Cardiac ischemia–reperfusion injury (IRI) occurs when blood flow is restored to the myocardium after a period of ischemia, leading to oxidative stress and subsequent myocardial cell damage, primarily due to the accumulation of reactive oxygen species (ROS). In our previous research, we identified that miR-25 is significantly overexpressed in pressure overload-induced heart failure, and its inhibition improves cardiac function by restoring the expression of SERCA2a, a key protein involved in calcium regulation. In this study, we aimed to investigate the role of miR-25 in the context of ischemia–reperfusion injury. We found that miR-25 was markedly upregulated under hypoxic conditions in both in vitro and in vivo models. Through in silico analysis, we identified Sestrin3 (SESN3), an antioxidant protein known for its protective effects against oxidative stress, as a novel target of miR-25. Based on these findings, we hypothesized that inhibiting miR-25 would restore Sestrin3 expression, thereby reducing ROS-induced myocardial cell damage and improving cardiac function. To test this hypothesis, we employed two model systems: a hypoxia/reoxygenation (H/R) stress model using H9c2 myoblasts and a surgically induced ischemia–reperfusion injury mouse model. In our previous study, circ_015343 was found to inhibit the viability and proliferation of ovine mammary epithelial cells (OMECs) and the expression levels of milk fat synthesis marker genes, but the regulatory mechanism underlying the processes is still unclear. Accordingly in this study, the target relationships between circ_015343 with miR‐25 and between miR‐25 with insulin induced gene 1 (INSIG1) were verified, and the functions of miR‐25 and INSIG1 were investigated in OMECs. The dual‐luciferase reporter assay revealed that miR‐25 mimic remarkably decreased the luciferase activity of circ_015343 in HEK293T cells cotransfected with a wild‐type vector, while it did not change the activity of circ_015343 in HEK293T cells cotransfected with a mutant vector. These suggest that cic_015343 can adsorb and bind miR‐25. The miR‐25 increased the viability and proliferation of OMECs, and the content of triglycerides in OMECs. In addition, INSIG1 was found to be a target gene of miR‐25 using a dual‐luciferase reporter assay. Overexpression of INSIG1 decreased the viability, proliferation, and level of triglycerides of OMECs. In contrast, the inhibition of INSIG1 in expression had the opposite effect on activities and triglycerides of OMECs with overexpressed INSIG1. A rescue experiment revealed that circ_015343 alleviated the inhibitory effect of miR‐25 on the mRNA and protein abundance of INSIG1. These results indicate that circ_015343 sponges miR‐25 to inhibit the activities and content of triglycerides of OMECs by upregulating the expression of INSIG1 in OMECs. In the present study, we prepared new sixteen different derivatives. The first series were prepared (methylene)bis(2-(thiophen-2-yl)-1H-indole) derivatives which have (indole and thiophene rings) by excellent yield from the reaction (2 mmol) 2-(thiophen-2-yl)-1H-indole and (1 mmol) from aldehyde. The second series were synthesized (2-(thiophen-2-yl)-1H-indol-3-yl) methyl) aniline derivatives at a relatively low yield from multicomponent reaction of three components 2-(thiophen-2-yl)-1H-indole, N-methylaniline and desired aldehydes. The anticancer effect of the newly synthesized derivatives was determined against different cancers, colon, lung, breast and skin. The counter screening was done against normal Epithelial cells (RPE-1). The effect on cell cycle and mechanisms underlying of the antitumor effect were also studied. All new compounds were initially tested at a single dose of 100 μg/ml against this panel of 5 human tumor cell lines indicated that the compounds under investigation exhibit selective cytotoxicity against HCT-116 cell line and compounds (4g, 4a, 4c) showed potent anticancer activity against HCT-116 cell line with the inhibitory concentration IC50 values were, 7.1±0.07, 10.5± 0.07 and 11.9± 0.05 μΜ/ml respectively. Also, the active derivatives caused cell cycle arrest at the S and G2/M phase with significant(p < 0.0001) increase in the expression levels of tumor suppressors miR-30C, and miR-107 and a tremendous decrease in oncogenic miR-25, IL-6 and C-Myc levels. It is to conclude that the anticancer activity could be through direct interaction with tumor cell DNA like S-phase-dependent chemotherapy drugs. Which can interact with DNA or block DNA synthesis such as doxorubicin, cisplatin, or 5-fluorouracil and which were highly effective in killing the cancer cells. This data ensures the efficiency of the 3 analogues on inducing cell cycle arrest and preventing cancer cell growth. Idiopathic intellectual disability (IID) encompasses the cases of intellectual disability (ID) without a known cause and represents approximately 50% of all cases. Neural progenitor cells (NPCs) from the olfactory neuroepithelium (NEO) contain the same information as the cells found in the brain, but they are more accessible. Some miRNAs have been identified and associated with ID of known etiology. However, in idiopathic ID, the effect of miRNAs is poorly understood. The aim of this study was to determine the miRNAs regulating the expression of mRNAs that may be involved in development of IID. Expression profiles were obtained using NPC–NEO cells from IID patients and healthy controls by microarray. A total of 796 miRNAs and 28,869 mRNAs were analyzed. Several miRNAs were overexpressed in the IID patients compared to controls. miR-25 had the greatest expression. In silico analysis showed that ROBO2 was the target for miR-25, with the highest specificity and being the most down-regulated. In vitro assay showed an increase of miR-25 expression induced a decrease in ROBO2 expression. In neurodevelopment, ROBO2 plays a crucial role in episodic learning and memory, so its down-regulation, caused by miR-25, could have a fundamental role in the intellectual disability that, until now, has been considered idiopathic. Dystrophic cardiomyopathy is a significant feature of Duchenne muscular dystrophy (DMD). Increased cardiomyocyte cytosolic calcium (Ca2+) and interstitial fibrosis are major pathophysiological hallmarks that ultimately result in cardiac dysfunction. MicroRNA-25 (miR-25) has been identified as a suppressor of both sarcoplasmic reticulum calcium ATPase 2a (SERCA2a) and mothers against decapentaplegic homolog-7 (Smad7) proteins. In this study, we created a gene transfer using an miR-25 tough decoy (TuD) RNA inhibitor delivered via recombinant adeno-associated virus serotype 9 (AAV9) to evaluate the effect of miR-25 inhibition on cardiac and skeletal muscle function in aged dystrophin/utrophin haploinsufficient mice mdx/utrn (+/−), a validated transgenic murine model of DMD. We found that the intravenous delivery of AAV9 miR-25 TuD resulted in strong and stable inhibition of cardiac miR-25 levels, together with the restoration of SERCA2a and Smad7 expression. This was associated with the amelioration of cardiomyocyte interstitial fibrosis as well as recovered cardiac function. Furthermore, the direct quadricep intramuscular injection of AAV9 miR-25 TuD significantly restored skeletal muscle Smad7 expression, reduced tissue fibrosis, and enhanced skeletal muscle performance in mdx/utrn (+/−) mice. These results imply that miR-25 TuD gene transfer may be a novel therapeutic approach to restore cardiomyocyte Ca2+ homeostasis and abrogate tissue fibrosis in DMD.

