Energy metabolism plays a crucial role in supporting cancer cell growth and driving tumor progression. Our objective was to create a unique gene signature based on metabolic genes that could accurately predict the prognosis of patients with ovarian cancer (OC). We accessed microarray data of patients with OC from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases. Patients from the TCGA dataset were divided into training and internal validation sets, maintaining a ratio of 3:1. Based on Least absolute shrinkage and selection operator Cox regression analysis, twenty-nine metabolism-related genes were identified for the development of the metabolic signature. Patients in the training set were successfully divided into lowand high-risk groups with a significantly different prognosis (Hazard Ratio (HR): 2.76, 95% Confidence Interval (CI): 2.12-3.59, p < 0.001). The prognostic value of this signature was confirmed in the internal (HR: 3.06, 95% CI: 1.80-5.17, p < 0.001) and external validation sets (HR: 2.17, 95% CI: 1.57-2.99, p < 0.001). The timedependent receiver operating characteristic (ROC) at the 5-year interval demonstrated that the prognostic accuracy of this metabolic signature (Area under curve (AUC) = 0.723) was superior to that of any other clinicopathological features, including the Federation of Gynecology and Obstetrics stage (AUC = 0.509), grade (AUC = 0.536), and debulking status (AUC = 0.637). Further immune cell infiltration analysis showed that low-risk patients had a higher enrichment of immune-activating cells. In conclusion, a novel metabolic signature with good performance was established in this study. Terbium scandium aluminum garnet (TSAG) crystals have been widely used in magneto-optical systems. We investigate the complex refractive index of the TSAG crystal in the terahertz frequency range using terahertz (THz) time-domain spectroscopy in the temperature range 100–300 K. It is observed that the refractive index and the absorption coefficient increase with the THz frequency. The refractive index increases with the temperature. We measure the temperature coefficient of the refractive index of the TSAG crystal in the frequency range 0.4–1.4 THz. Furthermore, the loss tangent, i.e., the ratio of experimental values of the imaginary and real part of the dielectric permittivity, is found to be almost independent of frequency. TSAG is very promising for applications in THz optoelectronics because it has a high dielectric constant, low loss, and low thermal coefficient of the dielectric constant. [ABSTRACT FROM AUTHOR]

Background. Controversies on the utility of laparoscopic mini gastric bypass (LMGB) in weight loss and type 2 diabetes mellitus (T2DM) control still exist. Methods. We conducted a comprehensive literature search of PubMed, EMBASE, and Cochrane Library. Review Manager was used to perform the meta-analysis and the weighted mean difference (WMD) and/or odds ratio with 95% confidence interval (95% CI) were used to evaluate the overall size effect. Considering the wide utilization of multi-component supplementary cementitious materials in recycled aggregate concrete, an intelligent prediction of comprehensive mechanical properties of this sustainable low-carbon concrete was proposed in this study using hybrid machine learning algorithms, rather than conducting tremendous experiments to establish a statistical empirical model previously. The database consisting of 980 datasets was used to train, validate and test the machine learning algorithms with various evaluation indicators. SHapley Additive exPlanations (SHAP) values were used for interpretability analysis of the predictive results of algorithms to quantify the contribution of each input feature. The results show that the hybrid algorithms possess higher prediction accuracy in terms of compressive strength, flexural strength, splitting tensile strength and elastic modulus compared to the single algorithms, among which Sparrow Search Algorithm-Extreme Gradient Boosting (SSA-XGB) stands out as the most promising algorithm model with satisfactory stability and generalization performance. For all mechanical properties, the effective water-binder ratio, cement content, and superplasticizer dosage are extremely important influencing factors. This study provides a systematic assessment framework for predicting the comprehensive mechanical properties of recycled aggregate concrete with supplementary cementitious materials, beneficial for the data-driven intelligent mix proportions design of sustainable low-carbon concrete in construction sector.

