M4DOTA, [(2S,5S,8S,11S)-4,7,10-tris-carboxymethyl-2,5,8,11-tetramethyl-1,4,7,10-tetraazacyclododecan-1-yl]acetic acid (2e), and M4DOTMA, (R)-2-[(2S,5S,8S,11S)-4,7,10-tris-((R)-1-carboxyethyl)-2,5,8,11-tetramethyl-1,4,7,10-tetraazacyclododecan-1-yl]propionic acid (3e), are derivatives of ligand DOTA (1e) that form sterically crowded lanthanide chelates. M4DOTMA forms highly symmetric and totally rigid single Y(3+) and Yb(3+) species in which the ring substituents occupy corner positions in a square antiprismatic arrangement as shown by molecular mechanics calculations and by a quantitative interpretation of the relative magnitudes of the paramagnetic (1)H NMR shifts of dipolar origin. The NMR spectrum of YbM4DOTMA(-) displays two intense methyl peaks outside the 0-10 ppm range whose shift difference is strongly temperature dependent. YbM4DOTMA(-) (3d) could be a useful probe in magnetic resonance thermometric imaging. With only four methyl substituents on the tetraaza ring, M4DOTA forms three Yb(3+) species in solution. The methyl substituents prevent the inversion of configuration of the ethylenic groups but not of the acetate arms. Although the methyl groups are likely to preferably occupy ring corner positions, the dipolar equations do not allow one to distinguish with certainty between the two available corner (equatorial) orientations. Reliably applying the dipolar equations is less obvious than usually assumed. A single methyl substituent as in ligand MDOTA (5e) suffices to rigidify the tetraaza cycle but not the acetate arms. Racemic YbMDOTA(-) (5d) is present in solution as four totally asymmetric topomers with the methyl groups occupying either one of the two equatorial positions. A complete assignment of the solution structures on the basis of the dipolar equations is again uncertain. The nuclear magnetic relaxation dispersion curves of the Gd(3+) chelates of all the methylated DOTA ligands including DOTMA, (R)-2-[4,7,10-tris-((R)-carboxyethyl)- 1,4,7,10- tetraazacyclododecan-1-yl]propionic acid, are very similar, and intermolecular conformational processes appear to have no influence on the relaxivity of these small complexes for which the relaxation T(1) is mainly determined by the rotational correlation time (tau(r)). The hydration number of the Tb(3+) chelates measured by fluorescence decreases from DOTMA to M4DOTMA presumably because steric crowding leads to an increase of the metal-water distance.

By exploiting substrate protection of Cys148 in lactose permease, a methanethiosulfonate nitroxide spin-label was directed specifically to one of two Cys residues in a double-Cys mutant, followed by labeling of Cys148 with a thiol-reactive chelator that binds Gd(III) quantitatively. Distances between bound Gd(III) and the nitroxide spin-label were then studied by electron paramagnetic resonance. The results demonstrate that the Gd(III)-induced relaxation effects on nitroxides at positions 228, 226 (helix VII), and 275 (helix VIII) agree qualitatively with results obtained by studying spin-spin interactions [Wu, J., Voss, J., et al. (1996) Proc. Natl. Acad. Sci. U.S.A. 93, 10123-10127]. Thus, a nitroxide attached to position 228 (helix VII) is closest to the lanthanide at position 148 (helix V), a nitroxide at position 275 (helix VIII) is further away, and the distance between positions 226 (helix VII) and 148 is too long to measure. However, the Gd(III)-spin-label distances are significantly longer than those estimated from nitroxide-nitroxide interactions between the same pairs due to the nature of the chelator. Although the results provide strong confirmation for the contention that helix V lies close to both helices VII and VIII in the tertiary structure of lactose permease, other methods for binding rare earth metals are discussed which do not involve the use of bulky chelators with long linkers.

