ObjectiveExisting brittle source identification methods have such problems as their single-perspective approach, disregard for the local and global characteristics of nodes, and insufficient consideration of the loads carried by nodes.MethodsThe method proposed herein identifies the inherent brittleness of the ship power system as the root cause of interruption and chain collapse. To establish a brittleness index, the method first considers node degree, eigenvector centrality, node cohesiveness, closeness centrality and brittle electrical betweenness from a brittleness perspective based on complex network theory. The combinatorial intuitionistic fuzzy analytic hierarchy process (IFAHP）and entropy weight method (EWM) with the Nash equilibrium as the coordination objective. The five indexes are then combined through comprehensive assignment to obtain a comprehensive brittleness index. Finally, the proposed brittle source identification method is validated using a typical four-station annular ship grid. ResultsThe method identifies four levels of brittle sources based on the network efficiency of different brittleness indexes, and the results demonstrate its ability to identify brittle sources more comprehensively and reasonably.ConclusionThe results of this study can provide effective guidance for the prevention and control of inter-locking faults in ship power networks.

Antibiotics release into the water environment through sewage discharge is a significant environmental concern. In the present study, we investigated the removal of ciprofloxacin (CIP) in simulated sewage by biological aeration filter (BAF) equipped with Fe3O4-modified zeolite (Fe3O4@ZF). Fe3O4@ZF were prepared with impregnation method, and the Fe3O4 particles were successfully deposited on the surface of ZF in an amorphous form according to the results of XPS and XRD analysis. The modification also increased the specific surface area (from 16.22 m²/g to 22 m²/g) and pore volume (from 0.0047 cm³/g to 0.0063 cm³/g), improving the adsorption efficiency of antibiotics. Fe3O4 modified ZF improved the treatment performance significantly, and the removal efficiency of CIP in BAF-Fe3O4@ZF was 79%±2.4%. At 10ml/L CIP, the BAF-Fe3O4@ZF reduced the relative abundances of antibiotics resistance genes (ARGs) int, mexA, qnrB and qnrS in the effluent by 57.16%, 39.59%, 60.22%, and 20.25%, respectively, which effectively mitigate the dissemination risk of ARGs. The modification of ZF increased CIP-degrading bacteria abundance, such as Rhizobium and Deinococcus-Thermus, and doubled bacterial ATP activity, promoting CIP degradation. This study offers a viable, efficient method to enhance antibiotic treatment and prevent leakage via sewage discharge. HIGHLIGHTS Fe3O4 modification can improve the removal efficiency of CIP by BAF.; Fe3O4@ZF reduced the released resistance genes in the effluent.; Fe3O4@ZF enriched the abundance of CIP-degrading bacteria.; Fe3O4@ZF increased the microbial activity on the fillers rather than the biomass.;

Background: The detailed clinical phenotype of patients carrying the α-galactosidase gene (GLA) c.548 G > A/p.Gly183Asp (p.G183D) variant in Fabry disease (FD) has not been thoroughly documented in the existing literature. Methods: This paper offers a meticulous overview of the clinical phenotype and relevant auxiliary examination results of nine confirmed FD patients with the p.G183D gene variant from two families. Pedigree analysis was conducted on two male patients with the gene variant, followed by biochemical and genetic screening of all high-risk relatives. Subsequently, evaluation of multiple organ systems and comprehensive instrument assessment were performed on heterozygotes of the p.G183D gene variant. Results: The study revealed that all patients exhibited varying degrees of cardiac involvement, with two demonstrating left ventricular wall thickness exceeding 15 mm on echocardiography, and the remaining six exceeding 11 mm. Impaired renal function was evident in all six patients with available blood test data, two of whom underwent kidney transplantation. Eight cases reported neuropathic pain, and five experienced varying degrees of stroke or transient ischemic attack (TIA). Conclusion: This study indicates that the GLA p.G183D gene variant can induce premature organ damage, particularly affecting the heart, kidneys, and nervous system.

Abstract Aims Recurrent heart failure hospitalization (HFH) is an important feature of the progression of heart failure (HF). In the Treatment of Preserved Cardiac Function Heart Failure with an Aldosterone Antagonist (TOPCAT) trial, we analysed risk factors for recurrent HFH events in HF patients with preserved ejection fraction (HFpEF) and developed a risk prediction model for recurrent HFH. Methods and results This analysis focused on the subset of TOPCAT participants enrolled in the Americas (n = 1767). Recurrent HFH was defined as two or more hospitalizations for HF during the follow‐up period. Lasso regression and multivariate logistic regression were used to screen the risk factors, and the risk prediction model of recurrent HFH was established. During a median follow‐up period of 3.4 (95% confidence interval: 3.3–3.6) years, 72.2% (542 of 751 total hospitalizations) of HFH events occurred in 9.4% (n = 163) of patients with recurrent HFHs. Patients in the recurrent HFH group had higher cardiovascular mortality rate [6.2 per 100 patient‐years (PY) vs. 3.8 per 100 PY, P = 0.016] and all‐cause mortality rate (10.0 per 100 PY vs. 6.8 per 100 PY, P = 0.015) than those in the non‐recurrent HFH group. The model consisting of nine predictors has moderate predictive power for recurrent HFH events in patients with HFpEF (AUC = 0.75, Brier score = 0.08). Decision curve analysis showed a net clinical benefit from the application of the prediction model. Conclusions In patients with HFpEF, the majority of HFHs occur in a small proportion of patients with repeated hospitalizations, who typically have more comorbidities and are at higher risk of death. The predictive model developed in this study helps to identify patients at high risk of recurrent HFH.

As global surface water pollutants, tire-wear particles (TWPs) are increasingly concerning, with TWP leachate hepatotoxicity poorly understood. In this study, based on environmental TWP concentrations, TWP leachate exposure (0, 0.0005, 0.005, 0.05, and 0.5 mg/mL) in black-spotted frogs was investigated over a 21 day period. TWP leachates at realistic environmental levels disturbed intestinal microbiome homeostasis, which manifested as decreased and increased Chloroflexi and Proteobacteria abundance, respectively, and elevated lipopolysaccharide (LPS) levels in plasma. Also, the content of lipopolysaccharide-binding protein, the binding site of LPS, was increased, and downstream LPS immune pathways, such as toll-like receptor 4 (TLR4)-nuclear factor (NF)-κB (TLR4/NF-κB) signaling, were activated. Subsequently, inflammation reactions, oxidative damage, and histopathology were affected in liver samples. These results shed new light on the potential mechanisms underpinning TWP leachate-associated liver injury via the gut–liver axis, and contribute to a better understanding of emerging TWP ecotoxicology.

