Brief daily telephone calls were evaluated as a method of enhancing adolescents' compliance with self-monitoring and physiological assessments of cigarette smoking in the natural environment. Seven direct and indirect measures of compliance were devised. Results indicated that subjects receiving telephone calls complied significantly more than control subjects in providing saliva samples and recording detailed self-monitoring data.

The circadian plasma melatonin profile of German Landrace sows was determined at 1-h intervals for 31 h after exposure to each of four different artificial photoperiods. Under 12L:12D, melatonin concentrations in four sows during the light phase ranged from 22 +/- 5.9 to 96 +/- 25.1 pg/ml (mean +/- SEM). During the dark phase (total dark) the individual concentrations increased two- to fivefold over the peak individual light phase values in three sows. This nocturnal surge was of 3.8 h duration and peaked at 190, 294, and 546 pg/ml at 0100 h, which was 5.5 h after the onset of dark. The surge was abolished in these animals after exposure to 16L:8D and could not be reinstated by the subsequent exposure to 8L:16D. During the latter two photoperiods the mean concentrations were consistently less than the maximum mean value of 30.8 +/- 10 pg/ml during the dark phase and 55 +/- 21.9 pg/ml during the light phase. All the sows displayed regular estrous cycles during the study, and the day of the estrous cycle on which the samples were obtained under each photoperiod was not significantly correlated with the presence or absence of the nocturnal surge. A separate experiment using different animals confirmed the nocturnal surge under 12L:12D. In two out of four cycling sows the peak concentrations during the dark phase (122 and 110 pg/ml at 0400 and 0500 h) were two- to sixfold higher than those of the light phase. Absence of the nocturnal surge under long and short daylengths may be a factor contributing to the decline in reproductive performance during the summer and winter months.

The present study uses data from a nationally administered multiple choice examination (MCE) to assess the performance of Canadian medical students in rheumatology. An average of 1722 medical students undertook to write the Medical Council of Canada (MCC) examination each year between 1978-82. Between 17 and 29 of the 516 MCE questions were of a rheumatological nature and the success rate of students from the 16 Canadian medical schools varied between 65.5 and 78.5% in any given year. No direct correlation was noted between the students' MCE performance and the amount of rheumatology education given at an individual university. Analysis of Arthritis Society Undergraduate Statistics indicated that the average medical student in Canada receives 17.5 h of didactic lectures, 10.4 h of clinical bedside demonstration and 8.9 h of direct supervision of rheumatologic patients by rheumatologists.

Three hundred and twenty-seven animals comprised of deer, mouflon sheep and bighorn sheep were vaccinated with an experimental quadrivalent, modified-live, bluetongue virus (BTV) vaccine. No untoward effects due to the vaccine were noted nor were abortions observed in vaccinated pregnant mouflon sheep. Precipitating antibodies to BTV and epizootic hemorrhagic disease (EHD) virus were identified in the serum of several animals prior to vaccination. Virus neutralizing antibodies to 3 of the 5 serotypes of BTV which exist in the US were identified in the serum of 2 peninsular bighorn sheep.

Ten gnotobiotic lambs and 4 gnotobiotic kids inoculated with virulent bluetongue virus (BTV) including BTV international types 10, 11, 13, and 17 did not show clinical signs of bluetongue. Seroconversion was observed in all gnotobiotes and conventional control sheep inoculated with the appropriate challenge BTV. Viremia was detected in 1 BTV-infected gnotobiote. Conventional control sheep inoculated with a given BTV international serotype showed a marked febrile response with hyperemia of the nasal and buccal mucosa.

During the development of the nervous system, growing axons must traverse considerable distances to find their targets. In insects, this problem is solved in part by pioneer neurons, which lay down the first axonal pathways when distances are at a minimum. Here the existence of a similar kind of neuron in the developing mammalian telencephalon is described. These are the subplate cells, the first postmitotic neurons of the cerebral cortex. Axons from subplate neurons traverse the internal capsule and invade the thalamus early in fetal life, even before the neurons of cortical layers 5 and 6, which will form the adult subcortical projections, are generated. During postnatal life, after the adult pattern of axonal projections is firmly established, most subplate neurons disappear. These observations raise the possibility that the early axonal scaffold formed by subplate cells may prove essential for the establishment of permanent subcortical projections.

Two strains of the same virus (isolates AR 168 and 7856), were isolated in 1981 from an apparently healthy cow and a sick sheep in TX, U.S.A. These isolates were shown to be members of the Bunyamwera serogroup (family Bunyaviridae, genus Bunyavirus) by complement-fixation tests. Serum dilution-plaque reduction neutralization test results indicated that the isolates are closely related to Cache Valley virus. The virus isolates were characterized by sensitivity to lipid solvent, size (50-100 nm by filtration and 70 nm by electron microscopy), heat (56 degrees C) and pH 3 lability, cytopathic effects or plaques in cultures of Vero, LLC-MK2, embryonic bovine testicle and PS cells, and pathogenicity for suckling and weaned mice by the intracranial but not the intraperitoneal route. Gnotobiotic and conventional sheep and goats were experimentally infected by inoculation with one of the isolates given either intravenously or intraperitoneally. Elevation of body temperature, depression, tremors, muscle spasms, disorientation, feeding anomalies, convulsions, or other signs of central nervous system disturbances were observed.

