Understanding the ecological determinants of assemblage diversity in freshwater insects is of crucial importance for conservation biology and environmental management. This is particularly true in Haiti, where severe and ongoing environmental degradation of aquatic ecosystems is a major threat to the local invertebrate fauna. Here, using Odonates assemblages sampled from 67 waterbodies belonging to 14 different Haitian watersheds, we assessed species uniqueness (U) from both rarity based on spatial occupancy and taxonomic species distinctness (TSD), and species contribution to total β diversity (SCBD). We assessed between-site variation in species richness (SR), relative taxonomic distinctness (RTD), mean species uniqueness per site (Mean U), and the contribution of individual sites to total β diversity (LCBD), based on presence-absence data. We then examined the relationships between these variables and the physico-chemical characteristics of the sites. In total, we recorded 49 different odonate species (including two endemic species of conservation interest), belonging to 40 genera and 7 families, with a relatively high percentage (69.4%) of rare species (i.e. occurring in 10% or less of sampled water bodies). TSD was negatively correlated with spatial occupancy, while U was significantly higher in Zygopeteran species compared to Anisopteran species. SR varied among the 67 sites, ranging from 2 to 17 species, while SCBD was positively correlated with site occupancy. Differences in assemblage composition between sites were mainly due to species replacement, while only a few sites had assemblages consisting of a subset of larger assemblages found at other sites. The observed positive correlation between dissimilarity and geographic distance between sites further suggests that the turnover of species may take place over some ecological gradient and/or might be explained by species ecological requirements and limited ability to disperse. LCBD and Mean U increased with increasing altitude and decreasing temperature and conductivity, whereas SR and RTD did not. All variables were independent of pH. Overall, SR and RTD were higher in lotic than in lentic sites, and in sites with natural riparian vegetation compared to sites with anthropogenic or mixed riparian vegetation. Mean U provided a relevant index for site selection for odonates conservation in Haiti, whereas β diversity appeared to be of limited interest for this purpose. Globally, our results advocate the importance of conserving a wide diversity of waterbodies to protect Odonate diversity in Haïti, and particularly in forested areas at > 1000 m altitude.

Ion homeostasis is a crucial process in plants that is closely linked to the efficiency of nutrient uptake, stress tolerance and overall plant growth and development. Nevertheless, our understanding of the fundamental processes of ion homeostasis is still incomplete and highly fragmented. Especially at the mechanistic level, we are still in the process of dissecting physiological systems to analyse the different parts in isolation. However, modelling approaches have shown that it is not individual transporters but rather transporter networks (homeostats) that control membrane transport and associated homeostatic processes in plant cells. To facilitate access to such theoretical approaches, the modelling of the potassium homeostat is explained here in detail to serve as a blueprint for other homeostats. The unbiased approach provided strong arguments for the abundant existence of electroneutral H+/K+ antiporters in plants.

Host actin cytoskeleton is often targeted by pathogenic bacteria through the secretion of effectors. Legionella pneumophila virulence relies on the injection of the largest known arsenal of bacterial proteins, over 300 Dot/Icm type 4 secretion system effectors, into the host cytosol. Here, we define the functional interactions between VipA and LegK2, two effectors with antagonistic activities towards actin polymerization that have been proposed to interfere with the endosomal pathway. We confirmed the prominent role of LegK2 effector in Legionella infection, as the deletion of legK2 results in defects in the inhibition of actin polymerization at the Legionella-containing vacuole, as well as in endosomal escape of bacteria and subsequent intracellular replication. More importantly, we observed the restoration of the ΔlegK2 mutant defects, upon deletion of vipA gene, making LegK2/VipA a novel example of effector-effector suppression pair that targets the actin cytoskeleton and whose functional interaction impacts L. pneumophila virulence. We demonstrated that LegK2 and VipA do not modulate each other’s activity in a “metaeffector” relationship. Instead, the antagonistic activities of the LegK2/VipA effector pair would target different substrates, Arp2/3 for LegK2 and G-actin for VipA, to temporally control actin polymerization at the LCV and interfere with phagosome maturation and endosome recycling, thus contributing to the intracellular life cycle of the bacterium. Strikingly, the functional interaction between LegK2 and VipA is consolidated by an evolutionary history that has refined the best effector repertoire for the benefit of L. pneumophila virulence.

