A synthetic method to prepare partially hydrogenated isoquinolines efficiently from silver-mediated [3,3]-sigmatropic rearrangement/Diels-Alder reaction of 1,9-dien-4-yne esters is described. The reactions were shown to be robust with a wide variety of substitution patterns tolerated to provide the corresponding nitrogen-containing heterocyclic products in good to excellent yields. This includes examples containing a bridgehead sp 3 quaternary carbon center as well as the cycloisomerization of one substrate to give the corresponding bicyclic adduct in excellent yield at the gram scale.

The synthesis of 1,4-amino alcohols from THF treated with N-tosyliminobenzyliodinane (PhINTs) followed by a Grignard reagent under mild reaction conditions at room temperature is described herein. Various Grignard reagents were shown to be compatible, furnishing the corresponding 4-substituted-N-1,4-tosylamino alcohols in good to excellent yields. A partial or full detosylation of the N-tosyl-1,4-amino alcohol was observed in instances involving a sterically bulky Grignard reagent, leading to the deprotected 1,4-amino alcohol product in moderate to good yields. The synthetic utility of this protocol was demonstrated by the synthesis of a 5-substituted-N-tosyl-1,5-amino alcohol from THP and the conversion of two examples to their corresponding γ-lactam and pyrrolidine adducts.

A method for the efficient preparation of hydronaphthalene and -cinnoline derivatives by Au(I)-catalyzed cycloisomerzation of 1,6-diyne esters followed by a Diels-Alder reaction with alkenes or diazenes under mild conditions at room temperature with catalyst loadings as low as 1 mol % is described.

A one-pot, two-step approach to prepare 2-tetrahydrofuran and -pyran substituted 1,3-dicarbonyl compounds by PhI=NTs-mediated amination/Brønsted base-catalyzed cross dehydrogenative coupling (CDC) reaction of the cyclic ether and 1,3-dicarbonyl derivative under mild conditions is reported. The reaction is compatible with a variety of cyclic ethers and 1,3-dicarbonyl compounds, affording the corresponding coupled products in moderate to good yields of up to 80% over two steps.

The mol-ecule of the title compound, C25H24O, obtained by acid-catalysed 1,3-migration of an alcohol group, is T-shaped. The planes of the two phenyl rings are inclined to one another by 81.9 (2)°. In the crystal, mol-ecules are linked by O-H⋯O hydrogen bonds, forming chains along [001].

A synthetic method to chemoselectively prepare 1H-cyclopenta[b]naphthalenes, cis-cyclopenten-2-yl δ-diketones, and bicyclo[3.2.0]hepta-1,5-dienes efficiently by gold-catalyzed cycloisomerization of 1,6-diyne esters is described. These three product classes were accessed divergently by taking advantage of the electronic and steric differences between a phosphine and NHC (NHC = N-heterocyclic carbene) ligand in the respective gold(I) complexes and that of gold(III) complex combined with substrate substitution patterns and optimized reaction conditions. In the presence of [PhCNAuIPr](+)SbF6(-) (IPr = 1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidine) as the catalyst, substrates with a pendant aryl group at the acetate alkynyl position were found to undergo preferential 1,3-acyloxy migration/5-exo-dig cyclization/Friedel-Crafts reaction to give 1H-cyclopenta[b]naphthalenes. In contrast, the analogous reactions with PicAuCl2 (Pic =2-picolinate) were found to proceed by selective 1,3-acyloxy migration/5-exo-dig cyclization/1,5-acyl migration to afford cis-cyclopenten-2-yl δ-diketones. Changing the catalyst to [MeCNAu(JohnPhos)](+)SbF6(-) (JohnPhos = (1,1'-biphenyl-2-yl)-di-tert-butylphosphine) and the acetate alkynyl position from an aryl to vinyl substituent in the starting ester led to 1,3-acyloxy migration/5-exo-dig cyclization/Prins-type [2 + 2]-cycloaddition to provide bicyclo[3.2.0]hepta-1,5-dienes.

