1. [7alpha-(3)H(1)]- and [7beta-(3)H(1)]-Cholesterol were synthesized by a modified method. 2. The stereochemistry of Delta(7)-bond formation during ecdysone and ecdysterone biosynthesis in the insect, Calliphora erythrocephala and the plants, Taxus baccata and Polypodium vulgare was investigated by using [4-(14)C,7alpha-(3)H(1)]cholesterol and [4-(14)C,7beta-(3)H(1)]cholesterol. 3. In each case, the 7beta hydrogen was stereospecifically eliminated. 4. The possible significance of the results is discussed in relation to double-bond formation in other systems and the stage at which the Delta(7) bond is introduced during ecdysone biosynthesis.

Rat-liver supernatant preparations are capable of achieving the biological sulphation of l-tyrosine methyl ester, the reaction proceeding maximally at a substrate concentration of 30 mm and at pH 7.0. 2. Two sulphated products are formed, one of which has been identified as l-tyrosine O-sulphate. On the basis of indirect evidence the other product can be assumed to be l-tyrosine O-sulphate methyl ester. 3. An enzyme present in rat-liver supernatant preparations is capable of converting l-tyrosine O-sulphate methyl ester into l-tyrosine O-sulphate. This enzyme is inhibited by l-tyrosine methyl ester. 4. l-Tyrosine ethyl ester also yields two sulphated products when used as an acceptor in the liver sulphating system. One of these has been identified chromatographically as l-tyrosine O-sulphate and the other may be presumed to be l-tyrosine O-sulphate ethyl ester.

1. A rat-liver supernatant preparation can achieve the biological O-sulphation of l-tyrosylglycine and l-tyrosyl-l-alanine at pH7.0. 2. The optimum concentrations of l-tyrosylglycine and l-tyrosyl-l-alanine in this system are 50mm and 60mm respectively. 3. l-Tyrosylglycine yields two sulphated products, whereas l-tyrosyl-l-alanine yields three sulphated products, when used as acceptor for sulphate in the rat-liver system. 4. With both substrates, one of the sulphated products has been identified as the O-sulphate ester of the corresponding parent peptide.

Selectivity coefficients, K(H)(M), for the exchange of alkali metal ions and ammonium ions with the hydrogen form of ZeoKarb 225 cation-exchange resin have been determined in various organic solvent-water and organic solvent-water-phenol mixtures. There is an increase in K(H)(M) in passing from purely aqueous conditions to media of increasing organic solvent content, frequently with the effect of producing magnified differences and selectivity reversals between one ion and another. The results are discussed in terms of the relationship between K(H)(M) and the reciprocal of the dielectric constant of the medium and also in the light of free energies of transfer of cations between methanol-water systems. The effect of phenol giving a lowering of K(H)(Cs) in methanol-water, of K(H)(K) and K(H)(Na) in ethanol-water and of K(H)(Na) and K(H)(L1) in acetone-water systems cannot be fully reconciled with the mechanism involving bonding proposed by earlier workers.

1. l-Tyrosylglycine O[(35)S]-sulphate is metabolized by the rat to yield the O[(35)S]-sulphate esters of l-tyrosine, p-hydroxyphenylpyruvic acid and p-hydroxyphenylacetic acid. 2. The proportion of the administered peptide which is excreted as l-tyrosine O[(35)S]-sulphate is greater at a higher dose. 3. An enzyme capable of hydrolysing the peptide bond of l-tyrosylglycine O[(35)S]-sulphate to yield l-tyrosine O[(35)S]-sulphate has been detected in rat liver and kidney. 4. The activity of this enzyme is completely inhibited by a large excess of l-tyrosylglycine.
