Background: White matter (WM) fiber tracts in the brainstem communicate with various regions in the cerebrum, cerebellum, and spinal cord. Clinically, small lesions, malformations, or histopathological changes in the brainstem can cause severe neurological disorders. A direct and non-invasive assessment approach could bring valuable information about the intricate anatomical variations of the white matter fiber tracts and nuclei. Although tractography from diffusion tensor imaging has been commonly used to map the WM fiber tracts connectivity, it is difficult to differentiate the complex WM tracts anatomically. Both high field MRI methods and ultrahigh-field MRI methods at 7T and 11.7 T have been used to enhance the contrast of WM fiber tracts. Despite their promising results, it is still challenging to achieve wide clinical adoption at 3T. In this study, we explored a clinically feasible method using a proton density weighted (PDW) 3D gradient echo (GRE) sequence to directly image the WM fiber tracts in the brainstem at 3T in vivo. Methods: We optimized a 3D high resolution, double echo, short TR, PDW GRE sequence on 5 healthy volunteers using a clinical 3T scanner to visualize the complicated anatomy of WM fiber tracts in the brain stem. Tissue properties including T1, proton density and T2* from in vivo quantitative MRI data were used for simulations to determine the optimal flip angle for the sequence. The visualization of multiple WM fiber tracts in the brainstem was assessed qualitatively and quantitatively using relative contrast and contrast-to-noise ratio (CNR). To improve the CNR, the final images were created by averaging over all echoes from two consecutive scans at the optimal flip angle. The results were compared to anatomical atlases and histology sections to identify the major fiber tracts. All the identified major fiber tracts were labeled on axial, sagittal and coronal slices. Results: The WM fiber tracts were found to have distinct hypointense signal throughout the brainstem and most of the major WM fiber tracts, such as the corticospinal tract, medial lemniscus, medial longitudinal fasciculus, and central tegmental tract, in the brainstem up to and including the thalamus were identified in all subjects. Both qualitative and quantitative evaluations showed that the 3° scan offered the best contrast for WM fiber tracts for a TR of 20 ms. The average over the first two echo times and two consecutive 3° scans gave a CNR of 47.8 ± 6.2 for the pyramidal tracts in particular and CNRs values greater than 6.5 ± 2.4 for the rest of the fiber tracts. Conclusions: All the major fiber tracts in the brainstem could be visualized. Given the reasonably short scan time of 10 min at 3T, double echo PDW GRE sequence is a very practical approach for clinical adoption.

Abstract Waste biorefining processes face significant challenges related to the variability of feedstocks. The supply and composition of multiple feedstocks in these processes can be uncertain, making it difficult to achieve economically feasible and sustainable waste valorization for large-scale production. Here, we introduce a reinforcement learning-based framework that aims to control these uncertainties and improve the efficiency of the process. The framework is tested on an anaerobic digestion process and is found to perform better than traditional control strategies. In the short term, it achieves faster target tracking with increased precision and accuracy, while in the long term, it shows adaptive and robust behavior even under additional seasonal supply variability, meeting downstream demand with high probability. This reinforcement learning-based framework offers a promising and scalable solution to address uncertainty issues in real-world biorefining processes. If implemented, this framework could contribute to sustainable waste management practices globally, making waste biorefining processes more economically viable and environmentally friendly.

Mapping the small venous vasculature of the hippocampus in vivo is crucial for understanding how functional changes of hippocampus evolve with age. Oxygen utilization in the hippocampus could serve as a sensitive biomarker for early degenerative changes, surpassing hippocampal tissue atrophy as the main source of information regarding tissue degeneration. Using an ultrahigh field (7T) susceptibility-weighted imaging (SWI) sequence, it is possible to capture oxygen-level dependent contrast of submillimeter-sized vessels. Moreover, the quantitative susceptibility mapping (QSM) results derived from SWI data allow for the simultaneous estimation of venous oxygenation levels, thereby enhancing the understanding of hippocampal function. In this study, we proposed two potential imaging markers in a cohort of 19 healthy volunteers aged between 20 and 74 years. These markers were: 1) hippocampal venous density on SWI images and 2) venous susceptibility (Δχvein) in the hippocampus-associated draining veins (the inferior ventricular veins (IVV) and the basal veins of Rosenthal (BVR) using QSM images). They were chosen specifically to help characterize the oxygen utilization of the human hippocampus and medial temporal lobe (MTL). As part of the analysis, we demonstrated the feasibility of measuring hippocampal venous density and Δχvein in the IVV and BVR at 7T with high spatial resolution (0.25 × 0.25 × 1 mm3). Our results demonstrated the in vivo reconstruction of the hippocampal venous system, providing initial evidence regarding the presence of the venous arch structure within the hippocampus. Furthermore, we evaluated the age effect of the two quantitative estimates and observed a significant increase in Δχvein for the IVV with age (p = 0.006, r2 = 0.369). This may suggest the potential application of Δχvein in IVV as a marker for assessing changes in atrophy-related hippocampal oxygen utilization in normal aging and neurodegenerative diseases such as AD and dementia.

Background: Parkinson's disease (PD) is associated with the loss of neuromelanin (NM) and increased iron in the substantia nigra (SN). Magnetization transfer contrast (MTC) is widely used for NM visualization but has limitations in brain coverage and scan time. This study aimed to develop a new approach called Proton-density Enhanced Neuromelanin Contrast in Low flip angle gradient echo (PENCIL) imaging to visualize NM in the SN. Methods: This study included 30 PD subjects and 50 healthy controls (HCs) scanned at 3T. PENCIL and MTC images were acquired. NM volume in the SN pars compacta (SNpc), normalized image contrast (Cnorm), and contrast-to-noise ratio (CNR) were calculated. The change of NM volume in the SNpc with age was analyzed using the HC data. A group analysis compared differences between PD subjects and HCs. Receiver operating characteristic (ROC) analysis and area under the curve (AUC) calculations were used to evaluate the diagnostic performance of NM volume and CNR in the SNpc. Results: PENCIL provided similar visualization and structural information of NM compared to MTC. In HCs, PENCIL showed higher NM volume in the SNpc than MTC, but this difference was not observed in PD subjects. PENCIL had higher CNR, while MTC had higher Cnorm. Both methods revealed a similar pattern of NM volume in SNpc changes with age. There were no significant differences in AUCs between NM volume in SNpc measured by PENCIL and MTC. Both methods exhibited comparable diagnostic performance in this regard. Conclusions: PENCIL imaging provided improved CNR compared to MTC and showed similar diagnostic performance for differentiating PD subjects from HCs. The major advantage is PENCIL has rapid whole-brain coverage and, when using STAGE imaging, offers a one-stop quantitative assessment of tissue properties.

