Benzhydrazone (BH) is an inhibitor of glycoprotein biosynthesis. It acts selectively in Herpes simplex virus (HSV)-infected cells and does not significantly affect glycoprotein synthesis in uninfected cells and in cells infected with other viruses. Previously, we reported on a syncytial (syn) mutant, designated HSV-1(13)S11, resistant to BH, and showed that BH-resistance is encoded in the mutant virus DNA and therefore can be transferred into the genome of wild type HSVs. The present paper reports on a preliminary mapping in HSV-1(13)S11 genome of the loci which confer resistance to BH and of three distinct syn mutations present simultaneously in this mutant. Two of them were mapped in previously described syn loci localized in BamHI fragment L (map units 0.707-0.745) (locus syn 1) and BamHI fragment Q (map units 0.296-0.317) (locus syn 5). A third mutation not described before and mapping in BamHI fragment SP (c.a. map units 0.81-0.85) conferred the syn phenotype to both HEp-2 and Vero cells. This novel mutation has been designated herein locus syn 6. Transfer of BH-resistance could be achieved in cotransfection experiments involving two HSV-1(13)S11 fragments, BamHIL and BamHISP.

The pattern of ocular disease produced in the rabbit eye by HSV-1 (F) and HSV-1(MP) strains and recombinants F(MP)A, F(MP)B, F(MP)C, F(MP)D, F(MP)E, and F(MP)F was studied. The characteristics of ocular herpetic disease such as morphology of dendritic ulcers, severity of epithelial disease and incidence and duration of stromal disease produced in the rabbit eye are genetically determined by the virus strain. Our studies show that transfer of a defined part of the genome of the stromal disease-producing virus, HSV-1(MP), to the genome of an epithelial disease-producing virus, HSV-1(F), yielded recombinants with one or more of the disease characteristics of the donor strain. Specifically, recombinant F(MP)D produced lesions characteristic of the donor HSV-1(MP) strain; recombinants F(MP)C and F(MP)E produced stromal disease approaching the severity of the disease produced by the donor HSV-1(MP) strain, and only recombinants F(MP)A and F(MP)B retained the typically elongate lesions of the recipient HSV-1(F), whereas the recombinant strain F(MP)F produced no disease. The viral functions pertaining to the ocular disease pattern map between 0.70 and 0.83 map units in HSV-1 DNA within the BglII F DNA fragment. The pattern of stromal disease is independent of the production of glycoprotein C and fusion of HEp-2-infected cells. The functions relating to these aspects of ocular disease segregate but are closely linked.

This paper reports on the variability of structural polypeptides of 32 strains of Herpes simplex virus 2 (HSV-2) isolated from various locations in the United States and Italy. Most strains were passaged a limited number of times at low multiplicity outside the human host; a few strains were characterized by numerous passages at variable multiplicities in cell culture. The acrylamide gel electrophoresis of polypeptides from purified virions revealed very few differences. Considering these differences the HSV-2 strains analysed can be classified into 3 groups according to the variability of three polypeptides. The study of polypeptides in HSV-2 infected cells revealed more differences than those seen in the structural proteins. Comparisons of the structural proteins specified by HSV-2 and HSV-1 virions, revealed clear variant features of the electrophoretic profiles, especially when pH of main gel was 8.9.

The possible involvement of herpes simplex virus type 1 (HSV-1) glycoprotein C (gC) in the maintenance of persistence in vitro and the establishment of latency in the mouse was investigated by the use of virus mutans which do not express glycoprotein C (gC-). A persistent infection of MDBK cells could be established with 3 of 3 gC- mutans derived from HSV-1 (MP) and with 1 of 2 gC- mutans derived from HSV-1 (HFEM)H6; cells infected with the other gC- mutant derived from HSV-1(HFEM)H6 survived for 25 days only. Of the 4 lines of persistence obtained, only one showed the appearance of revertants expressing gC (gC+). gC+ revertants, which appeared also in the "short term infection", rapidly became the majority population once they had arisen. gC- mutans could establish latency in mice, and the virus recovered from the latently infected ganglia maintained its gC- phenotype. The results show that expression of gC is not essential for the maintenance of persistence in MDBK cells and for the establishment of latency in the mouse. In addition, the results indicate that MDBK cells could offer a model to study the role of gC in the virus replication cycle in vitro.

Virus clones which express glycoprotein gC (gC+) were obtained from two persistently infected (p.i.) MDBK cell lines which had been independently established by infection with HSV-1(MP)10311, a gC- syncytial (syn) variant of herpes simplex virus type 1 strain MP [HSV-1(MP)]. The gC+ revertants were syn in MDBK, HEp-2, and Vero cell lines and in primary human fibroblasts; this offers further evidence that glycoprotein gC does not inhibit cell fusion. The gC+ revertants represented from 70 to 100 percent of the virions present in the virus populations examined, thus suggesting a possible selective advantage of the gC+ revertants in this system of persistent infection.

