No. of bitstreams: 1 2019 - Ariane da Silva Favareto.pdf: 3138607 bytes, checksum: 608cb285340e09c37c121e1f00b62a02 (MD5) Previous issue date: 2019-10-18 CAPES FAPERJ This thesis aims to analyze the way and in which dimensions of social life women are affected by the dynamics of contemporary rurality and how this is reflected in the social configurations of gender and, consequently, in the position of women in society. it was chosen, theoretically and methodologically, inspired by Norbert Elias's work, the use of the notion configurations instead of gender relations, in order to encompass greater complexity of social life, ranging from the initial socialization processes to interactions more recent, without forget exogenous factors that affect social constructions. The category way of life is used to understand rurality, which, translated into identities and representations, guides the strategies and use of opportunities placed in the exercise of past activities, linked to agriculture and other recently developed that take advantage of the use and appreciation of natural resources. Such conditions were observed in the municipality of Cunha, located in the Vale do Para?ba, state of S?o Paulo, universe of this research. As a method, we used the analysis of the life trajectory of women working in the development of activities related to tourism, agroecology and livestock, from semi-structured interviews; as well as participant observation. The search shows that in the dynamics of contemporary rurality are open new possibilities for insertion for women. The use of these opportunities is not unequivocal, if expressed heterogeneously, given their own life trajectories and the interdependencies that determine the use or embarrassment of opportunities that arise and will focus on their social position, especially with regard to work and income, education and training, and the use of time, dimensions of social life that affect women's capacities. Esta tese tem por objetivo analisar a maneira e em quais dimens?es da vida social as mulheres s?o afetadas pelas din?micas da ruralidade contempor?nea e como isso se reflete nas configura??es sociais de g?nero e, consequentemente, na posi??o da mulher na sociedade. Optou-se, de forma te?rica e metodol?gica, com inspira??o na obra de Norbert Elias, pelo uso da no??o configura??es ao inv?s de rela??es de g?nero, de modo a abarcar maior complexidade da vida social, remetendo desde os processos de socializa??o iniciais at? as intera??es mais recentes, sem deixar de lado fatores ex?genos que incidem nas constru??es sociais. Utiliza-se a categoria modo de vida para compreender a ruralidade que, traduzida em identidades e representa??es, orienta as estrat?gias e aproveitamento de oportunidades colocadas no exerc?cio de atividades pret?ritas, ligadas ? agropecu?ria e outras desenvolvidas mais recentemente que se aproveitam da utiliza??o e valoriza??o dos recursos naturais. Tais condi??es foram observadas no munic?pio de Cunha, localizado no Vale do Para?ba, estado de S?o Paulo, universo desta pesquisa. Como instrumento, utilizou-se a an?lise da trajet?ria de vida de mulheres que trabalham no desenvolvimento de atividades ligadas ao turismo, ? agroecologia e ? pecu?ria, a partir de entrevistas semiestruturadas; bem como da observa??o participante. A tese que se busca demonstrar ? que nas din?micas da ruralidade contempor?nea s?o abertas novas possibilidades de inser??o para as mulheres. O aproveitamento destas oportunidades n?o ? un?voco, se expressa de forma heterog?nea, dadas suas pr?prias trajet?rias de vida e as interdepend?ncias realizadas, que determinam o aproveitamento ou o constrangimento das oportunidades surgidas e v?o incidir na sua posi??o social, especialmente, no que se refere a trabalho e renda, escolaridade e capacita??o, e o uso do tempo, dimens?es da vida social que afetam as capacidades femininas.