Renal cortical slices were used to determine the toxicity of N-acetyl-S-(1,2-dichlorovinyl)-L-cysteine (N-acetyl-DCVC) as well as to investigate the transport and metabolism of S-(1,2-dichlorovinyl)-L-cysteine (DCVC) and the N-acetyl derivative. N-Acetyl-DCVC produced dose- and time-dependent decreases in intracellular K+ content and lactate dehydrogenase activity. Histopathology demonstrated an initial S3 lesion followed by a lesion inclusive of all proximal tubules. N-Acetyl-DCVC was shown to be transported via the organic anion system by its ability to inhibit PAH transport by the cells and the ability of probenecid to decrease uptake (80%) and toxicity of N-acetyl-DCVC. DCVC, in contrast, was not transported by the organic anion system, but may be transported by one or more amino acid systems. N-Acetyl-DCVC must be deacetylated before undergoing metabolism by beta-lyase. This process must occur since covalent binding of a 35S-labeled reactive product from N-acetyl [35S]DCVC is observed within 1 hr. Both the uptake inhibitor, probenecid, and aminooxyacetic acid (AOAA), a beta-lyase inhibitor, decreased the covalent binding from N-acetyl [35S]DCVC (80 and 50%, respectively), but only AOAA inhibited the covalent binding of DCVC. AOAA also partially inhibited the toxicity of DCVC and N-acetyl-DCVC as determined by intracellular K+ content, lactate dehydrogenase activity, and histopathology. Despite the fact that a separate transport system and an additional enzymatic step (deacetylation) are required, N-acetyl-DCVC produces a lesion with similar intratubular specificity to that seen with DCVC. Therefore, the S3 specificity seen in vivo could be produced by either compound.

The standard Bradford protein assay is insensitive to collagen. But if a small, sub-threshold amount of SDS is added to the sample, the response to collagen is increased by at least an order of magnitude, while, on average, the sensitivity for non-collagens is decreased by approximately a factor of 2. As a result, comparable color formation is achieved with both collagen and non-collagens. The addition of protein to a sub-threshold amount of SDS results in the formation of a green color measurable as an increase in absorbance at 700 nm, in contrast to the blue color measured at 595 nm in the standard assay. Depending upon the source, the threshold level for SDS varies from 30 to 50 microgram. The response to protein is linear up to approximately 40 microgram of protein per ml of reagent.

The effects of insulin and glucagon on cAMP accumulation, protein kinase activation, and glycogenolysis were investigated in isolated rat hepatocytes. Glucagon (0.01 nM to 10 micro M) increased the activation state of protein kinase and the rate of glucose accumulation. Addition of 1.0 nM insulin to cells preincubated with 0.1 nM glucagon attenuated the rate of glucose accumulation, but did not alter the protein kinase activity ratio. Addition of 0.1 nM glucagon to cells preincubated with 1.0 nM insulin caused a rapid activation of protein kinase; however, glycogenolysis was not immediately affected. These effects were enhanced with pharmacological concentrations of glucagon and insulin. These data indicate that the degree of protein kinase activation does not always correlate temporally or quantitatively with rates of glycogenolysis in liver cells exposed to insulin and glucagon.

A rapid and simple procedure for the preparation, directly from clotted whole blood, of bovine fibrin free of cell membrane contaminants is described. The procedure exploits the ability of detergents to solubilize and extract cell membrane proteins from homogenized clot fragments. The non-denaturing detergents, TX-100 and DOC, used in sequence, or the denaturing detergent, SDS, used alone, proved equally effective. The resulting fibrin has the solubility properties and the subunit composition on SDS gels of highly cross-linked fibrin.

Isolated hepatocytes carry out the N-demethylation of dansylamide at near linear rates for up to 8 h. This reaction was measured by following the release of tritium into water on hydroxylation of 3H-labeled methyl groups. The competitive inhibition of dansylamide by dansylated amino acids was studied in this system as an example of competing drug metabolism in a series of compounds which are identical around the site of metabolism and different remote to that site. A correlation between lipid solubility and the Ki was not found over the entire range of substrate analogues. While most of the high Km inhibitors seem to correlate with lipid solubility, the highly lipophilic derivatives of the leucines and phenylalanine are in a separate group. Lipid solubilities of the dansylated amino acids were little affected by changes in pH and thus behaved as "essential nonelectrolytes".

To reduce structural complexity, rabbit kidneys were sliced perpendicular to their cortical-papillary axis to isolate four distinct cell groupings. This positional orientation allows identification of each renal cell type based on its location within the slice. A mechanical slicer was used to make several precision-cut slices rapidly from an oriented cylindrical core of renal tissue, with minimal tissue trauma. Slices were then submerged under a gently circulating oxygenated media in a fritted glass support system that maintains viability (intracellular K+/DNA ratio) and structural integrity (histology) for at least 30 h. A high dose of mercuric chloride (10(-3) M) was used to demonstrate the structural and biochemical changes of intoxicated slices. This method provides a controlled subchronic in vitro system for the study of the individual cell types involved in cell-specific renal toxicities and may also be a useful tool for addressing other pharmacological and physiological research questions.

An all-glass apparatus for perfusing isolated rat livers was constructed for use in studies of the metabolism by, or action on, the liver of lipophilic substrates. Other designs, which generally include plastic connections or cannulae, can have unnecessarily complicated kinetics because of adsorption of the agent to the plastic parts. Two versions of an all-glass perfusion apparatus are described in detail. Both instruments operate on the same basic principle, but differ in size, total recirculating volume, pump rate, perfusion head pressures, maximum perfusate flow rate, maximum linear velocity in the perfusion head, and oxygenation efficiency. Although the maximum protein handling capacity differs for the two apparatuses, this capacity is so limited that it disallows the use of albumin in the perfusate.