Global fertilizer production and mismanagement significantly contribute to many harmful environmental impacts, revealing the need for a greater understanding of crop growth and nutrient uptake, which can be used to optimize fertilizer management. This study experimentally adapts first-principles microbial modeling techniques to the hydroponic cultivation of Bibb lettuce (Lactuca sativa) under nitrogen-limited conditions. Monod and Michaelis–Menten’s approaches are applied to predict biomass productivity and nutrient uptake and to evaluate the feasibility of reclaimed wastewater as a nutrient source of nitrogen. Experimental and modeling results reveal significantly different kinetic saturation constants (Ks = 1.331 and Km = 17.887 mg L−1) and a corresponding cell yield strongly dependent on nutrient concentration, producing visually and compositionally distinct tissue between treatments receiving ≤26.2 and ≥41.7 mgN L−1. The resulting Monod model overestimates dry mass predictions during low nutrient conditions, and the collective results support the development of a dynamic Monod curve that is temporally dependent during the plants’ lifecycle. Despite this shortcoming, these results support the feasibility of reclaiming nitrogen from wastewater in hydroponic agriculture, expecting to produce lesser biomass lettuce exhibiting healthy tissue. Furthermore, this study provides a mathematical foundation for agricultural simulations and nutrient management.

IntroductionCognitive impairment (CI) is a common complication of end-stage renal disease (ESRD) that is associated with structural and functional changes in the brain. However, whether a joint structural and functional alteration pattern exists that is related to CI in ESRD is unclear.MethodsIn this study, instead of looking at brain structure and function separately, we aim to investigate the covariant characteristics of both functional and structural aspects. The reuse reclaimed water for irrigation and the application of reclaimed biosolids in agriculture have sparked global concerns regarding the potential contamination of farmland and crops with pharmaceuticals and personal products (PPCPs). This concern is especially relevant to vegetables that are consumed raw by humans. To address this issue, we have developed a straightforward and robust method for concurrently screening of eight antibiotic compounds, including quinolones, tetracyclines, and sulfonamides, in vegetables. The detection and quantification of antibiotics were accomplished using high-performance liquid chromatography-electrospray tandem mass spectrum (HPLC-ESI-MS/MS) in positive mode. We optimized the sample preparation and instrumental operations to enhance the signal peak area of the target analytes. The recoveries of antibiotics in the five tested vegetables ranged from 84 % to 127.5 %, with a relative standard deviation(RSD) below 12.11 %. Additionally, the method exhibits limits of detection (LODs) ranging from 1.834 to 4.97 ng/g dry weight (dw). Following validation, we successfully applied the method to analyze antibiotic accumulation in hydroponic lettuce in a greenhouse. All antibiotics were detected in three different concentration treatment groups (0.1ppm, 1ppm and 3ppm), and the bioconcentration increased with higher antibiotic concentrations spiked to the nutrient solution. Given the various physicochemical properties of the antibiotics used in this study, our work suggests that the developed method offers a simple, reliable and effective means to identify and quantify antibiotic residues in leafy vegetables and other crops that may be impacted by contaminated treated wastewater or biosolids.

Abstract Given that bromine possesses similar properties but extra merits of easily synthesizing and polarizing comparing to homomorphic fluorine and chlorine, it is quite surprising very rare high-performance brominated small molecule acceptors have been reported. This may be caused by undesirable film morphologies stemming from relatively larger steric hindrance and excessive crystallinity of bromides. To maximize the advantages of bromides while circumventing weaknesses, three acceptors (CH20, CH21 and CH22) are constructed with stepwise brominating on central units rather than conventional end groups, thus enhancing intermolecular packing, crystallinity and dielectric constant of them without damaging the favorable intermolecular packing through end groups. Consequently, PM6:CH22-based binary organic solar cells render the highest efficiency of 19.06% for brominated acceptors, more excitingly, a record-breaking efficiency of 15.70% when further thickening active layers to ~500 nm. By exhibiting such a rare high-performance brominated acceptor, our work highlights the great potential for achieving record-breaking organic solar cells through delicately brominating.

Recent research has demonstrated the positive impact of herbal tea consumption on postprandial blood glucose regulation. This study conducts a comparative analysis of aqueous and ethanol extractions on four herbal teas (Mallotus, Cyclocarya, Rubus, and Vine) to assess their phytochemical profiles and functional attributes. Phytochemical contents, antioxidant activities, α-glucosidase inhibitory activities, and chemical compositions are investigated via colorimetric analyses and UPLC-Q-Orbitrap HRMS/MS, respectively. Results indicate that Vine, among the teas studied, exhibits the most pronounced glucose-regulating effects under both extraction methods. While ethanol extractions yield higher phytochemical content overall, the compositions vary. Conversely, aqueous extracts demonstrate unexpectedly potent antioxidant activities and comparable α-glucosidase inhibitory activities to ethanol extracts. Phytochemical contents correlate positively with antioxidant activities and α-glucosidase inhibitory activities. However, antioxidant activities exhibit a weak positive correlation with α-glucosidase inhibitory activities. These findings provide evidence that aqueous extracts from herbal teas contain valuable phytochemical compositions beneficial for antioxidants and individuals with hyperglycemia, suggesting their potential as functional ingredients to enhance the nutritional value of herbal food products.

The goal of this work was to explore the total iron burden of cerebral microbleeds (CMBs) using a semi-automatic quantitative susceptibility mapping and to establish its effect on brain atrophy through the mediating effect of white matter hyperintensities (WMH). A total of 95 community-dwelling people were enrolled. Quantitative susceptibility mapping (QSM) combined with a dynamic programming algorithm (DPA) was used to measure the characteristics of 1309 CMBs. WMH were evaluated according to the Fazekas scale, and brain atrophy was assessed using a 2D linear measurement method. Histogram analysis was used to explore the distribution of CMBs susceptibility, volume, and total iron burden, while a correlation analysis was used to explore the relationship between volume and susceptibility. Stepwise regression analysis was used to analyze the risk factors for CMBs and their contribution to brain atrophy. Mediation analysis was used to explore the interrelationship between CMBs and brain atrophy. We found that the frequency distribution of susceptibility of the CMBs was Gaussian in nature with a mean of 201 ppb and a standard deviation of 84 ppb; however, the volume and total iron burden of CMBs were more Rician in nature. A weak but significant correlation between the susceptibility and volume of CMBs was found (r = -0.113, P < 0.001). The periventricular WMH (PVWMH) was a risk factor for the presence of CMBs (number: β = 0.251, P = 0.014; volume: β = 0.237, P = 0.042; total iron burden: β = 0.238, P = 0.020) and was a risk factor for brain atrophy (third ventricle width: β = 0.325, P = 0.001; Evans's index: β = 0.323, P = 0.001). PVWMH had a significant mediating effect on the correlation between CMBs and brain atrophy. In conclusion, QSM along with the DPA can measure the total iron burden of CMBs. PVWMH might be a risk factor for CMBs and may mediate the effect of CMBs on brain atrophy.