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES Este estudo teve como objetivo analisar as particularidades dos trabalhos produtivo e reprodutivo não remunerados das mulheres no meio rural brasileiro. A totalidade de trabalhos que estas mulheres realizam de forma gratuita, tanto na esfera da produção quanto da reprodução/doméstica. Os pressupostos teóricos se referenciam na tradição marxista em diálogo com as elaborações no campo do feminismo materialista e da economia feminista. Refletiu-se sobre a consubstancialidade das relações sociais de sexo, raça e classe, a divisão sexual do trabalho, o trabalho doméstico e de cuidados como base de sustentação da subordinação e exploração das mulheres na sociedade capitalista-patriarcal-racista. A base empírica do nosso estudo foram os microdados da Pesquisa Nacional por Amostra de Domicílios – PNAD Contínua, realizada pelo Instituto Brasileiro de Geografia e Estatística – IBGE referente ao ano de 2018. Caracterizaram-se as diferentes formas de trabalhos realizados pelas mulheres rurais com ênfase nas variáveis cor ou raça. O resultado evidenciou que homens e mulheres participam do trabalho reprodutivo/doméstico, mas as mulheres continuam como responsáveis majoritárias por esse tipo de trabalho. Características como regularidade e eventualidade diferenciam o trabalho reprodutivo/doméstico de mulheres e homens, respectivamente. A participação dos homens obedece a uma hierarquia nos tipos e lugar em que as tarefas são realizadas. Portanto, realizam tarefas mais do espaço público e mantém distanciamento daquelas consideradas ―femininas‖. O trabalho das mulheres não reconhece fronteiras geográficas e é realizado seja em espaços públicos ou privados e na casa de parentes. Assim, o estudo mostrou que os homens, detém certa liberdade, convertida em privilégio que concede a eles o direito de escolher que tipo de atividade deseja realizar. No trabalho para o próprio consumo, revelou-se dinâmica uma pouco diferente. Nele as mulheres avançam mais na realização das atividades consideradas ―masculinas‖, mas no geral, permanece a desigualdade, na medida em que as horas que elas dedicam ao conjunto dos trabalhos não remunerados (reprodutivo e produtivo) é superior a dos homens. A maioria dos que realizam trabalhos não remunerados no meio rural são negros (pretos e pardos) com evidente divisão sexual e racial do trabalho. A condição das mulheres rurais em trabalhos duplamente não remunerados nos desafiam a aprofundar a reflexão sobre o processo de subordinação e exploração que estas vivenciam em seus cotidianos. This study aimed to analyze the particularities of the unpaid productive and reproductive work of women in rural Brazil. The totality of work that these women do free of charge, both in the sphere of production and reproduction / domestic. Theoretical assumptions are referenced in the Marxist tradition in dialogue with the elaborations in the field of materialist feminism and feminist economics. It reflected on the consubstantiality of social relations of sex, race and class, the sexual division of labor, domestic and care work as a basis for sustaining the subordination and exploitation of women in the capitalist-patriarchal-racist society. The empirical basis of our study was the microdata from the National Household Sample Survey - Continuous PNAD, conducted by the Brazilian Institute of Geography and Statistics - IBGE in 2018. We characterized the different forms of work performed by rural women with an emphasis on color or race variables. The result evidenced that men and women participate in reproductive / domestic work, but women remain the majority responsible for this type of work. Respectively, characteristics such as regularity and eventuality differentiate the reproductive / domestic work of women and men, respectively. The participation of men follows a hierarchy in the types and places where tasks are performed. Therefore, they perform more tasks in the public space and keep their distance from those considered ―female‖. Women's work does not recognize geographical boundaries and is executed either in public or private spaces and in the homes of relatives. Thus, the study showed that men, have a certain freedom, converted into a privilege that gives them the right to choose what type of activity they want to do. In the work for own consumption, dynamics were a little different. In it, women make more progress in activities considered ―masculine‖, but in general, inequality remains, as the hours they dedicate to all unpaid jobs are higher than that of men. Most of those who perform unpaid work in rural areas are black (black and brown) with an evident sexual and racial division of labor. The condition of rural women in doubly unpaid jobs challenges us to deepen the reflection on the process of subordination and exploitation that they experience in their daily lives.

