Acetylcarnitine is an endogenous substance, active in the CNS, but its exact role in neurotransmission is still unknown. The present study was made to analyze by means of iontophoretic administration on single neurones of the medullary-pontine reticular formation in the rat the effects of the D- and L-acetylcarnitine on spontaneous and glutamate-evoked firing and on responses to applications of some other putative transmitters, such as acetylcholine, 5-hydroxytryptamine (5-HT), gamma-aminobutyric acid (GABA) and noradrenaline. L-Acetylcarnitine increased both spontaneous and evoked discharges, and potentiated cholinergic and serotonergic responses. The D-isomer was found to be almost ineffective. Neuronal responses to L-acetylcarnitine were reduced by dihydro-beta-erythroidine and enhanced by eserine, while atropine was ineffective. L-Acetylcarnitine increased both excitatory and inhibitory responses to 5-HT and its effect on the increase in firing rate induced by serotonergic drugs was reduced by dihydro-beta-erythroidine, but not by atropine. L- and D-Acetylcarnitine did not affect GABAergic or noradrenergic transmission. These results suggest a stereospecific facilitatory action of acetylcarnitine on neuronal responses to acetylcholine and 5-HT. It is hypothesized that this action could be exerted at a presynaptic level, at least with regard to serotonergic-induced responses, and that nicotinic receptors may be involved in the synaptic mechanism of release or reuptake of both acetylcholine and 5-HT.

Protein patterns of mitochondrial outer membrane, inner membrane, and matrix from non-synaptic (free) mitochondria from rat cerebellum at different ages (4, 8, 12, 16, 20, and 24 months) were analyzed by gel electrophoresis. Acute L-acetylcarnitine treatment was performed by a single i.p. injection (100 mg/kg body weight) of the substance 60 min before the sacrifice of the animals. Different age-dependent changes were obtained for the proteins of the three fractions. The amount of some protein subunits increased and/or decreased after drug treatment. In particular, protein composition of the inner mitochondrial membrane showed significant age-related modifications. This result probably indicates differences in protein synthesis and/or turnover rates in the various mitochondrial compartments during aging. Acute L-acetylcarnitine treatment caused: a high increase in the amount of one inner membrane protein with Mw 16 kDa, at all the ages studied; a decrease in the amount of many other inner membrane proteins; modifications of some matrix proteins. Our results show that in vivo administration of L-acetylcarnitine affects mainly the inner membrane protein composition of cerebellar mitochondria.

In this pharmacological study the authors give an extensive review of existing literature regarding the activity of carnitine and acetyl carnitine on the central nervous system. Carnitine and acetyl carnitine enter the neuro-biochemical patterns in the endoneuronal system, and affect neurotransmission in synapsis.

Using the microelectrode technique, the effects of l-carnitine (LC) and acetyl-l-carnitine (ALC) on the changes in the transmembrane action potential of the canine ventricular muscle induced by palmityl-l-carnitine (PLC) were studied in comparison with those of disopyramide (D). LC (5 X 10(-3) M) itself had no effect on the electrophysiological parameters of the ventricular muscle. ALC (5 X 10(-3) M) increased the maximum rate of rise (dV/dt max) slightly and decreased the action potential duration (APD), although these changes were not statistically significant. D (1.5 X 10(-3) M) decreased dV/dt max and prolonged APD and the absolute refractory period (ARP). PLC (3 X 10(-4) M) decreased the resting membrane potential, action potential amplitude and dV/dt max, and it shortened APD and ARP. LC and ALC (5 X 10(-3) M) improved the electrophysiological derangement produced by PLC to the same degree. On the other hand, application of D (1.5 X 10(-5) M) resulted in no improvement of the electrophysiological derangement produced by PLC.

By using fluorescence microscopy and microfluorimetric techniques, the effects of ageing and of 11 months acetyl-L-carnitine (ALCAR) treatment on lipofuscin deposition within the cytoplasm of pyramidal neurons of rat prefrontal cortex and hippocampus (CA3 field) were assessed. No lipofuscin autofluorescence was observed in the nerve cell bodies of neurons under study in young rats (3 months of age), but lipopigment had accumulated in the same nerve cells of senescent rats (22 months of age). ALCAR administration significantly reduced the accumulation of lipofuscin within pyramidal neurons of the brain areas examined.

Intracerebral injection of L-acetylcarnitine in rats induced interictal and ictal epileptic phenomena with immediate onset, lasting up to 4 h. Pretreatment with systemic atropine prevents all epileptiform phenomena. Local injection of muscimol and THIP abolish ictal events, but not single spikes. L-carnitine induced only ictal discharges with a latency of 40-90 min. Acetylcarnitine epileptogenic properties are probably related to muscarinic agonism. The transition from interictal to ictal events may involve failure of GABAergic mechanisms.

D-L, Acetylcarnitine was applied microiontophoretically to neurones in the brainstem of the rat, to evaluate its possible effects. The results support the hypothesis that D-L, acetylcarnitine acts on the neuronal membrane, producing selective excitatory responses of the neurones examined. The possible role of this substance at the single neurone level is discussed in relation to the results obtained by investigating the interaction between D-L, acetylcarnitine and neurotransmitters, such as acetylcholine, gamma-aminobutyric acid (GABA), serotonin and noradrenaline.

