Plant phenological shifts on the Qinghai-Tibetan Plateau (QTP) have gained considerable attention over the last few decades. However, temporal changes in plant autumn phenology and the main driving factors remain uncertain. Most previous studies used satellite-derived phenological transition dates and climatic statistics during the preseason, which have relatively large uncertainties and may mask some important climate change characteristics at the intra-annual scale, thus affecting exploration of the underlying phenological change causes. This study collected 1685 phenological records at 27 ground stations on the QTP during 1983-2017. Temporal change trends and break points in leaf senescence date (LSD) of 23 herbaceous species were assessed using least squares regression, a meta-analysis procedure, and the Pettitt test. The main drivers and causes were investigated through correlation analysis and contribution calculation based on LSD observations and monthly climatic data. Results showed that, LSD of QTP herbaceous plants was significantly delayed at a rate of 4.45 days/decade during 1983-2017. Break points were concentrated during 1999-2003, with an overall mean in 2001. After 2001, the delay trend in LSD decreased, falling from 5.26 days/decade to 2.54 days/decade. Air temperature and precipitation were the most important climatic factors that showed closer and more extensive correlations with LSD and greater contributions to the inter-annual variations in LSD. August and September were the most critical period during which climatic factors had higher contributions to the LSD shifts. However, August was drier, with precipitation significantly decreasing and temperature increasing, and September was colder after 2001. Therefore, the declining trend in LSD may be attributed to the drier August and colder September. Plant photosynthesis plays an essential role in regulating the global carbon cycle. Therefore, it is essential to understand the limitations imposed by climate on plant photosynthesis to comprehend the impacts of climate change on land carbon dynamics. In this study, taking gross primary productivity as a direct representation of photosynthesis, we employed a light use efficiency model (i.e., the revised EC-LUE) and factorial analysis method to quantify the spatiotemporal variation of temperature- and water-limitations on plant photosynthesis over the Tibetan Plateau (TP) grasslands during growing season (May to October) in 1983-2018. Results revealed a clear spatiotemporal pattern of the temperature- and water-limitations: temperature is the primary climatic limiting factor in the eastern TP, while water is the primary climatic limiting factor in the western TP; the water- and temperature-limitations prevail in summer and spring/autumn, respectively. The water- and temperature-limitations intensified and alleviated, respectively, during 1983 through 2018. There also was a widespread shift from temperature-limitation to water-limitation in the TP, particularly in midsummer (August). Long-range transport and atmospheric deposition of gaseous mercury (Hg 0 ) result in significant accumulation of Hg in the Qinghai-Tibetan Plateau (QTP). However, there are significant knowledge gaps in understanding the spatial distribution and source contribution of Hg in the surface soil of the QTP and factors influencing Hg accumulation. In this study, we comprehensively investigated Hg concentrations and isotopic signatures in the QTP to address these knowledge gaps. Results show that the average Hg concentration in the surface soil ranks as follows: forest (53.9 ± 36.9 ng g -1 ) > meadow (30.7 ± 14.3 ng g -1 ) > steppe (24.5 ± 16.1 ng g -1 ) > shrub (21.0 ± 11.6 ng g -1 ). Hg isotopic mass mixing and structural equation models demonstrate that vegetation-mediated atmospheric Hg 0 deposition dominates the Hg source in the surface soil, with an average contribution of 62 ± 12% in forests, followed by 51 ± 10% in shrub, 50 ± 13% in steppe, and 45 ± 11% in meadow. Additionally, geogenic sources contribute 28-37% of surface soil Hg accumulation, and atmospheric Hg 2+ inputs contribute 10-18% among the four types of biomes. The Hg pool in 0-10 cm surface soil over the QTP is estimated as 8200 ± 3292 Mg. Global warming, permafrost degradation, and anthropogenic influences have likely perturbed Hg accumulation in the soil of QTP.

