The seasonal timing of recurring biological processes is essential for organisms living in temperate regions. While ample knowledge of these processes exists for terrestrial environments, seasonal timing in the marine environment is relatively understudied. Here, we characterized the annual rhythm of habitat use in six fish species belonging to the Sparidae family, highlighting the main environmental variables that correlate to such rhythms. The study was conducted at a coastal artificial reef through a cabled observatory system, which allowed gathering underwater time-lapse images every 30 minutes consecutively over 3 years. Rhythms of fish counts had a significant annual periodicity in four out of the six studied species. Species-specific temporal patterns were found, demonstrating a clear annual temporal niche partitioning within the studied family. Temperature was the most important environmental variable correlated with fish counts in the proximity of the artificial reef, while daily photoperiod and salinity were not important. In a scenario of human-induced rapid environmental change, tracking phenological shifts may provide key indications about the effects of climate change at both species and ecosystem level. Our study reinforces the efficacy of underwater cabled video-observatories as a reliable tool for long-term monitoring of phenological events.

Functional characterization of two novel endoglucanase genes, Pv-eng -5 and Pv-eng -8, of the root-lesion nematode Pratylenchus vulnus was carried out. In situ-hybridization experiments revealed that Pv-eng -8 transcript was localized in the pharyngeal glands. Silencing of Pv-eng -5 and Pv-eng -8 resulted in a significant reduction of expression level (52% and 67%, respectively). Furthermore, the silencing of Pv-eng -8 determined a reduction (41%) in nematode reproduction, suggesting that treated nematodes are much less able to process food. Surprisingly, no significant difference on reproduction rate was observed with Pv-eng -5 dsRNA nematodes, suggesting a neofunctionalization of Pv-eng -5 despite the high similarity with nematode endoglucanases. Pratylenchus species are poikilothermic organisms showing close relationships with the environmental temperature. The effects of different temperature ranges revealed that the reproductive potential of P. vulnus increased with increasing temperature from 23 °C to 28 °C, but no reproduction was observed at 33 °C. In real time, increasing temperature from 23 °C to 28 °C the heat shock gene Pv-hsp -90 was differentially expressed in adult stages, while the levels of the effector genes Pv-eng -1 and Pv-eng -8 in females showed no significant differences compared to those observed at 23 °C, only in males Pv-eng -8 level decreased (45%). The upregulation of Pv-hsp -90 in both adult stages suggests a protective mechanism in order to cope with unfavorable environmental conditions.

Marine cabled video-observatories allow the non-destructive sampling of species at frequencies and durations that have never been attained before. Nevertheless, the lack of appropriate methods to automatically process video imagery limits this technology for the purposes of ecosystem monitoring. Automation is a prerequisite to deal with the huge quantities of video footage captured by cameras, which can then transform these devices into true autonomous sensors. In this study, we have developed a novel methodology that is based on genetic programming for content-based image analysis. Our aim was to capture the temporal dynamics of fish abundance. We processed more than 20,000 images that were acquired in a challenging real-world coastal scenario at the OBSEA-EMSO testing-site. The images were collected at 30-min. frequency, continuously for two years, over day and night. The highly variable environmental conditions allowed us to test the effectiveness of our approach under changing light radiation, water turbidity, background confusion, and bio-fouling growth on the camera housing. The automated recognition results were highly correlated with the manual counts and they were highly reliable when used to track fish variations at different hourly, daily, and monthly time scales. In addition, our methodology could be easily transferred to other cabled video-observatories.

