This study aimed to produce yogurt with Ganoderma lucidum residues and selected probiotic bacteria. To select the most potent GABA-producing strain (GABA - ?-aminobutyric acid), nine probiotic bacteria were subjected to the glutamate decarboxylase (GAD) activity assay. Limosilactobacillus reuteri DSM 17938 was selected and used in preparing fresh yogurts, with and without supplementation of Ganoderma residues obtained after water extraction [GW (Ganoderma waste) 2% (w/v)]. A decrease in pH during fermentation and the occurrence of syneresis were investigated. Lactic acid bacteria (LAB) viability and anti-Escherichia activity were estimated. Further, the cytotoxic effect of yogurt extracts on the human colon cancer cell line (HCT116) was surveyed. Besides functionality, the sensory attributes were evaluated. The pH values did not significantly change with the GW addition while increasing the LAB counts [up to 9.76 ± 0.11 log colony-forming units (CFU) mL-1] and the cytotoxic effect on HCT116 cells. Yogurt produced with selected bacteria and GW had the most valuable anti-coli effect against E. coli 0157: H7 and E. coli ATCC 35218 (American Type Culture Collection, Rockville, Maryland) (4.81 ± 0.62 and 5.64 ± 0.29 CFU mL-1, respectively). Although the added GW increased the yogurt functionality, it had a slightly negative effect on the taste and texture of the partially modified recipe. Ethnobotanical and ethnopharmacological studies of many Centaurea species indicated their potential in folk medicine so far. However, investigations of different Centaurea calcitrapa L. extracts in terms of cytotoxicity and antimicrobial activity against phytopathogens are generally scarce. The phenolic profile and broad antimicrobial activity (especially towards bacterial phytopathogens) of methanol (MeOH), 70% ethanol (EtOH), ethyl-acetate (EtOAc), 50% acetone (Me2CO) and dichloromethane: methanol (DCM: MeOH, 1: 1) extracts of C. calcitrapa leaves and their potential toxicity on MRC-5 cell line were investigated for the first time. A total of 55 phenolic compounds were identified: 30 phenolic acids and their derivatives, 25 flavonoid glycosides and aglycones. This is also the first report of the presence of centaureidin, jaceidin, kaempferide, nepetin, flavonoid glycosides, phenolic acids and their esters in C. calcitrapa extracts. The best results were obtained with EtOAc extract with lowest MIC values expressed in µg/mL ranging from 13 to 25, while methicillin resistant Staphylococcus aureus was the most susceptible strain. The most susceptible phytopathogens were Pseudomonas syringae pv. syringae, Xanthomonas campestris pv. campestris and Agrobacterium tumefaciens. The highest cytotoxicity was recorded for EtOAc and Me2CO extracts with the lowest relative and absolute IC50 values between 88 and 102 µg/mL, while EtOH extract was the least toxic with predicted relative IC50 value of 1578 µg/mL. Our results indicate that all tested extracts at concentration considered as non-toxic can be one of great importance in combat towards phytopathogenic and human pathogenic strains, as well as natural sources of antimicrobials. Analytic thinking is reliably associated with lower belief in conspiracy theories. However, evidence for whether increasing analytic thinking can reduce belief in conspiracies is sparse. As an exception to this, Swami et al. (2014) showed that priming analytical thinking through a verbal fluency task (i.e., scrambled sentence task) or a processing fluency manipulation (i.e., difficult-to-read fonts) reduced belief in conspiracy theories. To probe the robustness of these effects, in this Registered Report, we present two highly powered (i.e., 95%) direct replications of two of the original studies (i.e., Studies 2 and 4). We found no evidence that priming analytic thinking through the scrambled sentence task (N = 302), nor the difficult-to-read fonts (N = 488) elicited more analytic thinking, nor reduced belief in conspiracy theories. This work highlights the need for further research to identify effective ways of inducing analytic thinking in order to gauge its potential causal impact on belief in conspiracies. Resorting to complementary/alternative medical (CAM) therapies can lead to bad health outcomes or interfere with officially recommended therapies. CAM use is, nevertheless, widespread and growing. This could be partially due to the perception of the CAM industry as powerless and non-profit oriented, in contrast to the pharmaceutical industry ("Big Pharma"). In reality, both industries are highly profitable and powerful; to highlight this similarity, science communicators coined the term "Big Suppla". Drawing from a sample of 242 participants upon all exclusions, we experimentally tested whether varying these attributes in presenting the industries impacts consumers' evaluation of the two categories of products (herbs and supplements) and their willingness to try and