Cytochrome P450 (CYP) enzymes are crucial for the detoxification of xenobiotics, cellular metabolism, and homeostasis. This study investigated the molecular characterization of CYP enzymes in the black-spotted frog, Pelophylax nigromaculatus, and examined the regulation of CYP expression in response to chronic exposure to the antibiotic sulfamethoxazole (SMX) at various environmental concentrations (0, 1, 10, and 100 μg/L). The full-length cDNA of Pn-CYP26B1 was identified. The sequence included open reading frames of 1,536 bp, encoding proteins comprising 511 amino acids. The signature motif, FxxGxxxCxG, was highly conserved when compared with a number of selected animal species. SMX significantly upregulated the expression of the protein CYP26B1 in frog livers at concentrations of 1 and 10 μg/L. SMX showed an affinity for CYP26B1 of −7.6 kcal/mol, indicating a potential mechanism for SMX detoxification or adaptation of the frog. These findings contributed to our understanding of the environmental impact of antibiotics on amphibian species and underscored the importance of CYP enzymes in maintaining biochemical homeostasis under exposure to xenobiotic stress.

Endocrine-disrupting chemicals (EDCs) are ubiquitous in ecological environments and have become a great issue of public health concern since the 1990 s. There is a deep scientific understanding of the toxicity of EDCs. However, recent studies have found that the abnormal physiological functions of the parents caused by EDCs could be transmitted to their unexposed offspring, leading to intergenerational toxicity. We questioned whether sustained epigenetic changes occur through the male germline. In this review, we (1) systematically searched the available research on the intergenerational impacts of EDCs in aquatic and mammal organisms, including 42 articles, (2) summarized the intergenerational genetic effects, such as decreased offspring survival, abnormal reproductive dysfunction, metabolic disorders, and behavioral abnormalities, (3) summarized the mechanisms of intergenerational toxicity through paternal interactions, and (4) propose suggestions on future research directions to develop a deeper understanding of the ecological risk of EDCs.

ObjectiveAn adaptive finite-time filtered backstepping sliding mode controller is designed to solve the rudder roll stabilization control problem of underactuated ships in unknown wave disturbance conditions. MethodFirst, in order to avoid differential operation for the virtual control law, a first-order filter is introduced to address the problem of differential explosion that affects the backstepping approach. By combining the filtered backstepping technique with adaptive sliding mode control, finite-time convergent control laws for rudder roll stabilization are designed on the basis of the finite-time control theory. Finally, the stability of the heading control subsystem and roll damping subsystem is proven by the Lyapunov stability theory. ResultsThe simulation results indicate that the designed controller can realize course tracking and roll damping simultaneously in finite time under different wave disturbance conditions. Compared with backstepping sliding mode control, the designed controller can improve the course-keeping control performance, and the roll reduction rate is increased by 5%. ConclusionThe proposed method can provide useful references for the rudder roll stabilization control problem of underactuated ships.

With the rapid development of mobile devices, aggregation security and efficiency topics are more important than past in crowd sensing. When collecting large-scale vehicle-provided data, the data transmitted via autonomous networks are publicly accessible to all attackers, which increases the risk of vehicle exposure. So we need to ensure data aggregation security. In addition, low aggregation efficiency will lead to insufficient sensing data, making the data unable to provide data mining services. Aiming at the problem of aggregation security and efficiency in large-scale data collection, this article proposes a data collection mechanism (VDCM) for crowd sensing in vehicular ad hoc networks (VANETs). The mechanism includes two mechanism assumptions and selects appropriate methods to reduce consumption. It selects sub mechanism 1 when there exist very few vehicles or the coalition cannot be formed, otherwise selects sub mechanism 2. Single aggregation is used to collect data in sub mechanism 1. In sub mechanism 2, cooperative vehicles are selected by using coalition formation strategy and auction cooperation agreement, and multi aggregation is used to collect data. Two sub mechanisms use Paillier homomorphic encryption technology to ensure the security of data aggregation. In addition, mechanism supplements the data update and scoring steps to increase the amount of available data. The performance analysis shows that the mechanism proposed in this paper can safely aggregate data and reduce consumption. The simulation results indicate that the proposed mechanism reduces time consumption and increases the amount of available data compared with existing mechanisms.

ABSTRACTSeeking economical, efficient and environmentally friendly nitrogen fertigation management strategies is extremely important for sustainable maize production. A two-year field experiment was conducted to investigate the effects of four nitrogen fertilization methods N0: no nitrogen application; FP: applying 234 kg N ha−1 at sowing; NF1: applying 210 kg N ha−1, 50% N at sowing and 50% N fertigation at the V8 stage; and NF4: applying 210 kg N ha−1 sowing: 30%, fertigation at V8: 20%, V12: 20%, R1: 20%, R3: 10% on maize yield, nitrogen and water use efficiency, economic benefits and the environmental assessment in Northeast China. The results showed that compared with the FP treatment, the NF1 and NF4 treatments significantly increased maize yield by 13.2% and 18.6%, increased recovery efficiency of nitrogen (REN) by 38.6% and 68.4%, agronomic nitrogen efficiency (AEN) by 58.6% and 78.2%, water use efficiency (WUE) by 23.8% and 26.9%, and net income by 16.1% and 25.7%, respectively. Additionally, the treatments reduced the apparent nitrogen loss by 69.5% and 96.1%, nitrogen footprint by 34.4% and 37.4% and carbon footprint by 17.0% and 20.8%, respectively. The yield, REN and net income of NF4 were significantly higher than those of NF1 by 4.8%, 21.6% and 8.3%, respectively, and the nitrogen and carbon footprints of NF4 were significantly lower than those of NF1 by 4.6% and 4.6%, respectively. The NF4 treatment was recommended as an effective method for improving yield and net income and reducing the environmental cost while achieving sustainable maize production.

The inevitability of faults arises due to prolonged exposure of photovoltaic (PV) power plants to intricate environmental conditions. Therefore, fault diagnosis of PV power plants is crucial to ensure the continuity and reliability of power generation. This paper proposes a fault diagnosis method that integrates PV power prediction and an exponentially weighted moving average (EWMA) control chart. This method predicts the PV power based on meteorological factors using the adaptive particle swarm algorithm-back propagation neural network (APSO-BPNN) model and takes its error from the actual value as a control quantity for the EWMA control chart. The EWMA control chart then monitors the error values to identify fault types. Finally, it is verified by comparison with the discrete rate (DR) analysis method. The results showed that the coefficient of determination of the prediction model of the proposed method reached 0.98. Although the DR analysis can evaluate the overall performance of the inverter and identify the faults, it often fails to point out the specific location of the faults accurately. In contrast, the EWMA control chart can monitor abnormal states such as open and short circuits and accurately locate the string where the fault occurs.

The use of human and veterinary drugs has led to the accumulation of pharmaceuticals in various aquatic environments at progressively increasing levels, exhibiting strong ecological risks. Metformin is widely used as a first-line prescription drug for the treatment of type 2 diabetes mellitus as well as a livestock drug. Unlike other drugs, metformin is not metabolized in the body, and almost all of its intake is excreted and released into the aquatic environment via urine and feces, causing adverse effects on aquatic ecosystems. This review provides an overview of the occurrence and detection of metformin in the aquatic environment and its toxic effects on different aquatic organisms (fish, daphnia, rotifers, chlorella). Metformin has been documented in a variety of aqueous environments such as wastewater, surface water, and groundwater as well as drinking water. The wide distribution of metformin in the aqueous environment calls for the development of more accurate detection methods. This paper reviews detection methods for metformin in the aqueous environment and evaluates their advantages and disadvantages. Toxicity studies have shown that metformin can cause adverse reactions in fish, such as oxidative stress, genotoxicity, disruption of intestinal flora, and morphological alterations; it also affects the growth and reproduction of small aquatic organisms. Knowledge gaps in the field of metformin research were assessed, and future research priorities were identified.