Benthic community succession patterns at whale falls have been previously established by means of punctual submersible and ROV observations. The contribution of faunal activity rhythms in response to internal tides and photoperiod cues to that community succession dynamism has never been evaluated. Here, we present results from a high-frequency monitoring experiment of an implanted sperm whale carcass in the continental slope (500 m depth) offshore Sagami Bay, Japan. The benthic community succession was monitored at a high frequency in a prolonged fashion (i.e. 2-h intervals for 2.5 months) with a seafloor lander equipped with a time-lapse video camera and an acoustic Doppler profiler to concomitantly study current flow dynamics. We reported here for the first time, to the best of our knowledge, the occurrence of strong 24-h day-night driven behavioral rhythms of the most abundant species (Simenchelys parasitica; Macrocheira kaempferi, and Pterothrissus gissu). Those rhythms were detected in detriment of tidally-controlled ones. Evidence of a diel temporal niche portioning between scavengers and predators avoiding co-occurrence at the carcass, is also provided. The high-frequency photographic and oceanographic data acquisition also helped to precisely discriminate the transition timing between the successional stages previously described for whale falls' attendant communities.

Dark marine habitats are often characterized by a food-limited condition. Peculiar dark habitats include marine caves, characterized by the absence of light and limited water flow, which lead to reduced fluxes of organic matter for cave-dwelling organisms. We investigated whether the most abundant and common cave-dwelling fish Apogon imberbis has the potential to play the role of trophic vector in Mediterranean marine caves. We first analysed stomach contents to check whether repletion changes according to a nycthemeral cycle. We then identified the prey items, to see whether they belong to species associated with cave habitats or not. Finally, we assessed whether A. imberbis moves outside marine caves at night to feed, by collecting visual census data on A. imberbis density both inside and outside caves, by day and by night. The stomach repletion of individuals sampled early in the morning was significantly higher than later in the day. Most prey were typical of habitats other than caves. A. imberbis was on average more abundant within caves during the day and outside during the night. Our study supports the hypothesis regarding the crucial trophic role of A. imberbis in connecting Mediterranean marine caves with external habitats.

Two different nematode species were recovered from pomegranate decaying fruit in two localities in Southern Italy: the mycetophagus nematode Sheraphelenchus sucus and a bacterial feeder nematode belonging to the Panagrolaimidae (Rhabditida) family. Morphometrics of the Italian population of S. sucus closely resemble that of the type population, whereas some differences were found when compared with another population from Iran. Molecular characterization of the Italian S. sucus using the 18S rRNA gene, D2-D3 expansion domains of the 28S rDNA, the ITS region, and the partial mitochondrial COI were carried out. Sequences of the 18S rRNA gene, the D2-D3 domains, and the ITS were analyzed using several methods for inferring phylogeny to reconstruct the relationships among Sheraphelenchus and Bursaphelenchus species. The bacterial feeder Panagrellus sp. was characterized at the molecular level only. The D2-D3 expansion domains and ITS sequences of this Italian panagrolaimid were determined. The D2-D3 sequences of the Italian panagrolaimid showed 99% similarity with the corresponding sequence of Panagrellus sp. associated with Rhynchophorus ferrugineus . This is the first report on the tritrophic association of S. sucus and Rhabditida that uses both insects and pomegranate fruit as hosts.

Longidorus piceicola , a new geographical and host record from Romania, was described and illustrated on the basis of two populations originating from a coniferous and a deciduous forest. The main morphological characters of specimens from Romania correspond very well with the type material collected from the soil around Picea abies L. (Slovakia) except for the shorter body and tail. The D2-D3 fragment of 28S rDNA from both populations was amplified and sequenced, and the sequences were identical to L. piceicola sequence from Slovakia. The partial 18S-ITS1-5.8S-ITS2 rDNA regions from one of the populations were sequenced for the first time. The evolutionary relationships between L. piceicola and the closest species L. intermedius based on D2-D3 sequence divergence and single-nucleotide polymorphisms are discussed. Although having very low sequence dissimilarity (0.3-0.9 %) both species have distinct morphology and biology. Longidorus piceicola differs from L. intermedius in having a much longer odontostyle, body, distance anterior end - guide ring, a wider lip region, more ventromedian supplements (11 vs 5-7) in the male, and develops through four rather than three juvenile stages. Furthermore, L. piceicola occurs more frequently in association with conifers, while L. intermedius is found mainly in oak forests.