Soil organic carbon (SOC) mineralization is a key component of the global carbon cycle. Its temperature sensitivity Q10 (which is defined as the factor of change in mineralization with a 10 °C temperature increase) is crucial for understanding the carbon cycle-climate change feedback but remains uncertain. Here, we demonstrate the universal control of carbon quality-availability tradeoffs on Q10. When carbon availability is not limited, Q10 is controlled by carbon quality; otherwise, substrate availability controls Q10. A model driven by such quality-availability tradeoffs explains 97% of the spatiotemporal variability of Q10 in incubations of soils across the globe and predicts a global Q10 of 2.1 ± 0.4 (mean ± one SD) with higher Q10 in northern high-latitude regions. We further reveal that global Q10 is predominantly governed by the mineralization of high-quality carbon. The work provides a foundation for predicting SOC dynamics under climate and land use changes which may alter soil carbon quality and availability. [ABSTRACT FROM AUTHOR]

Soil respiration (Rs), a key process in the terrestrial carbon cycle, is very sensitive to climate change. In this study, we synthesized 54 measurements of annual Rs and 171 estimates of Q10 value (the temperature sensitivity of soil respiration) in grasslands across China. We quantitatively analyzed their spatial patterns and controlling factors in five grassland types, including temperate typical steppe, temperate meadow steppe, temperate desert steppe, alpine grassland, and warm-tropical grassland. Results showed that the mean (± SE) annual Rs was 582.0 ± 57.9 g C m−2 yr−1 across Chinese grasslands. Annual Rs significantly differed among grassland types, and positively correlated with mean annual temperature, mean annual precipitation, soil organic carbon content and aboveground biomass, but negatively correlated with latitude and soil pH (P < 0.05). Among these factors, mean annual precipitation was the primary factor controlling the spatial variation of annual Rs in Chinese grasslands. The mean contributions of growing season Rs and heterotrophic respiration to annual Rs were 78.7 % and 72.8 %, respectively. Moreover, the mean (± SE) of Q10 across Chinese grasslands was 2.60 ± 0.08, ranging from 1.03 to 8.13, and varied largely within and among grassland types, and among soil temperature measurement depths. Generally, the seasonal variation of soil respiration in Chinese grasslands cannot be well explained by soil temperature using the van't Hoff equation. Longitude and altitude were the dominant driving factors and accounted for 26.0 % of the variation in Q10 derived by soil temperature at the depth of 5 cm. Overall, our findings advance our understanding of the spatial variation and environmental control of soil respiration and Q10 across Chinese grasslands, and also improve our ability to predict soil carbon efflux under climate change on the regional scale. We use transient terahertz photoconductivity measurements to demonstrate that upon optical excitation of CH3NH3PbI3 perovskite, the hole transfer from CH3NH3PbI3 into the organic hole-transporting material (HTM) Spiro-OMeTAD occurs on a sub-picosecond timescale. Second-order recombination is the dominant decay pathway at higher photo-excitation fluences as observed in neat CH3NH3PbI3 films. In contrast, under similar experimental conditions, second-order recombination weakly contributes the relatively slow recombination between the electrons in the perovskite and the injected holes in HTM, as a loss mechanism at the CH3NH3PbI3/Spiro-OMeTAD interface. In the crystal structure of the title polymeric complex, [Hg2(C10H6O6S2)(H2O)4] n, the metal atom of an H2O→Hg⋯Hg←H2O unit [Hg←Owater = 2.124 (7) Å] is linked to the metal atom of an adjacent unit by two water molecules [Hg←Owater = 2.619 (7) and 2.872 (7) Å], giving rise to a polycationic chain. Adjacent chains are linked through the dianion via a weaker interaction [Hg⋯Osulfonate = 3.062 (7) Å] to form layers. Growth differentiation factor 11 (GDF11) has been shown to promote stem cell activity and rejuvenate the function of multiple organs in old mice, but little is known about the functions of GDF11 in the diabetic rat model of hindlimb ischemia. In this study, we found that systematic replenishment of GDF11 rescues angiogenic function of endothelial progenitor cells (EPCs) and subsequently improves vascularization and increases blood flow in diabetic rats with hindlimb ischemia. Conversely, anti-GDF11 monoclonal antibody treatment caused impairment