A luta por terra e território que vem sendo travada, desde 2011, na porção norte-rio-grandense da Chapada do Apodi evidencia a disputa entre dois modelos de agricultura. De um lado, estão o agro e hidronegócio que, fundamentados na utilização de grandes áreas de terra e elevada quantidade de água, associada ao uso de agrotóxicos na produção, priorizam o lucro e ameaçam a vida no campo. Do outro, a agricultura familiar e camponesa que, a partir da produção, na maioria dos casos de base agroecológica, em pequenas propriedades, prioriza a vida e a biodiversidade no campo. Essa disputa mobiliza diversos movimentos e organizações locais, nacionais e internacionais contra as desapropriações e a favor das famílias ameaçadas de expulsão de suas terras. Constitui-se, dessa maneira, um movimento de luta e resistência na (e pela) Chapada do Apodi. Desse processo, destacamos, nesta dissertação, as ações de leitura, escrita e oralidade, tomadas como letramentos de resistência, desenvolvidas por trabalhadoras rurais de Apodi. Inserida no campo da Linguística Aplicada (MOITA-LOPES, 2006; PENNYCOOK, 2006), esta pesquisa assume uma abordagem qualitativa e interpretativista, amparada nos estudos de letramento de vertente etnográfica (KLEIMAN, 1995; TINOCO, 2008; SOUZA, 2009; SITO, 2010), na concepção dialógica da linguagem (BAKHTIN/VOLOSHINOV, 2006 [1929]) e no campo da educação popular e pedagogia crítica (FREIRE, 2014 [1974]; GIROUX, 1986). Seu objetivo geral é analisar os impactos sociais que resultam dos letramentos de resistência protagonizados pela Comissão de Mulheres do Sindicato dos Trabalhadores e das Trabalhadoras Rurais de Apodi (STTR) no processo de luta por terra e território na Chapada do Apodi norte-rio-grandense. Os objetivos específicos são (i) compreender o processo de construção de eventos de letramento de resistência na luta por terra e território na Chapada do Apodi e, nesses eventos, (ii) examinar estratégias e táticas de linguagem construídas por trabalhadoras rurais de Apodi para fortalecer suas práticas de agricultura familiar e, ao mesmo tempo, contrapor-se ao discurso das grandes empresas do agro e hidronegócio. Os dados foram gerados em seis eventos: duas oficinas com trabalhadoras rurais de Apodi; quatro sessões de entrevistas com coordenadoras e ex-coordenadoras da Comissão de Mulheres do STTR de Apodi. Os resultados apontam que práticas de letramento de resistência, mobilizadas em âmbito local e aliadas a outras práticas sociais de organização popular, podem transformar um problema local em uma luta coletiva internacional e contribuir para o fortalecimento da autonomia feminista de trabalhadoras rurais que, independentemente da escolaridade, atuam como agentes de letramento na luta por terra e território no Rio Grande do Norte e além-fronteiras. The striving for land and territory that has been occurring since 2011 at Chapada do Apodi evidences the dispute between two models of agriculture. On the one hand, agricultural and water businesses are based on the usage of large areas of land and large amount of water. Such models are associated with the employment of pesticides in agricultural production, prioritizing profit and threatening country life. On the other hand, most agro-ecological cases evidence that family and peasant agricultures prioritize life and field biodiversity based on the production conducted in small lands. The dispute between such models of agriculture mobilizes several movements and local, national and international organizations against expropriations and in favor of the families threatened with eviction of their own lands, thereby constituting striving and resistance movements at and for Chapada do Apodi. From this process, this dissertation emphasizes reading, writing and orality actions, which are denominated literacies of resistance and which were performed by farm workers of Apodi city. Inserted into the field of Applied Linguistics (MOITA-LOPES, 2006; PENNYCOOK, 2006), this research stands for a qualitative and interpretative approach, being backed by ethnographic literacy studies (KLEIMAN, 1995; TINOCO, 2008; SOUZA, 2009; SITO, 2010), as well as dialogical conception of language (BAKHTIN/VOLOSHINOV, 2006 [1929]), popular education and critical pedagogy (FREIRE, 2014 [1974]; GIROUX, 1986). The general objective of this research is to analyze the social impacts resulting from literacies of resistance featured by the Women’s Committee of the Female and Male Rural Workers Labor Union (STTR) of Apodi concerning the striving for land and territory at Chapada do Apodi. The specific objectives of this research are: (i) to comprehend the construction process of literacies of resistance concerning the striving for land and territory at Chapada do Apodi and in these events, (ii) to examine strategies and language policies developed by rural workers of Apodi to strengthen their practices of family agriculture and simultaneously to contradict the discourse of large companies that deal with agricultural and water business. The collection of data was conducted through six events: two workshops with female rural workers of Apodi and four interview sessions with coordinators and former coordinators of the Women’s Committee of STTR of Apodi. In conclusion, the results evidenced that practices of literacies of resistance at local level and allied with other social practices popularly organized may turn a local problem into an international collective striving and also contribute to strengthening the autonomy of the female rural workers that, regardless their education level, act as literacy agents concerning the striving for land and territory in the state of Rio Grande do Norte and beyond.

