactive evolution, remains largely unknown. Despite the complexity of the evolution of life on Earth, we explore the hypothesis that denitrification and aerobic respiration were responsible for local NO accumulation, which triggered primordial antagonistic biotic interactions, namely the first phytopathogenic interactions. N-oxides, including NO, could globally accumulate via lightning synthesis in the early anoxic ocean and constitute pools for the evolution of denitrification, considered an early step of the biological nitrogen cycle. Interestingly, a common evolution may be proposed for components of denitrification and aerobic respiration pathways, namely for NO and oxygen reductases, a theory compatible with the presence of low amounts of oxygen before the great oxygenation event (GOE), which was generated by Cyanobacteria. During GOE, the increase in oxygen caused the decrease of Earth’s temperature and the consequent increase of oxygen dissolution and availability, making aerobic respiration an increasingly dominant trait of the expanding mesophilic lifestyle. Horizontal gene transfer was certainly important in the joint expansion of mesophily and aerobic respiration. First denitrification steps lead to NO formation through nitrite reductase activity, and NO may further accumulate when oxygen binds NO reductase, resulting in denitrification blockage. The consequent transient NO surplus in an oxic niche could have been a key factor for a successful outcome of an early denitrifying prokaryote able to scavenge oxygen by NO/oxygen reductase or by an independent heterotrophic aerobic respiration pathway. In fact, NO surplus could result in toxicity causing “the first disease” in oxygen-producing Cyanobacteria. We inspected in bacteria the presence of sequences similar to the NO-producing nitrite reductase nirS gene of Thermus thermophilus, an extreme thermophilic aerobe of the Thermus/Deinococcus group, which constitutes an ancient lineage related to Cyanobacteria. Summary Nitrate reductase is considered to be a limiting factor for growth, development and protein production in plants. This enzyme is influenced by external conditions. The purpose of this study was to investigate the effect of different NO 3 -/NH 4 + ratios and salinity on NRA (nitrate reductase activity) in shoots and roots of wheat ( Triticum aestivum L. cv. Betres) seedlings. Plants were grown in a growth chamber under controlled conditions. Nutrient solution contained 4 mM N, applied as NO 3 - and NH 4 + in different ratios, and saline treatments consisted in two levels of NaCl: 1 mM and 60 mM. Wheat seedlings had most of their NRA in shoots. When NH 4 + was present in the nutrient solution NRA was almost completely inhibited in roots and decreased in shoots. Inhibition of NRA by NH4+ seems to be a direct effect on synthesis and activity of NR rather than through inhibition of NO 3 - uptake and thus by lack of substrate. Salinity significantly decreased NRA in shoots, this effect being influenced by NO 3 - in the and by NO 3 -/Cl - ratio in shoots. The effect on RNA may be due to the alteration of NO 3 - uptake, to an excessive CI - shoot content which may affect NO 3 - transport from vacuoles to cytoplasm and to the alteration of NO 3 - flux from root to shoot.

Key enzymes of the urea cycle and 15N-labeling patterns of arginine (Arg) were measured to elucidate the involvement of Arg in nitrogen translocation by arbuscular mycorrhizal (AM) fungi. Mycorrhiza was established between transformed carrot (Daucus carota) roots and Glomus intraradices in two-compartment petri dishes and three ammonium levels were supplied to the compartment containing the extraradical mycelium (ERM), but no roots. Time courses of specific enzyme activity were obtained for glutamine synthetase, argininosuccinate synthetase, arginase, and urease in the ERM and AM roots. 15NH4 + was used to follow the dynamics of nitrogen incorporation into and turnover of Arg. Both the absence of external nitrogen and the presence of l-norvaline, an inhibitor of Arg synthesis, prevented the synthesis of Arg in the ERM and resulted in decreased activity of arginase and urease in the AM root. The catabolic activity of the urea cycle in the roots therefore depends on Arg translocation from the ERM. 15N labeling of Arg in the ERM was very fast and analysis of its time course and isotopomer pattern allowed estimation of the translocation rate of Arg along the mycelium as 0.13 μg Arg mg−1 fresh weight h−1. The results highlight the synchronization of the spatially separated reactions involved in the anabolic and catabolic arms of the urea cycle. This synchronization is a prerequisite for Arg to be a key component in nitrogen translocation in the AM mycelium.

