Purpose: To compare the corneal biomechanical properties before and after small incision lenticule extraction (SMILE) and femtosecond laser-assisted LASIK in different levels of myopia with the Ocular Response Analyzer (Reichert Ophthalmic Instruments, Depew, NY)., Methods: A total of 187 and 79 eyes that received SMILE or LASIK, respectively, between January and June 2013 at Zhongshan Ophthalmic Center were enrolled in this study. Patients were grouped according to surgery type (SMILE or LASIK) and -6.00 diopters (D) or less (> -6.00 D) or myopia greater than -6.00 D (>-6.00 D). Corneal hysteresis, corneal resistance factor, and 37 waveform parameters were recorded and compared preoperatively and at 1 week and 1 and 3 months postoperatively., Results: There was a significant decrease of corneal hysteresis, corneal resistance factor, p1area, and p2area, and an increase of path1 and path2 in both SMILE and LASIK. In myopia -6.00 D or less, the differences between SMILE and LASIK were not significant (P > .05), but in myopia greater than -6.00 D, the corneal hysteresis, corneal resistance factor, p1area, and p2area decreased significantly more in LASIK than in SMILE (P < .05)., Conclusions: When comparing SMILE with LASIK, myopia was greater than -6.00 D, and the corneal hysteresis, corneal resistance factor, p1area, and p2area decrease was less after SMILE., (Copyright 2014, SLACK Incorporated.)

To extend the use of RNAi in chicken, we have developed a RNA interference (RNAi) system using a shortened chicken 7SK (ch7SK) promoter. The results stated that the cloned ch7SK promoter includes multiple Oct-1 motifs, SPH domain, PSE and TATA box, without CACCC box. All RNAi groups driven by ch7SK promoter showed significant mean fluorescence intensity (MFI) reduction. In the pch7SK-shEGFP transfected DF-EGFP cell culture, the MFI reduction ratio was smaller than the pmU6-shEGFP did. In the pmU6-shEGFP transfected Vero-EGFP cell culture, the MFI was reduced significantly than the pch7SK-shEGFP did. In summary, the essential part of ch7SK promoter was capable of efficiently expressing shRNAs with relatively different interfering degrees in avian and mammalian cells, respectively. Our results suggest that ch7SK promoter is an efficient alternative to commercially mouse U6 promoter in shRNA expression with chicken cells, and provide references for furthering functional genome analysis and disease resistant breeding in chicken., (Copyright © 2013 Elsevier Ltd. Poly(ethyleneglycol methacrylate) (PEGDMA) shell was used as a protective layer for the sandwiched gold nanoparticles, which were prepared through the in situ reduction in the HAuCl4 precursor in the presence of (aminopropyl)trimethoxysilane (APS) modified silica/PEGDMA core-shell microspheres. In this process, the silica/PEGDMA core-shell microspheres were afforded by the distillation-precipitation polymerization of the EGDMA monomer on the APS-modified silica particles with the aid of hydrogen-bonding interaction. The gold nanoparticles were formed at the interface between the silica core and the PEGDMA outer layer through the strong coordinate interaction between the gold nanoparticles and the amino groups on the surface of the silica during the reduction in HAuCl4. Background/aims: Endometrial carcinoma (EC) is the most common gynecological malignancy among women worldwide. Despite its prevalence, the molecular mechanisms underlying endometrial carcinogenesis are poorly understood. The purpose of this study was to examine the role of microRNA-182 and its target gene transcription elongation factor A-like 7 (TCEAL7) in EC., Methods: The expression of miR-182 in human normal endometrial epithelial cells (NEEC) and in three human endometrial carcinoma cell lines (HEC-1B, RL95-2 and AN3CA) was measured by qRT-PCR, and the mRNA and protein expression of TCEAL7 were assessed in the same three endometrial carcinoma cell lines and NEEC by qRT-PCR and western blotting, respectively. Subsequently, the target of miR-182 was predicted by bioinformatics and confirmed using a luciferase assay. Cell proliferation and colony formation of RL95-2 cells were examined by MTT assay and crystal violet staining, respectively. The expression of NFκB-p65, c-Myc and cyclin D1 proteins was determined by Western blot analysis., Results: MiR-182 was significantly upregulated and TCEAL7 was downregulated in EC cell lines compared to NEEC. We showed that MiR-182 binds directly to a conserved 8 bp sequence in the 3'-UTR of TCEAL7, and inhibition of miR-182 upregulated TCEAL7 mRNA and protein expression to levels comparable to those induced by lentiviral-mediated overexpression of TCEAL7. MiR-182 inhibition decreased cell proliferation and colony formation ability, downregulated the expression of the pro-proliferative genes c-Myc and cyclin D1, and inhibited NFκB activation, and these effects were mimicked by TCEAL7 overexpression., Conclusions: miR-182 acts as an oncogenic miRNA in EC, promoting cell proliferation by targeting the tumor suppressor gene TCEAL7 and modulating the activity of its downstream effectors c-Myc, cyclin D1 and NFκB., (© 2013 S. This study explored the possibility that the components in melanoma cytoplasm induce murine BMSCs transformation and expression of Melan-A by morphologically observing the changes of BMSCs and immunocytochemically detecting Melan-A in the cells after culturing BMSCs in medium containing melanoma cytoplasm components (MCC). MCC of B16 melanoma cells was prepared and BMSCs were cultured and induced by adding the MCC into culture medium. The cells were morphologically observed and Melan-A was immunohistochemically detected to confirm BMSCs transformation. MCC-induced BMSCs underwent morphological changes. A number of melanin granules appeared in the cytoplasm of the cells and some were released into surrounding areas. Several cells that might come from one cell formed a cluster, and their granules, together with those secreted by other induced BMSCs, formed a so-called "sphere-formed structure". The induced BMSCs expressed Melan-A. We are led to conclude that there might be some factors in the cytoplasm of melanoma cells that might induce BMSCs transformation toward melanogenic cell, or even melanoma.

