Abstract

It has been established that the M1, P and M2 pore-forming regions in glutamate receptors (GluRs) and potassium channels have very similar structure. Differences are mainly limited to the selectivity filter region. The amino-acid sequence and functional relationships between GluR0, eukaryotic GluRs and KcsA provide backing for the hypothesis that all GluRs are tetrameric in subunit stoichiometry. It was found that the N-terminal domain is not vital for channel gating.

484. Epithelial cells-enriched lncRNA SNHG8 regulates chromatin condensation by binding to Histone H1s

Abstract

Linker histone H1 proteins contain many variants in mammalian and can stabilize the condensed state of chromatin by binding to nucleosomes and promoting a more inaccessible structure of DNA. However, it is poorly understood how the binding of histone H1s to chromatin DNA is regulated. Screened as one of a collection of epithelial cells-enriched long non-coding RNAs (lncRNAs), here we found that small nucleolar RNA host gene 8 (SNHG8) is a chromatin-localized lncRNA and presents strong interaction and phase separation with histone H1 variants. Moreover, SNHG8 presents stronger ability to bind H1s than linker DNA, and outcompetes linker DNA for H1 binding. Consequently, loss of SNHG8 increases the amount of H1s that bind to chromatin, promotes chromatin condensation, and induces an epithelial differentiation-associated gene expression pattern. Collectively, our results propose that the highly abundant SNHG8 in epithelial cells keeps histone H1 variants out of nucleosome and its loss contributes to epithelial cell differentiation.

Abstract

Ionotropic glutamate receptors play a major role in synaptic transmission in both vertebrates and invertebrates. Transmembrane ion channels are opened through ligands. These receptors play a major role in the way the nervous system develops and functions, and in learning and memory. They have been linked to a number of dysfunctions such as Huntington's disease, stroke, Parkinson's and epilepsy. New ligands can be designed using information on the crystal structure of a ligand-binding region using these receptors.

Abstract

Expression of the S1S2 ligand binding domain [Kuusinen, A., Arvola, M. & Keinanen, K. (1995) EMBO J. 14, 6327-6332] of the rat [Alpha]-amino-3-hydroxy-5-methylisoxazole-4-propionic acid-selective glutamate receptor GluR2 in Escherichia coli under control of a T7 promoter leads to production of > 100 mg/liter of histidine-tagged S1S2 protein (HS1S2) in the form of inclusion bodies. Using a novel fractional factorial folding screen and a rational, step-by-step approach, multiple conditions were determined for the folding of the HS1S2 [Alpha]-amino-3-hydroxy-5-methylisoxazole-4-propionic acid binding domain. Characterization of the HS1S2 ligand binding domain showed that it is water-soluble, monomeric, has significant secondary structure, and is sensitive to trypsinolysis at sites close to the beginning of the putative transmembrane regions. Application of a fractional factorial folding screen to other proteins may provide a useful means to evaluate E. coli as an economical and convenient expression host.

Abstract

Patient X: A 67-year-old Caucasian man slips on a patch of ice. He has abrasions to his hands and has sustained significant damage to his hip. At the emergency room, he informs clinicians he takes atorvastatin, metformin and glimepiride to treat hypertension and Type 2 Diabetes Mellitus (T2DM). X-rays reveal a fractured hip, which will require total hip replacement surgery. Biotechnology is a major force poised to help us to live longer and healthier lives. In 2015, the United Nations defined 17 Sustainable Development Goals aimed at providing an all-encompassing framework for improving the state of the world (**). Promoting healthy living for all at all ages, is one of the principal objectives. Using the commonplace example described above, we examine how some of the most well-known biotechnologies (genome editing, stem cell therapy, tissue engineering and precision medicine) are able to benefit Patient X today, in 2018 and we also provide perspective on the additional value these same technologies could offer only ∼20 years into the future (Fig. 1). During this ∼20 year gap, both technical challenges as well as ethical, legal and socio-economic questions must be addressed before these technologies can achieve broader impact. Further, as both the successes and the failures will likely have enduring effects on society, responsible oversight of these and other biotechnologies are necessary as science and applications move out of the lab into clinical practice. In the next 20 years biotechnology will undoubtedly transform healthcare but how, when and, in some cases, whether it should, require careful consideration.
