Abstract

Platelet-derived growth factor receptor (PDGFR) signaling plays an important role in the biology of malignant gliomas. To investigate mechanisms modulating PDGFR signaling in gliomagenesis, we employed a Drosophila glioma model and genetic screen to identify genes interacting with Pvr, the fly homolog of PDGFRs. Glial expression of constitutively activated Pvr (lambda Pvr) led to glial over migration and lethality at late larval stage. Among 3316 dsRNA strains crossed against the tester strain, 128 genes shifted lethality to pupal stage, including tetraspanin 2A (tsp2A). In a second step knockdown of all Drosophila tetraspanins was investigated. Of all tetraspanin dsRNA strains only knockdown of tsp2A partially rescued the Pvr-induced phenotype. Human CD9 (TSPAN29/MRP-1), a close homolog of tsp2A, was found to be expressed in glioma cell lines A172 and U343MG as well as in the majority of glioblastoma samples (16/22, 73 %). Furthermore, in situ proximity ligation assay revealed close association of CD9 with PDGFR alpha and beta. In U343MG cells, knockdown of CD9 blocked PDGF-BB stimulated migration. In conclusion, modulation of PDGFR signaling by CD9 is evolutionarily conserved from Drosophila glia to human glioma and plays a role in glia migration.

Abstract

IDH wild type (IDHwt) anaplastic astrocytomas WHO grade III (AA III) are associated with poor outcome. To address the possibilities of molecular subsets among astrocytoma or of diagnostic reclassification, we analyzed a series of 160 adult IDHwt tumors comprising 120 AA III and 40 diffuse astrocytomas WHO grade II (A II) for molecular hallmark alterations and established methylation and copy number profiles. Based on molecular profiles and hallmark alterations the tumors could be grouped into four major sets. 124/160 (78 %) tumors were diagnosed as the molecular equivalent of conventional glioblastoma (GBM), and 15/160 (9 %) as GBM-H3F3A mutated (GBM-H3). 13/160 (8 %) exhibited a distinct methylation profile that was most similar to GBM-H3-K27, however, lacked the H3F3A mutation. This group was enriched for tumors of infratentorial and midline localization and showed a trend towards a more favorable prognosis. All but one of the 120 IDHwt AA III could be assigned to these three groups. 7 tumors recruited from the 40 A II, comprised a variety of molecular signatures and all but one were reclassified into distinct WHO entities of lower grades. Interestingly, TERT mutations were exclusively restricted to the molecular GBM (78 %) and associated with poor clinical outcome. However, the GBM-H3 group lacking TERT mutations appeared to fare even worse. Our data demonstrate that most of the tumors diagnosed as IDHwt astrocytomas can be allocated to other tumor entities on a molecular basis. The diagnosis of IDHwt diffuse astrocytoma or anaplastic astrocytoma should be used with caution.

Abstract

International audience; We investigated the formation of extended defects in CaFeO2.5, predominantly appearing as antiphase boundaries (APBs), as a function of the synthesis method and temperature. While CaFeO2.5 is known to adopt an ordered oxygen defect structure showing long range order of the (FeO4)∞ chains in its bulk form, interestingly, we demonstrated that the length of these (FeO4)∞ chains can be considerably scaled down to few nanometers by adopting a modified sol-gel method (low temperature synthesis) while the grain size of the resulting nano-phase CaFeO2.5 is around 50 nm. We discuss the synthesis dependent modulation of the length of APBs, characterized by X-ray diffraction and high resolution TEM, to be at the origin of an amplified switching dynamics of the (FeO4)∞ chains. This can accordingly explain the reduction of the onset temperature for oxygen diffusion to set in from 450 °C for bulk-CaFeO2.5 to 320 °C for nano-CaFeO2.5, as determined by 18O/16O oxygen isotope exchange reactions

Abstract

Gliosarcoma is a rare variant of glioblastoma characterized by a biphasic pattern of glial and mesenchymal differentiation. It is unclear whether mesenchymal differentiation in gliosarcomas is because of extensive genomic instability and/or to a mechanism similar to epithelial–mesenchymal transition (EMT). In the present study, we assessed 40 gliosarcomas for immunoreactivity of Slug, Twist, matrix metalloproteinase‐2 (MMP‐2) and matrix metalloproteinase‐9 (MMP‐9), which are involved in EMT in epithelial tumors. Nuclear Slug expression was observed in >50% of neoplastic cells in mesenchymal tumor areas of 33 (83%) gliosarcomas, but not in glial areas (P 50% of neoplastic cells in mesenchymal tumor areas of 35 (88%) gliosarcomas, but glial tumor areas were largely negative except in four cases (P 10% neoplastic cells. Thus, expression of Slug, Twist, MMP‐2 and MMP‐9 was characteristic of mesenchymal tumor areas of gliosarcomas, suggesting that mechanisms involved in the EMT in epithelial neoplasms may play roles in mesenchymal differentiation in gliosarcomas.

