Abstract

In this study, rice straw was used as the raw materials of biomass carbon to prepare biochar at different temperatures (400°C, 500°C, 600°C, 700°C, and 800°C). In addition, cetyl trimethyl ammonium bromide (CTAB) modified biochar was used to treat 2,4-dichlorophenol (2,4-DCP) in water. The influences of adsorbent dosage, solution pH, adsorption time, and initial solubility of the 2,4-DCP solution on the adsorption properties were investigated. The physicochemical properties of biochar were investigated using SEM, FT-IR, BET surface area, and pore size analysis. The results showed that the pyrolysis temperature had a great influence on the biochar structure. CTAB provided hydrophilic and hydrophobic groups to the modified biochar, which had increased adsorption capacity comparing to unmodified biochar. The pH also had a significant effect on the adsorption performance of biochar, and the adsorption performance of biochar decreased significantly under alkaline conditions. The maximum adsorption capacities of modified biochar and unmodified biochar were 59.81 and 20.89 mg/g, respectively, indicating that the adsorption capacity of modified biochar was significantly higher than that of unmodified biochar. The adsorption process of 2,4-DCP by rice straw biochar conformed to the Second-order kinetic model and the Freundlich isotherm adsorption model. [ABSTRACT FROM AUTHOR]

Abstract

The current method of extracting new login words ignores the diversity of constitution patterns of multi-word new login words (the number of words is greater than two). To solve this problem, this paper proposes an extraction method for new login word from Chinese micro-blog based on improved mutual information. First, micro-blog data are preprocessed, considering some nonsense signals such as web links and punctuation. Then, based on preprocessed data, the candidate strings are obtained by applying N-gram segmentation method. Finally, the extraction algorithm of new login words is proposed. In this algorithm, Multiword Mutual Information (MMI) is defined to describe the internal cohesion of the candidate string of multiple words in a variety of constitution patterns. And the candidate strings are extended and filtered according to frequency, MMI and right and left adjacency entropy to extract new login words. Experimental results show that this method has high feasibility for the extraction of new login words.

489. A data-fusion-model method for state of health estimation of Li-ion battery packs based on partial charging curve

Abstract

The estimation of State of Health (SOH) for battery packs used in Electric Vehicles (EVs) is a complex task with significant importance, accompanied by several challenges. This study introduces a data-fusion model approach to estimate the SOH of battery packs. The approach utilizes dual Gaussian Process Regressions (GPRs) to construct a data-driven and non-parametric aging model based on charging-based Aging Features (AFs). To enhance the accuracy of the aging model, a noise model is established to replace the random noise. Subsequently, the state-space representation of the aging model is incorporated. Additionally, the Particle Filter (PF) is introduced to track the unknown state in the aging model, thereby developing the data-fusion-model for SOH estimation. The performance of the proposed method is validated through aging experiments conducted on battery packs. The simulation results demonstrate that the data-fusion model approach achieves accurate SOH estimation, with maximum errors less than 1.5%. Compared to conventional techniques such as GPR and Support Vector Regression (SVR), the proposed method exhibits higher estimation accuracy and robustness.

Abstract

Phytoplasmas are insect-transmitted phytopathogenic bacteria, which secrete effector proteins that are often responsible for altering the plant morphology and behaviours of their vectors. Phytoplasma multifunctional effector proteins TENGU and SAP11 induce typical witches' broom symptoms, but their mode of action remains unknown. Previously, we have identified a SAP11-like effector from wheat blue dwarf phytoplasma, SWP1, which induces witches' broom symptoms in Nicotiana benthamiana. In this study, we observed that SWP1-expressing transgenic Arabidopsis thaliana plants showed typical witches' broom symptoms. On overexpression of SWP1 truncation mutants in N. benthamiana, we identified that the coiled-coil domain and nuclear localization were responsible for the induction of witches' broom symptoms. In addition, using yeast two-hybrid and bimolecular fluorescence complementation assays, we demonstrated that SWP1 interacts with A. thaliana transcription factor TCP18 (BRC1), the key negative regulator of branching signals in various plant species. Moreover, in planta co-expression analysis showed that SWP1 promotes the degradation of BRC1 via a proteasome system. These findings suggest that the phytoplasma effector SWP1 induces witches' broom symptoms through targeting of BRC1 and promoting its degradation.