A method to prepare 1-substituted 3-sulfonyl-1H-pyrroles efficiently that relies on the gold(I)-catalyzed cycloisomerization of N-substituted N-sulfonyl-aminobut-3-yn-2-ols is described. The method was shown to be applicable to a broad range of 1,7-enyne alcohols containing electron-withdrawing, electron-donating, and sterically demanding substrate combinations. The mechanism is suggested to involve activation of the propargylic alcohol by the Au(I) catalyst, which causes the intramolecular nucleophilic addition of the sulfonamide unit to the alkyne moiety. The resulting nitrogen-containing heterocyclic intermediate undergoes dehydration and deaurative 1,3-sulfonyl migration, a process that remains rare in gold catalysis, to give the aromatic nitrogen-containing product.

A synthetic method to prepare 2,4a-dihydro-1H-fluorenes efficiently from gold(I)-catalyzed 1,2-acyloxy migration/cyclopropenation/Nazarov cyclization of 1,6-diyne carbonates and esters is described. The suggested reaction pathway provides rare examples of [2,3]-sigmatropic rearrangement in this class of compounds as well as the involvement of an in situ formed cyclopropene intermediate in gold catalysis. Experimental and ONIOM(QM:QM') [our own n-layered integrated molecular orbital and molecular mechanics(quantum mechanics:quantum mechanics')] computational studies based on the proposed Au carbenoid species provide insight into this unique selectivity.

Don't cross me! 2-Alkynyl indoles were prepared efficiently by the AgOTf-catalyzed tandem heterocyclization/alkynylation of 1-(2-tosylamino)aryl)but-2-yne-1,4-diols under mild conditions (see scheme). The attractiveness of this approach lies in the fact that both the indole ring and alkyne side chain of the N-heterocycle are sequentially formed from low cost, readily available, and ecologically benign starting materials. Catalytic insertion or addition of a metal-imido/nitrene species, generated from reaction of a transition-metal catalyst with iminoiodanes, to C-H and C=C bonds offers a convenient and atom economical method for the synthesis of nitrogen-containing compounds. Following this groundbreaking discovery during the second half of the last century, the field has received an immense amount of attention with a myriad of impressive metal-mediated methods for the synthesis of amines and aziridines having been developed. This review will cover the significant progress made in improving the efficiency, versatility and stereocontrol of this important reaction. This will include the various iminoiodanes, their in situ formation, and metal catalysts that could be employed and new ligands, both chiral and non-chiral, which have been designed, as well as the application of this functional group transformation to natural product synthesis and the preparation of bioactive compounds of current therapeutic interest., (Copyright © 2011 The Japan Chemical Journal Forum and Wiley Periodicals, Inc.)

A synthetic method that relies on gold-catalysed cyclopropyl carbinol rearrangement of 2-tosylaminophenyl cyclopropylmethanols to prepare 2,3-dihydro-1H-benzo[b]azepines and 2-vinylindolines efficiently is reported. The reactions were shown to be chemoselective, with secondary and tertiary alcohol substrates exclusively providing benzo-fused five- and seven-membered ring products, respectively. The ring-forming process was also found to proceed in moderate to excellent yields under mild conditions only in the presence of the gold and silver catalyst combination. The mechanism is thought to involve activation of the alcohol by the (p-CF(3)C(6)H(4))(3)PAuCl/AgOTf (Tf = triflate) catalyst, resulting in ionization of the starting material. The tertiary carbocationic intermediate generated in situ in this manner then triggers ring-opening of the cyclopropane moiety and trapping by the tethered aniline group to give the 2,3-dihydro-1H-benzo[b]azepine. Cyclopropane ring fragmentation of the secondary carbocationic analogue, on the other hand, results in diene formation followed by subsequent intramolecular hydroamination to afford the 2-vinylindoline., (Copyright © 2011 WILEY-VCH Verlag GmbH & Co. A general and efficient method to prepare 2,4-di- and trisubstituted thiazoles via p-TsOH·H(2)O-catalyzed cyclization of trisubstituted propargylic alcohols with thioamides is described. The reaction was accomplished in moderate to excellent product yields under mild conditions that did not require the exclusion of air and moisture and offers an operationally simplistic and convenient route to this synthetically useful aromatic heterocycle.