The chemistry of conjugated fatty acids, specifically octadecadienoic acids (18:2; commonly referred to as conjugated linoleic acid, or CLA), has provided many challenges to lipid analysts because of their unique physical properties and the many possible positional and geometric isomers. After the acid-labile properties of CLAs during analytic procedures were overcome, it became evident that natural products, specifically dairy fats, contain one dominant (c9,t11-CLA), 3 intermediate (t7,c9-, t9,c11-, and t11,c13-CLA), and up to 20 more minor CLA isomers. The best analytic techniques to date include a combination of gas chromatography that uses 100-m highly polar capillary columns, silver ion-HPLC, and a combination of silver ion-thin-layer chromatography and gas chromatography to analyze the CLA and trans 18:1 isomers, because some of them serve as precursors of CLA in biological systems. These analytic techniques have assisted commercial suppliers to prepare pure CLA isomers and have permitted the evaluation of individual CLA isomers for their nutritional and biological activity in animal and human systems. It is increasingly evident that different CLA isomers have distinctly different physiologic and biochemical properties. These techniques are essential to evaluate dairy fats for their CLA content, to design experimental diets to increase the amount of CLA in dairy fats, and to determine the CLA profile in these CLA-enriched dairy fats. These improved techniques are used to evaluate the CLA profile in pork products from pigs fed different commercial CLA mixtures.

A survey of some notifiable fish diseases (enteric redmouth disease, furunculosis, bacterial kidney disease, proliferative kidney disease and whirling disease) was made at all trout culture sites in Portugal. All the analyses were negative for 95% confidence assuming an incidence of 10%. This emphasizes the need for fish import regulations in Portugal in order to maintain the situation.

Data on the seasonal occurrence and maturation of Rhabdochona gnedini Skrjabin, 1946 in its definitive host, the barbel Barbus bocogei, from the Sousa River, in northern Portugal, are presented. Monthly samples were taken from November, 1998 to October, 1999. A total of 178 fish specimens were inspected. R. gnedini occutred with high prevalences during whole year, attaining the highest values of intensity in summer(overall prevalence and mean intensity 85 % and 31 , respectively). A clear annual cycle of maturation was observed; females with mature eggs occurred in spring and summer. Unidentified Rhabdochona larvae were recorded from the trichopteran larvae Hydropsyche sp. (prevalence and intensity 29 % and one to 34, respectively). These larvae are presumptive intermediate host for R. gnedini.

The response of carob (Ceratonia siliqua L.) seedlings grown at different root zone temperatures affected by nitrate and ammonium nutrition was studied. When root temperatures ranged from 10 to 35°C, ammonium‐fed plants were significantly larger than nitrate‐fed plants. Ammonium‐fed plants displayed toxicity symptoms and were much smaller at 40°C root temperature in comparison with the nitrate‐fed plants grown at the same root temperature. Root/shoot ratio slightly increase with root temperature in ammonium‐ and nitrate‐fed plants in a similar way, and shoot demand per root unit decreased with root temperature between 15 and 25°C. There was a general increase in net photosynthesis with root temperature, though nitrate‐fed plants were more sensitive to low and ammonium‐fed plants to high temperatures. Increasing the root temperature of ammonium fed plants from 10 to 40°C leads to a 30% increase in the amount of photosynthates sent to the roots. The presence of ammonium resulted in the distribution of newly fixed carbon away from carbohydrates and into nitrogen compounds. Potassium, calcium, and nitrogen content of the plants also increased with increasing root temperature.