Endogenous components of evoked potentials resembling P300 in human beings were sequentially studied in four cynomolgus monkeys (Macaca fascicularis using an auditory "oddball" paradigm, after classical conditioning training. When robust P300-like signals were obtained, each animal underwent further recording sessions in which a placebo, L-carnitine or L-acetylcarnitine (LAC) was injected. Only LAC induced an amplitude increment of P300 waves in all the animals and a latency decrement in two animals. Furthermore a transient effect with amplitude reduction on the primary cortical auditory evoked potentials (AEP) both to rare and frequent tones was also recorded in all the animals. This effect resembled the AEP amplitude reduction induced by physostigmine administration. The findings suggest an effect of LAC on normal processes accompanying cognition.

The effects of long-term dosing with acetyl-L-carnitine (ALC) were examined in aged rats, and they were compared with those in young rats. ALC significantly reduced the lipofuscin deposition in the brain of aged rats. Emotional parameters such as locomotor activity and rearing behavior are lower in aged rats than in young rats, and these behaviors decreased in both age groups during the experiments. ALC diminished the decrease of these emotional behaviors, especially in rearing behavior in the aged rats. Furthermore, ALC had no effect on body weight gain. These results might reflect one of the main beneficial pharmacological mechanisms of ALC in clinical use.

The effect of methylazoxymethanol (MAM), administered on the 15th gestational day, on the density and pattern of muscarinic cholinergic receptors in some brain areas was assessed using combined radioreceptor binding and autoradiographic techniques. The effect of 15 days' treatment with acetyl-L-carnitine on the same parameter was also assessed. The density of muscarinic cholinergic receptors was found to be increased in the brain of MAM-microencephalic rats. Acetyl-L-carnitine administration caused a significant reduction in the density of receptors under study. A possible role of acetyl-L-carnitine in restoring central cholinergic neurotransmission is discussed.

Renshaw cells, which mediate the recurrent inhibition of spinal a-motoneurones, are activated by acetylcholine, both through motoneurone collaterals and the reticulo-spinal system. Since it is known that L-acetylcarnitine (L-AC) has central cholinergic effects, we tested in ten normal subjects and three spastic patients the ability of L-AC to induce changes in excitability of the Renshaw cells. These were activated by a conditioning monosynaptic reflex of the soleus muscle, evoked by electrical stimulation of the tibial nerve, and the resulting recurrent inhibition of the motoneurones was assessed by a subsequent monosynaptic reflex (H'). Recurrent inhibition was tested prior to, during and after an intravenous administration of a solution containing 2000 mg of L-AC. L-AC administration proved to be able to induce in all subjects a decrease in the H'-reflex. This effect ensued approximately 30 min after onset of L-AC administration, reached the peak after 40 min and vanished in about one hour. The extent of the decrease in H' varied among subjects, being on the average 22% of the control values. A relationship was found between duration of L-AC administration and time for reaching the maximal effect. These results show that L-AC is able to decrease a-motoneurone excitability by increasing Renshaw cell activity, both in normal and spastic subjects.

Spermatozoa were collected from the rat caput epididymidis by micropuncture and their motility assessed after dilution in physiological saline containing carnitine or related compounds. L(+)-Carnitine caused, 2 min after dilution, a transient stimulation of the motility of spermatozoa with low initial motility. No stimulatory effects were seen on spermatozoa which had high initial motility. The D-isomer inhibited the motility of spermatozoa with high initial motility after 2 min and all compounds tested appeared to inhibit, at 20 min after dilution, the motility of spermatozoa with high initial motility. Acetyl-L-carnitine and acetyl-D-carnitine stimulated the motility of spermatozoa with low initial motility. This study suggests that carnitine may be important in the development by spermatozoa of the potential for motility and also to maintain mature spermatozoa in a quiescent state.

We have incubated heart mitochondria with ferrous ions as catalyst of lipid peroxidation. Ferrous ions induced an increase of malondialdehyde formation and a reduction of mitochondrial oxygen consuming and calcium transporting capacities. L-Carnitine and Acetyl-L-Carnitine failed to prevent mitochondrial damage. Propionyl-L-Carnitine significantly improved mitochondrial function, but failed to reduce malondialdehyde formation. This protective effect was specific for Propionyl-L-Carnitine as propionic acid and L-Carnitine did not modify mitochondrial damage.

The distribution of choline acetyltransferase (ChAT, EC 2.3.1.6.) in the heart of adult rats has been reinvestigated in view of recent discoveries that acetylcholine (ACh) can be synthesized not only by ChAT, but also by carnitine acetyltransferase (CarAT, EC 2.3.1.7) and that it is possible to distinquish between the ACh-synthesizing activity of ChAT in intramuscular nerves and the CarAT-mediated extraneural synthesis of ACh by means of bromoacetylcholine (BrACh), a specific inhibitor of ChAT. BrACh (0.002 mmol/l) has been found to inhibit the synthesis of ACh in the atria by 66-85% and in the ventricles by only 19-29%. Bromoacetylcarnitine (BrACar, 0.02 mmol/l), and inhibitor of CarAT, inhibited the synthesis of ACh in the atria by 34% and in the ventricles by 74-80%. These findings indicate that ChAT is responsible for most of the synthesis of ACh observed in the homogenates of the atria; in the ventricles, it catalyses only a minor portion of the total ACh synthesis observed. In the investigation of the regional distribution of ChAT in the heart, the BrACh-sensitive part of ACh synthesis was taken as the measure of ChAT activity. The highest activity of ChAT (nmol ACh synthesized g-l.h-l) was found in the region of the sinoatrial node (1775); it decreased in the order: interatrial septum (781) greater than rest of the right atrium (712) greater than left atrium (416) greater than basal part of the right ventricle (366) greater than apical part of the right ventricle (250) greater than inter-ventricular septum (239) greater than basal and apical part of the left ventricle (208 and 205). The results indicate that earlier investigations of the distribution of ChAT in the heart provided a basically correct picture although the contribution of CarAT to the synthesis of ACh measured had not been excluded, and confirm that ChAT is present throughout the heart, including the apical parts of the ventricles, However, the sino-atrio-ventricular gradient of ChAT distribution is steeper when the contribution of CarAT to the synthesis of ACh is excluded.

