Brain is a central and pivotal organ of human body containing the highest lipids content next to adipose tissue. It works as a monitor for the whole body and needs an adequate supply of energy to maintain its physiological activities. This high demand of energy in the brain is chiefly maintained by the lipids along with its reservoirs. Thus, the lipid metabolism is also an important for the proper development and function of the brain. Being a prominent part of the brain, lipids play a vast number of physiological activities within the brain starting from the structural development, impulse conduction, insulation, neurogenesis, synaptogenesis, myelin sheath formation and finally to act as the signaling molecules. Interestingly, lipids bilayer also maintains the structural integrity for the physiological functions of protein. Thus, in light to all of these activities, lipids and its metabolism can be attributed pivotal for brain health and its activities. Decisively, the impaired/altered metabolism of lipids and its intermediates puts forward a key step in the progression of different brain ailments including neurodegenerative, neurological and neuropsychiatry disorders. Depending on their associated underlying pathways, they serve as the potential biomarkers of these disorders and are considered as necessary diagnostic tools. The present review discusses the role and level of altered lipids metabolism in brain diseases including neurodegenerative diseases, neurological diseases, and neuropsychiatric diseases. Moreover, the possible mechanisms of altered level of lipids and their metabolites have also been discussed in detail.

This study focusses upon simulation of building blocks for silicon photonic integrated circuits. The two necessary steps for the simulation are control of temperature in the integrated Mach-Zehnder Interferometer (MZI) or modulators (MZM), and stabilization and tuning of optical modulated spectrum in node-matched-diode (NMD) Fabry-Pérot (FP) resonator or modulators. Hence the thesis consists of two parts. The first part of this work deals with the control of temperature in the integrated Mach-Zehnder interferometer (MZI) or modulators (MZM). The thermodynamic model for lattice temperature or heat equation, Shockley-Read-Hall Recombination doping and temperature dependence, and mobility models have been used for thermal simulation in Synopsys Sentaurus device tool. These models were used to calculate the average temperature, electric field strength, charge carrier current density, mobility of carriers, and number of charge carriers per unit volume in different components of heaters and modulators. It is through these models that the Poisson, temperature, and continuity equations were solved. The complete analysis of above quantities and refractive index change in waveguide were computed from Sentaurus visual simulator tools. The steady state thermal simulation of Si- and aluminum heaters in Mach-Zehnder interferometer (MZI) or modulator (MZM) were conducted using Sentaurus TCAD software. The comparison of Si- and aluminum heaters in MZI or MZM thermal simulation analysis reveals that the value of current density of Si-heaters in waveguide 3.95 x 10^(-9) A/µm^2 are greater as compared to aluminum heater 1.8 x 10^(-9) A/µm^2 . But these low values of carrier’s current density do not affect the optical properties of MZI or MZM. Moreover, it was found that integrated Si-heaters (3.47 K/mW) are more thermally efficient than aluminum heaters (2.79 K/mW). It was computed that the refractive index changes per heater power of n-doped heater 6.42 x 10^(-4) (1/mW) is higher as compared to metal heater 5.2 x 10^(-4) (1/mW). These heaters are also used as thermo-optic phase shifter. The power for π-radian phase shift of thermo-optic phase shifter of simulated and measurement results have been verified. It was shown that low power (P_π) 26.42 mW and 24.07 