This paper presents a survey on the current state-of-the-art in Wireless Sensor Network (WSN) Operating Systems (OSs). In recent years, WSNs have received tremendous attention in the research community, with applications in battlefields, industrial process monitoring, home automation, and environmental monitoring, to name but a few. A WSN is a highly dynamic network because nodes die due to severe environmental conditions and battery power depletion. Furthermore, a WSN is composed of miniaturized motes equipped with scarce resources e.g., limited memory and computational abilities. WSNs invariably operate in an unattended mode and in many scenarios it is impossible to replace sensor motes after deployment, therefore a fundamental objective is to optimize the sensor motes’ life time. These characteristics of WSNs impose additional challenges on OS design for WSN, and consequently, OS design for WSN deviates from traditional OS design. The purpose of this survey is to highlight major concerns pertaining to OS design in WSNs and to point out strengths and weaknesses of contemporary OSs for WSNs, keeping in mind the requirements of emerging WSN applications. The state-of-the-art in operating systems for WSNs has been examined in terms of the OS Architecture, Programming Model, Scheduling, Memory Management and Protection, Communication Protocols, Resource Sharing, Support for Real-Time Applications, and additional features. These features are surveyed for both real-time and non-real-time WSN operating systems.

Satellite data suggest that summertime aerosol optical depth (AOD) over the southeastern USA depends on the air/land surface temperature, but the magnitude of the radiative effects caused by this dependence remains unclear. To quantify these radiative effects, we utilized several remote sensing datasets and ECMWF reanalysis data for the years 2005–2011. In addition, the global aerosol–climate model ECHAM-HAMMOZ was used to identify the possible processes affecting aerosol loads and their dependence on temperature over the studied region. The satellite-based observations suggest that changes in the total summertime AOD in the southeastern USA are mainly governed by changes in anthropogenic emissions. In addition, summertime AOD exhibits a dependence on southerly wind speed and land surface temperature (LST). Transport of sea salt and Saharan dust is the likely reason for the wind speed dependence, whereas the temperature-dependent component is linked to temperature-induced changes in the emissions of biogenic volatile organic compounds (BVOCs) over forested regions. The remote sensing datasets indicate that the biogenic contribution increases AOD with increasing temperature by approximately (7 ± 6) × 10−3 K−1 over the southeastern USA. In the model simulations, the increase in summertime AOD due to temperature-enhanced BVOC emissions is of a similar magnitude, i.e., (4 ± 1) × 10−3 K−1. The largest source of BVOC emissions in this region is broadleaf trees, thus if the observed temperature dependence of AOD is caused by biogenic emissions the dependence should be the largest in the vicinity of forests. Consequently, the analysis of the remote sensing data shows that over mixed forests the biogenic contribution increases AOD by approximately (27 ± 13) × 10−3 K−1, which is over four times higher than the value for over the whole domain, while over other land cover types in the study region (woody savannas and cropland/natural mosaic) there is no clear temperature dependence. The corresponding clear-sky direct radiative effect (DRE) of the observation-based biogenic AOD is −0.33 ± 0.29 W/m2/K for the whole domain and −1.3 ± 0.7 W/m2/K over mixed forests only. The model estimate of the regional clear-sky DRE for biogenic aerosols is similar to the observational estimate for the whole domain: −0.29 ± 0.09 W/m2/K. Furthermore, the model simulations showed that biogenic emissions have a significant effective radiative forcing (ERF) in this region: −1.0 ± 0.5 W/m2/K.

A controlled ammonia (NH3) release experiment was performed at a grassland site. The aim was to quantify the effect of dry deposition between the source and the receptors (NH3 measurement locations) on emission rate estimates by means of inverse dispersion modelling. NH3 was released for three hours at a constant rate of Q = 6.29 mg s−1 from a grid of 36 orifices spread over an area of 250 m2. The increase in line-integrated NH3 concentration was measured with open-path optical miniDOAS devices at different locations downwind of the artificial source. Using a backward Lagrangian stochastic (bLS) dispersion model (bLSmodelR), the fraction of the modelled release rate to the emitted NH3 ( Q bLS / Q ) was calculated from the measurements of the individual instruments. Q bLS / Q was found to be systematically lower than 1, on average between 0.69 and 0.91, depending on the location of the receptor. We hypothesized that NH3 dry deposition to grass and soil surfaces was the main factor responsible for the observed depletion of NH3 between source and receptor. A dry deposition algorithm based on a deposition velocity approach was included in the bLS modelling. Model deposition velocities were evaluated from a ‘big-leaf’ canopy resistance analogy. Canopy resistances (generally termed R c ) that provided Q bLS / Q = 1 ranged from 75 to 290 s m−1, showing that surface removal of NH3 by dry deposition can plausibly explain the original underestimation of Q bLS / Q . The inclusion of a dry deposition process in dispersion modelling is crucial for emission estimates, which are based on concentration measurements of depositing tracers downwind of homogeneous area sources or heterogeneously-distributed hot spots, such as, e.g., urine patches on pastures in the case of NH3.

The physico-chemical characteristics of 47 lakes from two regions in the lowlands of Central Yakutia (Vilyuysk and Yakutsk), Eastern Siberia were analysed. The aims of this study were (i) to classify the lakes according to their ionic compositions and their nutrient concentrations, (ii) to quantify environmental gradients representing the main directions of variation in the measured variables, and (iii) to explore the relationship between the investigated lakes and their spatial positions. Most of the study lakes are shallow, thermokarst lakes that are slightly alkaline to alkaline. The lakes are predominantly oligotrophic, with some mesotrophic and a few eutrophic exceptions in the study region Vilyuysk. There are four hypertrophic lakes in the study region Yakutsk which are strongly affected by anthropogenic inputs and, in one case, additionally by inputs of water birds. Most part of the variance in the data is represented by major ion concentrations and related variables such as electrical conductivity. There were clear differences in these variables between the lakes of both study regions partly due to regional differences in the climate-induced, negative water balance (i.e. evaporation exceeds precipitation). The statistical analysis has shown that a significant part of the variance can be attributed to the type of vegetation in the lake's catchment (11%), to the longitude (7.6%) and to local spatial differences in the lake water chemistry (2.8%). Hypothesis testing indicated that there are significant differences in the mean values of many variables according to vegetation type and to the region where the lakes are located. However, the hypothesis of spatial autocorrelation in the data had to be rejected. The results presented here have important implications for ongoing and future limnological and paleoenvironmental studies in Yakutia. The exploratory analysis has shown that the physico-chemical characteristics of Central Yakutian lakes are mainly influenced by vegetation and climate driven changes that provides the basis for paleoenvironmental studies.

