Skeletal maturation was evaluated in newborn and infant rhesus monkeys that had been subjected to a marginally zinc-deficient diet (4 ppm zinc) from conception through 12 months of postnatal life. Serial radiographic assessment of skeletal development was performed and compared to both ad libitum and pair-fed controls. Radiographs were obtained at birth and at 1, 3, 9, and 12 months of age. In each age group a maturation indicator was selected to identify individuals with abnormal skeletal maturation defined on the basis of presence of epiphyseal ossification centers. Animals were compared only within a given sex group. Additionally, to evaluate endochondral bone mineralization, the appearance of the zone of provisional calcification on the metaphyseal side of the growth plate and the width of the growth plate were assessed. A marginal level of zinc deprivation during gestation and during the 1st yr of life was found to be associated with significantly delayed skeletal maturation and defective mineralization. This abnormality of bone mineralization has many features similar to human rachitic syndromes and suggests that zinc plays an important role in endochondral bone formation.

An endogenous type C virus recently isolated from the Columbian black-tailed deer (Odocoileus hemionus) was used as a molecular probe to study the distribution of virus-related nucleotide sequences in cellular DNAs of mammalian species. By DNA-DNA hybridization, the most extensive homology was demonstrated between the viral complementary DNA and cellular DNA isolated from Odocoileus species. DNAs of representatives of other genera within the same family, Cervidae, were partially related to the virus, consistent with the phylogenetic relationship of these species to Odocoileus. O. hemionus viral sequences were also detected within cellular DNAs of members of a more distantly related artiodactyl family, Bovidae. These findings suggest the genetic transmission of type C viral genes within cervids and bovids for a period of at least 25 to 30 million years. There was no detectable nucleotide sequence homology between O. hemionus virus and representatives of other major groups of mammalian type C viruses. These results indicate that despite the known antigenic relatedness of mammalian type C viruses, the O. hemionus virus has diverged sufficiently to be considered the prototype of a separate group. By radioimmunological techniques, it was possible to detect and partially purify, from normal tissues of cervid species, antigens related to the major structural protein of the O. hemionus virus. The present findings, that O. hemionus virus has been genetically transmitted for millions of years and yet has maintained the ability to be expressed as infectious virus, argue for positive evolutionary selective pressures for the maintenance of type C viral genes.

We have previously reported that estrogens have the potential to induce new forms of renin substrate in addition to elevating the major circulating form of this protein. One of these estrogen-induced forms had a molecular weight in excess of 150,000. In this study we have compared the plasma concentration of the high-molecular-weight renin substrate in normotensive women receiving estrogen therapy and women with estrogenic hypertension. A statistically significant elevation of this protein was associated with estrogenic hypertension and normotensive pregnant women at term. This form of renin substrate differed from the major form with respect to electrophoretic mobility, isoelectric point, and immunologic cross-reactivity. In addition, kinetic analysis indicated that this high-molecular-weight substrate has a significantly higher affinity for the enzyme renin than the major circulating form (Km = 1800 +/- 290 versus 3520 +/- 260 ng angiotensin I equivalents/ml). These results suggest that in addition to renin substrate concentration, substrate composition may play an important role in blood pressure regulation.

This study examined the role of the lipoxygenase (LO) pathway in the maintenance of hypertension in rats with two-kidney, one clip (2K,1C) Goldblatt hypertension. A single dose of the lipoxygenase blocker phenidone was injected intraperitoneally to 2K,1C rats during the early phase (14 days) of the development of hypertension (mean intraarterial blood pressure 137 +/- 3.9 mm Hg). Phenidone (60 mg/kg) markedly decreased arterial pressure to nadir levels of 58.9% of resting blood pressure. The maximal changes were observed 15 min after injection and the hypotensive response was sustained for at least 2 h. Plasma renin concentration (PRC) increased from 74.3 +/- 18.9 to 281.0 +/- 6.5 ng/mL/h after injection (P less than .05). Thus, the hypotensive effect of phenidone was not due to suppression of renin secretion but presumably due to inhibition of its effects. It is suggested that arachidonate metabolites of the LO pathway at the vascular bed may be involved in maintenance of high arterial pressure in 2K,1C renovascular hypertension in rat.

Plasma renin substrate, a widely measured parameter of the renin reaction, is quantitated indirectly by the measurement of liberated angiotensin I upon exhaustive incubation of plasma with added renin. To overcome methodological problems of this assay system, we have developed a direct radioimmunoassay for this plasma protein using renin substrate purified from pooled plasma of normotensive subjects as the antigen. Comparison of substrate quantitated by the two assay systems (direct and indirect) indicates a 1:1 correlation with the exception of certain subjects with elevated substrate levels induced by estrogen therapy. To study the possibility of multiple substrate forms, we have made a comparison of substrate quantitated by both radioimmunoassays in conjunction with electrophoresis of plasma on polyacrylamide gel. One major form of substrate with a retardation factor (Rf) = 0.60 was found in normotensive and essential hypertensive subjects which gave a 1:1 correspondence on quantitation by the two methods. In contrast, six of 16 women on oral contraceptives demonstrated three forms of substrate (Rf = 0.16, 0.35, and 0.60) on electrophoresis. Substrate with Rf = 0.16 and 0.35 did not cross-react with the antiserum prepared against substrate from normotensive subjects, implying structural differences in these proteins.

