461. Integrated analysis of mRNA-seq and miRNA-seq reveals the potential roles of Egr1, Rxra and Max in kidney stone disease.

Abstract

Nephrolithiasis is one of the most common and frequent urologic diseases worldwide. The molecular mechanism of kidney stone formation is complex and remains to be illustrated. Transcript factors (TFs) that influenced the expression pattern of multiple genes, as well as microRNAs, important posttranscriptional modulators, play vital roles in this disease progression. Datasets of nephrolithiasis mice and kidney stone patients were acquired from Gene Expression Omnibus repository. TFs were predicted from differentially expressed genes by RcisTarget. The target genes of differential-expressed microRNAs were predicted by miRWalk. MicroRNA-mRNA network and PPI network were constructed. Functional enrichment analysis was performed via Metascape and Cytoscape identified hub genes. The assay of quantitative real-time PCR (q-PCR) and immunochemistry and the datasets of oxalate diet-induced nephrolithiasis mice kidneys and kidney stone patients’ samples were utilized to validate the bioinformatic results. We identified three potential key TFs (Egr1, Rxra, Max), which can be modulated by miR-181a-5p, miR-7b-3p and miR-22-3p, respectively. The TFs and their regulated hub genes influenced the progression of nephrolithiasis via altering the expression of genes enriched in the functions of fibrosis, cell proliferation and molecular transportation and metabolism. The expression changes of transcription factors were consistent in q-PCR and immunochemistry results. For regulated hub genes, they showed consistent expression changes in oxalate diet-induced nephrolithiasis mice model and human kidneys with stones. The identified and verified three TFs, which may be modulated by microRNAs in nephrolithiasis disease progression, mainly influence biological processes responding to fibrosis, proliferation and molecular transportation and metabolism. Abstract

Conductive hydrogels have attracted much attention because of their adjustable mechanical properties and excellent electrical conductivity. However, the variability and conductivity of traditional conductive hydrogels will be greatly reduced in low temperature environment, which seriously affects their application range. In this study, we fabricated a dual network conductive hydrogel with frost resistance. Here, water and glycerol binary solvents are introduced into the hydrogel to give the hydrogel antifreezing properties. The double-network structure of PAA/CMCs-Ca2+(W-G) hydrogel can provide good tensile and energy dissipation properties. The hydrogel strain sensor has high sensitivity (strain coefficient is 3.294 in the range of 100 ~ 250%, strain coefficient is 6.986 in the range of 250 ~ 400%) and good stability. Abstract

Covalent organic frameworks (COFs) have been extensively investigated due to their unique structure, porosity, and functionality. However, at the topological level, COFs remain as two‐dimensional (2D) or three‐dimensional (3D) structures, while COFs with one‐dimensional (1D) topology have not been systematically explored. In this work, we proposed a synthetic strategy for the construction of 1D‐COFs based on non‐linear edges and suitable high‐symmetry vertices. Compared with their 2D‐COFs counterparts, the 1D‐COFs with AIEgens located at the vertex of the frame exhibited enhanced fluorescence. The density functional theory (DFT) calculations revealed that the dimensional‐induced rotation restriction (DIRR) effect could spontaneously introduce additional non‐covalent interactions between the strip frames, which could substantially diminish non‐radiative transitions. Abstract

Inspired by the hierarchical fabrication technique, many self-assembly procedures have improved the construction of nanomaterials with unique physicochemical characteristics and multiple functions. The generation of multiple complexes is always accompanied by hierarchical structures and intriguing properties that are distinct from their individual segments. An interesting composite is amorphous magnetic Zn-Zr phosphate hydrated nanosheets (Zn-Zr APHNs), generated using templated synthesis and nanoparticle codeposition. The special porous structure of this construct, together with the abundance of metal ions and hydrate present, endows it with many interaction sites for proteins, provides high loading efficiency, and enhances bioactivity. Then, a series of proteins, including enzymes, was immobilized by the Zn-Zr APHNs by multiple interactions, high ionization, and larger surface of the nanosheets. In this study, novel methods for the enrichment of bioactive proteins while retaining the activity of protein payloads are presented. As a verification method, it is indicated that the Zn-Zr APHNs can deliver enzyme proteins (i.e., Cyt-c) to increase the catalytic activity with their biological function and structural integrity, resulting in a highly increased activity to free proteins.