DNA sequences homologous to HSV-1 and HSV-2 DNA fragments were searched in 64 genital, 35 labial and 34 cerebral tumors. Southern blot transfers of tumor and control DNAs were hybridized in stringent conditions with 32P labelled probes from HSV-1 and HSV-2 cloned DNA fragments. Specific hybridization to HSV-2 BglII N fragment was observed in six (9.4%) genital tumors. Labial and cerebral tumors did not show hybridization to any of the probes used. The technique employed allowed the detection of 0.1 copies of viral fragments per diploid genome.

Restriction enzyme analysis was utilized to investigate 11 HSV isolates from patients with herpetic keratitis. The electrophoretic profiles of the DNAs indicate that the isolates belong to HSV-1 and are epidemiologically unrelated. Two of the isolates, derived from a patient with simultaneous bilateral herpetic keratitis were further characterized by genetic and phenotypic analyses. These studies showed the following: (i) the two recrudescent lesions were caused by the same HSV-1 isolate; (ii) the isolate yielded two different plaque phenotypes; (iii) these plaques showed different sensitivity to I.D.U.

In this paper we report on the preliminary characterization of a mutant of herpes simplex virus type 1 (HSV-1) selected for acycloguanosine (acyclovir, ACV) resistance in vitro. The ACVr virus was examined for a series of parameters that include chemosensitivity assay, thymidine kinase (TK) activity, in vitro and in vivo growth, and mutation mapping. The data obtained indicate that a mutated TK gene is responsible for the ACVr phenotype. A distinctive feature of this mutant is the high level of resistance exhibited to ACV (100 microM) and the concomitant presence of a functional TK activity. Such a property makes this virus useful as a model for the study of viral resistance to nucleoside-type analogues in HSV.

The analysis of 23 clinical isolates of herpes simplex virus type 1 (HSV-1) showed that 15 of 15 isolates that had undergone a few passages in tissue culture (fresh isolates) and two of eight isolates that had never been passaged (new isolates) were composed of a mixed population with respect to plaque morphology in Vero cells. Cloning and characterization of 10 large plaque viruses (L variants) and nine small plaque viruses (S variants), obtained from seven different isolates, showed the following. BamHI DNA restriction patterns of the L and the S variants from a single isolate differed only with respect to the electrophoretic mobility of the fragments that contain reiteration of specific sequences; they did not differ regarding the presence or the absence of restriction endonuclease cleavage sites. The L and S variants differed with respect to the electrophoretic profiles of infected cell glycoproteins, thermosensitivity of growth and plaquing efficiency at 39 degrees C, and, at least in the case of the two couples of variants that we tested, pathogenicity for the mouse. The hypothesis that the L variants might arise from the S variant during in vivo replication is discussed.

In herpes simplex virus-infected cells, coreless capsids accumulate at the nuclear pores soon after infection, but subsequently disappear, suggesting that, as in adenovirus-infected cells (S. Dales and Y. Chardonnet, Virology 56:465-483, 1973), the release of viral DNA from nucleocapsids takes place at the nuclear pores. A nonlethal mutant, HSV-1(50B), produced by mutagenesis of HSV DNA fragments and selected for delayed production of plaques at 31 degrees C, accumulated coreless capsids at the nuclear pores late in infection in contrast to wild-type viruses. Recombinants selected for ability to produce plaques at 31 degrees C by marker rescue with digests of herpes simplex virus 2 DNA and selected clone fragments of HSV-1 DNA no longer accumulated empty capsids at nuclear pores late in infection. These results suggest that herpes simplex viruses encode a function which prevents accumulation of coreless capsids at nuclear pores, presumably by preventing uptake, unenvelopment, and DNA release from progeny virus, and indicate that the cold sensitivity of plaque formation and accumulation of coreless capsids might be related or comap in the S component of the genome.

The biochemical and functional properties of the thymidine kinase (TK) of the herpes simplex virus type 1 mutant R100, that is highly resistant to 9-(2-hydroxyethoxymethyl)guanine (acyclovir), are reported in comparison with the properties of its parental strain, wt. The mutant induced the production of a TK activity that accounted for only 10% of the wt one. This feature was not apparently related to a defective expression of the TK gene but it was rather connected to some functional characteristics of R100 enzyme. Although affinities of this enzyme for ATP and thymidine were unchanged, apparent Vmax values for thymidine were much reduced. In addition, affinities for antiviral analogues acyclovir, 9-(1,3-dihydroxymethyl)guanine (DHPG), 5-(2-bromovinyl)2'-deoxyuridine (BVdU), and 5-iodo-2'deoxycytidine (IdCyd) were drastically diminished (between 50-fold and more than 100-fold). This mutation therefore seems to affect the active site of the enzyme which is involved in the catalytic conversion of thymidine and in the binding of the analogues. The above features of HSV-1 R100 seem quite distinct from those of previously described HSV-1 resistant mutants.

Some hypotheses of the mechanism of action of benzhydrazone, a selective inhibitor of herpes simplex virus glycoprotein biosynthesis, are presented in this paper. Comparative analysis of different models indicates that the drug may interfere with herpes simplex virus DNA. Thus, benzhydrazone seems to act like a bifunctional alkylating agent.