Microplastics (MPs) are ubiquitous in the aquatic environment and have received widespread attention worldwide as emerging pollutants. Urbanization and anthropogenic activities are the main sources of MPs in rivers; however, the MPs in plateau rivers with less human activities are not well understood. In this study, the pollution of MPs in the surface water and shore sediment of the Lhasa River from the Qinghai-Tibet Plateau was investigated, and a risk assessment was conducted. The abundance of MPs in the surface water and shore sediment of Lhasa River were 0.63 n/L and 0.37 n/g, respectively. MPs in surface water were mainly dominated by films (43.23%) and fibers (31.12%) in shape, transparent (54.25%) in color, and 0-0.5 mm (75.83%) in size, while MPs in the shore sediment were mainly fibers (43.69%) and fragments (36.53%), transparent (71.91%), and 0-0.5 mm (60.18%). PP and PE were the predominant polymer types, accounting for 44.55% and 30.79% respectively in the surface water and 32.51% and 36.01% respectively in the shore sediment. More notably, the polymer pollution index (H) of MPs in the Lhasa River was at hazard level III due to the high risk caused by PVC, but the pollution load index (PLI) was low at hazard level I. Microorganisms play critical ecological roles in the global biogeochemical cycles. However, extensive information on the microbial communities in Qinghai-Tibet Plateau (QTP), which is the highest plateau in the world, is still lacking, particularly in high elevation locations above 4500 m. Here, we performed a survey of th e soil and water microbial communities in Bamucuo Lake, Tibet, by using shotgun metagenomic methods. In the soil and water samples, we reconstructed 75 almost complete metagenomic assembly genomes, and 74 of the metagenomic assembly genomes from the water sample represented novel species. Proteobacteria and Actinobacteria were found to be the dominant bacterial phyla, while Euryarchaeota was the dominant archaeal phylum. The largest virus, Pandoravirus salinus, was found in the soil microbial community. We concluded that the microorganisms in Bamucuo Lake are most likely to fix carbon mainly through the 3-hydroxypropionic bi-cycle pathway. To understand the mercury (Hg) biogeochemical cycle in alpine regions under global warming, it is critical to identify the distribution and sources of Hg in aquatic ecosystems of the Tibet Plateau. The spatial distribution pattern and potential sources of Hg species including total mercury (THg), particulate mercury (PHg), and dissolved mercury (DHg) were investigated in surface waters of the Yangtze and Yellow River source basins during the wet season. The results showed that average ρ (DHg) in surface water of the Yangtze and Yellow River source basins were comparable[(2.96±1.26) ng·L -1 and (2.47±0.83) ng·L -1 , respectively], whereas the average ρ (THg)[(10.69±11.14) ng·L -1 ] and ρ (PHg)[(8.46±11.41) ng·L -1 ] in the source basin of the Yangtze River were significantly higher than that in surface water of the Yellow River source basin[(3.37±2.03) ng·L -1 and (1.13±1.02) ng·L -1 , respectively]. It is worth noting that the ecological risk of Hg in the study area was limited because of low Hg concentration and methylation level. In addition, the correlation analysis illustrated that the THg was mainly concentrated by PHg in the source basin of the Yangtze River. Specifically, the concentration variations in Hg were mainly affected by the input of glacier meltwater, soil erosion, and precipitation. By contrast, the main species of Hg in the source basin of the Yellow River was DHg, the distribution pattern of which was mainly controlled by DOC. Spatially, a significant negative correlation was found between ρ (PHg) and longitude in the source basin of the Yangtze River ( R 2 =0.46, P <0.01). The spatial distribution differences of river slope and soil erosion intensity were identified as the key factors leading to the decreasing trend of ρ (PHg) and ρ (THg) along the river flow in the source basin of the Yangtze River. The results of the PMF model further demonstrated that in the surface waters of the source regions of the Yangtze and Yellow Rivers, 51.4% of Hg derived from long-distance atmospheric deposition, 38.8% from erosion of soil rock or sediment via stream flow, and 9.7% from soil runoff or seepage input.