Breast tumors show a complex structure and are highly heterogeneous. The study of cytokines, which exert great influence on tumor cells, and microRNAs, which, along with their influence on the proliferation and migration of neoplastic cells, may affect the work of cytokines, will contribute to a deeper understanding of pathological processes occurring in breast cancer. The aim of our work was to analyze the relationship of cytokine production with expression of miR-181a and miR-25in patients with invasive breast carcinoma of a non-specific type (IBC NST) with various molecular subtypes.Patients with IBC NST were divided into five subgroups according to the molecular genetics subtype of the tumor classified by immunohistochemical analysis of estrogen receptor (ER), progesterone (PR), epidermal growth factor 2 (HER2) and proliferation marker Ki-67. Using enzyme immunoassay, the concentration of 14 cytokines was determined in the supernatants of immunocompetent blood cells and tumors: IL-2, IL-6, IL-8, IL-10, IL-17, IL-18, IL-1β, IL-1ra, TNFα, IFNγ, G-CSF, GM-CSF, VEGF and MCP-1. The expression of miR-181a and miR-25 microRNAs isolated from the patients’ blood serum was evaluated using digital droplet polymerase chain reaction (ddPCR).In the luminal A subtype, cytokine concentrations and expression of miR-181a and miR-25 are significantly lower compared to other subtypes. Patients with the luminal B HER2-negative subtype were characterized by significantly increased expression of both studied microRNAs, especially when compared with the luminal A subtype. At the same time, patients with a triple negative molecular subtype, on the contrary, were characterized by high concentrations of cytokines in the supernatants of tumor samples and blood cells compared to other subtypes. In the general group of patients with IBC NST, direct correlations were found between the expression of both studied microRNAs and the concentration of vascular endothelial growth factor (VEGF) in the supernatant of tumor samples, which may presume mutual interactions existing between miR-181a and miR-25, and the process of angiogenesis in the tumor.The levels of cytokines in blood supernatants and tumors in invasive breast carcinoma may vary, depending on distinct molecular subtypes of the tumor. Moreover, they also have direct links with the levels of miR-181a and miR-25 in blood serum. Particularly noteworthy were the results of measuring the cytokines and microRNAs concentrations in luminal A, luminal B HER2-negative and triple negative molecular subtypes.

Circulating miR-181а and miR-25, which reflect regulation of the expression of carcinogenesis-related genes, were assayed in patients with invasive carcinoma of no specific type (ICNT) or benign breast diseases (BBDs) and in subjects without pathologies of the mammary gland (controls). miR-181а expression level proved to be higher compared to control in patients with fibroadenoma and adenosis with low, but not high, risk of malignant transformation, as well as in patients with luminal HER2-negative type B (Lum B HER2–), HER2-positive type (HER2+), and triple-negative breast cancer (TNBC) than in the controls and luminal-type (Lum A) breast cancer. MiR-25 expression level prevailed in patients with Lum B HER2- compared to control, Lum A, and TNBC patients compared to Lum A. Thus, miR-181а and miR-25 expression levels may be risk indicators of malignant transformation in some patients with BBD, whereas in patients with ICNT, these levels reflect pathological processes of different directions within the tumor. Significance of the observations was statistically verified using one-way ANOVA with a significance level of P<0.05. Annexin V-FITC/PI binding assay was utilized to measure cellular apoptosis. Results: The rate of cell apoptosis was significantly higher in MCF-7 cells compared to MDA-MB-231 cells at doses of 4Gy and 8Gy (P=0.013 and P=0.004, respectively). RNA expression analysis showed that circ-HIPK3 was increased in the MDA-MB-231 cell line compared to the MCF-7 cell line after exposure to 8Gy for 48 hours. Expression of circ-PVT1 was found to be higher in MDA-MB-231 cells compared to MCF-7 cells after exposure to 8Gy for 24 hours, likewise after exposure to 4Gy and 8Gy for 48 hours. After exposing 8Gy, expression of miR-25 was increased in MDA-MB-231 cells compared to MCF-7 cells at 24 and 48 hours. After exposing 8Gy dose, expression of miR-149 was increased in MCF-7 cells compared to MDA-MB-231 cells at 24 and 48 hours. Conclusion: circ-HIPK3, circ-PVT1, and miR-25 played crucial roles in the mechanisms of radioresistance in breast cancer. Additionally, miR-149 was involved in regulating cellular radiosensitivity. Objective: To study the spatiotemporal expression profiles of miR-25 in the Chinese perch(Siniperca chuatsi), to predict its targeting core Circadian clock genes, and to explore its possible biological significance. Methods: Real-time fluorescence quantitative PCR was used to analyze the expression characteristics of miR-25 in various tissues and embryos at different stages of Siniperca chuatsi, and TargetScan was used to predict its targeted core clock genes. Results: The expression level of miR-25 was higher in the red muscle and heart (P < 0.05), while it was lower in the spleen, kidney, intestine, and liver of the Chinese perch. However, the expression level difference in these four tissues was not statistically significant (P > 0.05). The expression level of miR-25 was higher in the phase 2 of cell cycle in embryos (P < 0.05), and it gradually decreased as the embryos developed. Target gene prediction results showed that the 3’UTR region of the rhythmic genes Arntl2 and Nr1d2b mRNA contained target sites for miR-25. Conclusion: miR-25 exhibited maternal expression during embryonic development and may have important regulatory significance in the red muscle and heart of Chinese perch. Additionally, miR-25 may regulate the circadian rhythm by targeting the expression of Arntl2 and Nr1d2b, providing a reference for further research on the physiological effects of miR-25 and the regulation of circadian rhythm to prevent related diseases. At present times, various kinds of literature have suggested the miR-25 acts as an oncogene in various types of human malignancies and until now, very less work has been performed pertaining to the role of miR-25 in esopharyngeal cancer. This study was performed to confirm that miR-25 is overexpressed in esophageal squamous cell carcinoma (ESCC) tumor tissue as a prognostic biomarker and to clarify the mechanism of miR-25. The expression levels of miR-25 and BTG2 were detected in esophageal squamous cell carcinoma tumor tissue. A stably knocked-down miR-25 cell line (miR-25KD) was established in esophageal squamous cell carcinoma cell lines. Moreover, a CCK-8 assay was performed for determining the role of miR-25 in proliferation. The Transwell assays were organized to detect metastasis. Later, a gene profiling study was carried out to identify the gene expression pertaining to tumor progression. The expression of miR-25 in the esophageal cancer tissues was much higher compared with that in paracarcinoma tissues (6.42±4.28 VS 3.36±2.63, p<0.001). A high level of miR-25 was identified to be correlated with postoperative metastasis (χ2=8.187, p =0.004). BTG2 levels were significantly lower in tumor tissues (3.24±2.79) than those in adjacent non-tumor tissues (1.96±1.56 VS 2.64±1.41, p<0.001). Negative signs of BTG2 were also associated with postoperative metastasis (χ2=7.766, p=0.005). Besides, BTG2-negative cancer tissues are often accompanied by increased miR-25 expression levels (χ2=18.379, p<0.001). Patients with high miR-25 levels were found with worse overall survival (OS) (χ2=6.906, p=0.009) and metastasis-free survival (MFS) (χ2=4.991, p=0.025). Patients with positive BTG2 had better OS (χ2=12.917, p <0.001) and MFS (χ2=14.173, p<0.001). Knockdown of miR-25 helped to inhibit the proliferation and metastatic ability of esophageal cancer cells. Also, MiR-25 inhibits the expression of BTG2 directly. Results also show that miR-25 also helps to suppress the expression of vimentin and increase the expressions of E-cadherin and BTG2. Cardiac hypertrophy is an adaptive response to various pathological insults, including hypertension. However, sustained hypertrophy can cause impaired calcium regulation, cardiac dysfunction, and remodeling, accompanied by cardiac fibrosis. Our previous study identified miR-25 as a regulator of SERCA2a, and found that the inhibition of miR-25 improved cardiac function and reduced fibrosis by restoring SERCA2a expression in a murine heart failure model. However, the precise mechanism underlying the reduction in fibrosis following miR-25 inhibition remains unclear. Therefore, we postulate that miR-25 may have additional targets that contribute to regulating cardiac fibrosis. Using in silico analysis, Krüppel-like factor 4 (KLF4) was identified