Summary: Background: While a low CD4/CD8 ratio during HIV treatment correlates with immunosenescence, its value in identifying patients at an increased risk for clinical events remains unclear. Methods: We analyzed data from the CoRIS cohort to determine whether CD4 count, CD8 count, and CD4/CD8 ratio at year two of antiretroviral therapy (ART) could predict the risk of serious non-AIDS events (SNAEs) during the next five years. These included major adverse cardiovascular events, non-AIDS-defining malignancies, and non-accidental deaths. We used pooled logistic regression with inverse probability weighting to estimate the survival curves and cumulative risk of clinical events. Abstract Background Fragmentation and deforestation are one of the greatest threats to forests, and these processes are of even more concern in the tropics, where the seasonal dry forest is possibly one of the most threatened ecosystems with the least remaining surface area. Methods The deforestation and fragmentation patterns that had occurred in Ecuadorian seasonal dry forests between 1990 and 2018 were verified, while geographic information systems and land cover shapes provided by the Ecuadorian Ministry of the Environment were employed to classify and evaluate three types of seasonal dry forests: deciduous, semi-deciduous, and transition. The study area was tessellated into 10 km2 hexagons, in which six fragmentation parameters were measured: number of patches, mean patch size, median patch size, total edge, edge density and reticular fragmentation index (RFI). The RFI was also measured both outside and inside protected natural areas (unprotected, national protected areas and protected forest). Moreover, the areas with the best and worst conservation status, connectivity and risk of disappearance values were identified by means of a Getis-Ord Gi* statistical analysis. Results The deforestation of seasonal dry forests affected 27.04% of the original surface area still remaining in 1990, with an annual deforestation rate of − 1.12% between 1990 and 2018. The RFI has increased by 11.61% as a result of the fact that small fragments of forest have tended to disappear, while the large fragments have been fragmented into smaller ones. The semi-deciduous forest had the highest levels of fragmentation in 2018. The three categories of protection had significantly different levels of fragmentation, with lower RFI values in national protected areas and greater values in protected forests. Conclusions The seasonal dry forest is fragmenting, deforesting and disappearing in some areas. An increased protection and conservation of the Ecuadorian seasonal dry forest is, therefore, necessary owing to the fact that not all protection measures have been effective.

PurposeTo analyze the impact of SARS-COV-2-specific memory B cells (MBC) on the immune response after two doses of mRNA-based Comirnaty COVID-19 vaccine in seronegative health care workers. This study is seeking a rationale for boosting vaccines.MethodsLongitudinal study including 31 seronegative health care workers with undetectable specific MBCs (IgG−MBC− group), 24 seronegative with detectable specific MBCs (IgG−MBC+ group), and 24 seropositive with detectable specific MBCs (IgG+MBC+ group). Labour market areas (LMAs) are a type of functional areas that provides the appropriate framework for spatially-explicit analyses of the labour market to guide policymaking in this field. This resource presents a data base with the most recent definition of LMAs for Spain using a state-of-the-art methodology. All defined LMAs are highly self-contained in terms of travel-to-work flows and exceed a minimum size compatible with the usual requirements in terms of confidentiality and representativeness, allowing the recoding of microdata geographic referencing and therefore a much greater granularity compared to what is usual for this type of data. Aim of the study: Traditional vineyards have, in the last few decades, been transformed into trellis systems, but little research has been carried out into the consequences as regards biodiversity. We compared the abundance and species richness of reptiles in conventional-traditional vineyards and trellis vineyards. Area of study: The study was conducted in a wine appellation area of origin denominated as Montilla-Moriles, Southern Spain. Material and methods: Reptile’s species richness and abundance were estimated by walking transects in 24 different vineyards (12 trellis and 12 traditional vineyards) in four consecutive years. Main results: The results showed an extremely low abundance in both management systems, since no reptiles were recorded in 43.1% of the transects. However, there was a greater abundance and diversity of reptiles in the traditional vineyards than in the trellis vineyards, with 7 vs. 3 species being found in traditional and trellis vineyards, respectively. Research highlights: The lack of refuge in trellis vineyards owing to the vertical growth of plants, whose branches grow higher from the ground, is probably the main cause of the lower abundance and species richness found in trellis systems, since both types of vineyard had bare ground owing to ploughing and the application of herbicides. Since the transformation of traditional vineyards into those with trellis systems is often subsidized, this modernization should be accompanied by certain agri-environmental measures (e.g., cover crops, artificial refuges or natural hedges) in order to compensate for the associated negative effects.