Disentangling the biogeographic patterns of rare and abundant microbes is essential in order to understand the generation and maintenance of microbial diversity with respect to the functions they provide. However, little is known about ecological assembly processes and environmental adaptation of rare and abundant microbes across large spatial-scale wetlands. Using Illumina sequencing and multiple statistical analyses, we characterized the taxonomic and phylogenetic diversity of rare and abundant bacteria and fungi in Qinghai-Tibet Plateau wetland soils. Abundant microbial taxa exhibited broader environmental thresholds and stronger phylogenetic signals for ecological traits than rare ones. By contrast, rare taxa showed higher sensitivity to environmental changes and closer phylogenetic clustering than abundant ones. The null model analysis revealed that dispersal limitation belonging to stochastic process dominated community assemblies of abundant bacteria, and rare and abundant fungi, while variable selection belonging to deterministic process governed community assembly of rare bacteria. Neutral model analysis and variation partitioning analysis further confirmed that abundant microbes were less environmentally constrained. Soil ammonia nitrogen was the crucial factor in mediating the balance between stochasticity and determinism of both rare and abundant microbes. Abundant microbes may have better environmental adaptation potential and are less dispersed by environmental changes than rare ones. Our findings extend knowledge of the adaptation of rare and abundant microbes to ongoing environmental change and could facilitate prediction of biodiversity loss caused probably by climate change and human activity in the Qinghai-Tibet Plateau wetlands., (© 2021 John Wiley & Sons Ltd.)

As global climate continues to warm, melting of glaciers releases a large quantity of mercury (Hg) originally locked in ice into the atmosphere and downstream ecosystems. Here, we show an opposite process that captures atmospheric Hg through glacier-to-vegetation succession. Our study using stable isotope techniques at 3 succession sites on the Tibetan Plateau reveals that evolving vegetation serves as an active "pump" to take up gaseous elemental mercury (Hg 0 ) from the atmosphere. The accelerated uptake enriches the Hg pool size in glacier-retreated areas by a factor of ∼10 compared with the original pool size in the glacier. Through an assessment of Hg source-sink relationship observed in documented glacier-retreated areas in the world (7 sites of tundra/steppe succession and 5 sites of forest succession), we estimate that 400 to 600 Mg of Hg has been accumulated in glacier-retreated areas (5‰ of the global land surface) since the Little Ice Age (∼1850). By 2100, an additional ∼300 Mg of Hg will be sequestered from the atmosphere in glacier-retreated regions globally, which is ∼3 times the total Hg mass loss by meltwater efflux (∼95 Mg) in alpine and subpolar glacier regions. The recapturing of atmospheric Hg by vegetation in glacier-retreated areas is not accounted for in current global Hg models. Similar processes are likely to occur in other regions that experience increased vegetation due to climate or land use changes, which need to be considered in the assessment of global Hg cycling., Competing Interests: The authors declare no competing interest.

Objectives: Meconopsis integrifolia (M. integrifolia) is one of the most popular members in Traditional Tibetan Medicine. This study aimed to investigate the anticancer effect of M. integrifolia and to detect the underlying mechanisms of these effects., Methods: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay and trypan blue assay were used to evaluate the cytotoxicity of M. integrifolia. Changes in cell nuclear morphology and reactive oxygen species (ROS) level were observed by fluorescent microscopy. Apoptosis ratio, DNA damage and mitochondrial membrane potential (MMP) loss were analyzed by flow cytometry. Western blotting assay was adopted to detect the proteins related to apoptosis. Immunofluorescence was used to observe the release of cytochrome C., Results: The obtained data revealed that M. integrifolia could significantly inhibit K562 cell viability, mainly by targeting apoptosis induction and cell cycle arrest in G2/M phase. Collapse in cell morphology, chromatin condensation, DNA damage and ROS accumulation were observed. Further mechanism detection revealed that mitochondrion might be a key factor in M. integrifolia-induced apoptosis., Conclusions: M. integrifolia could induce mitochondria mediated apoptosis and cell cycle arrest in G2/M phase with little damage to normal cells, suggesting that M. integrifolia might be a potential and efficient anticancer agent that deserves further investigation.