The conjugation of harmol and the disposition of harmol metabolites by isolated rat hepatocytes were investigated. Harmol sulfation was saturated at very low concentrations and exhibited a maximum velocity of 1.2 +/- 0.2 nmol/min/10(6) hepatocytes. Glucuronidation of harmol proceeded with a Km of 17 +/- 5.7 microM and a maximal velocity of 2.1 +/- 0.3 nmol/min/10(6) hepatocytes. After synthesis, harmol glucuronide and harmol sulfate were distributed between cells and incubation media, and at equilibrium, a large concentration gradient between cells and media existed for both conjugates. Experiments with preformed metabolites indicated that hepatocytes removed harmol glucuronide from the incubation media by single Michaelis-Menten process with a Km of 384 +/- 63 microM and Vmax of 1.8 +/- 0.3 nmol/min/10(6) hepatocytes. Harmol sulfate was taken up by two Michaelis-Menten processes, a high affinity process with a Km of 33 +/- 8 microM and a Vmax of 0.97 +/- 0.2 nmol/min/10(6) cells and a low affinity process with a Km of 452 +/- 71 microM and Vmax of 25.2 +/- 4.1/min/10(6) hepatocytes. Harmol sulfate was released from hepatocytes by a process which did not exhibit saturation. Hepatocytes converted preformed harmol glucuronide to harmol sulfate.

We describe a simple and rapid procedure for the isolation of proline and hydroxyproline from fossil bone based on the isolation of gelatin, hydrolysis, purification with XAD-2, deamination of primary amino acids with aqua regia, and separation of the imino acids by cation exchange chromatography. This procedure will provide material for accurate bone carbon dating, and stable carbon isotope ratio determinations for the evaluation of paleodiets.

A method is described for the rapid preparation of lung cell fractions enriched in type II alveolar pneumocytes. Isolated perfused rabbit lungs are exposed to Fe3O4 by tracheal lavage, which permits pulmonary alveolar macrophages to phagocytize the particles. Alveolar epithelial cells are then selectively freed from the basement membrane matrix by critical placement of collagenase and elastase. Detached cells are harvested either by repeated tracheal lavage or by mincing the lobes and filtering freed cells through a series of nylon mesh sieves. Iron oxide-containing macrophages are then removed from the harvested cells by a strong magnetic field. A final sizing of the macrophage-depleted suspension yields a preparation enriched in alveolar type II cells. Eight million viable cells (95% type II) were obtained per rabbit lung when harvested by lavage, while 32 x 10(6) (88% type II) cells were obtained from minced lungs. These values for cell yield and relative purity are comparable to previously described separation methods that depend upon differences in cell density or size. A major advantage of the magnetic separation procedure is the substantially shortened preparation time, typically 2 hr instead of 4. The viability (90-95%), oxygen consumption (88 nmol/10(6) cells/hr), and incorporation of [14C]acetate and [14C]choline (0.44 and 0.115 nmol/10(6) cells/hr, respectively) indicate that these cells will be suitable for pharmacologic and toxicologic investigation.

In human diabetes mellitus, thickening of the microvascular endothelial cell basement membrane has been documented by quantitative ultrastructural studies in a variety of vascular beds. We studied the thickness of the cerebral cortical capillary basement membrane in frontal and occipital lobes from 21 diabetics and 16 nondiabetics, and found that those from diabetics were significantly thicker. Skeletal muscle capillary basement membranes have been shown to thicken with increasing age. However, only in the frontal cortex of diabetics was a significant relationship observed between capillary basement membrane thickening and increasing age. We conclude that, along with many of the previously studied microvascular beds, the cerebral cortical microvasculature displays thickening of the endothelial cell basement membrane in diabetes mellitus. No currently known disease is associated with this microvascular change, unlike the closely related retinal vasculature, where the association with diabetic retinopathy is established.

Glucose intolerance has been observed following diphenylhydantoin (DPH) intoxication. Because of this association between DPH and hyperglycemia, the effect of DPH on insulin release in vitro using preparations of isolated islets of Langerhans and pancreatic pieces was examined. In concentrations identical with those considered necessary for adequate anticonvulsant therapy in man, DPH markedly decreases the insulin secretory response of pancreatic pieces to methacholine, 1 mug/ml, tolbutamide, 250 mug/ml, and glucose, 200 mg/100 ml, without any demonstrable alteration in the oxidative conversion of glucose-1-(14)C or glucose-6-(14)C to (14)CO(2) by isolated islets. This DPH-induced inhibition of insulin secretion is not reversed by higher concentrations of glucose (600 mg/100 ml) or by increasing concentrations of extracellular calcium ion (4-6 mmoles/liter). 0.1 mM potassium and 10(-4) M ouabain, however, effectively restore the DPH-induced block of insulin secretion in pancreatic pieces. 60 mM potassium ion, on the other hand, not only restores the insulin secretory response to glucose (200 mg/100 ml) but results in an added stimulation of insulin secretion in the presence of DPH. In the presence of DPH, (22)Na accumulation by isolated islets is decreased by 26-40% as compared with controls. Such evidence is considered to indirectly support the postulate that the electrophysiological properties of DPH on the pancreas are due to a stimulation of the membrane sodium-potassium-magnesium ATPase.