479. Carbon Dots from Lycium barbarumAttenuate Radiation-Induced Bone Injury by Inhibiting Senescence via METTL3/Clip3 in an m6A-Dependent Manner

Abstract

Radiation-induced bone injury management remains a challenge in clinical practice, and there is no effective medicine. Recently, biomass-derived carbon dots (CDs) have attracted attention in biomedical engineering due to the advantages of abundant heteroatoms, low toxicity, and no need to drug loading. Here, we report that CDs, synthesized from Lycium barbarumvia hydrothermal strategy, can effectively alleviate radiation-induced bone injury. CCK-8, apoptosis analysis, β-galactosidase staining, quantitative polymerase chain reaction, and western blots demonstrate that CDs can mediate radiation-induced damage and senescence of bone marrow mesenchymal stem cells (BMSCs). CDs regulate osteogenic- and adipogenic-balance after irradiation, shown by alizarin red and oil red O staining. In vivo experiments reveal that CDs prevent the occurrence of osteoradionecrosis in rats, demonstrated by micro-CT and histology examination. The osseointegration of titanium implants installed in irradiated bone is promoted by CDs. Mechanistically, CDs increase the N6-methyladenosine (m6A) level of irradiated BMSCs via the increased methyltransferase-like 3 (METTL3). High-throughput sequencing facilitates detection of increased m6A levels located in the 3′-untranslated regions (UTR) of the CAP-Gly domain containing linker protein 3 (Clip3) mRNA. The dual-luciferase reporter assay shows that 3′UTR is the direct target of METTL3. Subsequently, the increased m6A modification led to enhanced degradation of mRNA and downregulated CLIP3 expression, eventually resulting in the alleviation of radiation-induced bone injury. Interfering with the METTL3/Clip3 axis can antagonize the effect of CDs, indicating that CDs mediate radiation-induced bone injury via the METTL3/Clip3 axis. Taken together, CDs from L. barbarumalleviate radiation-induced bone injury by inhibiting senescence via regulation of m6A modification of Clip3. The present study paves a new pathway for the management of radiation-induced bone injury.

Abstract

Hierarchically porous Ce/TiO2/graphitic carbon microsphere composites (xCe/TiO2/GCM, where x = 0.2, 1.0, 2.0, 5.0 mmol·L–1) were prepared for the first time by using a simple colloidal crystal template and characterized by X-ray diffraction, nitrogen adsorption and desorption, scanning electron microscopy, and ultraviolet–visible diffuse reflectance spectra. In addition, the photocatalytic activity of CO2 reduction by H2O under simulated solar irradiation was studied. The results showed that the Ce/TiO2/GCM composite material was characterized by large porosity, high concentration of metal compounds and graphitized carbon matrix, and the content of acetone solvent having a great impact on its form. In terms of the photocatalytic CO2 reaction, the CH4 and CO productions were 4.587 and 357.851 μmol·g–1, respectively. The 2Ce/TiO2/GCM photocatalyst gave the highest production rate for three products. Under simulated solar irradiation, the Ce/TiO2/GCM has excellent photocatalytic activity in the photoreduction of CO2 from H2O, which was related to the special composition and the Ce/TiO2/GCM structure.

Abstract

A new rare-earth borate Na3YB8O15 was fabricated by the flux method. The compound crystallizes in the P1¯ group, and shows a three-dimensional network built by [B8O17] fundamental building blocks (FBBs) with [NaO6], [NaO7], and [YO7] polyhedra. The [B8O17] FBBs are composed of five planar BO3 and three BO4 tetrahedra and connected via corner-sharing to construct a unique one-dimensional (1D) [B8O15]∞ infinity chain that is observed for the first time in the Na2O–Y2O3–B2O3 system compound, enriching the structural diversity of rare-earth borate. The title compound exhibits a short ultraviolet (UV) cutoff edge of ∼195 nm and a wide experimental band gap of 5.50 eV. DFT calculations indicate that Na3YB8O15 is a direct band gap compound with a calculated GGA band gap of 5.14 eV, and the band gap is mainly determined by B 2p and O 2p orbitals. The results update the phase diagram of Na2O–Y2O3–B2O3 and enrich the diversity of rare-earth metal borates. [ABSTRACT FROM AUTHOR]
