The U.S. Department of Agriculture (USDA) Food Safety and Inspection Service (FSIS) conducts surveillance of metallic elements in U.S. meat, poultry, and Siluriformes fish samples collected immediately postmortem as part of its National Residue Program (NRP). From 2017 to 2022, 13,966 samples were analyzed under the NRP. The Federal Emergency Response Network (FERN) Cooperative Agreement Program (CAP) tests meat, poultry, and Siluriformes fish products collected at retail in the United States for metals. From 2018 to 2022, 2,902 samples were analyzed by FERN CAP laboratories. Meat and poultry samples collected by FSIS show that most metals were not detected at all or were detected infrequently. Meat is a rich source of iron and zinc, and iron was detected in 22% (1,255/5,623) and zinc was detected in 48% (2,742/5,676) of meat and poultry samples, respectively. The percentage of samples testing positive for manganese, molybdenum, lead, and cadmium were higher in the FERN CAP retail samples than in FSIS samples. Expected human exposure from average levels of lead and cadmium found in meat and poultry was compared to toxicological reference values and was not found to exceed these values. Detections of arsenic and mercury were found more often in Siluriformes fish samples (2017-2022) than in terrestrial animals. Trace amounts of arsenic and mercury were detected in 8% and 4% of Siluriformes samples, respectively, but were not detected at levels that raise concern. Purpose: To determine the contents of chicken nuggets from 2 national food chains., Background: Chicken nuggets have become a major component of the American diet. We sought to determine the current composition of this highly processed food., Methods: Randomly selected nuggets from 2 different national fast food chains were fixed in formalin, sectioned and stained for microscopic analysis., Results: Striated muscle (chicken meat) was not the predominate component in either nugget. In 2003-2004, the U.S. Department of Agriculture Food Safety and Inspection Service (FSIS) conducted an exploratory assessment to determine the occurrence and levels of cadmium and lead in randomly collected samples of kidney, liver, and muscle tissues of mature chickens, boars/stags, dairy cows, and heifers. The data generated in the study were qualitatively compared to data that FSIS gathered in a 1985-1986 study in order to identify trends in the levels of cadmium and lead in meat and poultry products. The exploratory assessment was necessary to verify that Hazard Analysis and Critical Control Point plans and efforts to control exposure to these heavy metals are effective and result in products that meet U.S. export requirements. A comparison of data from the two FSIS studies suggests that the incidence and levels of cadmium and lead in different slaughter classes have remained stable since the first study was conducted in 1985-1986. This study was conducted to fulfill FSIS mandate to ensure that meat, poultry, and egg products entering commerce in the United States are free of adulterants, including elevated levels of environmental contaminants such as cadmium and lead.

Antibiotics are used by the poultry industry to enhance the health and productivity of flocks. The use of antimicrobials is strictly regulated by the Food and Drug Administration (FDA) and the USDA to warrant their safety and efficacy. Prior to regulatory approval, the pharmacokinetics and tissue tolerances of an antimicrobial are determined to set the proper dosage. To ensure proper use, both the FDA and USDA have research, surveillance, and compliance programs to develop detection methods and monitor poultry tissues for antimicrobials. Unfortunately, there is the perception among many consumers that our food supply contains high concentrations of drug or hormone residues causing significant health concerns or problems. In fact, foods produced in this country (including poultry) are very safe and meet the highest standards to exclude chemical contaminants. An overview will be presented on the federal oversight and monitoring of antimicrobial residues in poultry tissues.

Arachidonic acid (AA) is an extremely important fatty acid involved in cell regulation. When provided in the diet, it is cogently incorporated in membrane phospholipids and enhances eicosanoid biosynthesis in vivo and in vitro; however, controversy exists as to the levels of AA in food and in the diet. This study determined the amount of AA in cooked and raw portions of beef (rib eye), chicken (breast and thigh), eggs, pork (loin), turkey (breast), and tuna; it compared these results to values published in Agriculture Handbook No. 8 (HB-8). The cooked portions were prepared as described in HB-8. With the exception of chicken thigh and tuna, the levels of AA (w/w) in the selected foods analyzed were significantly higher, in general, than those values published in HB-8. The greatest differences were observed in beef (raw and cooked), turkey breast (raw and cooked), and pork (cooked) where AA levels were twice that of the values in HB-8. In contrast, the AA and n-3 fatty acid contents in tuna were almost half the HB-8 values. The present data indicate that HB-8 tends to underreport the amounts of AA in a number of foods commonly consumed in the American diet, and new initiatives should be considered to validate and update the current database for fatty acid composition of foods.

An interlaboratory study of a liquid chromatographic (LC) method for determining monensin in premix (60-80 g/lb or 132-176 mg/g) and animal feeds (5-200 g/ton or 0.0055-0.22 mg/g) was conducted in laboratoriesin the United States, Canada, France, and Germany. The LC system used a reversed-phase column, postcolumn derivatization with vanillin, and UV detection. The method separates monensin from other ionophores such as narasin and salinomycin. Each laboratory analyzed a total of 20 samples of premix, liquid feed supplements, poultry, and cattle feeds. Concentrations of monensin in all samples ranged from 0 to 176 mg/g (80 g/lb). Reproducibility relative standard deviation (RSDR) for premix ranged from 2.8 to 3.4%. For feed samples containing monensin, repeatability standard deviation (sr) ranged from 0.9 to 7.0. Reproducibility standard deviation (sR) ranged from 1.2 to 11. Repeatability relative standard deviation (RSDr) ranged from 6.1 to 21% and RSDR values-ranged from 8.6 to 25%. Sample preparation for the LC method is less labor intensive than that for the microbiological assays. The LC assay is more efficient than the microbiological assays. This LC method for determination of monensin in premix and animal feeds has been adopted first action by AOAC INTERNATIONAL.

