Ultraviolet irradiation and actinomycin D impair the capacity of mouse embryo (ME) cells to support the replication of polyoma virus, but not of encephalomyocarditis (EMC) virus. The loss in capacity for polyoma virus synthesis was an "all-or-none" effect and followed closely upon the loss in cellular capacity for clone formation. Cells treated with either agent produced polyoma "T" antigen, but did not synthesize polyoma structural protein. Infection of untreated ME cells with polyoma virus produced marked stimulation of both deoxyribonucleic acid (DNA) synthesis and ribonucleic acid (RNA) synthesis. ME cell cultures irradiated with ultraviolet for 30 sec at 60 muw/cm(2) or treated with actinomycin D at 0.1 mug/ml for 6 hr prior to infection were incapable of synthesizing DNA or RNA, even after infection with polyoma virus. Irradiation of cells during infection produced cessation of synthesis of both RNA and DNA. Addition of actinomycin D during infection did not inhibit DNA synthesis but abolished RNA synthesis and reduced the yield of polyoma virus to 10% of that in untreated infected cultures. Both agents lost the ability to prevent replication of a full yield of polyoma virus when administered 30 hr after infection or later. The period after which neither agent inhibited polyoma replication corresponded with the period at which maximal RNA synthesis in untreated infected cultures had subsided. It can be concluded on the basis of the data presented that the functional integrity of the mouse embryo cell genome is required for the replication of polyoma virus, but not for EMC virus. Whereas the requirement for cellular DNA-dependent RNA synthesis for polyoma virus replication has been demonstrated, the exact nature of the host-cell function remains to be elucidated.

Neuraminidase (Nase) activity of chick embryo monolayer cell homogenates was determined by its rate of splitting of neuraminlactose, free neuraminic acid (NA) being determined by the thiobarbituric acid assay. Noninfected cells were found to have no detectable amount of Nase activity. Newcastle disease virus (NDV)-infected cells (multiplicity of infection, 20 to 75 plaque-forming units per cell) displayed a high level of Nase synthesis, the rate of synthesis being parallel to that of hemagglutinin (HA) synthesis (with a 1.5 hr delay in the latter). An "eclipse" of the Nase and HA activities associated with the virus that was adsorbed onto cells was observed. The data provide evidence that the Nase is not incorporated into the viral envelope from a pre-existing cell supply but that its synthesis is coded by the viral genome. The content of cell-bound sialic acid, determined simultaneously in infected-cell homogenates, showed characteristic features allowing certain conclusions concerning the renewal of NA-terminating cell receptors during the course of infection, and the intracellular action of the Nase of the virus introduced into cells by the inoculum and that of the newly synthesized Nase at different stages of infection.

Hemagglutinating and complement-fixing antigens of La Crosse virus (California arbovirus group) were produced in serum-free suspension cultures of BHK-21/13S cells. The appearance and production of these antigens were correlated with the titer of infectious virus. No significant differences in antigen titers were produced by varying virus dose 10-fold. Hemagglutinin appeared 6 to 8 hr after inoculation and reached peak titer in 14 to 22 hr. Both beta-propiolactone and Tween 80-ether treatment inactivated infectious virus in the antigens. Unlyophilized antigen was stable at -60, 5 and 24 C for at least 117 days but not for 1 year. Lyophilized antigen was stable for at least a year, however, at -20 and 5 C. Cell culture-produced antigen was more sensitive than brain-produced antigen in detecting hemagglutination inhibition antibody in human sera.

Cell envelope fractions from Salmonella can utilize exogenous lipid-linked intermediates for the synthesis of polymeric O-antigen. We describe a method for preparing aqueous suspensions of lipid intermediates and show that freezing and thawing of cell envelope-lipid intermediate mixtures is required for efficient synthesis. The lipid intermediates move freely in the hydrophobic environment of the membrane.

This paper describes the process by which the cell wall of Proteus mirabilis, as measured by the presence of the O antigen, develops during the differentiation of swarmers from short cells on an agar surface. The sequence was followed by fluorescent-antibody staining, with both the direct and reverse methods. When the organisms were labeled with fluorescent antibody by the direct method, they showed a progressive diminution of the marker along the cell surface and some increase in the length of the bacteria. However, the label had become completely diluted out before typical swarmers developed. When the bacteria were exposed initially to unlabeled antibody by the reverse technique, and then incubated with fluorescent antibody, they showed a progressive increase both in the intensity of the label along their entire periphery and in cellular length, culminating in the formation of swarmers. It is concluded that in P. mirabilis, as in the few other gram-negative bacteria examined so far, cell wall synthesis takes place diffusely, i.e., by intercalation of new with old components along the length of the cell wall.

