Toxoplasmosis is a parasitic zoonosis caused by Toxoplasma gondii which infects warm‐blooded species worldwide. Humans can be infected through ingestion of tissue cysts from raw or undercooked meat, including game meat. A nationwide large‐scale cross‐sectional study was conducted to assess exposure to T. gondii in seven wild ruminant species in Spain. A total of 2,040 serum samples from 77 sampling sites randomly distributed in the five bioregions (BRs) covering mainland Spain were tested for antibodies against T. gondii using the modified agglutination test. The overall seroprevalence was 22.0% (449/2,040). Seroprevalence by species in decreasing order was as follows: 39.6% (141/356) in roe deer (Capreolus capreolus), 37.1% (138/372) in fallow deer (Dama dama), 16.6% (92/553) in red deer (Cervus elaphus), 14.0% (26/186) in Southern chamois (Rupicapra pyrenaica), 11.5% (24/209) in mouflon (Ovis aries musimon), 7.8% (27/346) in Iberian wild goat (Capra pyrenaica) and 5.6% (1/18) in Barbary sheep (Ammotragus lervia). Seropositivity was detected in 74.0% (57/77) of the sampling sites. Results indicate widespread but not homogeneous exposure to T. gondii in wild ruminant populations in Spain during the last two decades and highlight differences related to animal species and spatial distribution of these species in this country; this implies potential consequences of T. gondii for animal health, conservation and public health. Barbary sheep (Ammotragus lervia) is a North African native wild Caprinae, introduced in the 1970s in new territories such as Spain, the USA, and Mexico. Here, we describe Sarcocystis species in Barbary sheep. Sarcocysts were found in 19 out of 56 adult A. lervia in Southern Spain and characterized morphologically and molecularly. By light microscopy, sarcocysts had thin (< 1 μm) or thick (> 2 μm) walls. By transmission electron microscopy, sarcocysts with thick walls had Type 14 villar protrusions corresponding to S. tenella/S. capracanis of domestic sheep (Ovis aries) or goats (Capra hircus). Sarcocysts with thin walls had Type 7b villar protrusions that corresponded to S. arieticanis/S. hircicanis of domestic sheep or goats. Molecular analyses allowed the identification of only thick-walled Sarcocystis species. Six sarcocysts were assigned to S. tenella (99.2–100% and 95.6–100% sequence similarity within 18S rRNA and COI, respectively) and 19 sarcocysts were assigned to S. capracanis (98.5–99.8% and 97.9–99.0% sequence similarity within 18S rRNA and COI, respectively). Further studies are needed for taxonomic identification of sarcocysts in Barbary sheep because Sarcocystis species in sheep and goats are not cross transmissible despite morphological similarities. Simple Summary: Toxoplasma gondii is a zoonotic intracellular parasite which infects a wide range of warm-blooded animals. Long-term studies provide the necessary perspective required to understand those processes which took place over many years in order to address epidemiology and ecology in complex host communities. This study is focused on evaluating what the main long-term determinants of the seroprevalence of T. gondii are in the wild ungulate community from Doñana National Park (southwestern Spain). With this purpose, we assayed sera from 1573 wild ungulates (wild boar, red deer, and fallow deer), collected for 13 years (from 2005 to 2018). We found high seroprevalence values of T. gondii (% ± CI 95%; wild boar 39 ± 3.3; red deer 30.7 ± 4.4; and fallow deer 29.7 ± 4.2. Several factors operating in the medium and long-term (individual, environmental, population and stochastic) explained the risk of T. gondii in wild boar and deer, some of them operating at the community level. Toxoplasma gondii is an obligate intracellular protozoan which infects warm-blooded vertebrates, including humans, worldwide. In the present study, the epidemiology of T. gondii was studied in the wild ungulate host community (wild boar, red deer, and fallow deer) of Doñana National Park (DNP, south-western Spain) for 13 years (2005–2018). We assessed several variables which potentially operate in the medium and long-term (environmental features, population, and stochastic factors). Overall, the wild ungulate host community of DNP had high seroprevalence values of T. gondii (STG; % ± confidence interval (CI) 95%; wild boar (Sus scrofa) 39 ± 3.3, n = 698; red deer (Cervus elaphus) 30.7 ± 4.4, n = 423; fallow deer (Dama dama) 29.7 ± 4.2, n = 452). The complex interplay of hosts and ecological/epidemiological niches, together with the optimal climatic conditions for the survival of oocysts that converge in this area may favor the spread of the parasite in its host community. The temporal evolution of STG oscillated considerably, mostly in deer species. The relationships shown by statistical models indicated that several factors determined species patterns. Concomitance of effects among species, indicated that relevant drivers