The lanthanide and Th4+ complexes with calix[4]arene ligands substituted either on the narrow or at the wide rim by four coordinating groups behave totally differently as shown by an NMR investigation of the dia- and paramagnetic complexes. Solutions of complexes were prepared by reacting anhydrous metal perchlorate salts with the ligands in dry acetonitrile (CAUTION). Relaxation time T1 titrations of acetonitrile solutions of Gd3+ by calixarenes indicate that ligands subsituted on the narrow rim form stable 1:1 complexes whether they feature four amide groups (1) or four phosphine oxide functions. In contrast, a ligand substituted by four (carbamoylmethyl)-diphenylphosphine oxide moieties on the wide rim (3) and its derivatives from polymeric species even at a 1:1 ligand/metal concentration ratio. Nuclear magnetic relaxation dispersion (NMRD) curves (relaxation rates 1/T1 vs magnetic field strength) of Gd3+, Gd3+.1 and Gd3+.3 perchlorates in acetonitrile are analyzed by an extended version of the Solomon-Bloembergen-Morgan equations. A comparison of the calculated rotational correlation times tau r shows that ligand 3 forms oligomeric Gd3+ species. The chelates of ligand 1 are axially symmetric (C4 symmetry), and the paramagnetic shifts induced by the Yb3+ ion are accounted for quantitatively. The addition of water or of nitrate ions does not modify the geometry of the complex. The metal chelates of 3 and its derivatives adopt a C2 symmetry, and the paramagnetic shifts are interpreted on a semiquantitative basis only. Water and NO3- ions completely labilize the complexes of the heavy lanthanides. The very high selectivity of ligand 3 through the lanthanide series stems from a complex interplay of factors.

The aim of this study was to detect possible alterations in semantic representation during normal aging. Forty young subjects (aged 20 to 30 years) and 40 elderly ones (divided into two groups of 60 to 70 years and 70 to 80 years), all in good health and with high education, completed a test protocol used for assessing semantic knowledge in the study of Alzheimer's disease. The same list of 42 words belonging to three different semantic categories was presented visually three times to the subjects, each time with different instructions: (a) find the words with one letter N (or more) (training task); (b) find the names of animals (test of categorical knowledge); (c) find the words representing something that prick (test of attribute knowledge of concepts). A two-way analysis of variance showed an effect of task (the times taken were significantly longer for the attribute-knowledge test), a group effect (the elderly subjects were slower) and an absence of interaction (the elderly subjects had the same profile as the young group). The results indicate the preservation of the semantic memory in normally aging groups and the usefulness is emphasized in the context of the data given in the literature on Alzheimer's disease.

The solution behavior of the Th(IV) chelate with a polyaza polycarboxylic ligand, HEHA (1,4,7,10,13,16-hexaazacyclooctadecane-N,N ',N ",N "',N "",N ""'-hexaacetic acid), is investigated by one- and two-dimensional NMR spectroscopy. [Th(HEHA)](2)(-) exhibits a very unusual irreversible topomerization process from a kinetically stable topomer of C(2) symmetry to a thermodynamically stable topomer of remarkably high symmetry (S(6)). The metal ion appears to be 12-coordinated in both geometries with a pseudoicosahedral arrangement of the chelating groups. The activation energy of the irreversible topomerization is exceedingly high (199 kJ.mol(-)(1)), and the slow kinetics of conversion is assigned to a complete reorganization of the chelate. U(IV) forms directly a stable entity of C(2) symmetry with HEHA and no reorganization of the complex is observed. The unusual topomerization phenomenon undergone by HEHA, is thus strongly dependant on the ionic radius of the encapsulated metal ion. The corresponding lanthanide chelates are totally asymmetric.

The selective dopamine D3 receptor antagonist [3H](+)S 14297 ((+)-[7-(N,N-dipropylamino)-5,6,7,8-tetrahydro-naphtho(2,3b)dihydro,2,3- furane]), labelled to high specific activity (145 Ci/mmol), bound to cloned human dopamine D3 receptors but displayed negligible binding to cloned human D2 receptors. [3H](+)S 14297 exhibited rapid association and dissociation, high affinity saturable binding (Kd = 7.0 nM) and a competition binding profile highly correlated with that of [125I]iodosulpride (r = 0.98).