Unlabelled: Acidovorax citrulli (Ac) is an important bacterium that occurs in watermelon, melon and other cucurbits. It mainly damages watermelon and melon, and can cause leaf blight, fruit rot, and even mortality., Objective: To verify the relationship between defects in the synthesis of histidine and the pathogenicity of Ac., Methods: We generated a transposon (Tn5) mutant library on the background of strain xjl12 of Ac. Then we used subclone technology to identify the gene., Results: The mutant could not elicit the hypersensitive response (HR) in nonhost tobacco, and its virulence was reduced. It is impaired in hisC, which encodes the protein histidinolphosphate aminotransferase. The other three genes (hisA, hisB and hisD) involved in the process of histidine synthesis were also studied. These mutants could not elicit the hypersensitive response (HR) in nonhost tobacco; their virulence was reduced significantly and disease symptoms caused by mutants were delayed for 48 hours when compared to the wild type strain. By adding exogenous histidine, pathogenicity of the mutants was restored., Conclusion: The change of the characteristics of the mutants was directly related to the synthesis of histidine.

This paper describes a new fluorescent family of branched dyes containing benzophenone unit including 4-N, N-diphenylamino-4'-phenacyl-stilbene (C1), 4,4'-di(4-benzoylphenylethylene)yl-triphenylamine (C2) and 4,4',4″-tri(4-benzoylphenylethylene)yl-triphenylamine (C3). Benzophenone part is coupled with core through C-C double bond. The chemical structures of the derivatives are characterized with (1)H and (13)C nuclear magnetic resonance and elemental analysis. Strong π-π stacking interactions are discovered with the analysis of the X-ray crystallographic data of C1. The absorption maxima and emission maxima of the derivatives exhibit gradual bathochromic shift from C1 to C3. The optical density of C1, C2 and C3 are shown to be related to the number of branches. The changes of dipole moments between the excited and ground states for C1, C2 and C3 were estimated to be 4.356, 8.091 and 8.479 Derby, respectively by Lippert equation, confirming that the internal charge transfer (ICT) dominates the process of excited singlet state. The possibility as fluorescence probes of the derivatives on the estimation of what region of micelles interacting with samples was evaluated.