Gastric ulcer (GU) is one of the most prevalent digestive diseases that seriously affects people’s health. Previous studies have demonstrated the anti-GU effect of Ruda-6 (RD-6), a classic formulae of traditional Mongolian medicine. However, the underlying mechanism of RD-6 against GU remains elusive. Thus, we conducted an integrative approach of network analysis, RNA-seq, and in vivo validation experiment to elucidate the therapeutic mechanisms of RD-6 in preventing GU. A network analysis was performed to predict the potential targets of RD-6. Rats were pretreated with RD-6 at different doses for 21 days, followed by GU induction with indomethacin injection. The ulcer index and inhibition rates were calculated, and the levels of inflammatory related factors were determined by ELISA. The gastroprotective mechanism of RD-6 against ulceration was verified by RNA-seq and the key pathway was detected by in vivo validation. As the network analysis predicted, RD-6 exerts anti-GU effects by regulating 75 targets and 160 signaling pathways. Animal experiment results suggested that pretreatment with RD-6 significantly ameliorated the gastric mucosal injury and inflammatory response, as evidenced by a reduced ulcer index, decreased interleukin (IL)-1β, IL-6, and IL-17 levels, and increased prostaglandin E2 (PGE2) levels in the GU model rats induced by indomethacin. RNA-seq data identified four potential hub genes that were primarily involved in the IL-17 signaling pathway. Furthermore, in vivo validation experiment showed that RD-6 inhibited the IL-17 signaling pathway by down-regulating the expression of IL17RA, proto-oncogene C-Fos (FOS), IL1B and prostaglandin-endoperoxide synthase 2 (PTGS2). Taken together, the present study provides evidence that RD-6 could effectively protect against indomethacin-induced GU, which might be attributed to suppressed inflammation. The IL-17 signaling pathway may be one of the crucial mechanisms that mediates the effect of RD-6.

Background: Intestinal pathogens are associated with xenotransplantation tolerance and rejection. However, changes in the gut microbiota in patients who have undergone peripheral nerve xenotransplantation and their association with immune rejection have not yet been reported. Objective: We aimed to explore intestinal microbes and their metabolites at different time points after peripheral nerve transplantation to provide new insight into improving transplant tolerance. Methods: A peripheral nerve xenotransplantation model was constructed by suturing the segmented nerves of Sprague Dawley rats to those of C57 male mice using xenotransplantation nerve bridging. Fecal samples and intestinal contents were collected at three time points: before surgery (Pre group; n = 10), 1 month after transplantation (Pos1 m group; n = 10), and 3 months after transplantation (Pos3 m group; n = 10) for 16S DNA sequencing and nontargeted metabolome detection. Results: Alpha diversity results suggested that species diversity was significantly downregulated after peripheral nerve xenotransplantation. There were six gut flora genera with significantly different expression levels after xenotransplantation: four were downregulated and two were upregulated. A comparison of the Pre vs. Pos1 m groups and the Pos1 m vs. Pos3 m groups revealed that the most significant differentially expressed Kyoto Encyclopedia of Genes and Genomes metabolite pathways were involved in phenylalanine, tyrosine, and tryptophan biosynthesis, as well as histidine metabolism. Metabolites with a strong relationship to the differentially expressed microbial flora were identified. Conclusion: Our study found lower gut microbiome diversity, with increased short-chain fatty acid (SCFA)-producing and sulfate-reducing bacteria at 1 month post peripheral nerve xenotransplantation, and these were decreased at 3 months post-transplantation. The identification of specific bacterial metabolites is essential for recognizing potential diagnostic markers of xenotransplantation rejection or characterizing therapeutic targets to prevent post-transplant infection.

Solar-driven interfacial evaporation for wastewater purification and seawater desalination is perceived an encouraging strategy to simultaneously address water scarcity and the energy crisis. Hydrogel materials can undertake the crucial tasks of water transport and evaporation based on their well-known hydrophilicity and water retention. In the past few years, on account of its merits of low cost, simple preparation method and low energy consumption, hydrogel-based solar energy water purification technology has gradually turned into a research hotspot. This article systematically introduces the latest progress in the field of solar evaporation using hydrogel materials. Three aspects are discussed, including various photothermal materials, different evaporation systems and practical applications. Finally, the challenges in the current development process of solar evaporation technology are analyzed, and the countermeasures are put forward.

MXene, a two-dimensional (2D) nanomaterial with diverse applications, has gained significant attention due to its 2D lamellar structure, abundance of surface groups, and conductivity. Despite various established synthesis methods since its discovery in 2011, MXenes produced through different approaches exhibit variations in structural and physicochemical characteristics, impacting their suitability for environmental application. This study delves into the effect of synthesis conditions on MXene properties and its adsorption capabilities for four commonly prescribed antibiotics. We utilized material characterization techniques to differentiate MXenes synthesized using three prevalent etchants: hydrofluoric acid (HF), mixed acids (HCl/HF), and fluoride salts (LiF/HCl). Our investigation of adsorption performance included isotherm and kinetic analysis, complemented by density functional theory calculations. The results of this research pinpointed LiF/HCl as an efficient etchant, yielding MXene with favorable morphology and surface chemistry. Electrostatic interactions and hydrogen bonding between MXene surface terminations and ionizable moieties of the antibiotic molecules emerge as pivotal factors in adsorption. Specifically, a higher presence of oxygen terminations increases the binding affinities. These findings provide valuable guidance for etchant selection in environmental applications and underscore the potential to tailor MXenes through synthesis conditions to design membranes capable of selectively removing antibiotics and other targeted substances.

Food is abundant in phenolic compounds and phenols are perceived as a rich source of natural antioxidants and vital functional factors. Bound polyphenols are a form of phenolic compounds that exist in the residue from which free polyphenols have been extracted and are combined with other substances through ester bonds, glycosidic bonds, ether glycosidic bonds, etc. As the forefront of human immune defense, the intestine is prone to exogenous and metabolic oxygen radical damage, which triggers intestinal oxidative stress. Intestinal oxidative stress can disrupt the tight junctions between intestinal epithelial cells through various pathways, leading to intestinal epithelial barrier functional impairment and increasing intestinal permeability, which can induce various diseases. Bound polyphenols, as a natural antioxidant, have structural integrity and activity stability in the intestine. These components feature an ameliorative effect on intestinal oxidative stress as well as impaired intestinal barrier function by modulating immune function and regulating the expression of inflammatory factors, antioxidant enzymes and barrier proteins.

Photovoltaic cells are one of the most promising renewable energy sources to address energy and environmental issues. Amongst the many photovoltaic technologies, organic solar cells (OSCs) have numerous advantages, such as low cost, light weight, semi-transparency, and flexibility. This last is a special merit of OSCs, arising due to the intrinsic flexibility of organic active layers. With promising applications in fields such as building-integrated photovoltaics and wearable electronics, flexible OSCs (F-OSCs) have developed rapidly, and significant process has been made in recent years. In this review, we summarize the recent progress in F-OSCs from the perspective of flexible transparent electrodes. In addition, large-area F-OSCs and their potential applications are briefly discussed. Finally, challenges for the further development of F-OSCs are presented.