Experiments involving the acute administration of a central cholinergic substance (L-acetylcarnitine) were performed on 6 healthy subjects to obtain additional evidence that the electrophysiological method developed by Bussel and Pierrot-Deseilligny in 1977 does actually assess recurrent inhibition in man. In all the subjects, the drug further decreased the amplitude of a test H reflex (H') following a conditioning H reflex (H1). The amount of this supplementary inhibition was found to be related to the size of H1 reflex. Experimental evidence is also presented that H' reflex supplementary depression is not contaminated by homonymous Ib effects. It is concluded that the method effectively tests the excitability of Renshaw cells in man.

The effects of L-propionylcarnitine on the recovery of cardiac contractile performance after global ischaemia and reperfusion were studied in isolated perfused rat hearts. The addition of either 5.5 or 11 mmol X litre-1 L-propionylcarnitine significantly improved the recovery of cardiac output, left ventricular pressure, and dP/dt after 90 min of ischaemia and 15 min of reperfusion. Myocardial adenosine triphosphate and creatine phosphate concentrations were significantly higher in the L-propionylcarnitine treated hearts than in controls, but the concentrations of long chain acyl carnitine and coenzyme A were unaffected. The protecting effects of L-propionylcarnitine were compared with those of L-carnitine and L-acetylcarnitine. A 11 mmol X litre-1 dose of L-propionylcarnitine and L-acetylcarnitine significantly improved the recovery of cardiac output after 90 min of ischaemia and 15 min of reperfusion, but L-carnitine did not. L-Propionylcarnitine was the most protective agent. The effects of these derivatives on L-3H-carnitine transport and 14C-palmitate oxidation were also measured. All of these derivatives competitively inhibited L-3H-carnitine transport in isolated cardiac myocytes, but L-propionylcarnitine was the most potent. Carnitine and L-propionylcarnitine stimulated palmitate oxidation in the homogenate, whereas L-acetylcarnitine inhibited it. In myocytes only L-propionylcarnitine affected palmitate oxidation. These data show that L-propionylcarnitine protects the ischaemic myocardium. Its protection is greater than that for L-carnitine or L-acetylcarnitine, and the difference in effectiveness may relate to the rate of transport into the cells and the effects on fatty acid utilisation.

L-acetylcarnitine plays a key role in sperm metabolism and in the whole spermatogenetic process. In the present work, the influence of L-acetylcarnitine, administered i.p. (100 mg/kg body weight), on the recovery processes of mouse spermatogenesis after local acute irradiation with 10 Gy X-rays has been investigated. The effects were monitored 28, 35, 40, 45, 50, 55, and 60 days after irradiation by flow cytometric analysis of cellular DNA content. In the LAC-treated animals, the fraction of tetraploid cells is higher at 28 (p less than 0.05) and 45 days (p less than 0.02). Corresponding with the timing of the stages of murine spermatogenesis, the round spermatid fraction is higher at 45 days (p less than 0.1) and the elongated spermatid fraction is higher at 50 days (p less than 0.1) after irradiation. In addition, the LAC-treated animals show a faster recovery throughout the maturation process, from tetraploid to round and elongated spermatids. These results indicate that the presence of exogenous LAC could enhance the recovery of spermatogonial cells.

The effect of aging and subchronic treatment with acetyl-L-carnitine (50 mg/kg per day) was studied on mitochondrial bioenergetics and cholinergic metabolism in non-synaptic mitochondria and synaptosomes isolated from cerebral cortex, hippocampus and striatum of rats aged 4, 11 and 18 months. Respiratory activity and cytochrome oxidase specific activity were unaffected by aging in non-synaptic mitochondria. In synaptosomes, pyruvate dehydrogenase, choline acetyltransferase and acetylcholinesterase specific activity remained unchanged, but the high-affinity choline uptake decreased in cerebral cortex and striatum of 18-month-old rats. Acetyl-L-carnitine treatment increased the high-affinity choline uptake in cerebral cortex of 18-month-old rats. The treatment caused also an increase in cytochrome oxidase activity in all the three cerebral regions and in choline uptake in the hippocampus, parameters that were not directly affected by aging processes.