This paper presents the molecular geometry optimization, two-photon absorption and electrochemistry of new dyes containing benzophenone part, including 4-(p-benzoyl-benzyloxy)yl-4'-nitro-diphenylethylene (C1), 4-[N-methyl-N-(2-(p-benzoyl-benzyloxy)yl-ethyl]-4'-nitro-diphenylethylene (C2), 4-[N-ethyl-N-(2-(p-benzoyl-benzyloxy)yl-ethyl]-4'-nitro-diphenylethylene (C3), and 4-N, N-bis[(2-(p-benzoyl-benzyloxy)yl-ethyl]-4'-nitro-diphenyl ethylene (C4). The molecular structural parameters show that the coplanarity of diphenylethylene moiety is diminished in the excited state for C1, while it is enhanced for C2, C3 and C4. The electron density distribution of frontier orbital suggests that the derivatives exhibit (π, π) transition with internal charge transfer character, and the extent of charge transfer of C2, C3 and C4 is larger than that of C1. The derivatives display remarkable two-photon absorption (TPA) induced up-converted emission under 800 nm Ti: Sapphire femtosecond laser excitation. The maximal TPA emission wavelength of C2, C3 and C4 is red-shifted with respect to that of C1. TPA cross sections of C2, C3 and C4 are larger than those of C1. The cyclic voltammograms and the fluorescence lifetimes of the derivatives were determined and discussed.

Inhibition of mitotic kinesins represents a novel approach for the discovery of a new generation of anti-mitotic cancer chemotherapeutics. We report here the discovery of the first potent and selective inhibitor of centromere-associated protein E (CENP-E) 3-chloro-N-{(1S)-2-[(N,N-dimethylglycyl)amino]-1-[(4-{8-[(1S)-1-hydroxyethyl]imidazo[1,2-a]pyridin-2-yl}phenyl)methyl]ethyl}-4-[(1-methylethyl)oxy]benzamide (GSK923295; 1), starting from a high-throughput screening hit, 3-chloro-4-isopropoxybenzoic acid 2. Compound 1 has demonstrated broad antitumor activity in vivo and is currently in human clinical trials.

Novel nitro-azobenzene dyes bearing one or two benzophenone branches were proposed and synthesized to improve their photophysical and photochemical properties. The new dyes exhibited double UV/visible bands, and they displayed weak fluorescence emission as excited at 350 nm. Single crystal X-ray diffraction data showed that two phenyl rings of azobenzene was almost coplanar, and the benzophenone part was neither coplanar nor linear connection with azobenzene via ether bridged bond, which have good fit with molecular geometry optimization calculation results. The cyclic voltammeric results of nitro-azobenzene dyes were firstly reported in this paper, which demonstrated that the electrochemical properties of nitro-azobenzene dyes was altered by the substitution of benzophenone part. Thermal stabilities of the new dyes were studied by the analysis of differential scanning calorimetry (DSC) and thermograving (TG) in this paper. Efficient visible-light photoinitiating polymerization of methyl methacrylate (MMA) by the novel nitro-azobenzene dyes was presented and discussed.

Objective: To probe a satisfactory surgical management of the limb salvage for osteogenic malignant tumors around the knees., Methods: From January 1989 to December 2001, 42 patients (19 males and 18 females, aged 12-46) with osteogenic malignant tumors around the knees underwent surgical management of the limb salvage, including prosthesis replacement, allogenous bone grafting, and bone cement with adriamycin filled. Based on the pathological examination, osteosarcoma was found in 11 patients, synoviosarcoma in 4 patients, malignant fibrous histiocytoma in 3 patients, and giant cell tumor of the bone in 19 patients. All the patients underwent neoadjuvant chemotherapy for 1-2 courses before operation except the patients with giant cell tumor of the bone. The patients underwent prosthesis replacement, allogenous bone grafting, bone cement with adriamycin filled, and postoperative chemotherapy. By the Enneking evaluating system, the patients were assessed on their reconstructed limb functions after the reconstructive operation for the musculoskeletal malignant tumors. Of the patients, 37 were followed up after operation., Results: According to the follow-up for 3-11 years (mean, 5.6 years) in the 37 patients, 2 patients had recurrence and metastasis of the tumor, and died 3 and 4 years after operation, respectively. One patient underwent amputation of the limb for local recurrence of the tumor. One patient had amputation of the limb for the preoperative radiotherapy and the infection and necrosis of the operative wound after the limb-salvage surgery. Two patients had amputation of the limb for the rejection of the allogenous bone graft and the formation of the fistula. Thirty-one patients had good wound healing and good functions of the limb. The results were evaluated by the Enneking evaluating system as follows: excellent in 7 patients, good in 14, fair in 10, and poor in 6., Conclusion: Making an early diagnosis, recognizing the operative indication, choosing the operative method, and performing the preoperative and postoperative chemotherapy and/or radiotherapy are the keys to achieving an ideal limb-salvage surgery for osteogenic malignant tumors around the knees.
