Abstract

Self-assembled drug-polymer micelles are attractive candidates for drug delivery. In this study, amphiphilic floxuridine (FdU)-poly ε-caprolactone (PCL) conjugates containing ester bond were synthesized and self-assembled with phenylsulfonyl furoxan (PSF) to form drug-loading micelles. The micelles had an average size of 91.2 nm and a critical micelle concentration (CMC) of 8.56 μg mL−1. The Micelles were stable in physiological environment, but would decompose under intracellular conditions, releasing chemotherapeutic drug FdU and NO precursor PSF in cancer cells. Intracellular glutathione (GSH) triggered NO release from the PSF, with which the FdU inhibited cancer cells synergistically. In vitro cell and in vivo small animal experiments demonstrated that the Micelles could accumulate in the tumor and be internalized by cancer cells. The micelles inhibited the cancer cell proliferation by releasing FdU and NO. The NO acted as an MDR reversal agent and hence increased the intracellular FdU concentration, through which synergistic therapy was achieved. This drug-polymer conjugate-based micelles had high therapeutic efficacy for cancer with low side effects, providing a new approach to cancer chemotherapy. [ABSTRACT FROM AUTHOR]

Abstract

Accurate representation of soil and snow state variables in the land surface model is crucial for good simulation results. In order to investigate the potential impacts of the overestimated snow cover over the Tibetan Plateau (TP) on precipitation over China, we designed comparative experiments over TP with and without updated soil texture data, soil hydrological parameters, and the fresh snow density scheme to address this issue with the Weather Research and Forecasting Model (WRF). The results show that snow cover fraction (SCF) and surface albedo in winter on the TP were overestimated in the WRF control experiment (CTL), with albedo bias exceeding 0.4 in the western TP. The SCF in the WRF improved experiment (WRFIM) is about 8% lower than that of the CTL, and the bias is decreased to 0.2–0.3 in the interior of the TP. The thermal and dynamic effects of the TP in winter have a significant impact on the weather and climate of East Asia during summer. Thus, the simulated summer rainfall bias is significantly reduced by WRFIM in North China, and the mid-lower reaches of the Yangtze River, along with the spatial correlation coefficient between the observed-based and WRF-simulated rainfall increased from 0.76 in CTL to 0.88 in WRFIM. The WRFIM-simulated reductions in snow cover and albedo cause a decrease in upward shortwave radiation and eventually increase net surface radiation. The horizontal temperature gradient of the atmosphere is increased in the WRFIM compared to the CTL, and the increased temperature in the lower and middle layers accommodates more water vapor, which is not conducive to precipitation formation. Correspondingly, the Western Pacific Subtropical High at 500 hPa in the middle troposphere weakened. The low-level anticyclonic circulation at 850 hPa also weakened to the east, affecting water vapor transport in eastern China. It leads to a decrease in the water vapor transported northward by southeasterly from the western Pacific Ocean and a decrease in the water vapor flux reaching North China. Abstract

Background: Parkinson's disease (PD) patients with tremor-dominant (TD) and non-tremor-dominant (NTD) subtypes exhibit heterogeneity. Rapid identification of different motor subtypes may help to develop personalized treatment plans. Methods: The data were acquired from the Parkinson's Disease Progression Marker Initiative (PPMI). Following the identification of predictors utilizing recursive feature elimination (RFE), seven classical machine learning (ML) models, including logistic regression, support vector machine, decision tree, random forest, extreme gradient boosting, etc., were trained to predict patients' motor subtypes, evaluating the performance of models through the area under the receiver operating characteristic curve (AUC) and validating by the follow-up data. Results: The feature subset engendered by RFE encompassed 20 features, comprising some clinical assessments and cerebrospinal fluid α-synuclein (CSF α-syn). ML models fitted in the RFE subset performed better in the test and validation sets. The best performing model was support vector machines with the polynomial kernel (P-SVM), achieving an AUC of 0.898. Five-fold repeated cross-validation showed the P-SVM model with CSF α-syn performed better than the model without CSF α-syn (P = 0.034). The Shapley additive explanation plot (SHAP) illustrated that how the levels of each feature affect the predicted probability as NTD subtypes. Conclusion: An interactive web application was developed based on the P-SVM model constructed from feature subset by RFE. Abstract

