Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES The present work aimed to investigate the potential of plants used empirically in agriculture, as well as evaluate different extractive methods, carry out the characterization of the obtained extracts and analyze their allelopathic potential. In the first experimental stage, the yield and chemical composition of the root and fruit matrices of Solanum viarum and flowers, leaves and stalks of Senecio brasiliensis were evaluated, using different extraction techniques: Soxhlet, maceration, supercritical fluids extraction (SFE), ultrasound-assisted extraction (UAE), pressurized liquid extraction (PLE) and microwave hydrodiffusion and gravity (MHG). For SFE, the solvent used was carbon dioxide with pressure values set between 150 and 250 bar and temperature values between 40°C and 60°C. In UAE, a hydroalcoholic solution (60% water/40% ethanol) was used as solvent and the effects of ultrasound intensity (17–85 W/cm²) and pulse cycle (0.5–1.0) in the extraction yields of matrices were evaluated. For PLE, ethanol, water and their mixtures were used as solvents at a pressure of 10 MPa and temperatures between 30°C and 60°C, with ethanol concentration of 70 to 100% (v/v). For MHG, water and ethanol were used as solvent in a proportion of 60 ml ethanol and 40 ml water, under atmospheric pressure and 400 W of power. For comparison, conventional extractions with Soxhlet and maceration were performed. All extracts obtained were analyzed for chemical composition in a gas chromatography system coupled to a mass spectrometer. In the second step, the allelopathic potential of the extracts obtained by ultrasound under the condition of 85 W/cm² power and 1 pulse cycle for the matrices of two species under study was evaluated, at a concentration of 100 mg/ml, using as a solvent ethanol. The fungicidal activity was tested in fungi Fusarium graminearum and Sclerotinia sclerotiorum and the herbicidal action was tested in pre-emergence of Cucumis sativus seeds. The results of this thesis demonstrate that the different extraction techniques are efficient technologies for extraction of compounds from S. viarum and S. brasiliensis, the matrix extracts are a potential source of bioactive compounds and both species studied show a great antifungal and herbicide potential. O presente trabalho teve como objetivo investigar o potencial de plantas utilizadas de forma empírica na agricultura, bem como avaliar diferentes métodos extrativos, realizar a caracterização dos extratos obtidos e analisar o seu potencial alelopático. Na primeira etapa experimental foram avaliados os rendimentos e a composição química das matrizes raízes e frutos de Solanum viarum e flores, folhas e caules de Senecio brasiliensis, utilizando diferentes técnicas de extração: Soxhlet, maceração, extração com fluidos supercríticos (EFS), extração assistida por ultrassom (EUS), líquidos pressurizados (LP) e hidrodifusão por micro-ondas e gravidade (MHG). Para a EFS, o solvente utilizado foi o dióxido de carbono com valores de pressão fixados entre 150 e 250 bar e os valores da temperatura entre 40°C e 60°C. Nas extrações com EUS, uma solução hidroalcoólica (60% água/40% etanol (v/v) foi utilizada como solvente e os efeitos da intensidade do ultrassom (17 a 85 Wcm−²) e do ciclo de pulso (0,5–1,0) nos rendimentos da extração das matrizes foram avaliados. Para as extrações com LP, etanol, água e suas misturas foram utilizados como solventes na pressão de 10 MPa em temperaturas entre 30°C e 60°C, com concentração de etanol de 70 a 100% (v/v). Para as extrações com hidrodifusão por micro-ondas e gravidade (MHG) foi utilizado como solvente água e etanol em uma proporção de 60 ml etanol e 40 ml de água, à pressão atmosférica e sob 400 W de potência. Ressalva-se que para comparação foram realizadas extrações convencionais utilizando Soxhlet e maceração. Todos os extratos obtidos foram analisados quanto a constituição química em um sistema de cromatografia gasosa acoplado a um espectometro de massa. Na 2ª etapa do trabalho, foi avaliado o potencial alelopático dos extratos obtidos por ultrassom na condição de 85 Wcm−² de potência e 1,0 de ciclo de pulso para as matrizes das duas espécies em estudo, a uma concentração de 100 mg/mL, utilizando como solvente etanol. A atividade fungicida foi testada nos fungos Fusarium graminearum e Sclerotinia sclerotiorum e a ação herbicida foi testada em pré emergência em sementes de Cucumis sativus. Os resultados desta Tese demonstram que as diferentes técnicas de extração são tecnologias eficientes para a extração de compostos de S. viarum e S. brasiliensis, os extratos das matrizes vegetais são uma fonte potencial de compostos bioativos e ambas as espécies estudadas mostram um grande potencial antifúngicos e herbicida.