Abstract

Mannan is an important renewable resource whose backbone can be hydrolyzed by β-mannanases to generate manno-oligosaccharides of various sizes. Only a few glycoside hydrolase (GH) 113 family β-mannanases have been functionally and structurally characterize. Here, we report the function and structure of a novel GH113 β-mannanase from Bacillus sp. N16-5 (Ba Man113A). Ba Man113A exhibits a substrate preference toward manno-oligosaccharides and releases mannose and mannobiose as main hydrolytic products. The crystal structure of Ba Man113A suggest that the enzyme shows a semi-enclosed substrate-binding cleft and the amino acids surrounding the +2 subsite form a steric barrier to terminate the substrate-binding tunnel. Based on these structural features, we conducted mutagenesis to engineer Ba Man113A to remove the steric hindrance of the substrate-binding tunnel. We found that F101E and N236Y variants exhibit increased specific activity toward mannans comparing to the wild-type enzyme. Meanwhile, the product profiles of these two variants toward polysaccharides changed from mannose to a series of manno-oligosaccharides. The crystal structure of variant N236Y was also determined to illustrate the molecular basis underlying the mutation. In conclusion, we report the functional and structural features of a novel GH113 β-mannanase, and successfully improved its endo -acting activity by using structure-based engineering. • The function and structure of a novel GH113 β-mannanase Ba Man113A was investigated. • Ba Man113A exhibits substrate preference toward oligosaccharides over mannans. • The structure shows a steric barrier at the reducing end of +2 subsite. • Rational design is applied to engineer Ba Man113A to improve specific activity. • The underlying molecular mechanism is elucidated by the structure of variant. Abstract

Unilateral moyamoya disease (MMD) represents a distinct subtype characterised by occlusive changes in the circle of Willis and abnormal vascular network formation. However, the aetiology and pathogenesis of unilateral MMD remain unclear. In this study, genetic screening of a family with unilateral MMD using whole-genome sequencing helped identify the c.1205 C > A variant of FOXM1, which encodes the transcription factor FOXM1 and plays a crucial role in angiogenesis and cell proliferation, as a susceptibility gene mutation. We demonstrated that this mutation significantly attenuated the proangiogenic effects of FOXM1 in human brain endothelial cells, leading to reduced proliferation, migration, and tube formation. Furthermore, FOXM1 c.1205 C > A results in increased apoptosis of human brain endothelial cells, mediated by the downregulation of the transcription of the apoptosis-inhibiting protein BCL2. These results suggest a potential role for the FOXM1 c.1205 C > A mutation in the pathogenesis of unilateral MMD and may contribute to the understanding and treatment of this condition. Abstract

Background: To investigate the role of CD47 expression and its relationship with tumor‐resident macrophages, specifically at the tumor margin, in patients with type II endometrial cancer. This study aims to elucidate whether CD47 could serve as a prognostic marker and to understand the dynamics between CD47 and macrophages, which could inform new therapeutic strategies. Methods: A retrospective cohort study was conducted involving 75 patients of type II endometrial. Immunohistochemical analysis was performed to assess CD47 expression and macrophage markers (CD68 and CD163). Results: The study found no direct correlation between CD47 expression levels and overall survival (p = 0.32), challenging its role as an independent prognostic marker in type II endometrial cancer. The higher expression of CD47 had significantly less incidence of endometrioid carcinoma G3 (p = 0.047). The negative correlation between CD47 H‐score and the density of CD68‐positive macrophages at tumor margin was statistically significant (p = 0.049). A high density of CD68‐positive macrophages at the tumor margin but a low density of CD163‐positive macrophages at the tumor margin were associated with poorer prognosis (p = 0.036). Conclusions: The complex interaction between CD47 and macrophages, particularly at the tumor margin, suggests new avenues for targeted therapy in type II endometrial cancer. [ABSTRACT FROM AUTHOR]

497. Cultural differences in hospitality service evaluations: mining insights of user generated content

Abstract

Online peer-to-peer platforms have been widely utilized to facilitate sharing vast amounts of user-generated content (UGC) in the hospitality industry. However, research on the cultural impacts of the broad spectrum of UGC remains nascent and limited. This study therefore elaborates how the hospitality service dimensions that are reflected in UGC both predict service evaluation and are contingent on a reviewer’s prevailing culture. We analyzed a usable sample of 9,257 hotel reviews from 148 countries with latent Dirichlet allocation and aspect-based sentiment analysis algorithms to identify the top service features and corresponding polarity scores. Machine learning identified three dimensions of hospitality service quality: adaptability, reliable delivery, and tangibles. These three dimensions predict customers’ overall hotel ratings, and the magnitude and direction of their effects depend on alignments of cultural orientations. Following Hofstede’s cultural dimensions, reliable delivery and tangibles align with individualism and uncertainty avoidance cultures. Adaptability aligns with power distance, masculinity, long-term orientation, and indulgence cultures.