A total of 19 government and private industry laboratories in Canada and the United States participated in the collaborative study. Naturally contaminated ground poultry and animal meals, as well as inoculated raw shrimp, were examined for presence of Salmonella by both the modified immunodiffusion method and the Bacteriological Analytical Manual culture method, resulting in an agreement rate of 93.1%. The 2 methods are statistically equivalent for all food types at each inoculation level and for all lots of naturally contaminated foods evaluated in this study. The modification of the AOAC Official Method 989.13, immunodiffusion (1-2 TEST) method for detection of motile Salmonella in all foods, has been adopted revised first action by AOAC INTERNATIONAL.

The protein nutritional value of meat, poultry, and their products must be considered in relation to humans. Because direct assessment of protein nutritional value in human subjects is impractical for regulatory purposes, methods based on in vitro (chemical) and animal bioassays for assessment of protein quality have been developed. Herein, a case is made for the use of amino acid data as a basis for regulation of meat and poultry products, where considerations of protein nutritional value are included in the regulation. Current data on the protein nutritional quality of these products are based almost entirely on the use of the rat PER assay procedure, supplemented by data on the amino acid content of these products. Because of the inadequacy of the PER assay, the available data base provides an inadequate body of knowledge from which to draw final recommendations concerning policies for regulation of the protein nutritional quality of meat and poultry products. However, if it is necessary to include a measure of protein nutritional quality in a regulation, these data provide a reasonably sound basis for tentatively recommending use of amino acid composition data rather than the standard rat bioassay procedure. In such an approach based on amino acid composition data, consideration might be given to the contents of hydroxyproline (or possibly proline), 3-methylhistidine, lysine, and nitrogen, with a view to establishing a composite or minimum value for the noncollagen muscle protein content of these products.

Since 1964 the Food and Drug Administration Total Diet study has reported residues of pesticides and other chemicals ingested in the diet of a young adult male, statisically the Nation's largest eater. During the tenth year of the study, pesticide residues remained at the relatively low levels previously reported. Thirty market baskets were collected in 30 cites which ranged in population from less than 50,000 to 1,000,000 or more. Averages and ranges of residues are reported from August 1973 through July 1974 by food class. Individual items in the dairy and meat composites in four market baskets were analyzed for pesticides; results are included. Data for lead, cadmium, selenium, mercury, arsenic, and zinc are also included. Results of recovery studies within various classes of residues are also presented.

FDA is engaged in a systematic review of approved animal drugs to determine whether upgrading of safety data is required to provide continuing assurance that the approved uses for these drugs are safe. These reviews will be undertaken on the basis of a priority of anticipated human health significance.

A detailed analysis of the amino acid composition data of meat and poultry products, in relation to PER, clearly reveals high correlations between amino acid data expressed in various forms and results of protein quality estimations obtained with the PER assay. The significance of these observations in reference to protein quality evaluation for regulatory purposes will be considered in Background Paper 5.

The U.S. Department of Agriculture has the responsibility of determining if a method for residues of drugs and pesticides in animal tissues is acceptable for regulatory control, reviewing data on interfering compounds, validating methods, monitoring drug residues in animal and poultry tissues, assigning priority for monitoring drugs and pesticide residues. These responsibilities are carried out in cooperation with the U.S. Food and Drug Administration in the areas of method development, determination of priorities, and punitive action.

The National Residue Program conducted by the Food Safety and Inspection Service (FSIS) of the USDA includes a comprehensive testing program for residues of pesticides, drugs and other chemical contaminants in meat and poultry. Prevention strategies encourage producers to adopt quality control measures in their production management to prevent illegal residues in food. These activities have been effective in reducing the occurrence of violative residues and the potential for adverse health effects. Overall, the number of domestic monitoring samples containing violative residues is low-about 1% of samples tested. Violative residues are found less frequently in poultry than in livestock. More occur in swine than in other species; the least number occur in fed cattle and broilers. Testing results over the last 10 yr show that most drugs and pesticides used to enhance agricultural productivity are not causing a residue problem in meat and poultry. However, the FSIS cannot be complacent about its program achievements. Unacceptably high incidences of violative residues of certain drugs, namely, sulfonamides and antibiotics, still occur in particular production classes. For example, the incidence of violative sulfonamide residues in liver samples from swine slaughtered in 1985 was about 6%, with significant differences between geographical areas. An estimated 2.5% of market hogs had violative sulfamethazine residues in the muscle tissue. The FSIS is taking steps to correct this and other residue problems by strengthening the link between residue detection and enforcement and by expanding its analytical capability to monitor for residues.