Epidermal cells are the main avenue for signal and material exchange between plants and the environment. Leaf epidermal cells primarily include pavement cells, guard cells, and trichome cells. The development and distribution of different epidermal cells are tightly regulated by a complex transcriptional regulatory network mediated by phytohormones, including jasmonic acid, and transcription factors. How the fate of leaf epidermal cells is determined, however, is still largely unknown due to the diversity of cell types and the complexity of their regulation. Here, we characterized the transcriptional profiles of epidermal cells in 3-day-old true leaves of Arabidopsis thaliana using single-cell RNA sequencing. We identified two genes encoding BASIC LEUCINE-ZIPPER (bZIP) transcription factors, namely bZIP25 and bZIP53, which are highly expressed in pavement cells and early-stage meristemoid cells. Densities of pavement cells and trichome cells were found to increase and decrease, respectively, in bzip25 and bzip53 mutants, compared with wild-type plants. This trend was more pronounced in the presence of jasmonic acid, suggesting that these transcription factors regulate the development of trichome cells and pavement cells in response to jasmonic acid. Abstract

For high-performance gas sensors to be used in practical applications, high sensitivity and ultrafast response are essential. Herein, metal–organic framework (MOF)-derived Pt@ZnO porous nanofibers (PNFs) were constructed by using Pt@ZIF-8 as an electrospun precursor. This brand-new Pt@ZnO PNFs combined the benefits of the two classes of porous materials (MOFs and PNFs) and therefore exhibited open porous characteristics. This mesh fiber structure also prevented the growth and aggregation of ultra-small Pt clusters of about 2 nm. As a typical case study, the 0.5 mL Pt@ZnO PNFs demonstrated good sensing performances characterized by high acetone responses (12.2-fold improvement @ 20 ppm) and fast response/recovery times (2 s/5 s) in comparison with ZnO nanocubes. The Pt@ZnO PNFs also displayed good stability and selectivity towards acetone. We credited these improved sensing signals to synergic interactions between Pt clusters and ZnO nanoparticles, the catalytic spillover effect of Pt clusters, and unique mesh nanofiber structure with open porous characteristics. This work provides a novel design method to construct MOF-derived ultra-small noble metal cluster modified mesh metal oxide PNFs with high gas-sensing performance. Abstract

Although there's increasing research on the health effects of air pollution in China, its direct influence on health-related expenditures, particularly during less severe pollution episodes, is still not well-understood. This study aims to quantify impact of three air pollutants (sulfur dioxide, nitrogen dioxide, and particulate matter) on individual healthcare spending in a city with relatively clean air. Using transaction-level medical social insurance data and temperature inversion as the instrumental variable, we estimate the current and cumulative effects of air pollutants on outpatient visits and medical expenses in an eastern city in China. We find a significantly positive effect on outpatient visits for 15-day moving average, and a significantly positive effect on outpatient expenditure for 25-day moving average. Therefore, the cumulative effect of pollutants on medical expenditure should not be ignored. The effects on total outpatient expenditure are larger for the elderly and men. The cumulative pollution exposure increases the visits to respiratory department but also increases the expenditure on non-respiratory diseases in the long term. Our findings provide empirical evidence that even for cities with good air quality, further improvements can improve patients' quality of life and lessen the expenditure burden on medical insurance. • The cumulative effect of pollutants on outpatient visits and expenditure are found. • The effects on total outpatient expenditure are larger for the elderly and men. • Improvements of air quality can improve patients' quality of life. • Reduction of air pollution can lessen the expenditure burden on medical insurance. Abstract

To investigate the dynamic change characteristics of dynamic functional connectivity (dFC) between the hippocampal subregions (anterior and posterior) and other brain regions in obstructive sleep apnoea (OSA) and its relationship with cognitive function, and to explore whether these characteristics can be used to distinguish OSA from healthy controls (HCs). Eighty-five patients with newly diagnosed moderate-to-severe OSA and 85 HCs were enrolled. All participants underwent resting-state functional magnetic resonance imaging (fMRI). The difference between dFC values between the hippocampal subregions and other brain regions in OSA patients and HCs was compared using the two-sample t tests. Correlation analyses were used to assess the relationship between dFC, clinical data, and cognitive functions in OSA patients. dFC values from different brain regions were used as classification features to distinguish between the two groups using a support vector machine. Compared with HCs, the dFC values between the left anterior hippocampus and right culmen of the cerebellum anterior lobe, right anterior hippocampus and left lingual gyrus, and left posterior hippocampus and left precentral gyrus were significantly lower, and the dFC values between the left posterior hippocampus and precuneus were significantly higher in OSA patients. The dFC values between the left posterior hippocampus and the precuneus of OSA patients were associated with sleep-related indicators and Montreal Cognitive Assessment scores. Support vector machine analysis results showed that dFC values in different brain regions could distinguish OSA patients from HCs. dFC patterns between the hippocampal subregions and other brain regions were altered in patients with OSA, including the cerebellum, default mode networks, sensorimotor networks, and visual function networks, which is possibly associated with cognitive decline. In addition, the dFC values of different brain regions could effectively distinguish OSA patients from HCs. Abstract