recommend them. We also tested whether the effect is moderated by conspiratorial thinking, and whether it is due to a change in trust. All hypotheses were pre-registered. As expected, participants who read the Big Suppla vignette decreased the endorsement of both supplements and herbs, whilst, against our hypotheses, there were no significant changes in endorsement in the contrasting "Baby Suppla" group. Conspiratorial thinking was related to more endorsement of CAM, but it did not moderate the experimental effects. We also did not observe the expected mediation by trust. Our most robust results corroborate the idea that challenging the myth of benevolence of the CAM industry makes people more critical in evaluating its products or considering their usage. [Display omitted] • Spent mushroom substrate (SMS) was hydrothermally carbonized. • The HHV, fuel ratio, and energy densification of hydrochars were upgraded. • The combustion kinetics and thermodynamic of SMS and hydrochars were tested. • The average activation energy was lowest for the SMS-180. • The SMS-180 showed the best overall combustion behavior. Hydrothermal carbonization (HTC) was employed for the conversion of the spent mushroom substrate (SMS) into a carbonaceous hydrochar. The effect of operating temperature (180, 200, 220, 240, and 260 °C) on the physicochemical, structural, and combustion properties of the obtained hydrochars was analyzed. The HTC treatment caused the increase of the higher heating value (HHV) and the lower heating value (LHV) of hydrochars for 58 % and 65 % in comparison with SMS, respectively. Analysis of morphology and functional groups showed the formation of microspheres and cracks on the hydrochar surface, which are predominantly dominated by aromatic and oxygen-rich functional groups. Thermal and kinetics analysis showed that HTC treatment improves the combustion behavior of the obtained solids. Combustion kinetic parameters of SMS and hydrochars were determined by the methods of Kissenger-Akahira-Sunose (KAS) and Flynn-Wall-Ozawa (FWO). Abstract A new biosorbent – alginate encapsulated with Myriophyllum spicatum – MsA was investigated for lead ions removal. This biosorbent was characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), zeta potential, X ray Diffraction (XRD) and size distribution analysis. FT-IR analysis demonstrated that the lead ions sequestration mechanism included ion exchange and lead complexation with the carboxyl, carbonyl and hydroxyl groups in MsA. In order to better understand the mechanisms of the binding of Pb(II) on immobilized M. spicatum beads, 3 reaction and one diffusion based kinetic models were applied on kinetic data removal lead ions on three materials: M. spicatum, Ca-alginate and MsA. Myriophyllum spicatum encapsulated with alginate – MsA have higher adsorption capacity than M. spicatum. Among examined six isotherms Redlich-Peterson and the Langmuir isotherm model exhibited the best fit to the experimental data, with capacities ranging from 230 to 268.7 mg/g. Among the various tested desorption agents, nitric acid has proven to be the best. The obtained results suggest that the immobilized M. spicatum biosorbent holds great potential for lead wastewater treatment applications. The health-promoting effects of berries have attracted attention due to the possible application of their extracts as functional ingredients in food products. Natural deep eutectic solvents (NADESs) are a new generation of environmentally friendly solvents for the extraction of natural products, and they are green alternatives to organic solvents, and they can improve the solubility, stability, and bioavailability of isolated biocompounds. In this study, an efficient eco-friendly method was used for the extraction of phenolic compounds from different berries: chokeberries, blueberries, and black goji berries with a range of eutectic solvents consisting of hydrogen bond acceptors (HBAs) such as choline chloride, L-proline, L-glycine, and L-lysine and hydrogen bond donors (HBDs) such as malic, citric, tartaric, lactic and succinic acids, glucose and glycerol. The obtained results indicated the ability of NADESs towards selective extraction of phenolics; the eutectic system choline chloride : malic acid showed selective extraction of anthocyanins, while choline chloride : glycerol and choline chloride : urea showed selectivity towards flavonoids and phenolic acids. The methodology for screening of the NADES extraction performance, which included chromatographic profiling via high-performance thin layer chromatography combined with chemometrics and spectrophotometric essays, allowed effective assessment of optimal eutectic solvents for isolation of different groups of phenolics. Great antioxidant and antimicrobial activities of extracts, along with the green nature of eutectic solvents, enable NADES berry extracts to be used as "green-labelled" functional foods or ingredients.

