Results from ion‐mobility (IM) separation experiments demonstrate that O‐ and N‐protomers of p‐(dimethylamino)chalcone (p‐DMAC) can coexist in the gas phase. The relative populations of the two protomers strongly depend on the ion‐generating settings and the conditions the precursor ions experience from the point of their gas‐phase inception to the time of their detection. Under relatively dry source conditions, the ratio of the gas‐phase protomers generated under helium‐plasma ionization (HePI) conditions is biased towards the thermodynamically favored O‐protomer. However, when the humidity of the enclosed ion source was increased, the IM arrival‐time distribution profile of the mass‐selected protonated precursor of p‐DMAC changed rapidly to one dominated by the N‐protomer. Under spray‐ionization conditions, the formation of the thermodynamically less favored protomer has been generally attributed to a phenomenon called kinetic trapping. Herein, we demonstrate that the population of thermodynamically less favored N‐protomer can be dramatically increased simply by introducing water vapor to the HePI ion source. [ABSTRACT FROM AUTHOR]

Protonated methyl benzoate, upon activation, fragments by three distinct pathways. The m/z 137 ion for the protonated species generated by helium-plasma ionization (HePI) was mass-selected and subjected to collisional activation. In one fragmentation pathway, the protonated molecule generated a product ion of m/z 59 by eliminating a molecule of benzene (Pathway I). The m/z 59 ion (generally recognized as the methoxycarbonyl cation) produced in this way, then formed a methyl carbenium ion in situ by decarboxylation, which in turn evoked an electrophilic aromatic addition reaction on the benzene ring by a termolecular process to generate the toluenium cation (Pathway II). Moreover, protonated methyl benzoate undergoes also a methanol loss (Pathway III). However, it is not a simple removal of a methanol molecule after a protonation on the methoxy group. The incipient proton migrates to the ring and randomizes to a certain degree before a subsequent transfer of one of the ring protons to the alkoxy group for the concomitant methanol elimination. The spectrum recorded from deuteronated methyl benzoate showed two peaks at m/z 105 and 106 for the benzoyl cation at a ratio of 2:1, confirming the charge-imparting proton is mobile. However, the proton transfer from the benzenium intermediate to the methoxy group for the methanol loss occurs before achieving a complete state of scrambling.ᅟ [ABSTRACT FROM AUTHOR]