Few data are available about the immune response to mRNA SARS-CoV-2 vaccines in patients with breast cancer receiving cyclin-dependent kinase 4/6 inhibitors (CDK4/6i). We conducted a prospective, single-center study of patients with breast cancer treated with CDK4/6i who received mRNA-1273 vaccination, as well as a comparative group of healthcare workers. The primary endpoint was to compare the rate and magnitude of humoral and T-cell response after full vaccination. A better neutralizing antibody and anti-S IgG level was observed after vaccination in the subgroup of women receiving CDK4/6i, but a trend toward a reduced CD4 and CD8 T-cell response in the CDK4/6i group was not statistically significant. There were no differences in the rate of COVID-19 after vaccination (19% vs. 12%), but breakthrough infections were observed in those with lower levels of anti-S IgG and neutralizing antibodies after the first dose. A lower rate of CD4 T-cell response was also found in those individuals with breakthrough infections, although a non-significant and similar level of CD8 T-cell response was also observed, regardless of breakthrough infections. The rate of adverse events was higher in patients treated with CDK4/6i, without serious adverse events. In conclusion, there was a robust humoral response, but a blunted T-cell response to mRNA vaccine in women receiving CDK4/6i, suggesting a reduced trend of the adaptative immune response.

Objective: T-cell responses against SARS-CoV-2 are observed in unexposed individuals, attributed to previous common human coronavirus (HCoV) infections. We evaluated the evolution of this T-cell cross-reactive response and the specific memory B-cells (MBCs) after the SARS-CoV-2 mRNA-based vaccination and its impact on incident SARS-CoV-2 infections. Methods: This was a longitudinal study of 149 healthcare workers (HCWs) that included 85 unexposed individuals that were subdivided according to previous T-cell cross-reactivity, who were compared to 64 convalescent HCWs. Changes in specific T-cell response and memory B-cell (MBC) levels were compared at baseline and after two doses of the SARS-CoV-2 mRNA-based vaccine. Results: A cross-reactive T-cell response was found in 59% of unexposed individuals before vaccination. Antibodies against HKU1 positively correlated with OC43 and 229E antibodies. Spike-specific MBCs was scarce in unexposed HCWs regardless of the presence of baseline T-cell cross-reactivity. After vaccination, 92% and 96% of unexposed HCWs with cross-reactive T-cells had CD4+ and CD8+ T-cell responses to the spike protein, respectively. Similar results to that were found in convalescents (83% and 92%, respectively). Contrarily, higher than that which was observed in unexposed individuals without T-cell cross-reactivity showed lower CD4+ and CD8+ T-cell responses (73% in both cases, p = 0.03). Nevertheless, previous cross-reactive T-cell response was not associated with higher levels of MBCs after vaccination in unexposed HCWs. During a follow-up of 434 days (IQR, 339–495) after vaccination, 49 HCWs (33%) became infected, with a significant positive correlation between spike-specific MBC levels and isotypes IgG+ and IgA+ after vaccination and a longer time to get infected. Interestingly, T-cell cross-reactivity did not reduce the time to vaccine breakthrough infections. Conclusion: While pre-existing T-cell cross-reactivity enhances the T-cell response after vaccination, it does not increase SARS-CoV-2-specific MBC levels in the absence of previous infection. Overall, the level of specific MBCs determines the time to breakthrough infections, regardless of the presence of T-cell cross-reactivity.

