Abstract

Network pharmacology method was adopted in this study to explore the active compounds and mechanism of Tongsaimai tablets for atherosclerosis. In molecular docking and molecular-target protein network analysis, 97 molecules in Tongsaimai tablets showed good interaction with the atherosclerosis-related target protein (docking score ≥ 7), and 37 molecules of them could act on more than 2 targets (≥ 2) with higher betweenness, suggesting that these 37 molecules might be the main active compounds group in Tongsaimai tablets for atherosclerosis treatment. Furthermore, the predicted active compounds contained more flavonoids and saponins, reminding more attention should be paid on flavonoids and saponins in study of effective compounds and quality standards of Tongsaimai tablets. Targets network analysis showed that, the active compounds of Tongsaimai tablets could regulate inflammation, stabilize plaque, protect vascular endothelial cell, regulate blood lipid and inhibit blood coagulation through acting on the main 22 target proteins, such as Toll-like receptors (TLR1, TLR2), matrix metalloproteinase (MMP1, MMP2, MMP3, MMP9), angiotensin converting enzyme (ACE), leukotriene A4 hydrolase (LTA4-H), 5-lipoxidase (5-LOX), peroxisome proliferators-activated receptors (PPARα, PPARγ). These active compounds can participate in regulating different pathologic stages of atherosclerosis and thus treat atherosclerosis finally. This study revealed the main active compounds and possible mechanism of Tongsaimai tablets for treatment of atherosclerosis and meanwhile, verified the characteristics of multi-components, multi-targets and integral regulation for Tongsaimai tablets, providing theoretical references for the following systematic laboratory experiments on effective compounds and action mechanism of Tongsaimai Tablet.

Abstract

A special beam line for high energy electron radiography is designed, including achromat and imaging systems. The requirement of the angle and position correction on the target from imaging system can be approximately realized by fine tuning the quadrupoles used in the achromat. The imaging system is designed by fully considering the limitation from the laboratory and beam diagnostics devices space. Two kinds of imaging system are designed and both show a good performance of imaging by beam trajectory simulation. The details of the beam optical requirement and optimization design are presented here. Background: This study was to investigate the health literacy and influencing factors of older population in pension institution of Xinjiang, China. Methods: Elderly people were selected from 44 pension institutions in Urumqi, Changji, Karamay and Shihezi and from September 2011 to June 2012 using random layer sampling method. The investigation was carried out by Chi-nese citizen health literacy questionnaire prepared by the China Health Education Center. Data were analyzed by One-way ANOVA, multiple linear regression and Pearson correlation analysis. Results: A total of 1396 elderly people met the inclusion criteria and their average age was (77.37 ± 8.48) years. Abstract

Background: Acute cerebrovascular disease (ACVD) could cause abnormal metabolism of thyroid hormones (TH), mostly represented as a euthyroid sick syndrome or low T3 syndrome. However, the changes in serum thyroid-stimulating hormone (TSH) are controversial. The aim of this study is to investigate the clinical significance of TSH alteration in patients with ACVD. Method: Patients with ACVD admitted in our hospitals between January 2013 and September 2017 were enrolled in this study (n = 245, including 176 cerebral infarctions and 69 cerebral hemorrhages). Their thyroid hormones were measured and compared with healthy individuals (n = 75). The correlation of TSH with severity and prognosis of ACVD were analyzed by receiver operating characteristic curve. Results: Serum TSH in ACVD group was higher than the control group (1.64 ± 1.08 vs. 1.26 ± 0.36 μIU/mL, P < 0.05). The TSH levels in intermediate and severe patients with ACVD were higher than in mild patients (1.72 ± 1.18 vs. 2.71 ± 0.93 vs. 1.02 ± 0.47 μIU/mL, P < 0.05). Receiver Operating Characteristic curve (ROC) of TSH in determining the severity of patients were 0.863 (Area under the curve, AUC), 1.496 μIU/L (optimal threshold), 76.5% (sensitivity) and 87.3% (specificity). TSH levels in improved and unchanged groups were significantly higher than the primarily healing group (2.27 ± 1.11 vs. 2.88 ± 1.07 vs. 0.86 ± 0.46 μIU/mL, P < 0.05). ROC of TSH in determining the prognosis of patients was 0.910 (AUC), 1.681 mIU/L (optimal threshold), 79.8% (sensitivity) and 90.5% (specificity) correspondingly. Conclusion: Since elevated TSH in ACVD patients affects the outcome of thyroid function evaluation, it is preferable to re-check after the acute period. A correlation between a high TSH level and the severity and prognosis of ACVD was detected, but the mechanism of this correlation needs to be further studied. [ABSTRACT FROM AUTHOR]