Background: Impaired cerebral waste clearance (CWC) has been associated with a broad range of both physiological and pathophysiological neurologic conditions.1,2 Because of the unique anatomy of the brain parenchyma, theoretically, in the brain parenchyma, biochemically inert waste such as magnetic resonance imaging (MRI) contrast agents can only be removed through two possible pathways: cerebrospinal fluid (CSF) pathway, and/or vascular pathway. Despite the controversy, there seems to be a solid consensus on the participation of the CSF pathway in CWC.3 In contrast to the CSF system, the current consensus is that the parenchymal vascular system does not participate in CWC. Considering there is a big difference in flow rate between the blood (2 mL/min) and the CSF (3.7 µL/min) and the brain is the most bioactive, energy-consuming organ (20% nutrition for about 5% of body weight) in the body, it is illogical that the brain would rely on the slow CSF circulation for CWC while less bioactive tissues outside the brain require both the fast vascular and slow lymphatic systems to remove waste in a timely manner.4,5 Methods: Superparamagnetic iron oxide–enhanced susceptibility-weighted imaging (SPIO-SWI) and quantitative susceptibility mapping (QSM) methods were used to simultaneously study 7 T MRI signal changes in parenchymal veins, arteries, and their corresponding para-vascular spaces in 26 rats, following intra-cisterna magna (ICM) infusion of different CSF tracers (FeREX, ferumoxytol, Fe-Dextran) to determine the amount of tracer in the artery and vein quantitatively. Results: The parenchymal venous system participated in CSF tracer clearance following ICM infusion of different MRI tracers with different concentrations of iron. Parenchymal venous participation was more obvious when 75 μg iron was injected. In this paper, we develop the cell-average based neural network (CANN) method to solve third order and fifth order Korteweg-de Vries (KdV) type equations. The CANN method is based on the weak or integral formulation of the partial differential equations. A simple feedforward network is forced to learn the cell average difference between two consecutive time steps. One solution trajectory corresponding to a generic initial value is used to generate the data set to train the network parameters, which uniquely determine a one-step explicit finite volume based network method. Once well-trained, the CANN method can be generalized to a suitable family of initial value problems. Comparing with conventional explicit methods, where the time step size is restricted as Δt = O(Δx3) or Δt = O(Δx5), the CANN method is able to evolve the solution forward accurately with a much larger time step size of Δt = O(Δx). A large group of numerical tests are carried out to verify the effectiveness, stability and accuracy of the CANN method. Wave propagation is well resolved with indistinguishable dispersion and dissipation errors. The CANN approximations agree well with the exact solution for long time simulation.

Abstract With practical interest in the future applications of next-generation electronic devices, it is imperative to develop new conductive interconnecting materials appropriate for modern electronic devices to replace traditional rigid solder tin and silver paste of high melting temperature or corrosive solvent requirements. Herein, we design highly stretchable shape memory self-soldering conductive (SMSC) tape with reversible adhesion switched by temperature, which is composed of silver particles encapsulated by shape memory polymer. SMSC tape has perfect shape and conductivity memory property and anti-fatigue ability even under the strain of 90%. It also exhibits an initial conductivity of 2772 S cm−1 and a maximum tensile strain of ~ 100%. The maximum conductivity could be increased to 5446 S cm−1 by decreasing the strain to 17%. Meanwhile, SMSC tape can easily realize a heating induced reversible strong-to-weak adhesion transition for self-soldering circuit. The combination of stable conductivity, excellent shape memory performance, and temperature-switching reversible adhesion enables SMSC tape to serve two functions of electrode and solder simultaneously. This provides a new way for conductive interconnecting materials to meet requirements of modern electronic devices in the future.

Cortex Mori Radicis (CMR) is enriched in various phenolics, this study aimed to estimate the antioxidant effect, enzyme (lipase, α-amylase, α-glucosidase and acetyl-cholinesterase) inhibition and the anti-proliferative effect of the phenolic compounds in CMR. However, the amount of these compounds obtained from CMR is highly dependent on the processing conditions. In this study, the processing parameters of extracting the phenolics from CMR using ultrasonic technique pooled with high-speed shearing extraction (UTPHSE) were optimized. Subsequently, the phenolics from Cortex Mori Radicis (PCMR) were purified using AB-8 macroporous resin, and their chemical analysis, antioxidant, enzyme inhibition, and antiproliferative activities were studied. Based on our findings, the optimal parameters of UTPHSE were: L/S ratio 25.8:1 (mL/g), voltage 81.0 V, ultrasonic temperature 51.8 ℃ and ultrasonic time 289 s. And under the optimal extraction conditions, the extraction rate of the PCMR was 0.531 ± 0.004%. Compared with PCMR, the contents of “total phenolics, flavonoids, flavonols, flavanols and phenolic acids” increased 2.30, 2.67, 2.59, 3.63 and 2.72 times in the purified phenolics from Cortex Mori Radicis (PPCMR), respectively. In addition, PPCMR depicted significant DPPH, ABTS+ and superoxide anion radicals’ scavenging capability, reducing power, ferric ion reducing antioxidant power (FRAP) and remarkable inhibitory activities on “lipase, alpha-amylase, alpha-glucosidase, and the proliferation of HeLa, HepG2 and NCI-H460”. At the same time, the morphological changes of HeLa, HepG2 and NCI-H460 cells suggested that PPCMR could effectively inhibit the proliferation of tumor cells in vitro. Therefore, PPCMR have good potential as natural antioxidants, antilipidemic, hypoglycemic, and antineoplastic agents in functional foods and pharmaceuticals.

Background: Renal cell carcinoma (RCC) is a prevalent malignancy with growing mortality and high metastasis. Ferroptosis has been identified as an essential process in cancer development, but the regulatory mechanism underlying the RCC progression remains obscure. The nanomaterial zinc oxide nanoparticles (ZONs) have presented anti-cancer function. Here, we identified the critical role of ZONs in promoting ferroptosis of RCC cells by regulating miR-27a-3p/YAP axis. Methods: The effect of ZONs on RCC was analyzed by qPCR, Western blot, MTT assays, colony formation assays, Flow cytometry analysis, transwell assays, wound healing assays, iron assays, lipid ROS detection, luciferase reporter gene assays, and tumor xenograft. Results: The treatment of ZONs repressed expression of GPX4 and SLC7A11 and enhanced ROS accumulation and iron/Fe2+ levels in RCC cells. Ferroptosis activator erastin repressed RCC cell viabilities and ZONs further repressed this effect. ZONs inhibited invasion and migration of RCC cells and treatment of ZONs represses RCC cell survival in vitro. ZONs suppressed RCC cell growth in tumorigenicity mouse model. Mechanically, ZONs down-regulated YAP expression by inducing miR-27a-3p, in which YAP overexpression and miR-27a-3p inhibition reverse ZONs -inhibited RCC cell survival in vitro. Discussion: Thus, we concluded that ZONs induced RCC cell ferroptosis to suppress RCC cell survival by targeting miR-27a-3p/YAP axis. The clinical significance of ZONs for the treatment of RCC is required to further study and may benefit the targeted therapy of RCC.

Designing ultra small metal nanoclusters or single atoms with metallic state is a challenge. Here, the authors demonstrate the stabilization of ultra small silver clusters in the nearly-metallic state by oxygen adsorption at high temperature, using in situ spectroscopy and microscope technologies.