SARS-CoV-2 spike mRNA vaccines have shown remarkable clinical efficacy in the general population, although the nature of T-cell priming is not fully understood. We performed longitudinal spike-, membrane-, and nucleocapsid-specific T-cell analysis in individuals with past infection and infection-naïve individuals with cross-reactivity. We found an additional enhancement of T-cell response to the structural membrane (M) and nucleocapsid (N) SARS-CoV-2 proteins after mRNA vaccine in these individuals. Thus, despite the spike-specific response, we found that the first dose of the vaccine boosted a significant CD8 cell response to M and N proteins, whereas no cellular response to those proteins was found in infection-naïve individuals without pre-existing cross-reactivity who were tested for eventual asymptomatic infection. These findings highlight the additional benefit of mRNA vaccines as broad boosters of cellular responses to different viral epitopes in these individuals and suggest extended protection to other viral variants.

Evidências epidemiológicas sugerem que a infecção por SARS-CoV-2 apresenta menor gravidade e melhor desfecho clínico em crianças. Por outro lado, complicações respiratórias agudas e falência de múltiplos órgãos podem ocorrer. A Síndrome Inflamatória Multissistêmica Pediátrica (SIM-P) é entidade que ocorre pós infecção por COVID-19, sendo considerada reação imunológica exacerbada, podendo evoluir com elevação de marcadores inflamatórios, alterações coronarianas e disfunções cardíacas. APRESENTAÇÃO: FMLP, 7 anos, natural de Patos-PB, previamente hígido, sem internamentos prévios e cartão vacinal atualizado. Iniciou febre em 02/07/2021 associada a vômitos, diarreia, artralgia, exantema, dispneia e sonolência. SatO2 87% em ar ambiente, hipotenso, dor abdominal e edema difusos. Transferido para UTI de serviço de referência do Estado de Pernambuco. À admissão: sorologia SARS-CoV-2 IgG reagente (2826), IgM não reagente, RT-PCR para COVID-19 negativo, D-dímero 3110, Ferritina > 2000, PCR 264.2mg/dl, albumina 2.4, Ht 36%, leucopenia com linfopenia e plaquetas 75.000/UL, troponina 143.1pg/ml. USG de abdome: discreta ascite, esplenomegalia homogênea. Iniciados: ATB venoso, droga vasoativa, corticoterapia, imunoglobulina endovenosa e enoxaparina. Solicitadas sorologias para dengue e chikungunya, RT-PCR para COVID-19, Ecocardiograma e hemoculturas normais. Após 24h apresentou melhora clínica importante com queda dos marcadores inflamatórios. DISCUSSÃO: A SIM-P foi inicialmente relatada no Reino Unido, com casos semelhantes em vários países da Europa correlacionados à infecção prévia por COVID-19, apresentando características clínicas e laboratoriais semelhantes à Doença de Kawasaki e Síndrome do Choque Tóxico. Neste paciente, observou-se necessidade de uso de droga vasoativa, porém não necessitou de suporte ventilatório, com boa resposta em 24h após tratamento. Diante da situação endêmica para arboviroses no Brasil não se pôde afastar tal diagnóstico diferencial, sobretudo, Dengue e Chikungunya. CONCLUSÕES: A SIM-P é condição grave, com potencial de morbimortalidade elevado. Há necessidade de os profissionais médicos estarem atentos para essa condição. Não é incomum que pacientes com SIM-P, tal qual o do caso relatado acima, necessitem de corticoterapia, drogas vasoativas, imunoglobulina endovenosa e suporte em unidades de terapia intensiva.