ObjectiveIn order to solve the problem of an underactuated ship responding slowly to heading changes during rudder roll stabilization (RRS) caused by fixed weight values in model predictive control (MPC), a RRS control method based on the finite time extended states observer (FTESO), fuzzy rules and robust predictive control is proposed. MethodsA fixed speed linear underactuated ship model is established for controller design. The FTESO is used to estimate the ship's motion states and external disturbances. By analyzing the conditions of the ship's course-keeping and heading change, the objective function weights under the two conditions and the fuzzy rules between the states and weights are designed respectively. Robust predictive control is used to solve the multi-objective cooperative control problem with constraints. The closed-loop stability of the proposed control method is then proven theoretically. ResultsAccording to a numerical simulation of a multi-purpose naval vessel, the proposed control method is compared with a disturbance compensation MPC and disturbance observer enhanced MPC, and is shown to have a higher roll stabilization rate by 5.74% and 0.898 3%, respectively. The response time of the proposed method for a 30° heading change is also reduced by 1.8 s and 7.3 s respectively. ConclusionThe effectiveness of the proposed method in underactuated ship rolling reduction is proven.

Abstract Background Adenine base editors (ABEs) are promising therapeutic gene editing tools that can efficiently convert targeted A•T to G•C base pairs in the genome. However, the large size of commonly used ABEs based on SpCas9 hinders its delivery in vivo using certain vectors such as adeno-associated virus (AAV) during preclinical applications. Despite a number of approaches having previously been attempted to overcome that challenge, including split Cas9-derived and numerous domain-deleted versions of editors, whether base editor (BE) and prime editor (PE) systems can also allow deletion of those domains remains to be proven. In this study, we present a new small ABE (sABE) with significantly reduced size. Results We discovered that ABE8e can tolerate large single deletions in the REC2 (Δ174-296) and HNH (Δ786-855) domains of SpCas9, and these deletions can be stacked together to create a new sABE. The sABE showed higher precision than the original ABE8e, with proximally shifted protospacer adjacent motif (PAM) editing windows (A3- A15), and comparable editing efficiencies to 8e-SaCas9-KKH. The sABE system efficiently generated A-G mutations at disease-relevant loci (T1214C in GAA and A494G in MFN2) in HEK293T cells and several canonical Pcsk9 splice sites in N2a cells. Moreover, the sABE enabled in vivo delivery in a single adeno-associated virus (AAV) vector with slight efficiency. Furthermore, we also successfully edited the genome of mouse embryos by microinjecting mRNA and sgRNA of sABE system into zygotes. Conclusions We have developed a substantially smaller sABE system that expands the targeting scope and offers higher precision of genome editing. Our findings suggest that the sABE system holds great therapeutic potential in preclinical applications.

ObjectivesIn this study, an adaptive non-singular fast terminal sliding mode rudder roll stabilization controller based on a multiple-layer recurrent neural network ( MLRNN) is proposed for the rudder roll stabilization control of an underactuated surface ship with unknown nonlinear system functions and random external disturbances. MethodsFirst, in view of the singularity and convergence problems in traditional sliding mode control, a non-singular fast terminal sliding surface is introduced, and the sliding mode control law is designed under the assumption that the ship model is known. The traditional radial basis function neural network （RBFNN） is then improved and used to approximate unknown nonlinear system functions in order to solve the problem of ship models being difficult to establish when the ship is sailing while also improving the control accuracy. The stability and finite time convergence of the system are proven by the Lyapunov theory, and the adaptive laws of the neural network parameters are derived. Finally, a numerical simulation analysis of a multi-purpose naval ship is carried out. ResultsThe results show that when the ship is under the course keeping condition, the roll reduction rate of the proposed controller is 50.41%, which is 19.2% larger than that of the non-singular fast terminal sliding mode controller (NFTSMC). When the ship is under the course changing condition, the roll reduction rate of the proposed controller is 23.46%, which is 12.59% larger than that of the NFTSMC. ConclusionsThis method can provide valuable references for the design of underactuated ship rudder roll stabilization controllers.

Summary: Fish require abundant nutrients to generate a large number of eggs for spawning. Based on the evolutionary conservation of human FBN2 and its C-terminal placensin-like sequences in fish, we identified a peptide hormone gonacin (GONAdal Cell placensIN) and found its high expression in early-stage germ cells in the ovary and testis of zebrafish. We demonstrated that gonacin is essential for food intake, glucose release, and ovarian development in zebrafish. Similar expression patterns and functions of gonacin were also demonstrated in rainbow trout. Gonacin represents the first hormone secreted by germ cells with endocrine functions in vertebrates, bridging the energy homeostasis and reproduction.

To solve the non-uniformity of stress in space membrane structure and the lack of shear compliant border configuration design method, shear compliant borders are designed, optimized, and verified in terms of configuration. Firstly, an orthotropic model of the borders is built by combining Hill and Christensen-Lo composite material models. Secondly, a finite element form-finding method is put forward by establishing rectangular and cylindrical coordinates in different areas. The configuration of borders is obtained and the influence of the borders on the edge of the membrane is 0.23%, which means that the borders are compatible with the existing tensegrity systems, especially the tensioning components and the cable sleeves. Thirdly, simulation verifies that borders can cut the spread of shear stress and improve the stress uniformity in membrane structure. The maximum stress in the membrane effective area is decreased by 35.6% and the stress uniformity is improved by 30.5%. Finally, a membrane extension experiment is committed to compare the flatness of membrane surface under shear stress with and without shear compliant borders. The borders decrease the increment speed of flatness by 58.1%, which verifies the amelioration of stress uniformity. The shear compliant border configuration design method provides a reference for space membrane structure stress-uniform design.

Abstract As a typical two‐dimensional (2D) transition metal dichalcogenides (TMDCs) material with nonzero band gap, MoS2 has a wide range of potential applications as building blocks in the field of nanoelectronics. The stability and reliability of the corresponding nanoelectronic devices depend critically on the mechanical performance and cyclic reliability of 2D MoS2. Although an in situ technique has been used to analyze the mechanical properties of 2D materials, the cyclic mechanical behavior, that is, fatigue, remains a major challenge in the practical application of the devices. This study was aimed at analyzing the planar cyclic performance and deformation behavior of three‐layer MoS2 nanosheets (NSs) using an in situ transmission electron microscopy (TEM) variable‐amplitude uniaxial low‐frequency and cyclic loading–unloading tensile acceleration test. We also elucidated the strengthening effect of the natural overlaying affix fragments (other external NSs) or wrinkle folds (internal folds from the NS itself) on cycling performances and service life of MoS2 NSs by delaying the whole process of fatigue crack initiation, propagation, and fracture. The results have been confirmed by molecular dynamics (MDs) simulations. The overlaying enhancement effect effectively ensures the long‐term reliability and stability of nanoelectronic devices made of few‐layer 2D materials.