of risk operated at the community level. Our focus, addressing factors operating at broad temporal scale, allows showing their impacts on the epidemiology of T. gondii and its trends. This approach is key to understanding the epidemiology and ecology to T. gondii infection in wild host communities in a context where the decline in seroprevalence leads to loss of immunity in humans. Abstract: Serum samples from 2970 (1400 sows, 1570 fattening) pigs, from 100 farms in the 10 main swine production regions in Spain were tested for antibodies against T. gondii by the modified agglutination test (MAT). Antibodies to T. gondii (MAT 1:25 or higher) were detected in 492 pigs (16.6%, 9.7% in fattening pigs and 24.2% in sows). The herd prevalence was 85.0% (95% CI: 78–92) and within-farm prevalence ranged from 2.9% to 92.8% (median=17.6%). Statistically significant differences were observed among sampling regions with seroprevalence significantly higher in pigs from Valencia Community (27.3%), Extremadura (23.3%) and Catalonia (21.2%). A generalized estimating equations model indicated that the risk factors associated with T. gondii seroprevalence were: age, sows compared to fattening pigs (OR=2.9; 95% CI=1.83–4.53), lack of rodent control (OR=1.9; 95% CI=1.04–3.60) and presence of cats (OR=1.6; 95% CI=1.12–2.34). The seroprevalence observed in the present study indicates a widespread, although variable, exposure to T. gondii among domestic pigs in Spain, which might have important implications for public health. Toxoplasma gondii is a widely distributed zoonotic protozoan parasite, which can affect most warm-blooded species. Some species of non-human primates (NHPs) are highly susceptible to T. gondii infection. The aim of the study was to determine the seroprevalence and risk factors associated with T. gondii infection in NHPs housed in zoos in Spain. Sera from 189 NHPs belonging to 33 species were collected in eight zoos. Additionally, 10 of the 189 animals were longitudinally sampled. Anti- T. gondii antibodies were detected in 48 NHPs (25.4%; confidence interval of 95% (CI 95%): 19.2–31.6) using a modified agglutination test (MAT; cut-off = 25). Seropositive animals had titers of 25 (6.3%), 50 (8.3%), 100 (8.3%) and ≥500 (68.8%). Seropositivity was detected in 15 of the 33 species (45.5%). Of the 10 NHPs sampled more than once, two animals (one Barbary macaque [ Macaca sylvanus ] and one common chimpanzee [ Pan toglodytes ]) seroconverted along the study period, while one seropositive chimpanzee increased antibody titers over time. The Hominidae family (OR = 5.9; CI 95% : 2.7–12.8) and sex (females) (OR = 2.1; CI 95% : 1.1–4.1) were risk factors potentially associated with seropositivity to T. gondii. Our results evince a widespread circulation of T. gondii in NHPs in zoos in Spain, which may be of conservation concern. Toxoplasmosis, caused by the protozoan Toxoplasma gondii , is a worldwide zoonotic disease, which affects most warm-blooded species. Besides the zoonotic relevance, toxoplasmosis is one of the major causes of reproductive disorders in small ruminants. A cross-sectional study was carried out to determine the seroprevalence, spatial distribution and risk factors associated with T. gondii infection in sheep and goats in southern Spain. During 2015–2017, a total of 1,943 small ruminants (998 sheep and 945 goats) from 127 flocks were tested for antibodies against T. gondii using the modified agglutination test (MAT, cut-off 1:25). Antibodies against T. gondii were detected in 464 of the 998 sheep (46.5 %; CI95 %: 43.4–49.6%) and 362 of the 945 goats (38.3 %; CI95 %: 35.2–41.4%) tested. The farm prevalence was 98.4 % (CI95 %: 95.4–100%) for sheep and 93.7 % (CI95 %: 87.6–99.7%) for goats. The generalized estimating equation analysis showed that presence of cats and existence of previous reproductive disorders were risk factors potentially associated with T. gondii seropositivity in small ruminants. Two statistically significant spatial clusters (P < 0.001) were identified. The seroprevalence observed in the present study indicates a widespread exposure to T. gondii in sheep and goats in southern Spain, which might have important implications for animal and public health. • Toxoplasma gondii is widespread in zoo animals in Spain, which could be of public health and conservation concerns. • The feeding habit (carnivorous) was a risk factor associated with T. gondii in captive animals. • Captive zoo animals can be useful sentinel species to monitor T. gondii. A serosurvey was performed to assess exposure to Toxoplasma gondii in zoo animals in Spain and to determine the dynamics of seropositivity in some zoo species over time. Sera from 393 zoo animals belonging to 91 species were collected in eight zoos in Spain between 2007 and 2019. Additionally, 39 of the 393 animals from five of the analyzed zoos were longitudinally sampled during the same study period. Antibodies to T. gondii