Thailand is a tropical country affected by global climate change and has high temperatures and humidity that cause heat stress in livestock. A temperature–humidity index (THI) is required to assess and evaluate heat stress levels in livestock. One of the livestock types in Thailand experiencing heat stress due to extreme climate change is crossbred dairy cattle. Genetic evaluations of heat tolerance in dairy cattle have been carried out for reproductive traits. Heritability values for reproductive traits are generally low (<0.10) because environmental factors heavily influence them. Consequently, genetic improvement for these traits would be slow compared to production traits. Positive and negative genetic correlations were found between reproductive traits and reproductive traits and yield traits. Several selection methods for reproductive traits have been introduced, i.e., the traditional method, marker-assisted selection (MAS), and genomic selection (GS). GS is the most promising technique and provides accurate results with a high genetic gain. Single-step genomic BLUP (ssGBLUP) has higher accuracy than the multi-step equivalent for fertility traits or low-heritability traits. [ABSTRACT FROM AUTHOR]

Grandparent roosters are crucial in poultry breeding programs and significantly influence future bird generations' genetic makeup and performance. However, these roosters face considerable challenges from heat stress, which can adversely affect their reproductive performance, semen quality, and overall health and welfare. Our study aimed to investigate the effects of heat stress on the genetics of semen characteristics, identify the appropriate temperature and humidity indices (THI), and determine the threshold point of heat stress to prevent thermal stress. We analyzed data from 3,895 records of 242 Thai native grandparent roosters in conjunction with the THI using 7 THI functions and the regression method. The threshold point of heat stress, genetic parameters, rate of decline of semen characteristics per level of THI, estimated breeding values and selection index values were analyzed using the multivariate test-day model in the AIREML and BLUPF90 programs. Based on the regression coefficient and statistical criteria of the lowest -2logL and AIC values, the results showed that a THI of 78 was considered the threshold point of heat stress. The estimated heritability values ranged from 0.023 to 0.032, 0.066 to 0.069, 0.047 to 0.057, and 0.022 to 0.024 for mass movement, semen volume, sperm concentration, and the semen index, respectively. The reduction rates of mass movement, semen volume, sperm concentration, and semen index at a THI of 78 were -0.009, -0.003, -0.170, and -0.083 per THI, respectively. The genetic correlations among the semen traits were moderately to strongly positive and ranged from 0.562 to 0.797. The genetic correlations between semen traits and heat stress were negative and ranged from -0.437 to -0.749. The permanent environmental correlations among the semen traits (0.648-0.929) were positive and greater than the genetic correlations. Permanent environmental correlations between semen traits and heat stress were negative and ranged from -0.539 to -0.773. The results of the selection indices showed that the higher the selection intensity was, the greater the degree to which the selection index corresponded to genetic progress. The recommendation for animal genetic selection is that the top 10% is appropriate because it seems most preferred. Thai indigenous roosters are exposed to unsuitable temperatures and humidity, resulting in a lower reproductive potential. Kaempferia parviflora (KP) extract containing methoxyflavones was fed to roosters to improve their reproductive performance. Thirty-two Thai native roosters were orally administered KP extract at 300, 450, and 600 mg, calculated according to their average body weight, for at least 14 d before semen collection and continued supplementation until the end of the experiment. The nonsupplemented group served as the control. Fresh semen in terms of semen volume, sperm concentration, mass movement score, and sperm viability were evaluated. Semen preservation at 5°C and fertility test were examined for total motility (MOT), progressive motility (PMOT), sperm viability, and lipid peroxidation up to 48 h of storage. Testosterone concentrations and testicular function were also determined. The results showed that the highest sperm concentration and sperm motility of fresh semen were observed in KP extract at 600 mg (P < 0.001). KP extract at 600 mg resulted in higher sperm viability than the control and KP extract at 300 mg (P < 0.05), but was not different from KP at 450 mg (P > 0.05). The highest MOT, PMOT, and viability were found in the roosters that received 600 mg oral KP extract (P < 0.05), while those of the roosters that received oral KP extract 300 mg and the control were the lowest (P < 0.05) at all storage times. Lipid peroxidation was significantly lower in the KP extract up to 24 h (P < 0.05). The fertility and hatchability of the KP extract at 600 mg at T48 showed a minor decrease compared to the control at T0. These results might be inferred as a result of good spermatogenesis, as revealed by the results of histological examination and testosterone activity. Skin hyperpigmentation is an aesthetic problem that leads to psychosocial issues. Thus, skin whitening agents from agro- and poultry-industrial co-products are considered high economic value ingredients of interest for sustainable application. Therefore, this study aimed to determine the cosmeceutical potential of anserine/carnosine-rich chicken extract (ACCE) from the Thai native chicken Pradu Hang Dam Mor Kor 55 (PD) meat. The chemical composition was identified and quantified using the HPLC-UV method. Then, the antioxidation potential of the extract was compared to that of L-anserine and L-carnosine, using 1,1-diphenyl-2-picrylhydrazyl assay and shikonin-induced production of reactive oxygen species in CCD-986Sk cell models, and the anti-melanogenesis effect in the MNT-1 melanoma cell line model was investigated. Furthermore, related mechanisms were identified using colorimetric tyrosinase assay and the Western blot technique. The ACCE was composed of L-anserine and L-carnosine as two major constituents. In a dose-dependent manner, ACCE, L-anserine, and L-carnosine manifested significant antioxidation potential and significant reduction of melanin production. Activation of the extracellular signal-regulated kinase (ERK) signaling pathway and inhibition of tyrosinase activity of ACCE were demonstrated as the mechanisms of the anti-melanogenesis effect. In conclusion, ACCE has been revealed as a potential cosmeceutical agent due to its antioxidation and anti-melanogenic activity in association with L-anserine and L-carnosine composition and biomolecular regulating ability. Therefore, further studies and development should be considered to support the utilization of anserine/carnosine-rich chicken extract in the cosmetic industry for economic value creation and sustainability.