This study examines the impact of bottom trawl fishing on the macrobenthic communities inhabiting the coastal terrigenous mud off the northern coast of Sicily (Western Mediterranean). Two intensely trawled gulfs were compared with two gulfs from which trawling has been excluded for 15 years. The results show a significant effect of trawling on the faunal assemblage and when comparing the mean biomass and the whole isotopic composition of the benthic communities. A similar pattern, although not significant, was found for total abundance, biomass, production/biomass ratio and diversity. Higher abundance and lower biomass were found in the untrawled areas, attributable to the presence of more numerous yet smaller individuals, possibly a consequence of more abundant larger predators that are not removed by trawling, and consequent higher predatory pressure on the benthic macrofauna. The SIMPER analysis evidenced a dominance of burrowing deposit feeding worms (Paraonidae and Cossuridae) in trawled areas, as a result of increased mechanical alteration and hence more organic matter available as food. In contrast, the response to trawling as drawn by the use of trophic markers (i.e., stable isotopes) was less clear. While δ 15 N of benthic taxa did not vary significantly between untrawled and trawled areas, δ 13 C was higher in trawled areas possibly due to high sediment resuspension and consequent intense microbial activity. Mixing models confirmed higher reliance to a detritus-based food web for benthic organisms in the trawled areas. Standard Ellipse Areas (SEAc) as a measure of community niche width were slightly larger in trawled areas, likely due to higher generalism triggered by alteration/removal of the original benthic community., (Copyright © 2016 Elsevier Ltd. This article provides the dataset of operating conditions of a hybrid organic Rankine plant generated by the optimization procedure employed in the research article "A genetic optimization of a hybrid organic Rankine plant for solar and low-grade energy sources" (Scardigno et al., 2015) [1]. The methodology used to obtain the data is described. The operating conditions are subdivided into two separate groups: feasible and unfeasible solutions. In both groups, the values of the design variables are given. Besides, the subset of feasible solutions is described in details, by providing the thermodynamic and economic performances, the temperatures at some characteristic sections of the thermodynamic cycle, the net power, the absorbed powers and the area of the heat exchange surfaces.

Oscheius onirici sp. n. (Nematoda: Rhabditidae) was isolated from a karst cave soil of Central Italy. Molecular and morphological analyses were performed. Total DNA was extracted from individual nematodes and the mitochondrial COI, the ITS containing region, the D2-D3 expansion domains of the 28S rRNA gene and the 18S rRNA gene were amplified and sequenced. BLAST search at NCBI by using all molecular markers revealed that this taxon is similar to Oscheius species. Phylogenetic trees of ITS, 28S and 18S rDNA revealed that O. onirici sp. n. belongs to Dolichura-group. Oscheius onirici sp. n. is characterized by small body size and stoma rhabditoid type. Female reproductive system is amphidelphic. Males are rare with peloderan bursa, spicules slender and small, nine pairs of papillae of different lengths, arranged in a 1+1+1/3+3 pattern. Entomopathogenicity bioassay revealed that this nematode is capable of infecting larvae of Galleria mellonella and Tenebrio molitor.