In the modern age of gas sensing technologies for broad applications such as internet of things, the capability to make selective, small form factor, and highly responsive sensors for applications such as wearable devices and cell phones could revolutionize the fields of gas sensing systems and fabrications. This thesis aims at developing a millimeter-sized and microwatt-powered sensor prototype using graphene field effect transistors (FETs) by exploring various DC and AC modulation techniques to realize the critical gas sensing features of selectivity and fast recovery speed at room temperature for practical applications.For the DC modulation, we applied the DC gate voltage on the graphene FET during the gas sensing events, and developed the concept of linear factor and the bandwidth enhanced noise method for improving graphene gas sensing performance at DC conditions. Specifically, we have experimentally demonstrated the label-free gas selectivity directly using a single graphene FET for NO2, NH3, H2O and CH3OH by measuring the linear factor parameter. Furthermore, we demonstrated the boosted sensing speed and linearity of the graphene resistance signal using the bandwidth-enhanced method to select the most gas sensitive frequency domain of the noise power density spectrum.For the AC modulation, we applied a hybrid AC+DC gate voltage on graphene FET, and studied the scattering effect and the speed of charge transfer of the gas-graphene interaction for improving the gas selectivity and sensing recovery speed, respectively. Particularly, we measured the scattering strength of the gas adsorbents on graphene and directly resolve the scattering strength spectrum of water, methanol and ethanol adsorption on graphene, achieving the label-free gas sensing using a single graphene transistor. On the other hand, the studies of charge transfer speed between the gas and graphene inspired us to develop the phase-sensitive scheme on graphene FET, achieving the ultrafast baseline recovery speed (~10s) on a defect-rich, chemical vapor deposition (CVD) grown monolayer graphene FET - almost ten times faster than the previous report

Abstract

The spectral reflectance measured in situ is often regarded as the "truth" of objects, which plays an important role in Earth observation applications. However, in situ measurements are influenced by several factors such as atmospheric conditions, illumination and view geometry (I&VG), cloud coverage, and adjacency effects. In order to avoid the influence of these factors, in situ measurements are usually carried out under sunny days and close to noon. However, the impact of I&VG is still present in most cases. At present, people still know little about the influence mechanism of I&VG. Moreover, correcting the impact of I&VG is also a problem that needs to be urgently solved in reflectance spectroscopy. In this work, experiments are carried out using the multi-directional hyperspectral remote sensing simulation facility (MHSRS2F), which allows adjustment and control of the I&VG parameters. This paper proposes an uncertainty evaluation model for I&VG and quantifies the uncertainty caused by different I&VG parameters. Then, the sensitivity of reflectance to I&VG at different wavelengths is explored based on uncertainty models. Finally, a correction model for reflectance under different I&VG conditions is proposed. The results reveal that the uncertainty and sensitivity caused by observation height are relatively high, regardless of the surface heterogeneity. It directly affects the size of the field of view and the physicochemical characteristics of the object. For objects that approximate the Lambertian surface, more attention should be paid to the selection and variation of solar and view zenith angles and view azimuth angles. For objects with surface heterogeneity, the selection and variation of solar azimuth angle, view azimuth angle, and solar zenith angle are more crucial. The correction model proposed in this paper has a 41.25% correction effect on different view zenith angles, but the correction effect on other environmental factors is not significant. Abstract

