452. Simvastatin Promotes Hematoma Absorption and Reduces Hydrocephalus Following Intraventricular Hemorrhage in Part by Upregulating CD36.

Abstract

We previously found that hematoma worsens hydrocephalus after intraventricular hemorrhage (IVH) via increasing iron deposition and aggravating ependymal cilia injury; therefore, promoting hematoma absorption may be a promising strategy for IVH. Recently, some investigations imply that simvastatin has the ability of accelerating hematoma absorption. Thus, this study was designed to examine the efficacy of simvastatin for IVH in rats. Intracerebral hemorrhage with ventricular extension was induced in adult male Sprague-Dawley rats after autologous blood injection. Simvastatin or vehicle was administered orally at 1 day after IVH and then daily for 1 week. MRI studies were performed to measure the volumes of intracranial hematoma and lateral ventricle at days 1, 3, 7, 14, and 28 after IVH. Motor and neurocognitive functions were assessed at days 1 to 7 and 23 to 28, respectively. Iron deposition, iron-related protein expression, ependymal damage, and histology were detected at day 28. Expression of CD36 scavenger receptor (facilitating phagocytosis) was examined at day 3 after IVH using western blotting and immunofluorescence. Simvastatin significantly increased hematoma absorption ratio, reduced ventricular volume, and attenuated neurological dysfunction post-IVH. In addition, less iron accumulation and more cilia survival was observed in the simvastatin group when compared with the control. What's more, higher expression of CD36 was detected around the hematoma after simvastatin administration. Simvastatin significantly enhanced brain hematoma absorption, alleviated hydrocephalus, and improved neurological recovery after experimental IVH, which may in part by upregulating CD36 expression. Abstract

The cannabinoid receptor-2 (CB2R) was initially thought to be the "peripheral cannabinoid receptor." Recent studies, however, have documented CB2R expression in the brain in both glial and neuronal cells, and increasing evidence suggests an important role for CB2R in the central nervous system inflammatory response. Intracerebral hemorrhage (ICH), which occurs when a diseased cerebral vessel ruptures, accounts for 10-15% of all strokes. Although surgical techniques have significantly advanced in the past two decades, ICH continues to have a high mortality rate. The aim of this study was to investigate the therapeutic effects of CB2R stimulation in acute phase after experimental ICH in rats and its related mechanisms. Data showed that stimulation of CB2R using a selective agonist, JWH133, ameliorated brain edema, brain damage, and neuron death and improved neurobehavioral outcomes in acute phase after ICH. The neuroprotective effects were prevented by SR144528, a selective CB2R inhibitor. Additionally, JWH133 suppressed neuroinflammation and upregulated the expression of microglial M2-associated marker in both gene and protein level. Furthermore, the expression of phosphorylated cAMP-dependent protein kinase (pPKA) and its downstream effector, cAMP-response element binding protein (CREB), were facilitated. Knockdown of CREB significantly inversed the increase of M2 polarization in microglia, indicating that the JWH133-mediated anti-inflammatory effects are closely associated with PKA/CREB signaling pathway. These findings demonstrated that CB2R stimulation significantly protected the brain damage and suppressed neuroinflammation by promoting the acquisition of microglial M2 phenotype in acute stage after ICH. In this study, we investigated the impact of smoking, a major risk factor of lung cancer, on DNA methylation by comparing the genome-wide DNA methylation patterns between lung adenocarcinoma samples from six smokers and six nonsmokers. We identified that smoking-induced DNA methylations were enriched in the calcium signaling and neuroactive ligand receptor signaling pathways, which are closely related to smoking-induced lung cancers. Interestingly, we discovered that two genes in the mitogen-activated protein kinase signaling pathway (RPS6KA3 and ARAF) were hypomethylated in smokers but not in nonsmokers. In addition, we found that the smoking-induced lung cancer-specific DNA methylations were mostly enriched in nuclear activities, including regulation of gene expression and chromatin remodeling. Moreover, the smoking-induced hypermethylation could only be seen in lung adenocarcinoma tissue but not in adjacent normal lung tissue. We also used differentially methylated DNA loci to construct a diagnostic model to distinguish smoking-associated lung cancer from nonsmoking lung cancer with a sensitivity of 88.9% and specificity of 83.2%. Abstract