The nerve growth factor protein (NGF) has been demonstrated to affect neuronal development and maintenance of the differentiated state in certain neurons of the peripheral and central nervous system (CNS) of mammals. In the CNS, NGF has sparing effects on cholinergic neurons of the rodent basal forebrain (BF) following lesions where it selectively induces choline acetyltransferase (ChAT). NGF also induces ChAT in the areas to which BF provides afferents. In aged rats, there is a reduction in the NGF-binding capacity of sympathetic ganglia. Here, we wish to report that there is a decrease in the NGF-binding capacity of the hippocampus and basal forebrain of aged (26-month-old) rats as compared to 4-month-old controls but no change in NGF binding in cerebellum. In all instances, equilibrium binding dissociation constants did not differ significantly. Treatment of rats with acetyl-L-carnitine, reported to improve cognitive performance of aged rats, ameliorates these age-related deficits.

An increase in motility of ejaculated human spermatozoa was observed after the addition of acetylcaritine or carnitine. Similar results were also obtained in the diluted semen with 25--30% initial motilities. However, indirect evidence suggests that carnitine is converted to acetylcarnitine prior to its stimulatory action. In addition, this stimulation was shown to be the result of no increase in ionic strength nor any change in the levels of ATP.

Three pharmacological agents, L-carnitine, L-acetylcarnitine and gangliosides, were tested for their ability to enhance the regeneration of the rat sciatic nerve following transection and microsurgical repair. The drugs were administered intraperitoneally at the dose of 50 mg/kg/d for 28 and 56 d postoperatively. At the end of treatment, the motor function recovery of the peroneal component of the sciatic nerve was assessed and the regenerated nerves were analysed morphometrically on histological semi-thin sections. Also, the reinnervated extensor digitorum longus (EDL) muscles were studied histochemically using the adenosine-triphosphatase (ATP-ase) technique 56 d after surgery. Motor function assessment at 56 d after nerve repair revealed that L-acetylcarnitine-treated animals recovered a clinical grade significantly higher (p less than 0.05) than the control animals. Twenty-eight days after nerve repair, the number of myelinated fibres was significantly higher (p less than 0.05) in L-acetylcarnitine and ganglioside-treated animals than in control animals. However, 56 d after nerve repair the number of regenerated fibres in all the drug-treated groups was not significantly different from that of the control group. The EDL muscles of the drug-treated animals did not show significant differences from those of control animals with respect to fibre composition and fibre diameter although the L-acetylcarnitine-treated animals exhibited a significantly lower (p less than 0.05) degree of muscle atrophy than did the control animals. The results of the present work seem to indicate that L-acetylcarnitine and to a lesser extent gangliosides exert some favourable effect on the regeneration of the transected sciatic nerve in rats.(ABSTRACT TRUNCATED AT 250 WORDS)

1. Untreated rats of different ages and old rats chronically treated with Acetyl-1-carnitine were subjected to the spatial learning task. 2. For this test, a circular pool filled with milk-opacified water was used. The animals were to reach an escape platform with the aid of visible environmental cues. 3. The experimental results indicated a clear-cut deterioration of the old animal's acquisition ability of a spatial learning task. 4. The long-term treatment (8 months) with Acetyl-1-carnitine was found to antagonize such a deterioration.

Brain autoradiography in adrenalectomized rats injected with 3H-corticosterone 2 hr before sacrifice was used to study the effect of aging and long-term acetyl-l-carnitine treatment on the hippocampal glucocorticoid receptor. Densitometric analysis of silver grains in individual nerve cells of the hippocampus showed that pyramidal neurones of the CA1 field and granular cells of the dentate gyrus are richest in 3H-corticosterone binding sites, whereas pyramidal neurons of the CA3 field have the lowest number of binding sites. There was a significant decline in the number of glucocorticoid receptors within the various hippocampal areas, both as the total number of 3H-corticosterone binding sites and as the number per single pyramidal or granule neuron associated with aging and perhaps due to loss of adrenocorticoid-competent neurons. The dentate gyrus and the CA1 region were mostly affected by the age-dependent decrease in glucocorticoid receptors of the hippocampus. Twenty-eight-month-old rats, treated with acetyl-l-carnitine for 7 months, showed a significantly higher number of 3H-corticosterone binding sites within the various hippocampal regions examined than did age-matched controls. The CA1 and the dentate gyrus were the regions most susceptible to amelioration by acetyl-l-carnitine treatment. These findings suggest a positive effect of acetyl-l-carnitine treatment on age-related changes which occur in the hippocampus.

Amphiphilic compounds such as long-chain acyl carnitines accumulate in ischemic myocardium and potentially contribute to the myocardial damage. To characterize alterations in membrane molecular dynamics produced by palmitoylcarnitine, human erythrocytes were spin-labeled with 5-doxylstearic acid, and membrane fluidity was quantified by measuring the changes in the order parameter derived from ESR spectra. Palmitoylcarnitine induced triphasic alterations in membrane fluidity of human erythrocytes. The membrane fluidity increased for 5 min, then decreased in a concentration-dependent manner. At higher concentrations (100 and 150 microM) of palmitoylcarnitine, membrane fluidity increased again after 30 and 20 min of the incubation, respectively. Addition of 2.8 mM CaCl2 resulted in a significant decrease in membrane fluidity and enhanced the alterations in membrane fluidity caused by palmitoylcarnitine. The results suggest that alterations in molecular dynamics are one mechanism through which long-chain acyl carnitine could play an important role in ischemic injury.