Abstract

Semi-supervised clustering is a new learning method which combines semi-supervised learning (SSL) and cluster analysis. It is widely valued and applied to machine learning. Traditional unsupervised clustering algorithm based on data partition does not need any property; however, there are a small amount of independent class labels or pair constraint information data samples in practice; in order to obtain better clustering results, scholars have proposed a semi-supervised clustering. Compared with traditional clustering methods, it can effectively improve clustering performance through a small number of supervised information, and it has been used widely in machine learning. Firstly, this paper introduces the research status and classification of semi-supervised learning and compares the four classification methods as follows: decentralized model, support vector machine, graph, and collaborative training. Secondly, the semi-supervised clustering is described in detail, the current status of semi-supervised clustering is analyzed, and the Cop-kmeans algorithm, Lcop-kmeans algorithm, Seeded-kmeans algorithm, SC-kmeans algorithm, and other algorithms are introduced. The introduction of several semi-supervised clustering methods in this paper can show the advantages of semi-supervised clustering over traditional clustering, and the related literature in recent years is summarized. This paper summarized the latest development of semi-supervised learning and semi-supervised clustering and discussed the application of semi-supervised clustering and the future research direction. Highlights • Coupling effect of mechanical load and environment load had influence on the durability performance of concrete structure. • Water and chloride penetration into concrete decreased firstly and then increased with the increase of applied load. • Applied load had an obvious influence on the frost resistance of concrete. • Coupling effect had more obvious influence on high water/ratio concrete. Abstract In cold coastal area, the destruction mechanism of reinforced concrete structures is mainly governed by a combination of factors such as self-loading, freeze–thaw and chloride erosion. In this study, ordinary cube concretes (C30 and C50, while w/c = 0.53 and 0.35 respectively) underwent a coupling effect of pressure load with stress ratio of 0, 0.3 and 0.5 and freeze–thaw cycles, following by capillary water absorption test and chloride penetration test. Concrete samples with 0.3 f c showed the best water and chloride penetration resistance under the coupling effect, followed by samples with 0.5 f c and 0 f c , which is consistent with the conclusion that under load only. Water and chloride ions penetration increased sharply when freeze–thaw cycles was over 100 times, which is different with samples without load. Outside part of concrete showed higher permeability and chloride content than inside part. MIP results confirmed that stress played an important role in the water absorption and chloride penetration of concrete under the coupling effect. These results provide important new insights into the permeability of concrete under a coupling effect. 587. Effects of aging on mechanical sensitivity threshold and thermal decomposition characteristic of RDX/HMX

Abstract

In order to analyze the influences of storage aging on the safety of typical elemental explosives, the aged cyclotrimethylene trinitramine (RDX) and cyclotetramethylene tetranitramine (HMX) were prepared by isothermal aging tests. The reaction thresholds of aged RDX and HMX under any ignition probability were studied by Langlie-Optimal D method. The thermal decomposition characteristics of RDX and HMX after aging were analyzed by DSC and ARC. Experimental results showed that compared with unaged RDX and HMX, on the one hand, the critical impact energy and critical friction of RDX and HMX aged for 14, 28 and 56 days are significantly reduced at an explosion probability of 50%, 0.01% and 0.0001%, respectively. With the increase of aging time, the mechanical sensitivity of RDX and HMX increases obviously. On the other hand, the initial decomposition temperature of RDX and HMX after 56 days of aging decreases, the decomposition heat decreases, the activation energy increases, and the reaction difficulty increases.