Endoplasmic reticulum (ER) stress, prompted by the accumulation of misfolded or unfolded proteins, triggers the activation of the unfolded protein response (UPR) pathway to restore ER homeostasis. This stress response is implicated in the development of hepatocellular carcinoma (HCC). A biallelic mutation in SPRTN is currently the only known single-gene mutation implicated in the early onset of HCC. However, the exact mechanism linking SPRTN mutations to HCC remains unclear. In our study, we analyzed SPRTN and UPR in 21 human HCC tissue samples using RT-qPCR, immunoblot, and immunohistochemistry. We found alterations in the expression levels of SPRTN and UPR-related genes and proteins in HCC samples. The impact of SPRTN on the ER stress response was assessed in SPRTN-depleted HepG2 cells through RNA sequencing, pull-down assay, comet assay, and mitotic index calculation. We demonstrated that SPRTN interacts with the UPR sensor GRP78. Furthermore, we observed a decrease in SPRTN levels during ER stress, and increased sensitivity to ER stress in SPRTN-depleted cells. These findings suggest an essential role for SPRTN in the ER stress response and provide new insights into HCC pathogenesis. Scientific evidence regularly guides policy decisions 1 , with behavioural science increasingly part of this process 2 . In April 2020, an influential paper 3 proposed 19 policy recommendations ('claims') detailing how evidence from behavioural science could contribute to efforts to reduce impacts and end the COVID-19 pandemic. Here we assess 747 pandemic-related research articles that empirically investigated those claims. We report the scale of evidence and whether evidence supports them to indicate applicability for policymaking. Two independent teams, involving 72 reviewers, found evidence for 18 of 19 claims, with both teams finding evidence supporting 16 (89%) of those 18 claims. The strongest evidence supported claims that anticipated culture, polarization and misinformation would be associated with policy effectiveness. Claims suggesting trusted leaders and positive social norms increased adherence to behavioural interventions also had strong empirical support, as did appealing to social consensus or bipartisan agreement. Targeted language in messaging yielded mixed effects and there were no effects for highlighting individual benefits or protecting others. No available evidence existed to assess any distinct differences in effects between using the terms 'physical distancing' and 'social distancing'. Analysis of 463 papers containing data showed generally large samples; 418 involved human participants with a mean of 16,848 (median of 1,699). That statistical power underscored improved suitability of behavioural science research for informing policy decisions. Sugar beet is the most important crop for sugar production in temperate zones. The plant microbiome is considered an important factor in crop productivity and health. Here, we investigated the bacterial diversity of seeds, roots, and rhizosphere of five sugar beet hybrids named Eduarda (ED), Koala (KO), Tibor (T), Tajfun (TF), and Cercospora-resistant (C). A culture-independent next-generation sequencing approach was used for the further investigation of seed-borne endophytes. Hybrid-associated bacteria were evaluated for their plant growth-promoting (PGP) characteristics, antagonistic activity towards Cercospora beticola and several Fusarium strains in dual culture assays, and drought and salinity tolerance. High-throughput sequencing revealed that the Proteobacteria phylum was most dominant in the seeds of all hybrids, followed by Cyanobacteria and Actinobacteriota. The predominant genus in all hybrids was Pantoea, followed by Pseudomonas, Acinetobacter, Chalicogloea, Corynebacterium, Enterobacter, Enterococcus, Glutamicibacter, Kosakonia, and Marinilactibacillus. Unique genera in the hybrids were Pleurocapsa and Arthrobacter (T), Klebsiella (TF), Apibacter (ED), and Alloscardovia (KO). The genera that were most represented in one hybrid were Weissella and Staphylococcus (TF); Streptococcus (T); Gardnerella, Prevotella, and Rothia (KO); and Gilliamella, Lactobacillus, and Snodgrassella (ED). Thirty-two bacteria out of 156 isolates from the rhizosphere, roots, and seeds were selected with respect to various plant growth-promoting activities in vitro, i.e., nitrogen fixation, phosphate solubilization, siderophore production, indole-3-acetic acid production, 1-aminocyclopropane-1-carboxylic acid deaminase activity, hydrogen cyanide production, exoenzymatic activity (amylase, protease, lipase, cellulase, xylanase, mannanases, gelatinase, and pectinase), mitigation of environmental stresses, and antifungal activity. Mixta theicola KO3-44, Providencia vermicola ED3-10, Curtobacterium pusillum ED2-6, and Bacillus subtilis KO3-18 had the highest potential to promote plant growth due to their multiple abilities (nitrogen fixation, phosphate