Abstract

Parasites threaten human and animal health globally. It is estimated that more than 60% of people on planet Earth carry at least one parasite, many of them several different species. Unfortunately, parasite studies suffer from duplications and inconsistencies between different investigator groups. Hence, groups need to collaborate in an integrated manner in areas including parasite control, improved therapy strategies, diagnostic and surveillance tools, and public awareness. Parasite studies will be better served if there is coordinated management of field data and samples across multidisciplinary approach plans, among academic and non-academic organizations worldwide. In this paper we report the first 'Living organism-World Molecular Network', with the cooperation of 167 parasitologists from 88 countries on all continents. This integrative approach, the 'Sarcoptes-World Molecular Network', seeks to harmonize Sarcoptes epidemiology, diagnosis, treatment, and molecular studies from all over the world, with the aim of decreasing mite infestations in humans and animals. © 2011 International Society for Infectious Diseases.

Abstract

The use of insect lipids as an alternative ingredient is an emergent topic in animal nutrition due to their antimicrobial activities. The present study evaluated the in vitro antimicrobial activities of two insect fats (black soldier fly (Hermetia illucens (HI) fat and yellow mealworm (Tenebrio molitor (TM) fat) and their effect as a total substitute for dietary soybean oil in cecal fermentation and gut microbiota of growing rabbits. The obtained results showed the potential of HI and TM fats as an antibacterial feed ingredient with a positive influence on the rabbit cecal microbiota. HI and TM fats therefore may be a sustainable lipid alternative to soybean oil in rabbit nutrition with possible interesting applications in the feed industry. This study aimed to evaluate the in vitro antimicrobial activities of two types of insect fats extracted from black soldier fly larvae (HI, Hermetia illucens L.) and yellow mealworm larvae (TM, Tenebrio molitor L.) and their effects as dietary replacement of soybean oil (S) on cecal fermentation pattern, and fecal and cecal microbiota in rabbits. A total of 120 weaned rabbits were randomly allotted to three dietary treatments (40 rabbits/group) -a control diet (C diet) containing 1.5% of S and two experimental diets (HI diet (HID) and TM diet (TMD)), where S was totally substituted by HI or TM fats during the whole trial that lasted 41 days. Regarding the in vitro antimicrobial activities, HI and TM fats did not show any effects on Salmonella growth. Yersinia enterocolitica showed significantly lower growth when challenged with HI fats than the controls. The insect fat supplementation in rabbit diets increased the contents of the cecal volatile fatty acids when compared to the control group. A metataxonomic approach was adopted to investigate the shift in the microbial composition as a function of the dietary insect fat supplementation. Abstract

The development of non-manipulative molecular tools to determine the origin of parasite infections in the animal trade (if infected before their export or import) is of great interest worldwide for both the animal trade industry and for animal welfare. Molecular tools have a wide range of applications, including forensic identification, wildlife preservation and conservation, veterinary public health protection, and food safety. Nonetheless, genetic markers were not reported to detect the source of infection in the animal trade. In this study we tested the applicability of molecular tools to detect the origin of Sarcoptes mite infection of wildebeest imported by the United Arab Emirate (UAE) from Tanzania. Using one multiplex of seven microsatellite markers and control samples from UAE, Kenya and Italy, we demonstrated the usefulness of the multiplex STR-typing as a molecular tool of pivotal interest to help commercialist, authorities, and conservationists, to identify the geographical origin of parasitic infections

20. Differential gene expression analysis in broiler chickens in response to dietary larvae Mealworm meal inclusion

Abstract

Recent studies on the exploitation of larvae meal inclusion in broiler chickens diet report a significant increase of breast muscle, carcass quality and growth performance. The purpose of this study was to investigate the effects of Mealworm (Tenebrio molitor, TM) meal inclusion in broiler chicken diet on global gene expression of four tissues, namely breast muscle, liver, jejunum and caecum. Isonitrogenous and isoenergetic diets were formulated with 0% (control group) and 15% (test group) of TM meal inclusion. 80 one-day-old male broiler chicks (Ross 708) were reared in 10 pens (eight birds/pen): the pens were divided in two groups including five experimental replicates for each diet. The birds were slaughtered at 53 days of age. The tissue samples were collected and stored in RNA Later solution (Ambion) for gene expression analysis. RNA-seq was carried out on Illumina NGS analyzer. The results showed that 117, 118, 119 and 182 RNA transcripts were upregulated and 120, 116, 168 and 126 RNA transcripts were downregulated in breast muscle, liver, jejunum and cecum respectively. In all tissues 25% of downregulated differentially expressed genes (DEG) had a foldchange> four; while the 25% of upregulated DEGs had a foldchange higher than four in breast and liver and>two in intestinal mucosa. A gene ontology analysis showed that only half of DEGs were involved in known biological processes including protein metabolism (30 DEGs), gene expression (30), signal transduction (15), and immune system (21). An expression pathway analysis with Reactome showed that the pathway with minor False Discovery Rate (FDR) was the striated muscle contraction in breast and the peptide chain elongation in the other tissues. The ubiquitin B gene (UBB), that plays an important role in protein's metabolism, was downregulated (-0.8) in breast and upregulated (þ0.57) in liver, whereas the expression of this gene did not change in intestinal mucosa. The differential expression of UBB might suggest an increase of non-lysosomal intracellular protein degradation related to a fast protein turnover in liver and a slow protein turnover in breast. In addition, the ribosomal protein (RPLP) profile, related to peptide chain elongation, was similar: -0.45 mean fold-change in breast and þ0.5 in liver while a moderated increase in intestinal mucosa was observed. No alterations in RNA expression were detected that could discourage the use of larvae mealworm inclusion in broiler chicken diet.

31. Host taxon-derived Sarcoptes mite in European wild animals revealed by microsatellite markers

Abstract

Ten markers specific to Sarcoptes mites were used in applying microsatellite genotyping to individual Sarcoptes mites collected in three European countries from 15 wild mammal populations belonging to 10 host species. The results showed that geographical separation had real biological significance for the definition of mite sub-populations, and that the degree of genetic exchange occurring between mites from different localities was apparently related to the geographical distance between locations. Wild host-derived mite populations were found to be clustered into three main groups: herbivore-, carnivore- and omnivore-derived Sarcoptes populations, with the omnivore-derived group located halfway between the herbivore- and carnivore-derived Sarcoptes populations. The separation between these three mite groups was better supported than the geographical separations; nevertheless, a kind of sub-clustering was detected within each of these three groups that separates mite populations into their geographical localities (countries). The lack of gene flow between Sarcoptes populations may have improved parasitic adaptations and led to what we refer to as a host-taxon-derived (carnivore host-, herbivore host- and omnivore host-derived) Sarcoptes mite found on European wild animals. Our results demonstrate that Sarcoptes is not a single panmictic population, even within each geographical location. This finding will have important ramifications for the study of the genetic structure of populations, life cycles, diagnosis and the monitoring protocols of the ubiquitous Sarcoptes mite, and could thus contribute to a better understanding of its associated epidemiology, which is of pivotal interest for wildlife biological conservation. © 2010 Elsevier Ltd.