498. Land Premium Effects of Urban Rail Transit and the Associated Policy Insights for TOD: A Case of Ningbo, China

Abstract

With considerable investments, mainly from local government budgets, the construction and operation of urban rail transit (URT) can exert significant spillover effects on the surrounding land use and land prices. In particular, China’s local governments are actively committed to developing their URT systems and promoting large-scale transit-oriented development (TOD) projects under the public land leasing policy. However, the connection between the land premium effects and TOD policy and practice is still lacking, particularly in the local government contexts, which exhibit significant policy and spatial heterogeneity. Thus, this research represents an attempt to better address this issue using the city of Ningbo as a case study. First, the premium effects of URT on land prices are examined, after which three crucial policy insights (land value capture [LVC], public–private cooperation [PPC], and urban regeneration) are proposed to enhance the effectiveness and efficiency of TOD, demonstrating its strong connection with the potential premium effects. The findings demonstrate that (1) local governments have adopted different innovative policies—with the ambition—to implement LVC; (2) assisted by PPC, the local rail transit authority can significantly amplify the premium effects, although it must still address the fair distribution of premiums across multiple stakeholders; and (3) transit-oriented urban regeneration can significantly influence land prices/land rents and subsequently generate significant gentrification, which will be further addressed by the TOD policy and practice.

499. Tea consumption and cerebral hemorrhage risk: a meta-analysis

Abstract

Background: Tea contains many polyphenols with biological properties such as antithrombosis and antioxidation. Recent observational studies on tea consumption concerning cerebral hemorrhage risk have reported inconsistent results. This meta-analysis aimed to summarize the accumulated evidence on the association between tea consumption and cerebral hemorrhage risk. Methods: Web of Science, PubMed, Embase, and Scopus databases were searched to identify relevant studies through December 2021. Relative risks (RRs) or odds ratios (ORs) from observational studies were synthesized. Results: Ten studies involving over 721,827 participants were included. Higher tea consumption was correlated with a 23% (RR = 0.77; 95% CI 0.66–0.89) lower risk of cerebral hemorrhage. Subgroup meta-analyses indicated higher tea consumption was beneficial in preventing cerebral hemorrhage risk for green tea, alcohol-adjusted, fruit/vegetables-adjusted, and physical activity-adjusted subgroups, respectively (P< 0.01). Dose–response analysis indicated each one-cup (120 ml/cup) increment in tea or green tea intake/day was correlated with an average of 2% (RR = 0.98, 95% CI 0.976–0.990), or 6% (RR = 0.94; 95% CI 0.92–0.97) lower cerebral hemorrhage risk. Conclusions: This study suggests that daily tea consumption is related to a lower risk of cerebral hemorrhage among adults. Green tea consumption appears to be more beneficial in preventing cerebral hemorrhage. Physical activity, fruit/vegetables, and alcohol may affect the relationship between tea consumption and hemorrhagic stroke. Future studies should investigate the interplay of tea with these factors.

Abstract

Cell migration plays a pivotal role in various physiological and pathological processes including angiogenesis, cancer metastasis, wound healing, inflammation, and embryogenesis. For cell migration in vitro , the conventional cell migration assays like the Boyden chamber and wound-healing assays are unable to meet the requirements of high-throughput, and are incapable of integrating complex environmental factors to comprehensively consider multi-parameters factors, such as cell matrix and concentration gradient, particularly those effecting cell migration. In contrast, microfluidic chips have the potential to take these challenges by allowing for precise and simultaneous control of multiple environmental factors mimicking in vivo environment of cells. Microfluidic devices successfully produce accurately controllable fluid and stable concentration gradient quickly, and manipulate gradients spatially and temporally. In addition, microfluidic devices enables accurate and reliable cell migration assay on real-time observation with limited amounts of reagents. It has exhibited tremendous potential due to its miniaturization, integration, high-throughput and high-precision. Owing to these particular advantages, over the past few years, a number of microfluidic chips combining two-dimensional (2D) and three-dimensional (3D) cell cultures have already been widely used in cell migration assays, demonstrating that microfluidic technology has significant implications for cell biology and cell-based assays. The microfluidic chips for cell migration assay using 2D platform could be classified into wound-healing and non-wound assays. Wound edges could be attained though a laminar flow of trypsin solution or removing solid barrier. After wound formed, cell migration could be detected and monitored using electric cell-substrate impedance sensing. Non-wound assays include chemotaxis and electrotaxis. The in vivo microenvironment is characterized by a 3D scaffold and multiple cell types. Therefore, increasing microfluidic devices with a 3D cell culture system have been developed for cell chemotaxis. These chips embed cells in a structure manufactured using agarose, collagen and hydrogel that mimics the extracellular matrix (ECM) of structural proteins found in real and living tissues. Various microfluidic devices with a 3D cell culture model can present cell-matrix and cell-cell interactions and provide reliable analysis platform for cell migration. Briefly, 2D and 3D microfludic chip-based models for concentration gradients, chemotaxis, electrotaxis and cells interactions have beneficial effects on cell migration assays. As a breakthrough to conventional methods, those microfluidic chip-based cell migration assays really promote the advanced progress in the area of cell biology and biomedicine. In this review, we describe the characterization of cell migration assays in microfluidic systems, highlight latest advances of the microfluidic device for researching in cell migration, focusing on both 2D and 3D cell cultures, and discuss advantages and disadvantages of this rapidly developed analysis technology.