of vascularization and thus, decreased blood flow. In vitro treatment of EPCs with recombinant GDF11 attenuated EPC dysfunction and apoptosis. Mechanistically, the GDF11-mediated positive effects could be attributed to the activation of the transforming growth factor-β/Smad2/3 and protein kinase B/hypoxia-inducible factor 1α pathways. These findings suggest that GDF11 repletion may enhance EPC resistance to diabetes-induced damage, improve angiogenesis, and thus, increase blood flow. The paucity of investigations of carbon (C) dynamics through the soil profile with warming makes it challenging to evaluate the terrestrial C feedback to climate change. Soil microbes are important engines driving terrestrial biogeochemical cycles; their carbon use efficiency (CUE), defined as the proportion of metabolized organic C allocated to microbial biomass, is a key regulator controlling the fate of soil C. It has been theorized that microbial CUE should decline with warming; however, empirical evidence for this response is scarce, and data from deeper soils are particularly scarce. Here, based on soil samples from a whole-soil-profile warming experiment (0 to 1 m, +4 °C) and 18O tracing approach, we examined the vertical variation of microbial CUE and its response to ~3.3-y experimental warming in an alpine grassland on the Qinghai-Tibetan Plateau. Microbial CUE decreased with soil depth, a trend that was primarily controlled by soil C availability. However, warming had limited effects on microbial CUE regardless of soil depth. Similarly, warming had no significant effect on soil C availability, as characterized by extractable organic C, enzyme-based lignocellulose index, and lignin phenol-based ratios of vanillyls, syringyls, and cinnamyls. Collectively, our work suggests that short-term warming does not alter microbial CUE in either surface or deep soils, and emphasizes the regulatory role of soil C availability on microbial CUE. Abstract Background Nutritional status and inflammation are closely associated with poor outcome in malignant tumors. However, the prognostic impact of postoperative in these variables on breast cancer (BC) remains inconclusive. We aimed to determine whether prognostic nutritional index (PNI), systemic immune‐inflammation index (SII), neutrophil–lymphocyte ratio (NLR) and platelet–lymphocyte ratio (PLR) affect two long-term outcomes among patients after curative resection of BC. Methods We retrospectively reviewed 508 patients with BC treated with curative surgery between February 5, 2013 and May 26, 2020. All patients were divided into 3 groups based on tertiles (T1-T3) of PNI, SII, NLR, and PLR. The effects of four indexes on disease-free survival (DFS) and overall survival (OS) have been evaluated using Cox proportional hazards models and Kaplan–Meier method. Results Compared with PNI-lowest cases, patients with highest PNI showed significantly longer DFS (multivariate adjusted hazard ratio [HR] = 0.37, 95% confident interval [CI] 0.19–0.70, P for trend = 0.002), whereas higher PLR seemed to be marginally associated with poorer DFS (P for trend = 0.086 and 0.074, respectively). Subgroup analyses indicate the potential modification effects of family history of BC and radiotherapy on the prognosis value of PNI to DFS in BC patients (P for interaction = 0.004 and 0.025, respectively). Global environmental changes can remarkably alter the amount of fine root litter inputs to the soil; these inputs affect soil CO2 and N2O emissions and net N mineralization processes by changing C and N supply to microorganisms. However, how these C and N processes respond to the amount of fine root litter input is yet not known. In this study, a year-long incubation experiment was conducted to investigate the impacts of changes in fine root litter biomass input on soil respiration, N2O emissions, and net N mineralization. Soil samples were obtained from forests of four biomes (boreal, temperate, subtropical, and tropical), and each sample was amended with fine roots from two species (either native tree species or maize roots) with four levels of root biomass input. The cumulative CO2 emissions increased linearly with root input levels, regardless of soil and root litter types. Soil respiration responded strongly to root inputs within the first 100 days and then leveled off. Root inputs retarded soil N2O emissions and net N mineralization, and the length of delay increased with root input levels, except for temperate and subtropical soils amended with tree roots, for which N2O emission dynamics were not