Soil ecoenzymatic stoichiometry reflects the dynamic equilibrium between microorganism's nutrient requirements and resource availability. However, uncertainties persist regarding the key determinants of nutrient restriction in relation to microbial metabolism under varying degrees of warming. Our long-term and multi-level warming field experiment (control treatment, +0.42 °C, +1.50 °C, +2.55 °C) in a typical alpine meadow unveiled a decline in carbon (C)- and nitrogen (N)-acquired enzymes with escalating warming magnitudes, while phosphorus (P)-acquired enzymes displayed an opposite trend. Employing enzymatic stoichiometry modeling, we assessed the nutrient limitations of microbial metabolic activity and found that C and N co-limited microbial metabolic activities in the alpine meadow. Remarkably, high-level warming (+2.55 °C) exacerbated microbe N limitation, but alleviate C limitations. The structural equation modeling further indicated that alterations in soil extracellular enzyme characteristics (SES) were more effectively elucidated by microbial characteristics (microbial biomass C, N, P, and their ratios) rather than by soil nutrients (total nutrient contents and their ratios). However, the microbial control over SES diminished with higher levels of warming magnitude. Overall, our results provided novel evidence that the factors driving microbe metabolic limitation may vary with the degree of warming in Tibet alpine grasslands. Steroid metabolism is a fundament to testicular development and function. The cytochrome P450, family 11, subfamily A, polypeptide 1 ( CYP11A1 ) is a key rate-limiting enzyme for catalyzing the conversion of cholesterol to pregnenolone. However, despite its importance, what expression and roles of CYP11A1 possesses and how it regulates the testicular development and spermatogenesis in Tibetan sheep remains largely unknown. Based on this, we evaluated the expression and localization patterns of CYP11A1 in testes and epididymides of Tibetan sheep at three developmental stages (three-month-old, pre-puberty; one-year-old, sexual maturity and three-year-old, adult) by quantitative real-time PCR (qPCR), western blot and immunofluorescence. The results showed that CYP11A1 mRNA and protein were expressed in testes and epididymides throughout the development stages and obviously more intense in one- and three-year-old groups than three-month-old group (except for the caput epididymidis). Immunofluorescence assay showed that the CYP11A1 protein was mainly located in Leydig cells and epididymal epithelial cells. In addition, positive signals of CYP11A1 protein were observed in germ cells, epididymal connective tissue and sperms stored in the epididymal lumen. Collectively, these results suggested that the CYP11A1 gene might be mainly involved in regulating spermatogenesis and androgen synthesis in developmental Tibetan sheep testis and epididymis.

Due to the lack of monitoring systems and water purification facilities, residents in western China may face the risk of drinking water pollution. Therefore, 673 samples were collected from Lhasa's agricultural and pastoral areas to reveal the status quo of drinking water. We used inductively coupled plasma-mass spectrometry to determine trace elements concentrations for water quality appraisal, source apportionment, and health risk assessment. The results indicate that concentrations of V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Cd, Ba, and Pb are below the guidelines, while As concentrations in a few samples exceed the standard. All samples were classified into "excellent water" for drinking purpose based on Entropy-weighted water quality index. Thereafter by principal component analysis, three potential sources of trace elements were extracted, including natural, anthropogenic, and mining activities. It is worth noting that geotherm and mining exploitation does not threaten drinking water safety. Finally, health risks were assessed using Monte Carlo technique. We found that the 95th percentiles of hazard index are 1.80, 0.80, and 0.79 for children, teenagers, and adults, indicating a non-carcinogenic risk for children, but no risks for the latter two age groups. In contrast, the probabilities of unacceptable cautionary risk are 7.15, 2.95 and 0.69% through exposure to Cr, Ni, As, and Cd for adults, children, and teenagers. Sensitivity analyses reveal As concentration and ingestion rate are most influential factors to health risk. Climate warming is predicted to considerably affect variations in soil organic carbon (SOC), especially in alpine ecosystems. Microbial necromass carbon (MNC) is an important contributor to stable soil organic carbon pools. However, accumulation and persistence of soil MNC across a gradient of warming are still poorly understood. An 8-year field experiment with four levels of warming was conducted in a Tibetan meadow. We found that low-level (+0-1.5°C) warming mostly enhanced bacterial