solubilization, production of siderophores, and IAA). The best antagonistic activity towards phytopathogenic fungi was found for Bacillus velezensis C3-19, Paenibacillus polymyxa C3-36 and Bacillus halotolerans C3-16/2.1. Only four isolates B. velezensis T2-23, B. subtilis T3-4, B. velezensis ED2-2, and Bacillus halotolerans C3-16/2.1 all showed enzymatic activity, with the exception of xylanase production. B. halotolerans C3-16/2.1 exhibited the greatest tolerance to salinity, while two B. subtilis strains (C3-62 and TF2-1) grew successfully at the maximum concentration of PEG. Background: This article presents a detailed analysis of a dataset consisting of 27 elements found in soils, soil eluates, and vegetables from private gardens in a region with a long history of coal mining and burning. With coal being one of the world's most significant energy sources, and previous studies highlighting elevated element levels in vegetables from this region, the objective of this study was to identify the factors that impact soil geochemistry and metal(loid) uptake in plants., Methods: Total major and trace element concentrations were analyzed in soils, soil eluates and vegetables by high resolution inductively coupled plasma mass spectrometry. The vegetable samples included six species: fennel, garlic, lettuce, parsley, onion, and radicchio. Each plant was divided into roots, stems, leaves, and/or bulbs and analyzed separately. In addition, the soil pollution status, bioavailable fractions and transfer factors from soil and soil eluates to different plant parts were determined., Results: The comprehensive dataset revealed that, apart from the substrate enriched with various elements (Al, As, Co, Cr, Mo, Ni, Pb, Sb, Sn, Ti, U, V, and Zn), other anthropogenic factors such as the legacy of coal mining and combustion activities, associated industries in the area, transport, and agricultural practices, also influence the elevated element concentrations (Cd, Cu, Fe, Mn, and Se) in locally grown vegetables. The transfer factors based on element concentrations in aqueous soil eluates and element bioavailable fractions confirmed to be an effective tool for evaluating metal uptake in plants, emphazising to some extent the effects of plant species and revealing unique patterns for each pollution source within its environmental context ( e.g. , Cd, Mo, S, and Se in this case). Health care policies often rely on public cooperation, especially during a health crisis. However, a crisis is also a period of uncertainty and proliferation of health-related advice: while some people adhere to the official recommendations, others tend to avoid them and resort to non-evidence based, pseudoscientific practices. People prone to the latter are often the ones endorsing a set of epistemically suspect beliefs, with two being particularly relevant: conspiratorial pandemic-related beliefs, and the appeal to nature bias regarding COVID-19 (i.e., trusting natural immunity to fight the pandemic). These in turn are rooted in trust in different epistemic authorities, seen as mutually exclusive: trust in science and trust in the "wisdom of the common man". Drawing from two nationally representative probability samples, we tested a model in which trust in science/wisdom of the common man predicted COVID-19 vaccination status (Study 1, N = 1001) or vaccination status alongside use of pseudoscientific health practices (Study 2, N = 1010), through COVID-19 conspiratorial beliefs and the appeal to nature bias regarding COVID-19. As expected, epistemically suspect beliefs were interrelated, related to vaccination status, and to both types of trust. Moreover, trust in science had both a direct and indirect effect on vaccination status through both types of epistemically suspect beliefs. Trust in the wisdom of the common man had only an indirect effect on vaccination status. Contrary to the way they are typically portrayed, the two types of trust were unrelated. These results were largely replicated in the second study, in which we added pseudoscientific practices as an outcome; trust in science and the wisdom of the common man contributed to their prediction only indirectly, through epistemically suspect beliefs. Economic inequality is associated with preferences for smaller, immediate gains over larger, delayed ones. Such temporal discounting may feed into rising global inequality, yet it is unclear whether it is a function of choice preferences or norms, or rather the absence of sufficient resources for immediate needs. It is also not clear whether these reflect true differences in choice patterns between income groups. We tested temporal discounting and five intertemporal choice anomalies using local currencies and value standards in 61 countries (N = 13,629). Across a diverse sample, we found consistent, robust rates of choice anomalies. Deposits remained after coal combustion are a well-known occurrence in the