OBJECTIVES: Detecting instantaneous stroke volume change in response to altered cardiac preload is the physiologic foundation for determining preload responsiveness. DESIGN: Proof-of-concept physiology study. SETTING: Research simulation laboratory. SUBJECTS: Twelve healthy volunteers. INTERVENTIONS: A wireless continuous wave Doppler ultrasound patch was used to measure carotid velocity time integral and carotid corrected flow time during a squat maneuver. The Doppler patch measurements were compared with simultaneous stroke volume measurements obtained from a noninvasive cardiac output monitor. MEASUREMENTS AND MAIN RESULTS: From stand to squat, stroke volume increased by 24% while carotid velocity time integral and carotid corrected flow time increased by 32% and 9%, respectively. From squat to stand, stroke volume decreased by 13%, while carotid velocity time integral and carotid corrected flow time decreased by 24% and 10%, respectively. Both changes in carotid velocity time integral and corrected flow time were closely correlated with changes in stroke volume (r 2 = 0.81 and 0.62, respectively). The four-quadrant plot found a 100% concordance rate between changes in stroke volume and both changes in carotid velocity time integral and changes in corrected flow time. A change in carotid velocity time integral greater than 15% predicted a change in stroke volume greater than 10% with a sensitivity of 95% and a specificity of 92%. A change in carotid corrected flow time greater than 4% predicted a change in stroke volume greater than 10% with a sensitivity of 90% and a specificity of 92%. CONCLUSIONS: In healthy volunteers, both carotid velocity time integral and carotid corrected flow time measured by a wireless Doppler patch were useful to track changes in stroke volume induced by a preload-modifying maneuver with high sensitivity and specificity.

Abstract Here we describe the structure, the high-pressure, low-temperature (HP-LT) metamorphism and tectonic evolution of the Briançonnais distal margin units from the south Western Alps. The studied area extends southwest of the Dora-Maira (U)HP basement units and east-southeast of the classical Briançonnais nappes. A new structural map accompanied by geological profiles shows the thrusting of the oceanic nappes (Monviso and Queyras units) onto the distal Briançonnais units (D1 and D2 late Eocene deformation phases) under blueschist-facies conditions. Subsequent deformation during the Early Oligocene (D3 deformation phase) took place under greenschist-facies conditions and was associated with back-folding and -thrusting in the units overlying the Dora-Maira massif and with exhumation related to normal reactivation of former thrusts within the latter massif. Two large cover units, detached from their former distal Briançonnais basement, are redefined as the Maira-Sampeyre and Val Grana Allochthons (shortly: Maira-Grana Allochthons = MGA) including, (i) the Val Maira-Sampeyre unit involving Lower and Middle Triassic formations, seemingly detached from the Dora-Maira units during the subduction process, and (ii) the Val Grana unit with Middle-Upper Triassic and Early-Middle Jurassic formations, which was probably detached from the Maira-Sampeyre unit and correlates with the “Prepiemonte units” known from the Ligurian Alps to the Swiss Prealps. Three major shear zones involving tectonic mélanges of oceanic and continental rocks at the base of the Val Grana, Maira-Sampeyre and Dronero units testify to an early phase of exhumation within the subduction channel in front of the Adria plate. We present a new metamorphic map based on published and new petrological data, including new thermometric data obtained by Raman spectroscopy of carbonaceous material (RSCM). The TRSCM values range from ~ 400 °C to > 500 °C, going from the most external Val Grana unit and overlying Queyras schists to the uppermost Dora-Maira unit. During the Late Triassic, the width of the Briançonnais s.l. domain can be restored at ~ 100 km, whereas it reached ~ 150 km after the Jurassic rifting. A significant, second rifting event affected the Briançonnais domain during the Late Cretaceous-Paleocene, forming the Longet-Alpet chaotic breccias, which deserve further investigations.
