Assessing the genetic diversity of yam germplasm from different geographical origins for cultivation and breeding purposes is an essential step for crop genetic resource conservation and genetic improvement, especially where the crop faces minimal attention. This study aimed to classify the population structure, and assess the extent of genetic diversity in 207 Dioscorea rotundata genotypes sourced from three different geographical origins. A total of 4,957 (16.2%) single nucleotide polymorphism markers were used to assess genetic diversity. The SNP markers were informative, with polymorphic information content ranging from 0.238 to 0.288 and a mean of 0.260 across all the genotypes. The observed and expected heterozygosity was 0.12 and 0.23, respectively while the minor allele frequency ranged from 0.093 to 0.124 with a mean of 0.109. The principal coordinate analysis, model-based structure and discriminant analysis of principal components, and the Euclidean distance matrix method grouped 207 yam genotypes into three main clusters. Genotypes from West Africa (Ghana and Nigeria) had significant similarities with those from Uganda. Analysis of molecular variance revealed that within-population variation across three different geographical origins accounted for 93% of the observed variation. This study, therefore, showed that yam improvement in Uganda is possible, and the outcome will constitute a foundation for the genetic improvement of yams in Uganda. [ABSTRACT FROM AUTHOR]

A proper understanding of the diversity of the available germplasm is an initial step for the genetic improvement of a crop through breeding. However, there is limited information on the diversity of Uganda's yam germplasm. The study sought to characterize the diversity of yam germplasm utilized for decades in Uganda together with germplasm recently introduced from West Africa using phenotypic traits. A germplasm collection of 291 genotypes was characterized using 28 phenotypic traits. Data were subjected to multivariate analysis using principal component analysis and cluster analysis. The traits assessed were informative and discriminating, with 62% of the total variation explained among the first six principal components. Results showed that the important phenotypic traits contributing to most of the variability among the genotypes were leaves, flowering, and tuber traits. Ugandan genotypes were identified with amorphous tuber shapes compared to West African genotypes. The study has shown that there is ample phenotypic variability within the major yam genotypes in Uganda yam germplasm that can be used for genetic improvement. More in-depth molecular and biochemical studies to further understand the diversity are recommended. Taro is an ancient nutritional and medicinal crop woven into the fabric of the socio-economic life of those living in the tropics and sub-tropics. However, West Africa (WA), which has been a major producer of the crop for several decades, is experiencing a significant decline in production as a result of taro leaf blight (TLB), a disease caused by Phytophthora colocasiae Raciborski. A lack of research on taro in WA means that available innovative technologies have not been fully utilized to provide solutions to inherent challenges and enhance the status of the crop. Improvement through plant breeding remains the most economically and environmentally sustainable means of increasing the productivity of taro in WA. With this review, we provide insights into the importance of the taro crop in WA, evaluate taro research to date, and suggest how to address research gaps in order to promote taro sustainability in the region. A core subset with a small number of accessions representing the genetic diversity of the base collection plays a vital role in facilitating efficient utilization of plant genetic resources. This is particularly relevant for vegetatively propagated large plant size tuber crops with a long growing period, such as white Guinea yam (Dioscorea rotundata Poir.). For the efficient utilization of D. rotundata genetic resources, this study was aimed at developing a mini‐core collection from a core collection of 447 D. rotundata accessions maintained at the International Institute of Tropical Agriculture (IITA). Accordingly, a D. rotundata mini‐core collection representing 102 accessions was selected using 16 simple sequence repeat (SSR) markers, retaining ∼98% of the SSR allelic diversity of the base collection. A similar level of diversity was captured within the mini‐core collection and the base collection with respect to 21 morphological traits, ploidy level, and geographic origin. The mini‐core collection demonstrated a wide range of variation in agronomic traits such as growth period, number of tubers, average tuber weight, and total yield per plant. This variation was considerable when compared with the variation observed for the same traits among the 10 lines or genotypes conventionally used in the breeding program at IITA, which were included in this study as checks. The selected mini‐core accessions could serve as a working collection to broaden the genetic variation for use in practical breeding programs, as well as in future genomic analyses aimed at the genetic improvement of D. rotundata in West Africa. White yam (Dioscorearotundata Poir.) is one of the most important tuber crops in West Africa, where it is indigenous and represents the largest repository of biodiversity through several years of domestication, production, consumption, and trade. In this study, the genotyping-by-sequencing (GBS) approach was used to sequence 814 genotypes consisting of genebank landraces, breeding lines, and market varieties to understand the level of genetic diversity and pattern of the population structure among them. The genetic diversity among different genotypes was assessed using three complementary clustering methods, the model-based admixture, discriminant analysis of principal components (DAPC), and phylogenetic tree. ADMIXTURE analysis revealed an optimum number of four groups that matched with the number of clusters obtained through phylogenetic tree. Clustering results obtained from ADMIXTURE analysis were further validated using DAPC-based clustering. Analysis of molecular variance (AMOVA) revealed high genetic diversity (96%) within each genetic group. A network analysis was further carried out to depict the genetic relationships among the three genetic groups (breeding lines, genebank landraces, and market varieties) used in the study. This study showed that the use of advanced sequencing techniques such as GBS coupled with statistical analysis is a robust method for assessing genetic diversity and population structure in a complex crop such as white yam.