Abstract Background Pancreatic cancer (PC) is a highly lethal malignancy regarding digestive system, which is the fourth leading factor of cancer-related mortalities in the globe. Prognosis is poor due to diagnosis at advanced disease stage, low rates of surgical resection, and resistance to traditional radiotherapy and chemotherapy. In order to develop novel therapeutic strategies, further elucidation of the molecular mechanisms underlying PC chemoresistance is required. Ribosomal RNA biogenesis has been implicated in tumorigenesis. Small nucleolar RNAs (snoRNAs) is responsible for post-transcriptional modifications of ribosomal RNAs during biogenesis, which have been identified as potential markers of various cancers. Here, we investigate the U3 snoRNA-associated protein RRP9/U3-55 K along with its role in the development of PC and gemcitabine resistance. Methods qRT-PCR, western blot and immunohistochemical staining assays were employed to detect RRP9 expression in human PC tissue samples and cell lines. RRP9-overexpression and siRNA-RRP9 plasmids were constructed to test the effects of RRP9 overexpression and knockdown on cell viability investigated by MTT assay, colony formation, and apoptosis measured by FACS and western blot assays. Immunoprecipitation and immunofluorescence staining were utilized to demonstrate a relationship between RRP9 and IGF2BP1. A subcutaneous xenograft tumor model was elucidated in BALB/c nude mice to examine the RRP9 role in PC in vivo. Results Significantly elevated RRP9 expression was observed in PC tissues than normal tissues, which was negatively correlated with patient prognosis. We found that RRP9 promoted gemcitabine resistance in PC in vivo and in vitro. Mechanistically, RRP9 activated AKT signaling pathway through interacting with DNA binding region of IGF2BP1 in PC cells, thereby promoting PC progression, and inducing gemcitabine resistance through a reduction in DNA damage and inhibition of apoptosis. Treatment with a combination of the AKT inhibitor MK-2206 and gemcitabine significantly inhibited tumor proliferation induced by overexpression of RRP9 in vitro and in vivo. Conclusions Our data reveal that RRP9 has a critical function to induce gemcitabine chemoresistance in PC through the IGF2BP1/AKT signaling pathway activation, which might be a candidate to sensitize PC cells to gemcitabine. Polycomb repressive complex 2 (PRC2) contributes to catalyze the methylation of histone H3 at lysine 27 and plays vital roles in transcriptional silencing and growth development in various organisms. In Magnaporthe oryzae, histone H3K27 is found to associate with altered transcription of in planta induced genes. However, it is still unknown whether and how H3K27me3 modification is involved in pathogenicity to rice and stress response. In this study, we found that core subunits of PRC2, Kmt6-Suz12-Eed, were required for fungal pathogenicity to rice in M. oryzae. Kmt6-Suz12-Eed localized in the nuclei and was necessary for the establishment of H3K27me3 modification. With ChIP-seq analysis, 9.0% of genome regions enriched with H3K27me3 occupancy, which corresponded to 1033 genes in M. oryzae. Furthermore, deletion of Kmt6, Suz12 or Eed altered genome-wide transcriptional expression, while the de-repression genes in the Δkmt6 strain were highly associated with H3K27me3 occupancy. Notably, plenty of genes which encode effectors and secreted enzymes, secondary metabolite synthesis genes, and cell wall stress-responsive genes were directly occupied with H3K27me3 modification and de-repression in the Δkmt6 strain. These results elaborately explained how PRC2 was required for pathogenicity, which is closely related to effector modulated host immunity and host environment adaption.

g-C3N4 has been regarded as a promising photocatalyst for photo-reforming antibiotics for H2 production but still suffers from its high charge recombination, which has been proven to be solvable by constructing a g-C3N4 homo-junction. However, those reported methods based on uncontrollable calcination for preparing a g-C3N4 homo-junction are difficult to reproduce. Herein, an amorphous/crystalline g-C3N4 homo-junction (ACN/CCN) was successfully synthesized via the electrostatic self-assembly attachment of negatively charged crystalline g-C3N4 nanorods (CCN) on positively charged amorphous g-C3N4 sheets (ACN). All the ACN/CCN samples displayed much higher photo-reforming of antibiotics for H2 production ability than that of pristine ACN and CCN. In particular, ACN/CCN-2 with the optimal ratio exhibited the best photocatalytic performance, with a H2 evolution rate of 162.5 μmol·g−1·h−1 and simultaneous consecutive ciprofloxacin (CIP) degradation under light irradiation for 4 h. The UV–vis diffuse reflectance spectra (DRS), photoluminescence (PL), and electrochemical results revealed that a homo-junction is formed in ACN/CCN due to the difference in the band arrangement of ACN and CCN, which effectively suppressed the charge recombination and then led to those above significantly enhanced photocatalytic activity. Moreover, H2 was generated from the water reduction reaction with a photogenerated electron (e−), and CIP was degraded via a photogenerated hole (h+). ACN/CCN exhibited adequate photostability and reusability for photocatalytic H2 production with simultaneous CIP degradation. This work provides a new idea for rationally designing and preparing homo-junction photocatalysts to achieve the dual purpose of chemical energy production and environmental treatment.

The functional groups and the ecological health of a constructed oyster reef were investigated during April 2016 to October 2017 in the Yangtze estuary. The data of the long-term monitoring program (14 years) confirmed the feasibility of creating an artificial oyster reef in Yangtze estuary. One or two functional groups were absent from moderate salinity conditions (5.17% - 8.73%), while macrobenthos functional group species richness, abundance, and biomass were greatest under high salinity conditions. The feeding evenness index (jFD) showed that 14 years after construction, the ecosystem health of the artificial oyster reefs were good or high, and was better under high salinity conditions. Ecological health was lower under moderate levels of salinity, possibly due to the absence of some functional groups. Redundancy analysis indicated ecosystem health was associated with changes in salinity and substrate factors. Our study confirmed artificial oyster reef construction is feasible and has a positive effect on estuarine ecosystem health.

Anti-deaminases can inhibit APOBEC3, a component of cytosine base editors. Here Zhanjun Li and colleagues repurposed anti-deaminase proteins derived from viruses to inhibit base editors for use in efficient regulation of base editors’ activity in gene modification and therapeutic applications.

ObjectivesTo improve the performance of course tracking and reduce the course errors of an underactuated surface ship, a heading control method based on a finite-time extended state observer (FTESO) and sliding mode control algorithm is studied. MethodsA pre-filter is proposed to reduce the influence of the large rate of speed change when steering. The time-varying sideslip angle is estimated by FTESO, and course error is amended by the estimated sideslip angle in a timely manner. To simplify the design of the controller, external disturbance and internal uncertainty in the yaw direction are estimated by the observer simultaneously, and compensated for in the controller design. Considering input saturation, a sliding mode control law is designed by combining FTESO and a sliding mode surface with an integral term. The stability of the control system is proven by the Lyapunov theory. Results The simulation results show that the proposed controller reduces course tracking error and makes it converge to zero in a shorter time. ConclusionsThe results of this study can provide references for the course tracking control design of surface ships.