Embedded in the tail base of all snakes is a pair of scent glands that open through ducts at the margin of the cloaca. Scent gland secretions (SGS), which typically are discharged when snakes are disturbed, are thought to deter predators. Previous chemical analyses have indicated carboxylic acids as the chief lipid constituents of SGS. We examined by GC-MS the SGS of the eastern small-scaled burrowing asp (Atractaspis fallax), a venomous, fossorial species in the Lamprophiidae, a diverse family containing more than 60 genera of mostly non-venomous African snakes. Our results confirm that acetic, propionic, 2-methylpropanoic, n -butanoic, and 3-methylbutanoic acids are the major constituents in the SGS samples of A. fallax. These compounds are widespread in the SGS of the Alethinophidia ('typical snakes'). Some of these short-chain acids also occur in the anal glands of mammals and the uropygial (preen) glands of birds, where they arise via microbial activity. Whether these compounds in SGS have a similar origin is unknown. In an enclosed atmospheric-pressure helium-plasma ionization (HePI) source engulfed with dehumidified ambient gases, molecular cations are generated from compounds such as toluene, bromobenzene, and iodobenzene. Evidently, the ionization is effected by a direct Penning mechanism attributable to interactions of the gas-phase analyte with metastable helium atoms. It is widely known that secondary ions generated from ambient gases also play an important role in the overall ionization process. For example, when the ambient gases bear even traces of moisture, the analytes are ionized by proton transfer reactions with gaseous HO. In this study, we demonstrate how a controlled variation of experimental conditions can manipulate the abundance of molecular ions and protonated molecules in a HePI source. Gas-phase addition of dioxygen to certain ions is a well-known phenomenon in mass spectrometry. For this reaction to occur, the presence of a distonic radical site on the precursor ion is thought to be a prerequisite. Herein, we report that oxygen adduct formation can take place also with deprotonated hydroquinone, which in fact is an even-electron species without a radical site. When the product-ion spectrum of the m/z 109 ion, generated by electrospray ionization from a solution of hydroquinone in acetonitrile, was recorded under ion-mobility conditions, a new peak was observed at m/z 141. However, an analogous peak was not visible in the spectrum acquired under nonmobility conditions (i.e., without any gas introduced to the mobility cell). Presumably, traces of oxygen present in the collision gas instigate an ion–molecule reaction to produce an adduct of m/z 141, which upon activation results in CO and H2O loss to form a product ion of m/z 95. Isotope-labeling studies confirmed that one of the hydrogen atoms from the hydroxy group and another from the aromatic ring contribute to the water loss instigated from the m/z 141 adduct. Furthermore, computational methods indicated the three-dimensional structure of the ground-state deprotonated hydroquinone to be distinctly different from those of its 1,2- and 1,3-isomers. Calculations predicted that all atoms in the two m/z 109 ions generated from catechol and resorcinol lie on one plane. In contrast, the structure of the m/z 109 ion from hydroquinone was significantly different. Computations predicted that the hydrogen atom on the intact hydroxyl group of deprotonated hydroquinone protrudes out of plane from rest of the atoms. Consequently, the exposed OH group can interact with an incoming dioxygen molecule. Computations conducted at the CAM-B3LYP/6-311++g-(2d,2p) level of theory detected a minimum energy crossing point (MECP) at −4.3 kJ mol–1 below the separated O2 + deprotonated hydroquinone triplet threshold. In contrast, similar calculations conducted for catechol and resorcinol yielded MECPs of +116.9 and +69.1 kJ mol–1, respectively, above the associated triplet thresholds. These results indicated that the curve crossing required to form singlet products upon reaction with triplet O2 is favorable in the case of hydroquinone and unfavorable in the cases of catechol and resorcinol. The ortho, meta, and para isomers of hydroxybenzyl alcohol can be unequivocally distinguished by the collision‐induced dissociation mass spectra of their anions. The presence of a prominent peak at m/z 121 for an elimination of a dihydrogen molecule renders the ortho‐isomer spectrum markedly different from those of its meta and para congeners. Investigations carried out with deuterium‐labeled isotopologues of the ortho isomer verified that the labile hydrogen atom on the hydroxyl group and one of the benzylic hydrogen atoms are specifically removed in the formation of the m/z 121 ion. The ortho‐isomer spectrum also showed a prominent peak at m/z 93. Experimental data indicated that the m/z 93 product ion originates either from a two‐step H2and CO elimination mechanism or from a direct loss of a HCHO molecule from the precursor anion. The intensity ratio of the m/z 93 and 94 peaks in the spectrum recorded from the m/z 124 ion generated from a sample of o‐hydroxybenzyl alcohol dissolved in D2O supported the notion that the direct HCHO loss is the more dominant pathway for the generation of the phenolate ion under low activation conditions. In contrast, the two‐step mechanism becomes the more dominant pathway under high collisional activation conditions. The spectrum also showed a weak peak at m/z 105 for a water loss. Based on computational data, the m/z 105 ion generated in this way appears to be a composite generated from a common ion‐neutral complex intermediate in which a hydroxyl anion is positioned equidistantly between one of the benzylic hydrogens and a nearby hydrogen atom of the benzene ring. Upon activation, the complex dissociates to form either a phenide or a quinone methide anion. Abstract: Tailoring the surfaces of a nanocontainer with polymer brushes that have different affinities to the components of a phase-separating polymer blend should impart self-directing properties to the nanocontainers. Such nanocontainers could then be used to deliver a variety of functional species in tunable amounts and in a site-specific manner to polymer systems. This paper describes the surface modification, subsequent characterization of nanocontainers derived from ferritin, and the effects of surface modification on their self-directing properties in a binary phase-separating homopolymer blend. Wild ferritin was either PEGylated or alkylated by zero-length cross-linking to its surface carboxylate groups that were activated by carbodiimide. Modification was confirmed by ion-exchange chromatography, ζ-potential measurement, and electrospray ionization mass spectrometry. FT-IR spectrometry was used to quantify the extent of PEGylation by ratioing the intensity of the C–O–C asymmetric stretching vibration from the grafted PEG to that of the carbonyl stretching vibration (amide I band) from the protein. Importantly, modified ferritin was soluble in the organic solvent dichloromethane (DCM). Modified ferritin was introduced into a polymer blend of hydrophobic and hydrophilic polymers made up of poly(desaminotyrosyl tyrosine dodecyl ester carbonate) (PDTD) and PEG by solvent casting from solution in the common solvent DCM. Polymer thin films with an average thickness of ∼200μm were obtained upon evaporation of the solvent. Transmission electron micrographs of microtomed polymer films demonstrated remarkable selectivity of PEGylated ferritin to PEG domains, while alkylated ferritin self-directs to the PDTD matrix. Selected-ion recording (SIR) or multiple-reaction monitoring (MRM) protocols are widely