Abstract The hydrolysis properties of lipase in castor was evaluated using two different substrate forms (tripalmitic glycerides and trioleic glycerides) to catalyze the reaction under different operational conditions. RcLipase was obtained from castor seeds and results show that RcLipase is a conservative serine lipase with a conserved catalytic center (SDH) and a conserved pentapeptide (GXSXG). This enzyme exhibited the greatest activity and tolerance to chloroform and toluene when it was expressed in Pichia pastoris GS115 at 40 ℃ and pH 8.0. Zn and Cu ions exerted obvious inhibitory effects on the enzyme, and displayed good hydrolytic activity for long-chain natural and synthetic lipids. HPLC analysis showed that this enzyme has 1,3 regioselectivity when glycerol tripalmitate and oleic acid are used as substrates. The fatty acid composition in the reaction product was 21.3% oleic acid and 79.1% sn-2 palmitic acid.

The compression molding performance of a substrate block has a significant effect on the quality and stability of mechanical transplanting. The physical experiment and DEM simulation were combined to evaluate the compression molding behavior of substrate block in this study. A calibration procedure of DEM parameters of peat particles was proposed at first. Then, the above parameters were brought into the contact model of the compression system–particles, and the effect of the loading speed on the compression behavior of the peat substrate block was investigated. The compressive force–displacement curves of the simulated and measured tests were all contained in the initial linear stage and non-linear stiffing stage. The particle number of central sections was higher than side section, and the variable coefficient was greater at higher loading speed. The substrate blocks all expanded after demolding. The higher the loading speed, the greater the expansion in the height’s direction, and the easier it was for cracks to be generated near the bottom. This study will provide a reference for the design of substrate block forming machines.

Background and Objectives: Aucklandiae Radix is a well-known medicinal herb that is often used to treat gastric ulcer, but its molecular mechanism of anti-ulcer action is poorly understood. This research aimed to reveal the potential active components, core targets, and mechanisms of Aucklandiae Radix in treating gastric ulcer by combining network pharmacology and animal experimentation. Materials and Methods: First, a network pharmacology strategy was used to predict the main components, candidate targets, and potential signaling pathways. Molecular docking was then used to confirm the binding affinity between the main components and primary targets. Finally, rats were treated with indomethacin 30 mg/kg to establish a gastric ulcer model. Aucklandiae Radix extract (0.15, 0.3, and 0.6 g/kg) was pre-treated in rats by oral gavage for 14 days, and the protective effect and candidate targets of network pharmacology were validated through morphological observation, pathological staining, and biochemical index detection. Results: A total of eight potential active components and 331 predicted targets were screened from Aucklandiae Radix, 37 of which were common targets with gastric ulcer. According to the component–target network and protein-protein interaction (PPI) network, stigmasterol, mairin, sitosterol, and dehydrocostus lactone were identified as the key components, and RAC-alpha serine/threonine-protein kinase (AKT1), prostaglandin-endoperoxide synthase 2 (PTGS2), interleukin 1 beta (IL1B), caspase-3 (CASP3), and CASP8 were selected as the core targets. Gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment results revealed the pharmacological mechanism of Aucklandiae Radix against gastric ulcer related to many biological processes and pathways, including antibacterial, anti-inflammatory, prostaglandin receptor response, and apoptosis. Molecular docking verification showed that the key components and core targets had good binding affinities. In the in vivo experiments, Aucklandiae Radix notably relieved the gastric ulcer by reducing the levels of tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and myeloperoxidase (MPO) while improving the gastric histopathological features. Conclusion: The overall findings suggest that Aucklandiae Radix treats gastric ulcer with a multi-component, multi-target, and multi-mechanism model.

The hippocampus is a small but complex grey matter structure that plays an important role in spatial and episodic memory and can be affected by a wide range of pathologies including vascular abnormalities. In this work, we introduce the use of Ferumoxytol, an ultra-small superparamagnetic iron oxide (USPIO) agent, to induce susceptibility in the arteries (as well as increase the susceptibility in the veins) to map the hippocampal micro-vasculature and to evaluate the quantitative change in tissue fractional vascular density (FVD), in each of its subfields. A total of 39 healthy subjects (aged 35.4 ± 14.2 years, from 18 to 81 years old) were scanned with a high-resolution (0.22×0.44×1 mm3) dual-echo SWI sequence acquired at four time points during a gradual increase in Ferumoxytol dose (final dose = 4 mg/kg). The volumes of each subfield were obtained automatically from the pre-contrast T1-weighted data. The dynamically acquired SWI data were co-registered and adaptively combined to reduce the blooming artifacts from large vessels, preserving the contrast from smaller vessels. The resultant SWI data were used to segment the hippocampal vasculature and to measure the FVD ((volume occupied by vessels)/(total volume)) for each subfield. The hippocampal fissure, along with the fimbria, granular cell layer of the dentate gyrus and cornu ammonis layers (except for CA1), showed higher micro-vascular FVD than the other parts of hippocampus. The CA1 region exhibited a significant correlation with age (R = −0.37, p 0.05) of the CA1 subfield, which would suggest that vascular degeneration may precede tissue atrophy.

Abstract Objective Sphingolipids are enriched in the nerves. Serine‐palmitoyltransferase (SPT) catalyzes the key step of sphingolipids biosynthesis. Mutations in SPT subunits (SPTLC) lead to the excessive production of neurotoxic deoxysphingolipids (DoxSLs) in patients with Hereditary Sensory Neuropathy Type‐1C (HSN1C). HSN1C is an autosomal dominant peripheral neuropathy characterized by sensory loss and distal muscle weakness. In this study, by leveraging a HSN1C family with a previously reported N177D mutation in SPTLC2, we aim to further define the spectrum of DoxSL species and the peripheral neve pathology of the disease. Methods Next‐generation sequencing along with Sanger confirmation was performed for family members and healthy controls. LC‐MS was used for lipidomic analysis in participants’ plasma. Quantitative magnetic resonance imaging (qMRI) was performed to study sciatic nerve pathologies. Results A heterozygous N177D mutation in SPTLC2 was co‐segregated in individuals with sensory‐motor deficits in the limbs. Nerve conduction studies (NCS) revealed nonuniform slowing of conduction velocities. In line with the NCS, qMRI detected a pattern of nerve changes similar to those in acquired demyelinating polyneuropathies. Additionally, we detected a significant increase in multiple species of deoxysphingoid bases and deoxyceramides in patients’ plasma. Interpretation Mutations in the SPTLC2 cause a demyelinating phenotype resembling those in acquired demyelinating polyneuropathy. The species of increased DoxSLs in HSN1C may be more diverse than originally thought.

Abstract Objective Hereditary Neuropathy with Liability to Pressure Palsies (HNPP) is caused by a heterozygous deletion of peripheral myelin protein‐22 (PMP22) gene resulting in focal sensorimotor deficits. Our lab has identified a disruption of myelin junctions in excessively permeable myelin that impairs action potential propagation. This mechanism is expected to cause fatigue in patients with HNPP. Therefore, the objective was to characterize fatigue in patients with HNPP and determine the relationship of fatigue to nerve pathology, disability, and quality of life. Methods Nine females with HNPP participated in a single visit that included genotyping, nerve conduction studies, neurological exam, quantitative magnetic resonance imaging, and a physical therapy exam incorporating upper and lower extremity function and survey measures of fatigue. This visit was followed by 2 weeks of ecological momentary assessment (wrist‐worn device) that captured fatigue ratings five times per day. Results Participants demonstrated mild neurological impairment (CMTNS: 5.7 ± 2.8), yet reported high fatigue levels (average fatigue intensity over 2 weeks: 5.9 out of 10). Higher fatigue levels were associated with poorer quality of life and more pain. Higher fatigue was associated with significantly greater distal nerve proton density changes on peripheral nerve MRI, which is in line with hyper‐permeable myelin in HNPP. Interpretation Fatigue is common and severe among patients with HNPP whose disabilities are minimal by conventional outcome measures. Therapeutic interventions targeting fatigue have the potential to improve quality of life and may serve as a robust outcome measure to show longitudinal changes for patients with HNPP.