Introduction: The aim of this survey was to evaluate the qualitative and quantitative relationship among women from Barletta--a national renowned wine center--and their alcohol consumption., Methods: The AUDIT questionnaire was used to assess the prevalence of alcohol hazardous consumption among women. Questionnaires were submitted from March to November 2012. The sample was composed of 150 women older than 13 years of age, selected by stratified sampling based on age group., Results: 107 women were enrolled with a total response rate of 71.3%. 62% of enrolled women consumes alcoholic beverages with a frequency that goes from 2-3 times a week to less than once a month, usually 1 or 2 alcoholic units. The binge-drinking was reported by 5% of women. Women who reported alcohol-related risk behaviors were less than 3%, they were single and between 18 and 60 years old and such behaviors occur less than once a month. The final score, calculated for all the women from their questionnaire answers, was not higher than 8, with an average score of 1.3 (SD = 1.5; range: 0 to 2.8). The comparison of the average scores of the three age groups showed a statistically significant difference (F = 5.8, p = 0.004)., Discussion: Data from literature showed a change in the habits of alcohol intake by the global, European and also Italian population. These changes also affect and involve female. Our study found a quite moderate alcohol consumption among women from Barletta, with only 1% who consumes 3 or more alcohol units and drink more than four times a week and 3% who had hazardous behavior related to their alcohol consumption. Statistical significance was found for the age and the lack of stable relationships. The analysis of characteristics of at risk women (old age and single-status) suggests that much attention should be paid to them and they should represent the main subject of future social interventions to prevent alcohol related problems in the city of Barletta.

This study examined the population structure, reproductive cycle and feeding pattern of the lanternfish Lampanyctus crocodilus in the Balearic Basin (north-west Mediterranean Sea) from a depth of 450 to 1800 m and at a seasonal scale. Juveniles were mainly located at shallower depths, but also at deepest stations in autumn, while adults mostly inhabited intermediate depths with their centre of population density (CPD) located at 800-1000 m of depth. The migration of adults to deeper depths was detected in late summer to autumn, probably linked to the occurrence of nepheloid layers at c. 1200 m, which in turn enhances the biomass of the zooplankton prey. The diet was mainly based on euphausiids and mysids, with marked seasonal variations both on the upper (450-800 m) and lower (1000-1800 m), where suprabenthic gammariids and pelagic decapods were also dominant. Stomach fullness increased from winter to autumn on the US, while it had a maximum in spring on the LS, in parallel with high consumption of gelatinous zooplankton, which is probably more available after the phytoplankton bloom in late winter. Reproduction occurred in winter, confirmed by the higher percentage of mature females and high gonadosomatic indices (I(G)) at both depth ranges. Hepatosomatic indices (I(H)) showed an inverse trend to I(G) on the US, except in autumn, and was almost parallel on the LS, probably attributable to the migration of adults, which determined different temporal schemes in energy use and storage for reproduction on the US v. LS. Consistent with the different patterns observed at the two depth ranges, environmental drivers of fullness (i.e. feeding intensity) and I(G) (as a proxy of reproductive cycle) differed on the US and LS. The biomass of mysids and euphausiids was the greatest explanatory variables of fullness on the US and LS, pointing to the increasing feeding intensity when a resource was more available. I(H) also explained fullness, suggesting that greater feeding intensity in pre-reproductive periods enabled energy storage in the liver. I(G) was linked directly (i.e. mysids) or indirectly (i.e. surface primary production recorded 2 months before sampling) to food availability, implying a rapid response to vertical food inputs by deep-sea predators. Also, I(G) in L. crocodilus was related to population density, which suggests aggregations for reproduction. Estimates of L. crocodilus trophic levels, and of other accompanying mesopelagic fishes, indicated that the species feed through a continuum spanning the third trophic level, confirming the key role of mesopelagic fishes in transferring organic carbon between trophic levels. Trophic niche segregation among mesopelagic species was pronounced and non-overlapping groups could be distinguished because of the different vertical distribution and migratory behaviour. The study highlights the important role of the benthic boundary layer in sustaining benthopelagic communities in the deep Mediterranean Sea and the need to study the biology of a species throughout its whole depth range and not just at exploited depths (i.e. fishing grounds)., (© 2014 The Fisheries Society of the British Isles.)