Amino acid and reduced glutathione (GSH) levels in substantia nigra (SN) as well as striatal monoamine levels were measured in acetyl-L-carnitine (ALCar) treated and control Swiss-Webster mice. ALCar, L carnitine, or saline were administered i.p. to mice for 5 days and mice were decapitated 24 hours following the last injection. Substantia nigra and striata were isolated within 2.5 and 3 min., respectively, and frozen immediately on dry ice. A significant dose-dependent increase of nigral GABA was observed following ALCar treatment; GABA levels were also increased by administration of carnitine. Nigral GSH levels were also increased. Striatal levels of dopamine and metabolites were not significantly affected by ALCar or carnitine. These results, suggest that ALCar may be useful in treating symptoms of neuronal dysfunction related to accumulation of metabolic waste.

The effect of methylazoxymethanol (MAM) administration at the 15th gestational day on some behavioural and morpho-functional parameters of rat brain was investigated. The effect of a 13-15-day treatment of acetyl-L-carnitine on the same parameters was also assessed. MAM microencephalic rats showed a significant impairment in water-maze and pole-climbing tests. The histochemical reactivity of the enzyme NADH2-tetrazolium reductase (NADHR) at the level of frontal and occipital cortex, neostriatum and hippocampus was remarkably reduced. Also cholinacetyltransferase (ChAT) immunoreactivity within nerve cell bodies of the pontine tegmentum was decreased in MAM-treated animals. On the contrary, acetylcholinesterase (AChE) reactivity was increased in all the investigated brain areas with the sole exception of the neostriatum. Nissl reactivity was decreased in the cytoplasm of the pyramidal neurons of the frontal cortex and hippocampus, and slightly increased in the cytoplasm of pyramidal neurons of the occipital cortex of MAM microencephalic rats. Acetyl-L-carnitine treatment improved the behaviour of microencephalic rats in water-maze and pole-climbing tests. Moreover the substance stimulated NADHR reactivity in the cerebral cortex and hippocampus as well as ChAT immunoreactivity in the cytoplasm of neurons of the raphe pontine nuclei. Pharmacological treatment reduced AChE reactivity in the cerebral cortex and the hippocampus, and improved the pattern of Nissl reactivity within all brain areas examined.

The effect of acetyl-L-carnitine (ALC) on the spontaneous release of acetylcholine (ACh) in the striatum and hippocampus of freely moving rats was investigated using brain microdialysis coupled with HPLC-electrochemical detection. Systemic administration of ALC, in a dose-dependent manner, stimulated ACh release in both areas, while the D-enantiomer was substantially ineffective. The effect of ALC was strongly Ca2+ dependent and tetrodotoxin (TTX) sensitive. These features of an exocytotic and impulse flow-dependent mechanism suggest that the increase in ACh release is the result of ALC activation of a physiological mechanism in cholinergic neurons.

DL-Aminocarnitine (3-amino-4-trimethylaminobutyric acid) and acetyl-DL-aminocarnitine (3-acetamido-4-trimethylaminobutyric acid) have been synthesized and the interactions of these compounds with carnitine acetyltransferase and carnitine palmitoyltransferase investigated. As anticipated from the low group transfer potential of amides, carnitine acetyltransferase catalyzes the transfer of acetyl groups from CoASAc to aminocarnitine (Km = 3.8 mM) but does not catalyze detectable transfer from acetylaminocarnitine to CoASH. Acetyl-DL-aminocarnitine is, however, a potent competitive inhibitor of carnitine acetyltransferase (Ki = 24 microM) and is bound to carnitine acetyltransferase about 13-fold more tightly than is acetylcarnitine, with which it is isosteric. DL-Aminocarnitine and, to a lesser extent, acetyl-DL-aminocarnitine are also inhibitors of the carnitine palmitoyltransferase activity of detergent-lysed rat liver mitochondria; in the presence of 1 mM L-carnitine, 5 microM aminocarnitine inhibits palmitoyl transfer by 64%. Significant acylation of aminocarnitine by palmitoyl-CoA was not observed. Neither aminocarnitine nor acetylaminocarnitine is significantly catabolized by mice; aminocarnitine is converted to acetylaminocarnitine in vivo. Both compounds are excreted in the urine. Mice given acetylaminocarnitine catabolize [14C]acetyl-L-carnitine and [14C]palmitate to 14CO2 more slowly than do control animals. Mice given acetylaminocarnitine and then starved are found to reversibly accumulate triglycerides in their livers; mice given the inhibitor but not starved do not show this effect.

The behavioral activity of carnitine acetylate derivative, acetyl-l-carnitine has been studied in the male rat. Intraperitoneal (IP) injection of acetyl-l-carnitine was followed by an increase in ambulation and rearing items in the open field behavior. Both the number of conditioned avoidance response (CARs) and the percentage of learners in the acquisition of shuttle-box active avoidance behavior appeared to be increased by IP or intracerebroventricular (ICV) injection of the drug at different doses. Subchronic administration of the drug mimicked the effects found after acute injection. The number of CARs in the extinction of shuttle-box active avoidance behavior appeared to be increased after acute IP or ICV injection, and after subchronic administration of acetyl-l-carnitine. The retention of passive avoidance behavior was facilitated by IP injection of the substance. The behavioral effects of acetyl-l-carnitine may involve central mechanisms, e.g., cholinergic neurotransmission in the brain.