Cuproptosis has been recently used to indicate unique biological processes triggered by Cu action as a new term. This study aimed to explore the relationship between cuproptosis-related lncRNA and hepatocellular carcinoma (HCC) with regard to immunity and prognosis. RNA sequencing and the clinical data were downloaded from the TCGA database. The cuproptosis-related genes were sorted out through literature study. The cuproptosis-related IncRNA signature was identified by Cox regression analysis and the least absolute shrinkage and selection operator analysis. The K-M survival analysis, receiver operating characteristic analysis, and C-index analysis were adopted to evaluate the prognostic prediction performance of the signature. The functional enrichment, immune infiltration and tumor mutation analysis were further analyzed. Subsequently, we predicted the differences in chemosensitivity from tumor gene expression levels for some chemotherapy drugs. The prognostic signature consisting of 5 overall survival-related CUPlncRNAs. It showed an extraordinary ability to predict the prognoses of patients with HCC. The signature can predict the abundance of immune cell infiltration, immune functions, expression of immune checkpoint inhibitors, m6A genes, which was supported by the GO biological process and KEGG analysis. And it may also have a guiding effect in the sensitivity of different chemotherapeutic drugs and tumor mutation burden. We constructed a new cuproptosis-related lncRNA signature for HCC patients. Abstract

Here, we propose a green and sustainable 3D porous aerogel based on citrus peel (CP), chitosan (CS), and bentonite (BT). This aerogel is prepared through a simple sol–gel and freeze-drying process and is designed for efficient capture of Cu(II) ions from water matrices. CCBA-2, with its abundance of active binding sites, exhibits an impressive Cu(II) adsorption yield of 861.58 mg/g. The adsorption isotherm and kinetics follow the Freundlich and pseudo-second-order models, respectively. In the presence of coexisting mixed-metal ions, CCBA-2 demonstrates a significantly higher selectivity coefficient (KdCu = 1138.5) for removing Cu(II) ions compared to other toxic metal ions. Furthermore, the adsorption of Cu(II) ions by CCBA-2 is not significantly affected by coexisting cations/anions, ionic strength, organic matter, or different water matrices. Dynamic fixed-bed column experiments show that the adsorption capacity of Cu(II) ions reaches 377.4 mg/g, and the Yoon-Nelson model accurately describes the adsorption process and breakthrough curve. Through experiments, FTIR, and XPS analyses, we propose a reasonable binding mechanism between CCBA-2 and metal cations, involving electrostatic attraction and chemical chelation between Cu(II) and the functional groups of the aerogel. CCBA-2 saturated with Cu(II) ions can be successfully regenerated by elution with 1 M HNO3, with only a slight decrease in adsorption efficiency (5.3%) after 5 adsorption–desorption cycles. Therefore, CCBA-2 offers a cost-effective and environmentally friendly material that can be considered as a viable alternative for the green and efficient removal of toxic Cu(II) ions from wastewater. Abstract

This study aimed to create a glycyrrhetinic acid (GA)-mediated, multi-component, self-assembled nano-drug delivery system co-loaded with syringopicroside (S) and hydroxytyrosol (H) obtained from Syringa Linn by synthesizing a GA-polyethylene glycol-poly (lactic acid-co-glycolic acid) (GPP) nanoparticle delivery carrier to actively target the liver. The nanoparticles were optimized using the central composite design. Nanoparticle characterization, cytotoxicity, pharmacodynamics, and tissue distribution study were performed. The optimized SH-GPP nanoparticle was a white solid powder, which was safe and non-toxic. The particle size and Zeta potential of the nanoparticles were 101.5 ± 3.18 nm and −23.3 ± 0.82 mV, respectively. The polydispersity index value (PDI) was 0.190 ± 0.005; the particle size distribution was comparatively uniform. The average total encapsulation efficiency of the optimized SH-GPP nanoparticle was 50.26% ± 1.29%, and drug loading was 15.47% ± 0.39%. After S and H were arranged into nanoparticles, the proliferation inhibition of HepG2.2.15 cells was improved, and the aim of drug-loaded synergism between GPP and SH was achieved. The GA-mediated nanoparticles were better targeted, were retained longer in vivo, and had higher concentrations in the liver than the unmodified nanoparticles. Abstract