One form of liver steatosis, namely Non-Alcoholic Fatty Liver Disease (NAFLD), is a worrisome health problem worldwide characterized by intrahepatic triacylglycerol (TG) overaccumulation. NAFLD is a common feature of metabolic syndrome being often associated with obesity, dyslipidemia and diabetes and mostly closely linked to insulin resistance. The mechanism of NAFLD pathogenesis is object of intense investigation especially regarding complex systems ultimately resulting in excessive TG deposition in hepatocytes. However, scarce is the attention about the relevance of hepatic import of glycerol, the other primary source (as glycerol-3-phosphate) of increased TG in hepatocytes. Obese leptin-deficient (ob/ob) mice, an animal model of NAFLD, were used to evaluate the functional involvement of Aquaporin-9 (AQP9), the major pathway of liver glycerol entry, in hepatosteatosis. By RT-PCR and qPCR, the level of Aqp9 mRNA in the liver of starved obese mice was comparable with the corresponding control lean littermates. By immunoblotting, the AQP9 protein at the hepatocyte sinusoidal plasma membrane of obese mice was markedly lower (33%) than lean mice, a finding fully confirmed by immunohistochemistry. By stopped-flow light scattering, the liver glycerol permeability of ob/ob mice was significantly lower (53%) than lean mice, a finding consistent with both the observed down-regulation of AQP9 protein and increased level of plasma glycerol characterizing obese mice. In summary, our results suggest implication of AQP9 in liver steatosis. The reduction of hepatocyte AQP9 and, consequently, glycerol permeability might be a defensive mechanism to counteract further fat infiltration in liver parenchyma.

Unlabelled: Nonalcoholic steatosis is a liver pathology characterized by fat accumulation and severe metabolic alterations involving early mitochondrial impairment and late hepatocyte cell death. However, mitochondrial dysfunction mechanisms remain elusive. Using four models of nonalcoholic steatosis, i.e., livers from patients with fatty liver disease, ob/ob mice, mice fed a high-fat diet, and in vitro models of lipotoxicity, we show that outer mitochondrial membrane permeability is altered and identified a posttranslational modification of voltage-dependent anion channel (VDAC), a membrane channel and NADH oxidase, as a cause of early mitochondrial dysfunction. Thus, in nonalcoholic steatosis VDAC exhibits reduced threonine phosphorylation, which increases the influx of water and calcium into mitochondria, sensitizes the organelle to matrix swelling, depolarization, and cytochrome c release without inducing cell death. This also amplifies VDAC enzymatic and channel activities regulation by calcium and modifies its interaction with proteic partners. Moreover, lipid accumulation triggers a rapid lack of VDAC phosphorylation by glycogen synthase kinase 3 (GSK3). Pharmacological and genetic manipulations proved GSK3 to be responsible for VDAC phosphorylation in normal cells. Notably, VDAC phosphorylation level correlated with steatosis severity in patients., Conclusion: VDAC acts as an early sensor of lipid toxicity and its GSK3-mediated phosphorylation status controls outer mitochondrial membrane permeabilization in hepatosteatosis., (Copyright © 2012 American Association for the Study of Liver Diseases.)

A class II hybrid sol-gel material was prepared starting from zirconium(IV) propoxide and 2,4-pentanedione and its catalytic activity in the removal of the herbicide 4-chloro-2-methylphenoxyacetic acid (MCPA) was revealed. The thermal and structural characterization, performed by thermogravimetry, differential thermal analysis, and diffuse reflectance Fourier transform infrared spectroscopy, demonstrated the hybrid nature of the material. The structure of the material can be described as a polymeric network of zirconium oxo clusters, on the surface of which large part of Zr(4+) ions are involved in strong complexation equilibria with acetylacetonate (acac) ligands. The incubation of MCPA in the presence of this material yielded an herbicide removal fraction up to 98%. A two-step mechanism was proposed for the MCPA removal, in which a reversible first-order adsorption of the herbicide is followed by its catalytic degradation. The nature of the products of the MCPA catalytic degradation as well as the reaction conditions adopted do not support typical oxidation pathways involving radicals, suggesting the existence of a different mechanism in which the Zr(4+):acac enol-type complex can act as Lewis acid catalyst.

