Abstract

Previous investigations have shown that Burkholderia pseudomallei secretes protease, lipase and phospholipase C into the extracellular milieu, but their mechanisms of secretion and roles in pathogenesis were not clear. B. pseduomallei 1026b transposon mutants deficient in secretion of protease, lipase and phospholipase C have been isolated and characterized. The relative virulence of 1026b and DD213, a representative secretion mutant, in the Syrian hamster model of melioidosis, were compared. Findings showed that exoproducts secreted by the type II pathway may serve a minor role in B. pseudomallei pathogenesis.

Abstract

Graphene foils improve angular and energy resolution in neutral atom detectors while also improving mass discrimination and usable energy ranges. We developed improved grid supports and achieved areas >10 cm2 with good foil coverage and significant improvements in secondary electron yield from ~1 nm metal oxide overcoats. We present Luxel’s characterization of large-area graphene foils for applications as transmission filters and detector components.

Abstract

The mutagenesis of Burkholderia pseudomallei with Tn5-OT182 was analyzed to identify the multiple genes that are involved in motility including the flagellin structural gene, fliC. Genetic analysis of the gram-negative bacillus via transposon mutagenesis with Tn5-OT182 indicated the presence of unique motility mutants that contained Tn5-OT182 integrations in a variety of genes encoding chemotaxis and flagellar proteins. However, there were no significant differences in the virulence of fliC-mutants and wild-type Burkholderia pseudomallei.

Abstract

This study examined the genetic and environmental architecture of early gambling involvement and substance use to determine whether genetic or environmental factors that contribute to substance use also put young adolescents at risk for early involvement in gambling. Self-reports of substance use and gambling involvement were collected at age 13 years from 279 Monozygotic and Dizygotic twin pairs. Univariate ACE modeling revealed that genetic and nonshared environmental factors almost equally accounted for gambling involvement, with no contribution from shared environmental factors. In contrast, both shared and nonshared environmental factors played important roles in substance use; the contribution of genetic factors was also substantial. Bivariate analyses identified a significant, albeit modest, overlap between the genetic influence on gambling involvement and the genetic influence on substance use. The results shed light on the etiology of early gambling involvement and substance use, suggesting that preventive interventions targeting common risk factors may also need to be complemented by modules that are specific to each behavior.

Abstract

This investigation examines the spread of problem behaviors (substance use and delinquency) between twin siblings. A sample of 628 twins (151 male twin pairs and 163 female twin pairs) drawn from the Quebec Newborn Twin Study completed inventories describing delinquency and substance use at ages 13, 14, and 15. A 3-wave longitudinal actor-partner interdependence model (APIM) identified avenues whereby problem behaviors spread from one twin to another. Problems did not spread directly between twins across domains. Instead, 2 indirect pathways were identified: (a) Problems first spread interindividually (between twins) within a behavioral domain, then spread intraindividually (within twins) across behavioral domains (e.g., Twin A delinquency → Twin B delinquency → Twin B substance use); and (b) problems first spread intraindividually (within twins) across behavioral domains, then spread interindividually (between twins) within a behavioral domain (e.g., Twin A delinquency → Twin A substance use → Twin B substance use). Controls for genetic effects, gene-environment correlations, friend substance use and delinquency, and parenting behaviors increase confidence in the conclusion that twin siblings uniquely contribute to the spread of problem behaviors during adolescence. Twin sibling influence is a risk factor for illicit substance use, both because substance use by one twin predicts substance use by the other twin, but also because delinquency in one twin predicts delinquency in the other twin, which then gives rise to greater substance use.

Melioidosis is a severe disease that can be difficult to diagnose because of its diverse clinical manifestations and a lack of adequate diagnostic capabilities for suspected cases. There is broad interest in improving detection and diagnosis of this disease not only in melioidosis-endemic regions but also outside these regions because melioidosis may be underreported and poses a potential bioterrorism challenge for public health authorities. Therefore, a workshop of academic, government, and private sector personnel from around the world was convened to discuss the current state of melioidosis diagnostics, diagnostic needs, and future directions.

Abstract

Several candidates for a vaccine against Burkholderia pseudomallei, the causal bacterium of melioidosis, have been developed, and a rational approach is now needed to select and advance candidates for testing in relevant nonhuman primate models and in human clinical trials. Development of such a vaccine was the topic of a meeting in the United Kingdom in March 2014 attended by international candidate vaccine developers, researchers, and government health officials. The focus of the meeting was advancement of vaccines for prevention of natural infection, rather than for protection from the organism's known potential for use as a biological weapon. A direct comparison of candidate vaccines in well-characterized mouse models was proposed. Knowledge gaps requiring further research were identified. Recommendations were made to accelerate the development of an effective vaccine against melioidosis.