were detected in 165 (42.0 %; CI95 %: 37.1–46.9) of 393 animals by the modified agglutination test (MAT, cut-off = 25). Antibody titers of 25, 50, 100 and ≥500 were found in 25 (15.2 %), 65 (39.4 %), 26 (15.8 %), and 49 (29.7 %) of the seropositive animals, respectively. Feeding habits (carnivorous species) was a risk factor (OR = 5.6; P < 0.001; CI95 %: 3.8–9.0) potentially associated with T. gondii exposure. Eighteen (46.2 %) of the 39 animals longitudinally sampled were always seropositive and five animals (12.8 %) (two spotted hyaena [ Crocuta crocuta ], one blesbok [ Damaliscus pygargus ], one white rhinoceros [ Ceratotherium simum ] and one mouflon [ Ovis aries musimon ]) seroconverted during the study period. Our results evince a wide circulation of T. gondii in zoos in Spain, including wild felids. Zoo animals can be useful sentinel species to monitor T. gondii circulation in epidemiological scenarios with a close human-wildlife interface. The possibility of Toxoplasma gondii transmitted from game meat to humans is of public health concern. Here we determined seroprevalence and risk factors associated with T. gondii in large game ungulates that cohabit in Sierras de Cazorla, Segura and Las Villas Natural Park (SCSV-NP) (Southern Spain), a natural park with high human-animal interaction. Antibodies against T. gondii in 328 wild ungulates were assayed by the modified agglutination test (MAT ≥ 1:25). Antibodies were found in 39 (11.9%, 95% CI: 8.4-15.4) wild ungulates, with seroprevalence levels of 20.8% in wild boars (Sus scrofa) (5/24), 19.0% in fallow deer (Dama dama) (12/63), 13.9% in Iberian ibex (Capra pyrenaica hispanica) (14/101), 7.9% in red deer (Cervus elaphus) (6/76), and 3.1% in mouflons (Ovis aries musimon) (2/64). Significantly higher seroprevalence was observed in fallow deer and wild boars compared to mouflons. Animals living close to urban areas (<2 km) had 4.6-times higher risk compared to those living at >5 km of urban areas. The results indicate high circulation of T. gondii in wild ungulates in SCSV-NP, which is of animal and public health concern. Toxoplasma gondii is a zoonotic intracellular protozoan parasite of worldwide distribution that infects many species of warm-blooded animals, including birds. To date, there is scant information about the seropositivity of T. gondii and the risk factors associated with T. gondii infection in wild bird populations. In the present study, T. gondii infection was evaluated on sera obtained from 1079 wild birds belonging to 56 species (including Falconiformes (n=610), Strigiformes (n=260), Ciconiiformes (n=156), Gruiformes (n=21), and other orders (n=32), from different areas of Spain. Antibodies to T. gondii (modified agglutination test, MAT titer ≥1:25) were found in 282 (26.1%, IC(95%:)23.5-28.7) of the 1079 birds. This study constitute the first extensive survey in wild birds species in Spain and reports for the first time T. gondii antibodies in the griffon vulture (Gyps fulvus), short-toed snake-eagle (Circaetus gallicus), Bonelli's eagle (Aquila fasciata), golden eagle (Aquila chrysaetos), bearded vulture (Gypaetus barbatus), osprey (Pandion haliaetus), Montagu's harrier (Circus pygargus), Western marsh-harrier (Circus aeruginosus), peregrine falcon (Falco peregrinus), long-eared owl (Asio otus), common scops owl (Otus scops), Eurasian spoonbill (Platalea leucorodia), white stork (Ciconia ciconia), grey heron (Ardea cinerea), common moorhen (Gallinula chloropus); in the International Union for Conservation of Nature (IUCN) "vulnerable" Spanish imperial eagle (Aquila adalberti), lesser kestrel (Falco naumanni) and great bustard (Otis tarda); and in the IUCN "near threatened" red kite (Milvus milvus). The highest seropositivity by species was observed in the Eurasian eagle owl (Bubo bubo) (68.1%, 98 of 144). The main risk factors associated with T. gondii seropositivity in wild birds were age and diet, with the highest exposure in older animals and in carnivorous wild birds. Kennel dogs can serve as sentinels and/or reservoirs of diseases of veterinary and zoonotic interest because they have often roamed free and lived outdoors, thus being exposed to pathogens. We tested dogs from the kennel of Inca (Majorca/Mallorca, Balearic Islands, Spain) for evidence of infection with three protozoan parasites: Leishmania infantum, Toxoplasma gondii, and Neospora caninum. Exposure to L. infantum was found in 56.3% of 48 dogs (37.5% by Western blot, 43.8% by PCR). Only 30% of infected dogs had leishmaniasis-like lesions. Seroprevalence to T. gondii was 58.7% of 46 dogs using the modified agglutination test (MAT, titer 1:25). None of the 44 dogs tested had N. caninum antibodies using a commercial competitive ELISA, probably because the surveyed dogs did not roam in the proximity of cattle farms. Results confirm the endemicity of L. infantum and also the widespread presence of T. gondii in the Mediterranean island of Majorca.