A single-stage sol-gel route was set to entrap yeast cells of Lipomyces starkeyi in a zirconia (ZrO(2)) matrix, and the remediation ability of the resulting catalyst toward a phenoxy acid herbicide, 4-chloro-2-methylphenoxyacetic acid (MCPA), was studied. It was found that the experimental procedure allowed a high dispersion of the microorganisms into the zirconia gel matrix; the ZrO(2) matrix exhibited a significant sorption capacity of the herbicide, and the entrapped cells showed a degradative activity toward MCPA. The combination of these effects leads to a nearly total removal efficiency (>97%) of the herbicide at 30 °C within 1 h incubation time from a solution containing a very high concentration of MCPA (200 mg L(-1)). On the basis of the experimental evidence, a removal mechanism was proposed involving in the first step the sorption of the herbicide molecules on the ZrO(2) matrix, followed by the microbial degradation operated by the entrapped yeasts, the metabolic activity of which appear enhanced under the microenvironmental conditions established within the zirconia matrix. Repeated batch tests of sorption/degradation of entrapped Lipomyces showed that the removal efficiency retained almost the same value of 97.3% after 3 batch tests, with only a subsequent slight decrease, probably due to the progressive saturation of the zirconia matrix.

The full-length cDNA and the corresponding gene of the heat shock protein 90, Mt-Hsp90, were isolated and characterized in the plant parasitic nematode Meloidogyne artiellia. The full-length Mt-Hsp90 cDNA contained a 5' untranslated region (UTR) of 45 bp with the 22 bp trans-spliced leader SL1, an ORF of 2172 bp encoding a polypeptide of 723 amino acids and a 3' UTR of 191 bp. The deduced amino acid sequence of Mt-hsp90 showed high similarity with other known Hsp90s. Five conserved amino acid signatures indicated that Mt-hsp90 is a cytosolic member of the Hsp90 family. The gene consists of 10 exons and 9 introns, a more expanded gene structure compared to the corresponding Caenorhabditis elegans gene, daf-21. Mt-hsp90 gene was constitutively expressed at high levels in all developmental stages of M. artiellia. Egg masses and second stage juveniles (J2s) were exposed at 5 degrees and 30 degrees C for different periods of times in order to explore the impact of adverse temperature on Mt-hsp90 gene expression. Expression levels of Mt-hsp90 were examined by fluorescent real-time PCR. At 30 degrees C a burst of expression for Mt-hsp90 was observed in J2s after 2 h of heat shock treatment, then expression dropped with longer exposing times, although remaining still relatively high after 24 h. This temperature did not affect Mt-hsp90 gene expression in the egg masses. However, egg masses exposed at 5 degrees C showed a little but gradual increase in the mRNA level with time. By contrast, no significant changes in the Mt-hsp90 level were observed in J2s exposed to cold. These data show that egg masses and J2s exposed to cold and heat stresses have different expression profiles suggesting that Mt-Hsp90 may provide a link between environmental conditions and the life cycle of the nematode.

The osmotic movement of water into and out of the mitochondrial matrix underlies the extraordinary plasticity that characterizes mitochondria, a feature of pivotal importance to cell bioenergetics and signaling, and of critical relevance to life-and-death cell decision. However, the biophysics and identity of mitochondrial water transport had remained mostly unexplored, until recent works suggesting high water permeability and the presence of multiple facilitated pathways of water diffusion in liver mitochondria. Here, we attempt to summarize our current view of the mechanisms of mitochondrial water transport and possible relevance of the channel-mediated pathways created by mitochondrial permeability transition, aquaporins and protein/lipid specializations. Assessing the molecular bases and dynamics of mitochondrial water permeability will help to answer the much-debated question over the role of mitochondria.