necromass carbon (BNC), fungal necromass carbon (FNC), and total MNC compared with control treatment across soil layers, while no significant effect was caused between high-level (+1.5-2.5°C) treatments and control treatments. The contributions of both MNC and BNC to soil organic carbon were not significantly affected by warming treatments across depths. Structural equation modeling analysis demonstrated that the effect of plant root traits on MNC persistence strengthened with warming intensity, while the influence of microbial community characteristics waned along strengthened warming. Overall, our study provides novel evidence that the major determinants of MNC production and stabilization may vary with warming magnitude in alpine meadows. Decades of intensifying human activities have caused dramatic changes in land use and land cover (LULC) in the ecologically fragile areas of the Qinghai-Tibet Plateau, which have led to significant changes in ecosystem service value (ESV). Taking the ecologically fragile Sanjiangyuan region of the Qinghai-Tibet Plateau as the research object, we focused on understanding the impact of LULC changes on the Sanjiangyuan's landscape pattern and its corresponding ESV, which was combined with a Markov-Plus model to predict LULC changes in 2030. The results showed: (1) from 2000 to 2020, the LULC of Sanjiangyuan has changed to varying degrees, respectively. In the central and southern regions where animal husbandry is the mainstay activity, the area of grass land converted to bareland had expanded; (2) from 2000 to 2010, the total regional ESV increased sharply. However, the total amount of ESV decreased from 2010 to 2020; (3) the overall ESV in the study area was observed to be trending down and is expected to decrease by approximately 4.25 billion CNY by 2030; (4) the fragmentation and complexity of regional landscape patterns will negatively affect local ecosystem stability and biodiversity. Overall, there is a strong temporal and spatial correlation between LULC and ESV. Glycolysis in Sertoli cells (SCs) can provide energy substrates for the development of spermatogenic cells. Triose phosphate isomerase 1 (TPI1) is one of the key catalytic enzymes involved in glycolysis. However, the biological function of TPI1 in SCs and its role in glycolytic metabolic pathways are poorly understood. On the basis of a previous research, we isolated primary SCs from Tibetan sheep, and overexpressed TPI1 gene to determine its effect on the proliferation, glycolysis, and apoptosis of SCs. Secondly, we investigated the relationship between TPI1 and miR-1285-3p, and whether miR-1285-3p regulates the proliferation and apoptosis of SCs, and participates in glycolysis by targeting TPI1. Results showed that overexpression of TPI1 increased the proliferation rate and decreased apoptosis of SCs. In addition, overexpression of TPI1 altered glycolysis and metabolism signaling pathways and significantly increased amount of the final product lactic acid. Further analysis showed that miR-1285-3p inhibited TPI1 by directly targeting its 3'untranslated region. Overexpression of miR-1285-3p suppressed the proliferation of SCs, and this effect was partially reversed by restoration of TPI1 expression. In summary, this study shows that the miR-1285-3p/TPI1 axis regulates glycolysis in SCs. Human cystic echinococcosis (CE) is known to be endemic in the Tibet Autonomous Region (TAR), China; however, there is relatively little data from hospital records or community prevalence studies, and the situation regarding occurrence of human alveolar echinococcosis (AE) is unclear. Here we review the available reports about human echinococcosis in the seven prefectures of TAR. In addition, two pilot studies by mass screening using ultrasound (with serology) were undertaken (2006/7) in Dangxiong County of Lhasa Prefecture (north central TAR) and Dingqing County of Changdu Prefecture (eastern TAR). In Dangxiong County a prevalence of 9.9% (55/557) for human CE was obtained but no human AE cases were detected. By contrast, in Dingqing County (N= 232 persons screened), 11 CE cases (4.7%) and 12 AE cases (5.2%) (including one mixed CE and AE case) were diagnosed by ultrasound. Hospital records and published reports indicated that CE cases were recorded in all of seven prefectures in Tibet Autonomous Region, and AE cases in four prefectures. Incidence rates of human CE were estimated to range from 1.9 to 155 per 100,000 across the seven prefectures of TAR, with a regional incidence of 45.1 per 100,000. Incidence of AE was estimated to be between 0.6 and 2.8 cases per 100,000. Overall for TAR, human AE prevalence appeared relatively low; however, the pilot mass screening in Dingqing in eastern TAR indicated that human AE disease is a potential public health problem, possibly similar to that already well described in Tibetan communities bordering TAR in north-west Sichuan and south-west Qinghai provinces.