In artificially ventilated beagle dogs a severe hypoglycemic condition was induced by insulin injection, while the posthypoglycemic recovery was induced by glucose treatment at the end of a 20-min period of spontaneous electroencephalographic silence. The motor area of the cerebral cortex was analyzed for glycolytic metabolites, related amino acids, energy mediators, fatty acids, phospholipids and free fatty acids. The effects on the posthypoglycemic recovery of a intracarotid infusion with some agents (i.e. uridine, cytidine, DL-carnitine, DL-acetylcarnitine, papaverine) were tested. Severe hypoglycemia induced an extensive derangement of the brain metabolism, with partial restitution during the posthypoglycemic recovery. During this condition, the intracarotid perfusion with some biological pyrimidines (uridine, cytidine) interfered with the glycolytic and amino acid metabolites, inducing a decrease in glucose, pyruvate and lactate contents, and an increase in succinate, alanine and glutamine cerebral concns. The lipid carriers (DL-carnitine, DL-acetylcarnitine) interfered with the fatty acid degradation inducing a magnification of the decrease in the individual (palmitic acid, oleic acid) and total fatty acids, the vasodilating agent (papaverine) being practically inactive.

The maximal rate (Vmax) of some mitochondrial enzymatic activities related to the energy transduction (citrate synthase, malate dehydrogenase, NADH cytochrome c reductase as total, cytochrome oxidase) and amino acid metabolism (glutamate dehydrogenase) were evaluated in non-synaptic (free) and synaptic mitochondria from rat brain hippocampus. Three types of mitochondria were isolated from rats subjected to single i.m. treatment with L-acetylcarnitine (308 mg X kg-1) or to sub-chronic i.m. treatment with L-acetylcarnitine at three different dose levels (38; 154; 614 mg X kg-1, 5 days a week, for 4 weeks). With respect to the enzymatic pattern of three types of non-synaptic and synaptic mitochondria, in hippocampus a different maximal rate of both total NADH-cytochrome c reductase and cytochrome oxidase was observed, these activities being lower in "synaptic heavy" mitochondrial subfraction rather than that in both "free" and "synaptic light" ones. This confirms that in various types of brain mitochondria a different metabolic machinery exists. Acute treatment with L-acetylcarnitine decreased citrate synthase and glutamate dehydrogenase activities only in mitochondria obtained from synaptosomes. The sub-chronic treatment with L-acetylcarnitine decreased the activity of citrate synthase and total NADH-cytochrome c reductase activities only in the same type of mitochondria, i.e. synaptic mitochondria. Therefore in vivo administration of L-acetylcarnitine mainly affects some specific enzyme activities (suggesting a specific molecular trigger mode of action) of the intrasynaptic mitochondria (suggesting a specific subcellular trigger site of action).

NMDA-sensitive glutamate receptors are involved in the regulation of neuronal plasticity, and contribute to the synaptic mechanisms underlying the learning process. Aging is associated with a reduction in the maximal density of NMDA-sensitive glutamate binding sites in rat hippocampus. This reduction is attenuated after long-term administration with L-acetylcarnitine (10 mg/Kg i.p. once a day for 4 months). These results support a neuroprotective and neurotrophic role for L-acetylcarnitine during aging.

The aim of the study was to evaluate the effects of acetyl-L-carnitine on learning and/or memory processes in laboratory animals. In the water maze test, acetyl-L-carnitine, given intraperitoneally at doses ranging from 0.3 to 100 mg/kg, improved performances in both mice and rats. In the latter the drug also proved active when administered orally in the 3-100 mg/kg dosage range. In the pole climbing test in the rat, acetyl-L-carnitine at doses ranging from 0.03 to 10 mg/kg i.p. increased the conditioned reflex learning rate. In the passive avoidance test in the rat, significant increases in retention were observed after treatment with acetyl-L-carnitine at doses ranging from 1 to 30 mg/kg i.p. In the passive avoidance plus electroconvulsive shock test in the mouse, acetyl-L-carnitine antagonized amnesia at doses ranging from 0.1 to 3 mg/kg i.p.

The synthesis of acetylcholine (ACh) has been measured in homogenates of the sciatic nerve, normal and denervated extensor digitorum longus (e.d.l.) muscles, and central (innervated) and peripheral (non-innervated) parts of the diaphragm of the rat. The synthesis proceeded under conditions accepted as optimal for the activity of choline acetyltransferase (ChAT). In view of the finding that cardiac carnitine acetyltransferase (CarAT) is able to acetylate choline (White & Wu, 1973), the possible contribution of CarAT to the synthesis of ACh in the muscles was investigated by using bromoacetylcholine (BrACh) as an inhibitor of ChAT and bromoacetylcarnitine (BrACar) as an inhibitor of CarAT.2. BrACh at a concentration of 2 mum inhibited the synthesis of ACh in nerve homogenates by 98%, in the homogenates of normal e.d.l. muscles by 53%, in denervated e.d.l. muscles by less than 5%; in the central part of the diaphragm BrACh inhibited ACh synthesis by 65%, and in the peripheral part by 13%. Comparative inefficiency of BrACh in inhibiting the synthesis of ACh in muscle homogenates was not due to its inactivation; the inhibitory effect of BrACh on the neural synthesis of ACh was preserved in the presence of muscle homogenates.3. BrACar at a concentration of 20 mum inhibited the synthesis of ACh in homogenates of the nerve by 18%, in those of normal e.d.l. muscles by 67%, in denervated e.d.l. muscles by 90%; in the central part of the diaphragm it inhibited the synthesis by 29%, and in the peripheral part by 76%.4. The inhibitory effects of BrACh and BrACar on the synthesis of ACh in muscle homogenates were roughly additive.5. Within 2 days of transection of the sciatic nerve, the BrACh-sensitive synthesis of ACh in the e.d.l. muscle diminished by 28%, whereas the BrACh-insensitive synthesis of ACh did not change. At 4 days after denervation, the rate of BrACh-sensitive synthesis decreased to 3% of control values.6. The results indicate that at least two enzymes are responsible for the synthesis of ACh in muscle homogenates is probably catalysed by CarAT. associated with intramuscular nerves, and probably corresponds to ChAT. The other enzyme is comparatively insensitive to BrACh, is sensitive to BrACar, and is probably localized in the muscle fibres. The BrACh-insensitive and BrACar-sensitive synthesis of ACh in muscle homogenates is probably catalysed by CarAT.7. Under the conditions used in the present experiments, CarAT was responsible for approximately one half of the synthesis of ACh in the homogenates of innervated e.d.l. muscles and for all ACh synthesized after denervation.8. The results provide an explanation for earlier findings of residual ACh synthesis in homogenates of denervated muscles, without resort to the idea that ChAT is localized in muscle fibres. It is proposed that CarAT catalyses some synthesis of ACh also in intact muscles, and that it is responsible for the synthesis of ACh observed during incubation of whole denervated muscles. It is not clear what is the physiological function of the synthesis of ACh catalysed by CarAT.9. Measurements of the BrACh-sensitive portion of the total ACh-synthesizing capacity of muscle homogenates provide a suitable procedure for obtaining information about the activity of neural ChAT in the muscles.