as an additional target of miR-25. Further experiments confirmed that KLF4 was directly targeted by miR-25 and that its expression was reduced by long-term treatment with Angiotensin II, a major hypertrophic inducer. Subsequently, treatment with an miR-25 inhibitor alleviated the cardiac dysfunction, fibrosis, and inflammation induced by Angiotensin II (Ang II). These findings indicate that inhibiting miR-25 not only enhances calcium cycling and cardiac function via SERCA2a restoration but also reduces fibrosis by restoring KLF4 expression. Mesenchymal stem cells (MSCs) have demonstrated promising therapeutic potential in the treatment of type 1 diabetes mellitus (T1DM); however, the underlying mechanism remains unclear. The primary pathological mechanism of T1DM involves activated T cells infiltrating the pancreas, leading to islet inflammation and the destruction of β-cells. However, the question of whether exosomes derived from MSCs can suppress the migration of T cells to the pancreas in the context of T1DM remains unresolved. In this study, we observed that miR-25 was highly expressed in MSCs exosomes and associated with signaling pathways related to cell migration. In vitro assay, we synthesized a miR-25 mimic and transiently transfected it into activated T cells, which revealed that miR-25 can effectively reduce the expression of CXCR3. Additionally, according to the in vivo T1DM mouse model, we found that there was a significant increase in miR-25 levels in T1DM mice treated with MSCs and the number of T cells decreased. Overall, our findings suggest that MSCs exosomes containing miR-25 can impede the infiltration of activated T cells into the pancreas in T1DM by repressing CXCR3 expression in these cells. The enzyme-free amplification technique using the Hybridization Chain Reaction (HCR) is gaining traction for its efficiency in miRNA analysis. Conventional HCR (C-HCR) with hairpin probes faces challenges due to enzymatic degradation in body fluids, leading to potential false-positive results. This study addresses the critical need for a more reliable method that resists enzymatic breakdown and improves diagnostic accuracy for detecting miRNA related to ischemic stroke. We have developed a novel DNA tetrahedral nanostructures-mediated HCR (DTN-HCR) platform for the precise detection of microRNA-25 (miR-25), a biomarker for ischemic stroke. Incorporating two unique DNA tetrahedral nanostructures with embedded hairpin structures (DTN-HP1 and DTN-HP2), this platform activates upon miR-25 binding, initiating a robust DTN-HCR reaction. This reaction forms extensive DNA tetrahedron clusters that significantly boost the fluorescence signal, enabling detection thresholds as low as 5.4 pM. The method showcases exceptional specificity by distinguishing target miRNA from close analogues and maintains structural integrity against DNase I and fetal bovine serum (FBS), verified through polyacrylamide gel electrophoresis (PAGE). It successfully differentiates ischemic stroke patients from healthy controls by analyzing peripheral blood-derived miRNAs. This study concludes that the DTN-HCR platform substantially enhances the specificity and stability of miRNA detection, marking a significant advancement in non-enzymatic miRNA analysis techniques. Abstract Background Mitochondrial calcium uniporter (MCU) is an important ion channel regulating calcium transport across the mitochondrial membrane. Calcium signaling, particularly via the Ca2+/NFATc1 pathway, has been identified as an important mediator of the osteoclast differentiation that leads to osteolysis around implants. The present study aimed to investigate whether down-regulation of MCU using microRNA-25 (miR-25) mimics could reduce osteoclast differentiation induced upon exposure to titanium (Ti) particles. Methods Ti particles were prepared. Osteoclast differentiation of RAW264.7 cells was induced by adding Ti particles and determined by TRAP staining. Calcium oscillation was determined using a dual-wavelength technique. After exposure of the cells in each group to Ti particles or control medium for 5 days, relative MCU and NFATc1 mRNA expression levels were determined by RT-qPCR. Vitiligo is an autoimmune skin disease characterized by presence of pale patchy areas of depigmentation. MicroRNAs (miRNAs) are important regulators of gene expression and play significant roles in diverse biological and pathological processes. Accumulating evidence has shown that miRNAs were differentially expressed in skin lesions and peripheral blood mononuclear cells of patients with vitiligo. In particular, miRNAs are significantly correlated with the development and progression of vitiligo. The abundance of some miRNAs in serum was also correlated with the vitiligo lesion severity, indicating that miRNAs might serve as prognostic biomarkers. Importantly, the direct involvement of miRNAs in the pathogenesis of vitiligo has been demonstrated. For example, increased expression of miR-25 contributes to vitiligo through promoting the dysfunction and oxidative stress-induced destruction of melanocytes. However, there are limited studies on the function and mechanism of deregulated miRNAs in vitiligo. Further studies are required to establish clinical applications of miRNAs for vitiligo. More in-depth investigations of miRNAs are needed for the understanding of the pathogenesis of vitiligo and the development of novel therapeutic targets. This present review summarizes the current literature on the deregulation and pathogenic roles of miRNAs in vitiligo. We also highlight the potential clinical applications of miRNAs in patients with vitiligo.

Purpose: Pancreatic β-cell failure is a central hallmark of the pathogenesis of diabetes mellitus; however, the molecular basis underlying chronic inflammation-caused β-cell failure remains unclear. This study reported here specifically assessed the association between miR-25/miR-92b family and β-cell failure in diabetes. Methods: IL-1β and two additional ER stress activators, palmitate and tunicamycin were applied to evaluate the expression level miR-25 by Taqman® RT-PCR. Glucose- and potassium-stimulated insulin secretion assays were performed to assess β-cell function. Dual-luciferase activity, and western blotting assays were utilized for miR-25 target gene verification. CCK-8 and TUNEL staining were used to evaluate β-cell viability and apoptosis. Results: miRNA ChIP identified the increased level of miR-25 in INS-1 cells by IL-1β treatment. Expression levels of miR-25 were significantly upregulated with the treatment of IL-1β, palmitate or tunicamycin in both INS-1 cells and human islets. Ectopic elevation of miR-25 recapitulated most featured β-cell defects caused by IL-1β, including inhibition of insulin biosynthesis and increased β-cell apoptosis. These detrimental effects of miR-25 relied on its seed sequence recognition and repressed expression of its target genes Neurod1 and Mcl1. The miR-25/NEUROD1 axis reduced insulin biosynthesis via transcriptional regulation of β-cell specific genes. The miR-25/MCL1 axis caused β-cell apoptosis in a CASPASE-3/PARP1-dependent manner. Comparable impairments were generated by miR-92b and miR-25, emphasizing the redundant biological roles of miRNA family members with the same seed sequence. Objective To explore the effect of miR-25 on invasion and migration of esophageal cancer and its potential ability as a biomarker for the diagnosis of esophageal cancer. Methods (1) The expression levels of miR-25 in cancer tissues and adjacent tissues of 54 patients with esophageal cancer were detected by fluorescence quantitative PCR (qPCR). (2) Human esophageal cancer cells EC109 were divided into the miR-25 mimic group, the NC mimic group, the miR-25 inhibitor group and the NC inhibitor group. After transfection of the corresponding sequences, miR-25 overexpression and interference efficiency were detected by qPCR. Transwell assay was used to detect the effect of overexpression or silencing of miR-25 on the invasion and migration of EC109 cells. Targetscan database predicted the target gene of miR-25 and selected the target gene salt-induced kinase 1 (SIK1). Western blot assay and double luciferase report experiment identified the targeting relationship between silencing and overexpression of miR-25 and SIK1. (3) EC109 cells were divided into the pcDNA 3.1 group, the SIK1 overexpression group and the miR-25 + SIK1 overexpression group. Transwell assay was used to detect the effect of miR-25 targeting SIK1 on the invasion and migration of EC109 cells. (4) The plasma exosomes of patients with esophageal cancer (54 cases) and healthy controls (54 cases) were extracted, the relative expression of miR-25 in plasma exosomes was compared between the two groups. The correlation between miR-25 in tissues and plasma exosomes of patients with esophageal cancer was analyzed. Results (1) The relative expression level of miR-25 was higher in esophageal