Sparse mobile crowdsensing (SMCS) has emerged as a promising sensing paradigm for urban sensing, leveraging the spatial and temporal correlation among data sensed in distinct sub-areas to cut sensing expenses dramatically. It intelligently selects only a tiny portion of the target regions for sensing and accurately infers the data for the remaining unsensed areas. SMCS confronts numerous challenges, such as sensing cell selection and missing data inference, when compared to mobile crowdsensing. Researchers in recent years have proposed plenty of strategies to solve these challenges. From the perspective of comparing MCS, we aim to provide a comprehensive literature review of recent advances in SMCS in this paper. We begin by going over the preliminary of SMCS and MCS, including their evolution, characteristics, and life-cycle stages. We then go through their common key techniques and recent developments. Furthermore, we give a review of the unique key techniques as well as the most recent advancements. We finally identify existing applications and highlight potential research opportunities for SMCS. Our objective is to provide researchers with a comprehensive understanding of SMCS.

Quinolone (QN) antibiotics are widely used all over the world and have been frequently detected in source water, but the occurrence in tap water and the treatment efficiencies of QNs by drinking-water treatment plants (DWTPs) were rarely reported. In the present study, the occurrence and distribution of six representative QNs in three urban DWTPs of China were investigated. The results showed that the concentrations of total QNs in the three source waters ranged from 26.4 ng/L to 313.8 ng/L and all of the six QNs were detectable with a detection frequency of 100% (4.6 to 121.7 ng/L). Enrofloxacin (ENR) and ofloxacin (OFL) were the dominant species of QNs and accounted for 40.1% to 79.5% of the total QNs. After the treatments, there were still considerable QNs in the finished water (total amounts of 74.9 ng/L to 148.4 ng/L). The adsorbed QNs could be readily treated with the removal of turbidity by DWTPs, but only a part of the dissolved QNs (13.6% to 68.5%) can be removed. This implies that the dissolved QNs were more hazardous in the source water. Pre-oxidation and disinfection could remove 15.8 ± 8.3% and 16.9 ± 10.8% of dissolved QNs, respectively, depending on the chemical structure of QNs and the types of oxidant. Chemical oxidation was more efficient than coagulation-sedimentation and filtration for the treatment of dissolved QNs. Ozone-granular activated carbon filtration may fail to remove dissolved QNs in the actual DWTPs, because of the insufficient dosage of oxidant and the competition effect of natural organic matter. Polycomb repressive complex 2 (PRC2) is responsible for the trimethylation of lysine 27 of histone H3 (H3K27me3)-mediated transcriptional silencing. At present, its biological roles in the devastating rice pathogenic fungus Ustilaginoidea virens remain unclear. In this study, we analyzed the function of a putative PRC2 catalytic subunit UvKmt6. The results showed that disruption of UvKMT6 resulted in reduced growth, conidiation and pathogenicity in U. virens. Furthermore, UvKmt6 is essential for establishment of H3K27me3 modification, which covers 321 genes in the genome. Deletion of UvKMT6 led to transcriptional derepression of 629 genes, 140 of which were occupied with H3K27me3 modification. Consistent with RNA-seq and ChIP-seq analysis, UvKmt6 was further confirmed to participate in the transcriptional repression of genes encoding effectors and genes associated with secondary metabolites production, such as PKSs, NRPSs and Cytochrome P450s. Notably, we found that UvKmt6 is involved in transcriptional repression of oxidative, osmotic, cell wall and nutrient starvation stresses response-related genes. From the perspective of gene expression and phenotype, in addition to the relatively conservative role in fungal development, virulence and production of secondary metabolites, we further reported that UvKmt6-mdediated H3K27me3 plays a critical role in the response to various stresses in U. virens.

Abstract Aims Predicting the risk of malignant arrhythmias (MA) in hospitalized patients with heart failure (HF) is challenging. Machine learning (ML) can handle a large volume of complex data more effectively than traditional statistical methods. This study explored the feasibility of ML methods for predicting the risk of MA in hospitalized HF patients. Methods and results We evaluated the baseline data and MA events of 2794 hospitalized HF patients in the HF cohort in Anhui Province and randomly divided the study population into training and validation sets in a 7:3 ratio. The Lasso‐logistic regression, multivariate adaptive regression splines (MARS), classification and regression tree (CART), random forest (RF), and eXtreme gradient boosting (XGBoost) algorithms were used to construct risk prediction models in the training set, and model performance was verified in the validation set. The area under the receiver operating characteristic curve (AUC) and Brier score were employed to evaluate the discrimination and calibration of the model, respectively. Clinical utility of the Lasso‐logistic regression model was analysed using decision curve analysis (DCA). The median (Q1, Q3) age of the study population was 70 (61, 77) years, and 39.5% were female. MA events occurred in 117 patients (4.2%) during hospitalization. In the training set (n = 1964), the AUC of the XGBoost model was 0.998 [95% confidence interval (CI) 0.997–1.000], which was higher than the other models (all P 0.05], which were higher than that of CART model [AUC: 0.743 (95% CI 0.661–0.824); all P

In this study, starches were isolated from inbred (sweet and waxy) and hybrid (sweet and waxy) sorghum grains. Structural and property differences between (inbred and hybrid) sweet and waxy sorghum starches were evaluated and discussed. The intermediate fraction and amylose content present in hybrid sweet starch were lower than those in inbred sweet starch, while the opposite trend occurred with waxy starch. Furthermore, there was a higher A chain (30.93–35.73% waxy, 13.73–31.81% sweet) and lower B2 + B3 chain (18.04–16.56% waxy, 24.07–17.43% sweet) of amylopectin in hybrid sorghum starch. X-ray diffraction (XRD) and Fourier transform infrared reflection measurements affirm the relative crystalline and ordered structures of both varieties as follows: inbred waxy > hybrid waxy > hybrid sweet > inbred sweet. Small angle X-ray scattering and 13C CP/MAS nuclear magnetic resonance proved that the amylopectin content of waxy starch was positively correlated with lamellar ordering. In contrast, an opposite trend was observed in sweet sorghum starch due to its long B2 + B3 chain content. Furthermore, the relationship between starch granule structure and function was also concluded. These findings could provide a basic theory for the accurate application of existing sorghum varieties precisely.