A pesquisa se dedica a aprender com as companheiras trabalhadoras rurais sobre o exercício feminista de narrar suas realidades e, para isto, se volta para a análise da produção de narrativas das mulheres do MMTR-NE, buscando, simultaneamente, compreender e visibilizar seu processo de autodefinição através dos “fuxicos” e das práticas de sistematização, da produção de materiais escritos e audiovisuais, além da centralidade das histórias próprias no processo de autoeducação e formação das bases no Movimento. A partir de uma perspectiva negra e decolonial, evidencia a potência epistêmica das narrativas contra hegemônicas produzidas pelas mulheres do MMTR-NE, e como estas utilizam suas histórias para avançar em autonomia e manifestar a interrupção, a denúncia e a reparação da coisificação a que foram submetidas as categorias subalternizadas: o deslocamento da condição desumana de “objeto” para a dignidade de se afirmar sujeito e cobrar o reconhecimento disso, em ousadia e desobediência política. The Women's Movement of Rural Workers from the Northeast (MMTR-NE) is a rural feminist, anti-racist and agroecological movement led for more than thirty years by rural women workers from the nine states of the Northeast Region of Brazil. The research aims to learn from fellow rural workers about the feminist practice of narrating their realities and, therefore, it focuses on analyzing narrative productions by MMTR-NE women; it seeks, simultaneously, to understand and make visible their process of self-definition through the making of “fuxicos,” and through practices of systematization, production of written and audiovisual materials, as well as the centrality of their own stories in self-education and political formation within the Movement’s bases. From a black and decolonial perspective, the project highlights the epistemic power of counter-hegemonic narratives produced by the women of MMTR-NE; how they use their stories to gain autonomy and manifest the interruption, denunciation and reparation of the objectification to which subaltern categories had been subjected to: displacing the inhuman condition of “object” to the dignity of affirming themselves as subjects and demanding such recognition with boldness and political disobedience.

