Brassica oleracea landraces are widely grown in northwestern Spain but also in other regions of the Spanish territory like the Canary Islands, where environmental conditions are very different from the peninsular ones. A collection of B. oleracea accessions from different Spanish locations is kept at the germplasm bank of Misión Biológica de Galicia (MBG, Spain). Considering that B. oleracea is the most diverse species within the Brassica genus, the study of these accessions is crucial to ensure the availability for breeding programs as a genetic variability source. In order to assess the genetic and biochemical diversity across this collection, we used simple sequence repeats (SSR) as molecular markers and the glucosinolate content and profile as a chemical marker. We concluded that there are significant genetic differences among accessions and that they are grouped by the same geographical provenance. Furthermore, different glucosinolate profiles were observed between peninsular and insular accessions, thus supporting the results obtained in the genetic analysis. The bioactive compounds, as well as the in vitro antioxidant activity of two Savoy cabbage cultivars, Dama and Leticia, grown in west of Spain under similar conditions were identified, quantified and compared. We found that cv. Dama presented in general betters results when compared with cv. Leticia. Cv. Dama presented higher concentrations of chlorophyll a (2.26 mg/100 g fresh weight), total phenolic content (102.71 mg of chlorogenic acid equivalent/100 g fresh weight) and total intact glucosinolates (195.22 µmol of sinigrin equivalent/100 g fresh weight). In northwest Spain and Portugal, there is a long tradition of cultivating B. rapa subsp. rapa to obtain turnip greens and turnip tops. Brassica rapa L. subsp. rapa (turnip greens and turnip tops) were grown under conventional and organic conditions in two Farms in southern Spain. Glucosinolatescontents were higher in Brassicas grown under conventional conditions than those grown under organic ones. Average Ca total and bioaccessible contents ranged between 14.6–23.4 mg/g; 8.9–12.0 mg/g for turnip greens and 6.4–8.9 mg/g; 4.3–4.8 mg/g for turnip tops. According to these concentrations, an intake of 100–200 g (fresh weight) of the studied Brassica rapa fulfills Ca dietary reference intakes (DRI) (considering the total content data) and complies with 72–100% Ca DRI percentage (considering the bioaccessible data). Se concentrations ranged between 0.061–0.073 µg/g and 0.039–0.053 µg/g for turnip greens and turnip tops respectively. Se bioaccessibility values were high, with percentages of around 90%. Finally, the total glucosinolate content ranged between 13.23–21.28 µmol/g for turnip greens and 13.36–20.20 µmol/g for turnip tops. In general, the bioaccessibility of the total glucosinolates analyzed in this study was high, with mean values of around 73% and 66% for turnip greens and turnip tops, respectively. Brassica rapa vegetables grown under both organic and conventional conditions in southern Spain are an excellent dietary source of Ca, Se, and glucosinolates with a high bioaccessibility. Abstract: Brassica rapa rapa group is widely distributed and consumed in northwestern Spain. The consumption of Brassica vegetables has been related to human health due to their phytochemicals, such as glucosinolates and phenolic compounds that induce a variety of physiological functions including antioxidant activity, enzymes regulation and apoptosis control and the cell cycle. For first time in Brassica crops, intact glucosinolates and phenolic compounds were simultaneously identified and characterized. Twelve intact glucosinolates, belonging to the three chemical classes, and more than 30 phenolic compounds were found in B. rapa leaves and young shoots (turnip greens and turnip tops) by LC–UV photodiode array detection (PAD)–electrospray ionization (ESI). The main naturally occurring phenolic compounds identified were flavonoids and derivatives of hydroxycinnamic acids. The majority of the flavonoids were kaempferol, quercetin and isorhamnetin glycosylated and acylated with different hydroxycinnamic acids. Quantification of the main compounds by HPLC-PAD showed significant differences for most of compounds between plant organs. Total glucosinolate content value was 26.84μmolg−1 dw for turnip greens and 29.11μmolg−1 dw for turnip tops; gluconapin being the predominant glucosinolate (23.2μmolg−1 dw). Phenolic compounds were higher in turnip greens 51.71μmolg−1 dw than in turnip tops 38.99μmolg−1 dw, in which flavonols were always the major compounds. [Copyright &y& Elsevier]