Film-mulched drip irrigation has become an important strategy in maize cultivation in the semiarid region of Northeast China. Most farmers concentrate nitrogen (N) fertilizer use early in maize growth, which leads to low N use efficiency and large N losses. Therefore, a three-year (2018 to 2020) field experiment was conducted to determine the optimal N management strategy for maize under film mulch with drip irrigation in the semiarid region of Northeast China. The experiment included five treatments with the total amount of N fertilizer (210 kg N ha−1) applied in different proportions at sowing, sixth-leaf (V6), twelfth-leaf (V12), tasseling (VT), and blister (R2) stages of maize growth: N1, 100-0-0-0-0; N2, 50-50-0-0-0; N3, 30-50-20-0-0; N4, 20-30-30-20-0; and N5, 10-20-30-20-20. The control (CK) did not receive N fertilizer. Maize yield, N uptake and use, changes in soil inorganic N content, and N balance were investigated. Compared with the single basal application (N1), split-N applications (N2, N3, N4, N5) increased maize yield from 13.8% to 24.5% by increasing kernel number per ear and 1000-kernel weight and also improved N accumulation from VT to physiological maturity (PM) stages and its contribution to grain N uptake. In addition, compared with N1, split-N applications also decreased N losses by increasing inorganic N contents in the 0–40 cm soil layer and by decreasing N leaching in the 60–200 cm soil layer. Regression analysis demonstrated that N accumulation after the VT stage was positively related with maize yield. Among treatments, N4 had the highest yield, N recovery efficiency, agronomic efficiency, and partial factor productivity, with respective increases of 24.5%, 14.7 percentage point, 11.4 kg kg−1, and 11.4 kg kg−1 compared with those in N1. As a result, N losses were also reduced by 33.7% in N4 compared with those in N1. In conclusion, the split-N management strategy with four N applications under film-mulched drip irrigation has great potential to improve maize yield, increase N use efficiency, and reduce N loss in the semiarid region of Northeast China.

Ensiling legume with cereal is an effective method to ensure the energy rich-feed, but no information is available on the microbial fermentation mechanism of intercropped Lablab purpureus (Lablab) and sweet sorghum in the saline-alkaline region. Therefore, the present study investigated the silage quality and microbial community of intercropped Lablab and sweet sorghum silages grown in the saline-alkaline region with or without inoculation of Lactobacillus plantarum (LP). The experimental treatments were prepared according to the Lablab and sweet sorghum planting patterns: Lablab and sweet sorghum sowing seed ratios were 1:1 (L), 5:1 (M), and 9:1 (H). After harvesting, each mixture was treated with LP or sterilized water (CK), followed by 60 days of fermentation. Results showed that both LP inoculation and intercropping significantly raised the lactic acid (LA) content and decreased the pH value, acetic acid (AA), and ammonia-N in intercropped silages. The LP addition and intercropping also improved the relative feed value by reducing structural carbohydrates. Moreover, LP silages had a greater relative abundance of Lactobacillus than CK silages, and its relative abundance increased with an increased seed-sowing ratio of Lablab in intercropping. LP was the prevalent species in LP silages compared to CK silages, and its relative abundance also increased with an increased seed-sowing ratio of Lablab in intercropping. The genus Lactobacillus was negatively correlated with ammonia-N (R = −0.6, p = 0.02) and AA (R = −0.7, p < 0.01) and positively correlated with LA (R = 0.7, p < 0.01) and crude protein (R = 0.6, p = 0.04). Overall, the intercropped seeding ratios of Lablab and sweet sorghum of ≥ 5:1 with LP inoculation resulted in better fermentation quality and preservation of nutritional components providing theoretical support and guidance for future intercropped protein-rich silage production in the saline-alkaline region.

Recently, a rationally engineered SpCas9 variant (SpCas9-NG) that can recognize a minimal NG protospacer adjacent motif (PAM) was reported to expand the targeting scope in genome editing. However, increased genome-wide off-target mutations with this variant compared with SpCas9 were reported in previous studies. In addition, lower base editing frequencies and higher unintended off-target mutations were also found in Hoxc13-ablated rabbits generated by NG-BE4max in our study. Here, a high-fidelity base editor, NG-HiFi, in comparison to NG-BE4max, showed retention of on-target activity while exhibiting significantly decreased off-target activity in Hoxc13-ablated rabbits. Collectively, the improved specificity and reduced off-target effect of SpCas9-NG assisted in cytidine base editing with the NG-HiFi system, providing a promising tool to precisely model human diseases in rabbits.

Antibiotics are commonly used to prevent and control diseases in aquaculture. However, long-term/overuse of antibiotics not only leaves residues but results in the development of antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs). Antibiotics, ARB, and ARGs are widespread in aquaculture ecosystems. However, their impacts and interaction mechanisms in biotic and abiotic media remain to be clarified. In this paper, we summarized the detection methods, present status, and transfer mechanisms of antibiotics, ARB, and ARGs in water, sediment, and aquaculture organisms. Currently, the dominant methods of detecting antibiotics, ARB, and ARGs are UPLC−MS/MS, 16S rRNA sequencing, and metagenomics, respectively. Tetracyclines, macrolides, fluoroquinolones, and sulfonamides are most frequently detected in aquaculture. Generally, antibiotic concentrations and ARG abundance in sediment are much higher than those in water. Yet, no obvious patterns in the category of antibiotics or ARB are present in organisms or the environment. The key mechanisms of resistance to antibiotics in bacteria include reducing the cell membrane permeability, enhancing antibiotic efflux, and structural changes in antibiotic target proteins. Moreover, horizontal transfer is a major pathway for ARGs transfer, including conjugation, transformation, transduction, and vesiculation. Identifying, quantifying, and summarizing the interactions and transmission mechanisms of antibiotics, ARGs, and ARB would provide useful information for future disease diagnosis and scientific management in aquaculture.

ObjectivesTo address the impact of non-zero sideslip angle and input saturation on the heading control of a surface ship, a method based on a backstepping control algorithm is proposed.Methods The actual sideslip angle is obtained through the relative velocity, and heading angle error is amended using the sideslip angle. A pre-filter is used to minimize the impact of speed change when the heading alters. Hyperbolic tangent and Nussbaum functions are introduced to approximate the input constraint, an adaptive law is designed to estimate approximation errors and disturbances in the yaw direction, and a command filter is proposed to simplify the design of the backstepping controller. The stability of the control system is proved by Lyapunov's theory.ResultsThe simulation results show that the proposed controller can effectively reduce the heading output error of a surface ship while simultaneously maintaining a small control input torque.ConclusionsThe study results can provide reference for the heading control design of surface ships.

Amphibian population declines are closely linked to increasingly serious environmental pollution. Field investigations revealed that perfluorooctane sulfonic acid (PFOS) distribution was detected in 100% of amphibians. In the present study, global transcriptome sequencing was determined on black-spotted frogs to quantify transcript expression levels and the development of an adverse outcome pathway for PFOS. A total of 1441 differentially expressed genes were identified in the PFOS exposure for 21 d, with 645 being downregulated and 796 upregulated. The gene functions and pathways for lipid metabolism, endocrine system, and immune defense were enriched. An adverse outcome pathway has been proposed, including PPAR (peroxisome proliferator-activated receptors) as the molecular initiating events; followed by changes in lipid metabolism, endocrine system, and immune defense; with an end result of liver damage or even population decline. This research provides molecular insight into the toxicity of PFOS. More research about differentially expressed genes is warranted to further provide the underlying mechanism that is altered as a result of PFOS toxicity in organisms.