Obst und Gemüse wirken sich positiv auf die Gesundheit aus. Dazu tragen auch die mit Pflanzen assoziierten Mikro­organismen bei. Allerdings kann Obst oder Gemüse, wenn es mit humanpathogenen Keimen kontaminiert ist und roh verzehrt wird, auch Lebensmittelvergiftungen verursachen. In diesem Übersichtsartikel zeigen wir anhand einer Literaturstudie, wie humanpathogene Stämme von Escherichia coli oder Salmonella enterica auf Obst und Gemüse gelangen können und dass ein kom­plexes Zusammenspiel verschiedener biotischer und abiotischer Faktoren ihr Überleben im Boden und auf Pflanzen beeinflusst. Insbesondere mobile genetische Elemente, die durch horizontalen Gentransfer erhalten werden, tragen zur Diversität der pathogenen Eigenschaften, aber auch zur Anpassung an Habitate außerhalb des Darms bei. Wichtig für die erfolgreiche Etablierung von Humanpathogenen an der Pflanze ist deren Anhaftung und Aufnahme sowie die Reak­tion der Pflanze. Die komplexe Interaktion von Human­pathogenen und Pflanze wird wesentlich durch die genetische Ausstattung des Humanpathogens, vom Pflanzengenotyp und -mikrobiom, aber auch von einer Vielzahl von Umweltfaktoren bestimmt. Für das Überleben von Humanpathogenen ist insbesondere die Verfügbarkeit von Nährstoffen kritisch, um die sie mit der natürlichen Mikroflora konkurrieren. Empfehlungen für die Praxis setzen ein besseres Verständnis der Ökologie von Humanpathogenen im Boden und an Pflanzen, aber auch entlang der Produktkette voraus. Dafür sind die Entwicklung und Nutzung empfindlicher und spezifischer Nachweistechniken, die sowohl die mögliche Diversifizierung der Humanpathogene durch horizontalen Gentransfer als auch das Problem der Nichtkultivierbarkeit berücksichtigen, besonders dringlich. We prospectively evaluated the activity and tolerance of lenalidomide-dexamethasone in 35 patients with acute light chain-induced renal failure. The lenalidomide dose was adapted to the estimated glomerular filtration rate and dexamethasone was given at high dose in cycle one and at low dose thereafter. Four patients died within the first two cycles, and five discontinued therapy leaving 26 patients for the per-protocol analysis. Responses were observed in 24/35 (68.6%) patients of the intent-to-treat population. Complete response was noted in seven patients (20%), very good partial response in three patients (8.6%), partial response in 14 patients (40%), and minimal response in one patient (2.9%). Renal response was observed in 16 (45.7%) patients: five (14.2%) achieved complete, four (11.4%) partial and seven (20%) minor renal responses. Five of 13 patients who were dialysis dependent at baseline became dialysis independent. The median time to myeloma and to renal response was 28 days for both parameters, while the median time to best myeloma and best renal response was 92 and 157 days, respectively. The median estimated glomerular filtration rate increased significantly in patients with partial response or better from 17.1 mL/min at baseline to 39.1 mL/min at best response (P=0.001). The median progression-free and overall survival was 5.5 and 21.8 months, respectively, in the intent-to-treat population and 12.1 and 31.4 months, respectively, in the per-protocol group. Infections, cardiotoxicity, anemia and thrombocytopenia were the most frequent toxicities. In conclusion, the lenalidomide-dexamethasone regimen achieved rapid and substantial myeloma and renal responses. The trial was registered under EUDRACT number 2008-006497-15.

We highlight different important factors that must be considered for an effective available-bandwidth-based flow admission control algorithm in ad hoc wireless networks. Moreover, we present BandEst; it is a combination of a measurement-based available bandwidth estimation technique and a flow admission control algorithm for ad hoc IEEE 802.15.4-based ad hoc networks that considers the identified factors. Extensive simulations are performed to compare BandEst with the state-of-the-art available-bandwidth-based flow admission control algorithms for ad hoc wireless networks. Our simulation results demonstrate that BandEst significantly outperforms the state-of-the-art available-bandwidth-based flow admission control algorithms for ad hoc wireless networks.

Allergen immunotherapy (AIT) is widely used in clinical practice for patients with moderate to severe allergic rhinitis due to inhalant allergens and may be delivered via subcutaneous (SCIT) and sublingual routes (SLIT). However, the quality of evidence for individual AIT products is very heterogeneous, and extensions of overall conclusions (“class effects”) on the efficacy and disease-modifying effects to all AIT products are unjustified. In contrast, each product needs to be evaluated individually, based on available study results, to justify efficacy and specific claims on sustained and disease modifying effects per allergen and targeted patient group (children vs. adults, allergic rhinitis vs. asthma). WAO intends to support the current development to evidence-based AIT, which ultimately will lead to a more efficacious treatment of allergic patients and the appropriate recognition of AIT.

OBJECTIVE:It was the aim of our study to evaluate the independent effect of preterm prelabor rupture of membranes (PPROM) as a cause of preterm delivery on mortality during primary hospital stay and significant morbidities in very-low-birth-weight (VLBW) infants < 32 weeks of gestation. DESIGN:Observational, epidemiological study design. SETTING:Population-based cohort, German Neonatal Network (GNN). POPULATION:6102 VLBW infants were enrolled in GNN from 2009-2012, n=4120 fulfilled criteria for primary analysis (< 32 gestational weeks, no pre-eclampsia, HELLP (highly elevated liver enzymes and low platelets syndrome) or placental abruption as cause of preterm birth). METHODS:Multivariable logistic regression analyses included PPROM as potential risk factors for adverse outcomes and well established items such as gestational age in weeks, birth weight, antenatal steroids, center, inborn delivery, multiple birth, gender and being small-for-gestational-age. RESULTS:PPROM as cause of preterm delivery had no independent effect on the risk of early-onset sepsis, clinical sepsis and blood-culture proven sepsis, while gestational age proved to be the most important contributor to sepsis risk. The diagnosis of PPROM was associated with an increased risk for bronchopulmonary dysplasia (BPD; OR: 1.25, 95% CI: 1.02-1.55, p=0.03) but not with other major outcomes. CONCLUSIONS:The diagnosis of PPROM per se is not associated with adverse outcome in VLBW infants < 32 weeks apart from a moderately increased risk for BPD. Randomized controlled trials with primary neonatal outcomes are needed to determine which subgroup of VLBW infants benefit from expectant or intentional management of PPROM.

Substrate dephosphorylation by the cyclin-dependent kinase (Cdk)-opposing phosphatase, Cdc14, is vital for many events during budding yeast mitotic exit. Cdc14 is sequestered in the nucleolus through inhibitory binding to Net1, from which it is released in anaphase following Net1 phosphorylation. Initial Net1 phosphorylation depends on Cdk itself, in conjunction with proteins of the Cdc14 Early Anaphase Release (FEAR) network. Later on, the Mitotic Exit Network (MEN) signaling cascade maintains Cdc14 release. An important unresolved question is how Cdc14 activity can increase in early anaphase, while Cdk activity, that is required for Net1 phosphorylation, decreases and the MEN is not yet active. Here we show that the nuclear rim protein Nur1 interacts with Net1 and, in its Cdk phosphorylated form, inhibits Cdc14 release. Nur1 is dephosphorylated by Cdc14 in early anaphase, relieving the inhibition and promoting further Cdc14 release. Nur1 dephosphorylation thus describes a positive feedback loop in Cdc14 phosphatase activation during mitotic exit, required for faithful chromosome segregation and completion of the cell division cycle.