Abstract Background Cell autophagy has been proposed to be involved in drug resistance therapy. However, how the long non-coding RNA (lncRNA) reduces risks of drug resistance in renal cancer (RC) cells needs a thorough inquiry. This study was assigned to probe the effect and mechanism of HOTAIR on sunitinib resistance of RC. Methods Clinical RC tissues and para-carcinoma tissues were obtained to detect the expressions of miR-17-5p, HOTAIR and Beclin1. Sunitinib-resistant cells (786-O-R and ACHN-R) were constructed using parental RC cells (786-O and ACHN). The resistance of 786-O-R and ACHN-R cells to sunitinib was examined. Western blot and qRT-PCR were assayed to obtain the expressions of miR-17-5p, HOTAIR and Beclin1. The effects of HOTAIR knockdown or miR-17-5p overexpression/knockdown on cell autophagy and sunitinib resistance were measured by MDC staining, immunofluorescence and Western blot. The sensitivity of RC cells to sunitinib and change in cell clone formation after sunitinib treatment were assessed by CCK-8 assay and colony formation assay, respectively. The relationships among HOTAIR, miR-17-5p and Beclin1 were verified by dual-luciferase reporter gene and RIP assay. The role of HOTAIR knockdown in sunitinib resistance was verified in nude mice. Results HOTAIR expression in sunitinib-resistant cells is higher than that in parental cells. Knockdown of HOTAIR in sunitinib-resistant cells lead to refrained sunitinib resistance and cell autophagy both in vivo and in vitro. Activation of autophagy could raise resistance to sunitinib in RC cells, while inhibition of autophagy could improve the sensitivity of sunitinib-resistant cells to sunitinib. HOTAIR could compete with miR-17-5p to regulate Beclin1 expression. Knockdown of miR-17-5p in parental cells increases cell resistant to sunitinib, and overexpression of miR-17-5p in sunitinib-resistant cells increases cell sensitive to sunitinib. Conclusion HOTAIR negatively targets miR-17-5p to activate Beclin1-mediated cell autophagy, thereby enhancing sunitinib resistance in RC cells.

Abstract Background The human leukocyte antigen (HLA) gene family plays a key role in the immune response and thus is crucial in many biomedical and clinical settings. Utilizing Sanger sequencing, the golden standard technology for HLA typing enables accurate identification of HLA alleles in high-resolution. However, only the commercial software, such as uTYPE, SBT-Assign, and SBTEngine, and very few open-source tools could be applied to perform HLA typing based on Sanger sequencing. Results We developed a user-friendly, cross-platform and open-source desktop application, known as SOAPTyping, for Sanger-based typing in HLA class I and II alleles. SOAPTyping can produce accurate results with a comprehensible protocol and featured functions. Moreover, SOAPTyping supports a more advanced group-specific sequencing primers (GSSP) module to solve the ambiguous typing results. We used SOAPTyping to analyze 36 samples with known HLA typing from the University of California Los Angeles (UCLA) International HLA DNA Exchange platform and 100 anonymous clinical samples, and the HLA typing results from SOAPTyping are identical to the golden results and 5.5 times faster than commercial software uTYPE, which shows the usability of SOAPTyping. Conclusions We introduce the SOAPTyping as the first open-source and cross-platform HLA typing software with the capability of producing high-resolution HLA typing predictions from Sanger sequence data.

The present study reports a rare primitive neuroectodermal tumor (PNET) of prostate. A 27-year-old male was admitted to Harbin Medical University Cancer Hospital (Harbin, China) for dysuria and dyschezia. Magnetic resonance imaging (MRI) revealed a large mass that may involve the bladder and rectum next to the prostate. Histopathological analysis of biopsy of prostate indicated mesenchymal origin tumor, and immunohistochemistric staining confirmed diagnosis of PNET of prostate. En bloc total pelvic exenteration and double barrel sigmoidostomy were performed. Double stomas in the skin incision were used for fecal and urinary diversion, respectively. Short-term outcome is satisfactory, while long-term efficacy remains to be poor. Clinical features of PNET of prostate should be paid much more attention and radical surgery and adjuvant chemotherapy should be recommended. Rheumatoid arthritis (RA) is a common autoimmune disease characterized by chronic and progressive inflammation of the synovium. Focused ultrasound therapy is an increasingly attractive alternative for treating RA owing to its noninvasiveness; however, it remains unclear which immune subsets respond to ultrasound stimulation. In this study, we showed that spleen-targeted low-frequency pulsed focused ultrasound (LFPFU) effectively improved the severity of arthritis in an arthritis mouse model established in DBA/1J mice. Additionally, we performed in-depth immune profiling of spleen samples from RA mice, RA mice that underwent ultrasound therapy, and healthy controls using mass cytometry along with extensive antibody panels and identified the immune composition of 14 cell populations, including CD4+/CD8+ T cells, B cells, natural killer cells, and dendritic cells. Moreover, multidimensional analysis according to cell-surface markers and phenotypes helped in identifying 4 and 5 cell subpopulations among T and myeloid cells, respectively, with 6 T cell subsets and 3 myeloid cell subsets responsive to ultrasound therapy among the 3 groups. Of these cell subsets, CD8+ T cell subsets showed a unique response to ultrasound stimulation in RA mice. Specifically, CD8+ T cells show a noticeable correlation with the degree of arthritis progression and could serve as an indicator for spleen-focused ultrasound-based therapy. Furthermore, single-cell RNA sequencing of spleen cells revealed the importance of T, B, and myeloid cell populations in the anti-inflammatory pathway. These results elucidated the unique cell subsets and transcriptome of splenic cells responsive to LFPFU and demonstrated the potential of spleen-focused ultrasound stimulation in the treatment of inflammatory diseases.

The growth of field crops needs appropriate soil nutrients. As a basic fertilizer, liquid manure provides biological nutrients for crop growth and increases the content of organic matter in crops. However, improper spraying not only reduces soil fertility but also destroys soil structure. Therefore, the precise control of the amount of liquid manure is of great significance for agricultural production and weight loss. In this study, we first built the model of spraying control, then optimized the BP neural network algorithm through a genetic algorithm. The stability and efficiency of the optimized controller were compared with PID, fuzzy PID and BPNN-PID control. The simulation results show that the optimized algorithm has the shortest response time and lowest relative error. Finally, platform experiments were designed to verify the four control algorithms at four different vehicle speeds. The results show that, compared with other control algorithms, the control algorithm described here has good stability, short response time, small overshoot, and can achieve an accurate fertilizer application effect, providing an optimization scheme for research on the precise application of liquid manure.

