Whitefly, Bemisia tabaci Gennadius (B cryptic species), transmits cucurbit leaf crumple virus (CuLCrV) in a persistent fashion. CuLCrV affects several crops such as squash and snap bean in the southeastern United States. CuLCrV is often found as a mixed infection with whitefly transmitted criniviruses, such as cucurbit yellow stunting disorder virus (CYSDV) in hosts such as squash, or as a single infection in hosts such as snap bean. The implications of different host plants (inoculum sources) with varying infection status on CuLCrV transmission/epidemics is not clear. This study conducted a series of whitefly mediated CuLCrV transmission experiments. In the first experiment, three plants species: squash, snap bean, and tobacco were inoculated by whiteflies feeding on field-collected mixed-infected squash plants. In the second experiment, three plant species, namely squash, snap bean, and tobacco with varying infection status (squash infected with CuLCrV and CYSDV and snap bean and tobacco infected with CuLCrV), were used as inoculum sources. In the third experiment, squash plants with differential CuLCrV accumulation levels and infection status (either singly infected with CuLCrV or mixed infected with CuLCrV and CYSDV) were used as inoculum sources. Irrespective of plant species and its infection status, CuLCrV accumulation in whiteflies was dependent upon the CuLCrV accumulation in the inoculum source plants. Furthermore, differential CuLCrV accumulation in whiteflies resulted in differential transmission, CuLCrV accumulation, and disease phenotype in the recipient squash plants. Overall, results demonstrate that whitefly mediated CuLCrV transmission between host plants follows a virus density dependent phenomenon with implications for epidemics. [ABSTRACT FROM AUTHOR]

The soil-borne pathogens, particularly Fusarium oxysporum f. sp. niveum (FON) and southern root-knot nematode (RKN, Meloidogyne incognita) are the major threats to watermelon production in the southeastern United States. The role of soil micronutrients on induced resistance (IR) to plant diseases is well-documented in soil-based media. However, soil-based media do not allow us to determine the contribution of individual micronutrients in the induction of IR. In this manuscript, we utilized hydroponics-medium to assess the effect of controlled application of micronutrients, including iron (Fe), manganese (Mn), and zinc (Zn) on the expression of important IR genes (PR1, PR5, and NPR1 from salicylic acid (SA) pathway, and VSP, PDF, and LOX genes from jasmonic acid (JA) pathway) in watermelon seedlings upon inoculation with either FON or RKN or both. A subset of micronutrient-treated plants was inoculated (on the eighth day of micronutrient application) with FON and RKN (single or mixed inoculation). The expression of the IR genes in treated and control samples was evaluated using qRT-PCR. Although, significant phenotypic differences were not observed with respect to the severity of wilt symptoms or RKN galling with any of the micronutrient treatments within the 30-day experimental period, differences in the induction of IR genes were considerably noticeable. However, the level of gene expression varied with sampling period, type and concentration of micronutrients applied, and pathogen inoculation. In the absence of pathogens, micronutrient applications on the seventh day, in general, downregulated the expression of the majority of the IR genes. However, pathogen inoculation preferentially either up- or down-regulated the expression levels of the IR genes at three days post-inoculation depending on the type and concentration of micronutrients. The results demonstrated here indicate that micronutrients in watermelon may potentially make watermelon plants susceptible to infection by FON and RKN. However, upon infection the IR genes are significantly up-regulated that they may potentially aid the prevention of further infection via SA- and JA-pathways. The sweetpotato whitefly, Bemisia tabaci MEAM1 Gennadius (Hemiptera: Aleyrodidae), and the complex of viruses it transmits are major limiting factors to squash production in the southeastern United States. At this time, insecticides are extensively relied upon for the management of whiteflies and, indirectly, whitefly-transmitted viruses. The development of a multi-faceted, integrated pest management (IPM) program is needed to increase the sustainability and profitability of squash production. Experiments in 2018 and 2019 evaluated the effects of insect exclusion netting (IEN) in combination with selected pesticides on whitefly population dynamics and virus incidence in greenhouse-grown squash seedlings. Field experiments from 2018 to 2021 evaluated the effects of mulch type (UV-reflective mulch, live mulch, and white plastic