mW for the simulated and real thermo-optic phase shifter to operate the signal in the operating range is required respectively. Furthermore, the high phase-shifting efficiency of simulated 26.42 ( mW)/π and real 24.07 mW/π phase shifter was determined. Consequently, simulated values of phase shifters were in a very good in agreement with experimental results. The silicon integrated heaters can be used in MZI or MZM due to their overall low temperature, uniform heating, high thermal efficiency, low current density in waveguide, low power consumption and high refractive index change per milliwatt power. Hence, highly efficient novel silicon heaters are appropriate to stabilize the optical spectrum and control the temperature in Mach-Zehnder Interferometer or Mach-Zehnder modulators. The second part deals with the stabilization and tuning of optical modulated spectrum in node-matched-diode (NMD) Fabry-Pérot (FP) resonator or modulators. These models were used as in the previous application of MZI. The steady state thermal simulation of aluminum and novel n-doped L- and U-shaped heaters were conducted in NMD FP-modulator using Synopsys Sentaurus software. It was computed that the thermal efficiency of n-doped silicon heaters 0.67 K/mW was high as compared to metal heaters 0.65 K/mW in NMD FP-modulator. It was shown that Si-heaters have high tuning efficiency 0.062 nm/mW as compared to metal heaters 0.060 nm/mW. The n-doped heaters tune the optical spectrum 3.4 nm to red-shift due to thermo-optic effect (TOE). Then the refractive index change of waveguide was computed in switch-on state of NMD. The influence on wavelength shift due to carrier plasma dispersion effect (PDE) and TOE was determined. The net wavelength shift amount to be 8.06 nm to blue-shift. In the third part, the combined thermal effects of NMD and Si-heaters have been computed. The silicon heaters use TOE and moves the spectrum to red-shift 3.8 nm. However, net wavelength shift is 4.26 nm in blue shift. The plasma dispersion effect dominates the thermo-optic effect. The trade-off spectrum due to switched-on state of NMD and Si-heaters was determined. In the end the simulated and estimated measurement results of Si- and aluminum heaters are verified. Hence it was proved that high thermal and tuning efficiency of n-doped heaters produce less temperature in the chip, generate uniform temperature in resonator and consume less power. The n-doped L- and U- shaped Si-heaters are appropriate to tune the spectrum in operating range in NMD FP-modulator. Diese Arbeit konzentriert sich auf die Simulation von Bausteinen für photonisch integrierte Siliziumschaltkreise. Die zwei notwendigen Schritte für die Simulation sind Temperatursteuerung in integrierten Mach-Zehnder-Modulatoren (MZM) und Stabilisierung und Abstimmung des optisch modulierten Spektrums in Node-Matched-Diode (NMD) Fabry-Pérot (FP) Resonatoren. Der erste Teil befasst sich mit der Temperaturregelung im integrierten MZM. Das thermodynamische Modell für die Gittertemperatur- oder Wärmegleichung, Shockley-Read-Hall-Rekombinationsdotierung und Temperaturabhängigkeit sowie Mobilitätsmodelle wurden für die thermische Simulation in Synopsys Sentaurus Device verwendet. Diese Modelle wurden zur Berechnung der Durchschnittstemperatur, der elektrischen Feldstärke, der Ladungsträgerstromdichte, der Mobilität der Ladungsträger und der Anzahl der Ladungsträger pro Volumeneinheit in verschiedenen Komponenten von Heizelementen und Modulatoren verwendet. Durch diese Modelle wurden die Poisson-, Temperatur- und Kontinuitätsgleichungen gelöst. Die vollständige Analyse der oben genannten Größen sowie der Brechungsindexänderung im Wellenleiter wurden mit den visuellen Simulationswerkzeugen von Sentaurus berechnet. Die stationäre thermische Simulation von Silizium (Si) - und Aluminiumheizelementen (Al) im MZM wurde mit Synopsys Sentaurus TCAD durchgeführt. Der Vergleich von