A method to prepare indenols efficiently by ytterbium(III) triflate catalyzed tandem Friedel-Crafts alkylation/hydroarylation of propargylic alcohols with phenols is described. The reaction was accomplished in moderate to excellent yields and regioselectivity under mild conditions and offers a straightforward and convenient one step synthetic route to bioactive indenols and its derivatives.

A general and efficient direct method for the alpha-substitution of Morita-Baylis-Hillman alcohols with carbon- and heteroatom-centred nucleophiles such as alcohols, arenes, 1,3-dicarbonyl compounds, and thiols in the presence of FeCl3.6H2O as catalyst has been developed. The reaction is operationally straightforward, accomplished in good to excellent product yields (40-99%) and with exclusive alpha-regioselectivity under mild conditions that did not need an inert and moisture-free environment.

A highly efficient triflic acid catalyzed ring opening of a wide variety of 1-cyclopropyl-2-propyn-1-ols with alcohols as an efficient synthetic route to conjugated enynes is reported herein. The reaction was operationally straightforward and accomplished in good to excellent yields (44-100%), high product turnovers (up to 10,000), and with complete regioselectivity under mild conditions with a low catalyst loading of 0.01 mol %. The mechanism is suggested to involve protonation of the alcohol substrate by the TfOH catalyst, followed by ionization of the starting material. This causes ring opening of the cyclopropane moiety and trapping by the alcohol nucleophile to give the conjugated enyne product. The synthetic utility of the present method was also exemplified by the efficient large-scale conversion in gram quantities of one example studied in this work to the corresponding conjugated enyne product in excellent yield and turnover number.

The hydrothermal reaction of (L)-ethyl lactate (Lig-Et) with Tb(ClO(4))(3).6H(2)O gives colorless block (Lig)(2)Tb(H(2)O)(2)(ClO(4)) (1), in which 1 displays a laminar 2D framework. Ferroelectric and magnetic property measurements reveal that 1 probably is the first example of two "ferroic" metal-organic frameworks. Ferroelectricity of its analogue, (Lig)(2)Tb(D(2)O)(2)(ClO(4)) (2), further confirms the presence of the ferroelectric deuterium effect.

An efficient synthetic route to pyrrolidines that relies on AuCl/AgOTf-catalyzed tandem amination/ring expansion of substituted cyclopropyl methanols with sulfonamides is reported herein. The reactions proceed rapidly at 100 degrees C with catalyst loadings as low as 2 mol % and produce the pyrrolidine products in yields of 30-95 %. The method was shown to be applicable to a broad range of cyclopropyl methanols, including unactivated ones, and sulfonamide substrates containing electron-withdrawing, electron-donating, and sterically-demanding substituents. The mechanism is suggested to involve activation of the alcohol substrate by the AuCl/AgOTf catalyst, followed by ionization of the starting material, which causes ring opening of the cyclopropane moiety and trapping by the sulfonamide nucleophile. The resultant aminated acyclic intermediate undergoes subsequent intramolecular hydroamination to give the pyrrolidine.