Objective: To compare the prognostic value of dynapenia, as evaluated by handgrip, and body mass index (BMI) on length of stay (LOS), days of bed rest, and other hospitalization-related outcomes in a population of older adults admitted to 12 italian acute care divisions. Methods: Data on age, weight, BMI, comorbidities, ADL, physical activity level, muscle strength, were recorded at hospital admission. Background: Recently the Berlin Aging Study II (BASE-II) showed that polypharmacy is associated with clinically relevant sarcopenia among community-dwelling older persons. Here we report findings from the GLISTEN study about the association of polypharmacy with sarcopenia among older medical in-patients. Methods: The GLISTEN study investigated prevalence and clinical correlates of sarcopenia in older patients admitted to geriatric and internal medicine acute care wards of 12 Italian hospitals. Results: In this sample of older medical in-patients with high prevalence of sarcopenia (34.7%) and polypharmacy (70.2%) we did not observe a significant association of polypharmacy with sarcopenia. Conclusions: Present findings demonstrate that the association of polypharmacy with sarcopenia, observed in the BASE-II study, is not evident in the GLISTEN sample, being our patients significantly older, more multi-morbid, with high prevalence of sarcopenia and polypharmacy, suggesting that this association might vary according to the heterogeneous health, functional, and nutritional characteristics of older people.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) No período 2004 e 2016, as mulheres rurais conquistaram um conjunto de políticas públicas, entre estas, a Assistência Técnica e a Extensão Rural para Mulheres, o programa ATER Mulheres, ofertado pela Diretoria de Políticas para as Mulheres Rurais (DPMR) do Ministério do Desenvolvimento Agrário (MDA). Objetiva atender as mulheres agricultoras na área de produção, gestão e comercialização. Nesse sentido, a pesquisa teve como objetivo geral avaliar a efetividade do ATER Mulheres no Rio Grande do Norte, no período de 2004 a 2016. A tese traz o resultado de um percurso teórico prático obtido durante a investigação. A abordagem proposta concentra-se em três formulações teóricas: o feminismo como uma teoria crítica e movimento social, a agroecologia nos aspectos ecológicos e socioeconômicos e avaliação de políticas públicas. É a partir dos estudos da economia feminista que a pesquisa dialoga com a construção da agroecologia como ciência e prática social. O debate conceitual é feito em colóquio com a trajetória do feminismo como movimento social e com seu deslocamento teórico-prático no processo histórico. Os aportes teóricos trazem ainda referenciais para essa avaliação, capazes de demonstrar que as relações sociais de sexo, em uma abordagem de consubstancialidade, devem ser parte integrante da leitura da agroecologia e âncora para a percepção da efetividade do programa estudado. Como parte da metodologia, a pesquisa busca realizar uma avaliação de políticas públicas compreendendo que essas são constituídas por teorias e técnicas científicas. O estudo utilizou ferramentas de investigação que tornam mais precisos o processo de julgamento e para alcançar os objetivos, os critérios são estabelecidos de acordo com o campo de pesquisa estudado (WEISS, 1975). A avaliação da efetividade do programa baseou-se no conceito de Arretche (2009), que parte da percepção se a voda das pessoas que acessaram a política mudou ou não. A partir desses referenciais e dos dados empíricos, a pesquisa afirma que o ATER Mulheres executado no RN, desenvolvido nos territórios Açu-Mossoró, Sertão do Apodi, Seridó e Mato Grande, teve efetividade na vida das mulheres pela capacidade de conjugar os aportes do feminismo e da agroecologia na sua execução. In the time between 2004 and 2016, rural women have achieved a set of social programs, among them, Technical Assistance and Rural Extension for Women, the ATER Women program, offered by the Directorate of Policies for Rural Women, DPMR, from the Ministry of Agrarian Development / MDA. It aims to assist women farmers in the fields of production, management and marketing. With this purport, the research had as its general objective: to evaluate the effectiveness of ATER Women in Rio Grande do Norte from 2004 to 2016. The thesis is the result of a practical theoretical course obtained during the investigation. The proposed approach focuses on three theoretical formulations: feminism as a critical theory and social movement, agroecology in ecological and socioeconomic aspects and evaluation of government social programs. It is from the studies of feminist economics that the research dialogues with the construction of agroecology as a science and social practice. The conceptual debate is held with the trajectory of feminism as a social movement and with its theoretical-practical shift in the historical process. The theoretical contributions also provide references for this evaluation, capable of demonstrating that the social relations of gender, in an approach of consubstantiality, should be an integral part of the reading of agroecology and anchor for the perception of the effectiveness of the studied program. As part of the methodology, the research seeks to carry out an evaluation of social programs understanding that these are constituted by scientific theories and techniques. The study used research tools that make the evaluation process more precise and to achieve the objectives, the criteria are established according to the field of studied research. (WEISS, 1975). The evaluation of the effectiveness of the program was based on the concept of ARRETCHE, (2009), which recognizes how the perception of whether or not the life of people who accessed the program changed. Based on these references and the empirical data, the research states that the ATER Women, executed in RN, developed in the territories Açu / Mossoró, Sertão do Apodi, Seridó and Mato Grande, had effectiveness in women’s life by the ability to combine the contributions of feminism and of agroecology in its execution.