Neuromorphic photonics devices based on phase change materials (PCMs) and silicon photonics technology have emerged as promising solutions for addressing the limitations of traditional spiking neural networks in terms of scalability, response delay, and energy consumption. In this review, we provide a comprehensive analysis of various PCMs used in neuromorphic devices, comparing their optical properties and discussing their applications. We explore materials such as GST (Ge2Sb2Te5), GeTe-Sb2Te3, GSST (Ge2Sb2Se4Te1), Sb2S3/Sb2Se3, Sc0.2Sb2Te3 (SST), and In2Se3, highlighting their advantages and challenges in terms of erasure power consumption, response rate, material lifetime, and on-chip insertion loss. By investigating the integration of different PCMs with silicon-based optoelectronics, this review aims to identify potential breakthroughs in computational performance and scalability of photonic spiking neural networks. Further research and development are essential to optimize these materials and overcome their limitations, paving the way for more efficient and high-performance photonic neuromorphic devices in artificial intelligence and high-performance computing applications. Abstract

The nervous necrosis virus (NNV) of the BFNNV genotype is the causative agent of viral encephalopathy and retinopathy (VER) in cold water fishes. Similar to the RGNNV genotype, BFNNV is also considered a highly destructive virus. In the present study, the RNA2 of the BFNNV genotype was modified and expressed in the EPC cell line. The subcellular localization results showed that the capsid and N-terminal (1–414) were located in the nucleus, while the C-terminal (415–1014) of the capsid was located in the cytoplasm. Meanwhile, cell mortality obviously increased after expression of the capsid in EPC. EPC cells were transfected with pEGFP-CP and sampled at 12 h, 24 h and 48 h for transcriptome sequencing. There are 254, 2997 and 229 up-regulated genes and 387, 1611, and 649 down-regulated genes post-transfection, respectively. The ubiquitin-activating enzyme and ubiquitin-conjugating enzyme were up-regulated in the DEGs, indicating that cell death evoked by capsid transfection may be related to ubiquitination. The qPCR results showed that heat stock protein 70 (HSP70) is extremely up-regulated after expression of BFNNV capsid in EPC, and N-terminal is the key region to evoke the high expression. For further study, the immunoregulation of the capsid in fish pcDNA-3.1-CP was constructed and injected into the Takifugu rubripes muscle. pcDNA-3.1-CP can be detected in gills, muscle and head kidney, and lasted for more than 70 d post-injection. The transcripts of IgM and interferon inducible gene Mx were up-regulated after being immunized in different tissues, and immune factors, such as IFN-γ and C3, were also up-regulated in serum, while C4 was down-regulated one week after injection. It was suggested that pcDNA-3.1-CP can be a potential DNA vaccine in stimulating the immune system of T. rubripes; however, NNV challenge needs to be conducted in the following experiments. Abstract

Polyacrylamide‐based hydrophobically associating polymers (HAPAM) containing capsaicin derivatives and itaconic acid with acrylamide (AM) and methacrylic acid (MAA) as the main bodies, and N‐vinylpyrrolidone (NVP) as the functional monomer are prepared by a one‐pot synthesis of the soap‐free emulsion polymerization process. The structure is characterized by fourier transform infrared (FTIR) and 1H nuclear magnetic resonance (1H NMR), and thermal gravimetric analyzer (TGA) explores thermal stability. For achieving proper purification, excellent preservation and convenient transportation of the polymer and to advance its practical engineering applications, two different treatments, namely ethanol washout and freeze‐drying, are carried out and compared. The surface morphology observation manifests that the freeze‐drying products (DHPAM) better protect the three‐dimensional cross‐linked network structure formed by various supramolecular forces such as hydrophobic association, electrostatic and hydrogen bonding inside the polymer than the ethanol washout products (EHPAM), which well explains why DHPAM has lower critical association concentration (CAC), greater resistance to high temperature and high salt, and better corrosion inhibition performance than EHPAM. Abstract

The fracture behavior of ultra‐high temperature ceramic matrix composites at high temperature has received increasing attention. However, few studies consider the effect of particle/crack interaction on the high temperature fracture strength of materials. In this work, the effect of particle/crack interaction is introduced into temperature‐dependent fracture strength model of monolithic ultra‐high temperature ceramic matrix composites, which also considers effects of flaw size, grain size, and residual thermal stress. Furthermore, by considering the influence of laminated structure, a theoretical model of the temperature‐dependent fracture strength of laminated ceramic matrix composites is developed. The effect of particle/crack interaction is also included in this model. It should be noted that the predictions of models agree well with the experimental data of both monolithic and laminated materials without using any fitting parameters. The effect of particle/crack interaction is found to have a significant weakening effect on the strength of materials at different temperatures.