carcinoma tissues than that in adjacent tissues. (2) After overexpression of miR-25, the invasion and migration ability of EC109 cells increased, while after silencing miR-25 expression, the invasion and migration ability of EC109 cells decreased. The Targetscan database predicted that SIK1 was the target gene of miR-25. Western blot assay showed that the expression of SIK1 protein decreased after miR-25 overexpression. On the contrary, the protein expression of SIK1 increased after knocking down the expression of miR-25. (3) Transwell experiment showed that compared with the pcDNA 3.1 group, the number of invasion and migration of EC109 cells decreased in SIK1 overexpression group, while the number of invasion and migration of EC109 cells increased in the miR-25 and SIK1 overexpression group compared with the SIK1 overexpression group. (4) The expression of miR-25 in plasma exosomes of esophageal cancer was higher than that in healthy controls, and the expression of miR-25 in esophageal cancer was positively correlated with that in plasma exosomes. Conclusion miR-25 may promote the invasion and migration of esophageal cancer through targeting the regulation of SIK1, and miR-25 has the potential as a biomarker for the early detection of esophageal cancer. Background: Mitochondrial calcium uniporter (MCU) is an important ion channel regulating calcium transport across the mitochondrial membrane. Calcium signaling, particularly via the Ca2+/NFATc1 pathway, has been identified as an important mediator of the osteoclast differentiation that leads to osteolysis around implants. The present study aimed to investigate whether down-regulation of MCU using microRNA-25 (miR-25) mimics could reduce osteoclast differentiation induced upon exposure to titanium (Ti) particles. Methods: Ti particles were prepared. Osteoclast differentiation of RAW264.7 cells was induced by adding Ti particles and determined by TRAP staining. Calcium oscillation was determined using a dual-wavelength technique. After exposure of the cells in each group to Ti particles or control medium for 5 days, relative MCU and NFATc1 mRNA expression levels were determined by RT-qPCR. MCU and NFATc1 protein expression was determined by western blotting. NFATc1 activation was determined by immunofluorescence staining. Comparisons among multiple groups were conducted using one-way analysis of variance followed by Tukey test, and differences were considered significant if p < 0.05. Results: MCU expression was reduced in response to miR-25 overexpression during the process of RAW 264.7 cell differentiation induced by Ti particles. Furthermore, osteoclast formation was inhibited, as evidenced by the low amplitude of calcium ion oscillation, reduced NFATc1 activation, and decreased mRNA and protein expression levels of nuclear factor-κB p65 and calmodulin kinases II/IV. Conclusions: Regulation of MCU expression can impact osteoclast differentiation, and the underlying mechanism likely involves the Ca2+/NFATc1 signal pathway. Abstract Background This study aimed to investigate the role of lncRNA MAGI2-AS3 in non-small cell lung cancer (NSCLC). Methods Expression levels of MAGI2-AS3 and RECK mRNA in two types of tissues (non-tumor and NCSLC) were measured by qPCR. To further investigate the interaction between MAGI2-AS3 and RECK, MAGI2-AS3 and RECK expression vectors were transfected into H1993 cells. Results We found that MAGI2-AS3 and RECK were upregulated and positively correlated in NSCLC. In NSCLC cells, MAGI2-AS3 overexpression led to upregulated RECK. Bioinformatics analysis showed that MAGI2-AS3 may bind miR-25, which can directly target RECK. In NSCLC cells, miR-25 overexpression led to downregulated RECK and attenuated the effects of MAGI2-AS3 overexpression, while MAGI2-AS3 and miR-25 failed to affect each other. Cell invasion and migration analysis showed decreased NSCLC cell invasion and migration rates after MAGI2-AS3 and RECK overexpression. MiR-25 showed opposite role and reduced the effects of MAGI2-AS3 overexpression. Conclusion Therefore, MAGI2-AS3 may sponge miR-25 to upregulate RECK, thereby inhibiting NSCLC cell invasion and migration. BACKGROUND: Studies have found that the down-regulation of miR-25 is related to the excessive activation of nuclear factor of activated T cells c1 (NFATc1), and NFATc1 is the core transcription factor in osteoclasts differentiation and maturation, suggesting that the expression level of miR-25 can regulate osteoclasts differentiation. OBJECTIVE: To explore the effect of miR-25 overexpression on titanium (Ti) particles-induced osteoclasts differentiation and investigate the signaling pathways. METHODS: RAW264.7 cell line was cultured in vitro. Cell counting kit-8 was used to determine the effect of Ti particles on cell proliferation. miR-25 mimics were transfected into RAW264.7 cells, and cultured with Ti particles to induce osteoclasts differentiation. Immunofluorescence was used to detect the activation status of NFATc1, TRAP staining and counting were adopted to verify the differentiation of osteoclasts, and real-time PCR was employed to detect and analyze the differential expression of NFATc1, CaMKII, and CaMKIV mRNAs. RESULTS AND CONCLUSION: The low concentration of Ti particles (0.1 g/L) had no significant effect on cell proliferation. Compared with the negative control group, there was less production of osteoclasts (TRAP positive) in the miR-25 overexpression group (P < 0.05). After intracellular transfection with miR-25 mimic, immunofluorescence detection showed that the activation of NFATc1 decreased and the fluorescence intensity decreased. The expression of signaling pathway molecules NFATc1, CaMKII, and CaMKIV mRNAs decreased significantly after miR-25 overexpression (P < 0.05). These findings indicate that miR-25 overexpression can inhibit the differentiation of Ti particles-induced osteoclasts. This process may be achieved through the Ca2+/NFATc1 pathway. HDAC3 plays important role in regulating memory and plasticity of neurons. We studied the role of miR-25 against HDAC3-induced neuronal injury in acute ischemic stroke. Subjects reported for acute stroke were included in the study. The rat model of middle cerebral artery occlusion was developed and received miR-25 agomir and antagomir treatments via intra-cerebroventricular injection. The brain tissues were processed and neuronal cells were isolated and submitted to oxygen glucose derivation-mediated injury. mRNA levels were studied by RT-PCR and protein levels by ELISA method. TUNEL and nuclear protein staining was done to find the ischemic area. Behavioral studies were carried out by Morris water maze test and beam balance test. Results suggested a significant increase in plasma miR-25 levels observed in acute ischemic stroke subjects. The levels of miR-25 were increased in the tissues of infarcted area of brain tissues of rats. However, the expression of miR-25 decreased in neuronal cells but increased in supernatant post-oxygen glucose deprivation. The treatment of miR-25 agomir decreased the infarct volume and apoptosis of neurons in MCAO rats, and it also improved the plasticity of neurons and axons, down-regulated ATXN3 and HDAC3 levels, and improved acetyl-H3K9 expression. In vitro outcomes suggested that miR-25 inhibited HDAC3 neurons and modulated the oxygen glucose deprivation injury in neurons. The treatment of RGFP966 increased the acetyl-H3K9 levels and prevented the miR-25 antagomir-induced injury. The study suggested miR-25 as an important predicting biomarker in acute ischemia injury. Abstract Background Acute myeloid leukemia (AML) pertains to a hematologic malignancy with heterogeneous therapeutic responses. Improvements in risk stratification in AML patients are warranted. MicroRNAs have been associated with the pathogenesis of AML. Methods To examine the prognostic value of miR-25, 162 cases with de novo AML were classified into two groups according to different treatment regimens. Results In the chemotherapy group, cases with upregulated miR-25 expression showed relatively longer overall survival (OS; P = 0.0086) and event-free survival (EFS; P = 0.019). Multivariable analyses revealed that miR-25 upregulation is an independent predictor for extended OS (HR = 0.556, P = 0.015) and EFS (HR = 0.598, P = 0.03). In addition, allogeneic hematopoietic stem cell transplantation (allo-HSCT) circumvented the poor prognosis that was related to miR-25 downregulation with chemotherapy. The expression level pattern of miR-25 coincided with AML differentiation and proliferation, which included HOXA and HOXB cluster members, as well as the HOX cofactor MEIS1. The MYH9 gene was identified as a direct target of miR-25. Conclusions The miR-25 levels are correlated with prognosis in AML independently of other powerful molecular markers. The expression of miR-25 may contribute to the selection of the optimal treatment regimen between chemotherapy and allo-HCST for AML patients.

