Abstract

In the summer of 2012, during a Pulsar Search Collaboratory workshop, two high-school students discovered J1930$-$1852, a pulsar in a double neutron star (DNS) system. Most DNS systems are characterized by short orbital periods, rapid spin periods and eccentric orbits. However, J1930$-$1852 has the longest spin period ($P_{\rm spin}\sim$185 ms) and orbital period ($P_{\rm b}\sim$45 days) yet measured among known, recycled pulsars in DNS systems, implying a shorter than average and/or inefficient recycling period before its companion went supernova. We measure the relativistic advance of periastron for J1930$-$1852, $\dot{\omega}=0.00078$(4) deg/yr, which implies a total mass (M$_{\rm{tot}}=2.59$(4) M$_{\odot}$) consistent with other DNS systems. The $2\sigma$ constraints on M$_{\rm{tot}}$ place limits on the pulsar and companion masses ($m_{\rm p}<1.32$ M$_{\odot}$ and $m_{\rm c}>1.30$ M$_{\odot}$ respectively). 19. Granulomatosis with Polyangiitis at Age 13 in an American Indian/Native American Patient

Abstract

Granulomatosis with polyangiitis (GPA), previously known as Wegener’s granulomatosis, is a rare small vessel vasculitis that is a part of the ANCA-associated family. First distinctly identified by Fredrick Wegener in 1936, GPA has had little light shed on the direct pathophysiology for the development of the disease. Classically the disease presents in Caucasian males during the fifth to seventh decades of life [1]. GPA can present with a wide array of symptoms and therefore the diagnosis can be easily missed. Unfortunately, without proper prompt treatment the disease can progress causing irreversible organ damage leading to chronic, relapsing dysfunction and possible death. Given this, early recognition and treatment of the disease is essential. This becomes difficult when given an atypical patient presentation compounded with nondescript symptoms. In this case-presentation, we review an atypical presentation of GPA from a rural rheumatology clinic in the United States with the intention of expanding the current limited literature. Through thorough literature-search we were able to find reports of a 12 year old female that was diagnosed with GPA, and a young 8 -year old American Indian/Native American (AI/NA) female diagnosed with microscopic polyangiitis, however, to our knowledge this is the youngest known AI/NA patient to be diagnosed with GPA at 13 years of age [2,3].

Abstract

Transcriptomic, proteomic and immune repertoire profiling reveals distinct peripheral features of MIS-C and pediatric COVID-19, including elevated soluble spike protein levels, more pronounced type II IFN-dependent gene expression and a higher B cell mutation rate in patients with MIS-C.Pediatric Coronavirus Disease 2019 (pCOVID-19) is rarely severe; however, a minority of children infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) might develop multisystem inflammatory syndrome in children (MIS-C), with substantial morbidity. In this longitudinal multi-institutional study, we applied multi-omics (analysis of soluble biomarkers, proteomics, single-cell gene expression and immune repertoire analysis) to profile children with COVID-19 (n = 110) and MIS-C (n = 76), along with pediatric healthy controls (pHCs; n = 76). pCOVID-19 was characterized by robust type I interferon (IFN) responses, whereas prominent type II IFN-dependent and NF-kappa B-dependent signatures, matrisome activation and increased levels of circulating spike protein were detected in MIS-C, with no correlation with SARS-CoV-2 PCR status around the time of admission. Transient expansion of TRBV11-2 T cell clonotypes in MIS-C was associated with signatures of inflammation and T cell activation. The association of MIS-C with the combination of HLA A*02, B*35 and C*04 alleles suggests genetic susceptibility. MIS-C B cells showed higher mutation load than pCOVID-19 and pHC. These results identify distinct immunopathological signatures in pCOVID-19 and MIS-C that might help better define the pathophysiology of these disorders and guide therapy.

25. EcAMSat: Effect of Space-Flight on Antibiotic Resistance of a Pathogenic Bacterium and its Genetic Basis

Abstract

Human immune response is compromised in space and incidence of urinary tract infections (UTI) in astronauts has been reported. We have found that the causative agent of UTI, the uropathogenic Escherichia coli, becomes more resistant to gentamicin (Gm), which is commonly used to treat this disease, under modeled microgravity conditions (MMG), the increase being controlled by the stress response master regulator, σs. While the wild type bacterium becomes virtually invincible under MMG, the strain missing this sigma factor barely survives. We report here preparatory ground work for testing this finding in space flight on a nanosatellite. We have shown that the effect of Gm treatment on culture viability is directly correlated to increased Alamar Blue (AB) reduction; we have identified conditions to keep the experimental elements - the bacterial cultures, Gm, and AB - in a state of viability and potency to permit successful spaceflight experimentation given the necessary constraints. Spaceflight kinetics of AB reduction will be transmitted from the satellite via telemetry. The PharmaSat hardware previously used for space experimentation with yeast was modified to permit studies with bacteria by reducing the filter pore size and increasing fluidics volume to enable more fluid exchanges. Several verification tests have been run using the nanosatellite's flight software and prototype hardware. Cells were grown to stationary phase to induce the σs-controlled stress resistance and treated with Gm. Without Gm, the mutant took longer than the wild type to reduce the AB; this time difference increased almost 8 fold at 55 μg/mL Gm concentration. Thus, using flight hardware the mutant shows similarly increased sensitivity to Gm compared to the wild type to that found in our pilot microtiter plate experiments. Previous inflight experiments have given contradictory results concerning bacterial antibiotic resistance; none has yet explored the involvement of specific genes in this phenomenon. Abstract

The design and performance evaluation of an airbag system capable of decelerating masses in the range of hundreds to thousands of kilograms with impact velocities in the range of tens to hundreds of kilometers per hour is presented. First, a simplified incompressible flow analysis of the airbag is utilized to derive the orifice venting area corresponding to the ideal deceleration for a given impact velocity and package mass. Second, testing with a small-scale model found three distinct control intervals during the deceleration. Finally, a full-scale airbag system was constructed and data is presented on the deceleration, deceleration force, deceleration velocity, airbag stopping power, and overall performance. The deceleration was experimentally, optimized for a single impact velocity and package mass and an approximate correction factor was developed to predict the actual air venting required for each of the three control intervals in order to achieve the optimum deceleration for any impact velocity and package mass.
