Abstract

This paper introduces a novel approach for high-quality deepfake detection called Localized Artifact Attention Network (LAA-Net). Existing methods for high-quality deepfake detection are mainly based on a supervised binary classifier coupled with an implicit attention mechanism. As a result, they do not generalize well to unseen manipulations. To handle this issue, two main contributions are made. First, an explicit attention mechanism within a multi-task learning framework is proposed. By combining heatmap-based and self-consistency attention strategies, LAA-Net is forced to focus on a few small artifact-prone vulnerable regions. Second, an Enhanced Feature Pyramid Network (E-FPN) is proposed as a simple and effective mechanism for spreading discriminative low-level features into the final feature output, with the advantage of limiting redundancy. Experiments performed on several benchmarks show the superiority of our approach in terms of Area Under the Curve (AUC) and Average Precision (AP). Abstract

The use of Deep Learning (DL) algorithms has improved the performance of vision-based space applications in recent years. However, generating large amounts of annotated data for training these DL algorithms has proven challenging. While synthetically generated images can be used, the DL models trained on synthetic data are often susceptible to performance degradation, when tested in real-world environments. In this context, the Interdisciplinary Center of Security, Reliability and Trust (SnT) at the University of Luxembourg has developed the 'SnT Zero-G Lab', for training and validating vision-based space algorithms in conditions emulating real-world space environments. An important aspect of the SnT Zero-G Lab development was the equipment selection. From the lessons learned during the lab development, this article presents a systematic approach combining market survey and experimental analyses for equipment selection. In particular, the article focus on the image acquisition equipment in a space lab: background materials, cameras and illumination lamps. The results from the experiment analyses show that the market survey complimented by experimental analyses is required for effective equipment selection in a space lab development project.

Abstract

Overexpression of cytokine receptor-like factor 2 (CRLF2) resulting from its genomic rearrangement is the most frequent genetic alteration found in Philadelphia chromosome-like (Ph-like) B-cell acute lymphoblastic leukemia (B-ALL), a high-risk leukemia. Detection of CRLF2 expression by multiparameter flow cytometry has been proposed as a screening tool for the identification of Ph-like B-ALL. However, the prognostic relevance of flow cytometric expression of CRLF2 in pediatric B-ALL is not very clear. Additionally, its association with common copy number alterations (CNA) has not been studied in detail. Hence, in this study, we prospectively evaluated the flow cytometric expression of CRLF2 in 256 pediatric B-ALL patients and determined its association with molecular features such as common CNAs detected using Multiplex ligation-dependent probe amplification and mutations in CRLF2, JAK2 and IL7RA genes. Further, its association with clinicopathological features including patient outcome was assessed. We found that 8.59% (22/256) pediatric B-ALL patients were CRLF2-positive at diagnosis. Among CNAs, CRLF2 positivity was associated with presence of PAX5 alteration (P=0.041). JAK2 and IL-7R mutations were found in 9% and 13.6% CRLF2-positive patients, respectively. IGH::CRLF2 or P2RY8::CRLF2 fusions were each found in 1/22 individuals. CRLF2-positive patients were found to have inferior overall (hazard ratio (HR) =4.39, P=0.006) and event free survival (HR=2.62, P=0.045), independent to other clinical features. Furthermore, concomitant CNA of IKZF1 in CRLF2 positive patients was associated with a greater hazard for poor overall and event free survival, compared to patients without these alterations or presence of any one of them. Our findings demonstrate that the surface CRLF2 expression in association with IKZF1 copy number alteration can be used to risk stratify pediatric B-ALL patients.

Abstract

Background Objectives Methods Results Conclusion Genomic variations associated with dystonia in Asian Indians remain largely unknown.To identify genomic alterations associated with dystonia in the Asian Indian population using next generation sequencing approaches.From September 2018 to December 2023, we enrolled 745 individuals including probands with dystonia and family members, in the Indian Movement Disorder Registry and Biobank. Clinical and demographic data were captured on a REDCap platform. We performed whole exome sequencing (WES) on DNA specimens obtained from 267 individuals with isolated, combined or complex dystonia. Variants were classified according to joint guidelines of American College of Medical Genetics and Genomics (ACMG) and Association of Molecular Pathology (AMP).The mean age of the WES cohort was 33.8 ± 16.2 years, and mean age at onset (AAO) of dystonia was 25.6 ± 17.7 years. 62.2% had isolated dystonia, 7.9% combined and 29.2% had complex phenotypes. WES identified pathogenic/ likely pathogenic variants in 54 patients (20.2%) including 14 novel variants in known dystonia genes. Variants in THAP1 were most common followed by PANK2, GLB1, PLA2G6, TOR1A, ANO3, VPS16, SGCE, SPG7, FTL and other genes. Multifocal/generalized distribution of dystonia [OR: 4.1; 95% CI 1.4–12.2, P = 0.011] and family history [OR: 4.3; 95% CI 2.1–8.9, P < 0.001] were associated with positive yield on WES.THAP1 was the most frequent dystonia associated gene in this cohort.