CRISPR-Cas9-mediated gene knockout and base-editing-associated induction of STOP codons (iSTOP) have been widely used to exterminate the function of a coding gene, while they have been reported to exhibit side effects. In this study, we propose a novel and practical alternative method referred to as CRISPR Start-Loss (CRISPR-SL), which eliminates gene expression by utilizing both adenine base editors (ABEs) and cytidine base editors (CBEs) to disrupt the initiation codon (ATG). CRISPR-SL has been verified to be a feasible strategy on the cellular and embryonic levels (mean editing efficiencies up to 30.67% and 73.50%, respectively) and in two rabbit models mimicking Otc deficiency (Otc gene) and long hair economic traits (Fgf5 gene).

CRISPR-Cas9 and base editors (BEs) systems are poised to become the gene-editing tool of choice in clinical contexts; however, large-fragment deletion was found in Cas9-mediated mutation cells and mice. In this study, by analyzing 16 gene-edited rabbit lines (including 112 rabbits) generated using SpCas9, BEs, xCas9, and xCas9-BEs with long-range PCR genotyping and long-read sequencing by the PacBio platform, we show the extension of thousands of base fragment deletions in single-guide RNA/Cas9 and xCas9 system mutation rabbits, but no deletions were found in BE-induced mutation rabbits. Thus, we first validated that no large-fragment deletion was induced by the BEs system, suggesting that BE systems can be beneficial tools for the further development of highly accurate and secure gene therapy for the clinical treatment of human genetic disorders.

Abstract Background Cytidine base editors (CBEs), composed of a cytidine deaminase fused to Cas9 nickase (nCas9), enable efficient C-to-T conversion in various organisms. However, current base editors can induce unwanted bystander C-to-T conversions when multiple Cs are present in the ~ 5-nucleotide activity window of cytidine deaminase, which negatively affects their precision. Here, we develop a new base editor which significantly reduces unwanted bystander activities. Results We used an engineered human APOBEC3G (eA3G) C-terminal catalytic domain with preferential cytidine-deaminase activity in motifs with a hierarchy CCC>CCC>CC (where the preferentially deaminated C is underlined), to develop an eA3G-BE with distinctive CC context-specificity and reduced generation of bystander mutations. Targeted editing efficiencies of 18.3–58.0% and 54.5–92.2% with excellent CC context-specificity were generated in human cells and rabbit embryos, respectively. In addition, a base editor that can further recognize relaxed NG PAMs is achieved by combining hA3G with an engineered SpCas9-NG variant. The A3G-BEs were used to induce accurate single-base substitutions which led to nonsense mutation with an efficiency of 83–100% and few bystander mutations in Founder (F0) rabbits at Tyr loci. Conclusions These novel base editors with improved precision and CC context-specificity will expand the toolset for precise gene modification in organisms.

In higher plants, vacuolar invertases play essential roles in sugar metabolism, organ development, and sink strength. In sorghum (Sorghum bicolor), two vacuolar invertase genes, SbVIN1 (Sobic. 004G004800) and SbVIN2 (Sobic. 006G160700) have been reported, but their enzymatic properties and functional differences are largely unknown. We combined molecular, biochemical and genomic approaches to investigate their roles in sorghum stem and grain traits. SbVIN1 and SbVIN2 showed different expression levels in internodes, leaves, and panicles, indicating that their importance in each organ was different. In an in vitro sucrose hydrolysis assay, proteins of both genes heterologously expressed in Pichia pastoris displayed similar enzyme properties including the same optimum reaction pH (5) and similar Km for sucroe (49 mmol L−1 and 45 mmol L−1 for SbVIN1 and SbVIN2, respectively). The optimum reaction temperatures of SbVIN1 and SbVIN2 were 45 °C and 65 °C, respectively. SbVIN2 showed higher tolerance to high temperature than SbVIN1. We characterized the sequence variation of these two vacuolar invertase genes in a panel of 216 diverse inbred lines of sweet and grain sorghum and performed gene-based association analysis. SbVIN1 showed significant associations with stem traits including stem length, stem diameter, internode number, stem fresh weight, and Brix, as well as grain traits including hundred-grain weight and grain width. Significantly associated variation sites were mainly in 5′ upstream and intron regions. SbVIN2 only associated with grain width and stem water-soluble carbohydrates (WSCs) content. We conclude that the vacuolar invertase genes SbVIN1 and SbVIN2 are differently associated with stem and grain traits in sorghum. In this paper, a nonlinear adaptive heading controller is developed for an underactuated surface vessel with constrained input and sideslip angle compensation. The controller design is accomplished in a framework of backstepping technique. First, to amend the irrationality of the traditional definition of the desired heading, the desired heading is compensated by the sideslip angle. Considering the actuator physical constrain, a hyperbolic tangent function and a Nussbaum function are introduced to handle the nonlinear part of control input. The error and the disturbance are estimated and compensated by an adaptive control law. In addition, to avoid the complicated calculation of time derivatives of the virtual control, the command filter is introduced to integrate with the control law. It is analysed by the Lyapunov theory that the closed loop system is guaranteed to be uniformly ultimately bounded stability. Finally, the simulation studies illustrate the effectiveness of the proposed control method.

As one of the most popular applications in recent years, cloud storage has been gradually integrated into all walks of life. In the field of communication techniques for the unmanned aerial vehicles (UAVs), UAVs use sensors to upload collected data to cloud servers during various exploration activities, but UAVs can only store and calculate valid information currently collected due to the limited storage and computing performance. In the actual exploration, UAVs need not only to upload complete data to the cloud server, but also to support the data dynamic update efficiently. Moreover, due to security requirements, the privacy of data uploaded by UAVs must be protected. However, the existing auditing schemes for dynamic data and integrity have many problems, such as high computation cost, low efficiency of dynamic update, low privacy and security. For this reason, we propose a public cloud audit scheme that supports dynamic data and privacy protection based on distributed string equality check protocol and Merkle-hash tree multi-level index structure. First of all, a third-party server (TPS) is set between the cloud service provider and users, which complete digital signature, integrity auditing, and data dynamic operations significantly in place of users to reduce the local computing cost. Users then locally upload data which has been encrypted to the TPS. Secondly, to further improve the security of the scheme, TPS implement signature for encrypted data based on the distributed string equality check protocol. By designing authorizations with time constraints, it is guaranteed that only the legitimate TPS with time constraints can operate with cloud servers. Finally, we implement dynamic data operation efficiently based on MHT multi-level index structure. The security proof and performance analysis show that our proposed scheme is safe and effective.

CuO NPs (cupric oxide nanoparticles) are widely used in various fields due to their high electrical conductivity, electronic correlation effect, and special physical property. Notably, CuO NPs have good application prospects in agricultural production because of its antifungal activity to prevent crop diseases. However, the increasing release of CuO NPs into the environment has resulted in a serious threat to the ecosystem, including plants. Previous studies have reported the toxicity of CuO NPs on rice, but little is known about the underlying molecular mechanisms or specific genes involved in the response to CuO NPs. In this study, we found that the rice well-known receptor Chitin Elicitor Receptor Kinase 1 (OsCERK1), which is essential for basal resistance against pathogens, is involved in CuO NPs stress in rice. Knockout of OsCERK1 gene resulted in enhanced tolerance to CuO NPs stress. Furthermore, it was revealed that OsCERK1 reduces the tolerance to CuO NPs stress by regulating the anti-oxidant system and increasing the accumulation of H2O2 in rice. In addition, CuO NPs treatment significantly enhances the basal resistance against M. oryzae which is mediated by OsCERK1. In conclusion, this study demonstrated a dual role of OsCERK1 in response to CuO NPs stress and M. oryzae infection by modulating ROS accumulation, which expands our understanding about the crosstalk between abiotic and biotic stresses.