Particle fluxes were measured from 2000 to 2001 with 3 integrating open traps (O-traps) and a sequencing trap (S-trap) in the 73-m deep, oligotrophic, high-mountain Estany (Lake) Redó (2240 m a.s.l.) over a period of 558 days. O-traps were deployed at 26, 46, and 66 m water depth to measure overall sedimentation rates, while the S-trap was deployed at 66 m water depth to detect dynamics of seasonal particle fluxes with a resolution of 4 days (during ice break-up, summer, ice formation) to 21 days (during ice cover). Our results show a high degree of seasonal variability in particle dynamics. Total particle fluxes vary from almost zero to more than 600 mg m-2 d-1. The highest fluxes occur during short time windows after ice-break-up (minerogenic particles), during spring (planktonic biomass), and during fall overturn (chrysophycean cysts). Particle fluxes also differed markedly from year to year in absolute values (2000: 644 mg m-2 d-1, 2001: 370 mg m-2 d-1) as well as in average values (2000: 76 mg m-2 d-1, 2001: 44 mg m-2 d-1). Annual and seasonal meteorological changes and events have a clear influence on the lake system and on the amount and composition of particles. C/N ratios during April and May increased significantly from 2000 (6-14) to 2001 (>28), reflecting the more intense soil erosion and transport of terrestrial plant remains into the lake caused by heavy precipitation in 2001. Air temperature strongly influences the timing of the occurrence of the main bio-productivity peak. Strong wind events shorten the period of ice cover. Our investigation shows that sediment trap studies lasting more than one limnological cycle are useful in studying the effects of short-term meteorological changes and weather events on high mountain lakes. However, long-term particle flux measurements would be necessary to determine amplitudes of natural seasonal cycles and for the interpretation of the decadal-scale environmental changes occurring in such lakes.

Background Electronic cigarettes (e-cigarettes) are advertised to tobacco users as a tool to decrease cigarette consumption and to reduce toxic exposure associated with conventional tobacco smoking. Little is known about the compounds contained in such products, their exposure and long-term health effects. Methods NMR spectroscopy was used to ascertain the content of several constituents of e-cigarette liquids including nicotine, solvents and some bioactive flavour compounds. Risk assessment was based on probabilistic exposure estimation and comparison with toxicological thresholds using the margin of exposure (MOE) approach. Results In 54 samples of e-cigarette liquids, the average nicotine content was 11 mg/ml. Only 18 from 23 samples were confirmed as nicotine-free samples and in one e-cigarette liquid nicotine was not detected while being declared on the labelling. Major compounds of e-cigarette liquids include glycerol (average 37 g/100 g), propylene glycol (average 57 g/100 g) and ethylene glycol (average 10 g/100 g). Furthermore, 1,3-propanediol, thujone and ethyl vanillin were detected in some samples. The average exposure for daily users was estimated as 0.38 mg/kg bw/day for nicotine, 8.9 mg/kg bw/day for glycerol, 14.5 mg/kg bw/day for 1,2-propanediol, 2.1 mg/kg bw/day for ethylene glycol, and below 0.2 mg/kg bw/day for the other compounds. The MOE was below 0.1 for nicotine, but all other compounds did not reach MOE values below 100 except ethylene glycol and 1,2-propanediol. Conclusions NMR spectroscopy is a useful and rapid method to simultaneously detect several ingredients in e-cigarette liquids. From all compounds tested, only nicotine may reach exposures that fall into a high risk category with MOE

Floating covers used in waste water treatment plants are one of the many structures formed with membrane materials. These structures are usually large and can spread over an area measuring 470 m × 170 m. The aim of this paper is to describe recent work to develop an innovative and effective approach for structural health monitoring (SHM) of such large membrane-like infrastructure. This paper will propose a potentially cost-effective non-contact approach for full-field strain and stress mapping using an unmanned aerial vehicle (UAV) mounted with a digital camera and a global positioning system (GPS) tracker. The aim is to use the images acquired by the UAV to define the geometry of the floating cover using photogrammetry. In this manner, any changes in the geometry of the floating cover due to forces acting beneath resulting from its deployment and usage can be determined. The time-scale for these changes is in terms of weeks and months. The change in the geometry can be implemented as input conditions to a finite element model (FEM) for stress prediction. This will facilitate the determination of the state of distress of the floating cover. This paper investigates the possibility of using data recorded from a UAV to predict the strain level and assess the health of such structures. An investigation was first conducted on a laboratory sized membrane structure instrumented with strain gauges for comparison against strains, which were computed from 3D scans of the membrane geometry. Upon validating the technique in the laboratory, it was applied to a more realistic scenario: an outdoor test membrane structure and capable UAV were constructed to see if the shape of the membrane could be computed. The membrane displacements were then used to calculate the membrane stress and strain, state demonstrating a new way to perform structural health monitoring on membrane structures.

In this contribution, we present a wavelength-sensitive method for the detection of extreme ultraviolet (XUV) photon energies between 30 eV and 120 eV. The method is based on 45° reflectivity from either a cesium iodide-coated or an uncoated metal surface, which directs the XUV beam onto an electron or ion detector and its signal is used to monitor the XUV beam. The benefits of our approach are a spectrally sensitive diagnosis of the XUV radiation at the interaction place of time-resolved XUV experiments and the detection of infrared leak light though metal filters in high-harmonic generation (HHG) experiments. Both features were tested using spectrally shaped XUV pulses from HHG in a capillary, and we have achieved excellent agreement with XUV spectrometer measurements and reflectivity calculations. Our obtained results are of interest for time-resolved XUV experiments presenting an additional diagnostic directly in the interaction region and for small footprint XUV beamline diagnostics.

The purpose of this study was to examine changes in haematopoietic stem cell transplant (HSCT) characteristics and outcome in our combined paediatric and adult programme over the past four decades, since its implementation in 1973. The total number of transplant procedures rose from 109 in the first decade (1973–82) to 939 in the last decade (2003–12). Transplant characteristics changed significantly over time: patient age increased, peripheral blood largely replaced bone marrow as stem cell source, unrelated donors became an alternative to matched siblings, and patients are increasingly transplanted in more advanced disease stages. Advances such as improved supportive care and histocompatibility typing resulted in a steady decrease of transplant-related mortality after allogeneic HSCT (43% in the first decade, 22% in the last decade). Despite this, unadjusted survival rates were stable in the last three decades for allogeneic HSCT (approximately 50% 5–year survival) and in the last two decades for autologous HSCT (approximately 60% 5–year survival). After adjustment for covariates such as donor type, age and stage, the relative risk of treatment failure continuously dropped (for allogeneic HSCT: first decade 1.0, second decade 0.58, third decade 0.51, last decade 0.41). Collectively, these data suggest that improvements in peri- and post-transplant care have allowed considerable extension of transplant indications without having a negative impact on outcome.