employed for the quantification of targeted analytes by liquid chromatography-mass spectrometry (LC-MS). After chromatographic separation, analytes are desolvated and converted to gaseous ions usually by electrospray-ionization. The chromatographic peaks generated in this way are then integrated for quantification. It is generally assumed that the chromatographic peak intensities are dependent only on the selected MRM-transition protocols and the instrumental parameters set on the mass spectrometer. Using p-aminosalicylic acid (PAS) as a model compound, we demonstrate that the nature of the LC mobile phase exerts a significant effect on the chromatographic peak intensities. Under identical mass spectrometric conditions, chromatographic peak intensities recorded with methanol as the mobile phase were drastically different from those acquired using acetonitrile as the eluent. In fact, the product-ion mass spectra recorded with protonated PAS under different solvent conditions were qualitatively different. The observed differences were attributed to the existence of different protomers of PAS in the gas phase in dissimilar ratios under different solvent-spray conditions. Results from ion-mobility mass spectrometry experiments confirmed this hypothesis. For example, when PAS was sprayed from an acetonitrile solution, the arrival-time profile recorded from the mass-selected m/z 154 ion for protonated PAS showed essentially one arrival-time peak for the N-protonated tautomer. In contrast, the profile recorded from a methanolic PAS solution showed a different arrival-time peak for a more mobile protomer, which was recognized as the carbonyl-protonated PAS. The coexistence of protomers in different and variable ratios in an ensemble of ions generated by electrospray ionization of a single pure compound wields strong ramifications on the identification and quantification of analytes by LC-MS. However, the inclusion of an ion-mobility separator before the mass-selected ions are fragmented and detected by mass spectrometry ameliorates the complications rendered by the coexistence of different protomers and deprotomers. Hydrocyanic acid (HCN) is a well-known defensive allomone in the chemical arsenal of millipedes in the order Polydesmida. The presence of HCN in the headspace vapor of adult Xystocheir dissecta (Wood, 1867), a common millipede from the San Francisco Bay Area, was traced by laser desorption/ionization-mass spectrometry (LDI-MS). To accomplish this, the headspace vapor surrounding caged, live millipedes was allowed to diffuse passively over gold-nanoparticle (AuNP) deposits placed at various distances from the emitting source. The stainless steel plates with AuNP deposits were removed and irradiated by a 355-nm laser. The gaseous ions generated in this way were detected by time-of-flight mass spectrometry. The intensity of the mass spectrometric peak detected at m/z 249 for the Au(CN)2− complex anion was compared to that of the residual Au− signal (m/z 197). Using this procedure, HCN vapors produced by the live millipedes could be detected up to 50 cm away from the source. Furthermore, the addition of H2O2, as an internal oxygen source for the gold cyanidation reaction that takes place in the AuNP deposits, significantly increased the detection sensitivity. Using the modified H2O2 addition procedure, HCN could now be detected at 80 cm from the source. Moreover, we found a decreasing intensity ratio of the Au(CN)2−/Au− signals as the distance from the emitting source increased, following an exponential-decay distribution as predicted by Fick's law of diffusion. Previous investigations have evaluated the efficacy of anions such as NO3−, Cl−, Br−, CH3COO−, and CF3COO− as additives to generate or enhance mass spectrometric signals from explosives under plasma ionization conditions. The results of this study demonstrate that for detecting nitramine-class explosives, such as 1,3,5-trinitroperhydro-1,3,5-triazine (RDX) and 1,3,5,7-tetranitro-1,3,5,7-tetrazacyclooctane (HMX), 1,4-benzoquinone (BQ) is a highly effective and efficient dopant. When used in conjunction with ambient-pressure negative-ion helium-plasma ionization (HePI), 1,4-benzoquinone readily captures an electron, forming an abundant molecular anion (m/z 108), which upon exposure to vapors of RDX and HMX generates adduct ions of m/z 330 and 404, respectively. The signal level recorded for RDX upon adduction to the radical anion of 1,4-benzoquinone under our experimental conditions was significantly higher than that realized by chloride adduction using dichloromethane (DCM) as the dopant. Upon laser irradiation, sulfur extracts chemically bound copper from its compounds, and forms Cu x S y − clusters, supporting Goldschmidt's classification that copper is a chalcophile. Moreover, an array of Cu–S ion clusters (with m / z ratios extending to over 8000) are formed when a mixture of copper and sulfur is irradiated with UV light. Samples of elemental copper, upon laser irradiation under negative-ion LDI-MS conditions, do not generate noticeable signals for copper-bearing gaseous ions. In contrast, a several thousand-fold enhancement of the generated ion current was observed when freshly made mixtures of copper and sulfur powders were laser-ablated. Time-of-flight mass spectra recorded showed a range of peaks with m / z ratios extending to over m / z 8000, indicating the formation in situ of an array of Cu–S ion clusters by a chemical reaction that takes place between the two elements upon laser irradiation. Evidently, sulfur acts as a potent reactive matrix and generates at least three distinguishable series of ion clusters: Cu 1 S x − (x = 2, 3, 4...), Cu 2 S y − (y = 3, 4, 5...), and Cu 3 S z − (z = 3, 4, 5...). Laser ablation of CuS alone did not produce signals beyond m / z 600. Some of the high-mass ions formed in this manner were isobaric; consequently, composite peaks were observed under low-resolution MS conditions. Under high-resolution conditions, however, we were able to separate and determine the composition of some isobaric mixtures. For example, the peak recorded at m / z 287 was resolved to two peaks that represented primarily the 63Cu 2 65Cu32S 3 − and 63Cu32S 7 − ions. Intriguingly, the spectra recorded from many copper-containing minerals mixed with sulfur were remarkably similar to those acquired from copper-and-sulfur mixtures, which demonstrated that elemental sulfur has the ability to extract chemically bound copper even from its compounds, and form gas-phase Cu–S clusters of varying composition. Abstract Pyrazines are an important group of natural products widely used as food additives and fragrants. Gas chromatography-mass spectrometry (GC MS) is the most widely applied analytical technique for characterization of alkylpyrazines. However, mass spectra of many positional isomers of alkylpyrazines are very similar. Consequently, an unambiguous identification of each positional isomer by spectral interpretation or database search protocols is practically unfeasible. In fact, there are many misidentifications in literature. To identify alkylpyrazines, chemists often resort to gas chromatographic retention indices (RIs). Although there are many compilations of retention indices of alkylpyrazines, these databases are often incomplete and the values reported are sometimes inconsistent. Herein, we present retention indices of fifty-six alkylpyrazines recorded on DB-1, ZB-5MS, DB-624, and ZB-WAXplus stationary phases, and compare them with those available in the NIST-2017 MS-RI database. Furthermore, we demonstrate how RI values can be used, together with mass spectral interpretations, to identify certain alkylpyrazines unambiguously. [ABSTRACT FROM AUTHOR]