altered by root input. Tree roots retarded net N mineralization more intensively than maize roots except for the tropical tree roots. Cumulative N2O emissions increased linearly with root input levels in only some soil type eroot specieseroot input level combinations. Taken together, our results suggest that increased fine root biomass production might result in a linear increase of soil C loss via heterotrophic respiration, indicating that the first-order kinetic functions that have been widely used in the soil C models are still valid for predicting C mineralization rates in response to the changes in the amount of root litter inputs. Fine roots in their initial decomposition stage could be the predominant sources of soil N2O emissions in some but not all terrestrial ecosystems. However, increased fine root input might retard net N mineralization, which might disrupt the temporal pattern of ecosystem N cycling, and thus have important consequences on plant N supply and growth. Purpose To explore the feasibility of microwave ablation (MWA) of the vertebral growth plate as a minimally invasive treatment for early-onset scoliosis. Materials and methods One side of the L1–L3 vertebral growth plates were ablated using different MWA powers. Ablation safety and size were examined. Subsequently, L1–L3 vertebral growth plates were ablated on one side for 40 s at 20 W. At 2, 4, and 6 weeks after the ablation, growth changes of the spine were observed. Context: Changium smyrnioides Wolff (Apiaceae) is an endangered medicinal plant with numerous pharmacological uses. Objective: To investigate the effect of light intensity levels on the growth and accumulation of secondary metabolites of C. smyrnioides, cultivated seedlings were subjected to different relative light intensities via sun-shading. Materials and methods: Changium smyrnioides seedlings were subjected to five irradiance treatments (100, 60.54, 44.84, 31.39, and 10.56% sunlight) in glasshouse for 9 months. Enzymatic and non-enzymatic antioxidants with spectrophotometric method, photosynthetic parameters with Li-6400XT, dry matter accumulation and active component contents in the root with spectrophotometric and HPLC method were analyzed. Results: With an increase in relative light intensity levels, activities of enzymatic and non-enzymatic antioxidants, and malondialdehyde (MDA) contents were increased overall, while net photosynthetic rate (Pn) and dry matter accumulation patter first increased and then declined. The highest net photosynthetic rate (30.68 μmol/m2·s) and dry root weight (5.07 g) were achieved under 60.54% sunlight. Lower relative light intensity levels stimulated the accumulation levels of bioactive compounds in the roots so that the highest contents of mannitol (1.35%) and choline (405.58 μg/g) were recorded under 31.39% sunlight, and the highest polysaccharide content (10.80%) were achieved under 44.84% sunlight. With a decrease in the relative light intensity levels, the water-soluble component content increased first and then decreased. Discussion and conclusion: The results revealed that 31.39–60.54% sunlight serve as appropriate relative light intensity conditions for cultivated C. smyrnioides.

This study used the finite element method (FEM) to analyze the stress field and seepage field of a roller-compacted concrete (RCC) dam, with an upstream impervious layer constructed with different types of concrete materials, including three-graded RCC, two-graded RCC, conventional vibrated concrete (CVC), and grout-enriched vibrated RCC (GEVR), corresponding to the design schemes S1 through S4. It also evaluated the anti-seepage performance of the imperious layer in the four design schemes under the normal water level and flood-check level. Stress field analysis of a retaining section and discharge section shows that the maximum tensile stress occurs near the dam heel, the maximum compressive stress occurs near the dam toe, and the stress distributions in the four schemes can satisfy the stress control criteria. Seepage field analysis shows that the uplift pressure heads in schemes S3 and S4 descend rapidly in the anti-seepage region, and that the calculated results of daily seepage flow under the steady seepage condition in these two schemes are about 30%–50% lower than those in the other two schemes, demonstrating that CVC and GEVR show better anti-seepage performance. The results provide essential parameters such as the uplift pressure head and seepage flow for physical model tests and anti-seepage structure selection in RCC dams.