The transient dark activation of opsin by 11-cis-13-desmethylretinal and its dihydro analogue was studied to determine the mechanism of dark activity of the 13-desmethylretinal/rod opsin complex. Three binding domains comprise the opsin cavity incorporating the retinals, namely, site I for the cyclohexene, site II for the polyene chain, and site III for protonated Schiff base (PSB). The cyclohexene moiety occupies site I as required for opsin activation, while 11-cis-retinals activate the protein during the binding process prior to pigment formation. The protein is inactivated by protonation of the Schiff base. The polyene chain then assumes a shape that results in a transient Meta-II-like conformation in opsin.

474. Accelerated angiogenesis by continuous medium flow with vascular endothelial growth factor inside tissue-engineered trachea

Abstract

Objective: To test the effects of a continuous medium flow inside DegraPol® scaffolds on the reepithelialization and revascularization processes of a tissue-engineered trachea prosthesis. Methods: In this proof-of-principle study a continuous medium flow was maintained within a tubular DegraPol® scaffold by an inserted porous catheter connected to a pump system. The impact of the intra-scaffold medium flow on the survival of a tracheal epithelial sheet wrapped around and on chondrocyte delivery to the DegraPol® scaffold was studied. In the chick embryo, chorioallantoic membrane (CAM) model angiogenesis within the biomaterial was investigated. Results: Scanning electronic microscopy (SEM) images showed an intact epithelial layer after a 2-week support by continuous medium flow underneath. On histology, three-dimensional cell growth was detected in the continuous delivery group. The CAM assay showed that angiogenesis was enhanced within the DegraPol® scaffolds when vascular endothelial growth factor/vascular permeability factor (VEGF/VPF) was added to the perfusate. Conclusions: Taken together, these results demonstrated that the built-in perfusion system within DegraPol® scaffolds was able to maintain an intact tracheal epithelial layer, allowed a continuous delivery of cells, and kept an efficient VEGF/VPF expression level which accelerated angiogenic response in the CAM assay. Abstract

Objective There are two types of endothelial progenitor cell (EPC) in bone marrow (BM) and circulation, BM-EPC and Circulation late endothelial progenitor cell (CL-EPC). Diabetes impairs the function of EPC, but it is not clear whether transplantation of EPC can rescue ischemic myocardium in diabetes. In this study, we compared the function of BM-EPC and CL-EPC in vitro. Then we administered BM-EPC and CL-EPC intramyocardially and compared their contribution to vasculogenesis in diabetic rabbits. Methods BM-EPC and CL-EPC from healthy and diabetic rabbits were isolated and subjected to in vitro proliferation, tube-forming, angiogenic cytokine assays. Exogenous BM-EPC and CL-EPC were analysed for therapeutic efficacy in an acute ischemia model of diabetic rabbits. LVfunction was assessed using echocardiography. The capillary density and fibrosis area were evaluated. mRNA expression of VEGF and bFGF was analysed using relative realtime quantitive PCR. Results Proliferation, tube-forming, secretion of VEGF and bFGF of diabetic EPC were significantly reduced compared with healthy EPC. In diabetic rabbits, healthy BM-EPC transplantation could increase capillary density and improve cardiac function, decrease fibrosis area compared with CL-EPC and the control group. Real time PCR indicated that mRNA expression of VEGF and bFGF were augmented more in the healthy BM-EPC group than those in the control and CL-EPC groups. Conclusions These findings suggest that diabetes impairs the function of EPC. Transplantation of healthy BM-EPC may rescue the ischemic myocardium by neovasculogenesis and paracrine effect in diabetic rabbits. However, autologous transplantation of CL-EPC could not enhance cardiac function.

Abstract

Eleutherobin is a marine natural product discovered in 1994. It serves as one of the most potent anti-cancer agents mimicking Taxol. The proposed research is focused on improving the total synthesis of eleutherobin, and based on that developing eleutherobin analogs for biological studies. A modified eleutherobin synthesis is being studied. Compared to the initial Synthesis, the new synthesis is designed to improve the stereoselectivity of key steps and shorten the sequence.