Cell-free DNA (cfDNA) in body tissues or fluids is extensively investigated in clinical medicine and other research fields. In this article we provide a direct quantitative real-time PCR (qPCR) as a sensitive tool for the measurement of cfDNA from plasma without previous DNA extraction, which is known to be accompanied by a reduction of DNA yield. The primer sets were designed to amplify a 90 and 222 bp multi-locus L1PA2 sequence. In the first module, cfDNA concentrations in unpurified plasma were compared to cfDNA concentrations in the eluate and the flow-through of the QIAamp DNA Blood Mini Kit and in the eluate of a phenol-chloroform isoamyl (PCI) based DNA extraction, to elucidate the DNA losses during extraction. The analyses revealed 2.79-fold higher cfDNA concentrations in unpurified plasma compared to the eluate of the QIAamp DNA Blood Mini Kit, while 36.7% of the total cfDNA were found in the flow-through. The PCI procedure only performed well on samples with high cfDNA concentrations, showing 87.4% of the concentrations measured in plasma. The DNA integrity strongly depended on the sample treatment. Further qualitative analyses indicated differing fractions of cfDNA fragment lengths in the eluate of both extraction methods. In the second module, cfDNA concentrations in the plasma of 74 coronary heart disease patients were compared to cfDNA concentrations of 74 healthy controls, using the direct L1PA2 qPCR for cfDNA quantification. The patient collective showed significantly higher cfDNA levels (mean (SD) 20.1 (23.8) ng/ml; range 5.1-183.0 ng/ml) compared to the healthy controls (9.7 (4.2) ng/ml; range 1.6-23.7 ng/ml). With our direct qPCR, we recommend a simple, economic and sensitive procedure for the quantification of cfDNA concentrations from plasma that might find broad applicability, if cfDNA became an established marker in the assessment of pathophysiological conditions.

Freshwater ecosystems are increasingly impacted by alien invasive species which have the potential to alter various ecological interactions like predator-prey and host-parasite relationships. Here, we simultaneously examined predator-prey interactions and parasitization patterns of the highly invasive round goby (Neogobius melanostomus) in the rivers Rhine and Main in Germany. A total of 350 N. melanostomus were sampled between June and October 2011. Gut content analysis revealed a broad prey spectrum, partly reflecting temporal and local differences in prey availability. For the major food type (amphipods), species compositions were determined. Amphipod fauna consisted entirely of non-native species and was dominated by Dikerogammarus villosus in the Main and Echinogammarus trichiatus in the Rhine. However, the availability of amphipod species in the field did not reflect their relative abundance in gut contents of N. melanostomus. Only two metazoan parasites, the nematode Raphidascaris acus and the acanthocephalan Pomphorhynchus sp., were isolated from N. melanostomus in all months, whereas unionid glochidia were only detected in June and October in fish from the Main. To analyse infection pathways, we examined 17,356 amphipods and found Pomphorhynchus sp. larvae only in D. villosus in the river Rhine at a prevalence of 0.15%. Dikerogammarus villosus represented the most important amphipod prey for N. melanostomus in both rivers but parasite intensities differed between rivers, suggesting that final hosts (large predatory fishes) may influence host-parasite dynamics of N. melanostomus in its introduced range.

A goal in human embryonic stem cell (hESC) research is the faithful differentiation to given cell types such as neural lineages. During embryonic development, a basement membrane surrounds the neural plate that forms a tight, apico-basolaterally polarized epithelium before closing to form a neural tube with a single lumen. Here we show that the three-dimensional epithelial cyst culture of hESCs in Matrigel combined with neural induction results in a quantitative conversion into neuroepithelial cysts containing a single lumen. Cells attain a defined neuroepithelial identity by 5 days. The neuroepithelial cysts naturally generate retinal epithelium, in part due to IGF-1/insulin signaling. We demonstrate the utility of this epithelial culture approach by achieving a quantitative production of retinal pigment epithelial (RPE) cells from hESCs within 30 days. Direct transplantation of this RPE into a rat model of retinal degeneration without any selection or expansion of the cells results in the formation of a donor-derived RPE monolayer that rescues photoreceptor cells. The cyst method for neuroepithelial differentiation of pluripotent stem cells is not only of importance for RPE generation but will also be relevant to the production of other neuronal cell types and for reconstituting complex patterning events from three-dimensional neuroepithelia.

BACKGROUND: Prognostic factors of melanoma with distant metastasis and systemic treatment are only poorly established. This study aimed to analyse the impact of S100B, lactate dehydrogenase (LDH) and the type of treatment on survival in advanced patients receiving systemic treatment. PATIENTS AND METHODS: We analysed overall survival of 499 patients from the university department of dermatology in Tuebingen, Germany, with unresectable melanoma at the time point of initiation of first-line systemic therapy. Only patients who started treatment between the years 2000 and 2010 were included. Disease-specific survival was calculated by bivariate Kaplan Meier survival probabilities and multivariate Cox hazard regression analysis. The ultrastructural characterization of neuronal compartments in intact tissue labeled with green fluorescent protein (GFP) remains a frequently encountered challenge, despite work establishing photooxidation of GFP in cultured cells. However, most applications require the detection of GFP or GFP fusion proteins expressed in intact tissue. Here, we report that illumination of GFP variants in oxygen-enriched environment reliably generated electron-dense 3,3'-diaminobenzidine (DAB) precipitates in slices from rat brain. The method is applicable to GFP variants tagged to presynaptic proteins as well as to soluble GFP in various brain regions. Serial section scanning electron microscopy was used to examine genetically labeled presynaptic terminals at high resolution and to generate three-dimensional representations of the synapses. Thus, we introduce a generally applicable correlative approach for the identification of presynaptic terminals genetically labeled with green fluorescent proteins in tissue slices and their ultrastructural characterization.

The calyx of Held, a large glutamatergic terminal in the mammalian auditory brainstem has been extensively employed to study presynaptic structure and function in the central nervous system. Nevertheless, the nanoarchitecture of presynaptic proteins and subcellular components in the calyx terminal and its relation to functional properties of synaptic transmission is only poorly understood. Here, we use stimulated emission depletion (STED) nanoscopy of calyces in thin sections of aldehyde-fixed rat brain tissue to visualize immuno-labeled synaptic proteins including VGluT1, synaptophysin, Rab3A and synapsin with a lateral resolution of approximately 40 nm. Excitation multiplexing of suitable fluorescent dyes deciphered the spatial arrangement of the presynaptic phospho-protein synapsin relative to synaptic vesicles labeled with anti-VGluT1. Both predominantly occupied the same focal volume, yet may exist in exclusive domains containing either VGluT1 or synapsin immunoreactivity. While the latter have been observed with diffraction-limited fluorescence microscopy, STED microscopy for the first time revealed VGluT1-positive domains lacking synapsins. This observation supports the hypothesis that molecularly and structurally distinct synaptic vesicle pools operate in presynaptic nerve terminals.

BACKGROUND: A direct comparison of prognosis between patients with regional lymph node metastases (LNM) detected synchronously with the primary melanoma (primary LNM), patients who developed their first LNM subsequently (secondary LNM) and those with initial LNM in melanoma with unknown primary site (MUP) is missing thus far. PATIENTS AND METHODS: Survival of 498 patients was calculated from the time point of the first macroscopic LNM using Kaplan Meier and multivariate Cox hazard regression analysis. RESULTS: Patients with secondary LNM (HR = 0.67; p = 0.009) and those with initial LNM in MUP (HR = 0.45; p = 0.008) had a better prognosis compared to patients with primary LNM (median survival time 52 and 65 vs. 24 months, respectively). A high number of involved nodes, the presence of in-transit/satellite metastases and male gender had an additional independent unfavourable effect. CONCLUSIONS: Survival of patients with LNM in MUP and with secondary LNM is similar and considerably more favourable compared to those with primary LNM. This difference needs to be considered during patient counselling and for stratification purposes in clinical trials. The assumption of an immune privilege of patients with MUP which is responsible for rejection of the primary melanoma, and results in a favourable prognosis is not supported by our data.