Este trabajo propone una tipología de territorios turísticos con relación a la incidencia de COVID-19, la disponibilidad de infraestructura física para enfrentar la pandemia y la disminución del empleo en el sector turístico en México, a escala municipal, en el período marzo-junio de 2020. Los hallazgos principales de la investigación, revelados mediante coordenadas cartesianas y representados en forma cartográfica, muestran16 combinaciones tipológicas territoriales que comprenden desde las que se asocian con condiciones de mayor incidencia, hasta las que gozan de una situación sanitaria comparativamente mejor.

Background Among HIV-infected individuals, screening for bone disease is encouraged to assess reversible risk factors and plan therapeutic interventions. Objective We assessed the usefulness of Fracture Risk Assessment (FRAX) tool to identify candidates for dual X-ray absorptiometry (DXA) scan, or individuals with bone loss progression. We further explored how secondary causes of osteoporosis are reflected on FRAX. Methods Longitudinal study of 217 consecutive individuals (mean, 45.8 years, 24% females) included after DXA scan. FRAX was calculated without/with femoral neck bone mineral density (BMD), checking the box of “secondary osteoporosis” for all the individuals. Results Low BMD was observed in 133/217 (61%) individuals, of whom 98.5% had not been selected as candidates for DXA by current FRAX thresholds. Specifically, 23% of individuals aged

Illegal wildlife trafficking has negative effects on biodiversity conservation at both global and local scale. Therefore, the establishment of appropriate conservation measures requires local studies that quantify this problem. The objective of this work was to quantify and characterize the species of birds and mammals seized in the period 2016-2017, at the Valle Alto Wildlife Rescue Centre and Wildlife Refuge. The study showed that 212 specimens belonging to 41 different species were confiscated. More birds than mammals were confiscated, and a greater proportion of birds were included in a national and international threat category. A clear preference for primates, parrots and squirrels was found. Furthermore, the presence of species with a distribution range outside the study area revealed the existence of the transportation of species from other parts of the country. Although these data are only a sample of what is actually trafficked in the country, they provide an approach of the type of species that are illegally trafficked in this biodiversity hotspot.

C. canephora (syn. C. robusta) is distinctive due to its rising industrial value and pathogen resistance. Both altitude and post-harvest methods influence coffee cup quality; however, modest information is known about this coffee species. Therefore, the aim of this study was to determine the relationship between four different altitudes and post-harvest processes (dry, honey, and wet) to the improvement of the organoleptic quality of the C. canephora congolensis and conilon drink. For dry processing, congolensis and conilon showed the lowest scores in terms of fragrance/aroma, flavour, aftertaste, salt–acid, bitter–sweet, and body. Above 625 m, coffees from dry, honey, and wet processes increased scores in their sensory attributes, but there was no difference at such high altitudes when comparing post-harvest samples. Dry-processed coffee samples had total scores over 80 points at high altitudes. Conilon was perceived to have the best sensory attributes at high altitudes using honey processing. In general, the wet-processed congolensis and conilon samples had a tastier profile than dry-processed ones.

Despite in vitro activity of interferon-β (IFN-β) against SARS-CoV-2 infection, its clinical efficacy remains controversial. We evaluated the impact of IFN-β treatment in a cohort of 3590 patients hospitalized with COVID-19 during March–April 2020. The primary endpoint was a composed variable of admission to intensive care unit (ICU)/death. Overall, 153 patients (4%) received IFN-β. They were significantly more severely ill, with a worse clinical and analytical situation, explaining a higher ICU admission (30% vs. 17%; p < 0.01), and a shorter time to the composed variable. In a Cox regression analysis, older age, lymphopenia, renal failure, or increased neutrophil-to-lymphocyte ratio were associated with a greater hazard ratio (HR) of admission at ICU/death. Notably, the HR of IFN-β for the outcome variable was no longer significant after adjustment (HR, 1.03; 95% CI, 0.82–1.30), and different sensitivity analysis (early IFN use, ICU admission) showed no changes in the estimates. A propensity score matching analysis showed no association of IFN-β therapy and outcome. In conclusion, in this large cohort of hospitalized COVID-19 patients, IFN-β was used mainly in patients with advanced disease, reflecting an important bias of selection. After adjusting by severity, IFN-β was not associated with a higher rate of ICU admission or mortality.