588. Precise Spectral Overlap-Based Donor–Acceptor Pair for a Sensitive Traffic Light-Typed Bimodal Multiplexed Lateral Flow Immunoassay

Abstract

Bimodal-type multiplexed immunoassays with complementary mode-based correlation analysis are gaining increasing attention for enhancing the practicability of the lateral flow immunoassay (LFIA). Nonetheless, the restriction in visually indistinguishable multitargets induced by a single fluorescent color and difficulty in single acceptor ineffectual fluorescence quenching due to the various spectra of multiple different donors impede the further execution of colorimetric–fluorescence bimodal-type multiplexed LFIAs. Herein, the precise spectral overlap-based donor–acceptor pair construction strategy is proposed by regulating the size of the nanocore, coating it with an appropriate nanoshell, and selecting a suitable fluorescence donor with distinct colors. By in situ coating Prussian blue nanoparticles (PBNPs) on AuNPs with a tunable size and absorption spectrum, the resultant APNPs demonstrate efficient fluorescence quenching ability, higher colloidal stability, remarkable colorimetric intensity, and an enhanced antibody coupling efficiency, all of which facilitate highly sensitive bimodal-type LFIA analysis. Following integration with competitive-type immunoreaction, this precise spectral overlap-supported spatial separation traffic light-typed colorimetric–fluorescence dual-response assay (coined as the STCFD assay) with the limits of detection of 0.013 and 0.152 ng mL–1for ractopamine and clenbuterol, respectively, was proposed. This work illustrates the superiority of the rational design of a precise spectral overlap-based donor–acceptor pair, hinting at the enormous potential of the STCFD assay in the point-of-care field.

589. Identification and characterization of long non-coding RNAs in mammary gland tissues of Chinese Holstein cows

Abstract

LncRNAs (Long non-coding RNA) is an RNA molecule with a length of more than 200 bp. LncRNAs can directly act on mRNA, thus affecting the expression of downstream target genes and proteins, and widely participate in many important physiological and pathological regulation processes of the body. In this study, RNA-Seq was performed to detect lncRNAs from mammary gland tissues of three Chinese Holstein cows, including three cows at 7 d before calving and the same three cows at 30 d postpartum (early lactation stage). A total of 1,905 novel lncRNAs were detected, 57.3% of the predicted lncRNAs are ≥ 500 bp and 612 lncRNAs are intronic lncRNAs. The exon number of lncRNAs ranged from 2 to 10. A total of 96 lncRNAs were significantly differentially expressed between two stages, of which 47 were upregulated and 49 were downregulated. Pathway analysis found that target genes were mainly concentrated on the ECM-receptor interaction, Jak-STAT signaling pathway, PI3K-Akt signaling pathway, and TGF-beta signaling pathway. Relevant studies in humans and model animals have shown that lncRNAs participate in mammalian mammary gland development and lactation, but there are few studies on lncRNAs regulation of mammary gland development and lactation in dairy cows. Therefore, this study aims to reveal the potential role of lncRNAs in the mammary gland of dairy cows through screening, identification, and functional research of differentially expressed lncRNAs at different periods of mammary gland development (pregnancy and early lactation period). It provides a reference for the follow-up study on the regulatory mechanism of dairy cows’ mammary gland health.

590. Fortified Dual-Spectral Overlap with Enhanced Colorimetric/Fluorescence Dual-Response Immunochromatography for On-Site Bimodal-Type Gentamicin Monitoring

Abstract

Immunochromatography (ICA) remains untapped toward enhanced sensitivity and applicability for fulfilling the nuts and bolts of on-site food safety surveillance. Herein, we report a fortified dual-spectral overlap with enhanced colorimetric/fluorescence dual-response ICA for on-site bimodal-type gentamicin (Gen) monitoring by employing polydopamine (PDA)-coated AuNPs (APDA) simultaneously serving as a colorimetric reporter and a fluorescence quencher. Availing of the enhanced colorimetric response that originated from the PDA layer, the resultant APDA exhibits less required antibody and immunoprobes in a single immunoassay, which facilitates improved antibody utilization efficiency and immuno-recognition in APDA-ICA. Further integrated with the advantageous features of fortified excitation and emission dual-spectral overlap for the Arg/ATT-AuNCs, this APDA-ICA with a “turn on/off” pattern achieves the visual limits of detection of 1.0 and 0.5 ng mL–1for colorimetric and fluorescence patterns (25- and 50-fold lower than standard AuNPs-ICA). Moreover, the excellent self-calibration and satisfactory recovery of 79.03–118.04% were shown in the on-site visual colorimetric–fluorescence analysis for Gen in real environmental media (including real river water, an urban aquaculture water body, an aquatic product, and an animal byproduct). This work provides the feasibility of exploiting fortified dual-spectral overlap with an enhanced colorimetric/fluorescence dual response for safeguarding food safety and public health.