We followed up the 1996 baseline parasitological and entomological studies on onchocerciasis transmission in eleven health districts in West Region, Cameroon. Annual mass ivermectin treatment had been provided for 15 years. Follow-up assessments which took place in 2005, 2006, and 2011 consisted of skin snips for microfilariae (mf) and palpation examinations for nodules. Follow-up Simulium vector dissections for larval infection rates were done from 2011 to 2012. mf prevalence in adults dropped from 68.7% to 11.4%, and nodule prevalence dropped from 65.9% to 12.1%. The decrease of mf prevalence in children from 29.2% to 8.9% was evidence that transmission was still continuing. mf rates in the follow-up assessments among adults and in children levelled out after a sharp reduction from baseline levels. Only three health districts out of 11 were close to interruption of transmission. Evidence of continuing transmission was also observed in two out of three fly collection sites that had infective rates of 0.19% and 0.18% and ATP of 70 (Foumbot) and 300 (Massangam), respectively. Therefore, halting of annual mass treatment with ivermectin cannot be done after 15 years as it might escalate the risk of transmission recrudescence.

Vegetable oils and fats may be used as cheap substitutes for milk fat to manufacture imitation cheese or imitation ice cream. In this study, 400 MHz nuclear magnetic resonance (NMR) spectroscopy of the fat fraction of the products was used in the context of food surveillance to validate the labeling of milk-based products. For sample preparation, the fat was extracted using an automated Weibull-Stoldt methodology. Using principal component analysis (PCA), imitation products can be easily detected. In both cheese and ice cream, a differentiation according to the type of raw material (milk fat and vegetable fat) was possible. The loadings plot shows that imitation products were distinguishable by differences in their fatty acid ratios. Furthermore, a differentiation of several types of cheese (Edamer, Gouda, Emmentaler, and Feta) was possible. Quantitative data regarding the composition of the investigated products can also be predicted from the same spectra using partial least squares (PLS) regression. The models obtained for 13 compounds in cheese (R2 0.75–0.95) and 17 compounds in ice cream (R2 0.83–0.99) (e.g., fatty acids and esters) were suitable for a screening analysis. NMR spectroscopy was judged as suitable for the routine analysis of dairy products based on milk or on vegetable fat substitutes.

Entry into mitosis is triggered by cyclinB/Cdk1, whose activity is abruptly raised by a positive feedback loop. The Greatwall kinase phosphorylates proteins of the endosulfine family and allows them to bind and inhibit the main Cdk1-counteracting PP2A-B55 phosphatase, thereby promoting mitotic entry. In contrast to most eukaryotic systems, Cdc14 is the main Cdk1-antagonizing phosphatase in budding yeast, while the PP2A(Cdc55) phosphatase promotes, instead of preventing, mitotic entry by participating to the positive feedback loop of Cdk1 activation. Here we show that budding yeast endosulfines (Igo1 and Igo2) bind to PP2A(Cdc55) in a cell cycle-regulated manner upon Greatwall (Rim15)-dependent phosphorylation. Phosphorylated Igo1 inhibits PP2A(Cdc55) activity in vitro and induces mitotic entry in Xenopus egg extracts, indicating that it bears a conserved PP2A-binding and -inhibitory activity. Surprisingly, deletion of IGO1 and IGO2 in yeast cells leads to a decrease in PP2A phosphatase activity, suggesting that endosulfines act also as positive regulators of PP2A in yeast. Consistently, RIM15 and IGO1/2 promote, like PP2A(Cdc55), timely entry into mitosis under temperature-stress, owing to the accumulation of Tyr-phosphorylated Cdk1. In addition, they contribute to the nuclear export of PP2A(Cdc55), which has recently been proposed to promote mitotic entry. Altogether, our data indicate that Igo proteins participate in the positive feedback loop for Cdk1 activation. We conclude that Greatwall, endosulfines, and PP2A are part of a regulatory module that has been conserved during evolution irrespective of PP2A function in the control of mitosis. However, this conserved module is adapted to account for differences in the regulation of mitotic entry in different organisms.

BACKGROUND: Prognosis of patients with loco-regional skin metastases has not been analyzed in detail and the presence or absence of concurrent lymph node metastasis represents the only established prognostic factor thus far. Most studies were limited to patients already presenting with skin lesions at the time of initial diagnosis. We aimed to analyze the impact of a broad penal of prognostic factors in patients with skin metastases at the time of first metastatic spread, including patients with synchronous lesions already present at the time of initial diagnosis, stage I/II patients with loco-regional recurrence and patients initially presenting with skin metastasis but unknown primary melanoma. PATIENTS AND METHODS: We investigated disease-specific survival of 380 patients treated at our department between 1996 and 2010 using Kaplan Meier survival probabilities and Cox-proportional hazard analysis. RESULTS: Five-year survival probability was 60.1% for patients with skin metastases only and 36.3% for those with synchronous nodal metastases. The number of involved nodes and a tumor thickness of at least 3 mm had independent negative impact on prognosis. A strong relationship was identified between the risk of death and the number of involved nodes. Neither ulceration nor the timing of the first occurrence of metastases as either in stage I/II patients, at the time of excision of the primary melanoma or initially in patients with unknown primary tumor, had additional effects on survival. CONCLUSION: Lymph node involvement was confirmed as the most important prognostic factor for melanoma patients with loco-regional skin metastasis including those with unknown primary tumor and stage I/II patients with skin recurrence. Consideration of the tumor thickness and of the number of involved lymph nodes instead of the exclusive differentiation into presence vs. absence of nodal disease may allow a more accurate prediction of prognosis for patients with satellite or in-transit metastases.

Synaptic long-term potentiation (LTP) at spinal neurons directly communicating pain-specific inputs from the periphery to the brain has been proposed to serve as a trigger for pain hypersensitivity in pathological states. Previous studies have functionally implicated the NMDA receptor-NO pathway and the downstream second messenger, cGMP, in these processes. Because cGMP can broadly influence diverse ion-channels, kinases, and phosphodiesterases, pre- as well as post-synaptically, the precise identity of cGMP targets mediating spinal LTP, their mechanisms of action, and their locus in the spinal circuitry are still unclear. Here, we found that Protein Kinase G1 (PKG-I) localized presynaptically in nociceptor terminals plays an essential role in the expression of spinal LTP. Using the Cre-lox P system, we generated nociceptor-specific knockout mice lacking PKG-I specifically in presynaptic terminals of nociceptors in the spinal cord, but not in post-synaptic neurons or elsewhere (SNS-PKG-I(-/-) mice). Patch clamp recordings showed that activity-induced LTP at identified synapses between nociceptors and spinal neurons projecting to the periaqueductal grey (PAG) was completely abolished in SNS-PKG-I(-/-) mice, although basal synaptic transmission was not affected. Analyses of synaptic failure rates and paired-pulse ratios indicated a role for presynaptic PKG-I in regulating the probability of neurotransmitter release. Inositol 1,4,5-triphosphate receptor 1 and myosin light chain kinase were recruited as key phosphorylation targets of presynaptic PKG-I in nociceptive neurons. Finally, behavioural analyses in vivo showed marked defects in SNS-PKG-I(-/-) mice in several models of activity-induced nociceptive hypersensitivity, and pharmacological studies identified a clear contribution of PKG-I expressed in spinal terminals of nociceptors. Our results thus indicate that presynaptic mechanisms involving an increase in release probability from nociceptors are operational in the expression of synaptic LTP on spinal-PAG projection neurons and that PKG-I localized in presynaptic nociceptor terminals plays an essential role in this process to regulate pain sensitivity.