People living with HIV-1 and HTLV-2 concomitantly show slower CD4+ T cell depletion and AIDS progression, more frequency of the natural control of HIV-1, and lower mortality rates. A similar beneficial effect of this infection has been reported on HCV coinfection reducing transaminases, increasing the spontaneous clearance of HCV infection and delaying the development of hepatic fibrosis. Given the critical role of CD8+ T cells in controlling HIV-1 infection, we analysed the role of CD8+ T cell-mediated cytotoxic activity in coinfected individuals living with HIV-1. One hundred and twenty-eight individuals living with HIV-1 in four groups were studied: two groups with HTLV-2 infection, including individuals with HCV infection (N = 41) and with a sustained virological response (SVR) after HCV treatment (N = 25); and two groups without HTLV-2 infection, including individuals with HCV infection (N = 25) and with a sustained virological response after treatment (N = 37). We found that CD8+ T cell-mediated HIV-1 inhibition in vitro was higher in individuals with HTLV-2. This inhibition activity was associated with a higher frequency of effector memory CD8+ T cells, higher levels of granzyme A and granzyme B cytolytic enzymes, and perforin. Hence, cellular and soluble cytolytic factors may contribute to the lower HIV-1 pre-ART viral load and the HIV-1 proviral load during ART therapy associated with HTLV-2 infection. Herein, we confirmed and expanded previous findings on the role of HTLV-2 in the beneficial effect on the pathogenesis of HIV-1 in coinfected individuals.

There are many local fragmentation metrics, but most can be grouped into four types (composition/area, isolation, edge, and shape), and none of them alone determines the degree of fragmentation of a patch. Here, we grouped together the main fragmentation metrics (area, edge, shape, and isolation) in order to propose a new metric/index, the Patch Fragmentation Index (PFI), with which to determine fragmentation at patch scale. The index was subsequently verified with the Ecuadorian seasonal dry forest by employing geographic information layers and temporal land uses changes in 1990, 2000, 2008, and 2018. The PFI was applied to calculate the fragmentation per patch, spatial and temporal changes of fragmentation based on PFI were assessed, and the spatial patterns (Getis-Ord Gi * analysis) were calculated. The Ecuadorian seasonal dry forest obtained a mean PFI value of 0.88 (median = 0.99) in 2018. This value has increased by 8.6% since 1990, and 3451 patches of forest disappeared between 1990 and 2018. The Getis-Ord Gi * analysis was effective with regard to describing the spatial patterns, and 62% of the patches that were classified as hot patches in 1990 had disappeared by 2018. The PFI has proven to be a useful tool with which to describe fragmentation patterns at patch scale (regardless of its size) and can be extrapolated to other landscapes. The PFI will provide a new vision and can help in the decision-making related to the conservation and management of fragmented ecosystems.

The risk of reinfection could be related to the initial SARS-CoV-2 clinical presentation, but there are no data about the risk change after SARS-CoV-2 vaccination. We evaluated the rate of reinfection in an inception cohort study of 4943 health care workers (HCWs) according to symptoms and serologic results during March–May 2020. Incidence rates (IR) and IR ratios (IRR) before and after SARS-CoV-2 vaccination were determined by adjusting Poisson models. Overall, 1005 HCWs (20.3%) referred COVID-19 suggestive symptoms during the first surge of disease, and 33.5% and 55% presented a positive PCR or serology result, respectively. Meanwhile, 13% of asymptomatic HCWs had specific antibodies. During a follow up of 3422.2 person-years before vaccination, the rate of reinfection among seropositive individuals was 81% lower for those who were symptomatic compared with those who were asymptomatic (IRR of 0.19; 95% CI, 0.05–0.67; p = 0.003). During the 3100 person-years period after vaccination, an overall 74% decrease in the rate of infection was observed (IRR of 0.26; 95% CI, 0.21–0.32; p < 0.001), with a significant 83% and 70% decrease in seropositive and seronegative HCWs, respectively. In conclusion, the risk of SARS-CoV-2 reinfections is closely related to the clinical and serological presentation of COVID-19. COVID-19 vaccination further decreases the risk of reinfection more markedly among seropositive.