Si- und Al-Heizelementen in der Thermosimulationsanalyse zeigt, dass der Wert der Stromdichte von Si-Heizelementen im Wellenleiter mit 3,95 x 10^(-9) A/μm^2 größer ist als im Aluminiumheizelement mit 1,8 x 10^(-9) A/μm^2 . Diese niedrige Stromdichte beeinflussen jedoch nicht die optischen Eigenschaften der MZM. Darüber hinaus wurde festgestellt, dass integrierte Si-Heizelemente mit einem thermischen Wirkungsgrad von 3,47 K/mW effizienter sind als Aluminiumheizelemente mit dem Wirkungsgrad 2,79 K/mW. Die Berechnungen zeigen, dass die Abhängigkeit des Brechungsindex von der Leistung des n-dotierten Heizelements mit 6,42 x 10^(-4) (1/mW) höher ist als bei Metallheizelementen mit 5,2 x 10^(-4) (1/mW). Die Heizelemente werden auch als thermooptischer Phasenschieber verwendet. Die Leistung für die π-Phasenverschiebung des thermooptischen Phasenschiebers aus den Simulationen wurde anhand von Messungen verifiziert. Es wurde gezeigt, dass die geringe Leistung (Pπ) des simulierten und realen thermooptischen Phasenschiebers von 26,42 mW bzw. 24,07 mW zum Betrieb des Signals im Betriebsbereich notig ist. Darüber hinaus wurde eine hohe Phasenverschiebungseffizienz von 26,42 mW/π für den simulierten und 24,07 mW/π für den realen fPhasenschieber bestimmt. Daher stimmten die simulierten Werte der Phasenschieber sehr gut mit den experimentellen Ergebnissen überein. Die integrierten Siliziumheizelemente können aufgrund ihrer niedrigen Gesamttemperatur, der gleichmäßigen Erwärmung, der hohen thermischen Effizienz, der geringen Stromdichte im Wellenleiter, dem niedrigen Energieverbrauch sowie einer hohen Brechungsindexänderung pro Milliwatt Leistung in MZM verwendet werden. Daher sind hocheffiziente neuartige Siliziumheizelemente geeignet, das optische Spektrum zu stabilisieren und die Temperatur in Mach-Zehnder-Modulatoren zu steuern. Der zweite Teil befasst sich mit der Stabilisierung und Abstimmung des optisch modulierten Spektrums in Node-Matched-Diode (NMD) -Fabry-Pérot (FP) –Resonatoren oder Modulatoren. Diese Modelle wurden wie in der vorherigen Anwendung von MZM verwendet. Die stationäre thermische Simulation von Aluminiumheizelementen und neuartigen n-dotierten L- und U-förmigen Heizelementen wurde mit einem NMD-FP-Modulator unter Verwendung der Synopsys Sentaurus Software durchgeführt. Es wurde berechnet, dass der thermische Wirkungsgrad von n-dotierten Siliziumheizelementen mit 0,67 K/mW im Vergleich zu Metallheizelementen mit 0,65 K/mW höher ist. Es wurde gezeigt, dass Si-Heizelementen eine hohe Abstimmeffizienz von 0,062 nm/mW aufweisen, verglichen mit Metallheizelementen von 0,060 nm/mW. Aufgrund des thermooptischen Effekts in n-dotierten Heizelementen ist das optische Spektrum um 3,4 nm rotverschoben. Die Brechungsindexänderung im Wellenleiters bei eingeschalteter NMD wurde berechnet. Der Einfluss des Plasma-Dispersionseffekts (PDE) und des TOE auf die Wellenlängenänderung wurde bestimmt. Die Nettowellenverschiebung beträgt 8,06 nm bis zur Blauverschiebung. Im dritten Teil wurden die kombinierten thermischen Effekte von NMD und Si-Heizelementen berechnet. Die Silikonheizelementen verwenden TOE und verschieben das Spektrum um 3,8 nm in Rotverschiebung. Die Nettowellenverschiebung beträgt jedoch bei der Blauverschiebung 4,26 nm. Der Plasmadispersionseffekt dominiert den thermooptischen Effekt. Das Kompromissspektrum aufgrund des Einschaltzustands von NMD- und Si-Heizelementen wurde bestimmt. Am Ende werden die simulierten und geschätzten Messergebnisse von Si- und Aluminiumheizelementen überprüft. In dieser Arbeit konnte gezeigt werden, dass die hohe termische- und Abstimmeffizienz der n-dotierten Heizelemente eine niedrigere Temperatur im Chip sowie eine gleichmäßige Temperatur im Resonator erzeugen und weniger Energie verbrauchen. Die n-dotierten L- und U-förmigen Siliziumheizelemente sind geeignet, um das Spektrum im Betriebsbereich des NMD-FP-Modulator abzustimmen.