Objective:To assess the intake of glucosinolates and cruciferous vegetables among Spanish adults.Design:Cross-sectional analysis of a prospective cohort study.Setting:The Spanish cohort of the European Prospective Investigation into Cancer and Nutrition (EPIC).Subjects:We analysed data from 40 684 men and women aged 35–64 years from the EPIC-Spain cohort. The usual diet was assessed by means of the dietary history method, and glucosinolate intake was calculated using a published food composition database.Results:The average intake of cruciferous vegetables was 11.3 g/day, accounting for about 5% of total vegetable consumption, whereas the daily intake of total glucosinolates was 6.5 mg, among which 35% were of indole type. The absolute intake of glucosinolates was in average higher in men than in women (6.8 vs 6.2 mg/day), whereas glucosinolate density per energy unit was higher in women's diet (3.4 vs 2.7 mg/4200 kJ). Northern regions consumed in average 36% more glucosinolates than Southern regions (7.3 vs 5.4 mg/day). Abstract: Brassica oleracea L. crops including kales, cabbages, and Tronchuda cabbages are widely grown in northwestern Spain and Portugal but little information is available on leaf glucosinolate content of these crops. The objectives were to determine the diversity for the total glucosinolate content and profile on leaves in a collection of 153 kales, 26 cabbages, and three Tronchuda cabbages varieties grown at two growing seasons and to determine the seasonal variation of glucosinolates in cabbages and Tronchuda cabbage varieties. Sinigrin, glucoiberin, and glucobrassicin were the major glucosinolates found in kales. Glucoiberin was the most common glucosinolate in Tronchuda cabbages in both planting seasons and in cabbages sown in fall season whereas glucobrassicin and glucoiberin were the most common glucosinolates in cabbages in spring season. In kales the total glucosinolate content ranged from 11.0 to 53μmolg−1 dw, with a mean value of 26.3μmolg−1 dw. Four kale varieties (MBG-BRS0468, MBG-BRS0476, MBG-BRS0060 and MBG-BRS0223) showed the highest total sinigrin or glucobrassicin contents. So, they could be good candidates for future breeding programs. In cabbages, the total glucosinolate content ranged from 10.9 to 27g−1 dw. Total glucosinolate concentration during spring sowing (22μmg−1 dw) was higher than those in fall sowing (13μmg−1 dw). Regarding both high glucosinolate content and the agronomic value, MBG-BRS0057 and MBG-BRS0074 could be good sources of beneficial glucosinolates. The presence of high concentrations of sinigrin, glucoiberin, and glucobrassicin warrant further search into their potential use to enhance the level of these important phytochemicals in these edible crops. Abstract: Glucosinolate levels in leaves were determined in a collection of 113 varieties of turnip greens (Brassica rapa L.) from northwestern Spain grown at two sites. Sensorial attributes were also assessed by a consumer panel. The objectives were to determine the diversity among varieties in total glucosinolate content and glucosinolate profile and to evaluate their sensory attributes in relation to glucosinolate content for breeding purposes. Sixteen glucosinolates were identified, being the aliphatic glucosinolates, gluconapin and glucobrassicanapin the most abundant. Other aliphatic glucosinolates, such as progoitrin, glucoalyssin, and gluconapoleiferin were relatively abundant in varieties with a different glucosinolate profile. Indolic and aromatic glucosinolate concentrations were low and showed few differences among varieties. Differences in total glucosinolate content, glucosinolate profile and bitterness were found among varieties, with a total glucosinolate content ranging from 11.8 to 74.0μmolg−1 dw at one site and from 7.5 to 56.9μmolg−1 dw at the other site. Sensory analysis comparing bitterness with variation in glucosinolate, gluconapin and glucobrassicanapin concentrations suggested that these compounds and their breakdown products are not the only determinants of the characteristic flavour of this vegetable. Other phytochemicals are probably involved on the characteristic bitter flavour. The varieties MBG-BRS0132, MBG-BRS0082, MBG-BRS0173, and MBG-BRS0184 could be good candidates for future breeding programs since they had high total glucosinolate content and good agronomic performance. The presence of glucoraphanin in some varieties should be studied more extensively, because this aliphatic glucosinolate is the precursor of sulforaphane, a potent anti-cancer isothiocyanate. Brassica rapa is grown in northwestern Spain to obtain turnip greens. The tops of the same plants (flower stems with buds) are cut and sell as turnip tops, increasing the value of the crop. This practice could be extended to other brassicas. The objectives of this work are to study the phytochemical potential of tops of coles (Brassica oleracea) and leaf rape (Brassica napus) compared to turnip tops and to compare tops of different coles (cabbage, kale, tronchuda cabbage), which differ in their morphology and use. We evaluated the content of glucosinolates and phenolic compounds and the antioxidant capacity in leaves and tops of the three species. We found that tops had higher amount of glucosinolates than leaves. Phenolic content and antioxidant capacity followed the opposite trend. Therefore, consumption of leaves and tops are complementary, since both type of organs are enriched with different types of compound. Local varieties of kale, curly kale, cabbage and curly leave cabbage are interesting because of their GSLs and phenolic content and antioxidant capacity in both leaves and tops. A comparative study for evaluating Italian and Spanish accessions of Brassica rapa