An ambient pressure ionization technique for mass spectrometric analysis of substances present on solid surfaces was developed. A nebulized spray containing molecular ions of a solvent such as toluene can be generated by passing the solvent through a stainless steel capillary held at a high voltage. When the stream of charged droplets produced in this way is directed onto a solid surface, the analytes present on the surface are desorbed and ionized by a charge exchange process. This technique was shown to desorb and generate positively charged molecular ions from compounds that are not readily ionized by some other ambient methods, under positive-ion generation mode. For example, intense signals representing radical cations of 1,4-hydroquinone, limonene, thymol, and several other compounds were observed when the analytes were deposited on a metal surface and exposed to a toluene spray nebulized from the metal capillary maintained at a potential of about +5 kV. In contrast, when the same samples were exposed to a spray of water/methanol/formic acid under customary DESI-like (positive-ion mode) conditions, no peaks representing the analytes were observed. The presence of a peak centered near m/z 2862, observed for the first time for the caged dodecatungstate radical-anion, [W12O41] −∙, enables distinguishing WO2 from WO3 by Laser Desorption Ionization mass spectrometry (LDI-MS). In addition to WO2, laser irradiation of dry deposits made from aqueous ammonium paratungstate, and calcium and lead orthotungstate also produce the [W12O41] −∙. In contrast, spectra recorded from deposits made from aqueous Na2WO4, sodium metatungstate, and WO3, or non-aqueous calcium and lead orthotungstate, and ammonium paratungstate, failed to show the m/z 2862 peak cluster. These observations support the hypothesis that polycondensation reactions to form [W12O41] −∙ occur solely in the presence of water. Although dry spots are irradiated for ionization, the solvent used for sample preparation plays an important role on the chemical composition endowed to ions detected. For example, the m/z 2862 peak seen from deposits made from aqueous ammonium paratungstate, and calcium and lead orthotungstate, is absent in the spectra recorded either from pristine deposits or those derived from solutions made with organic solvents such as acetonitrile or ethanol. The acceptance of Solanum surattenses as a host plant for the larvae of Manduca sexta was explained by the presence of feeding stimulants in foliage. Bioassay-guided fractionation of plant extracts resulted in the isolation of a highly active compound (1), which was identified as a furostan derivative {26-O-beta-D-glucopyranosyl-(25R)-furosta-5-ene-3-beta-yl-O-alpha-L-rhamnopyranosyl-(1''-2')-O-alpha-L-rhamnopyranosyl-(1'''-3'')-O-beta-D-glucopyranoside}. This compound has the same steroidal core substructure as that in a stimulant (indioside D) previously identified from potato foliage. However, the sugar substituents attached to the core are different.