A population of the gill parasite Hatschekia pagellibogneravei (Hesse, 1878) was studied on one of its sparid fish hosts, the blackspot seabream, Pagellus bogaraveo (Brunnich), off the coast of Madeira Island, Portugal, northeast Atlantic. Very high infection levels of this copepod were detected, with no significant seasonal differences. Abundance was negatively correlated with fish size. There were significant differences in the distribution of this copepod among the gill arches of the host, which seem to be best explained by differences in water flow within the gill habitat.

Carob seedlings were grown at different proportions of nitrate NO3) and ammonium (NH4) supplied at a concentration of 3.5 mM nitrogen (N). The effect of five different combinations of NH4- and NO3-N on growth rates, N and carbon (C) content of the plant and dry matter distribution were studied. The presence of NH4 in the nutrient solution enhanced plant growth rate, although net photosynthesis rates per unit area were lower than in NO3-fed plants. Nitrate –fed plants exbibited the highest rates of carbon dioxide (CO2) assimilation, water use, and N-use efficiency. Nevertheless, plants growing only with NO3 were more fragile and susceptible to fungus infections. The role of cotyledons on the regulation of the supply of photosynthates from leaves to roots in the different combinations of NO3- and NH4-N is described. Results suggest that the growth rates of carob may be improved using suitable combinations of NO3- and NH4-N. Equal concentrations of these two N sources allows good development and adequate allocation of low C and N to different developing sinks of the carob seedlings.

A severe case of ceroid deposition was observed in the kidney of a small number of feral Oncorhynchus mykiss from Alicura impoundment, Argentinian Patagonia. The pigments were observed intracellularly in the kidney tubules and in the parenchyma cells, varying from a large number of small droplets to one big droplet almost filling the cell and pushing the cytoplasm to one side of the cell. An extensive study of farmed fish, as well as wild specimens from the same impoudment, did not show any ceroid deposition. Further, no abnormal mortality rates were observed in the farms. The mechanism that caused this condition in just a small number of fish is not known

Despite their well-known benefits in electricity production, dams are also responsible for some adverse environmental impacts affecting particularly the wellbeing of residents of the local communities. These environmental damages have not been included in the cost-benefit analysis of hydropower developments mainly because of the difficulty to determine their value. The prime objective of this paper is to measure the economic values of several environmental impacts due to the dams’ activity in Portugal, using a discrete choice experiments approach. With the results of this research paper, we expect to contribute to a more efficient and thorough cost-benefit analysis within the complex process of deciding the optimal location of future dams to be built not only in Portugal, but elsewhere. The addition of this stage to the decision-making process allows the integration of economic, social and environmental dimensions, promoting a richer and more informed decision process, Financial support from PTDC/EGE-ECO/122402/2010 is greatly appreciated.

Crop management systems influence plant productivity and nutrient use efficiency, as well as plant growth-promoting rhizobacteria (PGPR), which are known to influence the growth of plants via phytohormone production, phosphate solubilization, nitrogen (N) fixation and antimicrobial activity. The objective of this study was to compare the influence of two crop management system on microbial PGPR features. PGPR isolated from the rhizospheres of Carica papaya L. grown under two distinct management systems (conventional and organic) were identified and characterized. The 12 strains most efficient in solubilizing inorganic phosphate belonged to the genera Burkholderia, Klebsiella, and Leclercia. N fixation was observed in the strains B. vietnamiensis from the conventional farming system and B. vietnamiensis, B. cepacia and Leclercia sp. from the organic farming system. The B. vietnamiensis, B. cepacia, Klebsiella sp. and Klebsiella sp. isolates showed antifungal activity, while Leclercia sp. did not. The strains B. vietnamiensis and Enterobcter sp. (isolated from the conventional farming system) and Klebsiella sp. (isolated from the organic farming system) were efficient at solubilizing phosphate, producing phytohormones and siderophores, and inhibiting the mycelial growth of various phytopathogenic fungi (Botrytis cinerea, Pestalotia sp., Alternaria sp., Phoma sp., Fusarium culmorum, Geotrichum candidum). Physiological differences between the isolates from the two crop management regimes were distinguishable after 10 years of distinct management.