In this pilot program of low-dose computed tomography (LDCT) for the screening of lung cancer (LC) in a targeted population of people with HIV (PWH), its prevalence was 3.6%; the number needed to screen in order to detect one case of lung cancer was 28, clearly outweighing the risks associated with lung cancer screening. While data from additional cohorts with longitudinal measurements are needed, PWH are a target population for lung cancer screening with LDCT.

La situación de los agricultores alinea objetivos macro, condiciones y actividades propias en su diario convivir; las variables interactúan en los diferentes escenarios que desarrollan los productores de maíz duro en Manabí-Ecuador, teniendo un efecto en el desarrollo productivo del país. Este trabajo se realizó a partir de la aplicación del modelo multivariante basado en un estudio multipropósito, obteniéndose conglomerados interrelacionados de las variables sociales, económicas, políticas, productivas y ambientales. Los resultados presentan una alta similitud de agrupamiento tanto en las observaciones y entre variables, demostrando el alto impacto que tienen las variables influyentes, especialmente las relacionadas a la producción (época de siembra, superficie de siembra, disponibilidad de agua de riego) y económicas (facturación anual, cantidad de empleados), en tanto la diversidad en las observaciones están afectadas con menores distancias, para las variables relacionadas en la toma de decisiones, sociales, políticas y ambientales generando un impacto positivo en los productores.

La expansión de los supermercados en México, favorecida por la llegada de Wal-Mart en los 1990s y su competencia con cadenas nacionales, ha impulsado su penetración tanto en localidades cada vez más pequeñas como en espacios de reciente urbanización, siguiendo así el proceso de desconcentración de la población; pero sobre todo los supermercados se han expandido al interior de las grandes ciudades. La proliferación de formatos más pequeños dirigidos a segmentos de menores ingresos ha permitido aumentar su cobertura poblacional, aunque siguen mostrando una clara preferencia por espacios de baja marginación y por tanto altos ingresos, así como por áreas densamente pobladas. Aunado a este proceso se ha asistido a una creciente concentración de estos establecimientos en unas pocas grandes empresas, ejerciendo Wal-Mart un claro dominio en México.

Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV-2) infection induces elevated levels of inflammatory cytokines, which are mainly produced by the innate response to the virus. The role of NK cells, which are potent producers of IFN-γ and cytotoxicity, has not been sufficiently studied in the setting of SARS-CoV-2 infection. We confirmed a different distribution of NK cell subsets in hospitalized COVID-19 patients despite their NK cell deficiency. The impairment of this innate defense is mainly focused on the cytotoxic capacity of the CD56dim NK cells. On the one hand, we found an expansion of the CD56dimCD16neg NK subset, lower cytotoxic capacities, and high frequencies of inhibitory 2DL1 and 2DL1/S1 KIR receptors in COVID-19 patients. On the other hand, the depletion of CD56dimCD16dim/bright NK cell subsets, high cytotoxic capacities, and high frequencies of inhibitory 2DL1 KIR receptors were found in COVID-19 patients. In contrast, no differences in the distribution of CD56bright NK cell subsets were found in this study. These alterations in the distribution and phenotype of NK cells might enhance the impairment of this crucial innate line of defense during COVID-19 infection.