Colorless block crystals of MOF (metal-organic framework) 1 and 2 were prepared in respective yields of 65 and 60% by thermal treatment of HQA (HQA=6-methoxyl-(8S,9R)-cinchonan-9-ol-3-carboxylic acid) with ZnBr(2) in either H(2)O or D(2)O and 2-butanol at 70 degrees C for 1-2 days. The MOFs 1 and 2 are isostructural, one-dimensional chains in which the local coordination geometry around the Zn center can be best described as a slightly distorted tetrahedron defined by two bromine atoms, one nitrogen atom of quinoline from HQA, and an oxygen atom of carboxylate from HQA. The nitrogen atom of the quinuclidine of HQA is protonated in a zwitterionic form. The MOFs 1 and 2 crystallize in a polar point group (C(2), space group P2(1)) which belongs to ferroelectric active compounds. MOFs 1 and 2 display both ferroelectric behavior and large dielectric constants. Interestingly, at low frequency range the dielectric response to water can achieve an approximate increase of more than 600%. Hydrothermal reaction of (l)-N-(4'-cyanobenzy)-(S)-proline with CdCl2 as a Lewis acid catalyst and NaN3 gives colorless block compound 1, in which 1 displays a complicated 3D framework. Ferroelectric and dielectric property measurements reveal that 1 exhibits physical properties comparable to that of a typical ferroelectric compound with a dipole relaxation process and a dielectric constant of ca. 38.6 that makes it, by definition, a high dielectric material.

[reaction: see text] Gold(III) porphyrin-catalyzed cycloisomerization of allenones gave the corresponding furans in good to excellent yields (up to 98%) and with quantitative substrate conversions. By recovering the Au(III) catalyst, a recyclable catalytic system is developed with over 8300 product turnovers attained for the cycloisomerization of 1-phenyl-buta-2,3-dien-1-one. The versatility of the gold(III) porphyrin catalyst was exemplified by its application to the hydroamination and hydration of phenylacetylene in 73% and 87% yield, respectively.

meso-Tetrakis(p-tolyl)porphyrinatoruthenium(II) carbonyl, [Ru(II)(TTP)(CO)], can effect intermolecular sulfonium and ammonium ylide formation by catalytic decomposition of diazo compounds such as ethyl diazoacetate (EDA) in the presence of allyl sulfides and amines. Exclusive formation of [2,3]-sigmatropic rearrangement products (70-80% yields) was observed without [1,2]-rearrangement products being detected. The Ru-catalyzed reaction of EDA with disubstituted allyl sulfides such as crotyl sulfide produced an equimolar mixture of anti- and syn-2-(ethylthio)-3-methyl-4-pentenoic acid ethyl ester. The analogous "EDA + N,N-dimethylcrotylamine" reaction afforded a mixture of anti- and syn-2-(N,N-dimethylamino)-3-methyl-4-pentenoic acid ethyl esters with a diastereoselectivity of 3:1. The observed catalytic activity of [Ru(II)(TTP)(CO)] for the ylide [2,3]-sigmatropic rearrangement is comparable to the reported examples involving [Rh(2)(CH(3)CO(2))(4)] and [Cu(acac)(2)] as catalyst. Similarly, cyclic sulfonium and ammonium ylides can be produced by intramolecular reaction of a diazo group tethered to allyl sulfides and amines under the [Ru(II)(TTP)(CO)]-catalyzed reaction conditions. The subsequent [2,3]-sigmatropic rearrangement of the cyclic ylides furnished 2-allyl-substituted sulfur and nitrogen heterocycles in good yields (>90%). By employing [Ru(II)(TTP)(CO)] as catalyst, the cyclic ammonium ylide [2,3]-sigmatropic rearrangement reaction was successfully applied for the total synthesis of (+/-)-platynecine starting from cis-2-butenediol.

[reaction: see text] Ruthenium(II) porphyrin-catalyzed amidation of aromatic heterocycles with iminoiodanes under mild conditions (CH(2)Cl(2), 4 A molecular sieves, ultrasound, 40 degrees C) was achieved in moderate to good yields (up to 84%) and conversions (up to 99%). Only the N,N-ditosylamidated product was obtained for reactions involving heteroarenes, where X = O, S, or NTs. N-Alkyl- and N-aryl-substituted pyrroles, on the other hand, were shown to give the 3,4-diaminated adduct.