The structure and function of alpine grassland ecosystems, including their extensive soil carbon stocks, are largely shaped by temperature. The Tibetan Plateau in particular has experienced significant warming over the past 50 y, and this warming trend is projected to intensify in the future. Such climate change will likely alter plant species composition and net primary production (NPP). Here we combined 32 y of observations and monitoring with a manipulative experiment of temperature and precipitation to explore the effects of changing climate on plant community structure and ecosystem function. First, long-term climate warming from 1983 to 2014, which occurred without systematic changes in precipitation, led to higher grass abundance and lower sedge abundance, but did not affect aboveground NPP. Second, an experimental warming experiment conducted over 4 y had no effects on any aspect of NPP, whereas drought manipulation (reducing precipitation by 50%), shifted NPP allocation belowground without affecting total NPP. Third, both experimental warming and drought treatments, supported by a meta-analysis at nine sites across the plateau, increased grass abundance at the expense of biomass of sedges and forbs. This shift in functional group composition led to deeper root systems, which may have enabled plant communities to acquire more water and thus stabilize ecosystem primary production even with a changing climate. Overall, our study demonstrates that shifting plant species composition in response to climate change may have stabilized primary production in this high-elevation ecosystem, but it also caused a shift from aboveground to belowground productivity. Objective: To evaluate the influence of long-term highly active antiretroviral therapy (HAART) on bone metabolism in patients with acquired immune deficiency syndrome (AIDS)., Methods: A total of 82 AIDS patients who received HAART in the First Affiliated Hospital of Zhejiang University Medical School during January 2012 and May 2016 were enrolled in the study. Patients were divided into TDF group (n=41) and AZT group (n=41), and glomerular filtration rate (GFR), serum calcium, serum phosphate and serum ALP levels in 6, 12, 18, 24 and 30-month during the treatment were analyzed in both groups., Results: Both TDF-based and AZT-based therapies significantly improved CD4(+) T-cell levels of the patients, and no significant difference was observed between the groups. Compared with the baseline, GFR [(120.71±62.85) vs(110.08±39.18) mL/min] and serum phosphate [(1.25±0.19) vs(1.22±0.21) mmol/L] levels rose after 6-month treatment in TDF group, but the difference was not statistically significant (P>0.05); Serum ALP [99.0(79.5-124.0) U/L vs 80.0(60.5-96.0) U/L] and serum calcium [(2.32±0.12) vs(2.25±0.17) mmol/L] levels rose significantly (P<0.05). As treatment continued, the levels of GFR, serum calcium and serum phosphate declined, while the serum ALP number was still increasing. After 30 months of HAART, the level of serum calcium [(2.16±0.15) vs(2.25±0.17) mmol/L], serum phosphate [(1.06±0.17) vs(1.22±0.21) mmol/L], GFR [(98.13±30.43) vs (110.08±39.18) mL/min] declined significantly (P<0.05) in TDF group, while serum ALP [110.0(98.5-120.5) U/L vs 80.0(60.5-96.0) U/L, P<0.05] increased. In AZT group, serum calcium and serum ALP levels rose and GFR level was declined (P<0.05), while serum phosphate level was not significantly changed during the treatment (P>0.05). Compared with AZT group, there were greater changes on the levels of GFR, serum calcium, serum phosphate and serum ALP in TDF group., Conclusion: HAART is effective for patients with AIDS, and TDF-based therapy may have significant impact on bone metabolism of the patients, which needs close monitoring and timely intervention or adjustment if necessary.

Latent reservoir (LR) of HIV is the cells (such as CD4(+)T cell) where HIV is able to hide. These cellular reservoirs are located throughout the body, including the spleen, lymph nodes, gastrointestinal lymphoid tissues, and become the major obstacle to cure HIV infection. To truly cure patients, a new strategy "shock and kill" was put forward by scientists, which is to shock HIV-infected cells out of hidden reservoirs in the body and then kill them. Quantitatively evaluating the size of long-lived LR is essential to this strategy. This paper reviews assays that measure the magnitude of the latent HIV reservoir, including Alu-gag PCR, quantitative viral outgrowth assay (Q-VOA) and tat/rev induced limiting dilution assay(TILDA). Alu-gag PCR can differentiate the integrated and un-integrated HIV DNA, however, it cannot distinguish defective virus from competent virus, leading to overestimate the real size of LR. TILDA measures cells with inducible msRNA as these transcripts are absent in latently infected cells but induced upon viral reactivation. It requires small blood sample size, does not need extraction of viral nucleic acids, can be completed in 2 d and covers a wide dynamic range of reservoir sizes, but has the disadvantage of overestimating the true size of LR.