The homeostatic oxygen sensing system (HOSS) optimizes systemic oxygen delivery. Specialized tissues utilize a conserved mitochondrial sensor, often involving NDUFS2 in complex I of the mitochondrial electron transport chain, as a site of pO 2 -responsive production of reactive oxygen species (ROS). These ROS are converted to a diffusible signaling molecule, hydrogen peroxide (H 2 O 2), by superoxide dismutase (SOD2). H 2 O 2 exits the mitochondria and regulates ion channels and enzymes, altering plasma membrane potential, intracellular Ca2+ and Ca2+-sensitization and controlling acute, adaptive, responses to hypoxia that involve changes in ventilation, vascular tone and neurotransmitter release. Subversion of this O 2 -sensing pathway creates a pseudohypoxic state that promotes disease progression in pulmonary arterial hypertension (PAH) and cancer. Pseudohypoxia is a state in which biochemical changes, normally associated with hypoxia, occur despite normal pO 2. Epigenetic silencing of SOD2 by DNA methylation alters H 2 O 2 production, activating hypoxia-inducible factor 1α, thereby disrupting mitochondrial metabolism and dynamics, accelerating cell proliferation and inhibiting apoptosis. Other epigenetic mechanisms, including dysregulation of microRNAs (miR), increase pyruvate dehydrogenase kinase and pyruvate kinase muscle isoform 2 expression in both diseases, favoring uncoupled aerobic glycolysis. This Warburg metabolic shift also accelerates cell proliferation and impairs apoptosis. Disordered mitochondrial dynamics, usually increased mitotic fission and impaired fusion, promotes disease progression in PAH and cancer. Epigenetic upregulation of dynamin-related protein 1 (Drp1) and its binding partners, MiD49 and MiD51, contributes to the pathogenesis of PAH and cancer. Finally, dysregulation of intramitochondrial Ca2+, resulting from impaired mitochondrial calcium uniporter complex (MCUC) function, links abnormal mitochondrial metabolism and dynamics. MiR-mediated decreases in MCUC function reduce intramitochondrial Ca2+, promoting Warburg metabolism, whilst increasing cytosolic Ca2+, promoting fission. Epigenetically disordered mitochondrial O 2 -sensing, metabolism, dynamics, and Ca2+ homeostasis offer new therapeutic targets for PAH and cancer. Promoting glucose oxidation, restoring the fission/fusion balance, and restoring mitochondrial calcium regulation are promising experimental therapeutic strategies. Introduction: Increasing evidence indicates that microRNAs (miRNAs) play a crucial role in modulating tumor growth. This study is centered on investigating the contribution of miR-25 to the progression of Renal Cell Carcinoma (RCC)., Methods: The investigators examined the expression levels of miR-25 and ADAMTS16 in RCC samples and cell lines. The association between miR-25 and ADAMTS16 was validated via a luciferase reporter assay. Cell viability, apoptosis, migration, and invasion were evaluated utilizing CCK-8 and flow cytometry techniques, while the expression levels of ADAMTS16, β-catenin, GSK-3β, and p-GSK-3β were assessed through western blot analysis., Results: The investigation revealed elevated expression levels of miR-25 in RCC tissues. Subsequently, ADAMTS16 was identified as a target of miR-25. Increased miR-25 levels were associated with decreased expression of ADAMTS16, resulting in enhanced cell viability and diminished apoptosis. Conversely, inhibition of miR-25 led to decreased cell viability, proliferation, and migration. Recent evidence has demonstrated that mesenchymal stem cells (MSCs) can release a large number of functionally specific microRNA (miRNA) microvesicles that play a role in promoting osteogenic differentiation, but the specific mechanism is not yet clear. Under such context, this study aims to elucidate the mechanism of bone marrow mesenchymal stem cell-derived exosomes (BMSC-Exo) promoting fracture healing in mice. We isolated and identified the BMSC-Exo. Bioinformatics analysis predicted high expression of miRNA in exosomes and verified the transfer of miR-25 in exosomes by immunofluorescence. Targeting relationship between miR-25 and Smad ubiquitination regulatory factor-1 (SMURF1) was predicted and verified by dual-luciferase reporter gene assay. Immunoprecipitation and protein stability assays were used to detect Runt-related transcription factor 2 (Runx2) ubiquitination and the effect of SMURF1 on Runx2 ubiquitination, respectively. The effect of miR-25 in BMSC-Exo on fracture healing in mice was assessed using X-ray imaging. alkaline phosphatase, alizarin red staining, EdU, CCK-8, and Transwell were used to evaluate the effects of exosomes transferred miR-25 on osteogenic differentiation, proliferation, and migration of osteoblasts. Bioinformatics analysis predicted that miR-25 expression in exosomes increased significantly. Moreover, the targeted regulation of SMURF1 by miR-25 was verified. SMURF1 inhibited Runx2 protein expression by promoting ubiquitination degradation of Runx2. Notably, miR-25 secreted by BMSC-Exo can accelerate osteogenic differentiation, proliferation, and migration of osteoblasts through SMURF1/Runx2 axis. Our results demonstrate that miR-25 in BMSC-Exo regulates the ubiquitination degradation of Runx2 by SMURF1 to promote fracture healing in mice.

Abstract Background The goat (Caprahircus) is one of the most important livestock animals. Goat milk fat is an important component in the nutritional quality of goat milk. Growing evidence points to the critical roles of microRNAs (miRNAs) in lipid metabolism. Results Using a highly sensitive method of S-poly(T) plus for miRNAs detection, we analyze the expression patterns of 715 miRNAs in goat mammary gland tissues at different stages of lactation. We observed that miR-25 expression had an inverse relationship with milk production. Overexpression of miR-25 significantly repressed triacylglycerol synthesis and lipid droplet accumulation. To explore the regulatory mechanism of miR-25 in milk lipid metabolism, we analyzed its putative target genes with bioinformatics analysis followed by 3′-UTR assays. Peroxisome proliferative activated receptor gamma coactivator 1 beta (PGC-1beta), a key regulator of lipogenics was identified as a direct target of miR-25 with three specific sites within its 3′-UTR. In addition, miR-25 mimics in goat mammary epithelial cells reduced the expressions of genes involved in lipid metabolism. Conclusions Taken together, our results show miR-25 is potentially involved in lipid metabolism and we reveal the function of the miR-25/PGC-1beta regulatory axis during lactation.