world; unfortunately, only a small percentage of such deposits are adequately regulated and, consequently, pose a serious threat to the local environment. Attenuation of negative consequences presupposes knowledge of a number of features, both of the deposit and the local environment as well the interaction with local biota. In this study, unregulated waste generated from decades of coal mining and combustion of superhigh-organic-sulfur Raša coal, enriched in Se-U-Mo-V and located in a vulnerable karst region, was investigated. To assess the impact of landfill on the environment, in addition to its general geochemical and mineralogical features, the human health risk was assessed and the leaching of elements from the landfill, local soil, and the coal itself was investigated. For the latter, three extraction procedures, ASTM, EP, and TCLP (pH 4.93 and 2.9), were employed, mimicking different environmental conditions, including the sporadic occurrence of acid rains in the region. The soil around the landfill displayed enrichment in the majority of elements compared to expected values, with exception of Se, Mo, U, V, Sr, and Cu found at the highest levels in landfill samples. Mobility of elements was found to be controlled by both pH and mineralogy (carbonates and sulfates), whereby the overall highest relative mobility was observed in landfill samples for elements prevalently bound to sulfate phases. Pervading global narratives suggest that political polarization is increasing, yet the accuracy of such group meta-perceptions has been drawn into question. A recent US study suggests that these beliefs are inaccurate and drive polarized beliefs about out-groups. However, it also found that informing people of inaccuracies reduces those negative beliefs. In this work, we explore whether these results generalize to other countries. To achieve this, we replicate two of the original experiments with 10,207 participants across 26 countries. We focus on local group divisions, which we refer to as fault lines. We find broad generalizability for both inaccurate meta-perceptions and reduced negative motive attribution through a simple disclosure intervention. We conclude that inaccurate and negative group meta-perceptions are exhibited in myriad contexts and that informing individuals of their misperceptions can yield positive benefits for intergroup relations. Drug resistance of Pseudomonas aeruginosa is a leading problem in hospital infections. The aim of this study was to determine the best molecular genetic discrimination method for Pseudomonas spp. isolates among 94 outpatients and inpatients and see their grouping by phenotype characteristics (biofilm formation, frequency of serotypes, pigmentation, production of different class of beta-lactamases, and susceptibility to different antibiotic classes) and genotype. The most common serotypes were P1, P6, and P11, while co-productions of pyoverdine and pyocyanin were observed in 70 % of isolates. A total of 77.66 % isolates were mostly weak and moderate biofilm producers. Isolates were susceptible to colistin (100 %), aztreonam (97.87 %), imipenem (91.49 %), doripenem (90.43 %), and meropenem (84.04 %). MICs values confirmed susceptibility to ceftazidime and cefepime and singled out meripenem as the most effective inhibitor. Most isolates were resistant to aminoglycosides and fluoroquinolones. Only two isolates produced ESBL, eight were carbapenemase producers, and five isolates produced MBLs. Twenty-nine isolates were multidrug-resistant; 82.8 % of which produced both pigments, 58.3 % were non-typeable, while the P6 and P11 serotypes were equally distributed (16.7 %). Thirteen MDR isolates were strong enzyme producers. RAPD PCR analysis using primer 272 proved the best at discriminatory fingerprinting for Pseudomonas isolates, as it allocated 12 clusters. A correlation between DNA patterns and antibiotic resistance, production of pigments, serotypes distribution, and biofilm formation was not observed, and only confirmed higher genetic heterogeneity among P. aeruginosa isolates, which suggests that other molecular methods are needed to reveal potential relations between genotypic patterns and phenotypic characteristics.

Respiratory-volume monitoring is an indispensable part of mechanical ventilation. Here we present a new method of the respiratory-volume measurement based on a single fibre-optical long-period sensor of bending and the correlation between torso curvature and lung volume. Unlike the commonly used air-flow based measurement methods the proposed sensor is drift-free and immune to air-leaks. In the paper, we explain the working principle of sensors, a two-step calibration-test measurement procedure and present results that establish a linear correlation between the change in the local thorax curvature and the change of the lung volume. We also discuss the advantages and limitations of these sensors with respect to the current standards.