Somatic (O) antigen 2 of Salmonella paratyphi A replaced somatic antigen 4 of an S. typhimurium recipient as the consequence of mating with an S. paratyphi A var. durazzo Hfr strain. The genetic determinants of these O antigens behaved in this cross as alleles of a common O locus, which is linked to the determinant of histidine biosynthesis, his. By employing phage lysates obtained by growth of P22 on an S. typhimurium hybrid which had received his and O-factor 2 determinants from the S. paratyphi A Hfr, it was possible to cotransduce the his and O-antigen 2 genes to both S. typhimurium and S. typhosa. S. typhimurium transductants which received somatic antigen 2 concurrently lost O-antigen 4, and S. typhosa transductants receiving O-antigen 2, lost their native O-antigen 9. These results indicate that the genetic determinants of O-antigens 2, 4, and 9 occupy the same O locus in S. paratyphi A, S. typhimurium, and S. typhosa, respectively, and are probably allelic.

Cytopathological, histochemical, and immunofluorescent studies. J. Bacteriol. 91:1977-1985. 1966.-Yaba tumor poxvirus synthesis in BSC-1 cell culture was followed sequentially with light microscopy, immunofluorescent microscopy, and various histochemical stains. The first evidence of infection was detected at 24 hr when nucleoli became hypertrophic, reflecting enhanced ribonucleic acid (RNA) synthesis. At 36 hr, deoxyribonucleic acid synthesis was detected in the cytoplasm. This was immediately followed by or associated with antigen synthesis at paranuclear sites and enhanced RNA synthesis in the cytoplasm. Cytoplasmic inclusions were readily apparent at 4 days in initially infected cells. Contiguous spread of virus was judged to have occurred around the third day of infection. The time required to complete the synthetic cycle from time of infection to production of infectious progeny was estimated to be of the order of 60 hr. This cycle is 6 to 10 times longer than for vaccinia virus. By light microscopy, cytopathic effects (CPE) were detectable in 5 days in heavily infected cultures. With 100 units or less of infectious virus, CPE was not readily detected until the 10th to 14th day. At this time, focal areas of infection classified either as microtumors or microplaques were present. Secondary foci appeared during the 4th week of incubation.

Induction of T antigen by adenovirus type 12 was studied in growing and growth-inhibited cultures of the Nil-2 line of Syrian hamster cells. At a viral input multiplicity of 10, neoantigen was present in 100% of the cells by 24 hr. T antigen gradually disappeared in descendants of these cells so that 2 weeks after infection only 1% gave specific immunofluorescence. When cellular replication was prevented by addition of fluorodeoxyuridine, T antigen persisted in all cells for the 2-week period. Upon infection of growing cultures with purified (3)H-labeled adenovirus type 12 and autoradiographic analysis of the cells at various times thereafter, a gradual reduction in labeled nuclear loci was noted which paralleled the decrease in T antigen-containing cells. In nongrowing cultures, no change in labeled loci was noted. Correlation of T antigen and labeled loci revealed that fluorescent cells contained, on the average, about 10 times more silver grains than nonfluorescent cells. All of 92 preselected fluorescent cells showed labeled loci, whereas, of 100 nonfluorescent cells, 18 were free of silver grains. The implications of these findings are discussed.

Methods are described for the preparation of complement-fixation (CF) and hemagglutination (HA) antigens from the Texas turkey ornithosis agent grown in McCoy cell culture monolayers. The particulate antigens prepared for this study were satisfactory for testing mammalian sera by direct CF tests and avian sera by indirect CF or modified direct CF tests. Comparison of titers were made on human, bovine and ovine sera using direct CF tests employing antigen prepared for this study, 6 BC yolk sac antigen, and a commercially available antigen. The HA antigen agglutinated mouse erythrocytes, but it was not of value in hemagglutination inhibition tests because of "nonspecific" inhibitors in both mammalian and avian sera.