mulch), row covers, and insecticides on whitefly population dynamics, silver leaf disorder (SSL) intensity, virus symptom severity, and marketable yield. IEN significantly reduced whiteflies and virus incidence on squash seedlings in the greenhouse study. In the field mulch study, lower whitefly abundance and SSL intensity, as well as reduced virus symptom severity, were observed in plots with reflective mulch compared with white plastic or live mulch. In the insecticide/row cover study, whitefly abundance, SSL intensity, and virus symptom severity were lowest in the row cover and cyantraniliprole- and flupyradifurone-treated plots. Field plots with row covers and those with UV-reflective mulch consistently produced the greatest marketable yields. These findings demonstrate that growers can reduce whitefly and virus pressure and preserve yields in squash production in the southeastern United States by combining cultural and chemical tactics, including row covers, UV-reflective mulch, and select insecticides. Watermelon (Citrullus lanatus var. lanatus) yield losses due to Fusarium wilt can be considerable and have been increasing in the United States, mainly due to the emergence of the new races of Fusarium oxysporum f. sp. niveum (FON). One of the potential management strategies to reduce Fusarium wilt is the use of fumigants and soil-applied fungicides. Field experiments were conducted in 2018 and 2020 to evaluate fumigant Pic-clor 60 (choloropicrin pre-mixed with 1,3 dicholoropropene) with post-plant soil-applied fungicides (prothioconazole and pydiflumetofen) for their effectiveness against Fusarium wilt. The field was fumigated 21 days before transplanting, and three applications of either fungicide were made at 14-day intervals via a drip irrigation system starting a day after transplanting. In 2018, based on the disease ratings at harvest maturity, all treatments significantly reduced final disease incidence compared to the non-treated control, and the highest reduction was observed when the fumigant was applied at 336.26 kg/ha along with either fungicides. The application of fumigant, together with either fungicides, had significantly higher reduction in disease incidence compared to the stand-alone application (fumigant or fungicides). Also, Pic-clor 60 at 336.26 kg/ha provided a higher reduction in disease incidence than Pic-clor 60 at 280.21 kg/ha. In 2020, results showed a similar trend to 2018, where Pic-clor 60 at 336.26 kg/ha in combination with either prothioconazole or pydiflumetofen had significantly lower final disease incidence. The reduction in disease incidence in Pic-clor 60 at 336.26 kg/ha was higher compared to a lower application rate of 280.21 kg/ha, either when applied alone or in combination with either fungicides. The results presented here are the first to demonstrate that the fumigant Pic-clor 60 along with soil-applied fungicides, pydiflumetofen or prothioconazole may provide an effective management option for Fusarium wilt of watermelon. • This study examined mixed virus infection in a plant host and acquisition of multiple viruses by the vector. • Effects of CuLCrV and/or CYSDV infection in squash on virus load, and whitefly preference and fitness were examined. • Effects of whitefly acquisition of TYLCV and/or CuLCrV on virus load, and whitefly preference and fitness were examined. • Vector-virus-host interactions were sometimes different in the presence of two viruses than one virus. Mixed virus infection in host plants can differentially alter the plant phenotype, influence vector fitness, and affect virus acquisition and inoculation by vectors than single-virus infection. Vector acquisition of multiple viruses from multiple host plants could also differentially affect vector fitness and virus inoculation than acquisition of one virus. Whitefly-virus pathosystems in the southern United States include both the above-stated facets. For the first facet, this study examined the effects of single and mixed infection of cucurbit leaf crumple virus (CuLCrV, a begomovirus) and cucurbit yellow stunting disorder virus (CYSDV, a crinivirus) infecting squash on whitefly (Bemisia tabaci Gennadius MEAM1) host preference and fitness. Mixed infection of CuLCrV and CYSDV in squash plants severely altered their phenotype than single infection. The CYSDV load was reduced in mixed-infected squash plants than in singly-infected plants. Consequently, whiteflies acquired reduced amounts of CYSDV from mixed-infected plants than singly-infected plants. No differences in CuLCrV load were found between singly- and mixed-infected squash plants, and acquisition of CuLCrV by whiteflies did not vary between singly- and mixed-infected squash plants. Both singly- and mixed-infected plants similarly affected whitefly preference, wherein