591. Bioretention Design Modifications Increase the Simulated Capture of Hydrophobic and Hydrophilic Trace Organic Compounds

Abstract

Stormwater rapidly moves trace organic contaminants (TrOCs) from the built environment to the aquatic environment. Bioretention cells reduce loadings of some TrOCs, but they struggle with hydrophilic compounds. Herein, we assessed the potential to enhance TrOC removal via changes in bioretention system design by simulating the fate of seven high-priority stormwater TrOCs (e.g., PFOA, 6PPD-quinone, PAHs) with log KOCvalues between −1.5 and 6.74 in a bioretention cell. We evaluated eight design and management interventions for three illustrative use cases representing a highway, a residential area, and an airport. We suggest two metrics of performance: mass advected to the sewer network, which poses an acute risk to aquatic ecosystems, and total mass advected from the system, which poses a longer-term risk for persistent compounds. The optimized designs for each use case reduced effluent loadings of all but the most polar compound (PFOA) to <5% of influent mass. Our results suggest that having the largest possible system area allowed bioretention systems to provide benefits during larger events, which improved performance for all compounds. To improve performance for the most hydrophilic TrOCs, an amendment like biochar was necessary; field-scale research is needed to confirm this result. Our results showed that changing the design of bioretention systems can allow them to effectively capture TrOCs with a wide range of physicochemical properties, protecting human health and aquatic species from chemical impacts.

ER signaling pathway-related proteins, such as TRX, AR, and cyclin D1, may have an important function in breast cancer. However, the ways that they influence breast cancer development and progression are still unclear.Patients and methods: A total of 101 Chinese female patients diagnosed with invasive ductal breast adenocarcinoma were retrospectively enrolled in the study. The expression levels of TRX, AR, and cyclin D1 were detected by immunohistochemistry and analyzed via correlation with clinicopathological characteristics and the expression status of ER, PR, and HER2.Results: The expression status of TRX, AR, and cyclin D1 was not associated with the patient’s age, menopausal status, tumor size, or histological differentiation (P>0.05), but was positively correlated with ER and PR (P<0.001, respectively). Most (66/76, 86.8) TRX-positive patients were also HER2-positive (P=0.003). Of AR- or cyclin D1-positive patients, most had relatively earlier I–II tumor stage (P=0.005 and P=0.047, respectively) and no metastatic lymph node involvement (P=0.008 and P=0.005, respectively).Conclusion: TRX was found to be positively correlated with ER and PR expression, whereas it was negatively correlated with HER2 expression. In addition, we found that the positive expression of AR and cyclin D1 was correlated with lower TNM stage and fewer metastatic lymph nodes, and it was more common in ER-positive breast cancer than in the basal-like subtype. Abstract

Etk/Bmx is a member of the Tec family of cytoplasmic non-receptor tyrosine kinases known to express in epithelial cells. We demonstrate herein that Etk activation in stably Etk-transfected epithelial Pa-4 cells resuited in a consistently increased transepithelial resistance (TER). After 24 h of hypoxic (1% [O.sub.2]) exposure, the TER and equivalent active ion transport rate ([I.sub.eq]) were reduced to [is less than] 5% of the normoxia control in Pa-4 cells, whereas both TER and [I.sub.eq] were maintained at comparable and 60% levels, respectively, relative to their normoxic controls in cells with Etk activation. Moreover, Pa-4 cells exhibited an abundant actin stress fiber network with a diffuse distribution of [Beta]-catenin at the cell periphery. By contrast, Etk-activated cells displayed a redistribution of actin to an exclusively peripheral network, with a discrete band of [Beta]-catenin also concentrated at the cell periphery, and an altered occludin distribution profile.