This study aimed to evaluate the effect of a high protein diet comprising breast meat from commercial broiler (BR), Thai native (PD), and commercial broiler × Thai native crossbred (KKU-ONE) chicken on serum uric acid, biochemical parameters, and antioxidant activities in rats. Male Sprague-Dawley rats were divided into four groups. The control group received a standard chow diet, and the other three groups were fed a high protein diet (70% standard diet + 30% BR, PD, or KKU-ONE chicken breast) for five weeks. The PD- and KKU-ONE-fed rats had lower plasma total cholesterol and triglyceride levels than the control rats. A decrease in HDL-c was also observed in rats fed a diet containing BR. Liver weight, liver enzyme, plasma ALP, xanthine oxidase activity, serum uric acid, creatinine, superoxide production, and plasma malondialdehyde levels increased in BR-fed rats. Plasma membrane lipids are the key factor in the ability of chicken sperm to be frozen. They ensure fluidity and flexibility of the sperm membrane for effective viability and motility during in vitro storage. The objective of this study was to compare the lipid profiles of different native and commercial chicken breeds: native Thai (Pradu Hang Dam) roosters and commercial (Rhode Island Red) roosters, with respect to their frozen sperm quality. In addition, lipid peroxidation and antioxidant enzymes superoxidase dismutase (SOD) and catalase (CAT) were also examined. Semen was collected from 12 roosters of each breed. For fresh semen, parameters assessed include semen volume, pH, sperm concentration, sperm motility, and viability, while for frozen semen, the parameters assessed were sperm motility and viability. Moreover, other parameters assessed included malondialdehyde (MDA) concentration, activities of SOD and CAT, and fatty acid profile. We found that sperm viability and motility of frozen semen were higher in the commercial breed than in the native breed (P < 0.05). The commercial chicken breed had higher MDA concentration than the native breed (P < 0.05), but antioxidant enzymes remained unchanged in both. Levels of arachidonic acid (AA; C20:4n-6) and docosahexaenoic acid (DHA; C22:6n-3) were significantly higher (P < 0.05) in the commercial than in the native breed; however, n-6 to n-3 ratios were not different. In conclusion, our study found that lipid profiles have an influence on frozen sperm viability and motility between the breeds. Polyunsaturated fatty acids, particularly AA and DHA, are beneficial to sperm quality.

Oxidative stress due to cryopreservation has been considered as a major factor in sperm damage. Supplementation of the diet with different concentrations of organic selenium has been proposed to improve the quality of fresh and frozen-thawed semen in different breeds of roosters. Sixteen Pradu Hang Dum (Thai native) and 16 Rhode Island Red roosters were used in this study. Four levels of selenium supplementation between 0 and 0.9 ppm were examined. After 14 days of feeding, semen samples were collected twice a week and the fresh semen was evaluated. Then semen from each group was pooled and cryopreserved. The fertility of frozen-thawed semen was determined by inseminating 48 layer hens. Supplementation of diets with 0.3, 0.6 and 0.9 ppm selenium improved the fresh semen in terms of sperm viability and normal morphology (P < 0.01). Sperm concentration increased (quadratically, P < 0.001) with increasing dietary selenium levels. Meanwhile, post-thawed semen quality in terms of sperm motility, viability, live with intact acrosome and functioning mitochondria improved significantly with selenium treatments of 0.6 and 0.9 ppm, and lipid peroxidation was decreased (P < 0.001) and fertility improved (P < 0.05) with those levels of selenium treatment. In addition, there were differences between breeds with respect to some fresh or frozen semen quality parameters (P < 0.05). In conclusion, the breed affected both fresh and frozen semen. Even there were no statistically significant differences in the parameters from groups 0.6 and 0.9 ppm on frozen-thawed semen quality, but the highest sperm concentration was found in 0.6 ppm. The objectives of this study were to compare the efficiency of a split single injection of follicle-stimulating hormone (FSH) given by either intramuscular (split-single IM) or ischiorectal fossa (split-single IRF) injection to the traditional treatment and to determine the concentrations of FSH. The temperature and humidity index (THI) values were interpreted together with the ovarian responses and embryo characteristics. The ovarian responses in the split-single IRF group were similar to those of the control group (p > .05) but higher compared with the split-single IM group (p < .05). Higher peak levels of plasma FSH in the split-single IRF group did not differ compared with the control group (p > .05) but were lower in split-single IM administration (p < .05). The results showed a significant decrease in the numbers of large follicles and corpora lutea (CLs) in the moderate THI compared with low and high THI (p < .05). The high THI affected ovulation rate as well as the numbers of transferable embryos and degenerated embryos (p < .05). In conclusion, the split-single IRF administration had a comparable superovulatory response to the traditional twice-daily protocol.