Severe acute respiratory syndrome (SARS) is a zoonotic disease caused by SARS-related coronavirus (SARS-CoV) that emerged in 2002 to become a global health concern. Although the original outbreak was controlled by classical public health measures, there is a real risk that another SARS-CoV could re-emerge from its natural reservoir, either in its original form or as a more virulent or pathogenic strain; in which case, the virus would be difficult to control in the absence of any effective antiviral drugs or vaccines. Using the well-studied SARS-CoV isolate HKU-39849, we developed a vaccinia virus-based SARS-CoV reverse genetic system that is both robust and biosafe. The SARS-CoV genome was cloned in separate vaccinia virus vectors, (vSARS-CoV-5prime and vSARS-CoV-3prime) as two cDNAs that were subsequently ligated to create a genome-length SARS-CoV cDNA template for in vitro transcription of SARS-CoV infectious RNA transcripts. Transfection of the RNA transcripts into permissive cells led to the recovery of infectious virus (recSARS-CoV). Characterization of the plaques produced by recSARS-CoV showed that they were similar in size to the parental SARS-CoV isolate HKU-39849 but smaller than the SARS-CoV isolate Frankfurt-1. Comparative analysis of replication kinetics showed that the kinetics of recSARS-CoV replication are similar to those of SARS-CoV Frankfurt-1, although the titers of virus released into the culture supernatant are approximately 10-fold less. The reverse genetic system was finally used to generate a recSARS-CoV reporter virus expressing Renilla luciferase in order to facilitate the analysis of SARS-CoV gene expression in human dendritic cells (hDCs). In parallel, a Renilla luciferase gene was also inserted into the genome of human coronavirus 229E (HCoV-229E). Using this approach, we demonstrate that, in contrast to HCoV-229E, SARS-CoV is not able to mediate efficient heterologous gene expression in hDCs.

Whether the dilatation of proximal aorta in patients with bicuspid aortic valve is secondary to hemodynamic effects related to the abnormal aortic valve or a primary manifestation of the genetic disorder remains controversial. We discuss in this paper the recent data on the BAV function and transvalvular flow patterns in relation with the dilatation type of the proximal aorta. Different morphological forms of bicuspid aortic valve in relation with the specific transvalvular blood flow patterns are focus of the first paragraph of this paper. In the second part of this paper we present the pathogenetic insight into the different clinically observed phenotypes of bicuspid aortic valve disease (i.e., association of proximal aortic shapes with the specific cusp fusion patterns), based on the data from recent rheological studies.

Transplantation of photoreceptor precursor cells (PPCs) into the retina represents a promising treatment for cell replacement in blinding diseases characterized by photoreceptor loss. In preclinical studies, we and others demonstrated that grafted PPCs integrate into the host outer nuclear layer (ONL) and develop into mature photoreceptors. However, a key feature of light detecting photoreceptors, the outer segment (OS) with natively aligned disc membrane staples, has not been studied in detail following transplantation. Therefore, we used as donor cells PPCs isolated from neonatal double transgenic reporter mice in which OSs are selectively labeled by green fluorescent protein while cell bodies are highlighted by red fluorescent protein. PPCs were enriched using CD73-based magnetic associated cell sorting and subsequently transplanted into either adult wild-type or a model of autosomal-dominant retinal degeneration mice. Three weeks post-transplantation, donor photoreceptors were identified based on fluorescent-reporter expression and OS formation was monitored at light and electron microscopy levels. Donor cells that properly integrated into the host wild-type retina developed OSs with the formation of a connecting cilium and well-aligned disc membrane staples similar to the surrounding native cells of the host. Surprisingly, the majority of not-integrated PPCs that remained in the sub-retinal space also generated native-like OSs in wild-type mice and those affected by retinal degeneration. Moreover, they showed an improved photoreceptor maturation and OS formation by comparison to donor cells located on the vitreous side suggesting that environmental cues influence the PPC differentiation and maturation. We conclude that transplanted PPCs, whether integrated or not into the host ONL, are able to generate the cellular structure for effective light detection, a phenomenon observed in wild-type as well as in degenerated retinas. Given that patients suffering from retinitis pigmentosa lose almost all photoreceptors, our findings are of utmost importance for the development of cell-based therapies.

Three-dimensional fluorescence imaging of thick tissue samples with near-molecular resolution remains a fundamental challenge in the life sciences. To tackle this, we developed tomoSTORM, an approach combining single-molecule localization-based super-resolution microscopy with array tomography of structurally intact brain tissue. Consecutive sections organized in a ribbon were serially imaged with a lateral resolution of 28 nm and an axial resolution of 40 nm in tissue volumes of up to 50 µm×50 µm×2.5 µm. Using targeted expression of membrane bound (m)GFP and immunohistochemistry at the calyx of Held, a model synapse for central glutamatergic neurotransmission, we delineated the course of the membrane and fine-structure of mitochondria. This method allows multiplexed super-resolution imaging in large tissue volumes with a resolution three orders of magnitude better than confocal microscopy.

High resolution, three-dimensional (3D) representations of cellular ultrastructure are essential for structure function studies in all areas of cell biology. While limited subcellular volumes have been routinely examined using serial section transmission electron microscopy (ssTEM), complete ultrastructural reconstructions of large volumes, entire cells or even tissue are difficult to achieve using ssTEM. Here, we introduce a novel approach combining serial sectioning of tissue with scanning electron microscopy (SEM) using a conductive silicon wafer as a support. Ribbons containing hundreds of 35 nm thick sections can be generated and imaged on the wafer at a lateral pixel resolution of 3.7 nm by recording the backscattered electrons with the in-lens detector of the SEM. The resulting electron micrographs are qualitatively comparable to those obtained by conventional TEM. S(3)EM images of the same region of interest in consecutive sections can be used for 3D reconstructions of large structures. We demonstrate the potential of this approach by reconstructing a 31.7 µm(3) volume of a calyx of Held presynaptic terminal. The approach introduced here, Serial Section SEM (S(3)EM), for the first time provides the possibility to obtain 3D ultrastructure of large volumes with high resolution and to selectively and repetitively home in on structures of interest. S(3)EM accelerates process duration, is amenable to full automation and can be implemented with standard instrumentation.