The European Union’s (EU) approach to safeguarding seed quality and ensuring agricultural productivity includes a range of regulatory measures related to seed marketing, including the restriction of market access to seed varieties that are properly certified. In order for varieties to be properly certified, they have to be registered in a Plant Variety Catalogue. EU seed laws, and their Member State implementation, have traditionally favoured uniform crop production of commercialised breeds over conservation varieties, which has been a contributing factor to crop genetic erosion. The United States’ (U.S.) approach to seed marketing regulation is often presented as an alternative model to the EU’s approach. In the United States, any seed can be indistinctly marketed if properly labelled. In this regulatory framework, certification and registration of seed is voluntary. In light of the continued issues regarding crop genetic erosion and the recent developments in EU seed law reform, this article examines the key elements of both regimes and considers their different approaches to market access for conservation varieties. The most important differences relate to in truth-in-labelling (U.S.) and ex ante quality control mechanisms (EU). These differences highlight that EU and U.S. seed laws must be placed in their respective broader regulatory context and that their relative comparability hinges on policy aims related to seed quality. This raises the broader question as to what the key policy aims of seed laws are and whether these oft-compared regimes are in fact analogous in terms of goals and structure.

This work assesses damage caused by Coraebus undatus (Fabricius) (Coleoptera: Buprestidae) and Reticulitermes grassei Clément (Blattodea: Rhinotermitidae) in managed cork oak forests from the southern Iberian Peninsula. Lesions were diagnosed and quantified in relation to the following features: height and orientation in the trunk, diameter at breast height, solar exposure, understory presence and orography. The distribution patterns of lesions in the study area across 12 plots and in the trunk of the trees were also analyzed. The study was performed in “Sa de Hornachuelos” Natural Park (Córdoba, Spain) and the research area encompassed 12 environmentally-restored sampling plots. Data were recorded from 2007 to 2014, with yearly sampling from late June to mid-September, corresponding to the cork extraction period, since lesions caused by these insects are well distinguishable promptly after bark removal. The results reveal that C. undatus has low population levels and a non-uniform distribution pattern in the study area as well as in the trunk of the tree. It shows preference for medium height but not by any trunk orientation. R. grassei also shows low infestation levels and a preference to affect the trunk’s base. Comparatively, C. undatus showed higher infestation levels than R. grassei. The location of damage was also different, since galleries made by C. undatus predominated at intermediate trunk heights while those of R. grassei were more frequent at lower heights. Our results further showed a low co-occurrence of both species in the same tree trunks, which could be explained in terms of overlapping in the distribution areas of both species. These results provide background information that will enable natural resources managers to detect changes and trends of these species and inform future management decisions.

Fanconi anemia (FA) patients have an exacerbated risk of head and neck squamous cell carcinoma (HNSCC). Treatment is challenging as FA patients display enhanced toxicity to standard treatments, including radio/chemotherapy. Therefore, better therapies as well as new disease models are urgently needed. We have used CRISPR/Cas9 editing tools in order to interrupt the human FANCA gene by the generation of insertions/deletions (indels) in exon 4 in two cancer cell lines from sporadic HNSCC having no mutation in FA-genes: CAL27 and CAL33 cells. Our approach allowed efficient editing, subsequent purification of single-cell clones, and Sanger sequencing validation at the edited locus. Clones having frameshift indels in homozygosis did not express FANCA protein and were selected for further analysis. When compared with parental CAL27 and CAL33, FANCA-mutant cell clones displayed a FA-phenotype as they (i) are highly sensitive to DNA interstrand crosslink (ICL) agents such as mitomycin C (MMC) or cisplatin, (ii) do not monoubiquitinate FANCD2 upon MMC treatment and therefore (iii) do not form FANCD2 nuclear foci, and (iv) they display increased chromosome fragility and G2 arrest after diepoxybutane (DEB) treatment. These FANCA-mutant clones display similar growth rates as their parental cells. Interestingly, mutant cells acquire phenotypes associated with more aggressive disease, such as increased migration in wound healing assays. Therefore, CAL27 and CAL33 cells with FANCA mutations are phenocopies of FA-HNSCC cells.