Este trabalho emergiu da necessidade de se voltar esforços para a superação do paradigma que considera que o desenvolvimento é unicamente ligado ao crescimento econômico alicerçado no uso intensivo de tecnologia convencional, o que tem invisibilizado saberes e práticas diversas no decorrer da história. Orientando-se, principalmente, em fundamentos de Sustentabilidade, Tecnologia Social e Convivência com o Semiárido, e buscando apresentar as contribuições, resistências e alternativas protagonizadas pelas mulheres nesse sentido, realizou-se um estudo de caso, de natureza aplicada, com o objetivo de conhecer os impactos ambientais, sociais e econômicos da utilização do Sistema Água Viva no Projeto de Assentamento Monte Alegre I – Upanema/RN a partir dos olhares das técnicas e agricultoras. Tal sistema é uma tecnologia social de reuso de água cinza que capta a água a ser reaproveitada a qual passa por uma filtragem até ser liberada para uso nos quintais e foi construída num esforço de entidades e de um grupo de mulheres agricultoras. Para atingimento do proposto, a coleta de dados foi realizada por meio de pesquisa exploratória, documental e bibliográfica, observação direta, entrevistas semi e não estruturadas, tendo uma abordagem qualitativa, que se valeu de categorização e análise de conteúdo para investigação dos dados. A compreensão dos dados à luz do referencial bibliográfico e das falas das sujeitas mostrou que o Sistema Água Viva contribuiu positivamente na questão da eliminação de resíduos poluentes e que as práticas de cultivo e criação foram alteradas após uso do sistema. A análise mostrou ainda que: a auto-organização local das mulheres é fator central nos processos relacionados a concepção, construção, utilização e manutenção da tecnologia social no local; que estes processos fortalecem a autonomia das mulheres e são vistos pelas agricultoras e técnicas como algo que possibilitou aprendizados vários, apesar de as agricultoras não se veem como pensantes da tecnologia; e que as agricultoras assimilam mais nitidamente o impacto monetário daquilo que produzem quando registram os dados referentes a sua produção, autoconsumo e trocas. A partir destes resultados, o trabalho aponta para a necessidade e importância de se pensar e construir coletivamente alternativas de convivência com o semiárido que possam alterar as relações entre os próprios sujeitos e destes com o ambiente. Indo mais além, evidencia-se que é urgente que se pense na sustentabilidade da vida a partir da criação e manutenção de condições materiais ou imateriais que se relacionam diretamente com a sobrevivência da natureza, propiciando uma valorização de bens comuns e uma socialização das tarefas do cuidado, considerando a realidade de grupos localizados sob a tripla dimensão de gênero, classe e raça. This research emerged from the need to focus efforts to overcome the paradigm that considers that development is only linked to economic growth based on the intensive use of conventional technology, which has made invisible diverse knowledge and practices throughout history. Focusing mainly on the foundations of Sustainability, Social Technology, and Human Coexistence with the Semi-Arid and seeking to present the contributions, resistances and alternatives performed by women in this sense, a case study of an applied nature was did with the objective of knowing the environmental, social and economic impacts of using Água Viva’s System at Monte Alegre I Settlement Project - Upanema/RN from the perspective of female technicians and women farmers. This system is a gray water reuse social technology that captures the water to be reclaimed which is filtered through to be released for use in backyards and was built in an effort by entities and a group of women farmers. To achieve the proposed, the data collection was performed through exploratory, documental and bibliographic research, direct observation, semi and non structured interviews with the subjects involved, having a qualitative approach, which was based on categorization and content analysis to investigate the data. The understanding of the data in the light of bibliographical references and female subjects's speeches showed the Água Viva System contributed positively to the issue of the elimination of pollutant residues and that the cultivation and breeding practices were altered after using the system. The analysis also showed that: the local women self-organization is a central factor in the processes related to the design, construction, use and maintenance of social technology in the place; that these processes strengthen the women autonomy and are seen by women farmers and technicians as something that made possible several apprentices, although women farmers do not see themselves as thinking about technology; and that the women farmers assimilate more sharply the monetary impact of their production when they register the data regarding their production, self-consumption and exchanges. From these results, the work points to the need and importance to think about and collectively construct alternatives of human coexistence with the semi-arid that can alter the relations between the subjects themselves and those with the environment. Going further, it is evident that it is urgent to think about the life sustainability from the creation and maintenance of material or immaterial conditions that are directly related to the survival of nature, propitiating a valuation of common goods and a socialization of the tasks of the care, considering the reality of groups located under the triple dimension of gender, class and race.