Animals and plants both respond rapidly to pathogens by inducing the expression of defence-related genes. Within this context, a prominent role has been assigned to the lysozyme. In the present study we isolated and carried out detailed analysis of the lysozyme gene in the plant nematode Meloidogyne artiellia. The expression of lysozyme was up-regulated following exposure of M. artiellia juveniles to the Gram-negative bacterium Serratia marcescens. On the other hand, when isolated eggs containing embryos at various developmental stages were challenged with bacteria, no increase in lysozyme expression was detected. Evidence of lysozyme expression regulation was obtained in the case of adult male and females worms collected from soil. The lysozyme gene was expressed solely in the nematode intestine and, as it is predicted to be secreted, may protect the nematode from microbial infections originating in the intestinal lumen or in the pseudocoelom. This paper demonstrates, to our knowledge for the first time, the immune response to infection in a plant parasitic nematode.

Seasonal and spatial variations in metal concentrations and pH were found in a stream at a restored copper mine site located near a massive sulfide deposit in the Foothill copper-zinc belt of the Sierra Nevada, California. At the mouth of the stream, copper concentrations increased and pH decreased with increased streamflow after the onset of winter rain and, unexpectedly, reached extreme values 1 or 2 months after peaks in the seasonal hydrographs. In contrast, aqueous zinc and sulfate concentrations were highest during low-flow periods. Spatial variation was assessed in 400 m of reach encompassing an acidic, metal-laden seep. At this seep, pH remained low (2-3) throughout the year, and copper concentrations were highest. In contrast, the zinc concentrations increased with downstream distance. These spatial patterns were caused by immobilization of copper by hydrous ferric oxides in benthic sediments, coupled with increasing downstream supply of zinc from groundwater seepage.

Selected key components of the continental shelf benthic and demersal communities were monitored for the two years following the Prestige oil spill (POS) in order to identify the possible ecological effects of the oil. This work includes the first results regarding changes in abundance, distribution and food habits of hake (Merluccius merluccius), four-spot megrim (Lepidorhombus boscii), Norway lobster (Nephrops norvegicus) and Pandalid shrimp (Plesionika heterocarpus) populations of Galician and Cantabrian Sea shelves following the POS. Significant reductions in the abundance of Norway lobster, Plesionika heterocarpus and four-spot megrim were detected in the POS maximum impact area, located over the Galician shelf. Noteworthy recoveries were observed in the 2004 abundance indices of four-spot megrim and Plesionika. On the other hand, no significant effects were detected in the abundance or distribution of hake juveniles even though the tar aggregates were bound by the same oceanographic drift events as the hake recruits were during the winter of 2003 (Navidad current) in different water column layers of the Cantabrian Sea. Feeding patterns of the four species analysed did not present apparent modifications that can be related to the POS.

A single chitin synthase gene is responsible for chitin production in the eggshells of the plant parasitic nematode Meloidogyne artiellia. In this paper we describe a functional analysis of this gene using RNAi as well as further analysis of two similar genes from the free-living nematode Caenorhabditis elegans. The parasitic life-style of M. artiellia required the development of a novel method for delivery of dsRNA to nematode eggs that may be of utility in other experimental systems. C. elegans chitin synthase genes were silenced by feeding nematodes bacteria expressing appropriate chitin synthase dsRNA from a plasmid vector, while M. artiellia egg masses were soaked in dsRNA solution. The results obtained demonstrated that the synthesis of chitin continues to take place in nematode eggs within the egg sac in the parasitic nematode, and that the removal of this activity affects egg development in both C. elegans and M. artiellia. The method described here provides a new way of investigating gene function in plant parasitic nematodes allowing the validity of potential target genes for novel control methods to be assessed. Furthermore, since intact egg cells within the gelatinous matrix of M. artiellia are permeable to dsRNA, eggs of other nematodes may also be similarly permeable to dsRNA and therefore amenable to use with dsRNAi.