var. rapa L., including turnip greens and turnip tops, was carried out at different locations with a view to determine the effect of earliness on crop production, antioxidant activity, glucosinolates amount, and profile (GLSs) and total phenolics content. The accessions evaluated were represented by two turnip top local varieties (one Italian variety and a Galician one), four new synthetic varieties established by Misión Biológica de Galicia (MBG-CSIC), and three commercial varieties widely used by growers in Galicia and in Italy. The results showed a great variability regarding flowering time, fresh and dry weight of the leaves and flower buds, and the branch number per plant. The highest turnip greens production was found in two synthetic varieties ("SIN07" and "SIN01") for both countries. Local varieties "BRS550" and "CM39" were also suitable for turnip greens production in Spain and Italy, respectively. For turnip tops, the highest production was found for "SIN07" in Spain, for "CM39" in Italy and for "BRS550" in both countries. We found a high diversity in the total and individual glucosinolate, phenolic content, and antioxidant activity among genotypes, geographical origins, and the different parts of the plant (leaf and flower). Varieties "SIN01" and "SIN07" showed the highest values in total GLSs, total aliphatic and gluconapin contents in turnip greens followed by the two commercial varieties. For turnip tops, the highest values in gluconapin, aliphatic, and total GLSs contents were found in "SIN01" and "BRS550". Even though different varieties stand out over the rest depending on the location, "SIN01", "SIN07", "CM39", and "BRS550" could be recommended for turnip greens production because of its high antioxidant activity. The study showed that the latest varieties are more productive and show higher bioactive compounds than the earlier ones and that it is possible to improve genotypes for different growing cycles. Total and individual glucosinolate (GSL) content of leaves of vegetable turnip rape ( Brassica rapa L. var. rapa) was determined in a set of 45 varieties consisting in early, medium and late types grown at two locations in northwestern Spain. The objectives were to determine the diversity among varieties in GSL content and to relate that variation with earliness and plant habit. Eight GSL were identified, being two aliphatic GSL, gluconapin (84.4 % of the total GSL) and glucobrassicanapin (7.2 % of the total GSL) the most abundant. Indolic and aromatic GSL content were low but also showed significant differences among varieties. Differences in total and individual GSL content were found among varieties, plant habit groups, and earliness groups. Total GSL content ranged from 19 to 37.3 μmol g dw in early and extra-late groups, respectively, and from 19.5 to 36.3 μmol g dw for turnips and turnip greens groups, respectively. These differences were consistent to values found for gluconapin content where the turnip group had the highest values (31.8 μmol g dw) and the turnip top group had the lowest (15.7 μmol g dw). Two varieties, MBG-BRS0429 and MBG-BRS0550 (from turnip greens and extra-late groups) and MBG-BRS0438 (from turnips and late groups), stood out as they had the highest total GSL content and could be used as a good source of these beneficial bioactive compounds. Elucidation of genetic diversity among crops can provide useful information to assist plant breeders to design improved breeding strategies in order to obtain varieties rich on GSL. Broccoli is a source of bioactive compounds that provide an important nutritional value. The content of these compounds can vary depending on agronomic and environmental conditions, as well as on elicitation. In this study, three crop trials were carried out to evaluate the effects of the cultivation season, the application of different dosages of methyl-jasmonate (MeJA) on the overall quality and on the total content of bioactive compounds of 'Parthenon' broccoli cultivated under the field conditions of southeastern Spain. Color parameters, chlorophyll content, total phenolic compounds, total flavonoids and antioxidant activity were measured to evaluate the overall quality. Moreover, individual carotenoids, phenolic compounds and glucosinolates were evaluated by high performance liquid chromatography with diode array detection (HPLC-DAD) and high performance liquid chromatography equipped with diode array detector coupled to mass spectrometer using electro spray ionization (HPLC-DAD-ESI/MSn). The content of total carotenoids, phenolic compounds and glucosinolates were higher in autumn compared with spring, showing increases of 2.8-fold, 2-fold and 1.2-fold, respectively. Moreover, a double application of MeJA increased the contents of total carotenoids, phenolic compounds and glucosinolates by 22%, 32% and 39%, respectively, relative to the untreated samples. Considering our results, the controlled and timely application of 250 µM MeJA to the aerial parts of the plants four days before harvest, on two consecutive days, seems to be a valid agronomic strategy to improve the health-promoting capacity of Parthenon broccoli, without compromising its overall quality.