Multiple sclerosis (MS) is a complex inflammatory disease in which demyelination occurs in the central nervous system affecting approximately 2.5 million people worldwide. Recent reports have shown that the gut microbiome plays a crucial role in the functioning of the immune system in inflammatory diseases such as MS. In this study, the cuprizone-induced demyelination mouse model was used to investigate the effect of Lactobacillus casei strain T2 (IBRC-M10783) on the alleviation of these mice. Female C57BL/6 mice (8–10 weeks old) were divided into 6 groups: group 1, normal control; group 2, cuprizone control (oral administration of cuprizone 0.2% w/w for 4 weeks); group 3, probiotic control (oral administration of 1 × 109 CFU/ml probiotic for 4 weeks); group 4, treatment 1 (probiotic for 4 weeks then cuprizone for 4 weeks); group 5, treatment 2 (cuprizone for 4 weeks then probiotic for 4 weeks); and group 6, treatment 3 (cuprizone for 4 weeks then probiotic for 4 weeks with vitamin D3 at a dose of 20 IU/day). Then, TGF-β and IL-17 were measured by ELISA, and the expression of miR-155, miR-25, and IDO-1 was evaluated by real-time PCR. Among the measured microRNAs, the results showed that there was a significant decrease in miR-155 expression between the treatment 1 group and the cuprizone group. In the case of IL-17, the results also showed a significant reduction between the three treatment groups and the cuprizone group. The present study aimed to investigate the expression levels and clinical value of miR-365 and miR-25 in serum of patients with non-small cell lung cancer (NSCLC). Patients (180) diagnosed with NSCLC at the Affiliated Hospital of Guangdong Medical University from July 2011 to December 2013 were used as the experimental group. Volunteers (90) undergoing health examinations were used as the control group. The serum of the patients was collected after fasting in the morning. The expression levels of miR-365 and miR-25 in the serum of patients was assessed by quantitative real-time PCR (qRT-PCR), and the relationship among miR-365, miR-25 and the postoperative survival rate of NSCLC patients was analyzed. The relative expression level of miR-25 of patients with peripheral infiltration was significantly higher than that of patients without peripheral infiltration (P<0.05). There were significant differences in the relative expression level of miR-25 in different pathological grades and TNM stages, as well as with lymph node metastasis (P<0.05). The survival rate of NSCLC patients with high expression of miR-25 was significantly lower than that of NSCLC patients with low expression of miR-25 (P<0.05). The relative expression level of miR-365 of patients with peripheral infiltration was significantly lower than that of patients without peripheral infiltration (P<0.05). There were significant differences in the relative expression level of miR-365 in different pathological grades and TNM stages, as well as with lymph node metastasis (P<0.05). The survival rate of NSCLC patients with high expression of miR-365 was significantly higher than that of NSCLC patients with low expression of miR-365 (P<0.05). In conclusion, the expression levels of miR-25 and miR-365 were different in the serum of NSCLC patients, and they were closely related to certain clinical characteristics such as peripheral infiltration, pathological grade, tumor diameter, TNM stage and lymph node metastasis. Background/aim: Osteosarcoma is the most common primary bone malignancy that occurs frequently in children and adolescents. Baicalein, a flavonoid that has attracted great attention in recent years with its strong antitumor activity, shows a wide range of biological and pharmaceutical effects.MicroRNAs have been found to be involved in many critical processes in cancers. This study aimed to investigate the effect of baicalein and miR-25 on Wnt/β-catenin signaling pathway of osteosarcoma cell line Saos-2. Materials and methods: Cell viability was assessed, and qRT-PCR and Western blot were performed to study the effects of baicalein on expression of Wnt/β-catenin signaling pathway-realted genes (β-catenin, GSK-3β, and Axin2) of Saos-2 cells. Results: Our results indicated that baicalein can inhibit the proliferation (IC50 value 35 μM), regulate Wnt/β-catenin pathway and also increase miR-25 expression of Saos-2. Baicalein and also miR-25 decreased the expression of β-catenin and Axin2, while increasing the expression of GSK-3β. Down regulation of miR-25 decreased the expression of GSK-3β, while β-catenin and Axin2 expression increased. Conclusion: These findings demonstrate that baicalein may target genes related to the Wnt/β-catenin pathway by regulating miR-25 expression and may be a potential Wnt/β-catenin pathway inhibitor for osteosarcoma therapy. MicroRNAs (miRNAs) are reported to play a critical role in the regulation of cancer cell proliferation; however, the role of microRNA-25 (miR-25) in pancreatic ductal adenocarcinoma (PDAC) remains unclear. In the present study, the role of miR-25 in PDAC cell proliferation was investigated. Upregulated expression of miR-25 was found in PDAC tissues and cell lines by reverse transcription-quantitative PCR. Cell proliferation was significantly enhanced by overexpression of miR-25 as shown by CCK-8 assay results. Meanwhile, overexpression of miR-25 also promoted G1-to-S phase transition of the cell cycle in Aspc-1 cells via flow cytometry analysis. However downregulation of miR-25 inhibited the tumor cell proliferation and cell cycle transition. Online software was used to predict the target gene for miR-25 and luciferase reporter assay confirmed that Abl interactor 2 (ABI2) was a target of miR-25 via direct binding of its 3' untranslated region with miR-25. Moreover, results of the western blot analysis demonstrated that miR-25 negatively regulated the expression of ABI2 at the protein level. In addition, introduction of ABI2 mRNA into cells overexpressing miR-25 attenuated the carcinogenic effects of miR-25. Background: This study aimed to investigate the role of lncRNA MAGI2-AS3 in non-small cell lung cancer (NSCLC).Methods: Expression levels of MAGI2-AS3 and RECK mRNA in two types of tissues (non-tumor and NCSLC) were measured by qPCR. To further investigate the interaction between MAGI2-AS3 and RECK, MAGI2-AS3 and RECK expression vectors were transfected into H1993 cells.Results: We found that MAGI2-AS3 and RECK were upregulated and positively correlated in NSCLC. In NSCLC cells, MAGI2-AS3 overexpression led to upregulated RECK. Bioinformatics analysis showed that MAGI2-AS3 may bind miR-25, which can directly target RECK. In NSCLC cells, miR-25 overexpression led to downregulated RECK and attenuated the effects of MAGI2-AS3 overexpression, while MAGI2-AS3 and miR-25 failed to affect each other. Cell invasion and migration analysis showed decreased NSCLC cell invasion and migration rates after MAGI2-AS3 and RECK overexpression. To evaluate miR-25 expression in serum of gastric cancer (GC) patients and whether it can be a potential biomarker for GC diagnosis.