Abstract

International audience; The structure of oxygen-intercalated La(2)CuO(4.07) has been investigated at 20 and 300 K by neutron diffraction on an electrochemically oxidized single crystal. At 20 K, reconstruction of the nuclear density by maximum entropy method shows strong displacements of the apical oxygen atoms towards [100] with respect to the F-centred unit cell, whilst displacements towards [110] and [100] were both found to be present at ambient temperature. Combining structural studies with first-principles lattice dynamical calculations, we interpret the displacements of the apical oxygen atoms to be at least partially of dynamic origin already at ambient temperature. Strong displacements of the apical oxygen atoms of stoichiometric and oxygen-doped La(2)CuO(4+delta) and corresponding associated lattice instabilities, i.e. low-energy phonon modes, are considered as a general prerequisite of low-temperature oxygen diffusion mechanisms. Lattice dynamical calculations on La(2)CuO(4+delta) suggest that the oxygen species diffusing at low temperature are not the interstitial but, more prominently, the apical oxygen atoms. The presence of interstitial oxygen atoms is, however, important to amplify via specific, low-energy phonon modes, a dynamic exchange mechanism between apical and vacant interstitial oxygen sites, thus allowing a dynamically triggered, shallow potential oxygen diffusion pathway. Abstract

Atypical teratoid/rhabdoid tumours (AT/RT) are malignant brain tumours. Unlike most other human brain tumours, AT/RT are characterized by inactivation of one single gene, SMARCB1. SMARCB1 is a member of the evolutionarily conserved SWI/SNF chromatin remodelling complex, which has an important role in the control of cell differentiation and proliferation. Little is known, however, about the pathways involved in the oncogenic effects of SMARCB1 inactivation, which might also represent targets for treatment. Here we report a comprehensive genetic screen in the fruit fly that revealed several genes not yet associated with loss of snr1, the Drosophila homologue of SMARCB1. We confirm the functional role of identified genes (including merlin, kibra and expanded, known to regulate hippo signalling pathway activity) in human rhabdoid tumour cell lines and AT/RT tumour samples. These results demonstrate that fly models can be employed for the identification of clinically relevant pathways in human cancer.

Abstract

Meningiomas are the second most common brain tumors in adults, and meningiomas exhibit a tendency to invade adjacent structures. Compared with high-grade gliomas, little is known about the molecular changes that potentially underlie the invasive behavior of meningiomas. In this study, we examined the expression and function of the membrane alanyl-aminopeptidase [mAAP, aminopeptidase N (APN), CD13, EC3.4.11.2] zinc-dependent ectopeptidase in meningiomas and meningioma cell lines, based on its prior association with tumor invasion in colorectal and renal carcinomas. We found a significant reduction of APNmRNA and protein expression, as well as enzymatic activity, in high-grade meningiomas. While meningioma tumor cell proliferation was not affected by either pharmacologic APN inhibition or siRNA-mediated APN silencing, APN pharmacologic and siRNA knockdown significantly reduced meningioma cell invasion in vitro. Next, we employed pathway-specific cDNA microarray analyses to identify extracellular matrix and adhesion molecules regulated by APN, and found that APN-siRNA knockdown substantially increased the expression of secreted protein, acidic and rich in cysteine (SPARC)/osteonectin. Finally, we demonstrated that SPARC, which has been previously associated with meningioma invasiveness, was increased in aggressive meningiomas. Collectively, these results suggest that APN expression and enzymatic function is reduced in aggressive meningiomas, and that alterations in the balance between APN and SPARC might favor meningioma invasion.

Abstract

The value of combined L-(methyl-[11C])methionine positron-emitting tomography (MET-PET) and magnetic resonance imaging (MRI) with regard to tumor extent, entity prediction, and therapy effects in clinical routine in patients with suspicion of a brain tumor was investigated. In n = 65 patients with histologically verified brain lesions n = 70MET-PET andMRI (T1-weighted gadolinium-enhanced [T1w-Gd] and fluid-attenuated inversion recovery or T2-weighted [FLAIR/T2w]) examinations were performed. The computer software "visualization and analysis framework volume rendering engine (Voreen)" was used for analysis of extent and intersection of tumor compartments. Binary logistic regression models were developed to differentiate between World Health Organization (WHO) tumor types/grades. Tumor sizes as defined by thresholding based on tumor-to-background ratios were significantly different as determined byMET-PET (21.6±36.8 cm3), T1w-Gd-MRI (3.9±7.8 cm3), and FLAIR/T2-MRI (64.8±60.4 cm3; P < .001). The METPET visualized tumor activity where MRI parameters were negative: PET positive tumor volume without Gd enhancement was 19.8± 35.0 cm3 and without changes in FLAIR/T2 10.3±25.7 cm3. FLAIR/T2-MRI visualized greatest tumor extent with differences to METPET being greater in posttherapy (64.6±62.7 cm3) than in newly diagnosed patients (20.5±52.6 cm3). The binary logistic regression model differentiated between WHO tumor types (fibrillary astrocytoma II n=10 fromother gliomas n=16) with an accuracy of 80.8% in patients at primary diagnosis. Combined PET and MRI improve the evaluation of tumor activity, extent, type/grade prediction, and therapy-induced changes in patients with glioma and serve information highly relevant for diagnosis and management. Abstract

High quality stoichiometric brownmillerite-type oxide single crystals have been successfully grown by the floating zone method using a mirror furnace. We report here on the growth conditions and structural characterization of two model compounds: Ca2Fe2O5 and Sr2ScGaO5. Both show oxygen deficiency with respect to the average perovskite structure, and are promising candidates for oxygen ion conductivity at moderate temperatures. While Sr2ScGaO5 single crystals were obtained in the cubic oxygen-deficient perovskite structure, Ca2Fe2O5 crystallizes in the brownmillerite framework. Having no cubic parent high temperature counterpart, Ca2Fe2O5 crystals were found to be not twinned. We report on structural characterization of the as-grown single crystals by neutron and X-ray diffraction, as well as scanning electron microscopy (SEM) coupled with EDX (Energy Dispersive X-Ray Spectroscopy) analysis and isotope exchange experiments.