non-viruliferous and viruliferous (CuLCrV and/or CYSDV) whiteflies preferred non-infected plants over infected plants. The fitness study involving viruliferous and non-viruliferous whiteflies revealed no differences in developmental time and fecundity. For the second facet, this study evaluated the effects of individual or combined acquisition of tomato-infecting tomato yellow leaf curl virus (TYLCV, a begomovirus) and squash-infecting CuLCrV on whitefly host preference and fitness. Whiteflies that acquired both CuLCrV and TYLCV had significantly lower CuLCrV load than whiteflies that acquired CuLCrV alone, whereas TYLCV load remained unaltered when acquired individually or in conjunction with CuLCrV. Whitefly preference was not affected following individual or combined virus acquisition. Viruliferous (CuLCrV and/or TYLCV) whiteflies preferred to settle on non-infected tomato and squash plants. The mere presence of CuLCrV and/or TYLCV in whiteflies did not affect their fitness. Taken together, these results indicate that mixed infection of viruses in host plants and acquisition of multiple viruses by the vector could have implications for virus accumulation, virus acquisition, vector preference, and epidemics that sometimes are different from single-virus infection or acquisition. Alternaria leaf blight and head rot is an important disease of broccoli and other cole crops. With no resistant host varieties, fungicides are utilized to manage this disease. However, anecdotal evidence suggests that, in southeastern U.S. broccoli-producing states, there is a loss of disease control through the use of quinone outside inhibitor (QoI) fungicides. To understand why there is a reduced sensitivity to QoI fungicides in these states, we isolated Alternaria spp. from symptomatic lesions on cole crops from Georgia and Virginia (two states with observations of loss of fungicide sensitivity) as well as New York (a state with no observations of loss of fungicide sensitivity). Using multilocus sequencing and phylogenetic analysis, we identified two species, Alternaria brassicicola and A. japonica . Whereas A. brassicicola was isolated in all states, A. japonica was only isolated in Georgia. Next, we wanted to determine the sensitivity of these isolates to azoxystrobin-an active ingredient in some QoI fungicides-by estimating the effective concentration at which only 50% of spores germinate (EC 50 ). The EC 50 of A. brassicicola ranged from 0.01 to 0.17 ppm, whereas that of A. japonica was 8.1 to 28.1 ppm. None of the known target-site mutations that confer resistance to QoI fungicides were identified during screening of either species. A. japonica was first reported on the east coast of the United States in 2020 in South Carolina. The substantially higher EC 50 value suggests that its emergence in the southeastern United States may play at least a part in the observed loss of disease control. Middle East-Asia Minor 1 (MEAM1) and Mediterranean (MED) are two of the most invasive members of the sweetpotato whitefly, Bemisia tabaci, cryptic species complexes and are efficient vectors of begomoviruses. Bemisia tabaci MEAM1 is the predominant vector of begomoviruses in open-field vegetable crops in the southeastern United States. However, recently B. tabaci MED also has been detected in the landscape outside of greenhouses in Florida and Georgia. This study compared the transmission efficiency of one Old-World (OW) and two New-World (NW) begomoviruses prevalent in the southeastern United States, viz., tomato yellow leaf curl virus (TYLCV), cucurbit leaf crumple virus (CuLCrV), and sida golden mosaic virus (SiGMV) between B. tabaci MEAM1 and B. tabaci MED. Bemisia tabaci MEAM1 efficiently transmitted TYLCV, CuLCrV, or SiGMV, whereas B. tabaci MED only transmitted TYLCV. Percent acquisition and retention of OW TYLCV following a 72 h acquisition access period was significantly higher for B. tabaci MED than B. tabaci MEAM1. In contrast, B. tabaci MEAM1 acquired and retained significantly more NW bipartite begomoviruses, CuLCrV or SiGMV, than B. tabaci MED. Quantitative analysis (qPCR) of virus DNA in whitefly internal tissues revealed reduced accumulation of CuLCrV or SiGMV in B. tabaci MED than in B. tabaci MEAM1. Fluorescent in situ hybridization (FISH) showed localization of CuLCrV or SiGMV in the midgut of B. tabaci MED and B. tabaci MEAM1. However, localization of CuLCrV or SiGMV was only observed in the primary salivary glands of B. tabaci MEAM1 and not B. tabaci MED. TYLCV localization was observed in all internal tissues of B. tabaci MEAM1 and B. tabaci MED. Overall, results demonstrate that both B. tabaci MEAM1 and B. tabaci MED are efficient vectors of OW TYLCV. However, for the NW begomoviruses, CuLCrV and SiGMV, B. tabaci MEAM1 seems to a better vector.