The structure of the fuscopeptins, bioactive lipodepsipeptides produced in culture by the gramineae pathogen Pseudomonas fuscovaginae, has been determined. The combined use of FAB mass spectroscopy NMR spectroscopy and chemical and enzymatic procedures allowed one to define a peptide moiety corresponding to Z-Dhb-D-Pro-L-Leu-D-Ala-D-Ala-D-Ala-D-Ala-D-Val-Gly-D-Ala-D-Val-D-Ala-D- Val-Z-Dhb-Da-Thr-L-Ala-L-Dab-D-Dab-L-Phe with the terminal carboxyl group closing a macrocyclic ring on the hydroxyl group of the allothreonine residue. The N-terminus is in turn acylated by 3-hydroxyoctanoate in fuscopeptin A and 3-hydroxydecanoate in fuscopeptin B. Some preliminary data on the biological activity of fuscopeptins are also reported.

The rapid acceptance of minimally invasive surgery in the United States has largely occurred without statistical proof of its superiority over traditional methods. All players in the healthcare field now see the need for valid outcome studies supporting the efficacy of new treatment techniques. Percutaneous laser disc decompression (PLDD) will gain wide acceptance only if it is statistically shown to be a safe and effective alternative treatment of a lumbar disc herniation. To that end, a central PLDD registry has been developed and implemented into clinical practice at several centers. This article reviews healthcare trends that motivated the creation of this data repository and discusses the history of its development. The currently used PLDD evaluation form is outlined and its utility is discussed. Most importantly, preliminary PLDD results and complications based on the data received are presented and analyzed.

DNA in cell nuclei is organized in large loops, which are formed by the binding of DNA to proteins present in a nuclear structure called matrix or scaffold. There is ample evidence that these interactions have not only a structural role, but also a functional one. Studies of these interactions have so far been carried out mainly in vitro. Their relevance, therefore, for the in vivo situation is not proven. We have analyzed the DNA-protein interactions directly in the intact nucleus by means of cross-linking reactions. Cross-linking by UV irradiation and by cis-diamine dichloro platinum (II) have been performed on nuclei from chicken liver and compared. The platinum complex has been found to be more efficient. The proteins complexed to DNA have been isolated and analyzed, and have been found to be enriched in species present in the nuclear scaffold, independently from its method of preparation.

Experimental results are reported on structural and thermodynamic modifications induced by gamma-irradiation in model membranes. Differential scanning calorimetry (DSC) and X-ray diffraction were used to study the different phases and associated transitions of dipalmitoylphosphatidylcholine after 60Co gamma-irradiation. Changes were observed in the shape of calorimetric peaks and in the corresponding enthalpy. The repetition distance of the layers increases while the distances related to the aliphatic chains decrease as a function of gamma-irradiation time. Moreover, an increase in the hexagonal symmetry with increasing dose was detected. No disappearance of a pre-transition was detected even at high doses.

An investigation of the structural and thermodynamical modifications induced by gamma-irradiation in model membranes is reported. Differential scanning calorimetry and X-ray diffraction were used to study the different phases and associated transitions of distearoylphosphatidylcholine multilamellar liposomes after 60Co gamma-irradiation. Changes were observed in the shape of the calorimetric peaks and in the corresponding phase transition temperatures. In particular a shoulder was observed at about 20 kGy. The three phases characteristic of lecithins with identical acyl chains were detected also for the highest radiation dose. The formation of lysolecithin and stearic acid upon phospholipid degradation was observed. The lysolecithin concentration increases as a function of irradiation dose, until a saturation value is reached at 40 kGy. These results correlate quite well with those obtained for interlayer and interchain distances and for the width of the main phase transition calorimetric peak. At the highest dose (approximately equal to 80 kGy) cross-linked adjacent radicals and other molecular species are also formed. Appreciable differences, and some similarities, in the behaviour of DSPC and DPPC liposomes under gamma-irradiation were observed.