Three methods of extracting bluetongue virus (BTV)-infected newborn mouse brains to prepare immunodiffusion (ID) test antigen were used. The most readily readable and reproducible results were obtained with fluorocarbon-extracted brains homogenized in 8.5% sucrose. Mouse brain- and reference cell culture-derived antigens gave a line of identity with anti-BTV serum. Extracts of noninfected brains were nonreactive. ID tests on field-collected bovine sera, comparing the two types of antigen, resulted in only 73% agreement due to a greater sensitivity of cell culture-derived antigen. A 70.5% agreement resulted when comparing mouse brain-derived antigen in ID tests with complement fixation tests, the latter being least sensitive. ID test results with sera from experimental sheep gave 95.9% agreement between cell culture- and mouse brain-derived antigens. Between ID, which detects antibody to the BTV common or group antigen, and virus neutralization, which detects type-specific antibody, the agreement was 71.4% with postchallenge sera. Data from pre- and postinjection sera, however, indicate the possible activity in Texas of viruses other than International BTV Types 10, 11, 13, and 17.

This experiment showed that phoria-induced displacement adds to or subtracts from prism-induced displacement. A near stimulus (25 cm) was apparently displaced more than the optical displacement when the base of a prism was out and less when the base was in. In contrast, a far stimulus (200 cm) was displaced less when the base was out and more when the base was in. Moreover, the between-subjects variability of the apparent displacement was greater with monocular than with binocular viewing. Some implications for studies on monocular prism adaptation are discussed.

In the mammalian cerebral cortex, neurons in a given layer are generated at about the same time in development. These cells also tend to share similar sets of morphological and physiological properties and have projection patterns characteristic of that layer. This correspondence between the birthday and eventual fate of a cortical neuron suggests the possibility that the commitment of a cell to a particular laminar position and set of connections may occur very early on in cortical development. The experiments described here constitute an attempt to manipulate the fates of newly generated cortical neurons upon transplantation. The first set of experiments addressed the normal development of neurons in the primary visual cortex (area 17) of the ferret. Injections of 3H-thymidine into newborn ferrets showed that neurons generated after birth are destined to sit in layer 2/3 of the cortex, whereas neurons born on embryonic day (E) 32 populate primarily layers 5 and 6. Many layer 2/3 neurons in adult ferrets could be retrogradely labeled with HRP from visual cortical areas 18 and 19, while about half of the neurons in layer 6 were found to project to the lateral geniculate nucleus (LGN). In the second set of experiments, presumptive layer 2/3 cells were labeled in vivo by injecting ferrets with 3H-thymidine on P1 and P2. Before the cells had a chance to migrate, they were removed from the donor brain, incubated in a fluorescent dye (DAPI or fast blue), and dissociated into a single-cell suspension. The labeled cells were then transplanted into the proliferative zone of a littermate host ferret ("isochronic" transplants). Over the next few weeks, many of these dye-labeled cells underwent changes in their position and morphology that were consistent with a radially directed migration and subsequent differentiation into cortical neurons. The final positions of isochronically transplanted neurons in the host brain were mapped out by using the 3H-thymidine marker after long survival periods. About 97% of radioactively labeled cells had migrated out into the visual cortex, where they attained a compact laminar distribution: 99% were found in layer 2/3, their normal destination. The labeled cells had normal, mostly pyramidal neuronal morphologies and appeared to be well integrated with host neurons when viewed in Nissl-stained sections. Ten isochronically transplanted neurons were successfully labeled after HRP injection into 2 normal target regions, areas 18 and 19.(ABSTRACT TRUNCATED AT 400 WORDS)

Earlier studies have indicated that percutaneous transluminal coronary angioplasty (PTCA) of chronic total occlusions has a low success rate. To determine success rate and assess clinical and angiographic variables associated with success and complications, 57 total occlusions in 56 patients undergoing PTCA were analyzed. The clinical duration of occlusion was 51 +/- 86 days. Success (less than 50% residual stenosis) was achieved at 40 of 57 (70%) dilatation sites. Of these 57 total occlusions, 5 were attempted within 24 hours of acute myocardial infarction, 35 between 1 day and 8 weeks of clinical occlusion, 13 greater than 8 weeks and 4 were of unknown duration. Success rates were 4 of 5, 25 of 35, 9 of 13 and 2 of 4, respectively, in each group (difference not significant, comparison of all time groups). Of the 9 narrowings with a successful PTCA for an occlusion greater than 8 weeks, the mean duration of occlusion was 93 +/- 41 days (range 60 to 180). None of the attempted dilatations of occlusions with a clinical duration of greater than 180 days (n = 3) was successful. None of the clinical or angiographic variables (including tortuosity, length of occlusion gap, distance of the occlusion from the vessel origin, thrombus, lesion calcium, collaterals, prior myocardial infarction, vessel dilated or diffuse disease) impacted on success rate (difference not significant for all). No patient died, had a Q-wave infarction, required emergency coronary artery bypass grafting or underwent repeat PTCA within 7 days of the procedure. Non-Q-wave infarction occurred in 2 of 56 patients (4%).(ABSTRACT TRUNCATED AT 250 WORDS)