Il testo illustra i recenti sviluppi dell'iconografia musicale, una disciplina giovane ma attualmente in piena espansione, e ne evidenzia l'apporto significativo alla conoscenza non solo di aspetti materiali e istituzionali del patrimonio musicale, ma anche delle diverse maniere in cui, in contesti ed epoche lontane, la musica è stata concepita, idealizzata, rivestita di significati allegorici e simbolici. Il contributo presenta inoltre alcuni progetti di documentazione e ricerca in corso a livello internazionale e ne prospetta possibili sviluppi, funzionali all'approfondimento della conoscenza e alla valorizzazione dei beni musicali.

Delineation of the immune correlates of protection in natural infection or after vaccination is a mandatory step for vaccine development. Although the most recent techniques allow a sensitive and specific detection of the cellular immune response, a consensus on the best strategy to assess their magnitude and breadth is yet to be reached. Within the AIDS Vaccine Integrated Project (AVIP http://www.avip-eu.org) we developed an antigen scanning strategy combining the empirical-based approach of overlapping peptides with a vast array of database information. This new system, termed Variable Overlapping Peptide Scanning Design (VOPSD), was used for preparing two peptide sets encompassing the candidate HIV-1 vaccine antigens Tat and Nef. Validation of the VOPSD strategy was obtained by direct comparison with 15mer or 20mer peptide sets in a trial involving six laboratories of the AVIP consortium. Cross-reactive background responses were measured in 80 HIV seronegative donors (HIV-), while sensitivity and magnitude of Tat and Nef-specific T-cell responses were assessed on 90 HIV+ individuals. In HIV-, VOPSD peptides generated background responses comparable with those of the standard sets. The genetic diversity of HIV-1 strains circulating among HIV-1-infected South Africans was investigated in a cohort of 420 individuals enrolled as part of the AIDS Vaccine Integrated Project (AVIP) study. Representative samples (10%) were randomly selected from treatment-naive participants. Viral RNA was extracted for reverse transcriptase-initiated amplification and population-based sequencing of partial pol (encompassing protease and reverse transcriptase) and full-length integrase. Overall, HIV-1 sequences confirmed that 97.1% and 96.9% were HIV-1 subtype C in pol and integrase, respectively. Two participants were infected with unique A1/C and C/A1 recombinants in pol/integrase. Further pol sequence analysis identified mutation patterns associated with high level resistance to NNRTIs in two participants, whereas no primary mutations conferring resistance to integrase inhibitors were detected. The predominance of HIV-1 subtype C in South African populations is therefore confirmed in the AVIP cohort finalized for testing preventive or therapeutic vaccines against HIV-1 infection.

Before performing phase II/III clinical trials in Africa, preliminary studies, including assessment and building up of clinical and laboratory infrastructures, estimates of human immunodeficiency virus incidence, investigation of the background immune response, and evaluation of the cross-clade immune response, need to be done.