L-acetyl carnitine administration increases the amplitude of the steady-state visual evoked potentials (VEPs) obtained with sinusoidal luminance stimulation at 8.4 Hz. This effect was studied with three different doses of L-acetyl carnitine (50, 25, 5 mg/kg) and was compared with the results of injection of similar doses of L-carnitine. The effect is most evident when 50 mg/kg L-acetyl carnitine are administered and has two different time peaks. The second peak of activity showed a dose dependency when 25 mg/kg and 5 mg/kg doses were tested. The effects of L-acetyl carnitine could be depressed by the simultaneous administration of atropine sulfate.

Rat cardiac microsomes treated with NADPH generated a chemiluminescence, detected by the chemilumigenic probe lucigenin. The chemiluminescent signal, which is an index of lipid peroxidation, was found to be inhibited by acetylcarnitine in a dose-dependent way. Superoxide dismutase (SOD) and inhibitors of arachidonate metabolism were also effective in preventing light emission. The combined action of acetylcarnitine plus SOD and acetylcarnitine plus indomethacin suggested a possible common target for the compounds. When tested on superoxide production from isolated human neutrophils detected both by luminol-amplified chemiluminescence and cytochrome C reduction, acetylcarnitine did not show any inhibitory effect. The results of these experiments demonstrate the antioxidant properties of acetylcarnitine, even if they cannot clarify the specific target of the drug action.

The ageing rat hippocampus undergoes ultrastructural changes, including the loss of axosomatic synapses of the granule cells. These synapses are supposed to take part in a feed-back regulation of granule cell activity, as they are inhibitory terminals of interneurons which receive afferences from the granules themselves via the giant synapses formed by the collaterals of the mossy fibres. In the present study the authors performed a quantitative analysis of axosomatic and giant synapses at the ultrastructural level in aged rats as compared with young animals. In aged rats a decrease both in axosomatic synapses and in giant synapsis vesicles was found, giving further support to the postulated feed-back mechanism. Both young and old rats were treated with L-acetyl-carnitine, a drug which favours the synthesis of acetylcholine, the main neurotransmitter deficient in old age. In aged rats the drug restored a normal number of both axosomatic synapses and giant bouton vesicles. The authors hypothesize that the drug, by a cholinergic-type mechanism, restores the excitatory afferences to the granule whose axon would thus form normal giant boutons with the interneuron, reestablishing the feed-back regulation. In young rats the drug induced a decrease only of the axosomatic synapses, suggesting that an "over-excitation" might impair the information to the local-circuit neurons, thus interrupting feed-back control.

The effect of amphiphilic lipid intermediates on the Na+-stimulatable activity of the Na+-K+-ATPase of sarcolemma from adult canine cardiac myocytes was studied. Sarcolemma (mean Na+-stimulatable ATPase activity of 73 mumol.mg sarcolemmal protein-1.h-1) was preincubated (37 degrees C for 10 min at pH 7.2) or rapidly mixed at 0 degrees C with amphiphilic lipid intermediates prior to dilution and assay of enzyme activity. Their effects were dependent on temperature, initial concentration, and the ratio of bound amphiphile to sarcolemmal protein. In particular, pretreatment of freshly prepared sarcolemma at 0 degrees C with arachidonyl CoA (up to 0.25 mM) caused 110% stimulation above control activity; palmitoyl CoA or palmitoyl carnitine under the same conditions caused no significant effect. Despite strong binding to the sarcolemmal vesicles, palmitoyl carnitine (up to 0.4 mM or 5 mumol/mg protein) and palmitoyl CoA (0.1 mM or 1.0 mumol of membrane-bound palmitoyl CoA/mg protein) were ineffective even with preincubation. Palmitoyl CoA was inhibitory above this level. Preincubation (22 degrees C for 10 min) with lysophosphatidylcholine only produced inhibition (40% at 0.75 mM). Thus fatty acyl thioesters of CoA and lysophosphatidyl choline but not palmitoyl carnitine perturb sarcolemmal Na+-K+-ATPase activity.