Background: Increasing evidence has shown that Helicobacter pylori is associated with coronary heart disease (CHD); however, the underlying mechanism remains unclear.Methods: The expression of miR-25 and mRNAs was measured using qRT-PCR. Protein levels were detected using western blotting and exosomes were assessed with an electron microscope. The target gene of miR-25 was identified using the luciferase report system.Results:H. pylori infection increased the expression of miR-25 in gastric epithelial cells and was associated with increased levels of exosome-transmitted miR-25 in human peripheral blood. Mechanistic investigation showed the Kruppel-like factor 2 (KLF2) was a direct target of exosome-transmitted miR-25 in vascular endothelial cells. In addition, the miR-25/KLF2 axis regulated the NF-κB signaling pathway, resulting in increased expression of interleukin 6 (IL6), monocyte chemoattractant protein-1 (MCP-1), vascular cell adhesion molecule-1 (VCAM-1), and intercellular adhesion molecule-1 (ICAM-1).Conclusion: Our findings suggest that the miR-25/KLF2 axis may be a potential therapeutic target for H. pylori-associated CHD. This study examined the molecular mechanisms underlying the roles of the microRNAs miR-18a and miR-25 in the progression of human liver cancer. Liver cancer biopsies obtained from early-stage liver cancer patients were examined by qRT-PCR and Northern blotting to examine the expression of miR-18a and miR-25. Both microRNAs were overexpressed in mouse primary hepatocytes following transfection of the cells with vectors encoding the microRNAs. An analysis of biopsy samples from liver cancer patients indicated that both miR-18a and miR-25 were overexpressed during the early stages of liver cancer. Further, qRT-PCR and Northern blotting confirmed that both of these microRNAs play crucial roles in the progression of liver cancer. Our findings clearly indicate that miR-18a and miR-25 can be used as prognostic biomarkers for early-stage liver cancer. OBJECTIVE: To study the mechanism of micro-ribonucleic acid (miR)-25 in regulating the fracture healing in rats. MATERIALS AND METHODS: A total of 45 male Sprague-Dawley (SD) rats were selected and randomly divided into group A [Phosphate Buffered Saline (PBS), n=15], group B (mimics NC, n=15) and group C (miR-25 mimics, n=15). The fracture model in rats was established via operation in all groups. From 1 d after the successful modeling, 50 µL (2 nmoL) of PBS was intraperitoneally injected into rats in group A, an equal amount of mimics NC was injected into rats in group B, and an equal amount of miR-25 mimics was injected into rats in group C. The above agents were injected once a week for consecutive 6 weeks. Fracture healing in rats was evaluated via X-ray imaging. At the same time, miR-25 expression in the three groups was detected via Reverse Transcription-Polymerase Chain Reaction (RT-PCR). Protein expressions of ß-catenin, proliferating cell nuclear antigen (PCNA) and bone morphogenetic protein-2 (BMP-2) in the three groups were detected via Western blotting. The OCN-, PCNA- and BMP-2-positive osteoblasts in the three groups were detected via immunohistochemical staining and were further quantified. Moreover, the biomechanical properties of femoral fracture healing in the three groups were analyzed via the 4-point bending flexural test. RESULTS: The X-ray examination of the femoral fracture healing at postoperative 1 and 7 weeks revealed that the fracture line disappeared, and both callus formation and fracture healing were good in miR-25 mimics group. In PBS group and mimics NC group, a few fracture lines could be observed, and both callus formation and fracture healing were poor. RT-PCR data showed that the expression level of miR-25 significantly increased in the miR-25 mimics group compared with that in the other two groups, and the differences were statistically significant (p<0.01). Western blotting analyses showed upregulated levels of ß-catenin, PCNA and BMP- 2 in the miR-25 mimics group compared with those in the control group, and the differences were statistically significant (p<0.01). Immunohistochemical staining manifested that the numbers of OCN-, PCNA- and BMP-2-positive osteoblasts in miR-25 mimics group markedly increased compared with that in the other two groups (p<0.01), suggesting that osteoblast differentiation in miR-25 mimics group was affected. The above immunohistochemical results indicated that the osteoblast differentiation at the fracture end in miR-25 mimics group was markedly enhanced compared with that in control groups. The results of the biomechanical test of femur specimens at 7 weeks after operation showed that in miR-25 mimics group, the maximum load, fracture energy and stiffness increased by 188%, 333% and 90%, respectively, compared with those in the PBS group (p<0.01). It is indicated that miR-25 promoted the mechanical properties of fracture healing. CCAAT enhancer binding protein α (C/EBP α), as an important transcription factor involved in cell proliferation, differentiation and metabolism, was up-regulated in primary hepatocellular carcinoma (HCC) and predicted poorer prognosis. In this study, we explored how histone deacetylases (HDACs) up-regulated C/EBP α in HCC. The protein expressions of HDAC1, HDAC2 were associated with C/EBP α by immunohistochemistry staining in a HCC tissue microarray. HCC cells were then treated with HDAC inhibitors or siRNAs to determine the roles of miR-124-3p and miR-25 in the regulation of C/EBP α mRNA expression. Both HDAC1 and HDAC2 proteins were significantly associated with C/EBP α. Inhibition of HDAC by either pharmacological inhibitors or siRNAs decreased C/EBP α mRNA expression in dose-dependent manners in HCC cells. HDAC inhibitors reduced C/EBP α mRNA stability as shown by pmiRGLO luciferase reporter assays. HDAC inhibition consistently induced miR-124-3p and miR-25 expression. Conversely, blockage of miR-124-3p and/or miR-25 by treatment with specific synthetic inhibitors abolished C/EBP α reduction. More importantly, C/EBP α mRNA stability could be rescued by site-directed mutations of miR-124-3p or miR-25 recognition sites in the C/EBP α 3′UTR sequence. OBJECTIVE: Ovarian cancer ranks 5th leading cause of cancer-related death in females worldwide. Physcion 8-O-β-glucopyranoside (PG) is an anthraquinone compound isolated from Rumex japonicus Houtt. This study aimed at investigating the effect of PG on ovarian cancer cells. PATIENTS AND METHODS: Cell viability was detected by CCK-8 assay. Colony formation assay evaluated whether PG could affect anchorage- independent growth. Whether PG affected cell cycle progression was examined by flow cytometry. The morphological changes caused by PG were visualized by microscopy. Apoptosis was quantitatively analyzed by flow cytometry. The effect of PG on cell migration and invasion was assessed by wound healing and transwell, respectively. The effect of PG on the expression of molecular markers was determined by Western blot. Microarray assay was performed to identify the potential target of PG. RESULTS: Results from the present study showed that PG decreased ovarian cancer cells viability. Colony formation assay also showed that PG suppressed the anchorage-independent growth of SKOV3 and OVCAR-3 cells. PG triggered cell cycle arrest at G1/G0 phase. The pro-apoptotic activity of PG was confirmed by flow cytometry, activation of caspase-3 and PARP, upregulation of Bax and downregulation of Bcl-2. The ability of PG to inhibit migration and invasion was evidenced by a decrease in wound healing and invasive cell number, as well as downregulation of MMP-2 and upregulation of TIMP-3. Microarray and qRT-PCR showed that miR-25 expression was downregulated by PG treatment. Moreover, our results indicated that the anti-cancer activities of PG were augmented by miR-25 knockdown and attenuated by ectopic miR-25 expression. BACKGROUND AND OBJECTIVE: MicroRNAs (miRNAs) are under investigation as novel diagnostic and prognostic biomarkers in several malignancies, including gastric cancer (GC). miR-25 was overexpressed in GC tissues, and higher miR-25 expression was statistically correlated with aggressive clinicopathological characteristics. In our study, we investigate the associations of serum miR-25 level with the clinicopathological characteristics, diagnosis and prognosis of GC patients. METHODS: Serum samples from 184 GC patients, 56 gastritis patients and 78 healthy controls were subjected to real-time quantitative polymerase chain reaction (RT-qPCR), and the relationship between micR-25 level and cliniopathological characteristics including diagnosis and prognosis was