The host plants of the native American butterfly, Pieris napi oleracea, include most wild mustards. However, garlic mustard, Alliaria petiolata, a highly invasive weed that was introduced from Europe, appears to be protected from this insect. Although adults will oviposit on the plant, most larvae of P. n. oleracea do not survive on garlic mustard. We used feeding bioassays with different larval stages of the insect to monitor the isolation and identification of two bioactive constituents that could explain the natural resistance of this plant. A novel cyanopropenyl glycoside (1), alliarinoside, strongly inhibits feeding by first instars, while a flavone glycoside (2), isovitexin-6"-D-beta-glucopyranoside, deters later instars from feeding. Interestingly, the first instars are insensitive to 2, and the late instars are little affected by 1. Furthermore, differential effects of dietary experience on insect responses suggest that 1 acts through a mechanism of post-ingestive inhibition, whereas 2 involves gustatory deterrence of feeding.

In wind-tunnel bioassays, dispensers loaded with 1 microg of the synthetic major component (3E,8Z, 11Z)-3,8,11-tetradecatrienyl acetate (TDTA) of the sex pheromone emitted by Tuta absoluta (Meyrick) females were found to be highly attractive to conspecific males. Field experiments were conducted to evaluate the efficacy of five trap designs. The best trap, baited with 100 microg of the synthetic sex pheromone caught on average 1,200 males per trap per night, while those baited with virgin females caught only 201 males. The male response to this pheromone is restricted to the same early-morning time window during which females exhibit calling behavior. The high biological activity of the synthetic pheromone suggests that it could be useful for pest monitoring and in mating disruption.

The bombardier beetle Metrius contractus discharges its defensive secretion as a froth that clings to its body. When attacked from the rear, it allows the froth to build up over the gland openings near the abdominal tip; when attacked from the front, it conveys the secretion forwards along special elytral tracks. M. contractus has two-chambered defensive glands typical of bombardier beetles, and its secretion, like that of other bombardiers, is quinonoid and hot. Its frothing mechanism, however, is unique for bombardiers and possibly illustrative of the ancestral glandular discharge mechanism of these beetles. M. contractus, thus, could be the least derived of extant bombardiers.

The African dwarf crocodile, Osteolaemus tetraspis (Crocodilidae, Reptilia), possesses a pair of skin glands, the paracloacal glands, the secretion of which is thought to be used to mark nest sites or attract mates. Ten aromatic steroidal esters were isolated from this secretion and characterized on the basis of NMR spectroscopic investigations, electrospray ionization-MS analyses, and chemical degradation. These esters, which account for more than 90% of the paracloacal glandular secretion, are derived from either cholesterol or cholestanol, esterified with a C-20 or C-22 acid closely related to dianeackerone, the only significant volatile compound found in this secretion.

The pupal defensive secretion of the 24-pointed ladybird beetle, Subcoccinella vigintiquatuorpunctata, consists of a mixture of macrocyclic polyamines, dominated by the three dimeric, 30-membered macrocycles 11-13, derived from the two building blocks 11-(2-hydoxyethylamino)-5-tetradecenoic acid (9) and 11-(2-hydoxyethylamino)-5,8-tetradecadienoic acid (10). Smaller amounts of the four possible cyclic trimers of 9 and 10 were also detected, corresponding to 45-membered macrocycles. Structural assignments were based on NMR-spectroscopic investigations and HPLC-MS analyses. In addition, the all-S absolute configuration of the S. vigintiquatuorpunctata macrocycles was determined by comparison of derivatives of the natural material with enantiomerically pure synthetic samples. Comparing this alkaloid mixture with that of the pupal defensive secretion in related ladybird beetle species indicates that the degree of oligomerization of the 2-hydroxyethylamino carboxylic acid building blocks can be carefully controlled by the insects.

The oily droplets on the pupal integumental hairs of the squash beetle Epilachna borealis contain a mixture of alpha-, beta-, gamma-, and delta-tocopheryl acetates as major constituents. In addition, the secretion contains a number of minor components that appear to be dehydrocongeners of the major components. This is the first report of the occurrence of acetate esters of any tocopherol in nature.