The bovine herpesvirus type 4 (BHV-4) group has a slow replication cycle, a narrow host range, and cytopathogenic effects characteristic of cytomegaloviruses (CMV), but a Group B genome structure similar to that of lymphotropic Herpesvirus saimiri (HVS). Reference BHV-4 strain DN599 and BHV-4 strains N124 and FHV-2 induced in the cytosol fraction of thymidine kinase-negative (TK-) rabbit skin (RAB-BU) cell mutants a novel TK activity. The BHV-4-induced thymidine kinase (TK) differed from the principal cytosol TK of mock-infected cells in PAGE mobility (Rm) under non-denaturing conditions and in the capacity to efficiently substitute CTP for ATP as a phosphate donor. The BHV-4 thymidine phosphorylating activity could also be distinguished from many common herpesvirus-induced TKs because it lacked iododeoxycytidine phosphorylating activity. Iododeoxyuridine, trifluorothymidine and bromovinyldeoxyuridine inhibited [3H]thymidine (0.01 mM) phosphorylation by the BHV-4 enzyme in a dose-dependent manner, but arabinosylthymine and 2'-fluoro-5-methyl-arabinosyluracil (FMAU) were poor inhibitors of [3H]thymidine phosphorylation, and acyclovir and (dihydroxy-2-propoxymethyl)guanine (DHPG) did not inhibit at all at 60 and 40 times the concentrations of [3H]thymidine, respectively.

The functional organization of geniculocortical afferents and the visual responses of neurons in primary visual cortex (area 17) were studied in barbiturate-anesthetized, paralyzed minks and cats. Responses of the afferents were studied after silencing intrinsic cortical activity with injections of kainic acid. In both species, afferents were segregated into patches on the basis of eye of origin. In the mink, but not in the cat, there was a further segregation on the basis of center type, with on- and off-center afferents terminating in alternating, partially overlapping patches. The visual responses of cortical neurons in the mink showed many similarities to those in the cat. Nearly all units were orientation-selective, and there was a columnar organization for preferred orientation. Many units were selective for one direction of movement. Within the binocular segment of cortex, although many units could be driven from either eye, there was a marked bias toward the contralateral eye compared to the cat. There was a columnar system for ocular dominance, but contralateral eye columns were wider than ipsilateral. In both species, a quantitative study was made of the responses of cortical neurons to stationary, flashing slits as a function of position in the receptive field. In the mink, and less clearly in the cat, units could be identified as simple or complex on the basis of the spatial separation or overlap of "on" and "off" discharge zones. In both species, simple cells were found most commonly in layers IV and VI, while layer V contained the greatest proportion of complex cells. The relative strengths of the on and off discharges of single cells were also measured. In the mink, many units gave better overall responses to the on or off phase of the stimulus, and 15% showed a strong (greater than 9:1) preference for one or the other, compared to 4% in the cat. In the mink, units with a common preference for the on or off phase of stationary stimuli were arranged in columnar aggregates, a feature of cortical organization that was not found in the cat. These columns probably result from the partial segregation of on-center and off-center geniculate afferents within layers IV and VI of the mink's cortex. On-dominated columns were, however, wider or more numerous than off-dominated columns.

One of the fundamental tasks of neurobiology is to understand how the precision and specificity of the adult nervous system is achieved during development. This paper reviews the progress that has been made toward this end in studies of the developing mammalian cerebral cortex. Particular attention is focused on the problem of how cortical neurons make decisions during development: the correlation between a neuron's 'birthday' and its final laminar destination and projection patterns has raised the possibility that young neurons may be committed to their adult fates very early on in development, perhaps prior to migration. Indeed, several lines of evidence reviewed here suggest that at least some of the decisions made by cortical neurons are intrinsic properties of the cell itself. These studies include experiments on the reeler mouse mutant, and more recent attempts to manipulate developmental fates by pharmacological interventions and transplantation techniques. It is concluded that early commitment events in the cerebral cortex may specify a neuron's laminar position and restrict the range of potential axonal projections that the cell may form, but that local positional cues direct neurons to select (or maintain) only certain of the possible projections.