explored. RESULTS: Compared with the gastritis and healthy patients, serum miR-25 level was significantly up-regulated in patients with GC. Using a cut-off of 0.042, the level of miR-25 was significantly increased in serum samples from cancer patients; Using this test cancer patients were identified with 67.3–69.4% sensitivity and 80.4%–81.0% specificity. High serum miR-25 level was significantly associated with depth of invasion, lymph node metastasis and stage of disease. In univariate and multivariate analyses, miR-25 was an independent prognostic factor for overall survival (OS). Moreover, high serum miR-25 level was correlated with poor prognosis in patients subgroups stratified by tumor size, depth of invasion and lymph node metastasis. Serum miR-25 level was increased in both prominent serosal invasion group and lymph node metastasis group. Furthermore, stratified analysis showed that the TNM stage I–VI patients with high serum miR-25 level had poor prognosis than those with low serum miR-25 level. Objectives: Micro-RNAs (miRs) affect the gene expression of the pain and inflammation mediators. Myofascial trigger points (MTrPs) are one of the most common causes of skeletal and muscle pain. In this regard, dry needling (DN) was favored by physiotherapists as an effective method of treatment for mucopolysaccharidosis. The present study aimed to assess the effectiveness of DN on serum concentration of miR-939 and miR-25 before and after the treatment in patients with muscular pain caused by MTrPs in their shoulder girdle muscles. Methods: Twenty patients with pain in their shoulders, upper limbs, along with heads and necks, who had 3-5 active MTrPs in their shoulder girdle muscles participated in the study. They were treated in 5 sessions with DN. One session was held per day every three days for 2 weeks. Before and after the treatment, the pain intensity of the patients was measured using the visual analogue scale (VAS), their peripheral blood samples were taken, and miR serum concentrations were estimated by real-time polymerase chain reaction. Results: Comparing pain intensity in patients revealed a significant reduction after the treatment (P<0.05). In addition, statistical analysis indicated up-regulation of miR-939 and miR-25 with a significant difference (P<0.05) after the treatment compared to the time before the treatment. Conclusion: According to the reduction in pain intensity and increased expression of miR-939 and miR-25 serum levels after treatment, it can be concluded that DN is an effective technique for treatment of mucopolysaccharidosis in shoulder girdle muscles. Cancer stem cells contribute to cancer progression, but the mechanisms underlying neuroblastoma stem cell development are unclear. Here, we examined the roles of the transcription factor SLC34A2 in regulating the stemness of neuroblastoma cells. We found that SLC34A2 expression was negatively correlated with the overall survival and relapse‐free survival probability of neuroblastoma patients. Additionally, SLC34A2 expression was observed to be remarkably increased in spheroids derived from neuroblastoma cells. Knockdown of SLC34A2 attenuated the expression of stemness markers and spheroid formation capacity of neuroblastoma cell‐derived spheroids, and overexpression of SLC34A2 exerted the opposite effects in neuroblastoma cells. Mechanistically, SLC34A2 was found to directly bind to the promoter of MIR25, which targets glycogen synthesis kinase 3β (Gsk3β), an antagonist of Wnt signaling. Transfection of miR‐25 inhibitor or a Gsk3β overexpression plasmid attenuated the effects of SLC34A2 overexpression on the stemness of neuroblastoma cells. Our results demonstrate that miR‐25/Gsk3β‐mediated activation of Wnt signaling is responsible for SLC34A2‐induced enhancement of neuroblastoma cell stemness. Novel reliable biomarkers of inflammatory bowel disease (IBD) are clinically imperative due to potential limitations of endoscopic techniques. MicroRNAs (miRNAs) have emerged as non-invasive biomarkers of IBD; however, the full disease-specific miRNAs signature for IBD subtypes remains elusive. We evaluated the diagnostic role of circulating miR-486 and miR-25 in IBD patients and their potential ability to discriminate IBD subtypes; ulcerative colitis (UC) and Crohn's disease (CD). Sixty UC patients, 60 CD patients, and 60 healthy controls were recruited. Serum miRNA expression was determined using RT-qPCR. Bioinformatics was employed for target gene and protein-protein interaction (PPI) network analyses. Serum miR-486 was upregulated in CD patients, but didn't change in UC patients compared to controls. Conversely, serum miR-25 was decreased in both CD and UC patients compared to controls. Only miR-486 was differentially expressed between UC and CD patients. Receiver-operating characteristic analysis revealed that serum miR-486 was superior in CD diagnosis (AUC=0.945) and significantly distinguished CD and UC patients, whereas miR-25 showed discriminative potential for both UC and CD from controls. In the multivariate logistic analysis only miR-486 was associated with the risk of CD diagnosis. Serum miR-486 was correlated with CD activity index and location of disease in CD patients, whereas miR-25 was correlated with the type/extent of UC. PPI network analysis revealed common target genes and signaling pathways for both miRNAs. Background: The aim of this study was to identify novel microRNAs for potential use in the diagnosis of pancreatic cancer (PaC). Methods: A total of 1063 serum samples from 303 patients with PaC were collected, and the expression level of miR-25 was measured using quantitative real-time PCR (qRT-PCR). Results: We found that miR-25 had significant diagnostic value for the differential diagnosis of PaC in normal controls with an AUC (the area under the ROC curve) of 0.915 (95% CI: 0.893-0.937) that was significantly higher compared with an AUC of 0.725 for serum tumor marker carcinoembryonic antigen (CEA) and an AUC of 0.844 for CA19-9. Conclusions: These data suggest that serum miR-25 has strong potential as a novel biomarker for the early detection of PaC.

Background: An appropriate personalized molecular testing ensures the most efficacious treatment in lung cancer. It is still controversial whether younger lung adenocarcinoma (LUAD) patients have different molecular features compared with their older counterparts. MicroRNAs have been involved in lung cancer and their altered expression has been suggested as a potential biomarker in the pathogenesis, diagnosis, prognosis, and therapy of LUAD. Materials and Methods: To analyze putative differences in miR-25 expression between young (with age ≤50 years) and old adenocarcinoma patients, we quantified miR-25 levels with NanoString technology in 88 LUAD specimens. We further investigated a cohort of 309 LUAD patients from the cancer genome atlas (TCGA) database to test our hypothesis. Results: miR-25 expression was upregulated in young LUAD patients in comparison to the older ones (P = 0.03) in our series. The analysis of public database TCGA confirmed our results, which miR-25 differentially expressed in the two aged groups (P = 0.0009). Moreover, a consequential pairing of miR-25 with a target region in phosphatase and tensin homolog (PTEN) 3' untranslated region (UTR) and actually low PTEN expression seemed to be associated with high miR-25 (P = 0.001) in young patients. Conclusions: The interaction of miR-25 and PTEN in young LUAD may define a subgroup of patients, highlighting the concept of molecular testing in different age subtypes.

The role of miRNAs with tumor suppressive activity in liver cancer has been well studied. However, little is known about potential oncomiRs in HCC. In our study, we conducted a systematic evaluation of candidate oncomiRs and found that upregulation of miR‐18a and miR‐25 in HCC was associated with poor patient survival and promoted proliferation in HCC cell lines. These two miRNAs belong to the polycistronic paralogous miR‐17‐92 and miR‐25‐106b clusters respectively. Although the members of both clusters are often upregulated in HCC, the contribution of individual miRNAs in these clusters to HCC tumorigenesis is not fully understood. We validated SOCS5 as a bona fide target of both miRNAs, and established, for the first time, the tumor suppressive role of SOCS5 in liver cancer. We further investigated the mechanism by which SOCS5 contributes to tumorigenesis, demonstrated that this SOCS5/miR‐18a/miR‐25 axis regulates the tumor suppressor TSC1 and downstream mTOR signaling, and highlighted the potential therapeutic use of miR‐18a and miR‐25 inhibition in restoring SOCS5 levels in HCC.