Perfluorooctanoic acid (PFOA) is frequently detected in the environment and accumulates in amphibians such as black-spotted frogs (Rana nigromaculata) with toxic effects; however, the mechanism underlying this toxicity is unclear. In this study, male black-spotted frogs were exposed to 10 μg/L waterborne PFOA for 21 days. Subsequently, the effect of PFOA exposure on gene expression in liver tissue was investigated using transcriptomic techniques. In total, 754 differentially expressed genes (DEGs; 389 up-regulated and 365 down-regulated) were identified. According to Kyoto Encyclopedia of Genes and Genomes enrichment and Gene Ontology functional enrichment analyses, the DEGs were mainly involved in lipid metabolism, endocrine functions, and immunity. Quantitative real-time polymerase chain reaction analysis of 15 selected DEGs revealed a high correlation (R2 = 0.9917) with the transcriptomic results. These results indicated that the PFOA hepatotoxicity in frogs is mediated mainly by lipid metabolism dysregulation, endocrine system disruption, and immunotoxicity. This study provides insights into the hepatotoxic mechanism of PFOA and other perfluorinated compounds in amphibians.

The Wnt gene family is of ancient origin and is involved in various biological processes. In this study, Wnt4 and Wnt16 were cloned from Daphnia pulex, named DpWnt4 and DpWnt16, respectively. In DpWnt4 cDNA, full-length 1684 bp, the open reading frame was 1122 bp and it encodes a 373 amino acid polypeptide. In DpWnt16 cDNA, full-length 1941 bp, the open reading frame was 1293 bp and it encodes a 430 amino acid polypeptide. The sequence analysis result showed that both DpWnt4 and DpWnt16 sequences contain a Wnt1 domain. Multiple sequence alignment and phylogenetic analysis revealed that DpWnt4 and DpWnt16 were most closely related to arthropods. The expression of DpWnt4 decreased at 0.5 mg/L group and was induced at 2 mg/L, while DpWnt16 was only induced at 2 mg/L nanoplastics group. These results help us understand more about the character of Wnt4 and Wnt16 in crustaceans and how Wnt genes respond to pollutants, especially nanoplastics.

Low temperature is a critical factor restricting the growth and survival of aquatic animals, but research on the mechanism of response to low temperature in Cherax destructor is limited. C. destructor is one of the most important freshwater crustaceans with strong adaptability in Australia, and it has been commercialized gradually in recent years. Here, growth indicators, antioxidant parameters, anti-stress gene expression, and transcriptome sequencing were used on crayfish following 8 weeks of low-temperature acclimation. The results showed that weight gain, length gain, and molting rates decreased as the temperature decreased. The activity of antioxidant enzymes decreased, while the content of antioxidant substances and the expression of anti-stress genes increased. Transcriptome sequencing identified 589 differentially expressed genes, 279 of which were upregulated and 310 downregulated. The gene functions and pathways for endocrine disorders, glucose metabolism, antioxidant defense, and immune responses were identified. In conclusion, although low-temperature acclimation inhibited the basal metabolism and immune ability of crayfish, it also increased the antioxidant substance content and anti-stress-gene expression to protect the organism from low-temperature damage. This study provided molecular insights into the study of low-temperature responses of low-temperature-tolerant crustacean species.

Zinc oxide nanoparticles (ZnO-NPs) are commonly used in various commercial applications, causing toxic effects on organisms and destroying biodiversity, but information about their protective approaches remains unknown. This study aims to evaluate the protective effects of theaflavins (TFs) and epigallocatechin gallate (EGCG) against ZnO-NP-induced cytotoxicity in rat tracheal epithelial (RTE) cells. Herein, RTE cells were exposed to 100 μg/L ZnO-NPs for 12 h, then treated with 0, 10, 100, and 1000 μg/L TFs or EGCG for another 12 h; subsequently, oxidative stress, inflammation, and apoptosis analyses were conducted. Relative to the control groups, TFs and EGCG treatment significantly inhibited the levels of reactive oxygen species and malondialdehyde content. Exposure to 1000 μg/L TFs or EGCG treatment downregulated cytochrome C gene expression levels by 59.10% and 77.27%; Caspase 3 gene expression by 50.03% and 60.01%; Caspase 8 gene expression by 45.11% and 55.57%; and Caspase 9 gene expression by 51.33% and 66.67%, respectively. Meanwhile, interleukin 1β and interleukin 6, tumor necrosis factor-α, and the other inflammatory chemokines such as C-C motif chemokine 2 and C-X-C motif chemokine 8 expression were all gradually rescued after the addition of TFs or EGCG. These results imply that TFs or EGCG possibly ameliorated ZnO-NPs-induced toxicity through antiapoptotic, antioxidant, and anti-inflammatory effects. This study provides novel approaches which mitigate the emerging nanoparticle pollutant toxicity in organisms, which may potentially slow down the destruction of biodiversity.

Global Navigation Satellite System (GNSS) radio occultation (RO) provides plentiful sounding profiles over regions lacking conventional observations. The Gridpoint Statistical Interpolation (GSI) hybrid system for assimilating RO data is integrated in this study with the Model for Prediction Across Scales–Atmosphere (MPAS) to improve tropical cyclone forecasts. After the MPAS-GSI assimilation cycles, dynamical vortex initialization (DVI) that may effectively spin up the initial inner typhoon vortex through cycled model integration is implemented to improve the initial analysis fit to the best track position as well as maximum wind or pressure intensity for Typhoon Nepartak (2016) that moved northwestward toward southern Taiwan. During the cycling assimilation, assimilation with RO data improves the temperature and moisture analysis, and largely reduces the forecast errors compared to those without RO data assimilation. The two RO operators that assimilate local bending angle or refractivity produce similar analyses, but the temperature and moisture increments from bending angle assimilation are slightly larger than those from refractivity assimilation. The MPAS forecasts at 60-15 km resolution show that the typhoon track prediction is improved with RO data, especially using bending angle data. The reduction in track deviations is explained by the wavenumber-one potential vorticity budget for several forecasts associated with the track deflection near southern Taiwan. Assimilation of RO data has fewer impacts on the typhoon intensity forecast compared to the DVI that largely improves the initial and thus forecasted intensity of the typhoon but at the cost of a slightly degraded track. Use of the enhanced 3 km resolution in the typhoon path also further improved the forecasts with and without the DVI. The feasible performance of the MPAS-GSI system with the RO data impact is also illustrated for Typhoon Mitag (2019), that passed around northern Taiwan.