Hemiacetylcarnitinium (2S,6R:2R,65)-6-carboxymethyl-2-hydroxy-2,4,4- trimethylmorpholinium) chloride is a relatively potent competitive inhibitor (Ki = 0.89 mM) of pigeon breast carnitine acetyltransferase (CAT) and of the crude rat liver CAT (Ki = 4.72 mM) but is neither an inhibitor nor an effective substrate for purified rat liver carnitine palmitoyltransferase (CPT). It does not inhibit state 3 oxygen consumption in isolated hepatic mitochondria using palmitoyl-CoA or palmitoylcarnitine as substrates. This compound is a reaction intermediate analogue of the proposed tetrahedral intermediate for acetyl transfer between acetylcarnitine and CoASH. Because the hemiketal carbon is chiral, a suggestion is made that one of the enantiomers has the same relative configuration as the proposed tetrahedral intermediate.

Age-related changes of the hippocampal mossy fibre system were studied in rats of different ages (4, 12, and 24 months) using the Timm's histochemical technique for the detection of tissue stores of zinc and heavy transitional metals. Quantitative image analysis and microdensitometry techniques were applied for the evaluation of the changes themselves. The area occupied by mossy fibres and the intensity of Timm's staining in field CA3 of hippocampus were greater in adult rats than in young and old animals. These findings are consistent with previous behavioural data indicating that young and old rats exhibited a reduced passive avoidance learning with respect to adult animals. The above techniques were also used in a second session of this study designed to investigate the effect of a 6-months treatment with acetyl-L-carnitine (ALC) on the hippocampal mossy fibre system. ALC was found to increase significantly the area occupied by mossy fibres and the intensity of Timm's staining in the hippocampus of old rats. These findings are in agreement with and support from an anatomical point of view, the data indicating that long-term treatment with ALC improves passive avoidance performance.

The efflux of L-[3H]carnitine was studied in cells from an established cell line from human heart (Girardi human heart cells, CCL 27). The cells were loaded with 4 mumol/l L-[3H]carnitine for 1 or 24 h, and the efflux of radioactivity into the medium was measured. The amount of intracellular L-[3H]-carnitine retained was expressed as a function of time. The results were fitted to an exponential equation, from which efflux rate constants were computed. Increasing the extracellular concentration of butyrobetaine, L-carnitine, D-carnitine, betaine, DL-norcarnitine or 3-dimethylamino-2-hydroxypropionic acid each increased the observed efflux. This is most likely due to accelerated exchange diffusion. The substrate specificity of this accelerated exchange diffusion is different from what previously has been found in competitive uptake studies of L-carnitine. L-Carnitine was preferentially released to L-acetylcarnitine, and blocking the sulfhydryl groups with 5,5-dithiobis-(2-nitrobenzoic acid) increased the efflux.

Using the two-stage coronary ligation method, first described by Harris, (1950), anti-arrhythmic effects (AAE) of (-)-carnitine chloride (LCC) and acetyl (-)-carnitine chloride (ALCC) were studied in conscious unrestrained dogs in comparison with those of disopyramide (D). Two-stage ligation of the coronary artery resulted in a significant decrease in the myocardial free carnitine content. Intravenous administration of LCC (300 mg kg-1) and D (5 mg kg-1) suppressed the ventricular arrhythmia induced by coronary ligation after 24 hours. ALCC (300 mg kg-1) was found to be less potent. An improvement of the mitochondrial function (respiratory control index (RCI) and oxidative phosphorylation rate (OPR) ) was noted with LCC and ALCC and there was a linear correlation between AAE expressed as reduction of arrhythmic ratio and improvement in the OPR, whereas there was no improvement in mitochondrial function after D. Plasma carnitine concentration was increased after administration of LCC, attaining the value of around 8 mM at 10 min after 300 mg kg-1. At 60 min, the plasma carnitine concentration was still about half as high as at 10 min. After ALCC, both acetyl carnitine and free carnitine were found in the plasma. The concentration of the former was decreased after attaining a peak value of around 0.2 mM at 10 min, while the plasma concentration of free carnitine was gradually increased. The anti-arrhythmic effects of LCC and ALCC were ascribed to the improvement of mitochondrial oxidative phosphorylation, while effects other than the improvement of the mitochondrial activity were suggested as mechanisms of anti-arrhythmic effects of D.

The acute effects of intravenously administered L-acetylcarnitine (LAC) were evaluated with single-photon emission computed tomography (SPECT) and 99mTc-hexamethylpropyleneamine oxime in 30 demented patients (21 with a clinical diagnosis of Alzheimer's dementia and 9 with mixed-type dementia). Two SPECT scans were performed: in basal conditions, and 30 min after the administration of 500, 1,000, 1,500, or 2,000 mg LAC intravenously. Tracer activity ratios were determined in 10 pairs of cerebellar, cortical and subcortical regions. After administration of the lowest dose of LAC, no changes from the basal values were observed in any of the regions examined. The higher doses of the drug significantly elevated the tracer activity in cortical regions, particularly in the parietal lobe, which showed an impaired regional cerebral blood flow in the basal study. These effects of LAC and their relation with the cholinomimetic properties of the drug are discussed.