Background Primary immune thrombocytopenia is a bleeding diathesis with an unknown etiology in predisposed individuals with immune disturbances. Although it is claimed that children and adults differ in clinical and laboratory aspects, few data exist to corroborate this observation. Our objective was to assess comparative data from children and adults with newly diagnosed immune thrombocytopenia.Design and Methods Clinical and laboratory data of 1,784 children and 340 adults were extracted from the Pediatric and Adult Registry on Chronic Immune Thrombocytopenia. The registry represents a prospective cohort of children and adults with newly diagnosed immune thrombocytopenia. Participating investigators registered their patients immediately after the diagnosis using a web based data transfer. Children aged under 16 years were compared with adults aged 16 years and over with descriptive statistical analyses.Results The presenting mean platelet count of children and adults was 18.1 and 25.4×109/L. Signs of bleeding were reported in 24% of children and in 23% of adults, and intracranial hemorrhage in 10 of 1,784 children and in 6 of 340 adults. Co-morbidity was observed in 3.9% of children and in 30% of adults. Bone marrow aspiration and laboratory tests (antinuclear antibodies, human immunodeficiency and hepatitis C virus) were performed more frequently in adults. Children and adults were followed with a ‘watch and wait’ strategy in 20% and in 29%, respectively. Immunoglobulins were used more frequently in children and corticosteroids in adults.Conclusions Comparative data of children and adults with newly diagnosed immune thrombocytopenia revealed similarities in presenting platelet counts and in bleeding, whereas differences occurred in co-morbidity, diagnostic procedures and therapy.

Coronaviruses (CoVs) are important human and animal pathogens that induce fatal respiratory, gastrointestinal and neurological disease. The outbreak of the severe acute respiratory syndrome (SARS) in 2002/2003 has demonstrated human vulnerability to (Coronavirus) CoV epidemics. Neither vaccines nor therapeutics are available against human and animal CoVs. Knowledge of host cell proteins that take part in pivotal virus-host interactions could define broad-spectrum antiviral targets. In this study, we used a systems biology approach employing a genome-wide yeast-two hybrid interaction screen to identify immunopilins (PPIA, PPIB, PPIH, PPIG, FKBP1A, FKBP1B) as interaction partners of the CoV non-structural protein 1 (Nsp1). These molecules modulate the Calcineurin/NFAT pathway that plays an important role in immune cell activation. Overexpression of NSP1 and infection with live SARS-CoV strongly increased signalling through the Calcineurin/NFAT pathway and enhanced the induction of interleukin 2, compatible with late-stage immunopathogenicity and long-term cytokine dysregulation as observed in severe SARS cases. Conversely, inhibition of cyclophilins by cyclosporine A (CspA) blocked the replication of CoVs of all genera, including SARS-CoV, human CoV-229E and -NL-63, feline CoV, as well as avian infectious bronchitis virus. Non-immunosuppressive derivatives of CspA might serve as broad-range CoV inhibitors applicable against emerging CoVs as well as ubiquitous pathogens of humans and livestock.

In mammals, myocardial cell death due to infarction results in scar formation and little regenerative response. In contrast, zebrafish have a high capacity to regenerate the heart after surgical resection of myocardial tissue. However, whether zebrafish can also regenerate lesions caused by cell death has not been tested. Here, we present a simple method for induction of necrotic lesions in the adult zebrafish heart based on cryoinjury. Despite widespread tissue death and loss of cardiomyocytes caused by these lesions, zebrafish display a robust regenerative response, which results in substantial clearing of the necrotic tissue and little scar formation. The cellular mechanisms underlying regeneration appear to be similar to those activated in response to ventricular resection. In particular, the epicardium activates a developmental gene program, proliferates and covers the lesion. Concomitantly, mature uninjured cardiomyocytes become proliferative and invade the lesion. Our injury model will be a useful tool to study the molecular mechanisms of natural heart regeneration in response to necrotic cell death.

Der Wintergerstenanbau wird in Deutschland durch eine Reihe insekten- und pilzübertragbarer Viren bedroht. Die Situation kann sich zukünftig durch ein weiteres bodenbürtiges Virus verschärfen, das aus Gerstenpflanzen, die gegenüber den Erregern der Gelbmosaikvirose resistent waren, isoliert und auf diese rückübertragen werden konnte. In symptomtragenden Pflanzen waren stäbchenförmige Viruspartikeln mit ca. 180 und 300 nm Länge und einem Durchmesser von ca. 20 nm vorhanden, die sich mit goldmarkierten Furovirus-spezifischen Anti­körpern dekorieren ließen. Die serologischen Tests und vorläufigen molekularen Analysen weisen auf eine enge Verwandtschaft des neuen Gerstenvirus mit Isolaten des Soil-borne cereal mosaic virus bzw. Soil-borne wheat mosaic virus hin, die aber bisher in Deutschland nicht an Wintergerste sondern nur in Roggen, Triticale und Winter­weizen gefunden wurden. Weitere Untersuchungen müssen zeigen, welche geographische Verbreitung und wirtschaftliche Bedeutung dieses neue Virus für den Getreideanbau in Deutschland besitzt. Außerdem muss seine genaue Identität zu ähnlichen Furoviren, die in Frankreich bzw. Japan aus Gerste isoliert wurden, aufgeklärt werden.

We introduce a new method for mesoscopic modeling of protein diffusion in an entire cell. This method is based on the construction of a three-dimensional digital model cell from confocal microscopy data. The model cell is segmented into the cytoplasm, nucleus, plasma membrane, and nuclear envelope, in which environment protein motion is modeled by fully numerical mesoscopic methods. Finer cellular structures that cannot be resolved with the imaging technique, which significantly affect protein motion, are accounted for in this method by assigning an effective, position-dependent porosity to the cell. This porosity can also be determined by confocal microscopy using the equilibrium distribution of a non-binding fluorescent protein. Distinction can now be made within this method between diffusion in the liquid phase of the cell (cytosol/nucleosol) and the cytoplasm/nucleoplasm. Here we applied the method to analyze fluorescence recovery after photobleach (FRAP) experiments in which the diffusion coefficient of a freely-diffusing model protein was determined for two different cell lines, and to explain the clear difference typically observed between conventional FRAP results and those of fluorescence correlation spectroscopy (FCS). A large difference was found in the FRAP experiments between diffusion in the cytoplasm/nucleoplasm and in the cytosol/nucleosol, for all of which the diffusion coefficients were determined. The cytosol results were found to be in very good agreement with those by FCS.

Analytical methods based on spectroscopic techniques were developed and validated for the determination of diethyl phthalate (DEP) and polyhexamethylene guanidine (PHMG), which may occur in unrecorded alcohol. Analysis for PHMG was based on UV-VIS spectrophotometry after derivatization with Eosin Y and 1H NMR spectroscopy of the DMSO extract. Analysis of DEP was performed with direct UV-VIS and 1H NMR methods. Multivariate curve resolution and spectra computation methods were used to confirm the presence of PHMG and DEP in the investigated beverages. Of 22 analysed alcohol samples, two contained DEP or PHMG. 1H NMR analysis also revealed the presence of signals of hawthorn extract in three medicinal alcohols used as surrogate alcohol. The simple and cheap UV-VIS methods can be used for rapid screening of surrogate alcohol samples for impurities, while 1H NMR is recommended for specific confirmatory analysis if required.