Three different alpha-D-glucosyltransferases (GTFs) were separated from culture filtrates of Streptococcus cricetus strain AHT grown in a complex, standard medium in batch culture or under defined conditions of growth in the chemostat. Two of the enzymes (GTF-S1 and GTF-S2) converted sucrose into branched, soluble dextrans, and the third (GTF-I) produced a relatively linear, water-insoluble, predominantly (1----3)-linked alpha-D-glucan. When the organism was grown in complex medium modified by the removal of the fraction of high molecular weight, only GTF-S1 and GTF-S2 were released, and no GTF-I was detected. The water-insoluble glucan fraction obtained by incubating the cell-free filtrate with sucrose contained from 17 to 25% of (1----3)-glucosidic linkages, and accounted for up to 78 and 4% of the total glucans derived from growth in standard and modified medium, respectively. The soluble glucans produced in the same reaction were fractionated with ethanol to give, from both media, two distinct dextrans comprising (1) a highly branched dextran similar to the S1-dextran product of GTF-S1 and (2) a dextran containing fewer branch linkages and up to 86% of (1----6)-alpha-D-glucosidic linkages. A GTF responsible for the synthesis of the latter dextran was not separated. The structures of the glucan fractions and the products of the separated GTF were examined by enzymic degradation and methylation analysis.

When Streptococcus salivarius was grown in batch culture in the presence of various Tween detergents, the fatty acid moiety of the detergent was incorporated into the lipids of its membrane. Tween 80 (containing primarily oleic acid) markedly stimulated the production of extracellular glucosyltransferase and also increased the degree of unsaturation of the membrane lipid fatty acids. The possibility that an increase in membrane unsaturated fatty acids promoted extracellular glucosyltransferase production was examined by growing cells at different temperatures in the presence or absence of Tween 80. The membrane lipids of cells grown at 30 degrees C, 37 degrees C and 40 degrees C without Tween 80 exhibited unsaturated/saturated fatty acid ratios of 2.06, 1.01 and 0.87 respectively. A significant increase in the production of extracellular glucosyltransferase was observed at 30 degrees C compared to cells grown at 40 degrees C. However, cells produced much more exoenzyme at all temperatures when grown with Tween 80. The results indicated that an increase in the unsaturated fatty acid content of the membrane lipids was not by itself sufficient to account for the stimulation of extracellular glucosyltransferase production by Tween 80, but that the surfactant also had to be present.

Previously we described physiological properties of pontine reticulo-spinal neurons which generate bursts and decaying tonic discharges related to eye movements and neck muscle activity during ipsiversive gaze shifts (Grantyn and Berthoz 1987). Two of these "eye-neck reticulo-spinal neurons" (EN-RSN) were labeled by intra-axonal injections of HRP. The present report provides a detailed description of their morphology with an emphasis on the topography of axon collaterals, bouton numbers, and the structure of preterminal ramifications in different target areas. The cell bodies of labeled EN-RSNs were located rostro-ventrally to the abducens nucleus. Their descending axons issued 8 and 13 collaterals (left and right EN-RSN, respectively) at different rostro-caudal levels, between the abducens nucleus and the pyramidal decussation. On the basis of the size of their cell bodies, the isodendritic type of dendritic branching and their multiple collateralization, EN-RSNs correspond to the class of "generalized" reticular neurons, often referred to as The Scheibels' neurons. Collaterals of EN-RSNs terminated in the following structures: the abducens and facial nuclei, the medial and lateral vestibular nuclei, the nn. prepositus and intercalatus, and the bulbar reticular formation. As judged from bouton numbers, the strongest connection of both neurons was with the abducens nuclei. Terminations in the rostral part of the medial vestibular and prepositus nuclei indicate that EN-RSNs may also influence oculomotor output activity through these indirect routes. In the facial nucleus, a majority of terminations was found in its medial subdivision containing motoneurons of ear muscles. However, other subdivisions were also contacted by EN-RSNs. Most terminations in the rostral bulbar reticular formation are distributed to the dorsal, gigantocellular field. Within this field, there is a substantial contribution to the zone characterized by the highest density of reticulo-spinal neurons projecting directly to neck motoneurons. Other target areas which may participate in the modulation of spinal cord activity by EN-RSNs are the ventral reticular nucleus in the caudal medulla and the lateral vestibular nucleus. EN-RSNs also establish connections with precerebellar structures: the prepositus and the paramedian reticular nuclei. The numbers of boutons on collaterals issued within 6 mm of the injection site varied between 37 and 469. The occurrence of presumed axo-somatic contacts was low (0-8.2%) and not characteristic for any particular target area. Local accumulations of boutons in the form of small and large field clusters was a common observation.(ABSTRACT TRUNCATED AT 400 WORDS)
