The singularity and statics performance of a 6-DOF mobile redundant parallel mechanism and its real-time optimization design are studied. The statics Jacobian matrix of this parallel mechanism is established,and the singularity and statics performance are analyzed. The statics transfer performance of parallel mechanism is improved by adjusting the leg position of the parallel mechanism. The performance spectrum based on the Jacobian matrix is drawn,the transfer performance of the statics force and moment of the parallel mechanism in different positions is observed intuitively. By above analysis shows that the redundant parallel mechanism can eliminate the singularity,while adjusting the leg position also greatly improves the static transfer performance of parallel mechanism.

Coumarin was first discovered in Tonka bean and then widely in other plants. Coumarin has an anticoagulant effect, and its derivative, warfarin, is a vitamin K analogue that inhibits the synthesis of clotting factors and is more widely used in the clinical treatment of endovascular embolism. At present, many artificial chemical synthesis methods can be used to modify the structure of coumarin to develop many effective drugs with low toxicity. In this study, we investigated the effects of six coumarin derivatives on the platelet aggregation induced by adenosine diphosphate (ADP). We found that the six coumarin derivatives inhibited the active form of GPIIb/IIIa on platelets and hence inhibit platelet aggregation. We found that 7-hydroxy-3-phenyl 4H-chromen-4-one (7-hydroxyflavone) had the most severe effect. In addition, we further analyzed the downstream signal transduction of the ADP receptor, including the release of calcium ions and the regulation of cAMP, which were inhibited by the six coumarin derivatives selected in this study. These results suggest that coumarin derivatives inhibit coagulation by inhibiting the synthesis of coagulation factors and they may also inhibit platelet aggregation.

The nonlinear vibration behaviors of stiffened cylindrical shells under electromagnetic excitations, transverse excitations, and in-plane excitations are studied for the first time in this paper. Given the first-order shear deformation theory and Hamilton principle, the nonlinear partial differential governing equations of motion are derived with considering the von Karman geometric nonlinearity. By employing the Galerkin discretization procedure, the partial differential equations are diverted to a set of coupled nonlinear ordinary differential equations of motion. Based on the case of 1 : 2 internal resonance and principal resonance-1/2 subharmonic parametric resonance, the multiscale method of perturbation analysis is employed to precisely acquire the four-dimensional nonlinear averaged equations. From the resonant response analysis and nonlinear dynamic simulation, we discovered that the unstable regions of stiffened cylindrical shells can be narrowed by decreasing the external excitation or increasing the magnetic intensity, and their working frequency range can be expanded by reducing the in-plane excitation. Moreover, the different nonlinear dynamic responses of the stiffened cylindrical shell are acquired by controlling stiffener number, stiffener size, and aspect ratio. The presented approach in this paper can provide an efficient analytical framework for nonlinear dynamics theories of stiffened cylindrical shells and will shed light on complex structure design in vibration test engineering.

The graphical progression method can obtain grand coordinated schemes with minimal computational complexity. However, there is no standardized solution for this method, and only a few related studies have been found thus far. Therefore, based on the in-depth discussion of the graphical optimization theory mechanism, a process-oriented and high-efficiency graphical method for symmetrical bidirectional corridor progression is proposed in this study. A two-round rotation transformation optimization process of the progression trajectory characteristic lines (PTC lines) is innovatively proposed. By establishing the updated judgment criteria for coordinated mode, the first round of PTC line rotation transformation realizes the optimization of coordinated modes and initial offsets. Giving the conditions for stopping rotation transformation and determining rotation points, rotation directions, and rotation angles, the second round of PTC line rotation transformation achieves the final optimization of the common signal cycle and offsets. Severe acute respiratory syndrome-associated coronavirus (SARS-CoV) emerged as the causal agent of an endemic atypical pneumonia, infecting thousands of people worldwide. Although a number of promising potential vaccines and therapeutic agents for SARS-CoV have been described, no effective antiviral drug against SARS-CoV is currently available. The intricate, sequential nature of the viral entry process provides multiple valid targets for drug development. Here, we describe a rapid and safe cell-based high-throughput screening system, dual envelope pseudovirion (DEP) assay, for specifically screening inhibitors of viral entry. The assay system employs a novel dual envelope strategy, using lentiviral pseudovirions as targets whose entry is driven by the SARS-CoV Spike glycoprotein. A second, unrelated viral envelope is used as an internal control to reduce the number of false positives. As an example of the power of this assay a class of inhibitors is reported with the potential to inhibit SARS-CoV at two steps of the replication cycle, viral entry and particle assembly. This assay system can be easily adapted to screen entry inhibitors against other viruses with the careful selection of matching partner virus envelopes.

The lung is a radiosensitive organ, which imposes limits on the therapeutic dose in thoracic radiotherapy. Irradiated alveolar epithelial cells promote radiation-related pneumonitis and fibrosis. However, the role of lung stem cells (LSCs) in the development of radiation-induced lung injury is still unclear. In this study, we found that both LSCs and LSC-derived type II alveolar epithelial cells (AECII) can repair radiation-induced DNA double-strand breaks, but the irradiated LSCs underwent growth arrest and cell differentiation faster than the irradiated AECII cells. Moreover, radiation drove LSCs to fibrosis as shown with the elevated levels of markers for epithelial-mesenchymal transition and myofibroblast (α-smooth muscle actin (α-SMA)) differentiation in in vitro and ex vivo studies. Increased gene expressions of connective tissue growth factor and α-SMA were found in both irradiated LSCs and alveolar cells, suggesting that radiation could induce the fibrogenic differentiation of LSCs. Irradiated LSCs showed an increase in the expression of surfactant protein C (SP-C), the AECII cell marker, and α-SMA, and irradiated AECII cells expressed SP-C and α-SMA. These results indicated that radiation induced LSCs to differentiate into myofibroblasts and AECII cells; then, AECII cells differentiated further into either myofibroblasts or type I alveolar epithelial cells (AECI). In conclusion, our results revealed that LSCs are sensitive to radiation-induced cell damage and may be involved in radiation-induced lung fibrosis.

Abstract Background Total knee arthroplasty (TKA) is gradually regarded as an effective choice for end-stage osteoarthritis or rheumatic arthritis. In the past, the management of tranexamic acid (TXA) using intravenous injection or topical application has been extensively researched. However, several studies have reported that oral TXA has an effect on blood loss. Therefore, a meta-analysis should be performed to determine whether oral TXA helps to prevent blood loss. Methods Randomized controlled trials or retrospective cohort studies about relevant studies were searched in PubMed (1996–April 2017), Embase (1980–April 2017), and the Cochrane Library (CENTRAL, April 2017). Six studies that compared oral TXA to non-TXA were included in our meta-analysis. Meta-analyses (PRISMA) guidelines, the Cochrane Handbook, and the Jadad scale were used to evaluate the included studies and the results to ensure that the meta-analysis was viable. Results In accordance with inclusion and exclusion, six studies with 2553 patients (oral TXA = 1386, without TXA = 1167) were eligible and accepted into this meta-analysis. Pooled data indicated that the oral TXA group was effective compared to the without TXA group in terms of hemoglobin (Hb) drop (P