Castor (Ricinus communis L.) seed oil is the commercial source of ricinoleate, a valuable raw material used in many industries. Oleoyl-12-hydroxylase (RcFAH12) is a key enzyme in the biosynthesis of ricinoleate, accumulating nearly 90% of the triacylglycerol in castor seeds. Little is known about the transcriptional regulation of RcFAH12. We used rapid amplification of cDNA 5′ ends (5′RACE) to locate the transcription start site (TSS) of RcFAH12, and the sequence of a 2605 bp region, −2506~+99, surrounding the TSS was cloned. We then investigated these regions to promote β-glucuronidase (GUS) expression in transgenic Arabidopsis by the progressive 5′ and 3′ deletions strategies. The GUS staining showed that the GUS accumulation varied in tissues under the control of different deleted fragments of RcFAH12. In addition, the GUS expression driven by the RcFAH12 promoter markedly accumulated in transgenic seeds, which indicated that RcFAH12 might play an important role in the biosynthesis of ricinoleic acid. This study will lay a potential foundation for developing a tissue-specific promoter in oil-seed crops.

Background: Recent emerging evidence has highlighted the potential critical role of cerebrospinal fluid (CSF) in cerebral waste clearance and immunomodulation. It is already very well-established that the central nervous system (CNS) is completely submerged in CSF on a macro-level; but to what extent is this true on a micro-level? Specifically, within the peri-neural and peri-vascular spaces within the CNS parenchyma. Therefore, the objective of this study was to use magnetic resonance imaging (MRI) to simultaneously map the presence of CSF within all peri-neural (cranial and spinal nerves) and peri-vascular spaces in vivo in humans. Four MRI protocols each with five participants were used to image the CSF in the brain and spinal cord. Our findings indicated that all CNS neuro- and vascular-communication channels are surrounded with CSF. In other words, all peri-neural spaces surrounding the cranial and spinal nerves as well as all peri-vascular spaces surrounding MRI-visible vasculature were filled with CSF. These findings suggest that anatomically, substance exchange between the brain parenchyma and outside tissues including lymphatic ones can only occur through CSF pathways and/or vascular pathways, warranting further investigation into its implications in cerebral waste clearance and immunity.

Diagnosing early stage Parkinson's disease (PD) is still a clinical challenge. Previous studies using iron, neuromelanin (NM) or the Nigrosome-1 (N1) sign in the substantia nigra (SN) by themselves have been unable to provide sufficiently high diagnostic performance for these methods to be adopted clinically. Our goal in this study was to extract the NM complex volume, iron content and volume representing the entire SN, and the N1 sign as potential complementary imaging biomarkers using a single 3D magnetization transfer contrast (MTC) gradient echo sequence and to evaluate their diagnostic performance and clinical correlations in early stage PD. A total of 40 early stage idiopathic PD subjects and 40 age- and sex-matched healthy controls (HCs) were imaged at 3T. NM boundaries (representing the SN pars compacta (SNpc) and parabrachial pigmented nucleus) and iron boundaries representing the total SN (SNpc and SN pars reticulata) were determined semi-automatically using a dynamic programming (DP) boundary detection algorithm. Receiver operating characteristic analyses were performed to evaluate the utility of these imaging biomarkers in diagnosing early stage PD. A correlation analysis was used to study the relationship between these imaging measures and the clinical scales. We also introduced the concept of NM and total iron overlap volumes to demonstrate the loss of NM relative to the iron containing SN. Furthermore, all 80 cases were evaluated for the N1 sign independently. The NM and SN volumes were lower while the iron content was higher in the SN for PD subjects compared to HCs. Interestingly, the PD subjects with bilateral loss of the N1 sign had the highest iron content. The area under the curve (AUC) values for the average of both hemispheres for single measures were: .960 for NM complex volume; .788 for total SN volume; .740 for SN iron content and .891 for the N1 sign. Combining NM complex volume with each of the following measures through binary logistic regression led to AUC values for the averaged right and left sides of: .976 for total iron content; .969 for total SN volume, .965 for overlap volume and .983 for the N1 sign. We found a negative correlation between SN volume and UPDRS-III (R2 = .22, p = .002). While the N1 sign performed well, it does not contain any information about iron content or NM quantitatively, therefore, marrying this sign with the NM and iron measures provides a better physiological explanation of what is happening when the N1 sign disappears in PD subjects. In summary, the combination of NM complex volume, SN volume, iron content and the N1 sign as derived from a single MTC sequence provides complementary information for understanding and diagnosing early stage PD.

Hydrogel-based material have been demonstrated promising potential for hemostasis. Herein, we prepared a composite hydrogel (CH-P 40%) by combining pectin and cellulose in ionic liquid. The superficial morphology of the CH-P 40% was explored by SEM; the internal chemical bonds, crystal form and thermal stability were determined via FTIR, XRD and thermogravimetric analysis, respectively. The biocompatibilities of the CH-P 40% hydrogel was evaluated by MTT, flow cytometry, and histological observation with H&E staining. Furthermore, the hemostatic effect was evaluated via the blood clotting index and mouse liver hemostatic model. The results showed that the CH-P 40% hydrogel exhibited a dense network structure and retained its chemical bonds, including the OH, CH, C=O, -CH2, CO, C1-H, and β-glycosidic bonds. Simultaneously, the hydrogel retained the Cellulose I and II crystal structure and favorable thermal stability. Moreover, the proliferation rates of CH-P 40%-treated cells increased (P > 0.05), and there were no pathological lesions in the mouse organs, which suggests favorable biocompatibility. The results showed less bleeding in the hydrogel-treated liver wound within 3 min. Overall, the pectin-cellulose hydrogel is stable and possesses favorable biocompatibility and hemostatic ability, further highlighting that the composite hydrogel has the potential to be rapid hemostatic biomedical material.

Background and Purpose: Multiple Sclerosis (MS) is a progressive, inflammatory, neuro-degenerative disease of the central nervous system (CNS) characterized by a wide range of histopathological features including vascular abnormalities. In this study, an ultra-small superparamagnetic iron oxide (USPIO) contrast agent, Ferumoxytol, was administered to induce an increase in susceptibility for both arteries and veins to help better reveal the cerebral microvasculature. The purpose of this work was to examine the presence of vascular abnormalities and vascular density in MS lesions using high-resolution susceptibility weighted imaging (SWI). Methods: Six subjects with relapsing remitting MS (RRMS, age = 47.3 ± 11.8 years with 3 females and 3 males) and fourteen age-matched healthy controls were scanned at 3 T with SWI acquired before and after the infusion of Ferumoxytol. Composite data was generated by registering the FLAIR data to the high resolution SWI data in order to highlight the vascular information in MS lesions. Both the central vein sign (CVS) and, a new measure, the multiple vessel sign (MVS) were identified, along with any vascular abnormalities, in the lesions on pre- and post-contrast SWI-FLAIR fusion data. The small vessel density within the periventricular normal-appearing white matter (NAWM) and the periventricular lesions were compared for all subjects. Results: Averaged across two independent raters, a total of 530 lesions were identified across all patients. The total number of lesions with vascularity on pre- and post-contrast data were 287 and 488, respectively. The lesions with abnormal vascular behavior were broken up into following categories: small lesions appearing only at the vessel boundary; dilated vessels within the lesions; and developmental venous angiomas. Summary: Ribonucleoside monophosphates (rNMPs) represent the most common non-standard nucleotides found in the genome of cells. The distribution of rNMPs in DNA has been studied only in limited genomes. Using the ribose-seq protocol and the Ribose-Map bioinformatics toolkit, we reveal the distribution of rNMPs incorporated into the whole genome of a photosynthetic unicellular green alga, Chlamydomonas reinhardtii. We discovered a disproportionate incorporation of adenosine in the mitochondrial and chloroplast DNA, in contrast to the nuclear DNA, relative to the corresponding nucleotide content of these C. reinhardtii organelle genomes. Our results demonstrate that the rNMP content in the DNA of the algal organelles reflects an elevated ATP level present in the algal cells. We reveal specific biases and patterns in rNMP distributions in the algal mitochondrial, chloroplast, and nuclear DNA. Moreover, we identified the C. reinhardtii orthologous genes for all three subunits of the RNase H2 enzyme using GeneMark-EP + gene finder.