Parasites of the nematode genus Anisakis are associated with aquatic organisms. They can be found in a variety of marine hosts including whales, crustaceans, fish and cephalopods and are known to be the cause of the zoonotic disease anisakiasis, a painful inflammation of the gastro-intestinal tract caused by the accidental consumptions of infectious larvae raw or semi-raw fishery products. Since the demand on fish as dietary protein source and the export rates of seafood products in general is rapidly increasing worldwide, the knowledge about the distribution of potential foodborne human pathogens in seafood is of major significance for human health. Studies have provided evidence that a few Anisakis species can cause clinical symptoms in humans. The aim of our study was to interpolate the species range for every described Anisakis species on the basis of the existing occurrence data. We used sequence data of 373 Anisakis larvae from 30 different hosts worldwide and previously published molecular data (n = 584) from 53 field-specific publications to model the species range of Anisakis spp., using a interpolation method that combines aspects of the alpha hull interpolation algorithm as well as the conditional interpolation approach. The results of our approach strongly indicate the existence of species-specific distribution patterns of Anisakis spp. within different climate zones and oceans that are in principle congruent with those of their respective final hosts. Our results support preceding studies that propose anisakid nematodes as useful biological indicators for their final host distribution and abundance as they closely follow the trophic relationships among their successive hosts. The modeling might although be helpful for predicting the likelihood of infection in order to reduce the risk of anisakiasis cases in a given area.

Innerhalb der medienanalytischen Beschäftigung mit dem Thema Migration bzw. Migrationshintergrund findet das Medium Jugendzeitschrift bislang nicht die Aufmerksamkeit, die ihm - angesichts seiner Verbreitung und Bedeutung für Jugendliche - gebührt. Insofern wird das Medium selbst als auch wie und in welcher Weise besagte Thematik dort verhandelt wird, bislang unterschätzt bzw. unterbewertet. Ist es doch in mehrfacher Weise bedeutsam: a) als Medium in welchem (genderspezifische) Jugendbilder und Rollenangebote unterbreitet und vermittelt werden, b) zudem als originär an Jugendliche adressiertes Medium, welches folglich - so die Annahme - ein besonderes Potenzial an Sozialisationswirkung auf Jugendliche mit und ohne Migrationshintergrund entfaltet sowie c) als ein Medienformat, welches als jugendtypisch gilt und dem folglich nicht selten in medienpädagogischen Settings (wie beispielsweise im Schulunterricht) besondere innovative Vermittlungspotenziale beigemessen werden. Vor dem Hintergrund dieser Annahmen wird am Beispiel einer sog. Foto-Love-Story - einem in Jugendzeitschriften beliebten und exponiert eingesetztem Format - exemplarisch verdeutlicht, auf welch unkritisch-drastische, teils dramatische Weise und mit höchst stereotypisierenden Bildern das Thema Migration auch in diesem Medium behandelt wird.. Stichworte an dieser Stelle sind: Verweise auf die besondere Religiosität von Jugendlichen mit Migrationshintergrund (hier: Islam), negativ konnotierte Geschlechterrollenklischees, Zuschreibungen vormoderner/traditionsfixierter Haltungen, Identitätskonflikte, Zwangsheirat, Gewalttätigkeit etc. Die genannten negativen Zuschreibungen gelten für die Jugendlichen mit Migrationshintergrund. Es lassen sich indes auch stereotypisierende Bilder mit Blick auf sog. Jugendliche ohne Migrationshintergrund identifizieren - jedoch mit positiver Konnotierung. Jene pauschalen Entgegensetzungen entlang der Achse mit/ohne Migrationshintergrund sind sehr markant und entsprechend zu kritisieren. Es überrascht zunächst, dass die vorfindbaren Klischees sich in solch einer überaffirmativen und ungebrochenen Weise zu den auch aus anderen Medien hinlänglich bekannten rassistischen Stereotypen und Applikationsvorgaben verhalten. Über die Vorstellung, Analyse und Kritik jener Konstruktionen hinaus ist aber auch zu fragen, wie solche medialen Vermittlungen sich zu Ansprüchen verhalten, im Mediendiskurs differenzierte und nicht-rassistische Darstellungen zu verankern.

Open source software and hardware has become an accepted way of developing new and interesting applications in many information and communication technology domains: operating systems, databases, Web infrastructure, and applications. It's not surprising that with the increasing popularity of mobile handheld devices, users and researchers have explored the power of open approaches to providing innovative new applications and services in this domain. However, unlike personal computers and the Internet, mobile handsets were tightly controlled by mobile network operators (MNOs) who developed a vertical ecosystem by integrating the communication infrastructure, the handheld device hardware, and often the applications installed on those devices. The software and protocols running the mobile communications infrastructure and devices are often standardized by membership-only bodies, where large MNOs and manufacturers have a predominant influence. These players invest significant financial resources into shaping the industry along their vision to gain a competitive advantage. A current example is the ongoing battle about the dominant radio access technology for 4G cellular systems: LTE vs. Wimax. These trends have changed recently. Companies such as Google, Nokia, or Openmoko and Industry Alliances such as the Open Handset Alliance are providing the core building blocks, both in hardware as well as software, of increasingly open mobile devices. This issue of the OSBR reviews the relevant trends in the open mobile platform space from a number of perspectives. As the articles in these issue show, there is a lot of exciting ongoing work that brings the power of open source development to the mobile space. This trend is not just confined to the mobile devices as there are also efforts in the development of open mobile infrastructure elements and whole systems.

BackgroundPhthalates are synthetic compounds with a widespread field of applications. For example, they are used as plasticizers in PVC plastics and food packaging, or are added to personal care products. Diethyl phthalate (DEP) may be used to denature alcohol, e.g., for cosmetic purposes. Public health concerns of phthalates include carcinogenic, teratogenic, hepatotoxic and endocrine effects. The aim of this study was to develop and validate a method for determining phthalates in alcohol samples and to provide a risk assessment for consumers of such products.Methodology/principal findingsA liquid-liquid extraction procedure was optimized by varying the following parameters: type of extraction solvent (cyclohexane, n-hexane, 1,1,2-trichlorotrifluoroethane), the ratio extraction solvent/sample volume (1 ratio 1 to 50 ratio 1) and the number of extraction repetitions (1-10). The best extraction yield (99.9%) was achieved with the solvent 1,1,2-trichlorotrifluoroethane, an extraction solvent volume/sample volume ratio of 10 ratio 1 and a double extraction. For quantification, gas chromatography/mass spectrometry with deuterated internal standards was used. The investigated samples were alcoholic beverages and unrecorded alcohol products from different countries (n = 257). Two unrecorded alcohol samples from Lithuania contained diethyl phthalate in concentrations of 608 mg/L and 210 mg/L.Conclusions/significanceThe consumption of the phthalate-positive unrecorded alcohols would exceed tolerable daily intakes as derived from animal experiments. Both positive samples were labelled as cosmetic alcohol, but had clearly been offered for human consumption. DEP seems to be unsuitable as a denaturing agent as it has no effect on the organoleptic properties of ethanol. In light of our results that DEP might be consumed by humans in unrecorded alcohols, the prohibition of its use as a denaturing agent should be considered.