Infant monkeys (Macaca mulatta) exposed to narcotic analgesics during labor after treatment of dams were evaluated for growth and development over a 14-week period. Ten infants exposed to meperidine (2 mg/kg maternal dose) or alfentanil (0.1 mg/kg maternal dose) were compared with seven controls whose dams received no analgesic. No group differences in weight gain or body growth were observed. Alfentanil-exposed infants appeared to have a higher incidence of infections requiring veterinary treatment. Measures of spontaneous activity showed a significantly higher incidence of immature rest pattern (laying) in the alfentanil group associated with lower overall locomotor activity relative to controls. Meperidine-exposed infants had a generally higher overall level of locomotor activity than controls that was statistically significant at 11 weeks of age. Alfentanil-exposed animals were impaired in performance of a simple cognitive task (object constancy) at 2 to 3 months of age and meperidine-exposed infants showed a similar nonsignificant trend. Gross and fine motor maturations were similar to those of controls. Thus, in this situation, the impact of obstetric analgesic treatment was demonstrable for several months after birth in some limited areas.

The use of the mini-airflow apparatus for the measurement of gossyplure release rates is described. The method involves the passage of air over controlled-release dispensers containing radiolabeled pheromone, through a coarse frit, and entrapment of the gossyplure on glass beads. Desorption of the beads with solvent is followed by quantification by liquid scintillation counting. The results of release rate measurements from hollow fibers, red rubber septa, and red rubber wicks are discussed.

How can health educators influence public policy so as to increase the resources for disease prevention? The following case history of the New York City Coalition to End Lead Poisoning (NYCCELP) describes how an advocacy group sought to educate parents, health workers and policy makers about a major urban health problem. Using health education techniques such as small group meetings, mass media coverage and community organizing, NYCCELP hoped to persuade city agencies to enforce more systematically existing laws for the prevention of lead poisoning and to allocate more resources for screening and education. By defining the problem of lead poisoning broadly, the Coalition was able to attract diverse constituencies including housing organizations, public health advocacy groups, public interest lawyers, elected officials and children's rights groups. In part as a result of NYCCELP's efforts, the city government has screened more children for lead poisoning, hired additional health educators, enforced relevant sections of the housing code more often, and allocated new resources for lead poisoning control.

This study was designed to determine if dopaminergic modulation of aldosterone secretion is mediated through the renin-angiotensin system. In rats, intraarterial administration of metoclopramide (MCP), a dopamine antagonist, resulted in a significant elevation of plasma aldosterone (PA) 5 min after administration and a peak response 10 min after administration. Pretreatment with L-dopa (30 mg/kg) 30 min before administration of MCP suppressed the early PA response to MCP. PRA after MCP showed no change at 5 min but increased significantly at 10 min, with peak responses occurring at 30 min. Preadministration of L-dopa blunted and delayed the PRA response to MCP. Preadministration of the angiotensin-converting enzyme inhibitor, SQ 14,225 (1 mg/kg), did not inhibit the PA response to MCP. Infusion of the angiotensin II antagonist, saralasin (10 micrograms/kg min-1), begun 30 min before MCP, depressed basal levels of PA slightly but did not significantly alter the PA response to MCP. Rats studied 36 h after bilateral nephrectomy displayed intact PA responses to MCP, but there was no PRA response to MCP. The results indicate that dopaminergic modulation of PA secretion occurs independently of alterations in renin secretion.

To investigate the effects of marginal zinc deficiency on early development, rhesus monkeys were fed a diet marginally deficient in zinc (M; 4 micrograms/g) throughout pregnancy and during the first month of lactation. Despite the low concentration of zinc in the diet. M dams did not develop overt signs of zinc deficiency. However, compared to control dams fed diets adequate in zinc (C; 100 micrograms Zn/g), M dams showed a low response to the mitogens concanavalin A and phytohemagglutinin. Pregnancy outcome was similar in the two groups and all of the neonates were judged to be healthy at delivery. From birth until d 30 of age, the infants were closely monitored for signs of zinc deficiency, and at d 30, they were killed and tissues were removed and analyzed for a number of parameters reported to be affected by zinc status. At birth, M infants had low plasma zinc concentrations compared to controls; however, this difference was not observed at d 30. D 30 M infants showed a normal response to the mitogens concanavalin A and phytohemagglutinin, but showed a low response to pokeweed mitogen. Tissue (liver, brain, spleen, kidney, and heart) trace element concentrations were similar in the two groups of infants, as were liver metallothionein concentrations and 65Zn uptake/retention by isolated hepatocytes. Infant wt gain was inversely correlated with plasma zinc, liver zinc, and liver metallothionein concentrations in both the M and C groups.(ABSTRACT TRUNCATED AT 250 WORDS)