Drought represents the most important limiting factor controlling performance of subtropical plants, leading to reduced growth and die-offs. However, the mechanisms underlying growth and physiological responses of subtropical woody plants to different types of drought are poorly understood. Here, potted seedlings of a typical evergreen shrub species Ardisia lindleyana D. Dietr. in subtropical China, were exposed to three water regimes: well watered (WW), fast drought (FD) and slow drought (SD) until death. Leaf gas exchange and water potential were measured over the experimental period. Dry mass production and allocation, leaf nitrogen and carbon isotopic composition, and non-structural carbohydrates (NSC) were measured at mortality. Results showed that growth, leaf water potential and photosynthesis were significantly reduced in both drought regimes. FD induced significant increase of root soluble sugars, but promoted significant depletion of leaf and root starch, finally displaying depletion of root NSC. By contrast, SD led to significant depletion of stem and root starch, whereas the accumulation of soluble sugars in stem and root was significantly higher than that of WW, thereby resulting in no significant change of NSC. Neither FD nor SD has affected leaf carbon isotopic composition or nitrogen concentration at mortality. Our results demonstrate the divergent effects of two different drought treatments on carbon and water economy of A. lindleyana, which may have implications for understanding the mortality mechanism of understory plants in subtropical forests in China.

Ion exchange membranes (IEMs), as a part of the reverse electrodialysis (RED) system, play an important role in salinity gradient power (SGP) generation. Structure optimization of IEMs is critical to increase the power production by RED. In this work, metal organic framework (MOF)-derived nanoporous carbons (hollow zeolitic imidazolate framework (ZIF)-derived nanoporous carbons, HZCs) were incorporated in a sulfonated poly (2, 6-dimethyl-1,4-phenylene oxide) (sPPO) membrane to prepare an organic−inorganic nanocomposite cation exchange membrane (CEM). Physicochemical properties, electrochemical properties, and power generation of the synthesized nanocomposite membranes with different HZCs loading were characterized. The results show that the incorporated HZCs could tailor the microstructure of the membrane matrix, providing a superior performance of the nanocomposite membrane. With a HZCs loading of 1.0 wt.%, the nanocomposite membrane possessed the highest permselectivity of 77.61% and the lowest area resistance of 0.42 Ω·cm2, along with a super gross power density of 0.45 W/m2, which was 87.5% (about 1.87 times) higher than that of the blank sPPO membrane. Therefore, incorporating of an appropriate amount of HZCs in the ion-exchange membrane can improve the performance of the membrane, providing a promising method to increase the power generation of the RED system.

We performed ultrasound-assisted extraction coupled with natural deep eutectic solvents (NADES) to achieve the green and efficient preparation of flavonoid extract from Ampelopsis grossedentata leaves. We then evaluated its antioxidant and antiproliferative activities. A NADES consisting of choline chloride and glucose at a molar ratio of 4:1 with 20% water was determined to be the most suitable solvent. The optimal extraction conditions were: a liquid-to-solid ratio of 30 mL/g, an ultrasonication power of 490 W, and an ultrasonication time of 6.5 min. The actual flavonoid yield was 83.93%, which was close to the predicted yield. Further, 86.75% of the flavonoids were recovered by adding the same volume of phosphate buffer saline (100 mM, pH of 7.0) to the extract solution. Although the chemical antioxidant activities of the flavonoid extract were slightly inferior to those of dihydromyricetin, the flavonoid extract could still effectively inhibit the proliferation of human breast MDA-MB-231 cells by inducing cell apoptosis, retarding the cell cycle, changing the mitochondrial membrane potential and scavenging intracellular reactive oxygen species (ROS). The obtained results can provide a reference in the development of plant-derived functional foods.

PurposeTo develop a method to reconstruct quantitative susceptibility mapping (QSM) from multi-echo, multi-flip angle data collected using strategically acquired gradient echo (STAGE) imaging.MethodsThe proposed QSM reconstruction algorithm, referred to as “structurally constrained Susceptibility Weighted Imaging and Mapping” scSWIM, performs an ℓ1 and ℓ2 regularization-based reconstruction in a single step. The unique contrast of the T1 weighted enhanced (T1WE) image derived from STAGE imaging was used to extract reliable geometry constraints to protect the basal ganglia from over-smoothing. The multi-echo multi-flip angle data were used for improving the contrast-to-noise ratio in QSM through a weighted averaging scheme. The measured susceptibility values from scSWIM for both simulated and in vivo data were compared to the: original susceptibility model (for simulated data only), the multi orientation COSMOS (for in vivo data only), truncated k-space division (TKD), iterative susceptibility weighted imaging and mapping (iSWIM), and morphology enabled dipole inversion (MEDI) algorithms. Goodness of fit was quantified by measuring the root mean squared error (RMSE) and structural similarity index (SSIM). Additionally, scSWIM was assessed in ten healthy subjects.ResultsThe unique contrast and tissue boundaries from T1WE and iSWIM enable the accurate definition of edges of high susceptibility regions. For the simulated brain model without the addition of microbleeds and calcium, the RMSE was best at 5.21ppb for scSWIM and 8.74ppb for MEDI thanks to the reduced streaking artifacts. However, by adding the microbleeds and calcium, MEDI’s performance dropped to 47.53ppb while scSWIM performance remained the same. The SSIM was highest for scSWIM (0.90) and then MEDI (0.80). The deviation from the expected susceptibility in deep gray matter structures for simulated data relative to the model (and for the in vivo data relative to COSMOS) as measured by the slope was lowest for scSWIM + 1%(−1%); MEDI + 2%(−11%) and then iSWIM −5%(−10%). Finally, scSWIM measurements in the basal ganglia of healthy subjects were in agreement with literature.ConclusionThis study shows that using a data fidelity term and structural constraints results in reduced noise and streaking artifacts while preserving structural details. Furthermore, the use of STAGE imaging with multi-echo and multi-flip data helps to improve the signal-to-noise ratio in QSM data and yields less artifacts.
